Approximately 660,000 women are diagnosed with cervical cancer annually. Current screening options such as cytology or human papillomavirus testing have limitations, creating a need to identify more effective ancillary biomarkers for triage. Here, we evaluated whether metabolomic analysis of cervical mucus metabolism could be used to identify biomarkers of cervical intraepithelial neoplasia (CIN) and cervical cancer. The case–control group consisted of 181 CIN, 69 squamous cell carcinoma (SCC) patients, and 48 healthy controls in the primary cohort. We undertook metabolomic analyses using ultra‐HPLC–tandem mass spectrometry. Univariate and multivariate analyses were carried out to profile metabolite characteristics, and receiver operating characteristic (ROC) analysis identified biomarker candidates. Five metabolites conferred the highest discriminatory power for SCC: oxidized glutathione (GSSG) (area under the ROC curve, 0.924; 95% confidence interval, 0.877–0.971), malic acid (0.914, 0.859–0.968), kynurenine (0.884, 0.823–0.945), GSSG/glutathione (GSH) (0.936, 0.892–0.979), and kynurenine/tryptophan (0.909, 0.856–0.961). Malic acid was the best marker for detection of CIN2 or worse (0.858, 0.793–0.922) and was a clinically useful metabolite. We confirmed the reproducibility of the results by validation cohort. Additionally, metabolomic analyses revealed eight pathways strongly associated with cervical neoplasia. Of these, only the tricarboxylic acid cycle was strongly associated with all CINs and cancer, indicating active energy production. Aberrant arginine metabolism by decreasing arginine and increasing citrulline might reduce tumor immunity. Changes in cysteine‐methionine and GSH pathways might drive the initiation and progression of cervical cancer. These results suggest that metabolic analysis can identify ancillary biomarkers and could improve our understanding of the pathophysiological mechanisms underlying cervical neoplasia. Plants acquire potassium (K) not only from exchangeable K fractions but also from non-exchangeable K fractions. However, factors affecting the amount of K supplied from the non-exchangeable K fraction have not been well understood. In this study, the effects of soils and exchangeable K on the amount of K supplied to rice from the non-exchangeable K fraction were investigated for six Japanese paddy soils diluted with different rates of quartz sand. The amounts of K supplied from the non-exchangeable K fractions (recharged K) ranged from 14.4 to 137 mg K2O kg−1, equivalent to 10 to 61% of the total K acquired by rice. The amounts of recharged K were affected by the exchangeable K levels in three out of six soils. Although the cultivation experiments were conducted under various K conditions including severe K deficiency, exchangeable K was detected after cultivation in all soils, and the contents of exchangeable K were almost identical for each soil regardless of the dilution ratio of soil with quartz sand. Accordingly, there would be at least three types of non-exchangeable K fractions which dissolve into soil solution to attain a specific level of exchangeable K: (i) a non-exchangeable K fraction that releases K regardless of the exchangeable K level; (ii) a non-exchangeable K fraction that easily releases K with the decrease in exchangeable K under sufficient K conditions without inducing K-deficient symptoms in rice; and (iii) a non-exchangeable K fraction that releases K with the decrease in exchangeable K only under severe K-deficient conditions with accompanying deficient symptoms in rice. The application of K fertilizer would reduce the amount of recharged K acquired by rice from fractions (ii) and (iii), whereas fraction (i) would remain unaffected by the fertilized K until the level of exchangeable K reaches a specific level for each soil. The release of K from a fraction (iii) only occurred under conditions of severe K deficiency; hence, farmers should not rely on this fraction for K supply. [ABSTRACT FROM AUTHOR]

Currently, human papillomavirus tests and cytology are used to screen for cervical cancer. However, more accurate ancillary screening tests are needed. MicroRNAs (miRNAs) and cytokines are promising biomarkers that are aberrantly expressed in cervical cancer. Therefore, the potential of developing new screening markers based on the levels of miRNAs and cytokines in serum and local mucus samples from the same patients with cervical neoplasia was investigated. miRNA screening was performed by microarray and measurement using real‐time reverse‐transcriptase PCR. Cytokine were measured using multiplex bead assay, and changes in expressions were analyzed based on disease severity. As lesions progressed, miR‐20b‐5p, −155‐5p, −144‐3p, −451a, and −126‐3p expression levels were increased in mucus, and miR‐16‐5p, −223‐3p, and ‐451a expression levels were decreased in serum. Regarding cytokines, IL‐6, IL‐8, monocyte chemoattractant protein‐1, Eotaxin, interferon‐γ, and RANTES were increased, whereas granulocyte–colony‐stimulating factor (G‐CSF) was significantly decreased in mucus. miRNAs and cytokines in serum did not have high diagnostic accuracy. However, a combination of miR‐20b‐5p, ‐451a, ‐126‐3p, Eotaxin, as well as G‐CSF in mucus samples, had high diagnostic accuracy with an area under the receiver operating characteristic curve of 0.989 (0.979–0.999). Aim: Minimally invasive surgery (MIS) has been introduced as an alternative to more radical surgical procedures. The Japan Society of Gynecologic and Obstetric Endoscopy and Minimally Invasive Therapy conducted a cross‐sectional questionnaire survey to ascertain the status of MIS for endometrial cancer. Methods: The survey was conducted between May 10 and June 30, 2022. The questionnaire included information on personal attributes, academic affiliations, qualifications, hysterectomies, and intraoperative procedures performed. Results: The total number of questionnaire respondents was 436 (9.2% of the membership). The hysterectomy methods and percentage performed were as follows: simple total hysterectomy (equivalent to benign surgery), 3%; simple total hysterectomy with care to avoid shaving the cervix, 31%; extended total hysterectomy, 48%; and modified radical hysterectomy, 15%. An analysis of hysterectomies performed using MIS for endometrial cancer by qualified gynecologists of endoscopy or board‐certified gynecologic oncologists showed a tendency not to choose simple total hysterectomy compared to the gynecologists who did not hold certification (p = 0.019, p = 0.045, and p = 0.010, respectively). Additionally, 67% of respondents did not use uterine manipulators, and 59% of the respondents did not perform lymph node dissection following the guidelines for treating endometrial cancer in Japan. Conclusion: This study provided the current status of MIS for endometrial cancer in Japan. The hysterectomy method, use of uterine manipulators, and criteria for omitting lymph node dissection were generally in agreement with the guidelines. Currently, an extra‐fascial simple hysterectomy, including at least not shaving the cervix, was a major method for early invasive endometrial cancer using MIS. [ABSTRACT FROM AUTHOR]

Actinernoidea is a superfamily of the suborder Anenthemonae of the order Actiniaria, subclass Hexacorallia. This superfamily has been diagnosed by a peculiar endocoeletic mesenterial arrangement and included two families, Actinernidae and Halcuriidae. Although the monophyly of this superfamily is anticipated based on preceding molecular phylogenetic works, the relationship between these two families was not certain because of the rarity of actinernoidean anemones. We conducted comprehensive sampling in Japan, where the highest diversity of actinernoideans is known, and conducted phylogenetic analyses using nuclear and mitochondrial gene markers. According to the comprehensive analyses, both Actinernidae and Halcuriidae were not indicated as monophyletic but rather as poly- or paraphyletic. Based on our phylogeny reconstruction, we propose a new classification for Actinernoidea composed of three families, including Isactinernidae fam. nov., and seven genera, including Isohalcurias gen. nov. We also describe four new species, Halcurias hiroomii sp. nov., H. fragum sp. nov., Isohalcurias citreum sp. nov., and I. malum sp. nov.; and propose a new combination, Isohalcurias carlgreni comb. nov. This work is the first study of Actinernoidea that comprehensively analyzes its phylogeny and rearranges its classification, showing that there is highly divergent fauna in the seas around Japan.
