Summary: Alpha-synuclein (α-Syn) is an important molecule in the pathogenesis of Parkinson’s and Alzheimer’s disease-related dementias such as Lewy body dementia, forming multiple pathological species. In vitro disease models, including human neurons and α-Syn-transfected cells, are instrumental to understand synucleinopathies or test new therapies. Here, we provide a detailed protocol to generate human neurons derived from induced pluripotent stem cells (iPSCs), and HEK cells, with α-Syn mutations. Abstract Brain–computer interfaces (BCIs) that reconstruct and synthesize speech using brain activity recorded with intracranial electrodes may pave the way toward novel communication interfaces for people who have lost their ability to speak, or who are at high risk of losing this ability, due to neurological disorders. Here, we report online synthesis of intelligible words using a chronically implanted brain-computer interface (BCI) in a man with impaired articulation due to ALS, participating in a clinical trial (ClinicalTrials.gov, NCT03567213) exploring different strategies for BCI communication. The 3-stage approach reported here relies on recurrent neural networks to identify, decode and synthesize speech from electrocorticographic (ECoG) signals acquired across motor, premotor and somatosensory cortices. We demonstrate a reliable BCI that synthesizes commands freely chosen and spoken by the participant from a vocabulary of 6 keywords previously used for decoding commands to control a communication board. Evaluation of the intelligibility of the synthesized speech indicates that 80% of the words can be correctly recognized by human listeners. Our results show that a speech-impaired individual with ALS can use a chronically implanted BCI to reliably produce synthesized words while preserving the participant’s voice profile, and provide further evidence for the stability of ECoG for speech-based BCIs.

Waveguide quantum electrodynamics represents a powerful platform to generate entanglement and tailor photonic states. We consider a pair of identical qubits coupled to a parity invariant waveguide in the microwave domain. By working in the one- and two-excitation sectors, we provide a unified view of decay processes and show the common origin of directional single-photon emission and two-photon directional bunching. Unveiling the quantum trajectories, we demonstrate that both phenomena are rooted in the selective coupling of orthogonal Bell states of the qubits with photons propagating in opposite directions. We comment on how to use this mechanism to implement optimized post-selection of Bell states, heralded by the detection of a photon on one side of the system.

Abstract Gene delivery to, and expression in, the mouse brain is important for understanding gene functions in brain development and disease, or testing gene therapies. Here, we describe an approach to express a transgene in the mouse brain in a cell-type-specific manner. We use stereotaxic injection of a transgene-expressing adeno-associated virus into the mouse brain via the intracerebroventricular route. We demonstrate stable and sustained expression of the transgene in neurons of adult mouse brain, using a reporter gene driven by a neuron-specific promoter. This approach represents a rapid, simple, and cost-effective method for global gene expression in the mouse brain, in a cell-type-specific manner, without major surgical interventions. The described method represents a helpful resource for genetically engineering mice to express a therapeutic gene, for gene therapy studies, or to deliver genetic material for genome editing and developing knockout animal models.

We examine the physical limitations and performance of spatially coherent imaging systems under defocused conditions using a modified two-point resolution criterion. By comparing coherent and incoherent illumination regimes, in defocused conditions, we highlight the peculiar advantages of coherent systems in resolving closely spaced features while maintaining a high depth of field. The analysis identifies two primary contributions to image degradation in coherent defocused systems: image broadening and interference-related contrast reduction. Through a specifically designed analytical framework, this study confirms that resolution degradation in coherent imaging is characterized by a square root dependence on defocusing distance, irrespective of the numerical aperture. This behavior starkly contrasts with incoherent imaging, where the resolution deteriorates linearly with defocusing and exhibits a strong dependence on the system’s numerical aperture. Furthermore, we provide explicit analytical expressions for contrast loss and minimum resolvable object size under defocused coherent conditions, offering deeper insights into their relation with system parameters. These results corroborate the superior performance of coherent imaging in resolving fine details in defocused environments, challenging conventional optical paradigms. Our findings not only extend the theoretical understanding of coherent imaging but also establish practical guidelines to optimize optical systems for high-resolution applications.

Abstract Correlation Plenoptic Imaging (CPI) is a novel volumetric imaging technique that uses two sensors and the spatio-temporal correlations of light to detect both the spatial distribution and the direction of light. This novel approach to plenoptic imaging enables refocusing and 3D imaging with significant enhancement of both resolution and depth of field. However, CPI is generally slower than conventional approaches due to the need to acquire sufficient statistics for measuring correlations with an acceptable signal-to-noise ratio (SNR). We address this issue by implementing a Deep Learning application to improve image quality with undersampled frame statistics. We employ a set of experimental images reconstructed by a standard CPI architecture, at three different sampling ratios, and use it to feed a CNN model pre-trained through the transfer learning paradigm U-Net architecture with VGG-19 net for the encoding part. We find that our model reaches a Structural Similarity (SSIM) index value close to 1 both for the test sample (SSIM = $$0.87 \pm 0.02$$ 0.87 ± 0.02 ) and in 5-fold cross validation (SSIM = $$0.92 \pm 0.07$$ 0.92 ± 0.07 ); the results are also shown to outperform classic denoising methods, in particular for images with lower SNR. The proposed work represents the first application of Artificial Intelligence in the field of CPI and demonstrates its high potential: speeding-up the acquisition by a factor 20 over the fastest CPI so far demonstrated, enabling recording potentially 200 volumetric images per second. The presented results open the way to scanning-free real-time volumetric imaging at video rate, which is expected to achieve a substantial influence in various applications scenarios, from monitoring neuronal activity to machine vision and security.

Foreign language acquisition within a highly interconnected context such as Europe is a complex challenge that can be influenced by many variables. This article presents TRIP a-Bike, an innovative didactic device designed to facilitate foreign language teaching for professionals and university students that combines artificial intelligence (AI) and gamification with an embodied based approach. TRIP a-Bike consists of an App for smartphones that integrates didactics with a video game, which in addition to offering the possibility of remote use, can be connected to a bicycle fixed to the ground and an LCD screen via a smartphone holder placed on the vehicle’s handlebar and a switch. Through physical interaction with the bicycle and the virtual environment, students can learn a new language in a dynamic and experiential way, while the AI generates customized content and activities based on their progress. The paper discusses the potential benefits of integrating AI and embodied theories and highlights how this system could represent an innovative and multidisciplinary educational research space. L’acquisizione di una lingua straniera all’interno di un contesto altamente interconnesso quale quello europeo, rappresenta una sfida complessa che può essere influenzata da tante variabili. Questo articolo presenta TRIP a-Bike, un dispositivo didattico innovativo pensato per facilitare l’insegnamento delle lingue straniere ai professionisti e agli studenti universitari che combina l’intelligenza artificiale (IA) e la gamification con un approccio embodied based. TRIP a-Bike consiste in un App per smartphone che integra la didattica ad un videogame, che oltre ad offrire la possibilità di utilizzo da remoto, può essere collegata a una bicicletta fissata al suolo e a uno schermo LCD mediante un supporto per smartphone posto sul manubrio del veicolo ed uno switch. Attraverso l’interazione fisica con la bicicletta e l’ambiente virtuale, gli studenti possono apprendere una nuova lingua in modo dinamico ed esperienziale, mentre l’IA genera contenuti e attività personalizzate in base ai loro progressi. L’articolo discute i potenziali benefici dell’integrazione tra IA ed embodied theories e sottolinea come questo sistema possa rappresentare uno spazio di ricerca didattico innovativo e multidisciplinare.

Abstract The correlation properties of light provide an outstanding tool to overcome the limitations of traditional imaging techniques. A relevant case is represented by correlation plenoptic imaging (CPI), a quantum-inspired volumetric imaging protocol employing spatio-temporally correlated photons from either entangled or chaotic sources to address the main limitations of conventional light-field imaging, namely, the poor spatial resolution and the reduced change of perspective for 3D imaging. However, the application potential of high-resolution imaging modalities relying on photon correlations is limited, in practice, by the need to collect a large number of frames. This creates a gap, unacceptable for many relevant tasks, between the time performance of correlated-light imaging and that of traditional imaging methods. In this article, we address this issue by exploiting the photon number correlations intrinsic in chaotic light, combined with a cutting-edge ultrafast sensor made of a large array of single-photon avalanche diodes (SPADs). This combination of source and sensor is embedded within a novel single-lens CPI scheme enabling to acquire 10 volumetric images per second. Our results place correlated-photon imaging at a competitive edge and prove its potential in practical applications.

Correlation plenoptic imaging is a procedure to perform light-field imaging without spatial resolution loss, by measuring the second-order spatiotemporal correlations of light. We investigate the possibility of using correlation plenoptic imaging to mitigate the effect of a phase disturbance in the propagation from the object to the main lens. We assume that this detrimental effect, which can be due to a turbulent medium, is localized at a specific distance from the lens, and is slowly varying in time. The mitigation of turbulence effects has already fostered the development of both light-field imaging and correlation imaging procedures. Here, we aim to merge these aspects, proposing a correlation light-field imaging method to overcome the effects of slowly varying turbulence, without the loss of lateral resolution, typical of traditional plenoptic imaging devices.

Abstract Background Combined antiretroviral therapy (cART) dramatically improved survival in people living with HIV (PLWH) but is associated with weight gain (WG), raising concern for a possible obesity epidemic in PLWH. This scoping review aims to identify the gaps in the existing evidence on WG in PLWH and generate a future research agenda. Methods This review was conducted according to the methodology for scoping studies and reported according to the PRISMA Extension for Scoping Review checklist. Articles published in English in the last 10 years indexed in Pubmed, WHO Global Index Medicus, or Embase were searched using specific queries focused on WG in PLWH. Results Following the selection process, 175 included articles were reviewed to search for the available evidence on four specific topics: (I) definition of WG in PLWH, (II) pathogenesis of WG in PLWH, (III) impact of ART on WG, (IV) correlation of WG with clinical outcomes. A summary of the data enabled us to identify gaps and clearly define the following research agenda: (I) develop a data-driven definition of WG in PLWH and define noninvasive assessment methods for body weight and fat composition; (II) further investigate the interaction between HIV/cART and immunity, metabolism, and adipose tissue; (III) establish the specific role of individual drugs on WG; (IV) clarify the independent role of WG, cART, HIV, and metabolic factors on clinical events. Conclusions The proposed research agenda may help define future research and fill the knowledge gaps that have emerged from this review.

Correlation Plenoptic Imaging (CPI) is an innovative approach to plenoptic imaging that tackles the inherent trade-off between image resolution and depth of field. By exploiting the intensity correlations that characterize specific states of light, it extracts information of the captured light direction, enabling the reconstruction of images with increased depth of field while preserving resolution. We describe a novel reconstruction algorithm, relying on compressive sensing (CS) techniques based on the discrete cosine transform and on gradients, used in order to reconstruct CPI images with a reduced number of frames. We validate the algorithm using simulated data and demonstrate that CS-based reconstruction techniques can achieve high-quality images with smaller acquisition times, thus facilitating the practical application of CPI.

Abstract Brain‐computer interfaces (BCIs) can be used to control assistive devices by patients with neurological disorders like amyotrophic lateral sclerosis (ALS) that limit speech and movement. For assistive control, it is desirable for BCI systems to be accurate and reliable, preferably with minimal setup time. In this study, a participant with severe dysarthria due to ALS operates computer applications with six intuitive speech commands via a chronic electrocorticographic (ECoG) implant over the ventral sensorimotor cortex. Speech commands are accurately detected and decoded (median accuracy: 90.59%) throughout a 3‐month study period without model retraining or recalibration. Use of the BCI does not require exogenous timing cues, enabling the participant to issue self‐paced commands at will. These results demonstrate that a chronically implanted ECoG‐based speech BCI can reliably control assistive devices over long time periods with only initial model training and calibration, supporting the feasibility of unassisted home use.

Metal halide perovskites are a promising photovoltaic technology for energy harvesting due to their potential for low cost via high-speed manufacturing and their flexible light form factors offering high power per weight. This study presents an investigation of the energy harvesting performance of perovskite solar cells under monochromatic illumination via finite element simulations and experimental validation with high-efficiency double cation perovskite solar cells. Device performance across a broad range of illumination intensity is analyzed, providing insights into the mechanisms limiting energy harvesting in medium- and long-range wireless power transfer. The simulations also provide a guideline for compositional engineering of wide bandgap perovskites to improve the spectral match to efficient monochromatic sources. Based on these results, we show how perovskite solar cells can become a platform for efficient (>33%) medium-range wireless power transfer at the 5–50 m scale for power levels of 1 mW to 1 W.

Abstract Light-field microscopy represents a promising solution for microscopic volumetric imaging, thanks to its capability to encode information on multiple planes in a single acquisition. This is achieved through its peculiar simultaneous capture of information on light spatial distribution and propagation direction. However, state-of-the-art light-field microscopes suffer from a detrimental loss of spatial resolution compared to standard microscopes. In this article, we experimentally demonstrate the working principle of a new scheme, called Correlation Light-field Microscopy (CLM), where the correlation between two light beams is exploited to achieve volumetric imaging with a resolution that is only limited by diffraction. In CLM, a correlation image is obtained by measuring intensity correlations between a large number of pairs of ultra-short frames; each pair of frames is illuminated by the two correlated beams, and is exposed for a time comparable with the source coherence time. We experimentally show the capability of CLM to recover the information contained in out-of-focus planes within three-dimensional test targets and biomedical phantoms. In particular, we demonstrate the improvement of the depth of field enabled by CLM with respect to a conventional microscope characterized by the same resolution. Moreover, the multiple perspectives contained in a single correlation image enable reconstructing over 50 distinguishable transverse planes within a 1 mm3 sample.

Summary: Background: Current approaches to support patients living with post-COVID condition, also known as Long COVID, are highly disparate with limited success in managing or resolving a well-documented and long-standing symptom burden. With approximately 2.1 million people living with the condition in the UK alone and millions more worldwide, there is a desperate need to devise support strategies and interventions for patients. Methods: A three-round Delphi consensus methodology was distributed internationally using an online survey and was completed by healthcare professionals (including clinicians, physiotherapists, and general practitioners), people with long COVID, and long COVID academic researchers (round 1 n = 273, round 2 n = 186, round 3 n = 138). Across the three rounds, respondents were located predominantly in the United Kingdom (UK), with 17.3–15.2% (round 1, n = 47; round 2 n = 32, round 2 n = 21) of respondents located elsewhere (United States of America (USA), Austria, Malta, United Arab Emirates (UAE), Finland, Norway, Malta, Netherlands, Iceland, Canada, Tunisie, Brazil, Hungary, Greece, France, Austrailia, South Africa, Serbia, and India). Respondents were given ∼5 weeks to complete the survey following enrolment, with round one taking place from 02/15/2022 to 03/28/22, round two; 05/09/2022 to 06/26/2022, and round 3; 07/14/2022 to 08/09/2022. A 5-point Likert scale of agreement was used and the opportunity to include free text responses was provided in the first round. Findings: Fifty-five statements reached consensus (defined as >80% agree and strongly agree), across the domains of i) long COVID as a condition, ii) current support and care available for long COVID, iii) clinical assessments for long COVID, and iv) support mechanisms and rehabilitation interventions for long COVID, further sub-categorised by consideration, inclusion, and focus. Consensus reached proposes that long COVID requires specialised, comprehensive support mechanisms and that interventions should form a personalised care plan guided by the needs of the patients. Supportive approaches should focus on individual symptoms, including but not limited to fatigue, cognitive dysfunction, and dyspnoea, utilising pacing, fatigue management, and support returning to daily activities. The mental impact of living with long COVID, tolerance to physical activity, emotional distress and well-being, and research of pre-existing conditions with similar symptoms, such as myalgic encephalomyelitis, should also be considered when supporting people with long COVID. Interpretation: We provide an outline that achieved consensus with stakeholders that could be used to inform the design and implementation of bespoke long COVID support mechanisms. Funding: None.

Abstract Understanding the cortical representations of movements and their stability can shed light on improved brain-machine interface (BMI) approaches to decode these representations without frequent recalibration. Here, we characterize the spatial organization (somatotopy) and stability of the bilateral sensorimotor map of forearm muscles in an incomplete-high spinal-cord injury study participant implanted bilaterally in the primary motor and sensory cortices with Utah microelectrode arrays (MEAs). We built representation maps by recording bilateral multiunit activity (MUA) and surface electromyography (EMG) as the participant executed voluntary contractions of the extensor carpi radialis (ECR), and attempted motions in the flexor carpi radialis (FCR), which was paralytic. To assess stability, we repeatedly mapped and compared left- and right-wrist-extensor-related activity throughout several sessions, comparing somatotopy of active electrodes, as well as neural signals both at the within-electrode (multiunit) and cross-electrode (network) levels. Wrist motions showed significant activation in motor and sensory cortical electrodes. Within electrodes, firing strength stability diminished as the time increased between consecutive measurements (hours within a session, or days across sessions), with higher stability observed in sensory cortex than in motor, and in the contralateral hemisphere than in the ipsilateral. However, we observed no differences at network level, and no evidence of decoding instabilities for wrist EMG, either across timespans of hours or days, or across recording area. While map stability differs between brain area and hemisphere at multiunit/electrode level, these differences are nullified at ensemble level.

Background: Intracortical microstimulation (ICMS) of somatosensory cortex can partially restore the sense of touch. Though ICMS bypasses much of the neuraxis, prior studies have found that conscious detection of touch elicited by ICMS lags behind the detection of cutaneous vibration. These findings may have been influenced by mismatched stimulus intensities, which can impact temporal perception. Objective: Evaluate the relative latency at which intensity-matched vibration and ICMS are perceived by a human participant. Methods: One person implanted with microelectrode arrays in somatosensory cortex performed reaction time and temporal order judgment (TOJ) tasks. To measure reaction time, the participant reported when he perceived vibration or ICMS. In the TOJ task, vibration and ICMS were sequentially presented and the participant reported which stimulus occurred first. To verify that the participant could distinguish between stimuli, he also performed a modality discrimination task, in which he indicated if he felt vibration, ICMS, or both. Results: When vibration was matched in perceived intensity to high-amplitude ICMS, vibration was perceived, on average, 48 ms faster than ICMS. However, in the TOJ task, both sensations arose at comparable latencies, with points of subjective simultaneity not significantly different from zero. The participant could discriminate between tactile modalities above chance level but was more inclined to report feeling vibration than ICMS. Conclusions: The latencies of ICMS-evoked percepts are slower than their mechanical counterparts. However, differences in latencies are small, particularly when stimuli are matched for intensity, implying that ICMS-based somatosensory feedback is rapid enough to be effective in neuroprosthetic applications.

Correlation plenoptic imaging (CPI) is an optical imaging technique based on intensity correlation measurement, which enables detecting, within fundamental physical limits, both the spatial distribution and the direction of light in a scene. This provides the possibility to perform tasks such as three-dimensional reconstruction and refocusing of different planes. Compared with standard plenoptic imaging devices, based on direct intensity measurement, CPI overcomes the problem of the strong trade-off between spatial and directional resolution. Here, we study the resolution limit in a recent development of the technique, called correlation plenoptic imaging between arbitrary planes (CPI-AP). The analysis, based on Gaussian test objects, highlights the main properties of the technique, as compared with standard imaging, and provides an analytical guideline to identify the limits at which an object can be considered resolved.

The pandemic has produced profound changes in teaching activities, with consequences on the educational processes and social development of students. In this sense, the following contribution explores the potential of Advanced Functional Didactics (DAF), a didactic proposal that aims to deliver traditional didactic contents through motor activity. Carried out in a difficult social context, namely the IC Ristori of Naples, located in the Forcella district, the study involves a second class of primary school and a first class of lower secondary school. The experimental hypothesis of this research is that the introduction of the DAF can bring benefits to the physical, psychological, social and learning abilities of the learners. This pilot study was the first to carry it out, thus proposing to verify the impact of this methodology on students to collect useful responses to improve it. Lo sviluppo di nuove abilità ed esperienze: uno studio pilota con la Didattica Avanzata Funzionale. La pandemia ha prodotto profondi cambiamenti nelle attività didattiche, con conseguenze sui processi formativi e di sviluppo sociale degli studenti. In tal senso, il seguente contributo esplora le potenzialità della Didattica Avanzata Funzionale (DAF), una proposta didattica che mira ad erogare i contenuti didattici tradizionali attraverso l’attività motoria. Realizzato in un contesto sociale difficile, ossia l’IC Ristori di Napoli, sito nel quartiere di Forcella, lo studio coinvolge una classe seconda della scuola primaria e una classe prima della scuola secondaria di primo grado. L’ipotesi sperimentale di questa ricerca è che l’introduzione della DAF possa apportare benefici alle capacità fisiche, psicologiche, sociali e di apprendimento dei discenti. Questo studio pilota è stato il primo a realizzarla, proponendosi quindi di verificare l’impatto di questa metodologia sugli studenti allo scopo di raccogliere risposte utili a migliorare quest’ultima.

The following document reports the experiences and life skills developed under the current didactic situation in Sports and Exercise Science. In particular, the manuscript discusses the pedagogic theories that should be fostered to improve the return to campus. The real-life experiences are presented in light of the Social and Emotional Learning and the Learning by Doing theories. The following document aims to reflect on social constructivism and embodied theories to improve the new (post-covid) learning environment. The introduction focuses on the Quality Assurance Agency for Higher Education benchmark statement and progresses to the most recent Blended approaches. With reflections upon in-class assessments, spaces and pace. In conclusion, the cartesian dualism (body-mind) is discussed and ideas for future research in pedagogy are presented to improve teaching and learning experiences in the post-covid scenario. Il documento ha come scopo quello di riflettere sul costruttivismo sociale e sulle embodied theories per migliorare il nuovo ambiente di apprendimento (post-covid). Il seguente documento riporta le esperienze e le abilità sviluppate nell‘attuale situazione didattica, nello specifico riguardo le scienze motorie. In particolare, nel testo sono discusse le teorie pedagogiche che dovrebbero essere incoraggiate per migliorare il ritorno in aula. Le esperienze didattiche sono presentate alla luce delle teorie dell‘apprendimento sociale ed emotivo e del Learning by Doing. L‘introduzione si concentra sul Quality Assurance Agency for Higher Education benchmark e presenta i più recenti approcci didattici definiti Blended, con l’aggiunta di riflessioni su valutazioni, spazi e ritmo della classe. In conclusione, viene discusso il dualismo cartesiano (corpo-mente) e vengono presentate idee per future ricerche in pedagogia al fine di migliorare le esperienze di insegnamento e di apprendimento nello scenario post-covid.

Nanoslits have various applications, including localized surface plasmon resonance (LSPR)-based nanodevices, optical biosensors, superfocusing, high-efficiency refractive index sensors and chip-based protein detection. In this study, the effect of substrates on the optical properties of gold nanoslits placed in free space is discussed; for this purpose, glass BK7 and Al2O3 are used as substrates and the wavelength of incident light is supposed to be 650 nm. The optical properties, power flow and electric field enhancement for gold nanoslits are investigated by using the finite element method (FEM) in COMSOL Multiphysics software. The effect of polarization of an incident electromagnetic wave as it propagates from a gold nanoslit is also analyzed. As special case, the effect of glass and alumina substrate on magnetic field, power flow and electric field enhancement is discussed. The goal of this research is to investigate the phenomenon of power flow and electric field enhancement. The study of power flow in gold nanoslits provides valuable insights into the behavior of light at the nanoscale and offers opportunities for developing novel applications in the field of nanophotonics and plasmonics. The consequences of this study show the significance of gold nanoslits as optical nanosensors.

Advances in intelligent robotic systems and brain-machine interfaces (BMI) have helped restore functionality and independence to individuals living with sensorimotor deficits; however, tasks requiring bimanual coordination and fine manipulation continue to remain unsolved given the technical complexity of controlling multiple degrees of freedom (DOF) across multiple limbs in a coordinated way through a user input. To address this challenge, we implemented a collaborative shared control strategy to manipulate and coordinate two Modular Prosthetic Limbs (MPL) for performing a bimanual self-feeding task. A human participant with microelectrode arrays in sensorimotor brain regions provided commands to both MPLs to perform the self-feeding task, which included bimanual cutting. Motor commands were decoded from bilateral neural signals to control up to two DOFs on each MPL at a time. The shared control strategy enabled the participant to map his four-DOF control inputs, two per hand, to as many as 12 DOFs for specifying robot end effector position and orientation. Using neurally-driven shared control, the participant successfully and simultaneously controlled movements of both robotic limbs to cut and eat food in a complex bimanual self-feeding task. This demonstration of bimanual robotic system control via a BMI in collaboration with intelligent robot behavior has major implications for restoring complex movement behaviors for those living with sensorimotor deficits.

Simulating the real-time dynamics of gauge theories represents a paradigmatic use case to test the hardware capabilities of a quantum computer, since it can involve non-trivial input states’ preparation, discretized time evolution, long-distance entanglement, and measurement in a noisy environment. We implemented an algorithm to simulate the real-time dynamics of a few-qubit system that approximates the Schwinger model in the framework of lattice gauge theories, with specific attention to the occurrence of a dynamical quantum phase transition. Limitations in the simulation capabilities on IBM Quantum were imposed by noise affecting the application of single-qubit and two-qubit gates, which combine in the decomposition of Trotter evolution. The experimental results collected in quantum algorithm runs on IBM Quantum were compared with noise models to characterize the performance in the absence of error mitigation.

Abstract Objectives Glyphosate (N-phosphonomethyl glycine) and its commercial herbicide formulations have been shown to exert toxicity via various mechanisms. It has been asserted that glyphosate substitutes for glycine in polypeptide chains leading to protein misfolding and toxicity. However, as no direct evidence exists for glycine to glyphosate substitution in proteins, including in mammalian organisms, we tested this claim by conducting a proteomics analysis of MDA-MB-231 human breast cancer cells grown in the presence of 100 mg/L glyphosate for 6 days. Protein extracts from three treated and three untreated cell cultures were analysed as one TMT-6plex labelled sample, to highlight a specific pattern (+/+/+/−/−/−) of reporter intensities for peptides bearing true glyphosate treatment induced-post translational modifications as well as allowing an investigation of the total proteome. Results Comparative statistical analysis of global proteome changes between glyphosate treated and non-treated samples did not show significant differences. Crucially, filtering of data to focus analysis on peptides potentially bearing glycine for glyphosate replacement revealed that the TMT reporter intensity pattern of all candidates showed conclusively that they are all false discoveries, with none displaying the expected TMT pattern for such a substitution. Thus, the assertion that glyphosate substitutes for glycine in protein polypeptide chains is incorrect.

Introduction. In March 2020, the Italian Ministry of Health prohibited individuals’ movement in the whole national territory. Schools closed, and teacher, educators, and students had to adapt to distanced learning ever since. However, the Italian National Institute of Statistic reported that only one student out of four could follow distanced learning, especially in the country’s poorest regions. Aim. To explore the evolution of distanced learning and produce evidence upon students’ perception of the past year’s educational experiences. Methods. The research team analysed two questionnaires conducted at a different time during the lockdown, in April and December 2020. The analysis was conducted using a quantitative approach and Cohen’s h effect size. Results. Data showed no improvements to the learning experience, leaving students with a poor, disadvantaged didactic experience. However, the data also showed an increment in students’ relationship abilities. Conclusion. It is then possible to conceive new intervention paradigms to foster the educational experience and prevent further students’ dropouts. Considering the embodied theories that have been neglected during the lockdown, the upcoming years of education are a call for responsibility to all adults (not only teachers and educators) since the health and education of the children, nationally and internationally, is at stake.

Correlation plenoptic imaging (CPI) is a technique capable of acquiring the light field emerging from a scene of interest, namely, the combined information of intensity and propagation direction of light. This is achieved by evaluating correlations between the photon numbers measured by two high-resolution detectors. Volumetric information about the object of interest is decoded, through data analysis, from the measured four-dimensional correlation function. In this paper, we investigate the relevant aspects of the refocusing algorithm, a post-processing method that isolates the image of a selected transverse plane within the 3D scene, once applied to the correlation function. In particular, we aim at bridging the gap between existing literature, which only deals with refocusing algorithms in case of continuous coordinates, and the experimental reality, in which the correlation function is available as a discrete quantity defined on the sensors pixels.

Diffraction-limited light-field imaging has been recently achieved by exploiting light spatial correlations measured on two high-resolution detectors. As in conventional light-field imaging, the typical operations of refocusing and 3D reconstruction are based on ray tracing in a geometrical optics context, and are thus well defined in the ideal case, both conceptually and theoretically. However, some properties of the measured correlation function are influenced by experimental features such as the finite size of apertures, detectors, and pixels. In this work, we take into account realistic experimental conditions and analyze the resulting correlation function through theory and simulation. We also provide an expression to evaluate the pixel-limited resolution of the refocused images, as well as a strategy for eliminating artifacts introduced by the finite size of the optical elements.

Abstract We present the experimental characterization of two distant double-slit masks illuminated by chaotic light, in the absence of first-order imaging and interference. The scheme exploits second-order interference of light propagating through two indistinguishable pairs of disjoint optical paths passing through the masks of interest. The proposed technique leads to a deeper understanding of biphoton interference and coherence, and opens the way to the development of novel schemes for retrieving information on the relative position and the spatial structure of distant objects, which is of interest in remote sensing, biomedical imaging, as well as monitoring of laser ablation, when first-order imaging and interference are not feasible.

COVID-19 has shaped and changed our normalities, and, with the discovery of new variants, the long-COVID syndrome, and stress disorders, the end of the pandemic seems distant. The current scenario is impacting all aspects of our lives. In particular, many studies reported that the pandemic resulted in increased psychiatric disorders and grief-related symptoms in adolescents. The project developed between Italy and Slovenia investigated students’ experiences during the lockdown and, in particular, reported the perception of distanced learning, producing a transparent qualitative analysis that can inform future research and open to discussions on learning strategies. A survey was conducted with secondary school students in Southern Italy between 1 April and 31 May 2020 and in Central Slovenia between 16 March and 28 February 2021. Qualitative and quantitative data were collected based on students’ perception of distanced learning, via an online survey platform. The results focused on three main aspects: learning experience, relationships with peers and teachers, and anxiety levels. The data showed similarities and differences in the two cohorts and suggested strategies to improve education (e.g., with blended approaches) to prepare students, teachers, and tutors for the challenges of returning to classes. In particular, data showed that it is recommended to foster collaboration between EU countries and to work to prevent students’ social isolation.

We review the advancement of the research toward the design and implementation of quantum plenoptic cameras, radically novel 3D imaging devices that exploit both momentum–position entanglement and photon–number correlations to provide the typical refocusing and ultra-fast, scanning-free, 3D imaging capability of plenoptic devices, along with dramatically enhanced performances, unattainable in standard plenoptic cameras: diffraction-limited resolution, large depth of focus, and ultra-low noise. To further increase the volumetric resolution beyond the Rayleigh diffraction limit, and achieve the quantum limit, we are also developing dedicated protocols based on quantum Fisher information. However, for the quantum advantages of the proposed devices to be effective and appealing to end-users, two main challenges need to be tackled. First, due to the large number of frames required for correlation measurements to provide an acceptable signal-to-noise ratio, quantum plenoptic imaging (QPI) would require, if implemented with commercially available high-resolution cameras, acquisition times ranging from tens of seconds to a few minutes. Second, the elaboration of this large amount of data, in order to retrieve 3D images or refocusing 2D images, requires high-performance and time-consuming computation. To address these challenges, we are developing high-resolution single-photon avalanche photodiode (SPAD) arrays and high-performance low-level programming of ultra-fast electronics, combined with compressive sensing and quantum tomography algorithms, with the aim to reduce both the acquisition and the elaboration time by two orders of magnitude. Routes toward exploitation of the QPI devices will also be discussed.

We explore the features of an equally-spaced array of two-level quantum emitters, that can be either natural atoms (or molecules) or artificial atoms, coupled to a field with a single continuous degree of freedom (such as an electromagnetic mode propagating in a waveguide). We investigate the existence and characteristics of bound states, in which a single excitation is shared among the emitters and the field. We focus on bound states in the continuum, occurring in correspondence of excitation energies in which a single excited emitter would decay. We characterize such bound states for an arbitrary number of emitters, and obtain two main results, both ascribable to the presence of evanescent fields. First, the excitation profile of the emitter states is a sinusoidal wave. Second, we discuss the emergence of multimers, consisting in subsets of emitters separated by two lattice spacings in which the electromagnetic field is approximately vanishing.

We study the out-of-equilibrium properties of $1+1$ dimensional quantum electrodynamics (QED), discretized via the staggered-fermion Schwinger model with an Abelian $\mathbb{Z}_{n}$ gauge group. We look at two relevant phenomena: first, we analyze the stability of the Dirac vacuum with respect to particle/antiparticle pair production, both spontaneous and induced by an external electric field; then, we examine the string breaking mechanism. We observe a strong effect of confinement, which acts by suppressing both spontaneous pair production and string breaking into quark/antiquark pairs, indicating that the system dynamics displays a number of out-of-equilibrium features.

Bacterial surface polysaccharides play significant roles in fitness and virulence. In Gram-negative bacteria such as Escherichia coli, major surface polysaccharides are lipopolysaccharide (LPS) and capsule, representing O- and K-antigens, respectively. There are multiple combinations of O:K types, many of which are well-characterized and can be related to ecotype or pathotype. In this investigation, we have identified a novel O:K permutation resulting through a process of major genome reorganization in a clade of E. coli. A multidrug-resistant, extended-spectrum β-lactamase (ESBL)-producing strain – E. coli 26561 – represented a prototype of strains combining a locus variant of O89 and group 1 capsular polysaccharide. Specifically, the variant O89 locus in this strain was truncated at gnd, flanked by insertion sequences and located between nfsB and ybdK and we apply the term O89m for this variant. The prototype lacked colanic acid and O-antigen loci between yegH and hisI with this tandem polysaccharide locus being replaced with a group 1 capsule (G1C) which, rather than being a recognized E. coli capsule type, this locus matched to Klebsiella K10 capsule type. A genomic survey identified more than 200 E. coli strains which possessed the O89m locus variant with one of a variety of G1C types. Isolates from our collection with the combination of O89m and G1C all displayed a mucoid phenotype and E. coli 26561 was unusual in exhibiting a mucoviscous phenotype more recognized as a characteristic among Klebsiella strains. Despite the locus truncation and novel location, all O89m:G1C strains examined showed a ladder pattern typifying smooth LPS and also showed high molecular weight, alcian blue-staining polysaccharide in cellular and/or extra-cellular fractions. Expression of both O-antigen and capsule biosynthesis loci were confirmed in prototype strain 26561 through quantitative proteome analysis. Further in silico exploration of more than 200 E. coli strains possessing the O89m:G1C combination identified a very high prevalence of multidrug resistance (MDR) – 85% possessed resistance to three or more antibiotic classes and a high proportion (58%) of these carried ESBL and/or carbapenemase. The increasing isolation of O89m:G1C isolates from extra-intestinal infection sites suggests that these represents an emergent clade of invasive, MDR E. coli.

We derive a light–matter interaction Hamiltonian to describe a quantum system embedded in a dispersive environment and coupled with the electromagnetic field. We include in this theory the spatial extension of the system, taken into account through its wavefunction. This enables us to overcome the divergence problem of the Green tensor propagator that arises from a point-like approximation of the quantum system. Thus the formalism can be applied to generalize the expressions for the spontaneous emission rate and the Lamb shift for a quantum system defined by a spatially extended dipole. In particular, these quantities can be modified by the asymmetry of the spatial structure of the atomic system as demonstrated in two test-bed examples.
