Background: Drought constitutes a major abiotic stress factor adversely affecting plant growth and productivity. Plant-microbe symbiotic associations have evolved regulatory mechanisms to adapt to environmental stress conditions. However, the interactive effects of different fungi on host growth and stress tolerance under drought conditions remain unclear. Objective: This study explored the effects of varying polyethylene glycol (PEG6000) concentrations (0%, 15%, 25%, and 35%) on the growth and physiological responses of two ectomycorrhizal fungi (Suillus granulatus (Sg) and Pisolithus tinctorius (Pt)) and two dark septate endophytes (Pleotrichocladium opacum (Po) and Pseudopyrenochaeta sp. (Ps)) isolated from the root system of Pinus tabuliformis. Specifically, the study aimed to evaluate six inoculation treatments, including no inoculation (CK), single inoculations with Sg, Pt, Po, Ps, and a mixed inoculation (Sg: Pt : Po: Ps = 1:1:1:1), on the growth and physiological characteristics of P. tabuliformis seedlings under different water regimes: well-watered at 70% ± 5%, light drought at 50% ± 5%, and severe drought at 30% ± 5% of the maximum field water holding capacity. Results: All four fungi exhibited the capacity to cope with drought stress by enhancing antioxidant activities and regulating osmotic balance. Upon successful root colonization, they increased plant height, shoot biomass, root biomass, total biomass, and mycorrhizal growth response in P. tabuliformis seedlings. Under drought stress conditions, fungal inoculation improved seedling drought resistance by increasing superoxide dismutase and catalase activities, free proline and soluble protein contents, and promoting nitrogen and phosphorus uptake. Notably, mixed inoculation treatments significantly enhanced antioxidant capacity, osmotic adjustment, and nutrient acquisition abilities, leading to superior growth promotion effects under drought stress compared to single inoculation treatments. Conclusion: All four fungi tolerated PEG-induced drought stress, with increased antioxidant enzyme activities and osmotic adjustment substances and they promoted the growth and enhanced drought resistance of P. tabuliformis seedlings. Introduction: Parasites can facilitate their own spread and reproduction by manipulating insect hosts behavior, as seen in the interaction between Thitarodes xiaojinensis and Ophiocordyceps sinensis. Infection by O. sinensis leads to the mummification of T. xiaojinensis larvae, but the underlying mechanisms remain mysterious. Methods: The morphology of O. sinensis infected larvae and fungal growth were first observed. Subsequently, the metabolite changes in the larvae before and after infection with the fungus were analyzed by LC/MS and targeted metabolomics. The expression of mannitol-related genes was detected using RT-qPCR, and morphological changes in larvae were observed after injection of different concentrations of mannitol into the O. sinensis-infected larvae. Results: Significant changes were found in phenotype, fungal morphology in hemocoel, larval hardness, and mannitol metabolites in infected, mummified 0 h larvae and larvae 5 days after mummification behavior. Surprisingly, the occurrence of mummification behavior was accompanied by fungal dimorphism, as well as the absence of mannitol in both infected and non-infected larvae, until the initial accumulation of mannitol and the expression of mannitol-associated genes occurred at the time ofmummification behavior. The presence of mannitol may promote fungal dimorphism to mediate changes in fungal toxicity or resistance, leading to the end of the fungus-insect coexistence period and the incidence of mummification behavior. Furthermore, mannitol injections increase the mummification rate of the infected larvae without significant difference from the normal mummification phenotype. Pepper is a vegetable that is used and cultivated worldwide. The quality and production of pepper are reduced by fruit anthracnose disease caused by Colletotrichum scovillei. The highly conserved fungal sucrose non‐fermenting 1 (SNF1)‐related AMPK protein kinases are implicated in the pathogenicity and differentiation of several destructive foliar fungal pathogens; in addition, they affect fungal growth in the presence of glucose at different concentrations and under different environmental stresses. However, the function of SNF1 in C. scovillei is unclear. We functionally characterized SNF1 of C. scovillei using a targeted deletion mutant (ΔCssnf1) generated by homology‐dependent replacement. ΔCssnf1 was severely impaired in mycelial growth under glucose‐limited conditions; multiple defects in conidiation, conidial germination, appressorium formation, lipid degradation and vacuole formation were also observed. Compared to the wild type, ΔCssnf1 caused anthracnose disease of reduced severity on intact and wounded pepper fruits. These defects were recovered in the complemented strain (Cssnf1c). Silage has been identified as a source of different microbial toxins, that may impair farm animal health and productivity as human health can also be compromised. In this sense, the aim of this study was to determine the impact of silage additives on the concentrations of deoxynivalenol (DON) and zearalenone (ZEN) mycotoxins and, eventually, to evaluate the hygienic quality of orchardgrass (Dactylis glomerata L.) silage based on the concentration of them compared to control silage. This study evaluated the influence of biological and chemical additives used in six different varieties of orchardgrass silage on DON and ZEN mycotoxin contents for the first time. The content of both fusariotoxins (DON and ZEN) in fresh matter and grass silage were below the threshold stipulated by the European Commission. The concentration of DON ranges from ~21.86 to 37.26 ng/kg, ~10.21 to 15 ng/kg, ~20.72 to 29.14 ng/kg; and ZEN range from ~3.42 to 7.87 ng/kg, ~3.85 to 8.62 ng/kg and ~2.15 to 5.08 ng/kg, in control, biological and chemical silages, respectively. In general, the biological additive was more efficient for preventing DON contamination, whereas the chemical additive was more efficient for preventing ZEN contamination in grass silage. In summary, the results obtained in this work demonstrate that biological and chemical additives can inhibit fungal growth and mycotoxin production on Dactylis glomerata L. silage and whose use could prevent animal and human diseases. Microbial volatile organic compounds (VOCs) emitted from fungi are known as their secondary metabolites from environmental sources. However, their physiological roles remain to be unclear. Even though the roles are still unknown, VOCs are deliberately released to convey information to both homologous and non-homologous organisms. We investigated the effects of single VOCs (hexanal, benzaldehyde, heptanal, 2-ethyl-1-hexanol, 3-octanone, 2-undecanone, 3-octanol, 2-Phenylethanol, 2-phenyl-2-propanol, phenylbenzaldehyde, 2-pentadecanone, β-trans-bergamotene, β-bisabolene, 2-methyl-5 -(1-methylethyl)pyrazine) on the fungal growth. In parallel, application of the co-culturing system in a growth chamber allowed free gas and VOCs exchange between emitter colonies of Fusarium solani and Aspergillus fumigatus, or between colonies of different growth stages of the same species. Distinct self-inhibition occurred by the emitters of fungal growing colonies against receiver ones on the stage of conidial germination or against the younger colonies at an earlier stage in both fungi. Similarly, the phenomenon of allelopathy appeared to work between growing colonies of F. solani and the germinating conidia or young colonies of A. fumigatus or vice versa. Solid phase microextraction—gas chromatography/mass spectrometry revealed VOCs compounds of each fungi. In F. solani, hexanal and benzaldehyde appeared to be significant inhibitors for colony growth. Benzaldehyde inhibited filamentous growth but not conidial germination. In A. fumigatus, heptanal seemed to be an equivalent effector. The objective was to develop an active chitosan‐based coating and to evaluate its effect on the shelf life and microbial safety of bread. Bacterial cellulose nanofibers (BCNF) and various levels (0.5%, 1%, and 2%) of silver nanoparticles (AgNPs) were in the chitosan (CS) film. Characterization of films was determined by analyzing WVP, ultraviolet barrier, and opacity as well as FTIR, XRD, DSC, TGA, and SEM. The water vapor permeability (WVP) of CS was remarkably (p < .05) decreased from 3.75 × 10−10 to 0.85 × 10−10 g/smPa when filled with BCNF and 2% AgNPs. Thermal and structural properties were enhanced in nanoparticle‐included films. Applying CS/BCNF/AgNPs coatings for bread samples demonstrated a significant improvement in moisture retention and a decrease in the hardness (from 10.2 to 7.05 N for CS and CS/BCNF/1% AgNPs coated samples, respectively). Moreover, microbial shelf life of bread sample increased from 5 to 38 days after packaging with CS/BCNF/2% AgNPs film. After a storage period of 15 days at 25°C, no fungal growth was detected in bread samples which were coated with nanocomposite suspensions containing 1% and 2% AgNPs. However, at the same condition, yeast and mold counts was 7.91 log CFU/g for control sample. In conclusion, the CS/BCNF/2% AgNPs film might have the potential for use as active packaging of bread. Oomycetes such as Saprolegnia sp. are responsible for serious economic losses in aquaculture, the strategies for their control lead to concerns about their efficacy and environmental repercussions, so alternatives for their regulation are being sought. The aim of this study was to evaluate the in vitro susceptibility of 2 representative strains of Saprolegnia sp. toward 2,4‐dihydroxy‐5′‐prenyl‐dihydrochalcone (A); dihydrochalcone isolated from the Chilean shrub Adesmia balsamica; and their derivatives oxyprenylated (B‐‐G) with the purpose to develop naturally occurring products in order to cope with emerging resistance phenomena. The minimum inhibitory concentration (MIC) and minimum oomycetocidal concentration (MOC) values were then determined for the series of compounds tested, in comparison with the conventional fungicides bronopol® and fluconazole plus the natural compound safrole, by quantifying fungal growth. Significant growth inhibition was observed for prenylated derivatives B–G, evidencing dihydrochalcone E as the most active. This compound presented MIC and MOC values (0.24–0.26 μM and 0.30–0.33 μM, respectively) superior to those determined for commercial antifungal in both of the tested pathogenic strains. Fungus-growing termites, like Odontotermes obesus, cultivate Termitomyces as their sole food source on fungus combs which are continuously maintained with foraged plant materials. This necessary augmentation also increases the threat of introducing non-specific fungi capable of displacing Termitomyces. The magnitude of this threat and how termites prevent the invasion of such fungi remain largely unknown. This study identifies these non-specific fungi by establishing the pan-mycobiota of O. obesus from the fungus comb and termite castes. Furthermore, to maximize the identification of such fungi, the mycobiota of the decaying stages of the unattended fungus comb were also assessed. The simultaneous assessment of the microbiota and the mycobiota of these stages identified possible interactions between the fungal and bacterial members of this community. Based on these findings, we propose possible interactions among the crop fungus Termitomyces, the weedy fungus Pseudoxylaria and some bacterial symbiotes. These possibilities were then tested with in vitro interaction assays which suggest that Termitomyces, Pseudoxylaria and certain potential bacterial symbiotes possess anti-fungal capabilities. We propose a multifactorial interaction model of these microbes, under the care of the termites, to explain how their interactions can maintain a predominantly Termitomyces monoculture. New evidence suggests a possible collaboration between the crop fungus, a weedy fungus and symbiotic bacteria in Odontotermes obesus fungus gardens that prevents the growth of unwanted fungi. Mold growth on body donations remains an underreported yet serious issue in anatomical teaching. Bacterial and fungal growth pose health risks to lecturers and students, alongside with ethical and aesthetic concerns. However, limited information exists on the presence of bacteria and fungi on body donations and their underlying causes. To investigate the potential impact of airborne germs on body donation contamination, we conducted indoor air measurements before, during, and after our anatomical dissection course, with outdoor measurements serving as a control. Tissue samples from the dissected body donations were collected to assess the germ load, with qualitative and quantitative microbiological analyses. Air samples from the dissection hall contained no fungi, but various fungal species were identified in the adjacent stairways and outdoors which implies that fungal occurrence in the dissection hall air was independent of lecturers' and students' presence. Moreover, our results indicate that adequate ventilation filters can effectively reduce indoor fungal germs during courses, while the bacterial load in room air appears to increase, likely due to the presence of lecturers and students. Additionally, the tissue samples revealed no bacterial or fungal germs which implies that our ethanol-formalin-based embalming solution demonstrates an effective long-term antimicrobial preservation of corpses. Phosphorus is essential in all cells' structural, metabolic and regulatory functions. For fungal cells that import inorganic phosphate (Pi) up a steep concentration gradient, surface Pi transporters are critical capacitators of growth. Fungi must deploy Pi transporters that enable optimal Pi uptake in pH and Pi concentration ranges prevalent in their environments. Single, triple and quadruple mutants were used to characterize the four Pi transporters we identified for the human fungal pathogen Candida albicans, which must adapt to alkaline conditions during invasion of the host bloodstream and deep organs. A high-affinity Pi transporter, Pho84, was most efficient across the widest pH range while another, Pho89, showed high-affinity characteristics only within one pH unit of neutral. Two low-affinity Pi transporters, Pho87 and Fgr2, were active only in acidic conditions. Only Pho84 among the Pi transporters was clearly required in previously identified Pi-related functions including Target of Rapamycin Complex 1 signaling, oxidative stress resistance and hyphal growth. We used in vitro evolution and whole genome sequencing as an unbiased forward genetic approach to probe adaptation to prolonged Pi scarcity of two quadruple mutant lineages lacking all 4 Pi transporters. Lineage-specific genomic changes corresponded to divergent success of the two lineages in fitness recovery during Pi limitation. Initial, large-scale genomic alterations like aneuploidies and loss of heterozygosity eventually resolved, as populations gained small-scale mutations. Severity of some phenotypes linked to Pi starvation, like cell wall stress hypersensitivity, decreased in parallel to evolving populations' fitness recovery in Pi scarcity, while severity of others like membrane stress responses diverged from Pi scarcity fitness. Among preliminary candidate genes for contributors to fitness recovery, those with links to TORC1 were overrepresented. Since Pi homeostasis differs substantially between fungi and humans, adaptive processes to Pi deprivation may harbor small-molecule targets that impact fungal growth, stress resistance and virulence. Author summary: Fungi must be able to access enough phosphate in order to invade the human body. Virulence of Candida albicans, the most common invasive human fungal pathogen, is known to decrease when one of the proteins that brings phosphate into the fungal cell, called Pho84, is disabled. We identified three more proteins in C. albicans that transport phosphate into the cell. We found that Pho84 plays the largest role among them across the broadest range of environmental conditions. After eliminating all 4 of these transporters, we let two resulting mutants evolve for two months in limited phosphate and analyzed the growth and stress resistance of the resulting populations. We analyzed genomes of representative populations and found that early adaptations to phosphate scarcity occurred together with major changes to chromosome configurations. In later stages of the adaptation process, these large-scale changes disappeared as populations gained small-scale mutations that increased cells' ability to grow in limited phosphate. Some but not all of these favorable mutations improved resistance of evolving populations to stressors like membrane- and cell wall stress. Bats are one of the most diverse mammal groups, with over 1400 species distributed across the planet, and they provide key ecological and economic services. In addition, bats provide an ideal habitat for fungal growth and harbour a high diversity of fungal species. Previous studies have described 423 fungal species associated with bats and bat carcasses. This paper describes a new fungal species, Fusarium rhinolophi, belonging to Nectriaceae, collected from bats in Mengla County, Xishuangbanna Dai Autonomous Prefecture, Yunnan Province, China. Phylogenetic analyses of the combined ITS, CAM, TEF-1α, TUB, RPB1, and RPB2 dataset confirmed that our fungus forms an independent lineage within the F. nisikadoi species complex. Thus, we describe our fungus as a new species of Fusarium based on morphology and molecular phylogeny. A description, phylogenetic tree showing species placement, and micrographs are provided. Many invertebrates and vertebrates secrete mucus from their body surface, which plays a crucial role in protecting themselves from attacks by microorganism or predators. This study reports on the potential functions of mucus secreted from the body surface of larvae of the moth Phauda flammans (Walker) (Lepidoptera: Phaudidae). We tested whether the mucus could inhibit the growth of the fungus Beauveria bassiana PfBb (Bals.‐Criv.) Vuill. (Hypocreales) and attract and/or deter predators such as the crazy ant, Paratrechina longicornis (Latreille) (Hymenoptera: Formicidae), and the predatory bug Eocanthecona furcellata (Wolff) (Hemiptera: Pentatomidae). Our results showed that de‐ and rehydrated mucus concentrations of 0.5–1.0 mg mL−1 had an inhibitory effect on the conidial germination and colony growth of B. bassiana PfBb; however, P. flammans mucus did not significantly affect fungal sporulation. Raw mucus and mucus concentrations of 1.0–4.0 mg mL−1 attracted P. longicornis workers in a Y‐tube olfactometer, whereas the physical viscosity of raw mucus hindered their foraging for food in a laboratory test. The E. furcellata adults did not display a preference for the odour of P. flammans mucus. Therefore, P. flammans mucus may inhibit pathogenic microorganisms and attract or repel predators, but the mucus does not act as a complete deterrent. [ABSTRACT FROM AUTHOR]

This study demonstrates that root-associated Kosakonia oryziphila NP19, isolated from rice roots, is a promising plant growth-promoting bioagent and biopesticide for combating rice blast caused by Pyricularia oryzae. In vitro experiments were conducted on fresh leaves of Khao Dawk Mali 105 (KDML105) jasmine rice seedlings. The results showed that NP19 effectively inhibited the germination of P. oryzae fungal conidia. Fungal infection was suppressed across three different treatment conditions: rice colonized with NP19 and inoculated by fungal conidia, a mix of NP19 and fungal conidia concurrently inoculated on the leaves, and fungal conidia inoculation first followed by NP19 inoculation after 30 h. Additionally, NP19 reduced fungal mycelial growth by 9.9–53.4%. In pot experiments, NP19 enhanced the activities of peroxidase (POD) and superoxide dismutase (SOD) by 6.1–63.0% and 3.0–67.7%, respectively, indicating a boost in the plant's defense mechanisms. Compared to the uncolonized control, the NP19-colonized rice had 0.3–24.7% more pigment contents, 4.1% more filled grains per panicle, 26.3% greater filled grain yield, 34.4% higher harvest index, and 10.1% more content of the aroma compound 2-acetyl-1-pyrroline (2AP); for rice colonized with NP19 and infected with P. oryzae, these increases were 0.2–49.2%, 4.6%, 9.1%, 54.4%, and 7.5%, respectively. In field experiments, blast-infected rice that was colonized and/or inoculated with NP19 treatments had 15.1–27.2% more filled grains per panicle, 103.6–119.8% greater filled grain yield, and 18.0–35.8% higher 2AP content. A higher SOD activity (6.9–29.5%) was also observed in the above-mentioned rice than in the blast-infected rice that was not colonized and inoculated with NP19. Following blast infection, NP19 applied to leaves decreased blast lesion progression. The mitochondrial distribution and morphology family 33 gene (MDM33) regulates mitochondrial homeostasis by mediating the mitochondrial fission process in yeast. The wheat head blight Fusarium graminearum contains an FgMdm33 protein that is orthologous to Saccharomyces cerevisiae Mdm33, albeit its function remains unknown. We have reported here the roles of FgMdm33 in regulating fungal morphogenesis, mitochondrial morphology, autophagy, apoptosis, and fungal pathogenicity. The ΔFgmdm33 mutants generated through a homologous recombination strategy in this study exhibited defects in terms of mycelial growth, conidia production, and virulence. Hyphal cells lacking FgMDM33 displayed elongated mitochondria and a dispensable respiratory-deficient growth phenotype, indicating the possible involvement of FgMDM33 in mitochondrial fission. The ΔFgmdm33 mutants displayed a remarkable reduction in the proteolysis of GFP-FgAtg8, whereas the formation of autophagic bodies in the hyphal cells of mutants was recorded under the induction of mitophagy. In addition, the transcriptional expression of the apoptosis-inducing factor 1 gene (FgAIF1) was significantly upregulated in the ΔFgmdm33 mutants. Cumulatively, these results indicate that FgMDM33 is involved in mitochondrial fission, non-selective macroautophagy, and apoptosis and that it regulates fungal growth, conidiation, and pathogenicity of the head blight pathogen. Candida auris, an emerging non-albicans multidrug-resistant yeast, has become a significant cause of invasive candidiasis in healthcare settings. So far, data on the metabolites of C. auris in different clades are minimal, and no studies have focused on clade V metabolites. Therefore, Gas chromatography–mass spectrometry (GC-MS) was used for the metabolomic profiling of clade I C. auris compared with fluconazole-resistant and susceptible C. auris in clade V strains. GC-MS chromatography revealed 28, 22, and 30 compounds in methanolic extracts of the fluconazole-susceptible and fluconazole-resistant C. auris clade V and C. auris clade I strain, respectively. Some compounds, such as acetamide and metaraminol, were found in fluconazole-susceptible and resistant C. auris clade V and clade I. N-methyl-ethanamine and bis(2-ethylhexyl) phthalate metabolites were found in both fluconazole -susceptible and resistant C. auris clade V, as well as 3-methyl-4-isopropylphenol, 3,5-bis(1,1-dimethyl)-1,2-benzenediol, and diisostyl phthalate metabolites in both fluconazole resistant C. auris clade V and I. Identifying these metabolites contributes to understanding the morphogenesis and pathogenesis of C. auris, highlighting their potential role in antifungal drug resistance and the control of fungal growth. However, further experiments are warranted to fully comprehend the identified metabolites' regulatory responses, and there may be potential challenges in translating these findings into clinical applications. The study was conducted to learn if the otoscope, a noncritical medical device, used by ENT residents harbor pathogenic organism. This study was conducted in a medical teaching hospital in India. Total of 38 otoscopes of ENT residents were examined after taking samples using sterile cotton swabs from otoscope speculum and otoscope head and both bacterial and fungal culture were studied. The study showed that 11 otoscope heads (28.94%) and 11 otoscope speculums (28.94%) out of the 38 otoscopes studied showed bacterial growth. Most commonly isolated bacteria were skin commensals followed by Klebsiella species. Fungal growth was seen in 3 out of 38 otoscope Speculums (7.89%). All 3 fungi isolated during the study belonged to Aspergillus species. Training residents regarding hygiene of medical equipments is necessary. The climate-change-coupled fungal burden in crop management and the need to reduce chemical pesticide usage highlight the importance of finding sustainable ways to control Aspergillus flavus. This study examines the effectiveness of 50 Pseudomonas isolates obtained from corn rhizospheres against A. flavus in both solid and liquid co-cultures. The presence and quantity of aflatoxin B1 (AFB1) and AFB1-related compounds were determined using high-performance liquid chromatography–high resolution mass spectrometry analysis. Various enzymatic- or non-enzymatic mechanisms are proposed to interpret the decrease in AFB1 production, accompanied by the accumulation of biosynthetic intermediates (11-hydroxy-O-methylsterigmatocystin, aspertoxin, 11-hydroxyaspertoxin) or degradation products (the compounds C16H10O6, C16H14O5, C18H16O7, and C19H16O8). Our finding implies the upregulation or enhanced activity of fungal oxidoreductases and laccases in response to bacterial bioactive compound(s). Furthermore, non-enzymatic reactions resulted in the formation of additional degradation products due to acid accumulation in the fermented broth. Three isolates completely inhibited AFB1 or any AFB1-related compounds without significantly affecting fungal growth. These bacterial isolates supposedly block the entire pathway for AFB1 production in the fungus during interaction. Myosins constitute a class of functional proteins that are widely found in eukaryotic organisms and play important roles in the process of various life activities in the cells of living organisms. In addition, myosins derived from phytopathogenic fungi significantly also have significant impacts on the growth and development of fungi and their pathogenicity. In light of these characteristics, more and more researchers are directing their attention towards the development of pesticides targeting myosin. Designing inhibitors based on myosins from phytopathogenic fungi to develop fungicides with novel mechanisms of action will be an important tool for preventing and controlling plant diseases, thereby ensuring food security. This paper focuses on the research progress of myosins and their inhibitors in phytopathogenic fungal and mainly reviews the structural features and biochemical functions, as well as myosin inhibitor fungicides reported in recent years, which can provide a reference for the discovery of myosin target-directed lead compounds of phytopathogenic fungi and the rational design of the active molecules of new pesticides. The quality of pistachio, one of the export products of Iran, will be decreased during storage as a result of mold spoilage, toxins production, and oil oxidation. This study aimed to investigate the capability of pistachio hull extract (PHE) loaded in fenugreek seed gum (FSG):whey protein isolate (WPI) nanoemulsion to control oil oxidation, and fungi growth in fresh pistachio nut during storage at 4°C. The total anthocyanin and total phenolic content of the PHE were 125.44 μg/g and 675.18 mg/g, respectively. The DPPH radical scavenging activity of PHE at 100 ppm was higher than that of tert‐butylhydroquinon (TBHQ). In comparison with other concentrations, 50 ppm showed the strongest antifungal activity against Aspergillus flavus, Aspergillus parasiticus, and Aspergillus nomius. All nanoemulsions have a mean size lower than 265 nm. The polydispersity index (PDI) of different nanoemulsions was lower than 0.3, and a negative zeta potential was observed. The encapsulation efficiency was higher than 67.0% and all nanoemulsions had spherical morphology. The pistachio nuts were coated with different coating solutions containing 0 and 100 ppm of PHE and stored at 4°C for 8 weeks. The results showed that the pistachio sample coated with a composite coating of WPI and FSG containing 100 ppm of PHE has a higher moisture content and lower changes in L*, a*, and b* indexes, oil oxidation, fungi development, and total mold and yeast count. This treatment exhibited higher overall acceptance than other samples at the end of storage time. Alternaria alternata is a prevalent postharvest pathogen that generates diverse mycotoxins, notably alternariol (AOH) and alternariol monomethyl ether (AME), which are recurrent severe contaminants. Nitrogen sources modulate fungal growth, development, and secondary metabolism, including mycotoxin production. The GATA transcription factor AreA regulates nitrogen source utilization. However, little is known about its involvement in the regulation of nitrogen utilization in A. alternata. To examine the regulatory mechanism of AaAreA on AOH and AME biosynthesis in A. alternata, we analyzed the impact of diverse nitrogen sources on the fungal growth, conidiation and mycotoxin production. The use of a secondary nitrogen source (NaNO3) enhanced mycelial elongation and sporulation more than the use of a primary source (NH4Cl). NaNO3 favored greater mycotoxin accumulation than did NH4Cl. The regulatory roles of AaAreA were further clarified through gene knockout. The absence of AaAreA led to an overall reduction in growth in minimal media containing any nitrogen source except NH4Cl. AaAreA positively regulates mycotoxin biosynthesis when both NH4Cl and NaNO3 are used as nitrogen sources. Subcellular localization analysis revealed abundant nuclear transport when NaNO3 was the sole nitrogen source. The regulatory pathway of AaAreA was systematically revealed through comprehensive transcriptomic analyses. The deletion of AaAreA significantly impedes the transcription of mycotoxin biosynthetic genes, including aohR, pksI and omtI. The interaction between AaAreA and aohR, a pathway-specific transcription factor gene, demonstrated that AaAreA binds to the aohR promoter sequence (5'-GGCTATGGAAA-3'), activating its transcription. The expressed AohR regulates the expression of downstream synthase genes in the cluster, ultimately impacting mycotoxin production. This study provides valuable information to further understand how AreA regulates AOH and AME biosynthesis in A. alternata, thereby enabling the effective design of control measures for mycotoxin contamination. Solid-state fermentation (SSF) is the bioprocess where microorganisms are cultivated in the absence of free water under controlled conditions. Lactic acid can be produced by Rhizopus oryzae SSF of grape stalks. During the microorganism's growth, the temperature and water content of the solid bed fluctuate, leading to areas of either dry or excessive moisture in the solid substrate. Therefore, it is crucial to control the water supply to the matrix. In this work, we obtain lactic acid through SSF of grape stalks using Rhizopus oryzae NCIM 1299. The SSF was conducted at a fixed temperature of 35 °C, with five constant relative humidity (RH) levels: 50, 57, 65, 72, and 80%RH. Mathematical models, including the Logistic and First-Order Plus Dead-Time models for fungal biomass growth and the Luedeking and Piret with Delay Time model for lactic acid production, were adjusted to kinetic curves. Growth kinetic parameters (Xmax, μmax, Tp, T0, Yp/x, and td) were determined for all conditions. These kinetic parameters were then correlated with relative humidity using a second-degree polynomial relationship. We observed a decrease in Xmax with an increasing %RH, while the value of Yp/x increased at a higher %RH. Finally, the optimal variable relative humidity profile was obtained by applying the dynamic optimization technique, resulting in a 16.63% increase in lactic acid production. [ABSTRACT FROM AUTHOR]

Radionuclides are essential for chemical and biological innovations, but their presence as toxic pollutant particles poses a significant threat to human health. To state this issue, bioremediation methodologies utilizing microorganisms have been implemented cost-effectively for removing elevated levels of heavy metals and radionuclides from the environment. The study's goal was to look into how bacteria (Bacillus cereus and Escherichia coli) and halophilic fungi (Aspergillus penicillioides, Engyodontium album, Penicillium imranianum, and Sterigmatomyces halophilus) tolerate radionuclides (Cesium, Strontium, and Uranium). The radionuclides inhibited bacteria and suppressed fungal growth. Interestingly, combinations of radionuclides with fungi were found to be more lethal against bacteria. Fungi demonstrated varying tolerance to radionuclides and exhibited efficient biosorption of radionuclides. Among the halophilic fungi, P. imranianum has shown the best performance in terms of tolerance against both bacteria and radionuclides. At the same time, S. halophilus exhibited the least ability to handle the stress conditions imposed by radionuclides. This study highlights the considerable potential for biosorption by halophilic fungi under extreme stress conditions, delivering a rate effective ecological solution for the elimination of toxic radionuclides. Signaling pathways in fungi offer a profound avenue for harnessing cellular communication and have garnered considerable interest in biomaterial engineering. Fungi respond to environmental stimuli through intricate signaling networks involving biochemical and electrical pathways, yet deciphering these mechanisms remains a challenge. In this review, an overview of fungal biology and their signaling pathways is provided, which can be activated in response to external stimuli and direct fungal growth and orientation. By examining the hyphal structure and the pathways involved in fungal signaling, the current state of recording fungal electrophysiological signals as well as the landscape of fungal biomaterials is explored. Innovative applications are highlighted, from sustainable materials to biomonitoring systems, and an outlook on the future of harnessing fungi signaling in living composites is provided. The study was conducted in order to explore the potential of fungi isolated from surface and bottom seawater collected from the fishing harbour of Bizerte on the bioremediation of industrial effluent (IE) contaminated by petroleum hydrocarbon. Among the 128 fungal isolates, 11 were isolated from surface seawater and 7 from bottom seawater, representing 18 taxa in total. The gas chromatography mass spectrometry (GC–MS) was used for the determination of hydrocarbon compounds in IE. An initial screening of fungal growth using six concentrations ranged between 20 and 70% (v/v) IE has allowed the identification of the optimal concentration for fungal growth as well as selection of species able to tolerate high amounts of hydrocarbon. Colorimetric test employing 2,6-dichlorophenol indophenol and gravimetric method was applied for the assessment of fungal growth using 20% EI. By checking the phylogenetic affiliation of the high-performing stains as identified using ITSr DNA sequence, a dominance of Ascomycetes was detected. Indeed, Aspergillus terreus and Penicillium expansum may degrade 82.07 and 81.76% of residual total petroleum hydrocarbon (TPH), respectively. Both species were collected from surface seawater. While, Aspergillus niger, Colletotrichum sp and Fusarium annulatum displayed comparable degradation rates 40.43%, 41.3%, and 42.03%, respectively. The lowest rate of degradation 33.62% was detected in Emericellopsis phycophila. All those species were isolated from bottom seawater, excepting A. niger isolated from surface water. This work highlighted the importance of exploring the potential of fungi isolated from the natural environment on the bioremediation of industrial effluent. In wheat and barley, Fusarium head blight is mainly caused by Fusarium graminearum, and its control is based on the agricultural practices of not leaving crop residues in the field, growing phytopathogenic fungi-resistant varieties, biological control, and chemical treatment, including using fungicides. Here, we investigated the antifungal and antimycotoxigenic activities of Massoia essential oil (MEO) and C10 Massoia lactone (C10) on Fusarium graminearum KACC 41047. Because DMSO, which was used as a solvent in this study, exhibited antifungal activity at 5% in a fungal growth medium, it was used in the antifungal and antimycotoxigenic experiments at 0.05%. Three assays were used to investigate the antifungal activities of MEO and C10, which exhibited potent antifungal activity in the agar dilution assay, with complete fungal growth inhibition at 100 mg/L. At 5–50 mg/L, MEO and C10 suppressed deoxynivalenol and 15-acetyl-deoxynivalenol production by >50% by downregulating the Tri10 gene, which expresses trichodiene synthase. Inflammatory bowel disease (IBD), which includes Crohn's disease and ulcerative colitis, involves chronic inflammatory disorders of the digestive tract. Oxidative stress, associated with increased reactive oxygen species generation, is a major risk factor for IBD pathogenesis. Industrialized lifestyles expose us to a variety of factors that contribute to deteriorating gut health, especially for IBD patients. Many alternative therapeutic strategies have been developed against oxidative stress along with conventional therapy to alleviate IBD pathogenesis. Polyphenol-rich foods have attracted growing interest from scientists due to their antioxidant properties. Polyphenols are natural compounds found in plants, fruits, vegetables, and nuts that exhibit antioxidant properties and protect the body from oxidative damage. This review presents an overview of polyphenol benefits and describes the different types of polyphenols. It also discusses polyphenols' role in inhibiting oxidative stress and fungal growth prevention. Overall, this review highlights how a healthy and balanced diet and avoiding the industrialized lifestyles of our modern society can minimize oxidative stress damage and protect against pathogen infections. Reactive carbonyl and oxygen species (RCS/ROS), often generated as metabolic byproducts, particularly under conditions of pathology, can cause direct damage to proteins, lipids, and nucleic acids. Glyoxal oxidases (Gloxs) oxidize aldehydes to carboxylic acids, generating hydrogen peroxide (H2O2). Although best characterized for their roles in lignin degradation, Glox in plant fungal pathogens are known to contribute to virulence, however, the mechanism underlying such effects are unclear. Here, we show that Glox in the insect pathogenic fungus, Metarhizium acridum, is highly expressed in mycelia and during formation of infection structures (appressoria), with the enzyme localizing to the cell membrane. MaGlox targeted gene disruption mutants showed RCS and ROS accumulation, resulting in cell toxicity, induction of apoptosis and increased autophagy, inhibiting normal fungal growth and development. The ability of the MaGlox mutant to scavenge RCS was significantly reduced, and the mutant exhibited increased susceptibility to aldehydes, oxidative and cell wall perturbing agents but not toward osmotic stress, with altered cell wall contents. The ΔMaGlox mutant was impaired in its ability to penetrate the host cuticle and evade host immune defense resulting in attenuated pathogenicity. Overexpression of MaGlox promoted fungal growth and conidial germination, increased tolerance to H2O2, but had little to other phenotypic effects. Transcriptomic analyses revealed downregulation of genes related to cell wall synthesis, conidiation, stress tolerance, and host cuticle penetration in the ΔMaGlox mutant. These findings demonstrate that MaGlox-mediated scavenging of RCS is required for virulence, and contributes to normal fungal growth and development, stress resistance. Objective Materials and Methods Results Conclusions To evaluate the efficacy of recombinant psoriasin as a novel treatment for oral candidiasis by eliminating Candida albicans growth on polymethyl methacrylate denture base.Recombinant psoriasin protein was expressed and purified from E. coli, and Candida growth was monitored in vitro with varying concentrations of psoriasin. Subsequently, denture‐base polymethyl methacrylate was immersed in psoriasin's solution or voriconazole, and fungal growth on the acrylic base and in the medium was examined by scanning electron microscopy and optical density, respectively. Cellular viability of HeLa and human gingival fibroblast cells treated with psoriasin was measured by methylene blue assay.The findings reveal an effective antifungal activity of psoriasin, completely inhibiting Candida albicans growth in RPMI at a protein concentration above 400 nM. Immersing the polymethyl methacrylate with 50 μM psoriasin completely eradicates fungal growth. Psoriasin has low cytotoxicity in HeLa cells at a concentration higher than 12 μM and no toxic effect on human gingival fibroblasts.This study marks psoriasin as an effective alternative to conventional antifungal treatments for denture stomatitis and a safe alternative to chemical antifungals in dental medicine and beyond. Using microorganisms as biocontrol agents against soilborne plant pathogens is a promising alternative to chemical pesticides. However, only some biocontrol agents have proven effective under field conditions. This study explores the potential of highly resilient microalgae isolated from harsh environments, such as Biological Soil Crusts and agricultural fields in semi-arid regions, as a novel and sustainable approach to biocontrol. Fifty-nine microalgal strains, including thirteen cyanobacteria and forty-six green algae, were isolated and identified. Dual-culture plate assays and toxicity tests of microalgal growth media were conducted to evaluate the antifungal activity of the isolates against eight representative soilborne pathogens. The results showed that many microalgae strains exhibited significant inhibitory effects on the growth of specific fungal pathogens, although the activity varied among different microalgal strains and pathogen species. Some strains even promoted the growth of certain fungi. The lack of a clear pattern in the antifungal activity highlights the complexity and specificity of the interactions between microalgae and soilborne pathogens. An "Inhibition Effectiveness" metric was developed to quantify biocontrol potential based on fungal growth inhibition. The green algal genus Desmodesmus, particularly Desmodesmus subspicatus isolates, showed higher antifungal efficacy than other genera. While the inhibitory mechanisms remain unclear, the results demonstrate the promising biocontrol capabilities of microalgae from extreme environments like BSCs. Further research could unlock novel opportunities for sustainable disease management by harnessing specific microalgal strains or synergistic strain combinations targeting soilborne pathogens. Azole antifungals inhibit the sterol C14-demethylase (CYP51/Erg11) of the ergosterol biosynthesis pathway. Here we show that the azole-induced synthesis of fungicidal cell wall carbohydrate patches in the pathogenic mold Aspergillus fumigatus strictly correlates with the accumulation of the CYP51 substrate eburicol. A lack of other essential ergosterol biosynthesis enzymes, such as sterol C24-methyltransferase (Erg6A), squalene synthase (Erg9) or squalene epoxidase (Erg1) does not trigger comparable cell wall alterations. Partial repression of Erg6A, which converts lanosterol into eburicol, increases azole resistance. The sterol C5-desaturase (ERG3)-dependent conversion of eburicol into 14-methylergosta-8,24(28)-dien-3β,6α-diol, the "toxic diol" responsible for the fungistatic activity against yeasts, is not required for the fungicidal effects in A. fumigatus. While ERG3-lacking yeasts are azole resistant, ERG3-lacking A. fumigatus becomes more susceptible. Mutants lacking mitochondrial complex III functionality, which are much less effectively killed, but strongly inhibited in growth by azoles, convert eburicol more efficiently into the supposedly "toxic diol". We propose that the mode of action of azoles against A. fumigatus relies on accumulation of eburicol which exerts fungicidal effects by triggering cell wall carbohydrate patch formation. Azole antifungals inhibit the ergosterol biosynthesis enzyme CYP51, but their effects on fungal viability and growth vary greatly among fungal species. Fusarium fujikuroi, the causal agent of rice bakanae disease (RBD), contains five succinate dehydrogenase (Sdh) subunits: FfSdhA, FfSdhB, FfSdhC1, FfSdhC2, and FfSdhD. However, the role of these subunits in regulating sensitivity to succinate dehydrogenase inhibitors (SDHIs) is largely unknown. Here, we conducted targeted gene disruption and phenotypic assays for all Sdh subunits and found that the deletion mutants of FfSdhA, FfSdhB, and FfSdhD exhibited severe defects in hyphal growth, conidiation, virulence, and sensitivity to CaCl2 and oxidative stresses. To a lesser extent, the mycelial growth rate and conidial production of ΔFfSdhC1 were also decreased as compared to those of the wild-type strain JS16. In addition, fungicide sensitivity assays showed that deletion of FfSdhA, B, C1, or D led to decreased sensitivity to all SDHIs tested. Unexpectedly, we were unable to obtain a FfSdhC1 + C2 double mutant and further found significant up-regulation of FfSdhC2 in ∆FfSdhC1, indicating that FfSdhC1 and -C2 might be essential for fungal growth although the FfSdhC2 deletion mutant was indistinguishable from the wild-type strain. Plant defense responses to the soil-borne fungus Verticillium longisporum causing stem stripe disease on oilseed rape (Brassica napus) are poorly understood. In this study, a population of recombinant inbred lines (RILs) using the Arabidopsis accessions Sei-0 and Can-0 was established. Composite interval mapping, transcriptome data, and T-DNA mutant screening identified the NITRATE/PEPTIDE TRANSPORTER FAMILY 5.12 (AtNPF5.12) gene as being associated with disease susceptibility in Can-0. Co-immunoprecipitation revealed interaction between AtNPF5.12 and the MAJOR LATEX PROTEIN family member AtMLP6, and fluorescence microscopy confirmed this interaction in the plasma membrane and endoplasmic reticulum. CRISPR/Cas9 technology was applied to mutate the NPF5.12 and MLP6 genes in B. napus. Elevated fungal growth in the npf5.12 mlp6 double mutant of both oilseed rape and Arabidopsis demonstrated the importance of these genes in defense against V. longisporum. Colonization of this fungus depends also on available nitrates in the host root. Accordingly, the negative effect of nitrate depletion on fungal growth was less pronounced in Atnpf5.12 plants with impaired nitrate transport. In addition, suberin staining revealed involvement of the NPF5.12 and MLP6 genes in suberin barrier formation. Objective- The microscope investigation of fungi is rapid but these diagnostic procedures show lack of sensitivity and specificity with 15% fallacious results and species or genus level identification is not feasible. Material and Methods- This 18-month prospective study included all dermatophytes that were isolated during that time. Dermatophytes were discovered using standard phenotypic techniques. Therefore, in recent years many molecular methods have been developed which are more specific, precise, rapid and cost effective than conventional phenotypic methods. Samples of skin and nails were analyzed under a microscope using 20% KOH and cultivated on Sabouraud dextrose agar supplemented with DTM Media, cycloheximide, and chloramphenicol. By sequencing the internal transcribed spacer (ITS) r-DNA region, the identity of the fungal isolates was verified. Results- Out Of the 320 samples 270 (84.37%) samples showed growth of fungi (dermatophytes) after culturing on SDA media. Whereas with AP-PCR, 303 (94.68%) samples were positive for dermatophytes. Seven species of dermatophyte namely Trichophyton mentagrophytes, Trichophyton rubrum, Epidermophyton floccosum, Trichophyton tonsurans, Microsporum gypseum, Trichophyton verrucosum and Microsporum canis identified after culturing on SDA media from these 270 samples were confirmed by AP-PCR also. The cheese ripening period, in which biochemical changes occur under specific temperature and humidity conditions, is crucial for making the finished product marketable. However, the food matrix can become a growth medium for developing spoilage microorganisms and fungi. In dairy factory, contamination of the crust area can be monitored with several wasteful and time-expensive methods, i.e., the use of saline solutions and/or the brushing of cheese wheels. This study investigates the effectiveness of different ozone treatments, 400 ppb, 8 hours (overnight) every other day until the end of ripening, and 300 ppb, 8 hours per day (overnight) for the entire ripening duration compared to control sample in normal atmosphere, in controlling microbial and fungal growth during the ripening of Toma Piemontese PDO cheese. Results showed that ozone treatments significantly affect spoilage microflora, preserving the overall quality parameters (i.e., weight loss, customer value, and greater sensory nuances). In the present study, we identified and characterized two defensin‐like peptides in an antifungal fraction obtained from Capsicum chinense pepper fruits and inhibited the growth of Colletotrichum scovillei, which causes anthracnose. AMPs were extracted from the pericarp of C. chinense peppers and subjected to ion exchange, molecular exclusion, and reversed‐phase in a high‐performance liquid chromatography system. We investigated the endogenous increase in reactive oxygen species (ROS), the loss of mitochondrial functioning, and the ultrastructure of hyphae. The peptides obtained from the G3 fraction through molecular exclusion chromatography were subsequently fractionated using reverse‐phase chromatography, resulting in the isolation of fractions F1, F2, F3, F4, and F5. The F1‐Fraction suppressed C. scovillei growth by 90, 70.4, and 44% at 100, 50, and 25 μg mL−1, respectively. At 24 h, the IC50 and minimum inhibitory concentration were 21.5 μg mL−1 and 200 μg mL−1, respectively. We found an increase in ROS, which may have resulted in an oxidative burst, loss of mitochondrial functioning, and cytoplasm retraction, as well as an increase in autophagic vacuoles. MS/MS analysis of the F1‐Fraction indicated the presence of two defensin‐like proteins, and we were able to identify the expression of three defensin sequences in our C. chinense fruit extract. The F1‐Fraction was also found to inhibit the activity of insect α‐amylases. In summary, the F1‐Fraction of C. chinense exhibits antifungal activity against a major pepper pathogen that causes anthracnose. BACKGROUND: Potatoes, a major economic crop, are significantly impacted by Fusarium dry rot, a prevalent postharvest disease. Despite the broad‐spectrum antimicrobial properties of cinnamaldehyde, a naturally‐derived plant substance, its efficacy against the causal pathogen of potato dry rot (Fusarium oxysporum) and the underlying mechanisms have not been extensively studied. RESULTS: Our study demonstrates that cinnamaldehyde effectively inhibits the growth of Fusarium oxysporum, the pathogen responsible for potato dry rot, and increases its sensitivity to environmental stress factors such as extreme temperatures and high salt stress. Treatment with cinnamaldehyde results in altered fungal mycelium morphology, compromised cell wall stability, and disrupted cell membrane integrity, thereby reducing spore viability. Specifically, it interferes with the cell membrane and cell wall structures of the fungus, potentially disrupting fungal growth by modulating signaling pathways involved in cell wall maintenance, chitin metabolism, and GPI‐anchored protein function. Notably, we show that cinnamaldehyde induces a form of regulated cell death in F. oxysporum, which is characterized not as typical apoptosis, as evidenced by Annexin V negative staining. However, the specific cell death type and underlying mechanism still needed to be further explored. CONCLUSION: Cinnamaldehyde, an environmentally friendly plant‐based active compound, exhibits strong inhibitory effects on F. oxysporum, indicating its potential use in the prevention and control strategies for potato dry rot. The objective of the current study was to evaluate the feasibility of Aloe vera gel as a plasticizer and crosslinker in improving the properties of starch–polyvinyl alcohol blends that could find applications in packaging. The concentration of Aloe vera gel was varied (1%, 3%, 5% and 7% wt/wt) to produce SPA‐1, SPA‐3, SPA‐5 and SPA‐7 films, respectively. The plasticizing and crosslinking characteristics associated with Aloe vera gel had a positive influence on the mechanical properties of the films. Addition of Aloe vera gel increased the tensile strength of films from 27.45 MPa (control) to 32.98, 32.53 and 32.32, for SPA‐1, SPA‐3 and SPA‐5 films, respectively. Among all the films, highest elongation at break (20.62%) was observed for SPA‐3 films. Due to crosslinking, degree of swelling, water solubility and water vapour permeability for SPA‐3 films decreased by 5.58%, 38.29% and 21.44%, respectively, compared to control films. The contact angle of the SPA‐3 film significantly increased by 49.25% when compared to control samples. Scanning electron microscope images revealed compact and smooth surface microstructure of control films, and crosslinking was evident in presence of Aloe vera gel. The rate of degradation for SPA‐3 films in soil after 40 days was enhanced by 32.25% compared to control films. SPA‐1 and SPA‐3 films were tested for use as packaging material for storage of green chillies. Chillies under unpacked conditions, in control and SPA‐1 films, turned red in 3 days. Those stored in films with 3% Aloe vera gel began to change colour later (after 5 days) with no visual evidence of microbial or fungal growth. Sophora davidii is a multipurpose, nitrogen-fixing shrub species. Phosphorus deficiency in the acidic soil of Southwest China has seriously affected its survival and growth, especially in the seedling stage. Evidence suggests that arbuscular mycorrhizal fungi (AMF) may improve the stress tolerance of plants. However, there is limited information on the systematic effects of AMF on phosphorus deficiency in S. davidii seedlings. We investigated the effects of three phosphorus levels (0.5, 0.25, 0 mmol/L) and two mycorrhizal inoculation (with Funneliformis mosseae and without Funneliformis mosseae) treatments on the growth and physiological performance of S. davidii using factorial design. The results showed that low-phosphorus stress significantly limited the growth of S. davidii seedlings and negatively affected their physiological properties. However, inoculation with F. mosseae significantly improved the plant height and shoot dry weight, promoted root growth, increased chlorophyll contents and osmoregulation substance contents, increased protective enzyme activity, and significantly reducing the accumulation of malondialdehyde, alleviated oxidative stress induced by low-phosphorus stress, improved the IAA and GA3 contents, and alleviated the negative effects of low-phosphorus stress. AMF-induced enhancement of aboveground growth and plant physiological characteristics is dependent on the P level and its impact on roots is regardless of phosphorus status. AMF inoculation significantly promoted the absorption of nitrogen and phosphorus in the roots (0.25- and 0-mmol/L treatments), thereby maintaining a higher biomass and relieving stress in S. davidii seedlings under low-phosphorus conditions. Our results demonstrated that AMF inoculation is useful for the promotion and cultivation of S. davidii in the karst area of Southwest China under low-phosphorus stress conditions. Fermented sprouted seeds are promising substrates for designing preservative-free, fortified baked goods. In the present study, predominant lactic acid bacteria (LAB) isolated from fermented sprouted clover seeds (FSC) were screened based on their potential ability to control resistant fungi towards potassium sorbate (PS). Then, controlled FSC (CFSC) containing the selected LAB isolate was used as an alternative for PS to produce clean-label enriched wheat bread. Phylogenetic evolutionary analysis led to the identification of Lacticaseibacillus rhamnosus as the selected LAB isolate with potent inhibitory effect on Aspergillus brasiliensis as the targeted PS resistant fungus. Furthermore, there was no significant difference (P > 0.05) between in situ inhibitory effects of the CFSC and PS on surface growth of the target fungus. The positive effects of sprouting/fermentation on the amino acid contents were also verified according to the HPLC-based analysis. Enriched bread with CFSC and spontaneous FSC had the highest amounts of arginine and lysine, respectively. Furthermore, sourdough fermentation led to the improvement of the crumb microstructure, increasing of antioxidant activity and reduction of phytate content compared to the control. Pleurotus ostreatus is one of the most consumed mushroom species, as it serves as a high-quality food, favors a rich secondary metabolism, and has remarkable adaptability to the environment and predators. In this study, we investigated the function of two key reactive oxygen species producing enzyme NADPH oxidase (PoNoxA and PoNoxB) in P. ostreatus hyphae growth, metabolite production, signaling pathway activation, and immune responses to different stresses. Characterization of the Nox mutants showed that PoNoxB played an important role in the hyphal formation of the multicellular structure, while PoNoxA regulated apical dominance. The ability of P. ostreatus to tolerate a series of abiotic stress conditions (e.g., osmotic, oxidative, membrane, and cell-wall stresses) and mechanical damage repair was enhanced with PoNoxA over-expression. PoNoxB had a greater responsibility in regulating the polysaccharide composition of the cell wall and methyl jasmonate and gibberellin GA1 biosynthesis, and improved mushroom resistance against Tyrophagus putrescentiae. Moreover, mutants were involved in the jasmonate and GA signaling pathway, and toxic protein defense metabolite production. Sclerotinia sclerotiorum causes white mold disease, resulting in losses of up to 50% in several host plants worldwide. It is a harmful fungus as it infects plants at any stage of development, in the field, or in storage; this includes young seedlings, mature plants, and fruits. The accessibility of molecular and biological methods has contributed significantly to our understanding of S. sclerotiorum etiology and progression. The present research aimed to characterize S. sclerotiorum causing white mold disease in eggplant (S. melobgena), evaluate the potentiality of some bio-agents for inhibiting the fungus growth and develop a strategy for dealing with the disease challenge. Eggplants grown in open fields and greenhouses showed cottony growth on their stems, followed by the emergence of sclerotia. A fungal growth with fluffy mycelium and considerable sclerotia was isolated from the infected tissues. Polyphasic characterization and phytopathological analysis identified the fungus as S. sclerotiorum. The symptoms previously seen in the field were reproduced by inoculating healthy eggplant roots, stems, and leaves with the fungus. The ITS rDNA sequencing (564 bp) homology and phylogenetic analysis showed 99% sequence similarity of the isolate with multiple S. sclerotiorum. A wide range of strategies are required for disease management. S. sclerotiorum is sensitive to competition from other bacteria. Such a technique might act as an initial basis for effective biological control of white mold. The results demonstrated that eight isolates have very strong antagonistic activity against S. sclerotiorum. Reduced rates ranged from 63.1% (65 D) to 39.0.1% (75 A1). At the same time, the effect of the Trichoderma harzianum fungus against the pathogen S. sclerotium was apparent in restricting mycelium growth and inhibiting the fungus growth. ITS sequencing identified the mycopathogen. The interaction of seven Bacillus spp isolates with S. sclerotiorum on PDA culture media highlighted these isolates' capacity to limit the pathogen's mycelial growth. Biodegradable packaging, both alone and in combination with acid whey protein coatings, has been used to pack fresh Gouda cheese to improve preservation prior ripening or storage. This study involved three key components: (i) the selection of biodegradable packaging (BP), (ii) the development of a plain liquid acid whey protein concentrate, pectin-based edible coating (BP + Ch + Coating), and (iii) the incorporation of at least 6 log10 CFU (colony forming units) mL−1 Lacticaseibacillus paracasei (BP + Ch + Coating + Lp) and Lactobacillus helveticus (BP + Ch + Coating + Lh) strains. The created compositions were compared with cheese packed in conventional polyethylene (PE) packaging to evaluate their overall synergy effect in reducing microbiological spoilage and influencing chemical parameters in Gouda cheese during 45 days of ripening and cold storage. The evaluation included microbiological analysis (total LAB, Enterobacteriaceae spp., and fungi CFU) and quality assessment of pH, moisture content, water activity, texture, and colour (CEI system) during ripening and shelf life. Although biodegradable packaging (BP) alone did not protect the cheese effectively compared to conventional packaging (EVA/PE/EPC/PVDC), the combination of biodegradable packaging with a coating (BP + Ch + Coating) showed protective properties against Enterobacteriaceae spp. and mould, maintaining moisture, pH, and colour during ripening and storage. The biocontrol potential of three native soil cyanobacteria from biological soil crusts (Nostoc commune, Scytonema hyalinum, and Tolypothrix distorta) was tested by means of in vitro mycelial growth inhibition assays for eighteen cyanobacteria-based products against three phytopathogenic soilborne fungi (Phytophthora capsici, Pythium aphanidermatum, and Fusarium oxysporum f. sp. radicis-cucumerinum). Three cyanobacteria-based production factors were considered: (i) cyanobacterium strain, (ii) cyanobacterial culture growth phase, and (iii) different post-harvest treatments: raw cultures, cyanobacterial filtrates, and cyanobacterial extracts. Results showed that any of the factors considered are key points for successfully inhibiting fungal growth. N. commune showed the highest growth inhibition rates for the three phytopathogens; stationary phase treatments produced higher inhibition percentages than logarithmic ones; and all the post-harvest treatments of N. commune at the stationary phase inhibited the growth of P. capsici, up to 77.7%. Thus, N. commune products were tested in planta against P. capsici, but none of the products showed efficacy in delaying the onset nor reducing the damage due to P. capsici, demonstrating the complexity of the in planta assay's success and encouraging further research to design an appropriate scaling up methodology. The aim of the research was to investigate the resistance and efficacy of the fungi Aspergillus awamori in removing heavy metals. We conducted an analysis of the fungus's tolerance to several heavy metals by measuring the dry weight and using a plate method. The fungus exhibited growth in concentrations and lowest growth was observed in a concentration of 400 ppm of Cd. The heavy metals were successfully removed with the following orders: Zn> Co, > Ag, > Hg and Cu. The optimal parameters for the elimination of 100 mg/L of heavy metals were a temperature of 28°C, a pH range of 4.0 to 5.5, a heavy metal concentration of 100 ppm, and a biomass of fungi of 1 g. Invasive fungal infections are a major health threat with high morbidity and mortality, highlighting the urgent need for rapid diagnostic tools to detect antifungal resistance. Traditional culture-based antifungal susceptibility testing (AFST) methods often fall short due to their lengthy process. In our previous research, we developed a whole-slide imaging (WSI) technique for the high-throughput assessment of bacterial antibiotic resistance. Building on this foundation, this study expands the application of WSI by adapting it for rapid AFST through high-throughput monitoring of the growth of hundreds of individual fungi. Due to the distinct "budding" growth patterns of fungi, we developed a unique approach that utilizes specific cell number change to determine fungi replication, instead of cell area change used for bacteria in our previous study, to accurately determine the growth rates of individual fungal cells. This method not only accelerates the determination of antifungal resistance by directly observing individual fungal cell growth, but also yields accurate results. Employing Candida albicans as a representative model organism, reliable minimum inhibitory concentration (MIC) of fluconazole inhibiting 100% cells of Candida albicans (denoted as MIC100) was obtained within 3h using the developed method, while the modified broth dilution method required 72h for the similar reliable result. In addition, our approach was effectively utilized to test blood culture samples directly, eliminating the need to separate the fungi from whole blood samples spiked with Candida albicans. Trichothecenes are sesquiterpenoid toxins produced by diverse ascomycetes, including Fusarium. The trichothecene analog deoxynivalenol (DON) produced by the Fusarium head blight (FHB) pathogen Fusarium graminearum is a virulence factor on wheat and a major food and feed safety concern. In Fusarium, the trichothecene biosynthetic gene (TRI) cluster consists of 7–14 genes. Most TRI cluster genes are conserved and their specific roles in trichothecene biosynthesis have been determined. An exception is TRI14, which is not required for DON synthesis in vitro but is required for spread of F. graminearum in wheat heads. In the current study, gene expression analyses revealed that TRI14 was highly induced in infected wheat heads. We demonstrated that TRI14 was not only required for F. graminearum spread but also important for initial infection in wheat. Although a prior study did not detect DON in infected seeds, our analyses showed significantly less DON and fungal biomass in TRI14-mutant (designated ∆tri14)-inoculated heads than wild-type-inoculated heads. Gene expression comparison showed that the level of expression of TRI genes was similar in the wheat tissues infected with ∆tri14 or the wild type, indicating the reduced toxin levels caused by ∆tri14 may be due to less fungal growth. ∆tri14 also caused less lesion and grew less in wheat coleoptiles than the wild type. The growth of ∆tri14 in carboxymethylcellulose medium was more sensitive to hydrogen peroxide than the wild type. The data suggest that TRI14 plays a critical role in F. graminearum growth, and potentially protects the fungus from plant defense compounds.

This study investigated the influence of bacterial cyclic lipopeptides (LP; surfactins, iturins, fengycins) on microbial interactions. The objective was to investigate whether the presence of bacteria inhibits fungal growth and whether this inhibition is due to the release of bacterial metabolites, particularly LP. Selected endophytic bacterial strains with known plant-growth promoting potential were cultured in the presence of Fusarium oxysporum f.sp. strigae (Fos), which was applied as model fungal organism. The extracellular metabolome of tested bacteria, with a focus on LP, was characterized, and the inhibitory effect of bacterial LP on fungal growth was investigated. The results showed that Bacillus velezensis GB03 and FZB42, as well as B. subtilis BSn5 exhibited the strongest antagonism against Fos. Paraburkholderia phytofirmans PsJN, on the other hand, tended to have a slight, though non-significant growth promotion effect. Crude LP from strains GB03 and FZB42 had the strongest inhibitory effect on Fos, with a significant inhibition of spore germination and damage of the hyphal structure. Liquid chromatography tandem mass spectrometry revealed the production of several variants of iturin, fengycin, and surfactin LP families from strains GB03, FZB42, and BSn5, with varying intensity. Using plate cultures, bacillomycin D fractions were detected in higher abundance in strains GB03, FZB42, and BSn5 in the presence of Fos. Additionally, the presence of Fos in dual plate culture triggered an increase in bacillomycin D production from the Bacillus strains. The study demonstrated the potent antagonistic effect of certain Bacillus strains (i.e., GB03, FZB42, BSn5) on Fos development. Myeloblastosis (MYB)-like proteins are a family of highly conserved transcription factors in animals, plants, and fungi and are involved in the regulation of mRNA expression of genes. In this study, we identified and characterized one MYB-like protein in the model organism Aspergillus nidulans. We screened the mRNA levels of genes encoding MYB-like proteins containing two MYB repeats in conidia and found that the mRNA levels of four genes including flbD, cicD, and two uncharacterized genes, were high in conidia. To investigate the roles of two uncharacterized genes, AN4618 and AN10944, deletion mutants for each gene were generated. Our results revealed that AN4618 was required for fungal development. Therefore, we further investigated the role of AN4618, named as mylA, encoding the MYB-like protein containing two MYB repeats. Functional studies revealed that MylA was essential for normal fungal growth and development. Phenotypic and transcriptomic analyses demonstrated that deletion of mylA affected stress tolerance, cell wall integrity, and long-term viability in A. nidulans conidia. In addition, the germination rate of the mylA deletion mutant conidia was decreased compared with that of the wild-type conidia. Overall, this study suggests that MylA is critical for appropriate development, conidial maturation, dormancy, and germination in A. nidulans. Fungal growth on construction materials in tropical climates can degrade aesthetics and manifestations on modern and historical sick buildings, affecting the health of their inhabitants. This study synthesized ZnO nanoparticles with enhanced antifungal properties using a precipitation method. Different concentrations (25%, 50%, and 100%) of Eichhornia crassipes aqueous extract were used with Zn(NO3)2·6H2O as the precursor to evaluate their spectroscopic, morphological, textural, and antifungal properties. X-ray diffraction confirmed the hexagonal wurtzite phase of ZnO with crystallite sizes up to 20 nm. Fourier-transform infrared spectroscopy identified absorption bands at 426, 503, and 567 cm−1 for ZnO-100, ZnO-50, and ZnO-25, respectively. Nitrogen physisorption indicated a type II isotherm with macropores and a fractal dimension coefficient near 2 across all concentrations. Polydispersity index analysis showed that ZnO-50 had a higher PDI, indicating a broader size distribution, while ZnO-25 and ZnO-100 exhibited lower PDI values, reflecting uniform and monodisperse particle sizes. FESEM observations revealed semi-spherical ZnO morphologies prone to agglomeration, particularly in ZnO-25. Antifungal tests highlighted ZnO-25 as the most effective, especially against Phoma sp. with an MFC/MIC ratio of 78 µg/mL. Poisoned plate assays demonstrated over 50% inhibition at 312 µg/mL for all tested fungi, outperforming commercial antifungals. The results indicate that ZnO NPs synthesized using E. crassipes extract effectively inhibit fungal growth on construction materials. This procedure offers a practical approach to improving the durability of building aesthetics and may contribute to reducing the health risks associated with exposure to fungal compounds.
