Abstract

Automated analysis of high-resolution transmission electron microscopy (HRTEM) images is increasingly essential for advancing research in organic electronics, where precise characterization of nanoscale crystal structures is crucial for optimizing material properties. This paper introduces an open-source computational framework called GRATEV2.0 (GRaph-based Analysis of TEM), designed for real-time analysis of HRTEM data, with a focus on characterizing complex microstructures in conjugated polymers, illustrated using Poly[N-9'-heptadecanyl-2,7-carbazole-alt-5,5-(4',7'-di-2-thienyl-2',1',3'-benzothiadiazole)] (PCDTBT), a key material in organic photovoltaics. GRATEV2.0 employs fast, automated image processing algorithms, enabling rapid extraction of structural features like d-spacing, orientation, and crystal shape metrics. Gaussian process optimization rapidly identifies the user-defined parameters in the approach, reducing the need for manual parameter tuning and thus enhancing reproducibility and usability. Additionally, GRATEV2.0 is compatible with high-performance computing (HPC) environments, allowing for efficient, large-scale data processing at near real-time speeds. A unique feature of GRATEV2.0 is a Wasserstein distance-based stopping criterion, which optimizes data collection by determining when further sampling no longer adds statistically significant information. This capability optimizes the amount of time the TEM facility is used while ensuring data adequacy for in-depth analysis. Abstract

This work proposes a geographically-based split of the community microgrids into clusters of members that tend to have similar consumption and generation profiles. Assuming a community microgrid divided into clusters, a two-layer architecture is developed to facilitate the greater penetration of distributed energy resources in an efficient way. The first layer, referred as the market layer, is responsible for creating local energy markets with the aim of maximising the economic benefits for community microgrid members. The second layer is responsible for the network reconfiguration, which is based on the energy balance within each cluster. This layer complies with the IEC 61850 communication standard, in order to control commercial sectionalizing and tie switches. This allows the community microgrid network to be reconfigured to minimise energy exchanges with the main grid. To implement this two-layer energy management strategy, an aperiodic market approach based on Blockchain technology, and the additional functionality offered by Smart Contracts is adopted. This embraces the concept of energy communities since it decentralizes the control and eliminates intermediaries. The use of aperiodic control techniques helps to overcome the challenges of using Blockchain technology in terms of storage, computational requirements and member privacy. The scalability and modularity of the Smart Contract-based system allow each cluster of members to be designed by tailoring the system to their specific needs. The implementation of this strategy is based on low-cost off-the-shelf devices, such as Raspberry Pi 4 Model B boards, which operate as Blockchain nodes of community microgrid members. Finally, the strategy has been validated by emulating two use cases based on the IEEE 123-node system network model highlighting the benefits of the proposal.

Abstract

ObjectivesThis study aimed to explore the acceptability, feasibility, usability, and preliminary effect of an electronic patient-reported outcome (ePRO) intervention for patients with breast cancer in Mexico.DesignWe conducted a multimethod non-randomised pilot study. We used a pre-test/post-test design for quantitative assessment of the intervention's effect on patients' supportive care needs and quality of life. We conducted in-depth interviews (IDIs) with participants and healthcare workers to explore the intervention's benefits and barriers and understand its feasibility.Participants50 women aged 20-75 diagnosed with stage I-III breast cancer were enrolled within 2 weeks of starting neoadjuvant or adjuvant treatment with chemotherapy or radiotherapy. We excluded illiterate women and those with visual impairment, cognitive disability or severe depression. IDIs were conducted with 18 participants and 10 healthcare providers.SettingOncology services in three public hospitals of the Mexican Social Security Institute.InterventionThe ePRO intervention consisted of a responsive web application for weekly symptom reporting combined with proactive follow-up by nurses guided by predefined clinical algorithms for 6 weeks.Results50 women were enrolled out of 66 eligible patients approached (75.8%). All 50 completed the 4-week follow-up assessment (100% retention). Completion of the symptom registry declined from 100% in week 1 to 66% in week 6. Participants experienced decreases in supportive care needs and increased quality of life. The ePRO application was rated highly usable. Participants and health professionals both perceived intervention benefits. Drawbacks included poor fit for women receiving radiotherapy and challenges using the application for women with low digital literacy or experiencing severe symptoms.ConclusionsThis pilot study provided evidence of the high usability and potential efficacy of a web-based ePRO intervention. We revised recruitment during the pilot to include multiple facilities, and we will further revise for the randomised trial to address barriers to successful ePRO implementation.Trial registration numberClinicalTrials.gov ID: NCT05925257.

Abstract

Conventional 2-D scanning electron microscopy (SEM) is commonly used to rapidly and qualitatively evaluate membrane pore structure. Quantitative 2-D analyses of pore sizes can be extracted from SEM, but without information about 3-D spatial arrangement and connectivity, which are crucial to the understanding of membrane pore structure. Meanwhile, experimental 3-D reconstruction via tomography is complex, expensive, and not easily accessible. Here, we employ data-science tools to demonstrate a proof-of-principle reconstruction of the 3-D structure of a membrane using a single 2-D image pulled from a 3-D tomographic data set. The reconstructed and experimental 3-D structures were then directly compared, with important properties such as mean pore radius, mean throat radius, coordination number and tortuosity differing by less than 15%. Abstract

Cardiotoxicity refers to the damage induced by antineoplastic treatments, leading to various cardiovascular conditions. [18F]FDG PET radiomics analysis could provide relevant information on early onset changes occurring in cardiac metabolism of chemotherapy-induced cardiotoxicity. Patients' sociodemographic data, cardiovascular risk factors, laboratory parameters, and left ventricle [18F]FDG PET radiomic features are analyzed. The HRad index for the quantification of the heterogeneity of the metabolic uptake patterns is proposed. Statistical analysis is performed by separating patients according to the diagnosis of cancer therapy-related cardiac dysfunction (CTRCD). Baseline, intermediate, and end-of-treatment scans are evaluated as separate groups. Overall, CTRCD+ patients show lower overall mean standardized uptake values (SUVmean) compared to CTRCD− patients, with statistically significant differences between groups only observed in the intermediate PET study (p = 0.025). A total of 34 radiomic features show statistically significant differences between the CTRCD+ and CTRCD− groups in the intermediate imaging studies. In the CTRCD− group, greater overall heterogeneity of metabolic uptake is observed in the intermediate PET image compared to the CTRCD+ groups (p = 0.025). The assessment of CTRCD through [18F]FDG PET radiomics analysis could be a potential tool for the identification of a predisposition to the later development of cardiac complications after cardiotoxic treatment. 34. Multi-Centre Study of Progression Factors and Intravesical Recurrence in Patients with Urothelial Carcinoma of the Upper Urinary Tract.

Abstract

Background/Objectives: A retrospective analysis was conducted to identify factors associated with disease progression and intravesical recurrence (IVR) in a multi-centre cohort of patients with upper urinary tract urothelial carcinoma (UTUC) treated surgically between 2015 and 2021. Methods: Progression-free survival (PFS) and IVR-free survival were evaluated using a Kaplan–Meier survival curve and a Log-Rank test. Prognostic factors for progression and IVR were analysed using Cox logistic regression analysis. Results: A total of 170 patients were analysed. Up to 32.9% developed progression within 65.64 ± 3.44 months. Multivariate analysis showed that pT (HR 2.9, 95%CI 1.54–5.48, p = 0.01), margin status (HR 2.89, 95%CI 2.88–57.68, p = 0.01), and lymphovascular involvement (HR 7.97, 95% CI 1.43–44.42, p = 0.02) were independent risk factors for PFS. Up to 25.9% of patients presented with IVR at a mean time of 68.33 ± 3.59 months. A previous diagnosis of bladder cancer (BC) (HR 3.73, 95% CI 1.24–11.22, p = 0.02) and the non-invasive appearance of the tumour on computed tomography were significant risk factors for IVR (HR 0.23, 95% CI 0.05–0.95, p = 0.03). Conclusions: pT stage, margin involvement, and lymphovascular involvement were independent risk factors for UTUC progression. Abstract

Non-Hodgkin lymphoma is a heterogeneous group of cancers that triggers bone marrow infiltration in 20–40% of cases. Bone marrow biopsy in combination with a visual assessment of [18F]FDG PET/CT images is used to assess the marrow status. Despite the potential of both techniques, they still have limitations due to the subjectivity of visual assessment. The present study aims to develop models based on bone marrow uptake in [18F]FDG PET/CT images at the time of diagnosis to differentiate bone marrow status. For this purpose, a model trained for skeleton segmentation and based on the U-Net architecture is retrained for bone marrow segmentation from CT images. The mask obtained from this segmentation together with the [18F]FDG PET image is used to extract radiomics features with which 11 machine learning models for marrow status differentiation are trained. The segmentation model yields very satisfactory results with Jaccard and Dice index values of 0.933 and 0.964, respectively. As for the classification models, a maximum F1_score_weighted and F1_score_macro of 0.962 and 0.747, respectively, are achieved.
