Abstract

We consider an inpainting model proposed by A. Bertozzi et al., which is based on a Cahn--Hilliard-type equation. This equation describes the evolution of an order parameter $u \in [0,1]$ representing an approximation of the original image which occupies a bounded two-dimensional domain $\Omega$. The given image $g$ is assumed to be damaged in a fixed subdomain $D \subset \Omega$ and the equation is characterized by a linear reaction term of the form $\lambda (u - g)$. Here $\lambda = \lambda_0 \chi_{\Omega \setminus D}$ is the so-called fidelity coefficient, $\lambda_0$ being a strictly positive bounded function. The idea is that, given an initial image $u_0$, $u$ evolves towards $g$ and this process properly diffuses through the boundary of $D$ restoring the damaged image, provided that $\lambda_0$ is large enough. Here, we formulate an optimal control problem based on this fact, namely our cost functional accounts for the magnitude of $\lambda_0$. Assuming a singular potential to assure that $u$ takes its values in $[0,1]$, we first analyse the control-to-state operator and prove the existence of at least one optimal control, establishing the validity of first-order optimality conditions. Then, under suitable assumptions, we demonstrate second-order optimality conditions. Abstract

In this paper, we tackle the problem of reconstructing earlier tumour configurations starting from a single spatial measurement at a later time. We describe the tumour evolution through a diffuse interface model coupling a Cahn-Hilliard-type equation for the tumour phase field to a reaction-diffusion equation for a key nutrient proportion, also accounting for chemotaxis effects. We stress that the ability to reconstruct earlier tumour states is crucial for calibrating the model used to predict the tumour dynamics and also to identify the areas where the tumour initially began to develop. However, backward-in-time inverse problems are well-known to be severely ill-posed, even for linear parabolic equations. Moreover, we also face additional challenges due to the complexity of a non-linear fourth-order parabolic system. Nonetheless, we can establish uniqueness by using logarithmic convexity methods under suitable a priori assumptions. To further address the ill-posedness of the inverse problem, we propose a Tikhonov regularisation approach that approximates the solution through a family of constrained minimisation problems. For such problems, we analytically derive the first-order necessary optimality conditions. Finally, we develop a computationally efficient numerical approximation of the optimisation problems by employing standard $C^0$-conforming first-order finite elements. We conduct numerical experiments on several pertinent test cases and observe that the proposed algorithm consistently meets expectations, delivering accurate reconstructions of the original ground truth., Comment: 54 pages

Abstract

The development of mathematical models of cancer informed by time-resolved measurements has enabled personalised predictions of tumour growth and treatment response. However, frequent cancer monitoring is rare, and many tumours are treated soon after diagnosis with limited data. To improve the predictive capabilities of cancer models, we investigate the problem of recovering earlier tumour states from a single spatial measurement at a later time. Focusing on prostate cancer, we describe tumour dynamics using a phase-field model coupled with two reaction-diffusion equations for a nutrient and the local prostate-specific antigen. We generate synthetic data using a discretisation based on Isogeometric Analysis. Then, building on our previous analytical work (Beretta et al., SIAP (2024)), we propose an iterative reconstruction algorithm based on the Landweber scheme, showing local convergence with quantitative rates and exploring an adaptive step size that leads to faster reconstruction algorithms. Abstract

The availability of cancer measurements over time enables the personalised assessment of tumour growth and therapeutic response dynamics. However, many tumours are treated after diagnosis without collecting longitudinal data, and cancer monitoring protocols may include infrequent measurements. To facilitate the estimation of disease dynamics and better guide ensuing clinical decisions, we investigate an inverse problem enabling the reconstruction of earlier tumour states by using a single spatial tumour dataset and a biomathematical model describing disease dynamics. We focus on prostate cancer, since aggressive cases of this disease are usually treated after diagnosis. We describe tumour dynamics with a phase-field model driven by a generic nutrient ruled by reaction-diffusion dynamics. The model is completed with another reaction-diffusion equation for the local production of prostate-specific antigen, which is a key prostate cancer biomarker. We first improve previous well-posedness results by further showing that the solution operator is continuously Fr\'echet differentiable. We then analyse the backward inverse problem concerning the reconstruction of earlier tumour states starting from measurements of the model variables at the final time. Since this problem is severely ill-posed, only very weak conditional stability of logarithmic type can be recovered from the terminal data. However, by restricting the unknowns to a compact subset of a finite-dimensional subspace, we can derive an optimal Lipschitz stability estimate., Comment: 25 pages

Abstract

We present the latest generation of MobileNets, known as MobileNetV4 (MNv4), featuring universally efficient architecture designs for mobile devices. At its core, we introduce the Universal Inverted Bottleneck (UIB) search block, a unified and flexible structure that merges Inverted Bottleneck (IB), ConvNext, Feed Forward Network (FFN), and a novel Extra Depthwise (ExtraDW) variant. Alongside UIB, we present Mobile MQA, an attention block tailored for mobile accelerators, delivering a significant 39% speedup. An optimized neural architecture search (NAS) recipe is also introduced which improves MNv4 search effectiveness. The integration of UIB, Mobile MQA and the refined NAS recipe results in a new suite of MNv4 models that are mostly Pareto optimal across mobile CPUs, DSPs, GPUs, as well as specialized accelerators like Apple Neural Engine and Google Pixel EdgeTPU - a characteristic not found in any other models tested. Finally, to further boost accuracy, we introduce a novel distillation technique. Enhanced by this technique, our MNv4-Hybrid-Large model delivers 87% ImageNet-1K accuracy, with a Pixel 8 EdgeTPU runtime of just 3.8ms.

7. Maximal regularity and optimal control for a non-local Cahn-Hilliard tumour growth model

Abstract

We consider a non-local tumour growth model of phase-field type, describing the evolution of tumour cells through proliferation in presence of a nutrient. The model consists of a coupled system, incorporating a non-local Cahn-Hilliard equation for the tumour phase variable and a reaction-diffusion equation for the nutrient. First, we establish novel regularity results for such a model, by applying maximal regularity theory in weighted $L^p$ spaces. This technique enables us to prove the local existence and uniqueness of a regular solution, including also chemotaxis effects. By leveraging time-regularisation properties and global boundedness estimates, we further extend the solution to a global one. These results provide the foundation for addressing an optimal distributed control problem, aimed at identifying a suitable therapy guiding the tumour towards a predefined target. Abstract

In this paper, we address an optimal distributed control problem for a non-local model of phase-field type, describing the evolution of tumour cells in presence of a nutrient. The model couples a non-local and viscous Cahn-Hilliard equation for the phase parameter with a reaction-diffusion equation for the nutrient. The optimal control problem aims at finding a therapy, encoded as a source term in the system, both in the form of radiotherapy and chemotherapy, which could lead to the evolution of the phase variable towards a desired final target. First, we prove strong well-posedness for the system of non-linear partial differential equations. In particular, due to the presence of a viscous regularisation, we can also consider double-well potentials of singular type and cross-diffusion terms related to the effects of chemotaxis. Moreover, the particular structure of the reaction terms allows us to prove new regularity results for this kind of system. Abstract

Weakly supervised object localization (WSOL) aims to learn representations that encode object location using only image-level category labels. However, many objects can be labeled at different levels of granularity. Is it an animal, a bird, or a great horned owl? Which image-level labels should we use? In this paper we study the role of label granularity in WSOL. To facilitate this investigation we introduce iNatLoc500, a new large-scale fine-grained benchmark dataset for WSOL. Surprisingly, we find that choosing the right training label granularity provides a much larger performance boost than choosing the best WSOL algorithm. Abstract

Abstract Background The optimal management of COVID-19 symptoms and their sequelae remains an important area of clinical research. Policy makers have little scientific data regarding the effects on the daily life of affected individuals and the identification of their needs. Such data are needed to inform effective care policy. Methods We studied 639 people with COVID-19 resident in France via an online questionnaire. They reported their symptoms, effects on daily life, and resulting needs, with particular focus on olfaction. Results The results indicate that a majority of participants viewed their symptoms as disabling, with symptoms affecting their physical and mental health, social and professional lives. 60% of the individuals reported having unmet medical, psychological and socio-professional support needs. Finally, affected individuals were concerned about the risk and invasiveness of possible treatments as shown by a preference for non-invasive intervention over surgery to cure anosmia. Conclusions It is important that policy makers take these needs into consideration in order to assist affected individuals to regain a normal quality of life.

Abstract

We find extremely general classes of nonsmooth open sets which guarantee Mosco convergence for corresponding Sobolev spaces and the validity of Sobolev inequalities with a uniform constant. An important feature of our results is that the conditions we impose on the open sets for Mosco convergence and for the Sobolev inequalities are of the same nature, therefore it is easy to check when both are satisfied. Our analysis is motivated, in particular, by the study of the stability of the direct acoustic scattering problem with respect to the scatterer, which we also discuss. Concerning Mosco convergence in dimension 3 or higher, our result extends all those previously known in the literature. Abstract

When interacting with objects through cameras, or pictures, users often have a specific intent. For example, they may want to perform a visual search. With most object detection models relying on image pixels as their sole input, undesired results are not uncommon. Most typically: lack of a high-confidence detection on the object of interest, or detection with a wrong class label. The issue is especially severe when operating capacity-constrained mobile object detectors on-device. In this paper we investigate techniques to modulate mobile detectors to explicitly account for the user intent, expressed as an embedding of a simple query. Compared to standard detectors, query-modulated detectors show superior performance at detecting objects for a given user query. Thanks to large-scale training data synthesized from standard object detection annotations, query-modulated detectors also outperform a specialized referring expression recognition system. Query-modulated detectors can also be trained to simultaneously solve for both localizing a user query and standard detection, even outperforming standard mobile detectors at the canonical COCO task.

Abstract

Urbanization transforms environments in ways that alter biological evolution. We examined whether urban environmental change drives parallel evolution by sampling 110,019 white clover plants from 6169 populations in 160 cities globally. Plants were assayed for a Mendelian antiherbivore defense that also affects tolerance to abiotic stressors. Urban-rural gradients were associated with the evolution of clines in defense in 47% of cities throughout the world. Variation in the strength of clines was explained by environmental changes in drought stress and vegetation cover that varied among cities. Sequencing 2074 genomes from 26 cities revealed that the evolution of urban-rural clines was best explained by adaptive evolution, but the degree of parallel adaptation varied among cities. Our results demonstrate that urbanization leads to adaptation at a global scale.

Abstract

Deep learning based object detectors are commonly deployed on mobile devices to solve a variety of tasks. For maximum accuracy, each detector is usually trained to solve one single specific task, and comes with a completely independent set of parameters. While this guarantees high performance, it is also highly inefficient, as each model has to be separately downloaded and stored. In this paper we address the question: can task-specific detectors be trained and represented as a shared set of weights, plus a very small set of additional weights for each task? The main contributions of this paper are the following: 1) we perform the first systematic study of parameter-efficient transfer learning techniques for object detection problems; 2) we propose a technique to learn a model patch with a size that is dependent on the difficulty of the task to be learned, and validate our approach on 10 different object detection tasks. Abstract

Introduction: The non-neoplastic kidney parenchyma from nephrectomies is often overlooked in routine examinations. We aimed to evaluate the associations between global glomerulosclerosis (GS), interstitial fibrosis (IF), or arteriosclerosis (AS) and estimated glomerular filtration rate (eGFR), dipstick proteinuria, and other clinical factors. Methods: We performed a cross-sectional analysis of 781 patients with nephrectomy. We used regression models with and without interaction factors. The tested exposures were GS, IF, or AS, and the outcome measures were GFR and dipstick proteinuria. Results: In multivariable analyses, increasing degrees of GS, IF, or AS were significantly associated with lower eGFR and proteinuria (p < 0.05 for each). Obesity and hypertension (HTN) modified the association between eGFR and degrees of GS, whereas proteinuria and cardiovascular disease (CVD) modified the association between eGFR and degrees of AS (p for interaction 50% was associated with lower eGFR in patients with (−45 mL/min/1.73 m2) than without (−19 mL/min/1.73 m2) obesity, and GS >50% was associated with lower eGFR in patients with (−31 mL/min/1.73 m2) than without (−16 mL/min/1.73 m2) HTN. Compared with AS 50% was associated with lower eGFR in patients with (−11 mL/min/1.73 m2) than without (−6 mL/min/1.73 m2) proteinuria, and AS >50% was associated with lower eGFR in patients with (−23 mL/min/1.73 m2) than without (−7 mL/min/1.73 m2) CVD. Conclusion: Greater degrees of each GS, IF, and AS are independently associated with proteinuria and lower eGFR. Obesity, HTN, proteinuria, and CVD modify the relationship between eGFR and specific histopathological features of nephrosclerosis.

33. Development of Auditory Cognition in 5- to 10-Year-Old Children: Focus on Musical and Verbal Short-Term Memory

Abstract

Developmental aspects of auditory cognition were investigated in 5-to-10-year-old children (n = 100). Musical and verbal short-term memory (STM) were assessed by means of delayed matching-to-sample tasks (DMST) (comparison of two four-item sequences separated by a silent retention delay), with two levels of difficulty. For musical and verbal materials, children's performance increased from 5 years to about 7 years of age, then remained stable up to 10 years of age, with performance remaining inferior to performance of young adults. Children and adults performed better with verbal material than with musical material. To investigate auditory cognition beyond STM, we assessed speech-in-noise perception with a four-alternative forced-choice task with two conditions of phonological difficulty and two levels of cocktail-party noise intensity. Partial correlations, factoring out the effect of age, showed a significant link between musical STM and speech-in-noise perception in the condition with increased noise intensity. Our findings reveal that auditory STM improves over development with a critical phase around 6-7 years of age, yet these abilities appear to be still immature at 10 years. Musical and verbal STM might in particular share procedural and serial order processes. Furthermore, musical STM and the ability to perceive relevant speech signals in cocktail-party noise might rely on shared cognitive resources, possibly related to pitch encoding. To the best of our knowledge, this is the first time that auditory STM is assessed with the same paradigm for musical and verbal material during childhood, providing perspectives regarding diagnosis and remediation in developmental learning disorders.

Abstract

Beyond the direct benefit that a transplanted organ provides to an individual recipient, the study of the transplant process has the potential to create a better understanding of the pathogenesis, etiology, progression and possible therapy for recurrence of disease after transplantation while at the same time providing insight into the original disease. Specific examples of this include: 1) recurrence of focal segmental glomerulosclerosis (FSGS) after kidney transplantation, 2) recurrent autoimmunity after pancreas transplantation, and 3) recurrence of disease after orthotopic liver transplantation (OLT) for cirrhosis related to progressive steatosis secondary to jejuno-ileal bypass (JIB) surgery. Our team has been studying these phenomena and their immunologic underpinnings, and we suggest that expanding the concept to other pathologic processes and/or transplanted organs that harbor the risk for recurrent disease may provide novel insight into the pathogenesis of a host of other disease processes that lead to organ failure.

Abstract

Background Knowledge of the physical and environmental conditions that may limit the migration of invasive species is crucial to assess the potential for expansion outside their native ranges. The cactus moth, Cactoblastis cactorum, is native to South America (Argentina, Paraguay, Uruguay and Brazil) and has been introduced and invaded the Caribbean and southern United States, among other regions. In North America there is an ongoing process of range expansion threatening cacti biodiversity of the genus Opuntia and the commercial profits of domesticated Opuntia ficus-indica. Methods To further understand what influences the distribution and genetic structure of this otherwise important threat to native and managed ecosystems, in the present study we combined ecological niche modeling and population genetic analyses to identify potential environmental barriers in the native region of Argentina. Samples were collected on the host with the wider distribution range, O. ficus-indica. Results Significant genetic structure was detected using 10 nuclear microsatellites and 24 sampling sites. At least six genetic groups delimited by mountain ranges, salt flats and wetlands were mainly located to the west of the Dry Chaco ecoregion. Niche modeling supports that this region has high environmental suitability where the upper soil temperature and humidity, soil carbon content and precipitation were the main environmental factors that explain the presence of the moth. Environmental filters such as the upper soil layer may be critical for pupal survival and consequently for the establishment of populations in new habitats, whereas the presence of available hosts is a necessary conditions for insect survival, upper soil and climatic characteristics will determine the opportunities for a successful establishment.

39. ABCA1 deficiency contributes to podocyte pyroptosis priming via the APE1/IRF1 axis in diabetic kidney disease

Abstract

Abstract Decreased ATP Binding Cassette Transporter A1 (ABCA1) expression and caspase-4-mediated noncanonical inflammasome contribution have been described in podocytes in diabetic kidney disease (DKD). To investigate a link between these pathways, we evaluated pyroptosis-related mediators in human podocytes with stable knockdown of ABCA1 (siABCA1) and found that mRNA levels of IRF1, caspase-4, GSDMD, caspase-1 and IL1β were significantly increased in siABCA1 compared to control podocytes and that protein levels of caspase-4, GSDMD and IL1β were equally increased. IRF1 knockdown in siABCA1 podocytes prevented increases in caspase-4, GSDMD and IL1β. Whereas TLR4 inhibition did not decrease mRNA levels of IRF1 and caspase-4, APE1 protein expression increased in siABCA1 podocytes and an APE1 redox inhibitor abrogated siABCA1-induced expression of IRF1 and caspase-4. RELA knockdown also offset the pyroptosis priming, but ChIP did not demonstrate increased binding of NFκB to IRF1 promoter in siABCA1 podocytes. Finally, the APE1/IRF1/Casp1 axis was investigated in vivo. APE1 IF staining and mRNA levels of IRF1 and caspase 11 were increased in glomeruli of BTBR ob/ob compared to wildtype. In conclusion, ABCA1 deficiency in podocytes caused APE1 accumulation, which reduces transcription factors to increase the expression of IRF1 and IRF1 target inflammasome-related genes, leading to pyroptosispriming.

Abstract

Background Albuminuria is a known marker of mortality risk. Whether the association between albuminuria and mortality differs by demographic and comorbidity factors remains unclear. Therefore, we sought to determine whether albuminuria is differentially associated with mortality. Methods and Results This study included 49 640 participants from the National Health and Nutrition Examination Survey (1999–2018). All‐cause mortality through 2019 was linked from the National Death Index. Multivariable‐adjusted Poisson regression models were used to determine whether levels of urine albumin‐to‐creatinine ratio (ACR) were associated with mortality. Models were adjusted for demographic, socioeconomic, behavioral, and clinical factors. Mean age in the population was 46 years, with 51.3% female, and 30.3% with an ACR ≥10 mg/g. 47. Adenine crosses the biomarker bridge: from ’omics to treatment in diabetic kidney disease

Abstract

Enabling the early detection and prevention of diabetic kidney damage has potential to substantially reduce the global burden of kidney failure. There is a critical need for identification of mechanistic biomarkers that can predict progression and serve as therapeutic targets. In this issue of the JCI, Sharma and colleagues used an integrated multiomics approach to identify the metabolite adenine as a noninvasive biomarker of progression in early diabetic kidney disease (DKD). The highest tertile of urine adenine/creatinine ratio (UAdCR) was associated with higher risk for end-stage kidney disease and mortality across independent cohorts, including participants with early DKD without macroalbuminuria. Spatial metabolomics, single-cell transcriptomics, and experimental studies localized adenine to regions of tubular pathology and implicated the mTOR pathway in adenine-mediated tissue fibrosis. Inhibition of endogenous adenine production was protective in a diabetic model. These findings exemplify the potential for multiomics to uncover mechanistic biomarkers and targeted therapies in DKD.

Abstract

V̇O2max is recognized as a key measure in exercise physiology and sports medicine. However, only 20–50% of maximal incremental exercise tests (IET) result in a plateau of V̇O2 (V̇O2pl). To our knowledge, no study has yet examined the possible difference in brain activity during an IET, in V̇O2pl and non-plateau athletes with the same V̇O2max and age. This study aimed to shed light on the central governor hypothesis, namely that the inability to reach a V̇O2pl may be dictated by the brain rather than by a peripheral physical limit. This hypothesis can now be explored using electroencephalography (EEG) during IET, measuring concomitant power in specific frequency bands. Forty-two athletes were divided into two groups: those who practiced endurance sports and those who did not, and were asked to perform an IET. EEG signals and gas exchange were recorded. A V̇O2pl was observed in twenty-two subjects (52%). EEG power increased in all subjects during IET, except in the alpha band, which showed variability, but not significantly (64% increase, 34% decrease, p = 0.07). No differences were found between endurance athletes and non-endurance athletes, except for V̇O2max (60.10 ± 6.16 vs. 51.77 ± 6.41, p < 0.001). However, the baseline-corrected ratio of EEG power to V̇O2 was found to decrease in all subjects during IET, in the alpha, beta and theta bands. In conclusion, the presence or absence of a V̇O2pl is not related to the type of EEG response during an IET. Nevertheless, the decline in brain and V̇O2 powers/ratios in all frequency bands suggests that aerobic power may be constrained by brain mobilization.

Abstract

Podocyte health is vital for maintaining proper glomerular filtration in the kidney. Interdigitating foot processes from podocytes form slit diaphragms which regulate the filtration of molecules through size and charge selectivity. The abundance of lipid rafts, which are ordered membrane domains rich in cholesterol and sphingolipids, near the slit diaphragm highlights the importance of lipid metabolism in podocyte health. Emerging research shows the importance of sphingolipid metabolism to podocyte health through structural and signaling roles. Dysregulation in sphingolipid metabolism has been shown to cause podocyte injury and drive glomerular disease progression. In this review, we discuss the structure and metabolism of sphingolipids, as well as their role in proper podocyte function and how alterations in sphingolipid metabolism contributes to podocyte injury and drives glomerular disease progression.
