Genetically encoded optical sensors and advancements in microscopy instrumentation and techniques have revolutionized the scientific toolbox available for probing complex biological processes such as release of specific neurotransmitters. Most genetically encoded optical sensors currently used are based on fluorescence and have been highly successful tools for single-cell imaging in superficial brain regions. However, there remains a need to develop new tools for reporting neuronal activity in vivo within deeper structures without the need for hardware such as lenses or fibers to be implanted within the brain. Our approach to this problem is to replace the fluorescent elements of the existing biosensors with bioluminescent elements. This eliminates the need of external light sources to illuminate the sensor, thus allowing deeper brain regions to be imaged noninvasively. Here, we report the development of the first genetically encoded neurotransmitter indicators based on bioluminescent light emission. These probes were optimized by high-throughput screening of linker libraries. The selected probes exhibit robust changes in light output in response to the extracellular presence of the excitatory neurotransmitter glutamate. We expect this new approach to neurotransmitter indicator design to enable the engineering of specific bioluminescent probes for multiple additional neurotransmitters in the future, ultimately allowing neuroscientists to monitor activity associated with a specific neurotransmitter as it relates to behavior in a variety of neuronal and psychiatric disorders, among many other applications.

Abstract Posterior Reversible Encephalopathy Syndrome (PRES) manifests as transient neurological symptoms and cerebral edema, commonly associated with immunosuppressive drugs (ISDs) in transplant recipients. ISDs can lead to endothelial dysfunction and compromise the blood–brain barrier. Typically, PRES exhibits identifiable MRI patterns, often demonstrating vasogenic edema in the bilateral parieto‐occipital white matter. Identifying unique presentations, such as the recently observed “lentiform fork sign,” commonly seen in uremic encephalopathy, emphasizes this syndrome's broad spectrum manifestations. A 19‐year‐old male, who underwent bilateral lung and liver transplantation, experienced a bilateral tonic–clonic seizure of unknown onset 47 days post‐surgery. MRI findings revealed an unconventional PRES pattern, featuring the “lentiform fork sign” as bilateral symmetrical vasogenic edema in the basal ganglia, surrounded by a hyperintense rim outlining the lentiform nucleus bilaterally. Subsequent management, including ISD modification and magnesium supplementation, resulted in clinical and neuroimaging resolution. An almost complete clinical and radiological resolution was achieved after 14 days. The occurrence of PRES in transplant recipients highlights the intricate interplay among ISDs, physiological factors, and cerebrovascular dynamics, potentially involving direct neurovascular endothelial toxicity and disruption of the blood–brain barrier. Neuroimaging plays a pivotal role in diagnosis. The distinctive “lentiform fork sign” was observed in this patient despite the absence of typical metabolic disturbances. Management strategies usually involve reducing hypertension, discontinuing ISDs, correcting electrolyte imbalances, and initiating antiseizure drugs if necessary. Identifying the presence of the “lentiform fork sign” alongside typical PRES edema in a patient lacking renal failure emphasizes that this manifestation is not solely indicative of uremic encephalopathy. Instead, it might represent the final common pathway resulting from alterations in the blood–brain barrier integrity within the deep white matter. Understanding such atypical imaging manifestations could significantly aid earlier and more precise diagnosis, influencing appropriate management decisions.

Abstract The state of Espirito Santo is the most important producer and exporter of ornamental quarry stone. In its various quarries, it uses bench heights that range from 5, 20, 50 and 100 meters. The high altitudes raise concerns about the stability of the slopes. Therefore, the study focuses on determining the value of the safety factor (SF) for these heights using the limit equilibrium method. To make this possible, the values of simple uniaxial compression, Geological Strength Index (GSI), the disturbance factor, and intact rock parameter must be obtained. To obtain the GSI, it is necessary to establish a correlation with the Rock Mass Rating, the values of which are determined using a scan line. The uniaxial compression values were treated statistically, obtaining classes at 75, 100, 125, 150, 175, and 200 Mpa. The 2D slide software was used to obtain the SF values for the respective heights. With the results obtained, these rocks do not present stability problems. The correct choice of GSI is of paramount importance for a good analysis, as it directly affects the results. Using this technique to determine safety factors is appropriate, but it is necessary to analyze more robust methods to compare the results.

En el presente artículo analizamos uno de los principales escenarios que Bolsonaro utilizó para comunicarse con sus bases durante toda su legislatura: los lives semanales en su cuenta oficial de Instagram. Así, estudiamos una muestra de diez lives, describiendo cómo su comunicación no verbal podía activar un proceso de humanización y aproximación del líder, normalizando formas de violencia y fortaleciendo la polarización política. La metodología empleada en el análisis se sustenta en la grounded theory, que sistematiza la investigación cualitativa a través de la construcción de hipótesis y teorías mediante la recolección y el análisis de datos. Este procedimiento de análisis propone una metodología inductiva que extrae las categorías de la comunicación no verbal a través de la descripción y clasificación del material y de las fuentes de datos por analizar. La contextualización e interpretación de los resultados se apoya en un trabajo etnográfico de campo durante la campaña electoral. Los resultados apuntan al uso de elementos amateurs en los videos que generan una aproximación de Bolsonaro con los ciudadanos, así como la inclusión de movimientos corporales que muestran su carisma como un líder imperfecto y del paralenguaje basado en el humor para la construcción de un personaje violento.

Pulmonary sequestration is an uncommon congenital malformation of the lung, generally diagnosed in childhood or adolescence, corresponding to dysplastic lung tissue not communicating with the rest of vascular or bronchial lung system but receiving an arterial blood supply from systemic arteries. Currently, surgical resection is usually indicated in order to prevent or treat related symptoms or complications, although controversy exists regarding its use in asymptomatic patients and adults. We present the case of a 32-year-old pregnant woman with acute chest pain and vomiting diagnosed with intralobar sequestration at 32+2 weeks of gestation and treated with pulmonary lobectomy after giving birth by cesarean section at 33+0 weeks of gestation.

One of the main challenges in super high-density olive is maintaining hedgerow structure suitable for mechanical harvesting. Deficit irrigation strategies (DI) can contribute to this maintenance. In this study, three DI treatments at 50% of the Control (DI-1: budbreak to pre-bloom, DI-2: fruit set to pit hardening, and DI-3: pit hardening to harvest) were applied to 6-year-old Arbequina hedgerows in a semi-arid environment of Argentina over three consecutive seasons. The mean applied water reduction was 12% for DI-1 and 16% for DI-2 and DI-3, compared to Control (100% of ETc throughout the cycle). DI-3 achieved the narrowest hedgerows at harvest, where canopy height was responsive to irrigation and canopy width was affected by fruit load but not by the irrigation regime. DI-1 demonstrated similar fruit and oil yields to Control throughout the experiment, while DI-2 only had comparable yields to Control in Off-season. DI-3 had the lowest oil yields, averaging – 27% in Off and On-seasons. The fruit number was similar among treatments in all seasons, indicating that differences in final yield were related to fruit size and oil concentration. Further studies are needed to determine the optimal duration and intensity of the water deficit for DI-3. [ABSTRACT FROM AUTHOR]

Plants rely on complex regulatory mechanisms to ensure proper growth and development. As plants are sessile organisms, these mechanisms must be flexible enough to adapt to changes in the environment. GROWTH-REGULATING FACTORS (GRFs) are plant-specific transcription factors that act as a central hub controlling plant growth and development, which offer promising biotechnological applications to enhance plant performance. Here, we analyze the complex molecular mechanisms that regulate GRFs activity, and how their natural and synthetic variants can impact on plant growth and development. We describe the biological roles of the GRFs and examine how they regulate gene expression and contribute to the control of organ growth and plant responses to a changing environment. This review focuses on the premise that unlocking the full biotechnological potential of GRFs requires a thorough understanding of the various regulatory layers governing GRF activity, the functional divergence among GRF family members, and the gene networks that they regulate. Therefore, 33 patients were included in the reassessment after nine years of follow-up and 92 patients were included in the Cox mortality analysis with IL-6 as a time-dependent covariate. Regarding the inflammatory profile, in patients who survived after nine years, there was a significant increase in IL-6 [0.4 (0.2– 0.8) vs 5.7 (3.4– 11) pg/mL; p < 0.001] and reduction in lymphocyte count [2.1 (1.6– 2.4) vs 1.4 (1.2– 2.1)10^9/L; p < 0.01] with an increase in the neutrophil/lymphocyte ratio (2.0 ± 0.7 vs 2.7 ± 1.2; p = 0.003). The Cox mortality model did not show a statistical significance influence of IL-6 assessed during the follow-up.Conclusion: There was a progressive increase in IL-6 during the follow-up, however, without influence on mortality.Keywords: chronic obstructive pulmonary disease, inflammation, interleukin-6, severity of illness index, mortality

El neuromarketing como objeto de estudio en el campo de la Comunicación está ganando cada vez más atención académica en el contexto español, sobre todo en los últimos años. En este texto analizamos la evolución de este tema de investigación en las tesis doctorales durante el período 2007-2018, justo en el momento de transición de las normativas sobre estudios de postgrado a nivel nacional. A partir de una metodología de metaanálisis, de tipología mixta, se evalúan los centros científicos de producción y los autores, las configuraciones de las líneas temáticas, los conceptos y objetivos más comunes, así como el diseño metodológico en cuanto a muestras, datos, técnicas y triangulaciones dominantes. Aunque el neuromarketing continúa teniendo una presencia residual en el campo académico, el interés epistemológico que genera sigue en aumento, en particular en ámbitos de investigación fronterizos directamente vinculados con la comunicación institucional u organizacional, la comunicación de eventos y la comunicación de marca.
