Retrotransposons copy their sequences via an RNA intermediate, followed by reverse transcription into cDNA and random insertion, into a new genomic locus. New retrotransposon copies may lead to cell transformation and/or tumorigenesis through insertional mutagenesis. Methylation is a major defense mechanism against retrotransposon RNA expression and retrotransposition in differentiated cells, whereas stem cells are relatively hypo‑methylated. Epithelial‑to‑mesenchymal transition (EMT), which transforms normal epithelial cells into mesenchymal‑like cells, also contributes to tumor progression and tumor metastasis. Cancer stem cells (CSCs), a fraction of undifferentiated tumor‑initiating cancer cells, are reciprocally related to EMT. In the present study, the outcome of long terminal repeat (LTR)‑Viral‑Like 30 (VL30) retrotransposition was examined in mouse mammary stem‑like/progenitor HC11 epithelial cells. The transfection of HC11 cells with a VL30 retrotransposon, engineered with an EGFP‑based retrotransposition cassette, elicited a higher retrotransposition frequency in comparison to differentiated J3B1A and C127 mouse mammary cells. Fluorescence microscopy and PCR analysis confirmed the specificity of retrotransposition events. The differentiated retrotransposition‑positive cells retained their epithelial morphology, while the respective HC11 cells acquired mesenchymal features associated with the loss of E‑cadherin, the induction of N‑cadherin, and fibronectin and vimentin protein expression, as well as an increased transforming growth factor (TGF)‑β1, Slug, Snail‑1 and Twist mRNA expression. In addition, they were characterized by cell proliferation in low serum, and the acquisition of CSC‑like properties indicated by mammosphere formation under anchorage‑independent conditions. Mammospheres exhibited an increased Nanog and Oct4 mRNA expression and a CD44+/CD24‑/low antigenic phenotype, as well as self‑renewal and differentiation capacity, forming mammary acini‑like structures. DNA sequencing analysis of retrotransposition‑positive HC11 cells revealed retrotransposed VL30 copies integrated at the vicinity of EMT‑, cancer type‑ and breast cancer‑related genes. The inoculation of these cells into Balb/c mice produced cytokeratin‑positive tumors containing pancytokeratin‑positive cells, indicative of cell invasion features. On the whole, the findings of the present study demonstrate, for the first time, to the best of our knowledge, that stem‑like epithelial HC11 cells are amenable to VL30 retrotransposition associated with the induction of EMT and CSC generation, leading to tumorigenesis.

Glucosylated liposomes composed of the natural saturated phospholipid 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC), cholesterol (Chol) and a cationic amphiphile featuring a glucosyl moiety (GL4), have been developed for delivering the antimicrobial trans-Resveratrol (RSV) to S. epidermidis, characterized by carbohydrate-specific adhesins able to recognize glucose. The cationic derivative of cholesterol, DC-Chol, was also included in liposome formulations, alone or in combination with GL4, in order to explore the role of both cationic charge and sugar moiety in the interaction of liposomes with bacterial cells. RSV was included inside glucosylated cationic liposomes by the thin film method, coupled with either extrusion or sonication; liposome mean diameter, polydispersity index, surface charge, RSV entrapment efficiency and concentration have been measured by DLS, electrophoretic mobility, and HPLC. The antimicrobial activity of RSV-loaded liposomes was evaluated by monitoring the bacterial growth curves of two cell lines of Staphylococcus epidermidis, a slime positive strain (i.e. a strain able to form a biofilm) and a slime negative one. Results point out that, when the glucosylamphiphile GL4 is included in the formulation, only the extrusion protocol allows obtaining monodisperse liposomes with high RSV entrapment efficiency. The mean diameters of empty and resveratrol-loaded liposomes are all around 120-140 nm and size distribution are narrow, except for samples including GL4 at 5 molar percentage. Here the higher polydispersity index may be the indication of the occurrence of a restructuring phenomenon. The microbiological tests put in evidence a different response of the two bacterial cell lines to liposome treatments, in fact, the slime negative bacterial cells, that are not able to produce the extracellular polymeric substances, are more susceptible to the cationic charge of the liposomes and to the detergent effect of GL4. The most interesting results concern DPPC/Chol/GL4 liposomes on the slime positive strain: this formulation, non-toxic in itself, displays an enhanced antibacterial efficacy with respect to free RSV, killing bacteria even at concentration tenfold under the MIC.

Tinzaparin is an anticoagulant and antiangiogenic drug with inhibitory properties against tumor growth. VEGF stimulates angiogenesis, while an association between reactive oxygen species (ROS) and angiogenesis is involved in tumor progression. The present study aimed to investigate the effect of tinzaparin on VL30 retrotransposition-positive mouse HC11 mammary stem-like epithelial cells, previously reported to be associated with induced mammosphere/cancer stem cell (CSC) generation and tumorigenesis. Under 24 h serum starvation, 15.2% nominal retrotransposition frequency was increased to 29%. Additionally, while treatment with 3–12 ng/ml VEGF further induced retrotransposition frequency in a dose-dependent manner (up to 40.3%), pre-incubation with tinzaparin (2 IU/ml) for 0.5–4 h reduced this frequency to 18.3% in a time-dependent manner, confirmed by analogous results in NIH3T3 fibroblasts. Treatment with 10–40 pg/ml glucose oxidase (GO) for 24 h induced HC11 cell retrotransposition in a dose-dependent manner (up to 82.5%), while a 3 h pre-incubation with tinzaparin (1 or 2 IU/ml) elicited a 13.5 or 25.5% reduction in retrotransposition, respectively. Regarding tumorigenic VL30 retrotransposition-positive HC11 cells, treatment with 2 IU/ml tinzaparin for 5 days reduced proliferation rate in a time-dependent manner (up to ~55%), and after 3 weeks, disaggregated soft agar-formed foci, as well as low-adherent mammospheres, producing single mesenchymal-like cells with a ~50% reduced retrotransposition. With respect to the VL30 retrotransposition mechanism: While 12 ng/ml VEGF increased the level of VL30 and endogenous reverse transcriptase (enRT) transcripts ~1.41- and ~1.16-fold, respectively, subsequent tinzaparin treatment reduced both endogenous/ROS- and VEGF-induced levels 1.15- and 0.40-fold (VL30) and 0.60- and 0.52-fold (enRT), respectively. To the best of our knowledge, these data demonstrate for the first time, the novel inhibition activity of tinzaparin against ROS- and VEGF-induced VL30 retrotransposition, and the proliferation and/or aggregation of mouse HC11 mammosphere/tumor-initiating CSCs, thus contributing to the inhibition of VL30 retrotransposition-induced primary tumor growth.

Οι ανθρώπινοι ενδογενείς ρετροϊοί (HERVs) θεωρούνται LTR-ρετροτρανσποζόνια ικανά να εισάγουν νέα τους αντίγραφα μέσω ενός μηχανισμού «αντιγραφής & ενσωμάτωσης». Αποτελούν το ~8% του ανθρώπινου γενώματος και δύνανται να επηρεάσουν την χρωμοσωμική ακεραιότητα όπως και την γονιδιακή έκφραση μετά την ρετρομετάθεσή των. Το γλοίωμα είναι ο πιο κοινός και επιθετικός ανθρώπινος τύπος καρκίνου, αποτελώντας το 60% όλων των περιπτώσεων πρωτογενών εγκεφαλικών όγκων. Χαρακτηρίζεται από έντονη ετερογένεια σε πολλαπλά επίπεδα που αφορούν στον φαινότυπο, στην γενετική και επιγενετική τους κατάσταση, στο δυναμικό πολλαπλασιασμού, μεταναστευτικότητας, ογκογενετικότητας και ανθεκτικότητάς τους. Πολλές από αυτές τις ιδιότητες αποδίδονται στα καρκινικά βλαστικά κύτταρα (Cancer Stem Cells: CSCs) τα οποία μπορούν να προκύψουν είτε μέσω αυξημένου αριθμού συμμετρικών κυτταρικών διαιρέσεων των CSCs είτε μέσω επαναπρογραμματισμού των μη-CSCs σε CSCs.Σκοπό της διατριβής αυτής αποτέλεσε η διερεύνηση τριών κρίσιμων ερωτημάτων, εάν: 1. γεγονότα ρετρομετάθεσης λαμβάνουν χώρα σε ανθρώπινα γλοιωματικά κύτταρα, 2. οι επιπτώσεις της ρετρομετάθεσης συσχετίζονται με μείζονα κυτταρικά φαινόμενα που απαντώνται σε γλοιωματικούς όγκους όπως είναι η φαινοτυπική ετερογένεια, το δυναμικό πολλαπλασιασμού, η ογκογενετικότητα και μεταστατικότητα και 3. η ρετρομετάθεση υπόκειται σε επιγενετικούς ή άλλους μηχανισμούς ρύθμισης σε γλοιωματικά κύτταρα.Τα κυριότερα ευρήματα της παρούσας διδακτορική διατριβής είναι τα ακόλουθα: - H διαμόλυνση δύο ανθρώπινων γλοιωματικών κυτταρικών σειρών (U-373- και U-87 MG) με το μη-αυτόνομο, LTR-ρετροτρανσποζόνιο, HERV-K10 οδήγησε στην παραγωγή υψηλής συχνότητας ρετρομετάθεσης σε αμφότερες τις κυτταρικές σειρές.- H ρετρομετάθεση HERV-K10 σε προγονικά γλοιωματικά κύτταρα U-373 συσχετίζεται με φαινόμενα διαφοροποίησής των προς ώριμα αστροκύτταρα που στοιχειοθετήθηκε τόσο μορφολογικά, όσο και μοριακά, μέσω ταυτοποίησης του πρωτεϊνικού δείκτη αστροκυτταρικής διαφοροποίησης GFAP (Glial Fibrillary Acidic Protein).- Η ρετρομετάθεση HERV-K10 επήγαγε φαινόμενα κυτταρικού επανα-προγραμματισμού αστροκυττάρων U87 προς νευρικού τύπου κύτταρα τα οποία απώλεσαν πλήρως την έκφραση της GFAP και συγχρόνως εμφανίστηκαν ισχυρά θετικά στην έκφραση της βIII-τουμπουλίνης (δείκτης χαρακτηρισμού νευρικών κυττάρων).- Ο HERV-K10-επαγόμενος επαναπρογραμματισμός επετεύχθη έμμεσα, μέσω από-διαφοροποίησής προς ένα ενδιάμεσο προγονικό/βλαστικό στάδιο (ενεργοποίηση CSCs). Πιο ειδικά, αποδείχθηκε ότι τα θετικά ρετρομετάθεσης κύτταρα U87 ήταν ικανά να παράξουν νευρόσφαιρες, οι οποίες: α. εξέφραζαν πρωτεϊνικούς δείκτες βλαστικότητας (νεστίνη, Sox2, Oct-4 και CD133) και β. έφεραν τις ιδιότητες της αυτό-ανανέωσης και του πολλαπλού δυναμικού διαφοροποίησης (αστρογλοιακή και νευρωνική γενεαλογία). - Η επαγωγή των CSCs σε κύτταρα U-87, οδήγησε σε in vitro ενίσχυση του ογκογενετικού και μεταστατικού τους δυναμικού, παρατηρήσεις που σημειώθηκαν και σε κύτταρα U373-θετικά ρετρομετάθεσης.- Επιπρόσθετη απόδειξη της συντελούμενης κυτταρικής διαφοροποίησης αποτέλεσε η σημαντική μείωση του ρυθμού πολλαπλασιασμού των θετικών ρετρομετάθεσης κυττάρων, που συνοδεύτηκε από εκτεταμένες διαταραχές του κυταρικού τους κύκλου.- Η επαγωγή της ρετρομετάθεσης HERV-K10 ως αποτέλεσμα της επίδρασης γλοιωματικών κυττάρων με τον παράγοντα κυτταρικής διαφοροποίησης, ρετινοϊκό οξύ, συνεπικουρεί στην εμπλοκή του φαινομένου της ρετρομετάθεσης στην διαφοροποίηση των γλοιωματικών κυττάρων.- Σε επίπεδο ρύθμισης της ρετρομετάθεσης HERV-K10 σε ανθρώπινα κύτταρα γλοιώματος, αποδείξαμε ότι: η επαγωγή της ρετρομετάθεσης από βαλπροϊκό οξύ και 5΄-αζακυτιδίνη συσχετίζονται με φαινόμενα κυτταρικής διαφοροποίησης και αναστολής του κυτταρικού κύκλου. - Η αναστολή της επαγόμενης από βαλπροϊκό οξύ ρετρομετάθεσης με την χρήση αναστολέων των ενδογενών αντίστροφων μεταγραφασών (Efavirenz), αποτρέπει την συντελούμενη από το βαλπροϊκό οξύ κυτταρική διαφοροποίηση. Η παρούσα μελέτη φιλοδοξεί να παρουσιάσει μία νέα πτυχή/διάσταση γύρω από τα κυριότερα κυτταρικά φαινόμενα που χαρακτηρίζουν/απαντώνται στα ανθρώπινα γλοιώματα, τοποθετώντας την ρετρομετάθεση στο επίκεντρο του γενικότερου μηχανισμού ρύθμισής των.

Triple-negative breast cancer (TNBC) is a highly aggressive type of cancer with limited therapeutic options. However, this type of cancer cells has shown overexpression of folate receptors, which bind with folic acid (FA) with high affinity. This feature can be used for targeting of nanocarriers, such as liposomes. In order to further examine the potential of increased efficacy by targeting the folate receptor, we prepared folate conjugated liposomes (DSPC/Chol/PEG/DSPE-PEG-FA) and loaded them with doxorubicin (DOX), an anticancer drug. For this, we first synthesized and verified the conjugate between FA and PEG-lipid (FA-PEG-lipid). After that, liposomes were prepared with thin film hydration method followed by probe sonication. Three different types of targeted liposomes having different concentrations of FA-PEG-lipid in their membranes (0.1 mol%, 0.5 mol%, and 1 mol%) were evaluated for their cytotoxicity (DOX-loaded liposomes) on MDA-MB-231 (epithelial human breast cancer cells), 4T1 (murine mammary carcinoma cells), and MCF7 (Human breast cancer cells); the two first are TNBC cells and overexpress FA receptor, the third does not. Cytotoxicity results proved that increasing amounts of FA on the surface of liposomes results in enhanced antitumor activity of liposomal-DOX in the case of the cancer cells, which overexpress the FA receptor.

BACKGROUND: Polymorphisms of the vitamin D receptor (VDR) gene have been linked to both multiple sclerosis (MS) and osteoporosis. We examined the frequency of the Taq-I and Bsm-I polymorphisms of the vitamin D receptor (VDR) gene in 69 patients with MS and 81 age and sex-matched healthy individuals. Genotyping of Taq-I (rs731236) and Bsm-I (rs1544410) was performed using TaqMan SNP Genotyping Assay. All patients and controls had determination of body mass index (BMI), bone mineral density (BMD) and smoking history. RESULTS: The mean age of patients was 39 +/- 10.5 years compared to 38.7 +/- 10.7 years of the controls (p = 0.86), the BMI was 24.8 +/- 4.2 kg/m2 compared to 25.7 +/- 4.8 kg/m2 of the controls (p = 0.23), the BMD in the lumbar spine 0.981 +/- 0.15 compared to 1.025 +/- 013 of the controls (p = 0.06) and the total hip BMD was 0.875 +/- 0.14 compared to 0.969 +/- 0.12 of the controls (p < 0.001). There were no differences of the Taq-I (TT, CT, CC) and Bsm-I genotypes (GG, GA, AA) and allelic frequencies between MS and control individuals. Multivariate analysis also failed to show any association of the Taq-I and Bsm-I polymorphisms and MS or sex, BMI, BMD and smoking history. CONCLUSIONS: This study suggests that the Taq-I and Bsm-I polymorphisms of the VDR gene are not associated with MS risk, BMI or BMD in the Greek population studied. J Negat Results Biomed
