Abstract

A multifidelity method for the nonlinear propagation of uncertainties in the presence of stochastic accelerations is presented. The proposed algorithm treats the uncertainty propagation (UP) problem by separating the propagation of the initial uncertainty from that of the process noise. The initial uncertainty is propagated using an adaptive Gaussian mixture model (GMM) method which exploits a low-fidelity dynamical model to minimize the computational costs. The effects of process noise are instead computed using the PoLynomial Algebra Stochastic Moments Analysis (PLASMA) technique, which considers a high-fidelity model of the stochastic dynamics. The main focus of the paper is on the latter and on the key idea to approximate the probability density function (pdf) of the solution by a polynomial representation of its moments, which are efficiently computed using differential algebra (DA) techniques. The two estimates are finally combined to restore the accuracy of the low-fidelity surrogate and account for both sources of uncertainty. Abstract

Abstract Peripheral T-cell lymphoma, not otherwise specified (PTCL-NOS) is a heterogeneous group of malignancies with poor outcome. Here, we identify a subgroup, PTCL-NOS SMARCB1- , which is characterized by the lack of the SMARCB1 protein and occurs more frequently in young patients. Human and murine PTCL-NOS SMARCB1- show similar DNA methylation profiles, with hypermethylation of T-cell-related genes and hypomethylation of genes involved in myeloid development. Single-cell analyses of human and murine tumors revealed a rich and complex network of interactions between tumor cells and an immunosuppressive and exhausted tumor microenvironment (TME). In a drug screen, we identified histone deacetylase inhibitors (HDACi) as a class of drugs effective against PTCL-NOS Smarcb1- . In vivo treatment of mouse tumors with SAHA, a pan-HDACi, triggered remodeling of the TME, promoting replenishment of lymphoid compartments and reversal of the exhaustion phenotype. These results provide a rationale for further exploration of HDACi combination therapies targeting PTCL-NOS SMARCB1- within the TME.

8. Receptivity of compressible boundary layers over porous surfaces

Abstract

Supersonic pre-transitional boundary layers flowing over porous flat and concave surfaces are studied using numerical and asymptotic methods. The porous wall is composed of thin equally-spaced cylindrical microcavities. The flow is perturbed by small-amplitude, free-stream vortical disturbances of the convected gust type. From the proximity of the leading edge, these external agents generate the compressible Klebanoff modes, i.e. low-frequency disturbances of the kinematic and thermal kind that grow algebraically downstream. For Klebanoff modes with a spanwise wavelength comparable with the boundary-layer thickness, the porous surface has a negligible effect on their growth. When the spanwise wavelength is instead larger than the boundary-layer thickness, these disturbances are effectively attenuated by the porous surface. For a specified set of frequency and wavelengths, the Klebanoff modes evolve into oblique Tollmien-Schlichting waves through a leading-edge-adjustment receptivity mechanism. The wavenumber of these waves is only slightly modified over the porous surface, while the growth rate increases, thus confirming previous experimental results. An asymptotic analysis based on the triple-deck theory confirms these numerical findings. When the wall is concave, the amplitude of the Klebanoff modes is enhanced by the wall curvature and is attenuated by the wall porosity during the initial development., Comment: Accepted for publication in Physical Review Fluids. 43 pages, 15 figures

Abstract

An algorithm for robust initial orbit determination (IOD) under perturbed orbital dynamics is presented. By leveraging map inversion techniques defined in the algebra of Taylor polynomials, this tool returns a highly accurate solution to the IOD problem and estimates a range centered on the aforementioned solution in which the true orbit should lie. To meet the specified accuracy requirements, automatic domain splitting is used to wrap the IOD routines and ensure that the local truncation error, introduced by a polynomial representation of the state estimate, remains below a predefined threshold. The algorithm is presented for three types of ground-based sensors, namely range radars, Doppler-only radars, and optical telescopes, by considering their different constraints in terms of available measurements and sensor noise. Finally, the improvement in performance with respect to a Keplerian-based IOD solution is demonstrated using large-scale numerical simulations over a subset of tracked objects in low Earth orbit., Comment: accepted for publication on Astrodynamics

CellProfiler is a widely used software for creating reproducible, reusable image analysis workflows without needing to code. In addition to the >90 modules that make up the main CellProfiler program, CellProfiler has a plugins system that allows for the creation of new modules which integrate with other Python tools or tools that are packaged in software containers. The CellProfiler-plugins repository contains a number of these CellProfiler modules, especially modules that are experimental and/or dependency-heavy. 15. A low-order automatic domain splitting approach for nonlinear uncertainty mapping

Abstract

This paper introduces a novel method for the automatic detection and handling of nonlinearities in a generic transformation. A nonlinearity index that exploits second order Taylor expansions and polynomial bounding techniques is first introduced to rigorously estimate the Jacobian variation of a nonlinear transformation. This index is then embedded into a low-order automatic domain splitting algorithm that accurately describes the mapping of an initial uncertainty set through a generic nonlinear transformation by splitting the domain whenever some imposed linearity constraints are non met. The algorithm is illustrated in the critical case of orbital uncertainty propagation, and it is coupled with a tailored merging algorithm that limits the growth of the domains in time by recombining them when nonlinearities decrease. The low-order automatic domain splitting algorithm is then combined with Gaussian mixtures models to accurately describe the propagation of a probability density function. A detailed analysis of the proposed method is presented, and the impact of the different available degrees of freedom on the accuracy and performance of the method is studied.

16. A Multifidelity Approach to Robust Orbit Determination

Abstract

This paper presents an algorithm for the preprocessing of observation data aimed at improving the robustness of orbit determination tools. Two objectives are fulfilled: obtain a refined solution to the initial orbit determination problem and detect possible outliers in the processed measurements. The uncertainty on the initial estimate is propagated forward in time and progressively reduced by exploiting sensor data available in said propagation window. Differential algebra techniques and a novel automatic domain splitting algorithm for second-order Taylor expansions are used to efficiently propagate uncertainties over time. A multifidelity approach is employed to minimize the computational effort while retaining the accuracy of the propagated estimate. At each observation epoch, a polynomial map is obtained by projecting the propagated states onto the observable space. Domains that do no overlap with the actual measurement are pruned thus reducing the uncertainty to be further propagated. Measurement outliers are also detected in this step. The refined estimate and retained observations are then used to improve the robustness of batch orbit determination tools. The effectiveness of the algorithm is demonstrated for a geostationary transfer orbit object using synthetic and real observation data from the TAROT network., Comment: accepted for publication in Acta Astronautica

The use of manual vs automatic segmentation in the performance of survival radiomic models was assessed, as well. METHODS A total of 899 NSCLC patients were included (2 proprietary: A and B, 1 public datasets: C). Automatic segmentation of lung lesions was performed by training a previously developed architecture, the nnU-Net, including 2D, 3D and cascade approaches. The quality of automatic segmentation was evaluated with DICE coefficient, considering manual contours as reference. The impact of automatic segmentation on the performance of a radiomic model for patient survival was explored by extracting radiomic hand-crafted and deep-learning features from manual and automatic contours of dataset A, and feeding different machine learning algorithms to classify survival above/below median. Models' accuracies were assessed and compared. RESULTS The best agreement between automatic and manual contours with DICE=0.78 +(0.12) was achieved by averaging predictions from 2D and 3D models, and applying a post-processing technique to extract the maximum connected component. No statistical differences were observed in the performances of survival models when using manual or automatic contours, hand-crafted, or deep features. The best classifier showed an accuracy between 0.65 and 0.78. CONCLUSION The promising role of nnU-Net for automatic segmentation of lung lesions was confirmed, dramatically reducing the time-consuming physicians' workload without impairing the accuracy of survival predictive models based on radiomics.

Abstract

This paper presents the solution approach developed by the team "theAntipodes" for the 11th Global Trajectory Optimization Competition (GTOC11). The approach consists of four main blocks: 1) mothership chain generation, 2) rendezvous table generation, 3) the dispatcher, and 4) the refinement. Blocks 1 and 3 are purely combinatorial optimization problems that select the asteroids to visit and allocate them to the Dyson ring stations. The rendezvous table generation involves interpolating time-optimal transfers to find all transfer opportunities between selected asteroids and the ring stations. The dispatcher uses the data stored in the table and allocates the asteroids to the Dyson ring stations optimally. The refinement ensures each rendezvous trajectory meets the problem accuracy constraints and introduces deep-space maneuvers to the mothership transfers. Background and objective: Registry-based studies for prostate cancer (PCa) document higher overall mortality (OM) after high-dose radiotherapy (RT) than after radical prostatectomy (RP). Our aim was to explore the association between pretreatment patient-reported health (“OverallHealth”: OH) and curative treatment type, and the impact on early OM. Methods: New PCa patients registered between 2017 and 2019 in the Cancer Registry of Norway (n = 1949) completed the European Organisation for Research and Treatment of Cancer Quality-of-Life Core 30 (QLQ-C30) questionnaire before RP (n = 592) or RT (n = 610) or after allocation to active surveillance (AS; n = 747). Abstract

A new multifidelity method is developed for nonlinear orbit uncertainty propagation. This approach guarantees improved computational efficiency and limited accuracy losses compared to fully high-fidelity counterparts. The initial uncertainty is modeled as a weighted sum of Gaussian distributions whose number is adapted online to satisfy the required accuracy. As needed, univariate splitting libraries are used to split the mixture components along the direction of maximum nonlinearity. Differential Algebraic techniques are used to propagate these Gaussian kernels and compute a measure of nonlinearity required for the split decision and direction identification. Taylor expansions of the flow of the dynamics are computed using a low-fidelity dynamical model to maximize computational efficiency and corrected with selected high-fidelity samples to minimize accuracy losses. Abstract

Background and purpose: There are few studies of personality traits in long-term Hodgkin lymphoma survivors (HLSs) treated according to contemporary stage-and risk-adapted approaches. The Distressed Personality (DP) Scale covers negative affectivity and social inhibition. We examined differences in self-reported late adverse effects (LAEs) between HLSs with and without DP and other explanatory variables. Material and methods: This cross-sectional questionnaire-based study included a population-based cohort of HLSs treated from 1997 to 2006, aged 8–49 years at diagnosis, and alive in 2016. Among 518 eligible HLSs, 303 responded (58%), and 294 completed the DP scale. DP was defined by scores above cut-off on both the negative affectivity and social inhibition subscales. LAEs studied were major depression, posttraumatic stress disorder, sleep problems, obesity, neuropathy, fatigue, memory problems, and general health. DP and 10 other explanatory variables were tested against LAEs as dependent variables in multivariable regression analyses. Results: The mean age at survey was 45.9 years (standard deviation [SD] 4.6), mean follow-up time 16.7 years (SD 3.0), and 48% were females. Eighty-two HLSs had DP (28%, 95% confidence interval 23% – 33%). All LAEs except obesity were significantly more common/had higher mean score in HLSs with DP. In multivariable analyses, presence of DP was significantly associated with all LAEs except obesity. Interpretation: The presence of DP is common among HLSs. The presence of DP was associated with most self-report LAEs examined. Including assessment of personality traits in the survivorship care plans of HLSs should be considered. Prospective studies assessing the influence of pretreatment DP on LAEs are warranted.
