Abstract

Quorum sensing (QS) strongly regulates the biofilm formation mechanism in several microorganisms. The blockade of violacein production in Chromobacterium violaceum has been employed to indicate QS inhibition. Nevertheless, quantifying violacein is challenging due to the reliance on high-cost equipment and supplies. Consequently, this study compared the production and inhibition of violacein using a traditional spectrophotometric method and an alternative approach utilizing a Photometrix UVC Android application integrated with a USB endoscopic camera. The study standardized the growth of C. violaceum in several culture media and incubation times, then evaluated the pigment inhibition against salicylic acid. Results indicated that C. violaceum produced more violacein when cultivated in BHI broth for 24 h. Salicylic acid inhibited violacein production, even at sub-inhibitory concentrations, with a minimum inhibitory pigment concentration (125 µg/mL) and a minimum bactericidal concentration (2000 µg/mL). A comparison between application-based readings and the traditional method showed no significant differences, highlighting the potential of this technology as an essential tool in for QS inhibition and biofilm research.

Abstract

Abstract Background The production of monoclonal antibodies for immunoglobulin detection is not cost-effective, while polyclonal antibody production depends on laboratory animals, raising concerns on animal welfare. The widespread use of immunoglobulins in the pharmaceutical industry and the increasing number and variety of new antibodies entering the market require new detection and purification strategies. The Tripartite motif-containing protein 21 is a soluble intracellular immunoglobulin G receptor that binds to the constant region of immunoglobulin G from various species with high affinity. We hypothesized that using this protein as an antibody-binding module to create immunoglobulin detection probes will improve the portfolio of antibody affinity ligands for diagnostic or therapeutic purposes. Results We created a chimeric protein containing a mutated form of the C-terminal domain of mouse Tripartite motif-containing protein 21 linked to streptavidin to detect immunoglobulin G from various species of mammals. The protein is produced by heterologous expression and consists of an improved molecular tool, expanding the portfolio of antibody-affinity ligands for immunoassays. We also demonstrate that this affinity ligand may be used for purification purposes since imidazole elution of antibodies can be achieved instead of acidic elution conditions of current antibody purification methods. Conclusion Data reported here provides an additional and superior alternative to the use of secondary antibodies, expanding the portfolio of antibodies affinity ligands for detection and purification purposes.

Abstract

Silica nanoparticles have been widely explored in biomedical applications, mainly related to drug delivery and cancer treatment. These nanoparticles have excellent properties, high biocompatibility, chemical and thermal stability, and ease of functionalization. Moreover, silica is used to coat magnetic nanoparticles protecting against acid leaching and aggregation as well as increasing cytocompatibility. This review reports the recent advances of silica-based magnetic nanoparticles focusing on drug delivery, drug target systems, and their use in magnetohyperthermia and magnetic resonance imaging. Notwithstanding, the application in other biomedical fields is also reported and discussed. Finally, this work provides an overview of the challenges and perspectives related to the use of silica-based magnetic nanoparticles in the biomedical field.

Surra is an infectious disease caused by Trypanosoma evansi, which affects a large number of domestic and wild animal species. Infection control is based on rapid diagnosis followed by treatment of sick animals. This study aimed to evaluate a buffered T. evansi antigen and rapid serum agglutination test (BA/Te) for the detection of anti-T. evansi antibodies in serum samples of horses. For this purpose, 445 serum samples from horses were evaluated and the results compared with the diagnosis by CATT/T. evansi. Abstract

The mevalonate pathway is an essential part of isoprenoid biosynthesis leading to production of a diverse class of >30,000 biomolecules including cholesterol, heme, and all steroid hormones. In trypanosomatids, the mevalonate pathway also generates dolichols, which play an essential role in construction of glycosylphosphatidylinositol (GPI) molecules that anchor variable surface proteins (VSGs) to the plasma membrane. Isoprenoid biosynthesis involves one of the most highly regulated enzymes in nature, 3-hydroxy-3-methylglutaryl-CoA reductase (HMGCR), which catalyzes the conversion of HMG-CoA to mevalonic acid. The enzyme mevalonate kinase (MVK) subsequently converts mevalonic acid to 5-phosphomevalonic acid. Trypanosoma evansi is a flagellate protozoan parasite that causes the disease “Surra” in domesticated large mammals, with great economic impact. T. evansi has only a trypomastigote bloodstream form and requires constant modification of the variant surface glycoprotein (VSG) coat for protection against the host immune system. We identified MVK of T. evansi (termed TeMVK) and performed a preliminary characterization at molecular, biochemical, and cellular levels. TeMVK from parasite extract displayed molecular weight ~36 kDa, colocalized with aldolase (a glycosomal marker enzyme) in glycosomes, and is structurally similar to Leishmania major MVK. Interestingly, the active form of TeMVK is the tetrameric oligomer form, in contrast to other MVKs in which the dimeric form is active. Despite lacking organized mitochondria, T. evansi synthesizes both HMGCR transcripts and protein. Both MVK and HMGCR are expressed in T. evansi during the course of infection in animals, and therefore are potential targets for therapeutic drug design.

Abstract

Herein, tetracycline adsorption employing magnetic chitosan (CS·Fe3O4) as the adsorbent is reported. The magnetic adsorbent was synthesized by the co-precipitation method and characterized through FTIR, XRD, SEM, and VSM analyses. The experimental data showed that the highest maximum adsorption capacity was reached at pH 7.0 (211.21 mg g−1). The efficiency of the magnetic adsorbent in tetracycline removal was dependent on the pH, initial concentration of adsorbate, and the adsorbent dosage. Additionally, the ionic strength showed a significant effect on the process. The equilibrium and kinetics studies demonstrate that Sips and Elovich models showed the best adjustment for experimental data, suggesting that the adsorption occurs in a heterogeneous surface and predominantly by chemical mechanisms. The experimental results suggest that tetracycline adsorption is mainly governed by the hydrogen bonds and cation–π interactions due to its pH dependence as well as the enhancement in the removal efficiency with the magnetite incorporation on the chitosan surface, respectively. Thermodynamic parameters indicate a spontaneous and exothermic process. Finally, magnetic chitosan proves to be efficient in TC removal even after several adsorption/desorption cycles.

Abstract

Introduction: adhesive capsulitis (frozen shoulder) is a common disease that causes significant morbidity. Chiropractic care is based on technical adjustments that restore normal micro-motion to the spine, reducing neural compression cause of painful symptoms that particular dermatome. The aim of this paper is to review the scientific literature the effects of chiropractic in patients with frozen shoulder. Method: This is an integrative review article, made from primary sources. As criteria for inclusion of articles, we selected only those whose methodology was Clinical Trial Controlled Randomized, published in the period 2010 to 2016, which made reference to features and methods of the relevant chiropractic treatment of frozen shoulder, which compared a group chiropractic and another that applied conventional physiotherapy or other similar technique. Results: The search in databases totaled 18 randomized controlled trials. According to the inclusion criteria, only three articles were selected. The three studies have shown benefits of chiropractic for improvements in mobility, pain and function. Conclusion: Each article has shown benefits with the application of chiropractic techniques frozen shoulder.

Abstract

Background: The term temporomandibular dysfunction (TMD) applies to functional changes related to temporomandibular joint (TMJ) and it is associated to masticatory structures. The main symptoms are pain in the TMJ region and / or the masticatory musculature. Objective: to perform a systematic review on the effects of manual therapy in the treatment of TMD. Methods: We analyzed 231 articles from the databases of Bireme, BVS, Lilacs, MedLine, PEDro, Pubmed and, Scielo. Of these, only 30 were selected for the elaboration of the article and 6 were used for the qualitative synthesis. Studies of controlled clinical trials of high methodological quality and high clinical relevance on the PEDro scale were included, which used the same evaluation measures, VAS (visual analogic scale), PPT (pressure pain threshold) and maximum mouth opening (MMO), and only one treatment technique comparing the control group with placebo. Result: The studies presented positive results for pain in VAS, PPT and MMO; however, there are significant differences depending on the technique. Myofascial release has a strong efficacy in reducing the symptoms of acute pain in the long term, but does not show clinically significant changes in mouth opening amplitude and MMO. Mobilization / manipulation presents significant changes in MMO, significantly improving the efficacy of mouth range of motion in the treatment of the short-term pain symptom, however does not sustain in the long term. Conclusion: The tool for the individual treatment of patients is based on the symptoms addressed, significantly improving the effectiveness of the range of motion of the mouth in the treatment of joint mobilization / manipulation, and treatment of pain by myofascial release.

G-type immunoglobulins (IgGs) are extensively used in the pharmaceutical industry against various diseases, being also crucial in multiple immunoassays. The production of secondary monoclonal antibodies (Abs) for IgG detection is not cost-effective, while polyclonal antibody production still depends on laboratory animals, which raises concerns regarding animal welfare. As alternatives, bacterial proteins (A and G) have been widely exploited; however, several difficulties are encountered regarding their use for IgG detection and purification. The widespread use of IgGs in the pharmaceutical industry and the increasing number and variety of new Abs entering the market impose the need to develop new detection and purification strategies. The TRIM21 protein is a soluble intracellular IgG receptor that binds to the Fc region of many species with high affinity. We created a chimeric protein containing a mutated form of the C-terminal domain of mouse TRIM21 linked to a streptavidin moiety to detect IgGs from a wide range of species. The protein is promptly produced by heterologous expression and consists of an improved molecular tool, expanding the portfolio of Ab-affinity ligands for immunoassays.

Abstract

Trypanosoma evansi is the agent of "surra," a trypanosomosis endemic in many areas worldwide. Trypanosoma proteins released/secreted during infection are attractive biomarkers for disease detection and monitoring. Using liquid chromatography-tandem mass spectrometry (LC-MS/MS), we performed a comprehensive analysis of the serum proteome of mice infected with T.evansi and detected changes in the abundance of parasite and host serum proteins during infection. Following bioinformatics analysis, 30 T. evansi proteins were identified in the mice serum including known targets such as pyruvate kinase 1, β-tubulin, actin A, heat shock protein 70, and cyclophilin A. We also identified two exclusive VSG epitopes which are novel putative biomarker targets. In addition, upregulation of 31 mouse proteins, including chitinase-like protein 3 and monocyte differentiation antigen CD14, were observed. Identification of parasite-specific biomarkers in the host serum is critical for the development of reliable serological/ assays for differential diagnosis.
