The development of biofouling is a major problem for marine industries. The conception of antifouling and fouling release coatings, with controlled physical-chemical properties is a promising strategy. Among them, amphiphilic systems, such as those composed of a hydrophobic polydimethylsiloxane matrix and a hydrophilic polyethyleneglycol additive are the most efficient and up to date. Despite their effectiveness, these systems are questioned due to the petrochemical origin of PDMS. The aim of this project was to substitute the PDMS matrix with a biopolymer, poly(3-hydroxybuyrate-co-3-hydroxyvalerate) and to improve its anti-adhesion properties through the elaboration of an amphiphilic system, via the addition of PEG or PHBHHx-b-PEG copolymer. The results, including the physico-chemical properties of PHBHV based coatings and static adhesion tests on a marine bacterium, Bacillus 4J6 and a diatom, Phaeodactylum tricornutum are compared with those of PDMS and PEG-modified PDMS coatings. Real antiadhesion activity was obtained for the PHBHV/PHBHHx-b-PEG system for a promising eco-friendly strategy.

The rising attention to Moon-based Earth observation provides a new way to monitor Earth. Among Moon-based detectors, the SAR method has the advantage of long-term, stable, unique observations compared to space-borne SAR. Unlike space-borne SAR, however, the complicated relative motion of a Moon-based sensor makes its Doppler centroid estimation more difficult. This paper proposes a method for two-dimensional attitude steering to compensate for the Doppler centroid of Moon-based SAR. In the periodic movement between Earth and the Moon, the track speed of sites distributed around the near side of the Moon are almost perpendicular to vectors from the sites to the Earth centroid. This situation indicates the feasibility of attitude steering to guide antennas into a zero Doppler plane intersecting Earth’s surface. Therefore, this paper proposes a method for two-dimensional attitude steering to compensate the Doppler centroid to zero. In this method, the Moon is no longer deemed as a point but a natural celestial body with an average radius. Sites distributed around the hemisphere of the near side are also considered separately because of differences in spatial position and speed. Through the proposed method, the pointing of SAR antennas will be steered to a place on Earth where the Doppler centroid is compensated to zero. The JPL DE430 and EOP data are closer to reality and are used for accurate simulation. In this paper, the twodimensional attitude steering method is compared with no attitude steering and a onedimensional attitude steering method, and the results show its effectiveness.

Notch signaling plays a critical role in the development and function of macrophages. The aim of the present study was to investigate the relationship between Notch signaling pathway and macrophage apoptosis after LPS stimulation. In RAW 264.7 cells, the mRNA expression of Jagged1, Hes1, Hes 5 and GM-CSF, and protein expression of NICD1 and GM-CSF were increased after LPS stimulation. Inhibition of Notch signaling by γ-secretase inhibitor DAPT and the suppression of Notch1 expression using siRNA both significantly prevented LPS induced activation of JNK and NF-kB, and simultaneously the expression of GM-CSF was also down regulated significantly. JNK inhibitor SP600125 was used to block the phosphorylation of JNK signaling, Western blot results showed that the activation of NF-kB was blocked and expression of GM-CSF was down-regulated. Finally, flow cytometry analyses showed that the Notch signaling was involved in the regulation of macrophage apoptosis after LPS stimulation. Our study showed that the Notch signaling pathway was activated and involved in the regulation of macrophage apoptosis after LPS stimulation through JNK/ NF-kB signaling regulated GM-CSF expression.

Increasing evidence has suggested that microRNAs (miRNAs) played critical roles in cancer development by acting as a tumor suppressor or tumor-promoting genes. However, the role of microRNA-330-3p (miR-330-3p) in hepatocellular carcinoma (HCC) is still unknown. This study aimed to investigate the expression and role of miR-330-3p in hepatocarcinogenesis.A total of 30 human hepatocellular carcinoma tissues and adjacent normal tissues were obtained from 30 hepatocellular carcinoma patients. Quantitative Real Time-Polymerase Chain Reaction (qRT-PCR) assay was carried out to measure the expression of miR-330-3p in HCC tissues and cell lines. The relation between B-cell translocation gene 1 (BTG1) and miR-330-3p was predicted by TargetScan and confirmed by dual-luciferase reporter assay. Cell Counting Kit-8 (CCK-8), flow cytometry analysis, and transwell assay were used to determine cell viability, apoptosis, cell migration, and invasion, respectively. In addition, the mRNA and protein expression of Cyclin D1, Bcl-2, Bax, and matrix metalloproteinase (MMP)9 were detected using qRT-PCR and Western blotting.We found that miR-330-3p expression was up-regulated in HCC tissues and cell lines. BTG1 was a direct target of miR-330-3p and it was down-regulated in HCC tissues and cell lines. Moreover, down-regulation of miR-330-3p suppressed HCCLM3 cell viability, migration, invasion, and enhanced cell apoptosis, while the tumor-suppressive effects were reversed by BTG1-siRNA. In addition, miR-330-3p inhibitor decreased the expression of Cyclin D1, Bcl-2, and MMP9 while enhanced the expression of Bax. Meanwhile, BTG1-siRNA led to the opposite effects.The data suggested that miR-330-3p acted as a tumor gene in HCC by targeting BTG1 and it might be a potential therapeutic target for the HCC treatment.

In this paper, the weld appearance, microstructures and mechanical properties of electron beam welding-welded joints of SiCp/Al composite and Ti-6Al-4V were studied. The results showed that the weld appearance and mechanical properties were poor when the heat source was located at the middle of the butt joint. The weld was mainly composed of Ti3Al intermetallic compounds, but large amounts of Al4C3 brittle phase appeared at the fusion line close to the SiCp/Al composite. The weld appearance was improved by deviating the heat source towards the Ti-6Al-4V side. The microstructures of the weld changed into an α(α')+β dual-phase structures and the amounts of Al4C3 brittle phase decreased. A well-formed joint with a tensile strength of 108·9 MPa, 60% that of the base metal, was obtained when the heat source was deviated by 0·3 mm towards the Ti-6Al-4V side.

Summary Portulaca oleracea, an r-strategist, is one of the world's most troublesome weeds. During hot seasons, P. oleracea frequently becomes monodominant in choy sum (Brassica parachinensis) fields in Guangzhou city, southern China. Here, we studied the seasonal dynamics of P. oleracea's germinable soil seedbank, population density and above-ground biomass in choy sum fields that had been cultivated continuously for several years. Using P. oleracea seeds collected from these fields, we tested seed dormancy, survival and germination, seedling growth and generation time. Portulaca oleracea occurred at high levels during the hot season, but its occurrence was low, and the germinable soil seedbank was much greater during the cold season. The weed's opportunistic characteristics allowed it to avoid freezing and to proliferate during optimal conditions. Portulaca oleracea's generation time was very flexible, as short as 31 days during the hot season, but longer than 100 days during the colder season. Seed dormancy tended to be shorter when the seeds were stored at a higher temperature. At a temperature of 35°C, both seed germination and seedling growth showed advantages over those of choy sum. Storage for one year at a temperature of −20°C or burial in a paddy field did not significantly reduce P. oleracea seed germination. Nevertheless, seed storage at a temperature of 15°C and soil coverage of 0.5 cm on top of the seeds significantly constrained seed germination. ‘Stale seedbed’ and/or coverage of the surface with soil are recommended during the hot season. However, rotation of rice and upland crops is not an efficient method for managing Portulaca oleracea infestation.

Dense β-Si3N4 ceramics were fabricated by hot-pressed sintering at 1800°C with a mixture of α-Si3N4 powders and different amount of CeO2 additive, of which the latter is less frequently used as sintering aid to Si3N4. The effects of sintering additives concentration on grain boundary phase composition, phase transformation and the grain size of silicon nitride ceramics were investigated, followed by the influences of sintering additives content on the densification and mechanical properties of silicon nitride ceramics. The results show that increased CeO2 addition leads to greater growth anisotropy of elongated grains, and the effects of the microstructure on fracture toughness of silicon nitride ceramics were closely related to CeO2.

Therapeutic resistance has been a great obstacle for successful treatment of breast cancer. Our study aimed to explore the role of microRNA-760 (miR-760) in chemoresistant breast cancer cells.Real-time PCR was performed to measure the mRNA expression of miR-760 and Nanog. Western blot was used to determine the protein expression of Nanog and mesenchymal and epithelial markers. Cell viability was measured by the CCK-8 assay.Our results showed that the expression of miR-760 was significantly reduced the doxorubicin (DOX)-resistant MCF-7/DOX cells and chemoresistant breast cancer tissues. Moreover, up-regulation of miR-760 sensitized breast cancer cells to the anti-cancer agents. The MCF-7/DOX cells exhibited increased expression of Snail, a mesenchymal marker, and decreased levels of E-Cadherin, an epithelial marker. In addition, overexpression of miR-760 suppressed the expression of Nanog, a transcriptional factor involved in chemoresistance, and resulted in the reversal of EMT in breast cancer cells.Our study demonstrated that miR-760 modulated chemoresistance through the epithelial-mesenchymal transition in breast cancer cells, providing a potential therapeutic target for treatment of drug-resistant breast cancer.

High temperature interfacial evolution by reaction between liquid Al and solid Ti3Al has been studied by observation of reaction couples isothermally reacted at 700, 800 and 900°C for different time intervals between 1 and 60 min respectively. Only TiAl3 phase was detected in Al/Ti3Al reaction couples up to 900°C. The reaction zone consisted of aluminide particles dispersed in an Al rich matrix. However, microstructures in the inner, central and marginal regions of reaction zone were different and were continuously changing across the width of reaction zone. A model corresponding to evolution progress was drawn schematically. A water quenched experiment confirmed the restriction effect of dispersed TiAl3 particles to Al matrix and suggested that TiAl3p/Al composite could be fabricated by this in situ reaction method.

To characterise anomalies embedded in graphite fibre reinforced aluminium matrix composite, chirplet parameters of ultrasonic signals were obtained by chirplet signal decomposition method. It was confirmed that the parameters of decomposed chirplets can represent the characteristic of ultrasonic signals in time and frequency domains compared with conventional time and frequency analysis method. Chirplet parameters were used to identify three types of anomalies embedded in graphite fibre reinforced aluminium matrix composite. The variations of centre frequency and phase of three anomalies were interpreted by frequency dependent attenuation coefficient of composite and frequency dependent reflection coefficient of thin anomalies embedded in composite.

Summary Solidago canadensis is a noxious invasive species in China. We examined the influences of herbicides on pollen germination and pollen tube growth and the effects of cutting flowers and herbicides on seed production and quality. We also determined the seed production and resource allocation to different organs of S. canadensis after uprooting the plants during their reproductive periods. The main results were as follows: (i) 49% pollen of S. canadensis was able to germinate on an appropriate medium; (ii) compared with paraquat, ethephon and 2,4-D, glyphosate had an ideal inhibitory effect on pollen germination and the formation of seeds of S. canadensis; (3) cut flowering branches, when placed in water or nutrient solution, were still able to produce viable seeds; (4) resources were allocated more to the inflorescences to facilitate seed maturation after plants were uprooted at the flowering stage. Uprooted flower branches were still able to produce viable seeds, especially those in full flower. Therefore, in order to reduce the seed production of S. canadensis and spread of the species we suggest: (i) apply glyphosate at the flower bud stage, (ii) completely destroy uprooted plants at flowering stages and (iii) avoid using the flowering branches as ornamental materials.

Purpose The regimens of weekly irinotecan with platinum have been used for treatment of metastatic small-cell lung cancer (SCLC). We conducted a multi-institution phase II trial to evaluate a novel 21-day schedule of irinotecan and carboplatin in patients with relapsed or extensive SCLC. Patients and Methods Eighty patients were enrolled with the following characteristics: 39 male patients, 41 female patients; median age, 65 years; and Zubrod performance status, 0 to 1 in 85% and 2 in 15% of patients. Dosing schemas were based on the maximum-tolerated dose derived in a previous phase I study. Chemotherapy-naive patients with extensive SCLC were treated with irinotecan 200 mg/m2 and carboplatin area under the curve (AUC) of 5 (arm A). Patients, who had previously been treated with chemotherapy and had relapsed disease received irinotecan 150 mg/m2 and carboplatin AUC of 5 (arm B). In each study arm, the treatment was given every 21 days for up to six cycles. Results The most common grade 3 to 4 toxicities included neutropenia (54%), thrombocytopenia (22%), anemia (13%), diarrhea (22%), and nausea/emesis (11%) in both study arms. There were three treatment-related deaths owing to neutropenic sepsis. Among 72 assessable patients, response rates of 65% and 50% were observed, respectively, for arm A and arm B. The median survival for both study arms was identical at 10 months (95% CI, 6 to 14 months). A response rate of 65% was observed in the intracranial disease of 14 patients with known brain metastases. Conclusion This 21-day regimen of irinotecan and carboplatin seems promising for the treatment of relapsed SCLC.

Spinal cord repair is a problem that has long puzzled neuroscientists. The failure of the spinal cord to regenerate and undergo reconstruction after spinal cord injury (SCI) can be attributed to secondary axonal demyelination and neuronal death followed by cyst formation and infarction as well as to the nature of the injury environment, which promotes glial scar formation. Cellular replacement and axon guidance are both necessary for SCI repair. Multipotent neural stem cells (NSCs) have the potential to differentiate into both neuronal and glial cells and are, therefore, likely candidates for cell replacement therapy following SCI. However, NSC transplantation alone is not sufficient for spinal cord repair because the majority of the NSCs engrafted into the spinal cord have been shown to differentiate with a phenotype which is restricted to glial lineages, further promoting glial scaring. Olfactory ensheathing cells (OECs) are a unique type of glial cell that occur both peripherally and centrally along the olfactory nerve. The ability of olfactory neurons to grow axons in the mature central nervous system (CNS) milieu has been attributed to the presence of OECs. It has been shown that transplanted OECs are capable of migrating into and through astrocytic scars and thereby facilitating axonal regrowth through an injury barrier. Given the complementary properties of NSCs and OECs, we predict that the co-transplantation of NSCs and OECs into an injured spinal cord would have a synergistic effect, promoting neural regeneration and functional reconstruction. The lost neurocytes would be replaced by NSCs, while the OECs would build "bridges" crossing the glial scaring that conduct axon elongation and promote myelinization simultaneously. Furthermore, the two types of cells could first be seeded into a bioactive scaffold and then the cell seeded construct could be implanted into the defect site. We believe that this type of treatment would lead to improved neural regeneration and functional reconstruction after SCI.

This paper introduces the design of a new type multielectrode capacitance sensor for liquid level. The system regards electric field sensor MC33794 as the core and applies microcontroller MC9S12DJ128 to realize intelligent liquid level monitoring system, which overcomes the disadvantages of the traditional capacitance sensor, improves on the anti-jamming ability and the measurement precision and simplifies the system structure. Finally, the paper sums up the design of the system.

We have constructed transgenic Arabidopsis lines that contain a gene for green fluorescent protein (GFP) under the control of auxin-responsive domains A and B of the promoter from the pea PS-IAA4/5 gene. The chimeric transgene was named BA-mgfp5-ER. GFP was detected after the application of indole-3-acetic acid at concentrations as low as 100 nM in epidermal cells in the root elongation zone. The induction of the reporter gene was highly specific to auxin and was correlated with the auxin-induced change in epidermal cell shape. No GFP accumulation was observed in the lateral root meristem that was formed as a result of exogenous auxin application. These results suggest that auxin signals were transmitted through several distinct pathways depending on the cell type. The intensity of the GFP signal was strong enough to be observed through the plastic lid of the culture dish using a dissecting microscope, thereby enabling GFP expression to be monitored in an aseptic environment. Thus, the BA-mgfp5-ER transgenic plant can be a powerful tool for screening mutants that are defective in auxin signaling and the expression of early auxin-response genes.

8013 Background: The proteasome inhibitor (PI) ixazomib (Ixz) is the first orally administered PI approved for treating multiple myeloma (MM). It has shown clinical activity as a single agent and when used in other combinations. In this phase 1/2 trial, we evaluated Ixz as a replacement therapy for bortezomib or carfilzomib for MM patients who were refractory to a bortezomib- or carfilzomib-containing combination regimen. Methods: This was a phase 1/2, intra-patient, multicenter, open-label trial evaluating the replacement of ixazomib for bortezomib or carfilzomib for MM patients who were refractory in combination with the other agents that the patients had received and failed. Patients received Ixz on days 1, 8 and 15 on a 28-day schedule and the other drugs were administered using the same doses and schedules as they were receiving during their prior regimen. If the Ixz maximum tolerated dose (MTD) for a particular combination regimen was previously determined, then patients were enrolled directly into Phase 2 (PhII). If not, MTD was determined during the Phase 1 (PhI) portion of the trial. Results: To date, a total of 40 patients have been enrolled; 37 patients (21 were enrolled in PhI and 16 in PhII) had completed at least one cycle of this treatment. Patients received a median of 5 prior treatments (range, 1-22). The median follow-up time for all patients was 1.6 months (range, 0.1-10.7 months), whereas that of PhII was 2.2 months (range, 0.2-10.7 months). There was no clinical benefit (CBR; 0%) nor any overall response rate (ORR; 0%) for patients receiving Ixz 3 mg (PhI). Nine patients (43%) showed stable disease (SD) while 12 (57%) exhibited disease progression (PD). In PhII (4mg Ixz) portion of the trial, ORR and CBR were both 18.7% with 16 (43.2%) patients showing SD, and 18 (48.6%) patients displaying PD. Common ≥ Gr3 adverse events were anemia (11%), thrombocytopenia (5.4%), hyponatremia (5.4%), dehydration (5.4%) and neutropenia (2.7%). Conclusions: Replacement of bortezomib or carfilzomib with Ixz infrequently leads to responses among RRMM patient who have progressed while on proteasome inhibitor -containing combination regimens. Roundness error assessment is always a nonlinear optimization problem without constraints. The method of particle swarm optimization (PSO) is proposed to evaluate the roundness error. PSO is an evolution algorithm derived from the behavior of preying birds. PSO regards each feasible solution as a particle (point in n-dimensional space). It initializes a swarm of random particles in the feasible region. All particles always trace two particles in which one is the best position itself; another is the best position of all particles. According to the inertia weight and two best particles, all particles update their positions and velocities according to the fitness function. After iterations, it converges to an optimized solution. The reciprocal of the error assessment objective function is adopted as the fitness. In this paper the calculating procedures with PSO are given. Finally, an assessment example is used to verify this method. The results show that the method proposed provides a new way for other form and position error assessment because it can always converge to the global optimal solution.

Bacteriorhodopsin (bR) is a light-driven proton pump from Halobacterium salinarium and is a model system for studying membrane protein folding, stability, function, and structure. bR is composed of bacterio-opsin (bO), the 248-amino acid apo protein, and all-trans retinal, which is linked to lysine 216 via a protonated Schiff base. A bO gene (sbOd) possessing 29 unique restriction sites and a carboxyl-terminal purification epitope (1D4, nine amino acids) has been designed and synthesized. Overexpression of bO was achieved by fusion to the carboxyl terminus of maltose binding protein (MBP). The expressed fusion protein (MBP-sbO-1D4) formed inclusion bodies in Escherichia coli and, following solubilization with urea and removal of the urea by dialysis, approximately 170 mg of approximately 75% pure MBP-sbO-1D4 was obtained from 1 L of culture. MBP-sbO-1D4 formed high molecular weight (> or = 2,000 kDa) oligomers that were water-soluble. The synthetic bO with the 1D4 tag (sbO-1D4) was separated from MBP by trypsin cleavage at the factor Xa site between the MBP and sbO-1D4 domains. Selective trypsin cleavage at the factor Xa site, instead of at the 14 other potential trypsin sites within bO, was accomplished by optimization of the digestion conditions. Both MBP-sbO-1D4 and sbO-1D4 were regenerated with all-trans retinal and purified to homogeneity. In general, 6-10 mg of sbR-1D4 and 52 mg of MBP-sbR-1D4 were obtained from 1 L of cell culture. No significant differences in terms of UV/vis light absorbance, light/dark adaptation, and photocycle properties were observed among sbR-1D4, MBP-sbR-1D4, and bR from H. salinarium.

e18195 Background: MMC have traditionally evaluated pt management and potential areas for quality improvement. We hypothesized that this approach could be adapted and extrapolated to gain increased insight into how pts with common solid tumors are being treated in general oncology practice and how the approaches used align with evidence-based guidelines. Methods: 102 randomly recruited US-based general medical oncologists from a proprietary database of 4,019 agreed to consult their medical records and provide information on biomarkers assessed, systemic therapies administered and other treatment-related factors for the last pt in their practice to die from OC, CRC and/or WTLC. An IRB exemption was obtained and a modest honorarium was provided. Results: Pts with OC, CRC and WTLC received a median of 3, 3 and 2 lines of therapy, respectively, the majority of which were supported by guidelines, but in more than half of cases, the regimen administered required dose adjustment/discontinuation due to toxicity. Less than half of pts were offered trial participation and only 15 (6%) entered a trial. 19 pts with OC (25%), 25 pts with CRC (32%) and 58 pts with WTLC (73%) did not undergo BRCA, MSI and multiplex genomic testing, respectively, despite the recommendation of available guidelines. Conclusions: The MMC model may be an effective means to document variability as well as consistency in current oncologic care. Data generated via this assessment juxtaposed with current clinical evidence and available guideline recommendations could potentially serve as a powerful educational intervention. These findings require additional validation but provide initial support for future iterations of this exercise. [Table: see text]

Thoracic ganglioneuroma is sporadic and rarely reported. Pre-operative misdiagnosis often occurs in clinical practice. To improve diagnostic accuracy and facilitate differential diagnosis, we summarised the CT and MRI findings of thoracic ganglioneuroma.22 cases of thoracic ganglioneuroma confirmed by surgery and pathology were retrospectively analysed in terms of CT (16 cases) and MRI data (6 cases).Of 22 lesions, 19 occurred in the posterior mediastinum, 2 in the lateral pleura and 1 in the right chest. The CT value of the plain scans ranged from 20 to 40 HU (mean 29.1 HU) in 16 cases. Punctate calcification was noted in four cases. Patchy fat density shadow was found in one case. Arterial-phase CT found nearly no enhancement (6 cases) or slight enhancement (10 cases) with a CT value of 0-12 HU (mean 5.8 HU). In the delayed phase, enhancement was strengthened progressively, and CT value of 10-20 HU (mean 13.6 HU) was achieved after 120 s. T(1) weighted images showed homogeneous hypointense signals in five cases and hypointense signals mixed with patchy hyperintense signal shadow in one case. T(2) weighted images demonstrated heterogeneous hyperintense signals in all six cases, of which the whorled appearance was noted in one case. Gadolinium-diethylenetriaminepentaacetic acid (Gd-DTPA)-enhanced MRI found mildly heterogeneous enhancement in the arterial phase, and progressive mild enhancement in the delayed phase.Thoracic ganglioneuroma shows hypodensity in plain CT. On CT and MRI, non-enhancement or slight enhancement in artery phase and progressive mild enhancement in delay phase are characteristic manifestations of ganglioneuroma in the thorax.

In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field. © 2012 Landes Bioscience.

Preflare phenomena have been analysed in 76 radio bursts observed in four frequencies (1.42, 2.13, 2.84, and 4.26 GHz). For 23 in the 76 events the rapid fluctuation rate of the radio radiation in one of the frequencies started to rise prior to the flare onsets by a time range from several minutes to tens of minutes. Three examples of the 23 events are presented in detail to demonstrate this kind of precursor.

Human serum albumin (HSA) microspheres have been produced in the size range of 200 nm to 10 microns by the controlled addition of acetone to an aqueous solution of HSA, followed by stabilization of the formed microspheres at an elevated temperature. Microspheres produced by this acetone-heat denaturation method could be stabilized at relatively low temperatures (75 degrees C) over a short time period (15-30 min). The acetone-heat denaturation method is different from the traditional oil/water technique for preparation of HSA microspheres, both in terms of production method and the avoidance of high temperatures (100 degrees C) and extended heating times (30 min) for stabilization. This paper describes the influence of process conditions, such as volumes of acetone and HSA concentration, on the formation of the microspheres and their morphological characteristics. A loading efficiency of 8 per cent was achieved for the HSA microspheres when using rose bengal as a model compound. The release of rose bengal from the microspheres in phosphate buffered saline at 37 degrees C was dependent on the presence of the surfactant Tween 80, and varied from a 5 per cent release in 14 days in the absence of surfactant to a 50 per cent release in 8 h in the presence of surfactant.

Polyhydroxyalkanoates (PHAs) are a family of bacterially synthesized biopolyesters with biodegradability, biocompatibility, thermoprocessibilty, and flexible strengths. Poly-(R)-3-hydroxybutyrate (PHB) was the first PHA discovered and has been the best-studied PHA member. PHAs have found applications in the form of packaging materials including films, boxes, coating, fibers and foam materials, biofuels, medical implants, and drug delivery carriers. For large-scale applications such as biodegradable packaging materials, the cost and properties of PHAs are very important. Over the past years, process development and metabolic engineering approaches have been adopted to develop recombinant PHA production strains for improving the strains’ ability to produce PHA, and for changing the PHA structures to obtain better thermal and mechanical properties. This article will describe major past and recent achievements in PHA/PHB.

Coupled heat and mass transfer with double moving interfaces, taking place in a porous half-space, is defined, and exact solutions for the temperature and moisture distributions, as well as the positions of two moving fronts, are obtained. As in Part 2 of the paper, the interest is focused on the case of moderate vacuum environment pressure, where the moisture transfer is due to the result of the vapour concentration and pressure gradients. The temperature of sublimation is assumed to be unknown, whereas the temperature of desorption is still assumed to be known, and the pressure of desorption is determined by a linear assumption among pressures of sublimation, desorption and surface. The Clapeyron equation is incorporated on the sublimation front to close the equation system. The effects of the surface pressure and the permeability on the sublimation and desorption are analysed. Conclusions are drawn on the basis of the results.

A perturbational h4 compact exponential finite difference scheme with diagonally dominant coefficient matrix and upwind effect is developed for the convective diffusion equation. Perturbations of second order are exerted on the convective coefficients and source term of an h2 exponential finite difference scheme proposed in this paper based on a transformation to eliminate the upwind effect of the convective diffusion equation. Four numerical examples including one- to three-dimensional model equations of fluid flow and a problem of natural convective heat transfer are given to illustrate the excellent behavior of the present exponential schemes, the h4 accuracy of the perturbational scheme is verified using double precision arithmetic.

The expression of hTERT gene, encoding the catalytic subunit of telomerase, is a feature of most cancer cells. Changes in the chromatin environment of its promoter and binding of transcriptional factors have been reported in differentiating cells when its transcription is repressed. However, it is not clear whether these changes are directly involved in this repression or only linked to differentiation. In a maturation-resistant acute promyelocytic leukemia (APL) cell line (NB4-LR1), we have previously identified a new pathway of retinoid-induced hTERT repression independent of differentiation. Using a variant of this cell line (NB4-LR1(SFD)), which resists to this repression, we show that although distinct patterns of histone modifications and transcription factor binding at the proximal domain of hTERT gene promoter could concur to modulate its expression, this region is not sufficient to the on/off switch of hTERT by retinoids. DNA methylation analysis of the hTERT promoter led to the identification of two distinct functional domains, a proximal one, fully unmethylated in both cell lines, and a distal one, significantly methylated in NB4-LR1(SFD) cells, whose methylation was further re-enforced by retinoid treatment. Interestingly, we showed that the binding to this distal domain of a known hTERT repressor, WT1, was defective only in NB4-LR1(SFD) cells. We propose that epigenetic modifications targeting this distal region could modulate the binding of hTERT repressors and account either for hTERT reactivation and resistance to retinoid-induced hTERT downregulation.

Castor oil is 90% ricinoleate (12-hydroxy-oleate) and has numerous industrial uses. Components of castor bean (Ricinus communis L.) pose serious problems to processors. We are evaluating two complementary approaches to providing a safe source of castor oil.

PURPOSE Brain metastasis occurs commonly in patients with small-cell lung cancer (SCLC). Herein, we report the efficacy of irinotecan and carboplatin in the treatment of brain metastases from SCLC. In addition, we review the existing data on chemotherapy for brain metastases in SCLC. PATIENTS AND METHODS Eighty patients with metastatic or relapsed SCLC were enrolled in a phase II trial of irinotecan and carboplatin. Patients naive to chemotherapy were treated with irinotecan 200 mg/m 2 and carboplatin AUC of 5, and patients previously treated with chemotherapy received irinotecan 150 mg/m 2 and carboplatin AUC of 5, every 21 days for 6 cycles. RESULTS Among the 80 patients, 15 (19%) presented with brain metastases. An analysis of 14 assessable patients with brain metastases revealed an overall response rate of 65% after 2 cycles of chemotherapy and a median survival of 6 months (range, 1-24 months). Upon review of the literature, 8 studies were identified as having > 10 patients who received chemotherapy for brain metastases from SCLC. Based on these studies, the response rate of brain metastases from SCLC to a variety of chemotherapy and median survival of patients ranged from 22% to 85% and 3 months to 9 months, respectively. CONCLUSION Chemotherapy, including the regimen of irinotecan and carboplatin, is an effective treatment for SCLC brain metastases.

Films made from microbial polyesters polyhydroxybutyrate (PHB) and poly(hydroxybutyrate-co-hydroxyhexanoate) (PHBHHx) were treated by lipases and NaOH solution. The change of the polyester biocompatibility was evaluated by inoculating mouse fibroblast cell line L929 on films of PHB, PHBHHx and their blends. Polylactic acid (PLA) was used as a control. It was found that untreated PHB and PLA films gave a poor support to the growth of L929 cells, viable cell density ranged from 0.1x10(4}) to 0.7x10(4) per ml only. While films of pure PHBHHx and PHB blended with PHBHHx showed improved biocompatibility, viable cell density observed increased from 9.6x10(2) to 6x10(4) on blended films of PHB/PHBHHx in ratios of 0.9/0.1 to 0/1, respectively. This result showed PHBHHx has a better biocompatibility compared with PHB. Films of PHB, PLA and the blends treated with lipases and 1 N NaOH, respectively, showed an improved ability to support cell growth. Biocompatibility of PHB was approximately the same as PLA after the treatment, while PHBHHx and its dominant blends showed improved biocompatibility compared with PLA. The sensitivity of the treatments was reduced when PHBHHx content increased in the PHB/PHBHHx blends. All three lipase treatments demonstrated more biocompatibility increase on all the films compared with the results of NaOH treatment. Scanning electron microscopy showed that PHB films changed its surface from multi-porous to rough non-porous after the lipase or NaOH treatment. While PHBHHx films showed little change after these treatments. The results showed that the polyester surface morphology played an important role in affecting cell attachment and growth on these materials.

OBJECTIVE:To analysis post-treatmental reaction and long-term effect of one-visit root canal therapy by microwave instrument. METHODS:Applying heat effect and the other ones of microwave,the changes of dentine tissue of extracted teeth were observated by SEM examination.48hs and 1 year post-treatmental effect of 47 patients of periapical periodontitis treated by one-visit root canal therapy were observated and compared with the results of 54 cases treated with common method.RESULTS:In the cases of acute and chronic periapical periodontitis microwave applied in root canal therapy can close the openings of dentine,enhance the effects of sterilization and decrease times of treatment.CONCLUSION:Microwave applied in one-visit root canal therapy is competent.

A strain of Bacillus sp. coded JMa5 was isolated from molasses contaminated soil. The strain was able to grow at a temperature as high as 45 degrees C and in 250 g/l molasses although the optimal growth temperature was 35-37 degrees C. Cell density reached 30 g/l 8 h after inoculation in a batch culture with an initial concentration of 210 g/l molasses. Under fed-batch conditions, the cells grew to a dry weight of 70 g/l after 30 h of fermentation. The strain accumulated 25-35%, (w/w) polyhydroxybutyrate (PHB) during fermentation. PHB accumulation was a growth-associated process. Factors that normally promote PHB production include high ratios of carbon to nitrogen, and carbon to phosphorus in growth media. Low dissolved oxygen supply resulted in sporulation, which reduced PHB contents and dry weights of the cells. It seems that sporulation induced by reduced supply of nutrients is the reason that PHB content is generally low in the Bacillus strain.

The main thrust of this work is developing the basis for a mixed formulation of turbulence modeling, combining analytical theories and engineering modeling, which includes second order two-point correlations of velocity and pressure. Related issues such as the different outcomes that stem from differently chosen sets of ensemble averaging, the approximate nature and the advantages and disadvantages of such a formulation, and the choices of closure schemes are addressed.
