Bacterial endometritis is among the most common causes of subfertility in mares. It has a major economic impact on the equine breeding industry. The sensitivity of detecting uterine microbes using culture-based methods, irrespective of the sample collection method, double-guarded endometrial swab, endometrial biopsy, or uterine low-volume lavage (LVL), is low. Therefore, equine bacterial endometritis often goes undiagnosed. Sixteen individual mares were enrolled, and an endometrial sample was obtained using each method from all mares. After trimming, quality control and decontamination, 3824 amplicon sequence variants were detected in the dataset. We found using 16S rRNA sequencing that the equine uterus harbors a distinct resident microbiome during estrus. All three sampling methods used yielded similar results in composition as well as relative abundance at phyla (Proteobacteria, Firmicutes, and Bacteroidota) and genus (Klebsiella, Mycoplasma, and Aeromonas) levels. A significant difference was found in alpha diversity (Chao1) between LVL and endometrial biopsy, suggesting that LVL is superior at detecting the low-abundant (rare) taxa. These new data could pave the way for innovative treatment methods for endometrial disease and subfertility in mares. This, in turn, could lead to more judicious antimicrobial use in the equine breeding industry.

Wave-transported boulders represent important records of storm and tsunami impact over geological timescales. Their use for hazard assessment requires chronological information on their displacement that in many cases cannot be achieved by established dating approaches. To fill this gap, this study investigated, for the first time, the potential of optically stimulated luminescence rock surface exposure dating (OSL-RSED) for estimating cliff-detachment ages of wave-transported coastal boulders. The approach was tested on calcarenite clasts at the Rabat coast, Morocco. Calibration of the OSL-RSED model was based on samples with rock surfaces exposed to sunlight for ∼ 2 years, and OSL exposure ages were evaluated against age control deduced from satellite images. Our results show that the dating precision is limited for all targeted boulders due to the local source rock lithology which has low amounts of quartz and feldspar. The dating accuracy may be affected by erosion rates on boulder surfaces of 0.02–0.18 mm yr−1. Nevertheless, we propose a robust relative chronology for boulders that are not affected by significant post-depositional erosion and that share surface angles of inclination with the calibration samples. The relative chronology indicates that (i) most boulders were detached from the cliff by storm waves; (ii) these storms lifted boulders with masses of up to ∼ 24 t; and (iii) the role of storms in the formation of boulder deposits along the Rabat coast is more significant than previously assumed. Although OSL-RSED cannot provide reliable absolute exposure ages for the coastal boulders in this study, the approach has large potential for boulder deposits composed of rocks with larger amounts of quartz or feldspar and less susceptibility to erosion.

Tectonically active regions are characterized by complex landscapes comprising soils with heterogeneous physicochemical properties. Spatial variability of nutrient sources enhances landscape biodiversity and creates heterogeneous habitats potentially attractive for animals and humans. In this study, we analyze the role of geological processes in the distributions of soil nutrients in the southern Kenya Rift, a key region in the interpretation of early human-landscape interactions. Our aim is to determine how spatial variations in rock chemistry, as well as topographic gradients and localized zones of rock fracturing from tectonic faulting determine the distributions of plant-available soil nutrients in soils. We hypothesize that present-day soil nutrient levels reflect the long-term chemical and geomorphological characteristics of the landscape and underlying parent material, and that regions with high nutrient availability occur along pathways correlating with locations of hominin fossil sites. Analyses of 91 topsoil samples from the main geological units show that Calcium (Ca) deficiencies predominately occur in shallow soils developed on trachytic volcanic rocks and granitic gneisses, while high Ca levels are associated with basaltic parent material and sedimentary deposits of mixed sources. XRF analysis of rock samples confirms that CaO levels in trachyte rocks are significantly lower than those in basalts, and Ca mobilization in basalt is more effective than in trachyte. Along two toposequences in densely faulted basaltic and trachytic rocks, we observed slope dependent soil nutritional gradients and a systematic increase of the concentrations of Ca, Mg and SOC in topsoils of colluvial sediments downslope of active normal faults. Known hominin sites in the region are located either along corridors of long-term Ca availability or at short-term nutrient hotspots potentially related to active CO2 degassing along active fault zones. This implies a strategic advantage of Ca-rich regions for hominin subsistence strategies, such as provision of predictable constraints on the distribution and mobility of grazing animals in complex tectonic landscapes. Our study implies that geological processes impact nutrient distributions in the southern Kenya Rift. Results of this study have further implications for understanding the role of soils in the interpretation of hominin-landscape interactions in the early stages of human evolution.

The work presented in this pilot study aimed to identify potential risk factors associated with bovine periodontitis development. Bovine periodontitis is a multifactorial polymicrobial infectious disease for which the aetiopathogenesis and risk factors are not fully understood. From cattle slaughtered in an abattoir in Scotland, 35 dental arcades with periodontal lesions and 40 periodontally healthy arcades were selected over seven visits for study. Multivariable logistic regression analysis was used to evaluate the association between periodontitis and the independent variables, gender, age and breed. For every increase in year of age, cattle were 1.5 times more likely to have periodontitis. A graphical analysis indicated that within the limits of this study, we could not detect any major influence of breed on the age-effect. Although logistic regression analysis demonstrated that periodontitis lesions are more prevalent with increasing age of cattle the underlying mechanisms remain unclear. It is likely that periodontitis is an important cause of oral pain in older cattle and can contribute to reduced productivity/performance. Further studies with a larger sample size are necessary to elucidate the associations between potential risk factors and periodontitis in cattle and to define its effects on animal welfare and productivity.

Rational number proficiency is predictive of later mathematics achievement, especially for rigorous mathematics courses, such as Algebra I. Students with learning disabilities (LD) in mathematics struggle to make adequate progress on rational number concepts and skills that lay the foundation for successfully completing secondary mathematics courses. However, recent research has demonstrated that utilizing effective instructional design, such as the use of a number line with systematic and explicit instruction, can result in academic progress for students with LD in mathematics. This article presents teachers with a four-step teaching sequence utilizing the number line and effective strategies for teaching students with LD in mathematics to introduce fraction-to-decimal relationships in upper elementary, a prerequisite for middle school mathematics.

Abstract With increasingly intense marine heatwaves affecting nearshore regions, foundation species are coming under increasing stress. To better understand their impacts, we examine responses of critical, habitat-forming foundation species (macroalgae, seagrass, corals) to marine heatwaves in 1322 shallow coastal areas located across 85 marine ecoregions. We find compelling evidence that intense, summer marine heatwaves play a significant role in the decline of foundation species globally. Critically, detrimental effects increase towards species warm-range edges and over time. We also identify several ecoregions where foundation species don’t respond to marine heatwaves, suggestive of some resilience to warming events. Cumulative marine heatwave intensity, absolute temperature, and location within a species’ range are key factors mediating impacts. Our results suggest many coastal ecosystems are losing foundation species, potentially impacting associated biodiversity, ecological function, and ecosystem services provision. Understanding relationships between marine heatwaves and foundation species offers the potential to predict impacts that are critical for developing management and adaptation approaches.

Aim. We investigated the relationship between sex (genetic/biological) and gender (environmental/ cultural) factors in relation to adolescent tobacco smoking. Methods. A representative sample of 11,582 students from French secondary public schools participated in the study by completing a self-administered, standardised questionnaire. Results. Using the WHO classification for smoking in the youth, 15.6% of the adolescents were regular smokers, 7.7% occasional smokers, 17.9% experimental smokers and 4.8% ex-smokers, with no statistically significant gender difference. Taking non-smoking as a reference, puberty had a much greater effect on the likelihood of being a regular smoker [OR=18.0 (95% Confidence Interval: 9.6- 32)] than of being an experimental/occasional smoker [OR=3.7 (2.9-4.6)] among girls. For boys, the effect of puberty was not as great [OR=4.7 (3.5-6.5)] for regular vs. [OR=2.1 (1.8-2.5)] for experimental/occasional smokers). Similarly, illicit drug use had a larger effect on the likelihood of being regular smoker vs. non-smoker [OR=15.0 (12.0-20.0) in boys and 12 (8.8-16.0) in girls] than of being experimental/occasional smoker vs. a non-smoker [OR=4.8 (3.7-6.1) and 2.9 (2.1-3.9) respectively]. Other factors related to regular smoking were exposure to passive smoking and regular alcohol consumption. Living with both parents was a protective factor for life and regular smoking in both genders. Conclusions. Our results show that influential factors of sex-related (puberty), gender-specific (environmental tobacco smoking, alcohol consumption, drug abuse) or sex/gender (regular sexual intercourse) are related to the smoking behaviour in French adolescents.

Although Indonesia has recorded good performance in its national economic development, especially in the agriculture sector during the Covid-19 pandemic, the impact of the pandemic on farming and food systems has not been evaluated yet. This study has evaluated the resilience of the two dominant existing farming systems in West Timor, i.e. (i) wetland farming system and (ii) dryland farming system. This research aims to understand the resilience of farming after the Covid-19 pandemic and to develop strategic policies that could be adopted to increase the resilience of the farming system in West Timor. A quantitative analysis using the Structural Equation Modelling (SEM) was employed to evaluate the relationship and impact of the following seven generic aspects: labour movement, sustainability, economy, socio- culture, output markets, input markets, farming system resilience, and 27 reflective indicators. The analysis shows that dryland farming systems are more resilient than wetland farming systems. It might be understood from the size of the regression coefficient, as the impact of exogenous construct variables of the environment, socioculture, input, and output on the resilience of dryland farming systems is more significant than on wetlands. Economic performance rather than labour movement factors will create better resilience of farming systems for wetland or dryland after the Covid-19 pandemic. Finally, the economic recovery process and the ongoing input supply mechanism after the Covid-19 pandemic have increased the resilience of the dryland food system more than the resilience of the wetland farming system.

ABSTRACT Background Back pain is a huge global problem. For some people, the pain is so severe that they feel the need to present to an emergency department (ED). Our aim was to explore patient and staff perspectives for the development of a digital care pathway (DCP) for people with back pain who have presented to ED, including acceptability, barriers and facilitators. Methods We used a descriptive phenomenology approach using semi‐structured interviews with patient and staff participants at a tertiary hospital. Interviews were transcribed and data codes were developed using inductive thematic analysis. Themes were discussed between researchers until consensus was achieved. Results A total of 16 interviews were carried out, half of which involved patient participants. We identified three major themes: (i) expectations and experiences of staff and patients with low back pain in ED; (ii) a digital care pathway can empower patients and support clinicians in providing care; and (iii) acceptability, barriers, facilitators and recommendations of engaging with a DCP to track the trajectory of back pain. Each theme was further categorised into subthemes. Conclusion Introducing a DCP was perceived as acceptable and beneficial by patients and staff. Both groups were aware of the potential participant burden if surveys were too long. Introducing a DCP could be a valuable adjunct to current management care models, providing a standardised source of education with the potential for individualised tracking and monitoring. The design and development of a DCP will need to consider reported facilitators and address perceived barriers for engagement. Patient or Public Contribution This project sought insights from patients and staff about a digital care pathway. This forms the first step of patient and consumer consultation before implementing a digital care pathway. All consumers were offered the opportunity to review their responses and our interpretation.

The efficacy of mathematics interventions for students with mathematics difficulties is a critical concern in educational research, particularly in light of the increasing demand for effective instructional strategies. While previous meta-analyses have explored various aspects of mathematics interventions, our study uniquely focuses on a broad range of intervention characteristics and their impacts on students' mathematics proficiency. This meta-analysis reviewed 223 reports comprising 286 studies, encompassing data from approximately 56,477 participants. We analyzed several intervention characteristics as potential moderators, including: (1) grade level; (2) group size; (3) content area; and (4) dosage. We also examined study level characteristics, including: (1) research design; (2) fidelity; (3) year of study; (4) type of measure; and (5) study quality. Our findings revealed significant variability in intervention effectiveness based on targeted content areas, with fraction interventions demonstrating a pronounced impact on the effectiveness of interventions for students with MD. The study also underscored the challenges in educational research, marked by heterogeneity in type of measure and study designs. This comprehensive analysis contributes to the field by providing nuanced insights into the effectiveness of mathematics interventions, aiding educators and policymakers in developing strategies that effectively address the diverse needs of students with MD in various educational settings.

Bed bugs: (Hemiptera: Cimicidae) are a globally distributed hematophagous pest that routinely feed on humans. Unlike many blood-sucking arthropods, they have never been linked to pathogen transmission in a natural setting, and despite increasing interest in their role as disease vectors, little is known about the viruses that bed bugs naturally harbor. Here, we present a global-scale survey of the bed bug RNA virosphere. We sequenced the metatranscriptomes of 22 individual bed bugs (Cimex lectularius and Cimex hemipterus) from 8 locations around the world. We detected sequences from two known bed bug viruses (Shuangao bedbug virus 1 and Shuangao bedbug virus 2) which extends their geographical range. We identified three novel bed bug virus sequences from a tenui-like virus (Bunyavirales), a toti-like virus (Ghabrivirales), and a luteo-like virus (Tolivirales). Interestingly, some of the bed bug viruses branch near to insect-transmitted plant-infecting viruses, opening questions regarding the evolution of plant virus infection. When we analyzed the viral sequences by their host's collection location, we found unexpected patterns of geographical diversity that may reflect humans’ role in bed bug dispersal. Additionally, we investigated the effect that Wolbachia, the primary bed bug endosymbiont, may have on viral abundance and found that Wolbachia infection neither promotes nor inhibits viral infection. Finally, our results provide no evidence that bed bugs transmit any known human pathogenic viruses.

Abstract A 64‐year‐old obese gentleman attended for further evaluation of ongoing dyspnoea in the context of a previous diagnosis of moderate COPD treated with dual long‐acting bronchodilators. A cardiopulmonary exercise test (CPET) was performed, which demonstrated reduced peak work and oxygen consumption with evidence of dynamic hyperinflation, abnormal gas exchange and ventilatory limitation despite cardiac reserve. The CPET clarified the physiological process underpinning the patient's dyspnoea and limiting the patient's activities. This, in turn, helped the clinician tailor the patient's management plan.

BackgroundGiven its putative roles in mediating prosocial behavior, attachment bonds, and stress physiology, oxytocin modulation has been hypothesized to be a biological correlate of the salubrious effects of meditation practice. Here we investigated the effects of a month-long silent meditation retreat on changes in oxytocin, and the related hormone and vasopressin, in relation to psychosocial changes in attachment style, anxiety, personality measures, and feelings of social connectedness with fellow meditators.MethodsPlasma oxytocin and vasopressin and self-report questionnaires were measured in retreat participants (n = 28) at the beginning of, and 3 weeks into, a residential meditation retreat. Control participants (n = 34), who were similar in age, gender, and meditation experience, were also assessed across a 3-week interval. Linear mixed effects models were used to assess outcomes.ResultsThe retreat group showed a small but significant decrease in oxytocin compared to controls who showed no change. In the retreat group, higher openness to experience at Time 1 predicted greater reductions in oxytocin during the retreat, and lower oxytocin at Time 2 was related to stronger feelings of personal connection with fellow meditators. The changes in oxytocin were not related to attachment style or anxiety. Vasopressin decreased over time across both groups, suggesting no specific effect of retreat.ConclusionThese preliminary findings suggest that meditation training in the context of a silent residential retreat may reduce circulating levels of oxytocin. We interpret this finding from multiple theoretical perspectives, discussing key measurement limitations and proposing future study designs that may help to differentiate the effects of different meditation practices and contexts on oxytocin signaling.

Disease of the peripheral (or small) airways is fundamental in asthma, being closely related to symptoms (or lack of control of them), airway hyperresponsiveness, spirometric abnormalities, risk of loss of control, or exacerbations and inflammation. Current technology now allows routine measurement of peripheral airway function. Having a working concept of peripheral airways disease in asthma is arguably very useful to clinicians and beneficial to patients because it allows a more comprehensive assessment of asthma severity (rather than just symptoms alone, which is the norm), tracking of progress or deterioration, and assessing response to treatment. Oscillometry is a sensitive way to monitor the peripheral airways, whereas multiple breath nitrogen washout parameters are excellent measures of future risk. In the longer term, physiologic measurements will be crucial in research to define causes and find new disease-modifying treatments.

Abstract Wildlife conservation involves making management decisions with incomplete knowledge of ecological relationships. Efforts to augment foraging resources for the endangered Mexican long‐nosed bat (Leptonycteris nivalis) are progressing despite limited knowledge about the species' foraging behavior and requirements. This study aimed to understand L. nivalis responses to floral resource availability, focusing on individual agave‐ and local‐scale characteristics influencing visitation rates to flowering agaves. We observed bat visitation at 62 flowering agaves around two roosts in northeast Mexico on 46 nights in the summers of 2017 and 2018. We found visitation rate had positive relationships with two agave‐scale characteristics: the number of umbels with open flowers and the lower vertical position on the stalk of those umbels (i.e., earlier phenological stages of flowering). However, these factors exhibited strong negative interaction: with few umbels with open flowers, the position of flowering umbels had little effect on visitation rate, but when umbels with open flowers were abundant, visitation rate was more strongly related to the lower flowering umbel position. We also found relationships between visitation rate and two local‐scale characteristics: negative for the density of flowering conspecifics within 30 m of the focal agave and positive for the density of dead standing agave stalks within 30 m. Our findings suggest opportunities to augment foraging resources for L. nivalis in ways that are consistent with their foraging behavior, including: increasing the supply of simultaneously blooming flowers by planting agave species that tend to have more umbels with simultaneously open flowers; planting multiple species of agaves with different flowering times to increase the availability of agaves with open flowers on lower‐positioned umbels throughout the period when bats are present in the region; planting agaves in clusters; and keeping dead standing agave stalks on the landscape. Our study points to useful management strategies that can be implemented and monitored as part of an adaptive management approach to aid in conservation efforts.

A significant problem in Arithmetic Unit design and particularly for systolic arrays remains the speed attainable in achieving high speed addition. The root of the problem is carry propagation and a method is presented which is relatively independent of word length. The problem is addressed by the description of a suggested radical design involving a hybrid optical and electronic approach. The method of carry chain addition through pass gates is explained and a suggested implementation utilising Fabry-Perott resonators, optical waveguides and voltage controlled couplers is described. The design is suitable for n-stage modular expansion.

Riboswitch architectures that involve the binding of a single ligand to a single RNA aptamer domain result in ordinary dose-response curves that require approximately a 100-fold change in ligand concentration to cover nearly the full dynamic range for gene regulation. However, by using multiple riboswitches or aptamer domains in tandem, these ligand-sensing structures can produce additional, complex gene control outcomes. In the current study, we have computationally searched for tandem riboswitch architectures in bacteria to provide a more complete understanding of the diverse biological and biochemical functions of gene control elements that are made exclusively of RNA. Numerous different arrangements of tandem homologous riboswitch architectures are exploited by bacteria to create more ‘digital’ gene control devices, which operate over a narrower ligand concentration range. Also, two heterologous riboswitch aptamers are sometimes employed to create two-input Boolean logic gates with various types of genetic outputs. These findings illustrate the sophisticated genetic decisions that can be made by using molecular sensors and switches based only on RNA.

La utilización de los estudios de montaña requiere de narrativas integradoras para la geoalfabetización sobre paisajes socioecológicos productivos y motiva más investigaciones transdisciplinares en el campo de la montología. Concebimos este artículo como la confluencia de la experiencia individual y el razonamiento colectivo hacia la formación de grupos de investigación sinérgicos que se ocupan de la ciencia de montaña convergente, para hacer avanzar la montología a un nuevo nivel, mediante el cual el pensamiento innovador sobre la ciencia de la sustentabilidad y el desarrollo regenerativo incorpora propuestas alternativas para el mantenimiento, la mejora, o regeneración de las condiciones de vida de los paisajes de montaña. Buscamos utilizar este marco contemporáneo de sustentabilidad y restauración ecológica como el ímpetu para comprender mejor las relaciones de la naturaleza y la cultura, desde una perspectiva transdisciplinar, en montañas habitadas que operan en cuatro dimensiones (largo, ancho, alto y tiempo). El artículo está orientado a potenciar la transversalidad de temáticas en torno a las montañas como sistemas socioecológicos productivos, en una nueva disciplina académica institucionalizada y convergente. Concluimos con un llamado a un desarrollo regenerativo, sustentable y consiliente en las montañas del mundo.

The effects of pre-meal whey protein consumption on acute food intake and subsequent energy balance measured over 48-h was investigated in males of healthy-weight (HW) or living with overweight and obesity (OV/OB). On two separate trial days, following a controlled breakfast (09:00) and lunch (13:00), 12 HW and 12 OV/OB males consumed either whey protein (20 g) or flavoured water beverages (16:40), and ad libitum test meal (17:00). A controlled 48-h assessment of energy intake and expenditure was used to determine any compensatory behaviour. Test meal energy intake reduced 15.9 % in HW (P = 0.003), and 17.8 % in OV/OB (P = 0.005) following whey protein, compared to placebo. We report no between-group differences and no changes in compensatory behaviour. A small dose of whey protein reduces energy intake at the next meal, without upregulating compensatory behaviours in both HW and OV/OB males. However, chronic effects on body composition and weight loss remain to be elucidated.

Introduction The aim of this randomised feasibility trial is to determine the feasibility of conducting an adequately powered randomised controlled trial (RCT) investigating the efficacy of prefabricated contoured foot orthoses in people with hip osteoarthritis (OA). The secondary aims of the trial are to compare the effect of prefabricated contoured foot orthoses to a flat shoe insert comparator on outcomes of hip-related pain, physical activity and quality of life. We hypothesise that the demand, implementation, acceptability and practicality of foot orthoses as a treatment option for people with hip OA will be deemed feasible, informing the development of an adequately powered RCT to evaluate the efficacy and long term outcomes.Methods and analysis We will recruit 28 people with hip OA who will be randomised to receive either prefabricated contoured foot orthoses or flat shoe inserts to use for a 6-week period. Both groups will receive standardised education on hip OA and physical activity. The study’s primary outcome is the feasibility domains of demand, implementation, acceptability and practicality. The secondary outcomes include the change in Hip Osteoarthritis Outcome Score-12, Patient Health Questionnaire-9, Brief Fear of Movement Scale for OA, Physical activity accelerometry and the Physical Activity Questionnaire-short form. Descriptive statistics will be used to describe feasibility outcomes with limited efficacy analysis used for the secondary outcomes. Linear mixed models will be used to analyse between-group differences at 6 weeks, with baseline values used as covariates, treatment allocation as a fixed factor and participant as a random factor.Ethics and dissemination This trial has been approved by the La Trobe University Human Research Ethics Committee (HEC20427), St. Vincent’s Hospital Melbourne, Human Research Ethics Committee (HREC 266/20) and Northern Health Research Governance (NH-2021-292862). The results will be disseminated via a peer-reviewed journal and presented at international conferences.Trial registration number NCT05138380.

Abstract Asthma with irreversible or fixed airflow obstruction (FAO) is a severe clinical phenotype that is difficult to treat and is associated with an accelerated decline in lung function and excess morbidity. There are no current treatments to reverse or prevent this excessive decline in lung function in these patients, due to a lack of understanding of the underlying pathophysiology. The current paradigm is that FAO in asthma is due to airway remodeling driven by chronic inflammation. However, emerging evidence indicates significant and critical structural and functional changes to the lung parenchyma and its lung elastic properties in asthma with FAO, suggesting that FAO is a ‘whole lung’ problem and not just of the airways. In this Perspective we draw upon what is known thus far on the pathophysiological mechanisms contributing to FAO in asthma, and focus on recent advances and future directions. We propose the view that structural and functional changes in parenchymal tissue, are just as (if not more) important than airway remodeling in causing persistent lung function decline in asthma. We believe this paradigm of FAO should be considered when developing novel treatments.

Recently, “Technical standards for respiratory oscillometry” was published, which reviewed the physiological basis of oscillometric measures and detailed the technical factors related to equipment and test performance, quality assurance and reporting of results. Here we present a review of the clinical significance and applications of oscillometry. We briefly review the physiological principles of oscillometry and the basics of oscillometry interpretation, and then describe what is currently known about oscillometry in its role as a sensitive measure of airway resistance, bronchodilator responsiveness and bronchial challenge testing, and response to medical therapy, particularly in asthma and COPD. The technique may have unique advantages in situations where spirometry and other lung function tests are not suitable, such as in infants, neuromuscular disease, sleep apnoea and critical care. Other potential applications include detection of bronchiolitis obliterans, vocal cord dysfunction and the effects of environmental exposures. However, despite great promise as a useful clinical tool, we identify a number of areas in which more evidence of clinical utility is needed before oscillometry becomes routinely used for diagnosing or monitoring respiratory disease.

A hallmark of adaptive immunity is CD4 T cells’ ability to differentiate into specialized effectors. A long-standing question is whether T cell receptor (TCR) signal strength can dominantly instruct the development of Th1 and T follicular helper (Tfh) cells across distinct infectious contexts. We characterized the differentiation of murine CD4 TCR transgenic T cells responding to altered peptide ligand lymphocytic choriomeningitis viruses (LCMV) derived from acute and chronic parental strains. We found that TCR signal strength exerts opposite and hierarchical effects on the balance of Th1 and Tfh cells responding to acute versus persistent infection. TCR signal strength correlates positively with Th1 generation during acute but negatively during chronic infection. Weakly activated T cells express lower levels of markers associated with chronic T cell stimulation and may resist functional inactivation. We anticipate that the panel of recombinant viruses described herein will be valuable for investigating a wide range of CD4 T cell responses.

Microstates reflect transient brain states resulting from the synchronous activity of brain networks that predominate in the broadband EEG. There has been increasing interest in how the functional organization of the brain varies across individuals, or the extent to which its spatiotemporal dynamics are state dependent. However, little research has examined within and between-person correlates of microstate temporal parameters in healthy populations. In the present study, neuroelectric activity recorded during eyes-closed rest and during simple visual fixation was segmented into a time series of transient microstate intervals. It was found that five data-driven microstate configurations explained the preponderance of topographic variance in the EEG time series of the 374 recordings (from 187 participants) included in the study. We observed that the temporal dynamics of microstates varied within individuals to a greater degree than they differed between persons, with within-person factors explaining a large portion of the variance in mean microstate duration and occurrence rate. Nevertheless, several individual differences were found to predict the temporal dynamics of microstates. Of these, age and gender were the most reliable. These findings not only suggest that the rich temporal dynamics of whole-brain neuronal networks vary considerably within individuals, but that microstates appear to differentiate persons based on trait individual differences. Rather than focusing exclusively on between-person differences in microstates as measures of brain function, researchers should turn their attention towards understanding the factors contributing to within-person variation.
