Refugee Tales IV, edited by Anna Pincus and David Herd and published in 2021, is the fourth volume to date of the Refugee Tales short-story collections. While these stories are told by individuals who have been in detention centers, it is renowned writers such as Dina Nayeri and Robert Macfarlane who generally help these refugees and asylum seekers capture their experiences on paper. The project was set up and is organized by the Gatwick Detainees Welfare Group “as a response to the silence that surrounded indefinite immigration detention in the U.K. [and other countries around the world]” (Herd 2017, 113), which criminalizes the asylum process, “mak[ing] the commitment of a crime (for instance, trying to leave the country under false papers) so much more likely” (117). In this article, I will explore how the short stories in Refugee Tales IV contribute to helping refugees and asylum seekers achieve a space of self-determination where their stories, which are often disesteemed or ignored, can be heard. By emulating some of the main themes and features of The Canterbury Tales, these stories shed light on the traumatic experiences many refugees face in the U.K. and other countries with regard to their indefinite detention and a legal system that marginalizes them. I will pay particular attention to the various forms in which the individuals express the impotence of not being able to decide things for themselves, as well as the ways in which the act of storytelling allows them to occupy a literary space from where to speak up and become active agents.

This study evaluates multisectoral energy planning to decarbonize Punta Arenas, Chile, transitioning from fossil fuels to renewable energies by 2050. Scenarios aligned with the Carbon Neutral 2050 (CN2050) plan, developed under the Long-Term Energy Planning (PELP) program by the Chilean Ministry of Energy, were assessed using EnergyPLAN. In 2019, Punta Arenas emitted 1.32 million tonnes of carbon dioxide equivalent (CO₂eq), projected to rise to 2.2 million tonnes by 2050 under a business-as-usual (BAU) scenario, with annual costs of 947 million euros. Implementing PELP CN2050 measures reduces emissions to 1.2 million tonnes and costs to 560 million euros, demonstrating that decarbonization can be achieved alongside economic savings. This validates the PELP CN2050 plan's effectiveness, highlighting that renewable energy integration supports sustainability and economic benefits. Reductions are achieved through energy efficiency, technological changes, and integrating renewable energies—particularly wind and solar thermal—in industrial, transport, and residential sectors. Electrification and green hydrogen for motive, thermal, and transport applications are crucial. Two cases were evaluated: importing green hydrogen or producing it locally via renewable-powered electrolysers. Sensitivity analyses increasing wind capacity from 13 MW to 310 MW showed that higher renewable integration reduces CO₂eq emissions and costs, indicating a negative abatement cost. Further decarbonization could be achieved by incorporating cogeneration, district heating, and synthetic fuels.

This paper presents a comprehensive techno-economical potential analysis for integrating solar heat for industrial processes (SHIP) in Chile. Utilizing TRNSYS software for simulation and Python for data analysis, the study evaluates the levelized cost of heat (LCoH) and solar fraction for various industrial sectors across different regions of the country. The findings indicate that regions such as Coquimbo, Antofagasta, and Metropolitan exhibit high solar fractions and competitive LCoH values, particularly in hot water production. The central zone, especially the Metropolitan region, demonstrates significant potential for steam generation, achieving an average solar fraction of 50 % and producing 295 GWh/year of solar thermal energy. Validation with real-world data demonstrated an average error margin of 21 %, underscoring the developed heat load calculation model's reliability. The study concluded that SHIP is a viable solution for reducing energy costs and greenhouse gas emissions in Chile. It has the potential to displace over 3,500 GWh/year of fossil fuel energy in the considered productive sectors. These findings highlight the strategic importance of supporting SHIP implementation through favorable policies and investments.

Patients present a wide range of clinical severities in response severe acute respiratory syndrome coronavirus 2 infection, but the underlying molecular and cellular reasons why clinical outcomes vary so greatly within the population remains unknown. Here, we report that negative clinical outcomes in severely ill patients were associated with divergent RNA transcriptome profiles in peripheral immune cells compared with mild cases during the first weeks after disease onset. Protein–protein interaction analysis indicated that early-responding cytotoxic natural killer cells were associated with an effective clearance of the virus and a less severe outcome. This innate immune response was associated with the activation of select cytokine–cytokine receptor pathways and robust Th1/Th2 cell differentiation profiles. In contrast, severely ill patients exhibited a dysregulation between innate and adaptive responses affiliated with divergent Th1/Th2 profiles and negative outcomes. This knowledge forms the basis of clinical triage that may be used to preemptively detect high-risk patients before life-threatening outcomes ensue.

Abstract Pitseed goosefoot (Chenopodium berlandieri) is a free-living North American member of an allotetraploid complex that includes the Andean pseudocereal quinoa (C. quinoa). Like quinoa, pitseed goosefoot was domesticated, possibly independently, in eastern North America (subsp. jonesianum) and Mesoamerica (subsp. nuttaliae). To test the utility of C. berlandieri as a resource for quinoa breeding, we produced the whole-genome DNA sequence of PI 433,231, a huauzontle from Puebla, México. The 1.295 Gb genome was assembled into 18 pseudomolecules and annotated using RNAseq data from multiple tissues. Alignment with the v.2.0 genome of Chilean-origin C. quinoa cv. ‘QQ74’ revealed several inversions and a 4A-6B reciprocal translocation. Despite these rearrangements, some quinoa x pitseed goosefoot crosses produce highly fertile hybrids with faithful recombination, as evidenced by a high-density SNP linkage map constructed from a Bolivian quinoa ‘Real-1’ × BYU 937 (Texas coastal pitseed goosefoot) F2 population. Recombination in that cross was comparable to a ‘Real-1’ × BYU 1101 (Argentine C. hircinum) F2 population. Furthermore, SNP-based phylogenetic and population structure analyses of 90 accessions supported the hypothesis of multiple independent domestications and descent from a common 4 × ancestor, with a likely North American Center of Origin.

IntroductionThe establishment of the rhizobium-legume nitrogen-fixing symbiosis relies on the interchange of molecular signals between the two symbionts. We have previously studied by RNA-seq the effect of the symbiotic regulators NodD1, SyrM, and TtsI on the expression of the symbiotic genes (the nod regulon) of Sinorhizobium fredii HH103 upon treatment with the isoflavone genistein. In this work we have further investigated this regulatory network by incorporating new RNA-seq data of HH103 mutants in two other regulatory genes, nodD2 and nolR. Both genes code for global regulators with a predominant repressor effect on the nod regulon, although NodD2 acts as an activator of a small number of HH103 symbiotic genes.MethodsBy combining RNA-seq data, qPCR experiments, and b-galactosidase assays of HH103 mutants harbouring a lacZ gene inserted into a regulatory gene, we have analysed the regulatory relations between the nodD1, nodD2, nolR, syrM, and ttsI genes, confirming previous data and discovering previously unknown relations.Results and discussionPreviously we showed that HH103 mutants in the nodD2, nolR, syrM, or ttsI genes gain effective nodulation with Lotus japonicus, a model legume, although with different symbiotic performances. Here we show that the combinations of mutations in these genes led, in most cases, to a decrease in symbiotic effectiveness, although all of them retained the ability to induce the formation of nitrogen-fixing nodules. In fact, the nodD2, nolR, and syrM single and double mutants share a set of Nod factors, either overproduced by them or not generated by the wild-type strain, that might be responsible for gaining effective nodulation with L. japonicus.

Green hydrogen has been considered as one of the energy carriers of the future, and Chile can become a production leader due to its great renewable energy potential. Cheap electricity is one of the key drivers for making green hydrogen a cost-effective energy carrier for many sectors. However, without energy storage, only a small operational electrolysis capacity can be achieved, and therefore, the share of the CAPEX in the levelized cost of hydrogen (LCOH2) increases [1]. This work set out to conduct a techno-economic analysis for the integration of large-scale green hydrogen production and a hybrid CSP+PV plant of 100 MWe in northern Chile, one of the world's solar hotspots. For a better understanding on the benefits of such integration, the performance of the hybrid solar plant was compared to the performance offered by each independent solar technology and with a grid-connection via a PPA mechanism. In addition, this study takes into account the costs of storage and transport to potential local and international consumers.

Introduction: Although it has been shown that the supportive periodontal therapy (SPT) is essential to maintain the results obtained by periodontal therapy, patient adherence to SPT is not usually satisfactory (4). The objective of this research is to explore the motivations in the adherence to periodontal maintenance of patients treated at Universidad Viña del Mar (UVM) during the year 2020. Material and Methods: A study based on the qualitative paradigm was carried out using a design based on discourse analysis through semi-structured interviews. A sample size of 10 participants was consolidated, by obtaining with them the saturation of data collected and of 4 main dimensions. Each interview was transcribed ad verbatim and later analyzed through the Atlas.ti 9 Software. Results: Four main categories or dimensions were created: Motivations of the patients to produce a change in their oral health; Information that the patients have regarding their oral health; Oral hygiene habits acquired after periodontal treatment; Situations that prevent adherence to periodontal treatment. Conclusion: The main motivations of patients to adhere to supportive periodontal therapy are related to the education and information they acquire in the first phase of periodontal treatment. Observing the changes during treatment, improving oral health, improving aesthetics and functionality, are other important motivations, as well as the patients’ fear of losing their teeth and the desire to educate their family.

To determine molecular epidemiology and clinical features of enterovirus D68 (EV-D68) infections, we reviewed EV-D68–associated respiratory cases at a hospital in Barcelona, Spain, during 2014–2021. Respiratory samples were collected from hospitalized patients or outpatients with symptoms of acute respiratory tract infection or suggestive of enterovirus infection. Enterovirus detection was performed by real-time multiplex reverse transcription PCR and characterization by phylogenetic analysis of the partial viral protein 1 coding region sequences. Shatila camp in Beirut was founded in 1949 and now houses up to 40,000 refugees. In 2017, the Peirene Press publisher Meike Ziervogel and London-based Syrian editor Suhir Hedal travelled to the camp to hold a three-day creative writing workshop in which nine Syrian and Palestinian refugees participated. The result is Shatila Stories (2018), a brilliant piece of collaborative fiction translated from Arabic to English by Naswa Gowanlock. It is a hybrid between a novel and a short story collection, in which refugee voices are given the chance to speak up, share their stories, and negotiate their identities. This article examines Shatila Stories (2018) as a book that highlights Shatila as a campscape (Diana Martín). These stories show that the camp, as Adam Ramadan argues, is not empty of law and political life, but rather it is a meaningful space produced by who and what is in it, and how they interrelate and interact. Shatila Stories is, indeed, an effective platform that allows readers to understand how refugees’ conflicts and thoughts are processed and the ways in which refugees in Shatila accept and embody the camp’s liminality and their border subject identity to gain agency and resist the restrained passivity to which they are often relegated. Ultimately, my analysis pays attention to how these stories encourage the renegotiation of the refugees’ selfhood and counter Agamben’s perception of refugees as “bare lives” by portraying them as autonomous, active and humanized individuals in the eyes of the international reader.

The short stories “En la espesura de la noche” (2010) and “Una camarera (expatriada a cualquier lugar del mundo)” (2011) depict the feeling of imprisonment that first-person narrators experience within the rigid borders of Equatorial Guinea and Cuba, respectively, and how they overcome this territorial oppression by loving a woman who dwells beyond the national borders they are unable to trespass. Bearing in mind that the love towards nation-states constitutes a way of perpetuating a nationalist ideology and the reinforcement of national borders over individual free agency (Morrison et al., 2021: 514), this article examines how, in both narratives, the exaltation of a transnational love over that of the nation challenges these nation-state’s “unique discourse,” based on the binary border thinking of inside/outside and us/them, in favor of individual agency and a more transnational way of understanding the world.

Polylactic acid is one of the most used biopolymers due to its overall properties and biodegradability. Nevertheless, polylactic acid has important drawbacks such as brittleness, low thermal stability, and higher cost than most commodity polymers. In order to overcome those disadvantages without compromising biodegradability, the addition of wood particles and thermal annealing on the crystallinity and impact strength of wood-polylactic acid biocomposites were studied. The samples were prepared by compression and rotational molding using two different wood particles: white ash and tzalam. The results showed that thermal annealing at 100 °C, 40 minutes, increased the crystallinity up to 60 % and also improved the thermal stability of polylactic acid and its biocomposites as determined via dynamic mechanical analysis. The specimens not exposed to thermal annealing exhibited important storage modulus loss above 60 °C, which mostly disappeared with the thermal treatment. Furthermore, the impact strength was substantially increased by the thermal treatment. Additionally, accelerated weathering tests showed that the thermally annealed samples had better dimensional stability growing their potential applications over a wider range of conditions.

Abstract Background Limited evidence exists on the role of glucose-lowering drugs in patients with COVID-19. Our main objective was to examine the association between in-hospital death and each routine at-home glucose-lowering drug both individually and in combination with metformin in patients with type 2 diabetes mellitus admitted for COVID-19. We also evaluated their association with the composite outcome of the need for ICU admission, invasive and non-invasive mechanical ventilation, or in-hospital death as well as on the development of in-hospital complications and a long-time hospital stay. Methods We selected all patients with type 2 diabetes mellitus in the Spanish Society of Internal Medicine’s registry of COVID-19 patients (SEMI-COVID-19 Registry). It is an ongoing, observational, multicenter, nationwide cohort of patients admitted for COVID-19 in Spain from March 1, 2020. Each glucose-lowering drug user was matched with a user of other glucose-lowering drugs in a 1:1 manner by propensity scores. In order to assess the adequacy of propensity score matching, we used the standardized mean difference found in patient characteristics after matching. There was considered to be a significant imbalance in the group if a standardized mean difference > 10% was found. To evaluate the association between treatment and study outcomes, both conditional logit and mixed effect logistic regressions were used when the sample size was ≥ 100. Results A total of 2666 patients were found in the SEMI-COVID-19 Registry, 1297 on glucose-lowering drugs in monotherapy and 465 in combination with metformin. After propensity matching, 249 patients on metformin, 105 on dipeptidyl peptidase-4 inhibitors, 129 on insulin, 127 on metformin/dipeptidyl peptidase-4 inhibitors, 34 on metformin/sodium-glucose cotransporter 2 inhibitor, and 67 on metformin/insulin were selected. No at-home glucose-lowering drugs showed a significant association with in-hospital death; the composite outcome of the need of intensive care unit admission, mechanical ventilation, or in-hospital death; in-hospital complications; or long-time hospital stays. Conclusions In patients with type 2 diabetes mellitus admitted for COVID-19, at-home glucose-lowering drugs showed no significant association with mortality and adverse outcomes. Given the close relationship between diabetes and COVID-19 and the limited evidence on the role of glucose-lowering drugs, prospective studies are needed.

Quinoa has been cultivated since centuries in the Andean region as a seed crop by indigenous communities. The crop has gained renewed interest because of its highly nutritious grain with high-quality protein rich in essential amino acids, and several bioactive compounds, along with its ability to grow under stress conditions. Despite the importance of the crop, limited research work on breeding aspects has been undertaken, leading to lack of information on the understanding of levels of variability of genotypes for different traits and their interactions. The aim of the present study was to assess and quantify the early response to mass selection in two quinoa landraces in highland conditions. Mass selection experiments were conducted during two successive crop seasons using eleven morphological traits. Correlation, genetic gain (gg) per selection cycle and principal component analysis was carried out. Only plant height (PH) and number of branches (NB) presented changes between selection cycles in both germplasm lines. Grain yield per plant (GYP) was positively correlated with inflorescence length (IL), stem diameter (SD) and plant weight (PW) for both quinoa lines. The results obtained would be useful to facilitate selection of the most relevant variables of quinoa considering its variation and interactions in the highland environment in Chile.

The coronavirus disease 2019 (COVID19) pandemic has left researchers scrambling to identify the humoral immune correlates of protection from COVID-19. To date, the antibody mediated correlates of virus neutralization have been extensively studied. However, the extent that non-neutralizing functions contribute to anti-viral responses are ill defined. In this study, we profiled the anti-spike antibody subtype/subclass responses, along with neutralization and antibody-dependent natural killer cell functions in 83 blood samples collected between 4 and 201 days post-symptoms onset from a cohort of COVID-19 outpatients. We observed heterogeneous humoral responses against the acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein. Overall, anti-spike profiles were characterized by a rapid rise of IgA and sustained IgG titers. In addition, strong antibody-mediated natural killer effector responses correlated with milder disease and being female. While higher neutralization profiles were observed in males along with increased severity. These results give an insight into the underlying function of antibodies beyond neutralization and suggest that antibody-mediated natural killer cell activity is a key function of the humoral response against the SARS-CoV-2 spike protein.

(1) Background: Some rhizobia, such as Rhizobium tropici CIAT 899, activate nodulation genes when grown under osmotic stress. This work aims to determine whether this phenomenon also takes place in Sinorhizobium fredii HH103. (2) Methods: HH103 was grown with and without 400 mM mannitol. β-galactosidase assays, nodulation factor extraction, purification and identification by mass spectrometry, transcriptomics by RNA sequencing, motility assays, analysis of acyl-homoserine lactones, and indole acetic acid quantification were performed. (3) Results: Non-ionic osmotic stress induced the production of nodulation factors. Forty-two different factors were detected, compared to 14 found in the absence of mannitol. Transcriptomics indicated that hundreds of genes were either activated or repressed upon non-ionic osmotic stress. The presence of 400 mM mannitol induced the production of indole acetic acid and acyl homoserine lactones, abolished swimming, and promoted surface motility. (4) Conclusions: In this work, we show that non-ionic stress in S. fredii HH103, caused by growth in the presence of 400 mM mannitol, provokes notable changes not only in gene expression but also in various bacterial traits, including the production of nodulation factors and other symbiotic signals.

Non-alcoholic fatty liver disease (NAFLD) is the primary cause of chronic liver disease. The range is extensive, including hepatocellular carcinoma, cirrhosis, fibrosis, fatty liver, and non-alcoholic steatohepatitis (NASH). NASH is a condition related to obesity, overweight, metabolic syndrome, diabetes, and dyslipidemia. It is a dynamic condition that can regress to isolated steatosis or progress to fibrosis and cirrhosis. Statins exert anti-inflammatory, proapoptotic, and antifibrotic effects. It has been proposed that these drugs could have a relevant role in NASH. In this review, we provide an overview of current evidence, from mechanisms of statins involved in the modulation of NASH to human trials about the use of statins to treat or attenuate NASH.

Abstract Background There is no randomized clinical trials with recurrence of atherosclerotic cardiovascular disease (ASCVD) as a major outcome with rosuvastatin. In order to analyze potential differences in the clinical response to atorvastatin and rosuvastatin in secondary ASCVD prevention, we have analyzed the clinical evolution of those subjects of the Dyslipemia Registry of the Spanish Society of Arteriosclerosis (SEA) who at the time of inclusion in the Registry had already suffered an ASCVD. Methods This observational, retrospective, multicenter, national study was designed to determine potential differences between the use of atorvastatin and rosuvastatin in the ASCVD recurrence. Three different follow-up start-times were performed: time of inclusion in the registry; time of first event if this occurred after 2005, and time of first event without date restriction. Results Baseline characteristics were similar between treatment groups. Among atorvastatin or rosuvastatin users, 89 recurrences of ASCVD were recorded (21.9%), of which 85.4% were coronary. At the inclusion of the subject in the registry, 345 participants had not suffered a recurrence yet. These 345 subjects accumulated 1050 person-years in a mean follow-up of 3 years. Event rates were 2.73 (95% CI: 1.63, 4.25) cases/100 person-years and 2.34 (95% CI: 1.17, 4.10) cases/100 person-years in the atorvastatin and rosuvastatin groups, respectively. There were no statistically significant differences between the two groups independently of the follow-up start-time. Conclusions This study does not find differences between high doses of rosuvastatin and atorvastatin in the recurrence of ASCVD, and supports their use as clinically equivalent in secondary prevention of ASCVD.

BackgroundNew World Hantaviruses (NWHs) are the etiological agent underlying hantavirus cardiopulmonary syndrome (HCPS), a severe respiratory disease with high mortality rates in humans. In Panama, infections with Choclo Orthohantavirus (CHOV) cause a much milder illness characterized by higher seroprevalence and lower mortality rates. To date, the cytokine profiles and antibody responses associated with this milder form of HCPS have not been defined. Therefore, in this study, we examined immune serological profiles associated with CHOV infections.MethodsFor this retrospective study, sera from fifteen individuals with acute CHOV-induced HCPS, were analyzed alongside sera from fifteen convalescent phase individuals and thirty-three asymptomatic, CHOV-seropositive individuals. Cytokine profiles were analyzed by multiplex immunoassay. Antibody subclasses, binding, and neutralization against CHOV-glycoprotein (CHOV-GP) were evaluated by ELISA, and flow cytometry.ResultsHigh titers of IFNγ, IL-4, IL-8, and IL-10 serum cytokines were found in the acute individuals. Elevated IL-4 serum levels were found in convalescent and asymptomatic seropositive individuals. High titers of IgG1 subclass were observed across the three cohorts analyzed. Neutralizing antibody response against CHOV-GP was detectable in few acute individuals but was strong in both convalescent and asymptomatic seropositive individuals.ConclusionA Th1/Th2 cytokine signature is characteristic during acute mild HCPS caused by CHOV infection. High expression of Th2 and IL-8 cytokines are correlated with clinical parameters in acute mild HCPS. In addition, a strong IL-4 signature is associated with different cohorts, including asymptomatic individuals. Furthermore, asymptomatic individuals presented high titers of neutralizing antibodies.

Hantavirus cardiopulmonary syndrome (HCPS) caused by Andes orthohantavirus (ANDV) in South America is a public health threat due to the significant rate of mortality and the lack of a specific treatment. Interestingly, the virus does not produce cytopathic effect, thereby the strong antiviral immune response is suspected to contribute to pathogenesis, hence is important to understand the balance between protective and harmfully immunity. CD4+ T regulatory cells (Treg) are essential to control an exacerbated immune response. In human ANDV infection, little is known about CD4+ Treg cells, which may be involved in control immunopathology associated to the infection. In this report, we characterize the phenotype of memory CD4+ Tregs in a HCPS survivor's cohort. Based on the expression of CXCR3, CCR4, and CCR6, we identified different Th-like Treg populations in ANDV survival's PBMCs. In addition, the effect of ANDV-glycoprotein virus like particles (VLP) was determined. We demonstrated that memory CD4+ Treg from HCPS present a specific phenotype, showing higher frequency of PD-1 compared to healthy donors (HD). In addition, it was observed a decrease in the frequency of Th1-like memory CD4+ Treg in HCPS, important to highlight that this signature could be preserved even years after resolution of infection. Moreover, to gain insight in the mechanism involved, we evaluated whether ANDV-glycoprotein (GP) VLP could modulate CD4+ Treg. Interestingly, ANDV-GP VLP induced a decrease in the frequency of CXCR3 (Th1-like) and an increase in CCR4 (Th2-like) memory CD4+ Treg in both HD and HCPS PBMCs, indicating that ANDV-GP could specifically act over CXCR3 and CCR4 in CD4+ Treg. This report contributes to the study of human CD4+ Treg cells in ANDV infection.

Here we aim to describe early mutational events across samples from publicly available SARS-CoV-2 sequences from the sequence read archive and GenBank repositories. Up until 27 March 2020, we downloaded 50 illumina datasets, mostly from China, USA (WA State) and Australia (VIC). A total of 30 datasets (60%) contain at least a single founder mutation and most of the variants are missense (over 63%). Five-point mutations with clonal (founder) effect were found in USA next-generation sequencing samples. Sequencing samples from North America in GenBank (22 April 2020) present this signature with up to 39% allele frequencies among samples (n = 1,359). Australian variant signatures were more diverse than USA samples, but still, clonal events were found in these samples. Mutations in the helicase, encoded by the ORF1ab gene in SARS-CoV-2 were predominant, among others, suggesting that these regions are actively evolving. Finally, we firmly urge that primer sets for diagnosis be carefully designed, since rapidly occurring variants would affect the performance of the reverse transcribed quantitative PCR (RT-qPCR) based viral testing.

For this, a plan of actions was elaborated supported by the qualitative methodology, starting from the theoretical indicators of emotional intelligence and the main deficiencies detected, valued in their initial state, by empirical methods of the investigation such as interview, observation, psychometric techniques and others, designed so that the athlete achieves an optimal psychological state thus raising their sporting performance.

Rhizobial NodD proteins and appropriate flavonoids induce rhizobial nodulation gene expression. In this study, we show that the nodD1 gene of Sinorhizobium fredii HH103, but not the nodD2 gene, can restore the nodulation capacity of a double nodD1/nodD2 mutant of Rhizobium tropici CIAT 899 in bean plants (Phaseolus vulgaris). S. fredii HH103 only induces pseudonodules in beans. We have also studied whether the mutation of different symbiotic regulatory genes may affect the symbiotic interaction of HH103 with beans: ttsI (the positive regulator of the symbiotic type 3 protein secretion system), and nodD2, nolR and syrM (all of them controlling the level of Nod factor production). Inactivation of either nodD2, nolR or syrM, but not that of ttsI, affected positively the symbiotic behavior of HH103 with beans, leading to the formation of colonized nodules. Acetylene reduction assays showed certain levels of nitrogenase activity that were higher in the case of the nodD2 and nolR mutants. Similar results have been previously obtained by our group with the model legume Lotus japonicus. Hence, the results obtained in the present work confirm that repression of Nod factor production, provided by either NodD2, NolR or SyrM, prevents HH103 to effectively nodulate several putative host plants.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease characterized by motor neuron death. A 20% of familial ALS cases are associated with mutations in the gene coding for superoxide dismutase 1 (SOD1). The accumulation of abnormal aggregates of different proteins is a common feature in motor neurons of patients and transgenic ALS mice models, which are thought to contribute to disease pathogenesis. Developmental morphogens, such as the Wnt family, regulate numerous features of neuronal physiology in the adult brain and have been implicated in neurodegeneration. β-catenin is a central mediator of both, Wnt signaling activity and cell-cell interactions. We previously reported that the expression of mutant SOD1 in the NSC34 motor neuron cell line decreases basal Wnt pathway activity, which correlates with cytosolic β-catenin accumulation and impaired neuronal differentiation. In this work, we aimed a deeper characterization of β-catenin distribution in models of ALS motor neurons. We observed extensive accumulation of β-catenin supramolecular structures in motor neuron somas of pre-symptomatic mutant SOD1 mice. In cell-cell appositional zones of NSC34 cells expressing mutant SOD1, β-catenin displays a reduced co-distribution with E-cadherin accompanied by an increased association with the gap junction protein Connexin-43; these findings correlate with impaired intercellular adhesion and exacerbated cell coupling. Remarkably, pharmacological inhibition of the glycogen synthase kinase-3β (GSK3β) in both NSC34 cell lines reverted both, β-catenin aggregation and the adverse effects of mutant SOD1 expression on neuronal differentiation. Our findings suggest that early defects in β-catenin distribution could be an underlying factor affecting the onset of neurodegeneration in familial ALS.

Su detección y tratamiento precoz reduce la morbimortalidad coronaria. A pesar de la disponibilidad de un tratamiento eficaz, la HF está poco diagnosticada y tratada. La identificación de los casos índices y la posterior detección en cascada familiar utilizando los niveles de cLDL y la detección genética es la estrategia más coste-efectiva para la detección de nuevos casos. Chile, isolated by a hyper-arid desert in the north, the Andes Range to the east and the Pacific and Antarctic waters (west and south), has a highly endemic flora. This hotspot of biodiversity is in danger not only due to increasing desertification, but also because human activities can diminish agrobiodiversity. Quinoa (Chenopodium quinoa Willd.) is an Andean species producing highly nutritious grains, which almost disappeared from Chile during the Spanish colonization. Today less than 300 small-scale and highly isolated farmers still grow it as a rain-fed crop. This review describes the biogeographical-social context of quinoa in Chile, and its high genetic diversity as a product of a long domestication process, resulting in numerous local landraces whose conservation and use for breeding improved varieties is of paramount importance. We suggest the term “lighthouse crop” to emphasize its contribution to small scale ecological and bio diverse agriculture, particularly in stressful environments, to promote a healthier nutrition and more equitable markets in the world. Furthermore this crop and its exceptional nutritional properties were invoked by the United Nations Food and Agriculture Organization (FAO) to promote its use worldwide, and to declare 2013 the International Year of Quinoa.

Excessive oxidative stress induced by reactive oxygen species (ROS), reactive nitrogen species (RNS), and reactive metabolites of carcinogens alters cellular homeostasis, leading to genetic/epigenetic changes, genomic instability, neoplastic transformation, and cancer initiation/progression. As a protective mechanism against oxidative stress, antioxidant/detoxifying enzymes reduce these reactive species and protect normal cells from endo-/exogenous oxidative damage. The transcription factor nuclear factor-erythroid 2 p45 (NF-E2)-related factor 2 (Nrf2), a master regulator of the antioxidative stress response, plays a critical role in the expression of many cytoprotective enzymes, including NAD(P)H:quinine oxidoreductase (NQO1), heme oxygenase-1 (HO-1), UDP-glucuronosyltransferase (UGT), and glutathione S-transferase (GST). Recent studies demonstrated that many dietary phytochemicals derived from various vegetables, fruits, spices, and herbal medicines induce Nrf2-mediated antioxidant/detoxifying enzymes, restore aberrant epigenetic alterations, and eliminate cancer stem cells (CSCs). The Nrf2-mediated antioxidant response prevents many age-related diseases, including cancer. Owing to their fundamental contribution to carcinogenesis, epigenetic modifications and CSCs are novel targets of dietary phytochemicals and traditional Chinese herbal medicine (TCHM). In this review, we summarize cancer chemoprevention by dietary phytochemicals, including TCHM, which have great potential as a safer and more effective strategy for preventing cancer.

Quinoa (Chenopodium quinoa Willd.) is a genetically diverse Andean crop that has earned special attention worldwide due to its nutritional and health benefits and its ability to adapt to contrasting environments, including nutrient-poor and saline soils and drought stressed marginal agroecosystems. Drought and salinity are the abiotic stresses most studied in quinoa; however, studies of other important stress factors, such as heat, cold, heavy metals, and UV-B light irradiance, are severely limited. In the last few decades, the incidence of abiotic stress has been accentuated by the increase in unpredictable weather patterns. Furthermore, stresses habitually occur as combinations of two or more. The goals of this review are to: (1) provide an in-depth description of the existing knowledge of quinoa’s tolerance to different abiotic stressors; (2) summarize quinoa’s physiological responses to these stressors; and (3) describe novel advances in molecular tools that can aid our understanding of the mechanisms underlying quinoa’s abiotic stress tolerance.

The APOA1/C3/A4/A5 gene cluster encodes important regulators of fasting lipids, but the majority of lipid metabolism takes place in the postprandial state and knowledge about gene regulation in this state is scarce. With the aim of characterizing possible regulators of lipid metabolism, we studied the effects of nine single nucleotide polymorphisms (SNPs) during postprandial lipid metabolism. Eighty-eight healthy young men were genotyped for APOA1 -2630 (rs613808), APOA1 -2803 (rs2727784), APOA1 -3012 (rs11216158), APOC3 -640 (rs2542052), APOC3 -2886 (rs2542051), APOC3 G34G (rs4520), APOA4 N147S (rs5104), APOA4 T29T (rs5092), and A4A5_inter (rs1263177) and were fed a saturated fatty acid-rich meal (1g fat/kg of weight with 60% fat, 15% protein and 25% carbohydrate). Serial blood samples were extracted for 11 h after the meal. Total cholesterol and fractions [HDL-cholesterol, LDL-cholesterol, trifacylglycerols (TGs) in plasma, TG-rich lipoproteins (TRLs) (large TRLs and small TRLs), apolipoprotein A-I and apolipoprotein B] were determined. APOA1 -2803 homozygotes for the minor allele and A4A5_inter carriers showed a limited degree of postprandial lipemia. Carriers of the rare alleles of APOA4 N147S and APOA4 T29T had lower APOA1 plasma concentration during this state. APOC3 -640 was associated with altered TG kinetics but not its magnitude. We have identified new associations between SNPs in the APOA1/C3/A4/A5 gene cluster and altered postprandial lipid metabolism.

In the El Volcan and Rodeo de los Bueyes areas, Andean Principal Cordillera (east of Santiago; 33°50'S), an Upper Oligocene-Lower Miocene volcanic series belonging to the Abanico Formation (Late Eocene-Early Miocene) is exposed. The rock successions outcropping in both areas, ca. 3,300 m total thickness, have been affected by very low-grade, non-deformative metamorphism in the prehnite-pumpellyite facies. This is represented by the widespread development of secondary mineral assemblages composed of epidote, mixed-layer chlorite-smectite, albite, quartz, white mica, and titanite. These mineral assemblages also contain pumpellyite, prehmte or prehnite+actinolite in a few samples. Chemical characteristics, such as low compositional variability of mixed-layer chlorite-smectite and actino-lite independent from the metadomain where these phases are hosted, along with a high proportion of chlorite layers in the former, suggest that these phases closely represent the whole rock effective bulk composition. On the contrary, pumpellyite compositions show a higher variability and a strong metadomain control evidencing its lower equilibration kinetics and leaving only those that grow in the rock matrix as the more closely representative of a whole rock effective bulk compositional control. Mineral assemblages from both areas show evidence of having been formed during the same metamorphic event where reactions have ocurred equivalently between them. However, local differences in chemical variables controlling this process are recognizable. A higher CO2 concentration in the fluids during the metamorphism in the El Volcán area than in the Rodeo de los Bueyes area is suggested by the scarce development of prehnite and the ubiquitous presence of calcite in the metamorphic assemblages of the former. Pressure and temperature of this metamorphic event have been roughly estimated to be lower than 2-3 kbar and 320°C, respectively.En las areas de El Volcán y Rodeo de los Bueyes, Cordillera Principal al este de Santiago (33°50'S), aflora una serie volcánica del Oligoceno Superior-Mioceno Inferior perteneciente a la Formación Abanico (Eoceno Tardío-Mioceno Temprano). Las sucesiones de roca expuestas en ambas areas, de ca. 3.300 m de espesor total, fueron afectadas por metamorfismo no deformativo de muy bajo grado en facies prehnita-pumpellyita. Esto está representado en el desarrollo extensivo de arreglos de minerales secundarios formados por epidota, inter-estratificados de clorita-esmectita, albita, cuarzo, mica blanca, y titanita. Estos arreglos minerales contienen además pumpellyita, prehnita o prehnita+actinolita en algunas muestras. Las características químicas tales como una baja variabilidad composicional de los interestratificados de clorita-esmectita y actinolita independientemente del dominio donde se hospedan estas fases, junto con una alta proporción de capas de clorita en las primeras, sugieren que estas fases representan cercanamente la composición efectiva de la roca total. Por el contrario, la composición de las pum-pellyitas muestra una mayor variabilidad y un fuerte control del metadominio donde éstas se desarrollan evidenciando una baja cinética de equilibrio y dejando sólo a aquellas desarrolladas en la matriz como formadas bajo un control composicional efectivo de la roca total. Los arreglos minerales de ambas areas muestran evidencias de haberse formado bajo un mismo evento metamórñco bajo el cual las reacciones se han desarrollado de manera equivalente. Aún así se reconocen diferencias químicas locales que controlan este proceso. Una mayor concentración de CO2 en los fluidos durante el metamorfismo en el area de El Volcán, con respecto al area de Rodeo de los Bueyes, se ve representada en la presencia escasa de prehnita, y abundante de calcita, en sus asociaciones minerales. La presion y temperatura de este evento metamórfico han sido gruesamente estimadas en valores inferiores a 2-3 kbar y 320°C, respectivamente.

The apolipoprotein E (apoE) gene promoter (−219G/T) polymorphism has been associated with increased risk of myocardial infarction, premature coronary heart disease, and decreased plasma apoE concentrations. We examined whether the −219G/T polymorphism could modify the postprandial response of triacylglycerol-rich lipoproteins (TRLs). Fifty-one healthy apoE 3/3 male volunteers (14GG, 29GT, and 8TT) were given a vitamin A fat-loading test consisting of 1 g of fat/kg body weight and 60,000 IU of vitamin A per m2 of body surface area. Blood samples were taken at time 0 and every hour until the sixth hour, and every 2 hours and 30 minutes until the eleventh hour. Cholesterol, triacylglycerols (TGs), and apoE were determined in plasma; and cholesterol, TG, apoB-100, apoB-48, and retinyl palmitate (RP) were analyzed in lipoprotein fractions. Postprandial lipemia data revealed that subjects with the −219TT genotype had a higher postprandial response of large TRL-cholesterol (P < 0.03), large TRL-triacylglycerols (P < 0.001), large TRL-RP (P < 0.004), and small TRL-apoB-48 (P < 0.03) than carriers of the −219G allele. Moreover, the −219TT subjects had the lowest postprandial levels of serum apoE (P < 0.05).In conclusion, the −219G/T polymorphism may influence TRL metabolism during the postprandial period, thus prolonging postprandial lipemia in subjects with the TT genotype.

The metabolic syndrome is a cluster of cardiometabolic alterations that include the presence of arterial hypertension, insulin resistance, dyslipidemia, and abdominal obesity. Obesity is associated with a chronic inflammatory response, characterized by abnormal adipokine production, and the activation of proinflammatory signalling pathways resulting in the induction of several biological markers of inflammation. Macrophage and lymphocyte infiltration in adipose tissue may contribute to the pathogenesis of obesity-mediated metabolic disorders. Adiponectin can either act directly on macrophages to shift polarization and/or prime human monocytes into alternative M2-macrophages with anti-inflammatory properties. Meanwhile, the chronic inflammation in adipose tissue is regulated by a series of transcription factors, mainly PPARs and C/EBPs, that in conjunction regulate the expression of hundreds of proteins that participate in the metabolism and storage of lipids and, as such, the secretion by adipocytes. Therefore, the management of the metabolic syndrome requires the development of new therapeutic strategies aimed to alter the main genetic pathways involved in the regulation of adipose tissue metabolism.
