Abstract

De acuerdo con un proyecto nacional promovido por el Grupo Nacional Italiano para la Protección contra Riesgos Hidrogeológicos han sido publicados los mapas de vulnerabilidad (1:50.000) de parte del Territorio Italiano, particularmente algunos mapas de Sicilia: Montes Iblei, Moadonie, Sicani y el Monte Etna. Estos mapas se han vuelto documentos públicos para el manejo local del agua subterránea. Algunos fenómenos muy peligrosos han sido reportados en algunas áreas de Sicilia. Intervenciones antrópicas han creado basureros de residuos tóxicos sobre áreas vulnerables. Esta investigación explica las ventajas del uso de mapas de vulnerabilidad en estudio de tipos peligrosos de contaminación para la situación encontrada en el área de Augusta-Priolo, Sicilia, en la cual un complejo petroquímico ha sido instalado. Aspectos hidrogeológicos y petroquímicos son examinados para evaluar las posibilidades de intervención preventiva y de reclamación. Abstract

Piana di Palermo es caracterizada por la presencia de dos acuíferos diferentes: uno somero constituido por calcarenitas y otro profundo en rocas carbonatadas fracturadas. El acuífero calcarenítico presenta un bajo potencial hídrico comparado con el acuífero carbonatico. El comportamiento químico del agua en ambos acuíferos refleja severos procesos como interacción agua-roca, intrusión marina. cambio iónico, etc. La aplicación del método SINTACS mostró que cerca del 80 % del área en estudio puede ser clasificada como de alta a muy alta vulnerabilidad. doi: https://doi.org/10.22201/igeof.00167169p.2004.43.4.850

Abstract

According to a national project promoted by the Italian National Group for the Protection against Hydrogeological Hazards, vulnerability maps (1: 50.000) of part of the Italian National territory have been published, particularly in Sicilia (Iblei Mounts, Madonie, Sicani and Etna). These maps have become a public document for local groundwater management. Dangerous phenomena have been reported in some zones of Sicily. Anthropic interventions have created toxic waste dumps in vulnerable areas. We explain the advantage of vulnerability maps in the study of dangerous types of pollution for the situation in the Augusta Priolo area, Sicilia, in which a petrochemical complex has been installed. Hydrogeological and petrochemical aspects are examined to assess the possibilities of a preventive intervention and reclamation. In the same areas, seawater intrusion is increasing in coastal regions.

Abstract

Piana di Palermo is characterized by the presence of two different aquifers: one shallow, constituted by calcarenites and the other deep in fractured carbonates. The calcarenitic aquifer presents a low potential compared to the carbonatic aquifer. The chemistry of the water in both aquifers reflects water-rock interaction, seawater intrusion, ionic exchange etc. The application of SINTACS method showed that about 80% of the study area can be classified as of high to very high vulnerability.

Abstract

A growing body of evidence shows that Insulin, Insulin Receptor (IR) and IR signaling are involved in brain cognitive functions and their dysfunction is implicated in Alzheimer’s disease (AD) degeneration. Thus, administration of insulin could be a strategy for AD treatment. For this aim we designed, synthesized and characterized a nanogel system (NG) to deliver insulin to the brain, as a tool for the development of a new therapy for AD. Abstract

Crosslinked xyloglucan-poly(vinyl alcohol) based hydrogel films have been synthetized and characterized for wound healing applications. Polyvinyl alcohol (PVA) and glycerol (Gro) were added to have an optimal combination of softness, conformability and resilience. Physical hydrogel have been transformed into permanent covalent hydrogels by reaction with glutaraldehyde (GA). Chemical and physical properties of the films have been analyzed by different technique. Moreover, biocompatibility of a selected formulation was assessed in vitro by investigating different aspects, such as cell viability, oxidative stress, mitochondrial dysfunction and specific stress biomarkers. Partial adhesiveness was demonstrated by performing different attaching assays and phalloidin staining. After interaction with blood the hemocompatibility of the XG-PVA film was evaluated by using a multiparametric approach. Antimicrobial activity of the film against Escherichia coli (E.coli) was tested, and we found that XG-PVA film promotes bacterial retentivity and provides mechanical protection against bacterial infiltration. After loading the film with ampicillin, an inhibitory E. coli growth zone was observed. All together these results indicate that the manufactured film is a promising material to be tested in vivo for wound healing applications

Abstract

Background The independent prognostic impact of diabetes mellitus ( DM ) and prediabetes mellitus (pre‐ DM ) on survival outcomes in patients with chronic heart failure has been investigated in observational registries and randomized, clinical trials, but the results have been often inconclusive or conflicting. We examined the independent prognostic impact of DM and pre‐ DM on survival outcomes in the GISSI ‐HF (Gruppo Italiano per lo Studio della Sopravvivenza nella Insufficienza Cardiaca‐Heart Failure) trial. Methods and Results We assessed the risk of all‐cause death and the composite of all‐cause death or cardiovascular hospitalization over a median follow‐up period of 3.9 years among the 6935 chronic heart failure participants of the GISSI ‐ HF trial, who were stratified by presence of DM (n=2852), pre‐ DM (n=2013), and non‐ DM (n=2070) at baseline. Compared with non‐ DM patients, those with DM had remarkably higher incidence rates of all‐cause death (34.5% versus 24.6%) and the composite end point (63.6% versus 54.7%). Conversely, both event rates were similar between non‐ DM patients and those with pre‐ DM . Cox regression analysis showed that DM , but not pre‐ DM , was associated with an increased risk of all‐cause death (adjusted hazard ratio, 1.43; 95% CI , 1.28–1.60) and of the composite end point (adjusted hazard ratio, 1.23; 95% CI , 1.13–1.32), independently of established risk factors. In the DM subgroup, higher hemoglobin A1c was also independently associated with increased risk of both study outcomes (all‐cause death: adjusted hazard ratio, 1.21; 95% CI , 1.02–1.43; and composite end point: adjusted hazard ratio, 1.14; 95% CI , 1.01–1.29, respectively). Conclusions Presence of DM was independently associated with poor long‐term survival outcomes in patients with chronic heart failure. Clinical Trial Registration URL : http://www.clinicaltrials.gov . Advanced simulation of the stability and handling properties of bicycles requires detailed road–tyre contact models. In order to develop these models, in this study, four bicycle tyres are tested by means of a rotating disc machine with the aim of measuring the components of tyre forces and torques that influence the safety and handling of bicycles. Abstract

Recent studies have suggested that astrocytes share with the macrophages several properties in vitro, among which is the ability to express MHC Class II molecules and to present some antigens to syngeneic primed lymphocytes (MHC-restricted presentation). It has been claimed in the literature that astrocytes cannot start the presentation and cannot express the related MHC Class II molecules if not previously stimulated with gamma IFN. In this paper we report that 2 human GFAP + glioma cell lines, incubated in culture with various soluble antigens for at least 24 h, were able, in the absence of gamma IFN or of activated lymphocytes, to express the MHC Class II and to expose the antigens on their surfaces. Moreover, when a lysosomotropic agent such as chloroquine was added during the incubation, no MHC Class II expression was observed. This last datum suggests that in astrocytes, as is already known in macrophages, the processing of antigens and their assembling with MHC Class II molecules probably involves the lysosomal apparatus.

Abstract

In the Rocca Busambra area (mid-west Sicily, Italy), from November 1999 to July 2002, 23 water points including wells and springs were sampled and studied for their chemical and isotopic compositions. Two rain gauges were also installed at different altitudes, and rainwater was collected monthly to determine the isotopic composition. The obtained results revealed the Rocca Busambra carbonate complex as being the main recharge area on account of its high permeability value. From a chemical view point, two main groups of water can be distinguished: calcium–magnesium–bicarbonate-type and calcium–magnesium–chloride–sulphate-type waters. The first group reflects the dissolution of the carbonate rocks; the second group probably originates from circulation within flyschoid sediments. Three water wells differ from the other samples due to their relatively high Na and K content, which probably is to be referred to a marked interaction with the “Calcareniti di Corleone” formation, which is rich in glauconite [(K, Na)(Fe3+, Al, Mg)2(Si, Al)4O10(OH)2]. In accordance with WHO guidelines for drinking water (2004), almost all the samples collected can be considered drinkable, with the exception of four of them, whose NO3−, F− and Na+ contents exceed the limits. On the contrary, the sampled groundwater studied is basically suitable for irrigation.

Abstract

A fundamental problem in maintaining Web contents accessibility, according to International and national issues, refers to nowadays common scenario of very-short-time dynamics of sites/portals which have an exponential growth of their pages and services, often produced by unskilled Web authors. A process-approach, rather than a product-approach in designing Web applications, services and contents, is the keystone to guarantee low cost effectiveness and efficacy in accessibility supervisory. In such a context, we're going to propose an Accessibility Management Assistant (AMA), as a Web application and suite of integrated instruments to alert and control accessibility for a content management framework and its instantiated content management systems, starting from the authoring/updating/modifying contents phases and continuing to the whole publishing process.

16 páginas, 19 figuras, 10 tablas.-- El PDF es el manuscrito aceptado para su publicación.-- et al., The purpose of this study is to show the results of the Italian research project of national interest (PRIN) launched in 2006 and finished in 2008, concerning the “assessment of groundwater contamination risk by nitrates assessment”. The project verified the IPNOA method for nitrate groundwater contamination risk assessment in four test-sites of Italy. The IPNOA is a parametric index which assesses the potential hazard of nitrate contamination originating from agriculture on a regional scale. The method integrates two categories of parameters: the hazard factors (HF), which represent all farming activities that cause, or might cause, an impact on soil quality in terms of nitrate (use of fertilisers, application of livestock and poultry manure, food industry wastewater and urban sludge), and the control factors (CF) which adapt the hazard factors to the characteristics of the site (geographical location, climatic conditions and agronomic practices). Finally, the Potential Risk Map is obtained by coupling the potential hazard of nitrate pollution (IPNOA) and the aquifer Contamination Vulnerability Map. The project was carried out by five Research Units (RU) from the Politecnico di Torino, Universities of Piacenza, Florence, Naples and Palermo. The geochemistry of groundwaters from the four test-sites was studied to determine the distribution of nitrate, and to evaluate groundwater chemical facies. All the study areas are affected by groundwater nitrate contamination and often by hydrogeochemical peculiarities. In some cases isotopic study, δ18O–NO3δ15N–NO3, allowed to differentiate nitrates of chemical fertilisers from those of biological origin, as well as denitrification processes.

Abstract

A complex approach in characterisation of submarine groundwater discharge (SGD) off south-eastern Sicily comprising applications of radioactive and non-radioactive tracers, direct seepage measurements, geophysical surveys and a numerical modelling is presented. SGD fluxes in the Donnalucata boat basin were estimated by direct seepage measurements to be from 4 to 12Ls(-1), which are comparable with the total SGD flux in the basin of 17Ls(-1) obtained from radon measurements. The integrated SGD flux over the Donnalucata coast estimated on the basis of Ra isotopes was around 60m(3)s(-1) per km of the coast. Spatial variations of SGD were observed in the Donnalucata boat basin, the average (222)Rn activity concentration in seawater varied from approximately 0.1kBqm(-3) to 3.7kBqm(-3) showing an inverse relationship with salinity. The continuous monitoring carried out at the site closest to the coast has revealed an inverse relationship of (222)Rn activity concentration on the tide. The (222)Rn concentrations in seawater varied from 2.3kBqm(-3) during high tides to 4.8kBqm(-3) during low tides, thus confirming an influence of the tide on submarine groundwater discharge. Stable isotopes (delta(2)H and delta(18)O) showed that SGD samples consist up to 50% of groundwater. Geo-electrical measurements showed a spatial variability of the salt/fresh water interface and its complex transformation in the coastal zone. The presented results imply that in the studied Donnalucata site there are at least two different sources of SGD, one superficial, represented by mixed fresh water and seawater, and the second one which originates in a deeper limestone aquifer.

Abstract

The main results of a hydrogeochemical survey carried out during 2002–2003 along the coast of the south-eastern Sicily, which aimed at geochemical characterization of both groundwater chemistry and submarine groundwater discharge (SGD) in the area are presented. A general frame of chemical processes affecting the studied groundwater and SGD point out that most samples fall within the calcite-anhydrite-dolomite field (CAD). The chemical composition of the samples within the CAD triangle is essentially controlled by calcite, dolomite and gypsum dissolution, which are the main minerals of the carbonate rocks hosting the aquifers. An additional process evidenced in this study is a mixing with seawater. Nitrate is the most typical ion significantly disturbed in the groundwater chemistry influenced by agricultural activities. The strong correlation with SO42− indicates that the use of ammonium sulphate fertilisers is widespread in the study area. The K+ vs. NO3− diagram evidences a correlation occurring at lower and higher concentrations, and implies that there is not a common source of both nitrate and potassium, at least on a regional scale. High-phosphate concentration is found in submarine springs along the coast, specifically in the Donnalucata and Avola areas, while its content in inland wells is generally lower. Phosphate is also associated with high-bicarbonate contents in the Donnalucata area, suggesting its possible origin is phosphate-rich carbonate rocks, which are commonly outcropping in the area.

Abstract

Although several attempts at the immunohistochemical characterization of histiocytosis have recently been made there is only one paper which reports a case of cerebral Langerhans cell histiocytosis (LCH) diagnosed by biopsy. This paper presents a bioptically diagnosed case of juvenile histiocytosis. The panel of antibodies used was as follows: anti-S-100, 2 different antibodies to anti-interleukin 2, anti-lysozyme, anti-LEU M1, anti-MAC 387, anti-major histocompatibility complex II and anti-GFAP. Microglia markers--Griffonia simplicifolia and RCA 1 lectins were also utilized. The proliferating cells produced a positive response to S-100, lysozyme and a partially positive response to HLA DR, but responded negatively to MAC 387, LEU M1, lectins, IL2R and GFAP. Our results were compared and analyzed in the light of those obtained by other authors.

Abstract

Altered prolactin and thyrotropin responses to the TRH test in parkinsonian patients are held to indicate an impairment of the tubero-infundibular dopaminergic axis (TIDA). We correlated the plasmatic prolactin (PRL), thyrotropin (TSH) and somatotropin (GH) responses to TRH and bromocriptine + TRH of 12 parkinsonian patients, who had never received anti-parkinsonian drugs, with the severity, the duration, the age of onset and the dopamine-dependence of the motor symptomatology as indicated by the therapeutic response to a six-month oral treatment with bromocriptine. Patients with basal motor impairment over 9 on the Webster Rating Scale (WRS), those with duration of the disease over 24 months and those with onset earlier than 55 years of age showed lower PRL responses than the respectively matched subgroups. Patients showing a therapeutic motor improvement over 50% on the WRS (dopamine-dependent or "responder") showed lower PRL and TSH and higher GH responses than the non-responders. These findings suggest that the TIDA impairment described in Parkinson's disease develops along with the progressive course of the extrapyramidal involvement and is strictly correlated with the dopamine-dependence of the motor impairment.

Abstract

Hallucinations occur fairly frequently in the course of the pharmacological treatment of Parkinson disease. Our aim in this study was to assess first the relation between hallucinations and mental deterioration and second the correlation between the perception disorder and the profile on the Minnesota Multiphasic Personality Inventory (MMPI). Of 304 parkinsonian subjects followed as outpatients at our center 27 (8.88%) had had hallucinations and 17 of these presented marked cognitive deficits (Mini Mental State less than 18) (62.9%) compared with 32 of the other 277 patients (11.5%) (X2 = 55.16, p less than 0.0001). A group of 9 patients who had had hallucinations and 10 controls who had not, all free from marked cognitive deficits, were assessed on the Luria-Nebraska Neuropsychological Battery (LNNB) and on the MMPI. The two groups did not differ significantly in respect of the LNNB but did in respect of the MMPI scales. On this evidence the frequency of mental deterioration is significantly higher in patients who have hallucinated. We discuss the meaning of the MMPI differences both from the biochemical angle and from that of personality factors. These data suggest that the MMPI might be useful for predicting hallucinations in undeteriorated parkinsonian patients on pharmacological therapy.

Abstract

In a double-blind cross-over study, the effects of a subthreshold dose of scopolamine (0.25 mg) on memory were compared in 32 control subjects and 32 parkinsonian patients who were without any sign of intellectual and mnemic impairment. Although the scores of the controls in the memory test battery showed no deterioration after the administration of scopolamine, the same dose resulted in significantly reduced memory performance in parkinsonian patients in two tests which involved the recognition of meaningless drawings. The selective vulnerability of parkinsonian subjects without cognitive impairment to a subthreshold dose of scopolamine suggests the existence of an underlying alteration of central cholinergic transmission. The neuropsychological findings in our study agree with postmortem biochemical data, which showed decreased cortical choline acetyltransferase activity in all parkinsonian patients, suggesting the existence of neuronal compensation in parkinsonian patients who are without cognitive impairment.

Abstract

The results of a double-blind cross-over clinical trial involving 27 patients with classical or common migraine are described to compare the prophylactic effect of the calcium entry-blocker flunarizine with that of pizotifen. Duration of the treatment was two months, with an evening single-dose administration of both drugs. For most parameters, there was no definite difference between flunarizine and pizotifen in migraine prophylaxis. It has been demonstrated previously that pizotifen is an effective drug in migraine prophylaxis, and these results suggest that flunarizine is effective, too. Weight gain as a side effect was less frequent and less severe with flunarizine than with pizotifen; other side effects showed the same incidence with both drugs.

Abstract

The irreversible polarographic reductions of various N -alkyl- N -nitrosoureas were studied from a pH of 1.1–6.7. Below pH 3.2, the limiting current, i lim. , of N -methyl- N -nitrosourea (NNMU) is controlled by diffusion. Above this pH, adsorption or chemical reaction is the electrode-controlling process and buffer effects are observed. Similar results were obtained for N -ethyl- N -nitrosourea (NNEU) and 1,3-dimethyl-1-nitrosourea (SRI-1384), except that the buffer effects were greatly diminished for the latter compound. The acetate buffer effects on i lim. have been quantified and have been attributed either to adsorption of the nitrosourea (NNMU) and the buffer components onto the microelectrode, or to a reaction between these species. The polarographic method offers a sensitive (1 × 10 −6 M ) and reliable (0.91 per cent standard error among days) assay procedure for the nitrosoureas. All of the compounds studied in this series were more easily reduced than the parent methyl compound. A correlation between the change in the half-wave potentials and the Taft substituent constants (δ*) was noted for simple alkyl groups such as methyl, ethyl, butyl, etc. A correlation between the half-wave potentials and the apparent first-order rate constants ( k , sec. −l ) for hydroxyl ion catalyzed solvolysis was noted for several of the compounds studied.

Various spray-congealed SETD-wax products were prepared by pneumatic atomization using six waxes (white wax USP, glyceryl tristearate, carnauba wax, hydrogenated castor oil, cetyl alcohol NF, and glyceryl monostearate), two nozzle sizes, 0.20 and 0.25 cm. (0.08 and 0.10 in.) i.d., and three concentrations of the surfactant sorbitan monostearate (0,1, and 5 percent w/w). The geometric mean diameters and the volume-surface diameters were affected primarily by the wax and the nozzle size used. All of the main effects, i.e. , wax, nozzle size, and concentration of surfactant, had a significant effect on the percent of SETD released in acid pepsin and alkaline pancreatin media. In general, the dissolution rate increased as the concentration of surfactant was increased. Faster dissolution rates were noted with formulations atomized through the smaller of the two nozzles. The most important factor that had an effect on the dissolution behavior of these products was the wax used in a particular formulation. This effect depends upon the following factors: the physical properties of the wax and the drug-wax particles, the chemical composition of wax, and the composition of the dissolution medium.

Abstract

In a double-blind cross-over study, the effects of a subthreshold dose of scopolamine (0.25 mg) on memory were compared in 32 control subjects and 32 parkinsonian patients who were without any sign of intellectual and mnemic impairment. Although the scores of the controls in the memory test battery showed no deterioration after the administration of scopolamine, the same dose resulted in significantly reduced memory performance in parkinsonian patients in two tests which involved the recognition of meaningless drawings. The selective vulnerability of parkinsonian subjects without cognitive impairment to a subthreshold dose of scopolamine suggests the existence of an underlying alteration of central cholinergic transmission. The neuropsychological findings in our study agree with postmortem biochemical data, which showed decreased cortical choline acetyltransferase activity in all parkinsonian patients, suggesting the existence of neuronal compensation in parkinsonian patients who are without cognitive impairment.

Abstract

In order to investigate the neuronal basis of cognitive disorders in Parkinson's disease, the neuropsychological performance of 120 patients with idiopathic Parkinson's disease was analysed in relation to motor symptoms as a function of their response to levodopa. Cognitive impairment was poorly correlated with akinesia and rigidity, symptoms which respond well to levodopa treatment, and was not correlated at all with that part of the patients' motor score that could be improved by the drug. In contrast, strong correlations were found between all neuropsychological test scores and axial symptoms such as gait disorder and dysarthria, which respond little if at all to levodopa treatment. The neuropsychological test scores were also strongly correlated with the motor score of patients estimated when clinical improvement was maximal under levodopa treatment. This score is assumed to represent residual non-dopaminergic motor dysfunctions. The correlations suggest that much of the cognitive impairment in Parkinson's disease results from the dysfunction of non-dopaminergic neuronal systems.

Abstract

To achieve a better understanding of CH and DCH, we used a multidisciplinary approach evaluating both the depression and anxiety scores and the ability of DEX to decrease plasma cortisol levels in patients with these two forms of headache. The Hamilton rating scale for depression, the Zung test for depression and Stai ×2 for anxiety showed scores within the control range in both groups of patients without any statistically significant difference between the groups. The DEX test showed significant cortisol suppression in both groups of patients either at 8 a.m. or at 4 p.m. (after DEX administration, 1 mg orally at 11 p.m. The night before). The results obtained indicate that in CM and in DCH, normal depression and anxiety scores exist in the present of the apparent integrity of the hypothalamic-pituitary-adrenal axis.

Moreover getting in connection high nervous functions with motor activity, authors suppose BC-PL have a primary and direct action on cortical cholinergic structures correlated with attention, learning and memory and consequently a secondary action on motor performances.

Abstract

A neuroendocrinological study was carried out by evaluating plasma prolactin levels after etoperidone i.m. (100 mg) and placebo. Fourteen male inpatients (mean age: 35.36 +/- 11.7 years) with chronic schizophrenia were selected for the study, whose aim was to improve interpretation of the pharmacological activity of etoperidone. The results suggest that etoperidone plays the role of an atypical psychotropic drug since it does not affect prolactin levels. In addition, the drug is devoid of anticholinergic effects, which facilitates its prospective clinical use for medium-term and long-term therapy.

Abstract

In this double-blind clinical trial we compared the prophylactic efficacy of the calcium-entry blocker flunarizine (15 mg nocte) with that of pizotifen (1,5 mg nocte). In 30 patients affected by classical and common migraine. During a two months treatment both drugs showed a good efficacy. Flunarizine tended to more markedly suppress severity of pain and duration of attacks than pizotifen. Daytime drowsiness and weight gain occurred with both drugs; the first side effect was more evident in the group treated with flunarizine, the second one in the group treated with pizotifen.

SYNOPSIS After neuropsychological evaluation, we administered the dexamethasone suppression test to eleven patientssuffering from muscle-contraction headache and to thirteen patients affected by migraine, in order to verify a possiblecorrelation between a chronic pain such as headache and a cortisolemic response to dexamethasone. None of themappeared to be suffering from depressive syndrome; all patients were DST suppressors. These data suggest that thedexamethasone suppression test does not correlate with chronic pain, but with depression, consistent with the highspecificity of this marker test for depression.

Abstract

The ARCO-YBJ experiment, (Astrophysical Radiation Ground-based Observatory at YaugBaJing) is under construction in Tibet, 90 kill to the north of Lhasa. The full coverage approach and the high altitude location allow the study of many physics items in the field of low energy cosmic rays, namely gamma-ray astronomy, diffuse gamma-rays, gamma-ray bursts (GRBs), proton/antiproton ratio, primary proton spectrum and heliosphere physics, In this paper the expected sensitivities of ARGO-YBJ for gamma astronomy and GRB physics are presented and compared with the present experimental techniques and results. The performance of a test-module of similar to 50 m(2) operated on-site is, also discussed.

Abstract

There is currently no prophylactic vaccine available for human immunodeficiency virus (HIV). Research efforts have resulted in improved immunogens that mimic the native envelope (Env) glycoprotein structure. Recently, a novel triple tandem trimer (TTT) platform has been used to generate a plasmid encoding Env immunogen (pBG505-TTT) that expresses only as trimers, making it more suitable for nucleic acid vaccines. We have previously demonstrated that adenosine deaminase-1 (ADA-1) is critical to the T follicular helper (TFH) function and improves vaccine immune responses in vivo . In this study, we demonstrate that co-delivery of plasmid-encoded adenosine deaminase 1 (pADA) with pBG505-TTT enhances the magnitude, durability, isotype switching and functionality of HIV-specific antibodies in a dose-sparing manner. Co-delivery of the molecular immune modulator ADA-1 also enhances HIV-specific T cell polyfunctionality, activation, and degranulation as well as memory B cell responses. Abstract

The glycosylation of IgG plays a critical role during human severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, activating immune cells and inducing cytokine production. However, the role of IgM N-glycosylation has not been studied during human acute viral infection. The analysis of IgM N-glycosylation from healthy controls and hospitalized coronavirus disease 2019 (COVID-19) patients reveals increased high-mannose and sialylation that correlates with COVID-19 severity. These trends are confirmed within SARS-CoV-2-specific immunoglobulin N-glycan profiles. Moreover, the degree of total IgM mannosylation and sialylation correlate significantly with markers of disease severity. We link the changes of IgM N-glycosylation with the expression of Golgi glycosyltransferases. Lastly, we observe antigen-specific IgM antibody-dependent complement deposition is elevated in severe COVID-19 patients and modulated by exoglycosidase digestion. Taken together, this work links the IgM N-glycosylation with COVID-19 severity and highlights the need to understand IgM glycosylation and downstream immune function during human disease., (© 2024. In the field of immunology, a systems biology approach is crucial to understanding the immune response to infection and vaccination considering the complex interplay between genetic, epigenetic, and environmental factors. Significant progress has been made in understanding the innate immune response, including cell players and critical signaling pathways, but many questions remain unanswered, including how the innate immune response dictates host/pathogen responses and responses to vaccines. To complicate things further, it is becoming increasingly clear that the innate immune response is not a linear pathway but is formed from complex networks and interactions. To further our understanding of the crosstalk and complexities, systems-level analyses and expanded experimental technologies are now needed. In this review, we discuss the most recent immunoprofiling techniques and discuss systems approaches to studying the global innate immune landscape which will inform on the development of personalized medicine and innovative vaccine strategies.

48. IgM N-glycosylation correlates with COVID-19 severity and rate of complement deposition.

Abstract

The glycosylation of IgG plays a critical role during human SARS-CoV-2, activating immune cells and inducing cytokine production. However, the role of IgM N-glycosylation has not been studied during acute viral infection in humans. In vitro evidence suggests that the glycosylation of IgM inhibits T cell proliferation and alters complement activation rates. The analysis of IgM N-glycosylation from healthy controls and hospitalized COVID-19 patients reveals that mannosylation and sialyation levels associate with COVID-19 severity. Specifically, we find increased di- and tri-sialylated glycans and altered mannose glycans in total serum IgM in severe COVID-19 patients when compared to moderate COVID-19 patients. This is in direct contrast with the decrease of sialic acid found on the serum IgG from the same cohorts. Moreover, the degree of mannosylation and sialylation correlated significantly with markers of disease severity: D-dimer, BUN, creatinine, potassium, and early anti-COVID-19 amounts of IgG, IgA, and IgM. Further, IL-16 and IL-18 cytokines showed similar trends with the amount of mannose and sialic acid present on IgM, implicating these cytokines' potential to impact glycosyltransferase expression during IgM production. When examining PBMC mRNA transcripts, we observe a decrease in the expression of Golgi mannosidases that correlates with the overall reduction in mannose processing we detect in the IgM N-glycosylation profile. Importantly, we found that IgM contains alpha-2,3 linked sialic acids in addition to the previously reported alpha-2,6 linkage. We also report that antigen-specific IgM antibody-dependent complement deposition is elevated in severe COVID-19 patients. Taken together, this work links the immunoglobulin M N-glycosylation with COVID-19 severity and highlights the need to understand the connection between IgM glycosylation and downstream immune function during human disease., Competing Interests: Declarations Competing Interests The authors have no competing interests to declare.

Abstract

This study investigates the effectiveness of a 6-month parent-mediated early intervention telehealth program for children with an autism spectrum disorder. The participants comprised a total of 27 parents. The findings showed that participation in the program promotes parents' empowerment and reduce parental stress and a general improvement in the parents' ability to stimulate children's learning. Moreover, the study reveals an effect of parents' age in mediating the relationship among the parents' ability, stress, and empowerment levels. These results suggest that an early intervention telehealth program may help parents become aware of how to benefit from interactions with their children, promote their empowerment, and reduce parenting stress., (© 2022. 50. Lipid nanoparticles (LNP) induce activation and maturation of antigen presenting cells in young and aged individuals.

Abstract

Despite the overwhelming success of mRNA-based vaccine in protecting against SARS-CoV-2 infection and reducing disease severity and hospitalization, little is known about the role lipid nanoparticles (LNP) play in initiating immune response. In this report we studied the adjuvantive impact of empty LNP with no mRNA cargo (eLNP) on anti-viral pathways and immune function of cells from young and aged individuals. We found that eLNP induced maturation of monocyte derived dendritic cells by measuring the expression of CD40, CD80, HLA-DR and production of cytokines including IFN-α,IL-6, IFN-γ, IL-12, and IL-21. Flow cytometry analysis of specific dendritic cell subsets showed that eLNP can induce CD40 expression and cytokine production in cDC1, cDC2 and monocytes. Empty LNP (eLNP) effects on dendritic cells and monocytes coincided with induction pIRF7 and pTBK1, which are both important in mitigating innate immune signaling. Interestingly our data show that in response to eLNP stimulus at 6 and 24 hrs, aged individuals have decreased CD40 expression and reduced IFN- γ output compared to young adults. Furthermore, we show that cDC1, cDC2, and CD14 dim CD16 + monocytes from healthy aged individuals have dysregulated anti-viral signaling response to eLNP stimulation as measured by the defect in type I IFN production, phosphorylation of IRF7, TBK-1, and immune function like phagocytosis. These data showed a novel function of eLNP in eliciting DC maturation and innate immune signaling pathways and that some of these functions are impaired in older individuals providing some suggestion of why older individuals (> 65 yrs of age) respond display lower immune responses and adverse events to SARS-CoV-2 mRNA-based vaccines.
