Abstract Objectives This research simulates an adaptive version of the IDAS‐II (IDAS‐CAT). Methods 2021 participants from both community (n = 1692) and patients (n = 329) samples completed the IDAS‐II. Item response theory metric properties of the IDAS‐II full test and the 20‐items of the general depression (GD) scale were obtained. The efficiency and accuracy of different computerized adaptive algorithms were simulated. Different subsamples completed additional external measures in order to gather evidence of validity of the scores estimated with the simulated adaptive algorithms selected. Results Both unidimensional computerized adaptive testing algorithm selected for the GD scale and the bifactor model chosen for the full test, allow 70% reduction in the length of administration, maintaining a measurement error below 0.30 on the general and 0.50 on the specific factors. Results show high correlations of the scores estimated with the adaptive algorithms and the estimates based on the full test, as well as correlations with external criteria almost equal to those generated with the full test. Conclusions IDAS‐CAT could be a reliable and fast tool for measuring internalizing spectrum.

Background: The diversity of patients' symptomatology among people seeking treatment on community-based mental health services poses significant challenges to traditional models of care. Recent approaches favor identifying transdiagnostic factors that allow a better understanding of patient heterogeneity and designing more effective and quality interventions. This study examines the heterogeneity of patients with internalizing symptoms based on profiles identified with cognitive and motivational control variables. Differences between these profiles on dimensional measures of psychopathology and quality of life are examined. Methods: 263 patients were selected by non-probabilistic sampling procedures on mental health services in the province of Huelva (Spain). A latent class analysis on the standardized scale scores of The Behavioral Inhibition/Behavioral Activation System Scales and the Effortful Control Scale of the Adult Temperament Questionnaire Short-Form was conducted. Profiles were compared on the scores of the Inventory of Depression and Anxiety Symptoms-II, the WHO Disability Assessment Schedule II, and the Health Assessment Questionnaire SF-36. Results: The four latent profile solution is the one that showed the best fit indicators and substantive interpretability, with a kappa of 0.94 in the cross-validation procedure with 75% of the sample. No sex differences were found between the profiles (χ32 5.17, p = .160). Profiles #1 and #3, both characterized by an imbalance between low activation and high inhibition, had lower well-being, lower functionality, and quality of life. When comparing profile #2 (featuring the highest inhibitory control) lower scores on most internalizing scales are observed, specially claustrophobia, social anxiety, panic mania. Profile #4 (low control, high activation, and high inhibition) showed greater scores on both mania and euphoria and lower scores on emotional role. Conclusions: We identified four distinctive profiles that had overly increased behavioral inhibition (as expected in internalizing disorders) and differed in the degree of imbalance between inhibition and activation systems, and between motivational systems and top-down cognitive control. The profile characterized by high activation and reduced cognitive (inhibitory) control was the one showing greater mood-related symptoms and lower levels of quality of life. These profiles could be generated by treatment providers to guide clinical management in an evidence-based manner.

The Dark Energy Spectroscopic Instrument (DESI) survey will measure spectroscopic redshifts for millions of galaxies across roughly $14,000 \, \mathrm{deg}^2$ of the sky. Cross-correlating targets in the DESI survey with complementary imaging surveys allows us to measure and analyze shear distortions caused by gravitational lensing in unprecedented detail. In this work, we analyze a series of mock catalogs with ray-traced gravitational lensing and increasing sophistication to estimate systematic effects on galaxy-galaxy lensing estimators such as the tangential shear $\gamma_{\mathrm{t}}$ and the excess surface density $\Delta\Sigma$. We employ mock catalogs tailored to the specific imaging surveys overlapping with the DESI survey: the Dark Energy Survey (DES), the Hyper Suprime-Cam (HSC) survey, and the Kilo-Degree Survey (KiDS). Among others, we find that fiber incompleteness can have significant effects on galaxy-galaxy lensing estimators but can be corrected effectively by up-weighting DESI targets with fibers by the inverse of the fiber assignment probability. Similarly, we show that intrinsic alignment and lens magnification are expected to be statistically significant given the precision forecasted for the DESI year-1 data set. Our study informs several analysis choices for upcoming cross-correlation studies of DESI with DES, HSC, and KiDS.

The Global Navigation Satellite System (GNSS) is a well-known and fundamental tool for crustal monitoring projects and tectonic studies, thanks to its high coverage and the high quality of the data they provide. In particular, at slowly convergent margins, where deformation rates are of the order of a few millimetres per year, GNSS monitoring proves to be beneficial in detecting the diffuse deformation responsible for tectonic stress accrual. Its strength lies in the high precision achieved by GNSS permanent stations, especially when long-term data and stable structures are available at the stations. North-eastern Italy is a tectonically active region located in the northernmost sector of the Adria microplate, slowly converging with the Eurasia plate, characterized by low deformation rates and moderate seismicity. It greatly benefits from continuous and high-precision geodetic monitoring, since it has been equipped with a permanent GNSS network providing real-time data and daily observations over 2 decades. The Friuli Venezia Giulia Deformation Network (FReDNet) was established in the area in 2002 to monitor crustal deformation and contribute to the regional seismic hazard assessment. This paper describes GNSS time series spanning 2 decades of stations located in north-eastern Italy and surroundings as well as the outgoing velocity field. The documented dataset has been retrieved by processing the GNSS observations with the GAMIT/GLOBK software ver10.71, which allows calculation of high-precision coordinate time series, position and velocity for each GNSS station by taking advantage of the high-performance computing resources of the Italian High-Performance Computing Centre (CINECA) clusters. The GNSS observations (raw and standard RINEX – Receiver INdependent EXchange – formats) and the time series estimated with the same procedure are currently daily continued, collected and stored in the framework of a long-term monitoring project. Instead, velocity solutions are intended for annual updates. The time series and velocity field dataset documented here are available on Zenodo (https://doi.org/10.5281/zenodo.8055800, Tunini et al., 2024).

Abstract Background Uncontrolled inflammation contributes to the progression of organ damage in acute conditions, such as acetaminophen-induced acute liver injury (APAP-ALI) and there are limited treatments for this condition. AT7519, a cyclic-dependent kinase inhibitor (CDKI), has been used successfully in several conditions, to resolve inflammation and return tissue homeostatic functions. AT7519 has not been assessed in APAP-ALI and its effect on APAP metabolism is unknown. Targeted chromatography and mass spectrometry can be used to assess multiple compounds simultaneously and this approach has not been applied yet to measure APAP and AT7519 in a mouse model. Results We show an optimised simple and sensitive LC–MS/MS method for determining concentrations of AT7519 and APAP in low volumes of mouse serum. Using positive ion mode electrospray ionisation, separation of AT7519 and APAP and their corresponding isotopically labelled internal standards [2H]8-AT16043M (d8-AT7519) and [2H]8-APAP (d4-APAP), was achieved on an Acquity UPLC BEH C18 column (100 × 2.1 mm; 1.7μm). A gradient mobile phase system of water and methanol was delivered at a flow rate of 0.5 mL/min with a run time of 9 min. Calibration curves were linear, intra-day and inter-day precision and accuracy were acceptable and the covariates of all standards and quality control replicates were less than 15%. The method was successfully applied to evaluate AT7519 and APAP levels 20 h post AT7519 (10 mg/mg) in C57Bl6J wild type mouse serum treated with either vehicle or APAP. Serum AT7519 was significantly higher in mice that had received APAP compared to control, but there was no correlation between APAP and AT7519 quantification. There was also no correlation of AT7519 and hepatic damage or proliferation markers. Conclusion We optimised an LC–MS/MS method to quantify both AT7519 and APAP in mouse serum (50 µL), using labelled internal standards. Application of this method to a mouse model of APAP toxicity proved effective in accurately measuring APAP and AT7519 concentrations after i.p. dosing. AT7519 was significantly higher in mice with APAP toxicity, indicating hepatic metabolism of this CDKI, but there was no correlation with markers of hepatic damage or proliferation, demonstrating that this dose of AT7519 (10 mg/kg) does not contribute to hepatic damage or repair. This optimised method can be used for future investigations of AT7519 in APAP in mice.

Abstract Conservation translocations are frequently inhibited by extensive dispersal after release, which can expose animals to dispersal‐related mortality or Allee effects due to a lack of nearby conspecifics. However, translocation‐induced dispersals also provide opportunities to study how animals move across a novel landscape, and how their movements are influenced by landscape configuration and anthropogenic features. Translocation among populations is considered a potential conservation strategy for lesser prairie‐chickens (Tympanuchus pallidicinctus). We determined the influence of release area on dispersal frequency by translocated lesser prairie‐chickens and measured how lesser prairie‐chickens move through grassland landscapes through avoidance of anthropogenic features during their dispersal movements. We translocated 411 lesser prairie‐chickens from northwest Kansas to southeastern Colorado and southwestern Kansas in 2016–2019. We used satellite GPS transmitters to track 115 lesser prairie‐chickens throughout their post‐release dispersal movements. We found that almost all lesser prairie‐chickens that survived from their spring release date until June undergo post‐translocation dispersal, and there was little variation in dispersal frequency by release area (96% of all tracked birds, 100% in Baca County, Colorado, 94% in Morton County, Kansas, n = 55). Dispersal movements (male: 103 ± 73 km, female: 175 ± 108 km, n = 62) led to diffusion across landscapes, with 69% of birds settling >5 km from their release site. During dispersal movements, translocated lesser prairie‐chickens usually travel by a single 3.75 ± 4.95 km dispersal flight per day, selecting for steps that end far from roads and in Conservation Reserve Program (CRP) grasslands. IntroductionGastric cancer (GC) is one of the main causes of death from cancer globally. Long-term survival, especially in Western countries, remains dismal, with no significant improvements in recent years. Therefore, precise identification of clinical and pathological risk factors is crucial for prognosis, as it allows a better selection of patients suitable for oncologically radical treatments and contributes to longer survivals.MethodsWe devised a retrospective observational longitudinal study over 10 years of experience with GC patients operated with curative intent.ResultsSeveral factors were thoroughly investigated in a multivariate analysis to look for significance as independent risk factors for disease-free survival. Our results showed that only BMI, pTNM, and lymph node ratio expressed hazard ratios with implications for survival in our series of patients.DiscussionAlthough limited by the retrospective nature of the study, this is one of the few cancer reports from Northern Italy showing results over 10 years, which may in our view, have an impact on decision-making processes for multidisciplinary teams dedicated to the care of gastric cancer patients.

Abstract Translocation is a management tool used to restore or augment wildlife populations, but outcomes of translocations are often poorly documented and can have varying levels of success for improving wildlife population declines. The lesser prairie‐chicken (Tympanuchus pallidicinctus) is a prairie grouse endemic to the southern Great Plains. In response to declining abundance and distribution, in 2023 lesser prairie‐chickens were listed as threatened or endangered under the Endangered Species Act in different states. Translocation is a potential management response to population declines when there is an availability of unoccupied habitats, but translocation efficacy has not been evaluated for lesser prairie‐chickens. We translocated 411 lesser prairie‐chickens seasonally from 2016‐2019 and monitored the translocated lesser prairie‐chicken population from 2017–2022. To assess translocation as a management tool for lesser prairie‐chickens, we estimated survival for 2017–2020 and conducted lek surveys during 2017–2022. Over a fifth (22.8%, n = 94) of translocated birds either died or went missing within the first 2 weeks following release. Survival rates of translocated birds during the breeding (0.44 ± 0.02 [SE]) and nonbreeding (0.55 ± 0.03 [SE]) seasons were relatively low compared to nontranslocated lesser prairie‐chickens in other studies (0.63–0.93 for breeding season; 0.43–0.87 for non‐breeding season). Twenty‐seven percent of translocated birds survived to the breeding season after release (i.e., >1 year). Translocated lesser prairie‐chickens initiated 28 lekking sites over the study period. We estimated 77% of males available >2 weeks post release participated in lekking activity. The number of leks and male high counts on leks in the study area increased after translocation, peaking one year post‐translocation (an overall increase of 250% and 340%, respectively). However, both the number of leks and male high counts decreased (48% and 39%, respectively) within 3 years after translocation cessation. Establishment of leks and increasing lek attendance directly following translocation initially suggested that translocation could be a viable management tool. However, survival rates after translocation and declining lek counts following translocation indicates that the increased population abundance and occupied range from this translocation effort may be unsustainable. Our results highlight the necessity of monitoring to determine outcomes of a large lesser prairie‐chicken translocation. Other management strategies, such as targeted grassland restoration and management in areas of greatest lesser prairie‐chicken density, could be more beneficial for conservation of lesser prairie‐chicken populations.

BackgroundSevere radiation-induced lymphopenia (RIL) in patients undergoing chemoradiotherapy (CRT) for non-small cell lung cancer (NSCLC) is associated with decreased immunotherapy efficacy and survival. At The Christie and MD Anderson Cancer Center (MDACC), prediction models for lymphopenia were developed in lung and esophageal cancer patients, respectively. The aim of this study was to externally validate both models in patients with stage III NSCLC.MethodsPatients who underwent concurrent CRT for stage III NSCLC in 2019–2021 were studied. Outcomes were grade ≥3 and grade 4 lymphopenia during CRT. The Christie model predictors for grade ≥3 lymphopenia included age, baseline lymphocyte count, radiotherapy duration, chemotherapy, mean heart and lung doses, and thoracic vertebrae V20Gy. MDACC predictors for grade 4 lymphopenia were age, baseline lymphocyte count, planning target volume (PTV), and BMI. The external performance of both models was assessed.ResultsAmong 100 patients, 78 patients (78%) developed grade ≥3 lymphopenia, with grade 4 lymphopenia in 17 (17%). For predicting grade ≥3 lymphopenia, the Christie and MDACC models yielded c-statistics of 0.77 and 0.79, respectively. For predicting grade 4 lymphopenia, c-statistics were 0.69 and 0.80, respectively. Calibration for the Christie and MDACC models demonstrated moderate and good agreement, respectively.ConclusionThe PTV-based MDACC prediction model for severe RIL demonstrated superior external performance in NSCLC patients compared to the dosimetry-based Christie model. As such, the MDACC model can aid in identifying patients at high risk for severe lymphopenia. However, to optimize radiotherapy planning, further improvement and external validation of dosimetry-based models is desired.

Background and Aims: Inflammatory bowel disease (IBD) is associated with increased circulating damage-associated molecular patterns, in particular, the highly pro-inflammatory mitochondrial DNA (mtDNA). Here, we study the importance of blood neutrophils in mtDNA release via neutrophil extracellular trap (NET) formation and mitochondrial NETosis, where neutrophils specifically expulse mtDNA as potential targetable biological pathways. Methods: We investigated the roles of A23187 (a known NET stimulant), granulocyte macrophage stimulating factor, lipopolysaccharide (LPS), and human IBD plasma in their ability to induce NET formation, mitochondrial NETosis, mtDNA, and total DNA release from human blood neutrophils; and the evidence for increased NET formation in IBD. Results: We demonstrated that NET formation resulted in significant DNA (P < .0001) and mtDNA release (P < .0001) with long DNA fragments (>1000 base pairs) with NETs containing high levels of mtDNA. Using previously described in vitro conditions for mitochondrial NETosis, granulocyte macrophage stimulating factor + LPS triggered neutrophil mtDNA release at lower levels but not NETosis. LPS alone can trigger neutrophilic DNA release without NET formation. Heterologous coculture with plasma from patients with active IBD (vs remission [n = 6/group]) were not associated with significantly higher levels of NETs and mtDNA release. During coculture with active IBD plasma (vs remission), citrullinated histone 3 (CitH3) (a NETs biomarker) levels were significantly lower (P < .001). Similarly, CitH3 levels were lower in stool supernatants of patients with active IBD vs remission (n = 19/12, P = .0001). Stool CitH3 negatively correlates with stool calprotectin, a biomarker for gut inflammation (r = −0.47, P = .03). Conclusion: Hence, although blood neutrophils remain an important source of circulating mtDNA with defined mechanisms for release via NET formation and during neutrophil activation, our data do not support excessive systemic NET formation as a dominant underpinning pathobiological process in IBD.

4D printing (4DP) is a growing branch of 3D printing technology that involves the design of composite material architectures capable of shape-change transformations, which occur post printing, in response to external stimulus. Among these, Wood Polymer Composites (WPCs) change their shape in reaction to changes of moisture content, shrinking or swelling like natural wood until the equilibrium with the environment is reached. Such intrinsic material behavior can be particularly useful in the development of passive moisture airflow controllers that can modulate humidity and airflow in indoor environments to improve air quality. Precise measurement of the time-based stimulus induced shape-change response of these composites is critical to assess the responsiveness, velocity of reaction and overall deformation of the designed 4DP composite mechanisms. Up until now, Digital Image Correlation (DIC) techniques have been widely used for such purpose. However, DIC methods require expensive equipment and costly commercial software. This paper presents a Low-Cost Depth-Camera (LCDC) method that uses a free custom algorithm that returns a 3D coloured displacement map with the corresponding meshes of the acquired object. The LCDC method does not require specialized equipment and allows for an overall understanding of the time-dependent deformation of 4DP actuators, this method also facilitates the comparison between composites with different properties under the same external conditions. This new LCDC method has the potential to further 4DP research by providing an open-source, accessible and reliable tool to assess 3D displacement measurements.

Abstract Background Macrophages play a central role in inflammation by phagocytosing invading pathogens, apoptotic cells and debris, as well as mediating repair of tissues damaged by trauma. In order to do this, these dynamic cells generate a variety of inflammatory mediators including eicosanoids such as prostaglandins, leukotrienes and hydroxyeicosatraenoic acids (HETEs) that are formed through the cyclooxygenase, lipoxygenase and cytochrome P450 pathways. The ability to examine the effects of eicosanoid production at the protein level is therefore critical to understanding the mechanisms associated with macrophage activation. Results This study presents a stable isotope labelling with amino acids in cell culture (SILAC) -based proteomics strategy to quantify the changes in macrophage protein abundance following inflammatory stimulation with Kdo2-lipid A and ATP, with a focus on eicosanoid metabolism and regulation. Detailed gene ontology analysis, at the protein level, revealed several key pathways with a decrease in expression in response to macrophage activation, which included a promotion of macrophage polarisation and dynamic changes to energy requirements, transcription and translation. These findings suggest that, whilst there is evidence for the induction of a pro-inflammatory response in the form of prostaglandin secretion, there is also metabolic reprogramming along with a change in cell polarisation towards a reduced pro-inflammatory phenotype. Conclusions Advanced quantitative proteomics in conjunction with functional pathway network analysis is a useful tool to investigate the molecular pathways involved in inflammation.

In vergelijking met andere domeinen binnen het persoonlijkheidsonderzoek staat het onderzoek naar persoonlijkheid en persoonlijkheidspathologie bij ouderen nog in zijn kinderschoenen. De laatste jaren is echter de interesse in dit onderwerp sterk toegenomen gezien het groeiende aandeel ouderen in de bevolking, ook al blijft de kennis hieromtrent nog relatief beperkt. Onderzoekers binnen dit domein staan voor enkele uitdagingen. Enerzijds is er nog maar weinig geweten over het beloop van persoonlijkheid en persoonlijkheidspathologie op latere leeftijd, daar longitudinale studies hieromtrent schaars, zo niet onbestaand zijn. Anderzijds blijkt zowel vanuit de praktijk als vanuit de onderzoeksliteratuur een grote nood aan betrouwbare en valide instrumenten voor het meten van persoonlijkheidskenmerken bij ouderen. In dit artikel worden de voornaamste conceptuele en methodologische knelpunten besproken, alsook recente ontwikkelingen in Nederland en Vlaanderen binnen dit onderzoeksdomein.

IntroductionHigh quality corn silage depends on factors such as corn type, stage of crop development at harvest time, fermentation time, in addition to use or not of inoculants. This study aimed to investigate the impact of maturity stage, bacterial inoculation, and storage time on fermentation, aerobic stability, and nutritional characteristics of flint corn silage and their implications for corn silage management.MethodsA flint corn hybrid was harvested very early, early, and medium (at 250, 300 and 350 g dry matter (DM)/kg as fed, respectively) and ensiled in mini-silos without (control) or with Lentilactobacillus buchneri CNCM I-4323 at 1 × 105 cfu/g for 120, 240 and 360 d to investigate how these factors interact with each other.Results and discussionThere was only a small increase (7 g/kg starch; p = 0.003) in starch digestibility (starch-D) in the silages stored for 360 d when compared to that stored for 240 d, but with no difference for 120 d. Despite the reduced starch-D (526 vs. 694 g/kg starch; p 120 d) with the goal of increasing silage digestibility did not occur. Harvesting whole-crop flint corn with 300 to 350 g/kg DM is desirable to have higher DM yield and starch accumulation. Inoculation with L. buchneri is recommended to preserve the silage against aerobic deterioration. This study has shown the importance of harvesting flint corn at the right time, and the need for inoculation with L. buchneri to ensure greater yield, starch accumulation, and silage preservation, if 120 days of storage are not exceeded.

The Dark Energy Spectroscopic Instrument (DESI) is a revolutionary instrument designed for precise measurements of cosmic distances and the investigation of dark energy. DESI utilizes 5000 optical fibers to simultaneously measure the spectra of distant objects and aims to measure 40 million galaxies and quasars in a 5 yr survey. One of the critical challenges to DESI’s success was ensuring that the fiber system was not only highly efficient but also delivered a highly stable beam enabling more reliable sky subtraction for measurements of faint objects. We achieved this stability by minimizing the stress on the fiber system during the manufacture and operation of the telescope and fiber positioning robots. We installed the DESI fiber system on the 4 m Mayall telescope with ≥99% of fibers intact, and the instrument has delivered superb optical performance throughout the initial years of the DESI survey, including ≥90% average throughput when injected with a focal ratio of ∼ f /3.9 as delivered by the primary focus corrector, excluding fiber absorption losses. The design of DESI required multiple innovations to achieve these requirements, such as cleaved fibers bonded with a UV-curing epoxy to glass ferrules in the focal plane and fusion splicing instead of physical connectors. In this paper, we describe the development, delivery, and installation of the fiber system, the innovations that made the state-of-the-art performance possible, and the key lessons learned that could benefit future projects.

We explore the influence of radio-mode feedback on the properties of the cool circumgalactic medium (CGM). To this end, we assemble a statistical sample of approximately 30,000 radio galaxies with background quasars by combining optical spectroscopic measurements of luminous red galaxies and quasars from the year 1 data set of the Dark Energy Spectroscopic Instrument and radio sources from the LOw-Frequency ARray Two-metre Sky Survey (LoTSS) DR2 catalog and the Very Large Array Sky Survey (VLASS) quick-look catalog. Galaxies with similar optical properties but with no radio counterparts in LoTSS and VLASS are selected as the control group. We measure the cool CGM properties of radio galaxies and their control samples traced by Mg ii absorption lines, including covering fraction, rest equivalent width, and gas kinematics. Our results show no significant difference in the properties of gas around radio galaxies and their control sample, indicating that the operating radio-mode feedback of massive galaxies does not produce detectable effects on the properties of the cool CGM. Finally, we show that the CGM of radio galaxies contains a nonnegligible amount of cool gas with approximately 10 ^10 M _⊙ . This abundance can place a stringent constraint on the radio-mode feedback models.

We present a computationally efficient galaxy archetype-based redshift estimation and spectral classification method for the Dark Energy Survey Instrument (DESI) survey. The DESI survey currently relies on a redshift fitter and spectral classifier using a linear combination of principal component analysis–derived templates, which is very efficient in processing large volumes of DESI spectra within a short time frame. However, this method occasionally yields unphysical model fits for galaxies and fails to adequately absorb calibration errors that may still be occasionally visible in the reduced spectra. Our proposed approach improves upon this existing method by refitting the spectra with carefully generated physical galaxy archetypes combined with additional terms designed to absorb data reduction defects and provide more physical models to the DESI spectra. We test our method on an extensive data set derived from the survey validation (SV) and Year 1 (Y1) data of DESI. Our findings indicate that the new method delivers marginally better redshift success for SV tiles while reducing catastrophic redshift failure by 10%–30%. At the same time, results from millions of targets from the main survey show that our model has relatively higher redshift success and purity rates (0.5%–0.8% higher) for galaxy targets while having similar success for QSOs. These improvements also demonstrate that the main DESI redshift pipeline is generally robust. Additionally, it reduces the false-positive redshift estimation by 5%−40% for sky fibers. We also discuss the generic nature of our method and how it can be extended to other large spectroscopic surveys, along with possible future improvements.

The Dark Energy Spectroscopic Instrument (DESI) completed its 5 month Survey Validation in 2021 May. Spectra of stellar and extragalactic targets from Survey Validation constitute the first major data sample from the DESI survey. This paper describes the public release of those spectra, the catalogs of derived properties, and the intermediate data products. In total, the public release includes good-quality spectral information from 466,447 objects targeted as part of the Milky Way Survey, 428,758 as part of the Bright Galaxy Survey, 227,318 as part of the Luminous Red Galaxy sample, 437,664 as part of the Emission Line Galaxy sample, and 76,079 as part of the Quasar sample. In addition, the release includes spectral information from 137,148 objects that expand the scope beyond the primary samples as part of a series of secondary programs. Here, we describe the spectral data, data quality, data products, Large-Scale Structure science catalogs, access to the data, and references that provide relevant background to using these spectra.

Highly multiplexed, fiber-fed spectroscopy is enabling surveys of millions of stars and galaxies. The performance of these surveys depends on accurately positioning fibers in the focal plane to capture target light. We describe a technique to measure the positioning accuracy of fibers by dithering fibers slightly around their ideal locations. This approach also enables measurement of the total system throughput and point-spread function delivered to the focal plane. We then apply this technique to observations from the Dark Energy Survey Instrument (DESI), and demonstrate that DESI positions fibers to within 0.″08 of their targets (5% of a fiber diameter) and achieves a system throughput within about 7% of expectations.

Extremely metal-poor galaxies (XMPGs) at relatively low redshift are excellent laboratories for studying galaxy formation and evolution in the early universe. Much effort has been spent on identifying them from large-scale spectroscopic surveys or spectroscopic follow-up observations. Previous work has identified a few hundred XMPGs. In this work, we obtain a large sample of 223 XMPGs at z < 1 from the early data of the Dark Energy Spectroscopic Instrument (DESI). The oxygen abundance is determined using the direct T _e method based on the detection of the [O iii ] λ 4363 line. The sample includes 95 confirmed XMPGs based on the oxygen abundance uncertainty; the remaining 128 galaxies are regarded as XMPG candidates. These XMPGs are only 0.01% of the total DESI observed galaxies. Their coordinates and other properties are provided in the paper. The most XMPGs have an oxygen abundance of ∼1/34 Z _⊙ , a stellar mass of about 1.5 × 10 ^7 M _⊙, and a star formation rate of 0.22 M _⊙ yr ^−1 . The two most XMPGs present distinct morphologies suggesting different formation mechanisms. The local environmental investigation shows that XMPGs preferentially reside in relatively low-density regions. Many of them fall below the stellar mass–metallicity relations (MZRs) of normal star-forming galaxies. From a comparison of the MZR with theoretical simulations, it appears that XMPGs are good analogs to high-redshift star-forming galaxies. The nature of these XMPG populations will be further investigated in detail with larger and more complete samples from the ongoing DESI survey.

The Dark Energy Spectroscopic Instrument (DESI) was designed to conduct a survey covering 14,000 deg ^2 over 5 yr to constrain the cosmic expansion history through precise measurements of baryon acoustic oscillations (BAO). The scientific program for DESI was evaluated during a 5 month survey validation (SV) campaign before beginning full operations. This program produced deep spectra of tens of thousands of objects from each of the stellar Milky Way Survey (MWS), Bright Galaxy Survey (BGS), luminous red galaxy (LRG), emission line galaxy (ELG), and quasar target classes. These SV spectra were used to optimize redshift distributions, characterize exposure times, determine calibration procedures, and assess observational overheads for the 5 yr program. In this paper, we present the final target selection algorithms, redshift distributions, and projected cosmology constraints resulting from those studies. We also present a One-Percent Survey conducted at the conclusion of SV covering 140 deg ^2 using the final target selection algorithms with exposures of a depth typical of the main survey. The SV indicates that DESI will be able to complete the full 14,000 deg ^2 program with spectroscopically confirmed targets from the MWS, BGS, LRG, ELG, and quasar programs with total sample sizes of 7.2, 13.8, 7.46, 15.7, and 2.87 million, respectively. These samples will allow exploration of the Milky Way halo, clustering on all scales, and BAO measurements with a statistical precision of 0.28% over the redshift interval z < 1.1, 0.39% over the redshift interval 1.1 < z < 1.9, and 0.46% over the redshift interval 1.9 < z < 3.5.

Despite the increasing exploitation of nanomaterials (NMs) in an array of consumer products, there are uncertainties regarding their potential adverse impact on human health. Investigation of whether NMs activate a pro-inflammatory response is routinely used to assess their toxicity in in vitro and in vivo (rodent) studies. The use of zebrafish ( Danio rerio) to investigate inflammatory responses to chemicals, pathogens and injury has increased considerably over recent years. Zebrafish have also been used to investigate the role of inflammation in disease pathogenesis and for drug discovery. Availability of transgenic strains which express fluorescent proteins in immune cells (e.g. macrophages and neutrophils) enables the visualization and quantification of immune cell accumulation in the target site(s) of interest. We therefore propose that transgenic zebrafish have great utility for screening the toxicity of NMs via investigation of inflammatory responses. Indeed, we have successfully used non-protected life stages of transgenic zebrafish with fluorescent neutrophils (Tg(mpx:EGFP 114) to investigate inflammatory responses to NMs. The more widespread use of transgenic zebrafish in nanotoxicology could reduce the reliance placed on rodents and thereby enhance the implementation of the 3Rs principles. As zebrafish continue to grow in popularity it is timely to offer guidance to new users on their use. Here we will reflect on: exposure routes that can adopted to mimic human/rodent exposure, what transgenic strains and life stages are best suited to investigate inflammatory responses, selection criteria for zebrafish embryos/larvae, the inclusion of appropriate controls, the importance of dose selection and sample size, and how the (inflammatory) response can be quantified. It is hoped that our recommendations will support the development of standard protocols that can be used to assess whether NMs activate inflammatory responses. Importantly, the themes discussed are not restricted to NMs but relevant also to zebrafish application in ecotoxicology or human health focused studies.

Abstract Water in rivers is delivered via the critical zone (CZ)—the living skin of the Earth, extending from the top of the vegetation canopy through the soil and down to fresh bedrock and the bottom of significantly active groundwater. Consequently, the success of stream‐rearing salmonids depends on the structure and resulting water storage and release processes of this zone. Physical processes below the land surface (the subsurface component of the CZ) ultimately determine how landscapes “filter” climate to manifest ecologically significant streamflow and temperature regimes. Subsurface water storage capacity of the CZ has emerged as a key hydrologic variable that integrates many of these subsurface processes, helping to explain flow regimes and terrestrial plant community composition. Here, we investigate how subsurface storage controls flow, temperature, and energetic regimes that matter for salmonids. We illustrate the explanatory power of broadly applicable, storage‐based frameworks across a lithological gradient that spans the Eel River watershed of California. Study sites are climatically similar but differ in their geologies and consequent subsurface CZ structure that dictates water storage dynamics, leading to dramatically different hydrographs, temperature, and riparian regimes—with consequences for every aspect of salmonid life history. Lithological controls on the development of key subsurface CZ properties like storage capacity suggest a heretofore unexplored link between salmonids and geology, adding to a rich literature that highlights various fluvial and geomorphic influences on salmonid diversity and distribution. Rapidly advancing methods for estimating and observing subsurface water storage dynamics at large scales present new opportunities for more clearly identifying landscape features that constrain the distributions and abundances of organisms, including salmonids, at watershed scales.

Abstract Objectives Inflammasomes induce maturation of the inflammatory cytokines IL‐1β and IL‐18, whose activity is associated with the pathophysiology of a wide range of infectious and inflammatory diseases. As validated therapeutic targets for the treatment of acute and chronic inflammatory diseases, there has been intense interest in developing small‐molecule inhibitors to target inflammasome activity and reduce disease‐associated inflammatory burden. Methods We examined the therapeutic potential of a novel small‐molecule inhibitor, and associated derivatives, termed ADS032 to target and reduce inflammasome‐mediated inflammation in vivo. In vitro, we characterised ADS032 function, target engagement and specificity. Results We describe ADS032 as the first dual NLRP1 and NLRP3 inhibitor. ADS032 is a rapid, reversible and stable inflammasome inhibitor that directly binds both NLRP1 and NLRP3, reducing secretion and maturation of IL‐1β in human‐derived macrophages and bronchial epithelial cells in response to the activation of NLPR1 and NLRP3. ADS032 also reduced NLRP3‐induced ASC speck formation, indicative of targeting inflammasome formation. In vivo, ADS032 reduced IL‐1β and TNF‐α levels in the serum of mice challenged i.p. with LPS and reduced pulmonary inflammation in an acute model of lung silicosis. Critically, ADS032 protected mice from lethal influenza A virus challenge, displayed increased survival and reduced pulmonary inflammation. Conclusion ADS032 is the first described dual inflammasome inhibitor and a potential therapeutic to treat both NLRP1‐ and NLRP3‐associated inflammatory diseases and also constitutes a novel tool that allows examination of the role of NLRP1 in human disease.

Background. The self-locating peritoneal dialysis (PD) catheter, contains a tungsten tip. The effects of magnetic resonance (MR) on the catheter were evaluated, emphasizing its MR signal, artifacts, ferromagnetism, and possible heating production during the MR sequences. Methods. The catheter was studied in an ex vivo model using a 1.5T MR system and placed into a plastic box containing saline solution. Acquisitions on coronal and axial planes were obtained on fast gradient-echo T1-weighted and fast spin-echo T2-weighted. In vivo abdominal MR exams were also carried out. Results. Overall, the catheter had good visibility. In all sequences, an extensive paramagnetic blooming artifact was detected at the level of the tip tungsten ballast, with a circular artifact of 5 cm in diameter. The catheter showed no magnetic deflection, rotation, or movements during all MR sequences. After imaging, the temperature of the saline solution did not change compared to the basal measurement. Patients safely underwent abdominal MR. Conclusions. The results point to the possibility of safely performing MR in PD patients carrying the self-locating catheter. The self-locating PD catheter is stable when subjected to a 1.5T MR system. However, it creates some visual interference, preventing an accurate study of the tissues surrounding the tungsten tip.

Abstract The ability of patients with hematologic malignancies (HM) to develop an effective humoral immune response after COVID-19 is unknown. A prospective study was performed to monitor the immune response to SARS-CoV-2 of patients with follicular lymphoma (FL), diffuse large B-cell lymphoma (DLBCL), chronic lymphoproliferative disorders (CLD), multiple myeloma (MM), or myelodysplastic/myeloproliferative syndromes (MDS/MPN). Antibody (Ab) levels to the SARS-CoV-2 nucleocapsid (N) and spike (S) protein were measured at +1, +3, +6 months after nasal swabs became PCR-negative. Forty-five patients (9 FL, 8 DLBCL, 8 CLD, 10 MM, 10 MDS/MPS) and 18 controls were studied. Mean anti-N and anti-S-Ab levels were similar between HM patients and controls, and shared the same behavior, with anti-N Ab levels declining at +6 months and anti-S-Ab remaining stable. Seroconversion rates were lower in HM patients than in controls. In lymphoma patients mean Ab levels and seroconversion rates were lower than in other HM patients, primarily because all nine patients who had received rituximab within 6 months before COVID-19 failed to produce anti-N and anti-S-Ab. Only one patient requiring hematological treatment after COVID-19 lost seropositivity after 6 months. No reinfections were observed. These results may inform vaccination policies and clinical management of HM patients.

This article discusses the oak mulch experiment conducted by researchers at the University of Florida Indian River Research and Education Center and USDA. The rationale behind the experiment and the experimental design are discussed. Results from the first 2.5 years of the study are described and explained. This includes impacts of oak mulch on both the soil and plant health. Lastly, this article discusses what’s next for both the experiment and the use of oak mulch in the future.

Rationale: Galectin-3 (Gal-3) drives fibrosis during chronic lung injury, however, its role in acute lung injury (ALI) remains unknown. Effective pharmacological therapies available for ALI are limited; identifying novel concepts in treatment is essential. GB0139 is a Gal-3 inhibitor currently under clinical investigation for the treatment of idiopathic pulmonary fibrosis. We investigate the role of Gal-3 in ALI and evaluate whether its inhibition with GB0139 offers a protective role. The effect of GB0139 on ALI was explored in vivo and in vitro.Methods: The pharmacokinetic profile of intra-tracheal (i.t.) GB0139 was investigated in C57BL/6 mice to support the daily dosing regimen. GB0139 (1–30 µg) was then assessed following acute i.t. lipopolysaccharide (LPS) and bleomycin administration. Histology, broncho-alveolar lavage fluid (BALf) analysis, and flow cytometric analysis of lung digests and BALf were performed. The impact of GB0139 on cell activation and apoptosis was determined in vitro using neutrophils and THP-1, A549 and Jurkat E6 cell lines.Results: GB0139 decreased inflammation severity via a reduction in neutrophil and macrophage recruitment and neutrophil activation. GB0139 reduced LPS-mediated increases in interleukin (IL)-6, tumor necrosis factor alpha (TNFα) and macrophage inflammatory protein-1-alpha. In vitro, GB0139 inhibited Gal-3-induced neutrophil activation, monocyte IL-8 secretion, T cell apoptosis and the upregulation of pro-inflammatory genes encoding for IL-8, TNFα, IL-6 in alveolar epithelial cells in response to mechanical stretch.Conclusion: These data indicate that Gal-3 adopts a pro-inflammatory role following the early stages of lung injury and supports the development of GB0139, as a potential treatment approach in ALI.

Background and aim: Based upon a person-centred approach, there is a growing interest in neurobiological transdiagnostic factors, such as reactive temperament (Behavioral Inhibition (BIS) and Behavioral Activation Systems (BAS)) and regulatory processes (Effortful Control (EC)). Three temperament-based personality types (Resilient, Undercontrolled and Overcontrolled type (RUO)) have been replicated in adolescents and younger adults with several clinical problems; with the resilient type (highest scores on EC) consistently showing less psychological symptoms. However, these RUO types have not been studied yet in older adults with mental disorders. Therefore, the current study investigates (1) whether these RUO types can be replicated based on the aforementioned reactive and regulative temperamental factors in older inpatients and (2) whether a higher EC is related to the presence of less psychopathology. Fluorescence spectroscopy coupled with chemometric tools is a powerful analytical method, largely used for rapid food quality and safety evaluations. However, its potential has not yet been explored in the novel food sector. In the present study, excitation emission matrices (EEMs) of 15 insect powders produced by milling insects belonging to 5 Orthoptera species (Acheta domesticus, Gryllus assimilis, Gryllus bimaculatus, Locusta migratoria, Schistocerca gregaria) from 3 different origins were investigated. Parallel factor (PARAFAC) analysis performed on the overall averaged dataset was validated for five components, highlighting the presence of five different fluorescence peaks. The presence of these peaks was confirmed on each species, suggesting that fluorescence compounds of edible insects are the same in several species. PARAFAC analysis performed on the overall averaged dataset after alternatively adding the EEM recorded from one standard compound allowed to speculate that edible insects fluorescence raises from mixtures of: tryptophan + tyrosine (PARAFAC component-1), tryptophan + tyrosine + tocopherol (PARAFAC component-2), collagen + pyridoxine + pterins (PARAFAC component-3). This study suggests that fluorescence spectroscopy may represent a powerful method for investigating composition and quality of insect-based foods.

Cyclin-dependent kinase (CDK) inhibitor drugs (CDKi), such as R-roscovitine and AT7519, induce neutrophil apoptosis in vitro and enhance the resolution of inflammation in a number of in vivo models. This class of compounds are potential novel therapeutic agents that could promote the resolution of acute and chronic inflammatory conditions where neutrophil activation contributes to tissue damage and aberrant tissue repair. In this study we investigated CDKi effects on macrophage pro-inflammatory mediator production and viability. Treatment of human monocyte-derived macrophages (MDMs) with the CDKi AT7519 and R-roscovitine at concentrations that induce neutrophil apoptosis had no significant effect on control or LPS-activated MDM apoptosis and viability, and did not detrimentally affect MDM efferocytosis of apoptotic cells. In addition, enhanced efferocytosis, induced by the glucocorticoid dexamethasone, was also unaffected after a short time treatment with R-roscovitine. Macrophage cytokine responses to inflammatory stimuli are also of importance during inflammation and resolution. As a key target of CDKi, CDK9, is involved in protein transcription via the RNA polymerase II complex, we investigated the effect of CDKi drugs on cytokine production. Our data show that treatment with AT7519 significantly downregulated expression and release of key MDM cytokines IL-6, TNF, IL-10 and IL-1β, as well as markers of pro-inflammatory macrophage polarisation. R-Roscovitine was also able to downregulate inflammatory cytokine protein secretion from MDMs. Using siRNA transfection, we demonstrate that genetic knock-down of CDK9 replicates these findings, reducing expression and release of pro-inflammatory cytokines. Furthermore, overexpression of CDK9 in THP-1 cells can promote a pro-inflammatory phenotype in these cells, suggesting that CDK9 plays an important role in the inflammatory phenotype of macrophages. Overall, this study demonstrates that pharmacological and genetic targeting of CDK9 inhibits an inflammatory phenotype in human MDMs. As such these data indicate that CDK9 may be key to therapeutically targeting pro-inflammatory macrophage functions during chronic inflammation.

This work studies the stability of wild-type frataxin and some of its variants found in cancer tissues upon Co2+ binding. Although the physiologically involved metal ion in the frataxin enzymatic activity is Fe2+, as it is customarily done, Co2+ is most often used in experiments because Fe2+ is extremely unstable owing to the fast oxidation reaction Fe2+ → Fe3+. Protein stability is monitored following the conformational changes induced by Co2+ binding as measured by circular dichroism, fluorescence spectroscopy, and melting temperature measurements. The stability ranking among the wild-type frataxin and its variants obtained in this way is confirmed by a detailed comparative analysis of the XAS spectra of the metal-protein complex at the Co K-edge. In particular, a fit to the EXAFS region of the spectrum allows positively identifying the frataxin acidic ridge as the most likely location of the metal-binding sites. Furthermore, we can explain the surprising feature emerging from a detailed analysis of the XANES region of the spectrum, showing that the longer 81-210 frataxin fragment has a smaller propensity for Co2+ binding than the shorter 90-210 one. This fact is explained by the peculiar role of the N-terminal disordered tail in modulating the protein ability to interact with the metal.

The University causes changes in the lifestyles of academics, during graduation and after graduation. Many do not work in the area that was graduated and some even do not finish college, today it is a reality observed in all higher education courses and some cases intensified due to the size of the hours and other peculiarities. In view of this premise, we sought to identify the influence of the dynamics of the Agronomic Engineering Course at the State University of Mato Grosso, New Xavantina campus on why students drop out of college, and feedback from those who have already graduated. Evaluate the professional perspective of the labor market by trained students. To observe the motivational factors in the avoidance of graduates in the course of agronomy. Analyze the trajectories of professional development throughout the university experience that lead to different expectations and ways of preparing for the job market. A questionnaire was applied through innovative teaching approaches using Web 2.0 tools, in particular Google Docs, "forms". The questionnaire link was provided by email, SMS, Facebook, Instagran and WhatsApp to the target audience. It is concluded that the evasion in the University of Mato Grosso State Nova Xavantina campus occurred for three reasons: the agronomy course was the second choice of the academic, and the lack of knowledge of the course and support of family members. Based on the perspectives of post-graduates, one can conclude that: (1) the greatest difficulty was to obtain employment, (2) about 81% are working in their training area, (3) the agronomy course was the first option of the trained students, that is, they began the course aware of the difficulties, and yet they persevered and concluded the course.

Scientific research, respect for the environment, and passion for photography merged into an exceptional heritage of images collected by the researchers and technicians of the National Institute of Oceanography and Applied Geophysics (OGS). The images were taken during past scientific expeditions conducted all over the world to widen scientific knowledge in the fields of Earth and ocean sciences, to raise awareness on the environment and conservation of natural resources, and to mitigate natural risks. In this paper, we describe a photographic exhibition organized using some of the OGS images to draw public attention to the striking effects of global warming. In the artistic images displayed, the glaciers were the protagonists. Their infinite greyish blue shades and impossible shapes were worthy of a great sculptor, and the boundaries with rocks or with the sea were sometimes sharp and dramatic and sometimes so nuanced that they looked like watercolours. The beauty of the images attracted the attention of the public to unknown realities, allowing us to document the dramatic retreat of the Alpine glaciers and to show the majesty of the Arctic and Antarctic landscapes, which are fated to vanish under the present climate warming trend. The choice of the exhibition location allowed us to reach a broad public of working-age adults, who are difficult to involve in outreach events. The creators of the images were present during the exhibition to respond to visitors' curiosity about research targets, the emotional and environmental context, and the technical details or aesthetic choices of the photographs.

Abstract Background Neutrophils rapidly respond to and clear infection from tissues, but can also induce tissue damage through excessive degranulation, when acute inflammation proceeds unchecked. A number of key neutrophil functions, including adhesion-dependent degranulation, are controlled by src family kinases. Dasatinib is a potent src inhibitor used in treating patients with chronic myeloid leukaemia and treatment-resistant acute lymphoblastic leukaemia. We hypothesized that dasatinib would attenuate acute inflammation by inhibiting neutrophil recruitment, degranulation and endothelial cell injury, without impairing bacterial clearance, in a murine model of bacteria-induced acute lung injury. C57BL/6 mice received intratracheal Escherichia coli, and were treated with intraperitoneal dasatinib or control. Bacterial clearance, lung injury, and markers of neutrophil recruitment and degranulation were measured. Separately, human blood neutrophils were exposed to dasatinib or control, and the effects on a range of neutrophil functions assessed. Results Dasatinib was associated with a dose-dependent significant increase in E. coli in the mouse lung, accompanied by impairment of organ function, reflected in significantly increased protein leak across the alveolar-capillary membrane. However, the number of neutrophils entering the lung was unaffected, suggesting that dasatinib impairs neutrophil function independent of migration. Dasatinib did not cause direct toxicity to human neutrophils, but led to significant reductions in phagocytosis of E. coli, adhesion, chemotaxis, generation of superoxide anion and degranulation of primary and secondary granules. However, no biologically important effect of dasatinib on neutrophil degranulation was observed in mice. Conclusions Contrary to our starting hypothesis, src kinase inhibition with dasatinib had a detrimental effect on bacterial clearance in the mouse lung and therefore does not represent an attractive therapeutic strategy to treat primary infective lung inflammation. Data from human neutrophils suggest that dasatanib has inhibitory effects on a range of neutrophil functions.
