Fenugreek (Trigonella foenum-graecum L.) is a pharmaceutically significant aromatic crop with health benefits linked to its phytochemicals. This article aims to overview progress in using emerging technologies to extract its bioactive compounds and extraction mechanisms. Also, the trends in the applications of this herb in the food industry and its therapeutical effects were explained. Fenugreek's flavor is the primary reason for its applications in the food industry. At the same time, it has antimicrobial, antibacterial, hepatoprotection, anticancer, lactation, and antidiabetic effects. Phytochemicals responsible for these effects include galactomannans, saponins, alkaloids, and polyphenols. Besides, data showed that emerging technologies boost fenugreek extracts' yield and biological activity. Among these, ultrasound (55.6%) is the most studied technology, followed by microwave (37.0%), cold plasma (3.7%), and combined approaches (3.7%). Processing conditions (e.g., treatment time and intensity) and solvent (type, ratio, and concentration) are significant parameters that affect the performance of these novel extraction technologies. Extracts obtained by sustainable energy-saving emerging technologies can be used to develop value-added health-promoting products. Guar is a typical legume cash crop native to India. Guar beans are ever-rising in the market share at present. Among them, the guar gum of its seed endosperm has been mainly used in the oil exploration industry. Water-based fracturing fluid can be formulated to increase the permeability of oil-bearing strata during oil production. In addition, guar gum is also widely used in paper making, textiles, food, spices, drugs, as well as mining and metallurgical industries. The nutrient-rich green seeds of guar beans are edible for food vegetables, due to a large amount of protein and a small amount of fat. The plant can be used as fodder and green manure. However, the low supply of guar gum on the market cannot fully meet the broad market space. Only a small number of planting areas of guar beans are suitable for the cultivation of guar beans in the food industry. Among them, Xinjiang located in the northwestern inland arid zone is very similar to the unique geographic conditions for the production and cultivation of guar beans. The temperature and heat conditions are suitable for the origin of guar, rich in the light and heat resources under the soil and water environment, with the high temperature difference between day and night, due to the scarce rainfall and high evaporation. But the relatively small region of guar bean planting still remains so far. Therefore a broad prospect can be expected to optimize the water and nitrogen management for the high crop yield and efficient use of water and nitrogen. This study aims to explore the water and nitrogen mode suitable for drip irrigation of guar beans under film in the Xinjiang area of China. Four irrigation levels were set: W1:1170 m³/hm², W2:1 530 m³/hm², W3:1 890 m³/hm², W4: 2 250 m³/hm²; Two levels of nitrogen application were: N1:30 kg/hm², N2:50 kg/hm². A systematic investigation was made to clarify the effects of water and nitrogen interaction on the growth index, yield, water, and nitrogen use efficiency, as well as the quality of guar bean during drip irrigation under film. The results showed that the coupling effect of water and nitrogen shared a significant effect on the yield, irrigation water use efficiency, and partial factor productivity of nitrogen fertilizer (W3>W4>W2>W1). There was an increase in the plant height, stem diameter, dry matter accumulation, yield, water use efficiency, galactomannan, soluble sugar, and polysaccharide content of Guar bean under the N2 level, compared with the N1 level. Only the partial productivity of nitrogen fertilizer decreased slightly. Principal component and membership function analysis showed that the optimal treatment was achieved in the irrigation amount of 1 890 m³/hm² and the nitrogen application rate of 50 kg/hm²(W3N2). Carob (Ceratonia siliqua) seeds are rich in diverse bioactive compounds, including galactomannan, β-sitosterol, unsaturated fatty acids and proteins with bioactive peptides in their sequence. This study delineates the compositional characterization of six carob seed flour types derived from different production processes, providing valuable insights for designing tailored nutraceutical products based on desired bioactive compound profiles. Our analysis indicated that a higher purity of galactomannan resulted in a greater mannose/galactose ratio, which increased the linearity of the galactomannan polymer and could enhance interchain interaction, thereby increasing aggregation capacity. A higher viscosity could potentially increase the capacity of galactomannan to create satiety and lower cholesterol levels. Among the different tested flours, those whose main compound was the endosperm were optimal for containing high galactomannan content, whereas those derived from the germ were ideal for having high concentrations of fatty acids (i.e., oleic and linoleic acids) and β-sitosterol. The presence of these lipids in carob flours could offer cardiovascular and metabolic health benefits, contributing synergistically. Additionally, flours that contain the germ have beneficial peptides included in proteins like glycinin and conglutin with potential anticholesterolemic and antidiabetic properties. Natural biopolymers become increasingly attractive as bio-based alternatives to petrol-based rheological modifiers, especially in personal care applications. However, many polysaccharides exhibit undesired properties in cosmetic applications such as limited viscosifying characteristics, unpleasant sensory properties, or incompatibility with certain formulation compounds. Here, a comprehensive rheological analysis of non-decorated acetan-like heteroexopolysaccharides derived from two Kozakia baliensis strains was performed in selected surfactant formulations. The results were compared to native xanthan gum and a genetically engineered xanthan variant, Xan∆ gumFGL , which lacks any acetyl- and pyruvyl moieties and whose rheological properties are unaffected by saline environments. All four polysaccharides displayed a highly similar rheological performance in the non-ionic surfactant lauryl glucoside, while the rheological properties differed in amphoteric and anionic surfactants cocamidopropyl betaine and sodium laureth sulfate due to minor changes in side chain composition. Polysaccharide precipitation was observed in the presence of the cationic surfactant. Nevertheless, the native heteroexopolysaccharide derived from K. baliensis LMG 27018 shows significant potential as a salt-independent rheological modifier compared to the genetically engineered Xan∆ gumFGL variant. In addition, blends of heteroexopolysaccharides from K. baliensis and several galactomannans displayed synergistic effects which were comparable to native xanthan gum-galactomannan blends. Pulmonary invasive fungal infections (pIFI) disproportionately affect patients with haematological malignancies (HM). Establishing a rapid and accurate diagnosis of pIFI is challenging. Multiple guidelines recommend diagnostic testing of invasive fungal infections but lack consensus and may contribute to inconsistent diagnostic approaches. To identify key diagnostic challenges and review metagenomic sequencing data. PubMed, professional consortium, and scientific society websites search to identify relevant, published, evidence-based clinical guidelines within the past 5 years. PubMed searchs for papers describing clinically relevant novel diagnostic technologies. Current guidelines for patients with HM and suspected pIFI recommend chest computed tomography imaging and specimen testing with microscopic examination (including calcofluor white stain, histopathology, cytopathology, etc.), Aspergillus galactomannan, β-D-glucan, PCR, and culture, each with certain limitations. Emerging real-world data support the adjunctive use of metagenomic sequencing-based tests for the timely diagnosis of pIFI. High-quality evidence from robust clinical trials is needed to determine whether guidelines should be updated to include novel diagnostic technologies. Trials should ask whether the combination of powerful novel diagnostics, such as pathogen-agnostic metagenomic sequencing technologies in conjunction with conventional testing can optimize the diagnostic yield for all potential pIFI pathogens that impact the health of patients with HM. Galactomannans are polysaccharides commonly extracted from the endosperm of various leguminous seeds, exhibiting variations in their man/gal ratio based on their botanical origin. These gums are highly valued for their biodegradability and excellent stability. However, their applications have been limited due to their high viscosity and molecular weight. In this study, enzymatic and ultrasound depolymerisation of galactomannans (guar gum, locust bean gum, and unconventional Neltuma ruscifolia seed gum) was carried out, resulting in hydrolysates, which were subsequently freeze-dried and characterised. The hydrolysis process leads to a decrease in viscosity and molecular weight, accompanied by a noticeable increase in polydispersity through enzymatic treatment. Thin-layer chromatography (TLC) analysis indicates the generation of at least eight-membered subunits. Fourier-transform infrared (FT-IR) spectroscopy analysis demonstrates no significant changes in functional groups following ultrasound hydrolysis. Furthermore, in agreement with TLC results, enzymatic hydrolysates display partial reactivity towards non-enzymatic browning (NEB), yielding mostly intermediate products with limited browning development. Guar gum is a kind of natural galactomannan extracted from the endosperm of seeds of guar beans. It has excellent water solubility thickening, gelling, and film-forming properties, as well as non-toxicity, good biocompatibility, and biodegradability. Moreover, its physical and chemical properties can be significantly improved and its application performance can be efficiently enhanced through molecular modification, chemical cross-linking, graft copolymerization, oxidation, and other modification treatments by reacting with the hydroxyl groups and glycosidic bonds in its molecular structure. Therefore, guar gum and its modified products have been found wide applications in industrial fields such as daily chemicals, papermaking, food processing, oil exploitation, biomedicine and water treatment. In this review, the name, source, chemical structure, main physicochemical properties and functions of guar gum along with its extraction technologies and modification methods were briefly presented. C-Glycosyl amino acids are a group of C-glycosides in which a carbohydrate molecule is attached to the side chain or backbone of the amino acid via a C–C bond. Despite the numerous methods that have been developed for their synthesis, the C-glycosyl α-amino acids and their oligomers are relatively unexplored. In this work, we presented a protocol for the synthesis of oligomers containing alternating C-glycosyl α-amino acids and proteinogenic α-amino acids. The methodology is based on the modification of α-acyloxyamides obtained from the Passerini reaction using α- D -galactopyranose- and α- L -sorbofuranose-derived aldehydes as C-glycosyl donors. Coix seed is a nutrient-rich food and traditional Chinese medicine with anti-inflammatory and analgesic properties. Polysaccharides from Coix seed have been rarely investigated for structure and activities. In this study, the analgesic and anti-inflammatory effects were investigated in vivo and in vitro. The results showed that Coix seed had a significant influence on reducing the number of writhing, increasing the pain threshold and alleviating the swelling degree caused by acute inflammation. Column chromatography was used to obtain two active compounds of Coix seed. Compound 1 was (1→6)-α-glucan with a molecular weight of 6.81 × 105 Da. The chemical connection of compound 2 was as follows: α-Frup (2→ [1)-α-Glcp (6]5→1)-α-Glcp (4→1)-α-Glcp, which was isolated in Coix seed for the first time. LPS-induced inflammation in RAW264.7 cells was well inhibited by compounds. Natural antimicrobial peptides (AMPs) and enzymes (AMEs) are promising non-antibiotic candidates against antimicrobial resistance but suffer from low efficiency and poor stability. Here, we develop peptide nanozymes which mimic the mode of action of AMPs and AMEs through de novo design and peptide assembly. Through modelling a minimal building block of IHIHICI is proposed by combining critical amino acids in AMPs and AMEs and hydrophobic isoleucine to conduct assembly. Experimental validations reveal that IHIHICI assemble into helical β-sheet nanotubes with acetate modulation and perform phospholipase C-like and peroxidase-like activities with Ni coordination, demonstrating high thermostability and resistance to enzymatic degradation. The assembled nanotubes demonstrate cascade antifungal actions including outer mannan docking, wall disruption, lipid peroxidation and subsequent ferroptotic death, synergistically killing >90% Candida albicans within 10 min on disinfection pad. These findings demonstrate an effective de novo design strategy for developing materials with multi-antimicrobial mode of actions. Natural antimicrobial peptides and enzymes are good candidates for application but suffer from low stability. Aim: Using atenolol as a model active pharmaceutical and galactomannan from the seeds of Trigonella foenumgraecum as a mucoadhesive polymer, an effort was made to develop a gastroretentive bioadhesive floating delivery system to improve the residence time in the stomach. Materials and Methods: Different formulations were made with Hydroxypropyl Methyl Cellulose (HPMC) and galactomannan. Galactomannan was chosen because of its exceptional swelling properties in aqueous environments. In this investigation, citric acid and sodium bicarbonate were utilized as gas-producing components. Pre- and postcompression evaluations were performed on the manufactured tablets. The floating properties as well as the atenolol release pattern were investigated. Results: The extracted polymer was characterized using various techniques and found to be similar to literature. The results from GC analysis showed a residual acetone content of 0.127 ppm, indicating that the extracted polymer is safer to use in the formulation of gastroretentive tablets. The physicochemical properties of the manufactured tablets were satisfactory in terms of swelling index, release characteristics, and buoyancy pattern. All of the manufactured batches had adequate in vitro buoyancy. The gastroretentive tablet exhibited axial and radial swelling throughout the in vitro buoyancy test. According to the testing, the formulation remained buoyant for approximately 8 to 12 hr. Conclusion: According to the results of the evaluation, the manufactured tablet has a pleasing look, is heat stable, and is therapeutically efficient. Rambutan seeds are by-products generated from fruit-processing factories; the leftover seeds are buried in landfills, generating methane emissions. This work aimed to extract polysaccharides (POLS) from rambutan seeds by using subcritical water extraction (SWE). The effects of defatting pretreatment and operating parameters in SWE were investigated using a Box–Behnken design. The results show that defatting pretreatment significantly enriched the POLS yield, while it had no significant effect on the total sugar content. Using the desirability approach, the suitable feedstock for SWE was defatted rambutan seeds. The maximum desirability of 0.86 was found at a temperature range of 145–150 °C, an extraction time of 15 min, and a liquid–solid ratio of 10:1. The POLS yield and total sugar content were in the range of 52.33–55.63 g/100 g feedstock and 83.37–87.45 g/100 g POLS, respectively. The extracted POLS had an equivalent molecular weight of 413.70 kDa that could be used as an extender in plant-based products. In conclusion, the defatting pretreatment of rambutan seeds not only improved the POLS yield obtained via SWE but also generated additional lipids that could be utilized as an unconventional source of specialty fat. The cell surface of Cryptococcus neoformans is covered by a thick capsular polysaccharide. The capsule is the most important virulence factor of C. neoformans; however, the complete mechanism of its biosynthesis is unknown. The capsule is composed of glucuronoxylomannan (GXM) and glucuronoxylomannogalactan (GXMGal). As GXM is the most abundant component of the capsule, many studies have focused on GXM biosynthesis. However, although GXMGal has an important role in virulence, studies on its biosynthesis are scarce. Herein, we have identified a GT31 family β-(1 → 3)-galactosyltransferase Ggt2, which is involved in the biosynthesis of the galactomannan side chain of GXMGal. Comparative analysis of GXMGal produced by a ggt2 disruption strain revealed that Ggt2 is a glycosyltransferase that catalyzes the initial reaction in the synthesis of the galactomannan side chain of GXMGal. The ggt2 disruption strain showed a temperature-sensitive phenotype at 37°C, indicating that the galactomannan side chain of GXMGal is important for high-temperature stress tolerance in C. neoformans. The widening of possible areas of practical uses for zero-valent tellurium nanoparticles (Te0NPs) from biomedicine to optoelectronic and thermoelectric applications determines the actuality of the development of simple and affordable methods for their preparation. Among the existing variety of approaches to the synthesis of Te0NPs, special attention should be paid to chemical methods, and especially to "green" approaches, which are based on the use of precursors of tellurium in their powder bulk form and natural galactose-containing polysaccharides—arabinogalactan (Ar-Gal), galactomannan—(GM-dP) and κ-carrageenan (κ-CG) acting as ligands stabilizing the surface of the Te0NPs. The use of basic-reduction system "N2H4 H2O-NaOH" for preliminary activation of bulk-Te and Ar-Gal, GM-dP and κ-CG allowed us to obtain in aqueous medium a number of stable nanocomposites consisting of Te0NPs stabilized by the polysaccharides' macromolecules. By varying the precursor ratio, different morphologies of nanoparticles were obtained, ranging from spheres at a polysaccharide/Te ratio of 100:1 to rice-like at a 10:1 ratio. The type (branched, combed, or linear sulfated) of polysaccharide and its molecular weight value determined the size of the nanoparticles. Thus, the galactose-containing polysaccharides that were selected for this study may be promising renewable materials for the production of water-soluble Te0NPs with different morphology on this basis. Galactomannans are polysaccharides obtained from legume seed extraction. They present a chemical structure consisting of D-mannose chains linked by glycosidic bonds and galactose branches. The main focus lies in their use as thickeners in the food industry, aimed at improving the dielectric properties of food during heating processes within the radiofrequency and microwave ranges. In this work, the prepared galactomannan samples were electrically analyzed through impedance spectroscopy, which is a powerful physical technique. From the experimental measurements, the dielectric permittivity and loss tangent of the galactomannan solutions were analyzed and the electrical modulus formalism was used to study the dielectric relaxations. Crude galactomannans exhibited higher values of permittivity, conductivity, and losses compared to purified galactomannans. Increasing ethanol concentration in galactomannan purification causes an increase in the permittivity and conductivity of galactomannan solutions. In a 1% solution, at 1 kHz, the permittivity increased from 378.56 to 538.09, while in the 2% solution, this increase was from 656.22 to 1103.24. Regarding the conductivity, at the same frequency, the increase was from 1.6 × 10−3 to 3.3 × 10−3 Ω−1m−1 and from 2.9 × 10−3 to 5.5 × 10−3 Ω−1m−1, respectively. The rise of the ethanol concentration in galactomannan purification led to a decrease in the relaxation time, from 448.56 to 159.15 μs and from 224.81 to 89.50 μs in the solution with 1 and 2%, respectively. Several studies have shown that lactic acid bacteria (LAB) fermentation plays an important role in the development and application of soy-based products and could increase their nutritional values and content of bioactive substances. Lactiplantibacillus plantarum LP95 has shown in previous studies to be a promising candidate as a probiotic and microbial culture in fermented soymilk production. In this study, the suitability of Lp. plantarum LP95 as a functional starter culture in tofu production was verified, with a focus on evaluating the isoflavone and amino acid content in the final product after 21 days of storage at 4 °C. Lp. plantarum LP95 was found able to ferment monosaccharides and disaccharides naturally present in soymilk (D-glucose, D-fructose, D-galactose and D-sucrose) after 24 h while leaving the content of galacto-oligosaccharides (stachyose and raffinose) unaffected. The rich amino acid profile of tofu has undergone some quantitative but not qualitative variations compared to the soy milk used, highlighting the high nutritional value of the product obtained. The enzymatic activity of Lp. plantarum LP95 allowed the release of isoflavone aglycones (daidzein, glycitein and genistein) that were not further metabolized during the fermentation phase of soymilk and during storage at 4 °C. In addition, Lp. plantarum LP95 showed a good viability after 21 days of tofu storage at 4 °C. The results obtained highlighted the suitability of this LAB strain to be used as a microbial culture capable of playing a pro-technological role in the production of fermented tofu, which has good nutritional and functional properties. Abstract Curcumin obtained from Curcuma longa is a polyphenol that has been used in traditional Indian and Chinese medicine for centuries. It was also incorporated into foods through encapsulation to include its health benefits in daily diet. The present study aimed to microencapsulate curcumin through complex coacervation using lysozyme (LSZ) and carboxymethylated tara gum (CMTG) and then incorporate it into bread. Curcumin encapsulation through complex coacervation was performed using different core/wall ratios and total biopolymer concentration. The complexes of LSZ and CMTG formed at pH 5.0 exhibited electrostatic attraction with high affinity and an excellent curcumin encapsulation efficiency of 74.86%. The microcapsules protected curcumin during the oral and gastric phases, with an average release of 77% to 94% in the intestinal phase. After gastrointestinal digestion, the bioaccessibility of the encapsulated curcumin was approximately 47%. The bread in which the curcumin was encapsulated exhibited antioxidant activity (FRAP and DPPH+), with 93.9% curcumin preservation after cooking and 31% bioaccessibility. Therefore, the microcapsules containing curcumin formed after the complex coacervation of lysozyme and carboxymethylated tara gum may be used for the enrichment of bakery products such as bread.

The lectin pathway (LP) of complement mediates inflammatory processes linked to tissue damage and loss of function following traumatic brain injury (TBI). LP activation triggers a cascade of proteolytic events initiated by LP specific enzymes called MASPs (for Mannan-binding lectin Associated Serine Proteases). Elevated serum and brain levels of MASP-2, the effector enzyme of the LP, were previously reported to be associated with the severity of tissue injury and poor outcomes in patients with TBI. To evaluate the therapeutic potential of LP inhibition in TBI, we first conducted a pilot study testing the effect of an inhibitory MASP-2 antibody (α-MASP-2), administered systemically at 4 and 24 h post-TBI in a mouse model of controlled cortical impact (CCI). Treatment with α-MASP-2 reduced sensorimotor and cognitive deficits for up to 5 weeks post-TBI. As previous studies by others postulated a critical role of MASP-1 in LP activation, we conducted an additional study that also assessed treatment with an inhibitory MASP-1 antibody (α-MASP-1). A total of 78 mice were treated intraperitoneally with either α-MASP-2, or α-MASP-1, or an isotype control antibody 4 h and 24 h after TBI or sham injury. An amelioration of the cognitive deficits assessed by Barnes Maze, prespecified as the primary study endpoint, was exclusively observed in the α-MASP-2-treated group. The behavioral data were paralleled by a reduction of the lesion size when evaluated histologically and by reduced systemic LP activity. Our data suggest that inhibition of the LP effector enzyme MASP-2 is a promising treatment strategy to limit neurological deficits and tissue loss following TBI. Our work has translational value because a MASP-2 antibody has already completed multiple late-stage clinical trials in other indications and we used a clinically relevant treatment protocol testing the therapeutic mechanism of MASP-2 inhibition in TBI. Yeast, crucial in beer production, holds great potential owing to its ability to transform into a valuable by-product resource, known as brewer's spent yeast (BSY), with potentially beneficial physiological effects. This study aimed to compare the composition and soluble polysaccharide content of Brewer's spent yeast with those of cultured yeast strains, namely Saccharomyces cerevisiae (SC) and S. boulardii (SB), to facilitate the utilization of BSY as an alternative source of functional polysaccharides. BSY exhibited significantly higher carbohydrate content and lower crude protein content than SC and SB cells. The residues recovered through autolysis were 53.11%, 43.83%, and 44.99% for BSY, SC, and SB, respectively. Notably, the polysaccharide content of the BSY residue (641.90 μg/mg) was higher than that of SC (553.52 μg/mg) and SB (591.56 μg/mg). The yields of alkali-extracted water-soluble polysaccharides were 33.62%, 40.76%, and 42.97% for BSY, SC, and SB, respectively, with BSY comprising a comparable proportion of water-soluble saccharides made with SC and SB, including 49.31% mannan and 20.18% β-glucan. Guar gum is a specialty galactomannan that serves as an abundant, renewable and sustainable feedstock. Nevertheless, its films cannot be used for sustained drug delivery due to certain drawbacks such as susceptibility towards microbial contamination, high swelling rate, non-porous structure, and brittleness. Therefore, functionalization of galactomannan has been achieved by crosslinking with malonic acid, a green, water-soluble, simple organic dicarboxylic acid with minimum chain length possessing natural plasticizer properties using a green chemistry approach. Interestingly, malonic acid also acts as an auto catalyst during acylation and subsequent crosslinking. The reaction conditions were optimized by varying the concentrations of malonic acid (0.5%, 1.5%, and 2% w/v), temperature (55 °C, 100 °C, 120 °C, and 140 °C) and time (5, 10, and 15 min) to introduce 1,3-propane dioates in the galactomannan chains resulting in flexible crosslinked films of galactomannans. Furthermore, the effect of low and high curing temperatures on crosslinking has been investigated by the analysis of the degree of crosslinking for the first time. The mechanism of crosslinking with a dicarboxylic acid has also been proposed. Detailed analysis of films has been achieved by NMR (1D & 2D), FTIR, XRD, TGA and DSC. Furthermore, curcumin has been loaded in these crosslinked films and kinetic studies via the Korsmeyer--Peppas model demonstrate that curcumin has a non-Fickian diffusion controlled release (DCR) mechanism. The antibacterial, antifungal and antioxidant studies show that the prepared formulation has potential utility in sustained drug delivery and other biomedical applications. The aim of this study was to develop microcapsules containing juniper or black pepper essential oils, using a combination of faba bean protein and chia seed polysaccharides (in ratios of 1:1, 1:2, 2:1). By synergizing these two polymers, our goal was to enhance the efficiency of essential oil microencapsulation, opening up various applications in the food industry. Additionally, we aimed to investigate the influence of different polymer mixing ratios on the properties of the resulting microcapsules and the course of the complex coacervation process. To dissolve the essential oils and limit their evaporation, soybean and rapeseed oils were used. The powders resulting from the freeze-drying of coacervates underwent testing to assess microencapsulation efficiency (65.64–87.85%), density, flowability, water content, solubility, and hygroscopicity. Additionally, FT-IR and DSC analyses were conducted. FT-IR analysis confirmed the interactions between the components of the microcapsules, and these interactions were reflected in their high thermal resistance, especially at a protein-to-polysaccharide ratio of 2:1 (177.2 °C). In the absence of naturally available galactofuranose-specific lectin, we report herein the bioengineering of GalfNeoLect, from the first cloned wild-type galactofuranosidase (Streptomyces sp. strain JHA19), which recognises and binds a single monosaccharide that is only related to nonmammalian species, usually pathogenic microorganisms. We kinetically characterised the GalfNeoLect to confirm attenuation of hydrolytic activity and used competitive inhibition assay, with close structural analogues of Galf, to show that it conserved interaction with its original substrate. We synthetised the bovine serum albumin-based neoglycoprotein (GalfNGP), carrying the multivalent Galf units, as a suitable ligand and high-avidity system for the recognition of GalfNeoLect which we successfully tested directly with the galactomannan spores of Aspergillus brasiliensis (ATCC 16404). In this work, an enzyme cocktail with β-mannanase as the main activity was immobilized on epoxy resin foams filled with fibers from annatto capsules. The catalytic system was characterized by SEM, FTIR, and a mechanical crush resistance test. The behavior of the pH and temperature for the hydrolysis of the locust bean gum were also studied. With the same substrate and with respect to the free enzyme, the immobilized enzyme showed an activity retention of 79.61%. Its operational stability in ten reuse cycles did not show any statistically significant loss of activity. This catalytic system was used to study the preferential release of MOS of two to five degrees of polymerization from mannan present in dried and ground açaí seeds, which were not subjected to any other pretreatment. Using an experimental response surface design, the predicted quadratic models for the M2–M5 MOS content were obtained and they fit well with the experimental data, predicting a production range between 0.435 and 20 g/L of MOS (M2–M5). In addition, the production reached about 12 g/L under the optimized conditions. These results indicate that the used foamed epoxy resin supports and immobilization methodology are suitable for catalyzing the hydrolysis of mannan from açaí seeds. [ABSTRACT FROM AUTHOR]

Biopolymers are gaining increased attention in the industry due to their unique characteristics, including being cost-effective, environmentally friendly, and biodegradable. It is also worth noting that natural polymers can be obtained in significant quantities from various renewable sources, whereas synthetic polymers are derived from non-renewable petroleum resources. Enhanced oil recovery (EOR) using biopolymers such as galactomannan, xanthan, welan gum, acacia gum, carboxy methyl cellulose, and corn starch is a developing trend and is projected to replace synthetic polymers (hydrophobically associated polyacrylamides) in the nearby future. The choice of polymers to be utilized in EOR technologies should be based on their cost and availability in addition to their functional properties. Biopolymers in enhanced oil recovery serve to enhance the mobility ratio by increasing the viscosity of displacing fluid and reducing permeability. Even though biopolymers have a tough structure and long polysaccharide chains that make them suitable for enduring severe reservoir conditions, they are highly susceptible to bacterial destruction. In this comprehensive review, we have illustrated the different techniques used to enhance the performance of biopolymers (xanthan gum, guar gum, and starch) in enhanced oil recovery and create new composites that can overcome the challenges faced by these biopolymers under reservoir conditions. We have found that the most famous and favorable techniques used in this approach are, grafting copolymerization, nanocomposites functionalization, amphiphilic style, and hydrogel formation. The review also discussed some other biopolymers (carboxy methyl cellulose, welan gum, and acacia gum) that can be utilized to improve oil recovery and evaluated how widely they have been applied in this field. In this review, we have addressed several important issues (knowledge gaps) that have not been covered in recent studies. We have also provided recommendations and prospects for the successful future implementation of these composites in the EOR field. Characterizing molecular interactions at the microscopic level remains difficult and, therefore, represents a key target to better understand macromolecule and biomacromolecule behaviors in solution, alone, or in mixtures with others. Therefore, accurate characterization in liquid media, especially in aqueous solutions, without causing any perturbation of the system in which they are studied, is quite difficult. To this purpose, the present paper describes an innovative methodology based on fluorescence spectrophotometry. Two molecular fluorescent probes, namely 8-anilino-1-naphtalenesulfonic acid (ANS) and 2-benzofuryl-3-hydroxy-4(1H)-quinolone (3HQ-Bf), were selected to characterize, respectively, the dipole-dipole interactions and hydrophobic micro-domains, for the first one, and hydrogen bonding, for the second. As a support to study molecular interactions, xanthan, galactomannan, and corresponding mixtures of these substances which are well known to exhibit a synergy of interactions in well-defined mixture conditions were chosen. Once the methodology was set up, the existence of the three types of interactions in these systems was demonstrated, thus allowing the elucidation of the mechanisms of interactions at the molecular scale. Introduction. Studying the functional properties of practically important enzymes, in particular α-galactosidases, is a necessary step towards obtaining stable preparations with high selectivity of action to be used in the food industry for biodegradation and modification of raw materials. Materials and methods. Gel filtration and ion exchange chromatography on TSK-gels were used to purify the microbial α-galactosidase. To determine the activity and specificity of the enzymatic action, p-nitrophenyl-α-D-galactopyranoside, raffinose, stachyose, and galactomannan were used as substrates. Results and discussion. α-Galactosidase with a specific activity of 49.1 IU/mg was isolated from the culture liquid of Penicillium restrictum and purified to a homogeneous state. The molecular weight of the enzyme was 17 kDa according to the data of gel filtration on Sepharose 6B. The optimum pH for the enzyme action was 4.0, and the thermal optimum was 60 °C. Enzyme showed high stability in the temperature ranged from 40 to 60 °C. The half-life of α-galactosidase from Penicillium restrictum at 70°C increased in 2-fold in the presence of 20-40% glycerol and 2.3-fold in the presence of 4 M sorbitol. Km for the hydrolysis of p-nitrophenyl-α- galactopiranoside, raffinose, stachyose, and galactomannan were 0.9 mM, 2.2 mM, 2.4 mM, and 3.8 mM, respectively. It was shown that the rate and efficiency of hydrolysis of galactose-containing natural substrates by Penicillium restrictum α-galactosidase decreased with increasing their molecular weight. The enzyme was completely inhibited in the presence of 10-3 M Ag+, Hg2+, Ca2+, and Cd2+. α-Galactosidase from Penicillium restrictum was resistant to action of proteinase K, pronase E, and trypsin. Conclusions. The high thermal stability and efficiency of α-galactosidase from Penicillium restrictum for the modification of galactooligosaccharides suggests its broadrange applicability for food and animal feed processing. The ability of recombinant, SARS-CoV-2 Spike (S) protein to modulate the production of two COVID-19 relevant, pro-inflammatory cytokines (IL-6 and IFN-γ) in PBMC cultures of healthy, pre-COVID-19 subjects was investigated. We observed that cytokine production was largely and diversely modulated by the S protein depending on antigen or mitogen stimulation, as well as on the protein source, insect (S-in) or human (S-hu) cells. While both proteins co-stimulated cytokine production by polyclonally CD3-activated T cells, PBMC activation by the mitogenic lectin Concanavalin A (Con A) was up-modulated by S-hu protein and down-modulated by S-in protein. These modulatory effects were likely mediated by the S glycans, as demonstrated by direct Con A-S binding experiments and use of yeast mannan as Con A binder. While being ineffective in modulating memory antigenic T cell responses, the S proteins and mannan were able to induce IL-6 production in unstimulated PBMC cultures and upregulate the expression of the mannose receptor (CD206), a marker of anti-inflammatory M2 macrophage. Our data point to a relevant role of N-glycans, particularly N-mannosidic chains, decorating the S protein in the immunomodulatory effects here reported. Background Invasive fungal infections (IFI) mainly caused by aspergillus species are one of the leading causes of death in patients undergoing immunosuppressive therapy for hematological malignancies. The presence of galactomannan in bronchoalveolar lavage fluid (BAL GM) is an important diagnostic marker. Some of the factors affecting the BAL GM are still unknown. Antifungal treatment administered before or after BAL also affect BAL GM results. Methods To investigate the effect of BAL GM timing on the diagnosis of IFE, 100 patients receiving immunosuppressive therapy with hematological malignancy at the Uludağ University Faculty of Medicine, Hematology Department, and underwent BAL over a 3-year period with the suspicion of IFE as well as 127 BAL procedures of these patients were examined. Results There were 70 patients who started antifungal therapy before BAL and 30 patients who did not. BAL GM was found positive in 33 (47.1%) of the 70 patients who received antifungal therapy compared to 22 (73.3%) of the 30 patients who did not receive antifungal therapy. There was a significant difference between the two groups in terms of BAL GM positivity (p = 0.016). Subsequently, 127 BAL procedures of these 100 patients were evaluated. When the second, third, and subsequent BAL procedures of the same patients were included in the study, BAL GM was positive in 41 (46.6%) of the 88 procedures in patients who received treatment before BAL and in 25 (64.1%) of the 39 procedures in patients who did not receive treatment before BAL. The rate of BAL GM positivity did not differ between groups (p = 0.068). Conclusions The balance between reducing the risk by initiating early antifungal therapy and maximizing the diagnostic value of BAL GM should be evaluated individually for each patient. Beta-mannans are insoluble plant polysaccharides with beta-1,4-linked mannose as the backbone. We used three forms of this polysaccharide, namely, pure mannan, glucomannan, and galactomannan, to enrich haloarchaea, which have the ability to utilize mannans for growth. Four mannan-utilizing strains obtained in pure cultures were closely related to each other on the level of the same species. Furthermore, another strain selected from the same habitats with a soluble beta-1,4-glucan (xyloglucan) was also able to grow with mannan. The phylogenomic analysis placed the isolates into a separate lineage of the new genus level within the family Natrialbaceae of the class Halobacteria. The strains are moderate alkaliphiles, extremely halophilic, and aerobic saccharolytics. In addition to the three beta-mannan forms, they can also grow with cellulose, xylan, and xyloglucan. Functional genome analysis of two representative strains demonstrated the presence of several genes coding for extracellular endo- beta-1,4-mannanase from the GH5_7 and 5_8 subfamilies and the GH26 family of glycosyl hydrolases. Furthermore, a large spectrum of genes encoding other glycoside hydrolases that were potentially involved in the hydrolysis of cellulose and xylan were also identified in the genomes. A comparative genomics analysis also showed the presence of similar endo-beta-1,4-mannanase homologs in the cellulotrophic genera Natronobiforma and Halococcoides. Based on the unique physiological properties and the results of phylogenomic analysis, the novel mannan-utilizing halolarchaea are proposed to be classified into a new genus and species Natronoglomus mannanivorans gen. nov., sp. nov. with the type strain AArc-m2/3/4 (=JCM 34861=UQM 41565). Summary: Galactomannan oligosaccharides (GMOS) have been designated as the second generation of prebiotics. This study aimed to systematically investigate the prebiotic effects of GMOS. GMOS were obtained by the hydrolysis of guar gum (5 g L−1) using β‐mannanase (160 U g−1) for 4 h, with the molecular weight significantly decreased to 665 Da. The prebiotic activity of GMOS was verified, which displayed a great proliferation effect on Lactobacillus and Bifidobacterium. After in vitro faecal fermentation, GMOS greatly improved the production of total short‐chain fatty acids (43.62 mM), especially butyric acid (7.44 mM). Meanwhile, the relative abundance of several beneficial bacteria, including Megasphaera, Lactobacillus, and Bifidobacterium, was greatly improved to 41.15%. Besides, the total relative abundance of several typical harmful bacteria decreased to 37.80%, indicating that GMOS can regulate the composition of intestinal flora. Nurr1 is a member of the orphan nuclear receptor family NR4A (nuclear receptor subfamily 4 group A) that modulates inflammation in several cell lineages, both positively and negatively. Macrophages are key regulators of inflammatory responses, yet information about the role of Nurr1 in human macrophages is scarce. Here we examined Nurr1 expression and activity in steady state and activated human macrophages. Pro- and anti-inflammatory macrophages were generated in vitro by culture of blood monocytes with granulocyte/macrophage colony-stimulating factor (GM-CSF) and macrophage colony-stimulating factor (M-CSF), respectively. Nurr1 expression was predominant in macrophages with the pro-inflammatory phenotype. Nurr1 activation with the agonists 1,1-bis(3ʹ-indolyl)-1-(p -chlorophenyl) methane (C-DIM12) or isoxazolo-pyridinone 7e (IP7e) did not globally modify the polarization status of pro-inflammatory macrophages, but they decreased their production of TNF, IL-1β, IL-6, IL-8, IL-12 p40, CCL2, IFN-β, and reactive oxygen species, with variable potencies. Conversely, Nurr1 deficient macrophages increased the expression of transcripts encoding inflammatory mediators, particularly that of IL6 , IFNB1 , and CCL2. Mechanistically, endogenous Nurr1 interacted with NF-κB p65 in basal conditions and upon lipopolysaccharide (LPS)-mediated activation. C-DIM12 stabilized those complexes in cells exposed to LPS and concurrently decreased NF-κB transcriptional activity and p65 nuclear translocation. Expression of high levels of Nurr1 was associated with a subset of dermal macrophages that display enhanced levels of TNF and lower expression of the anti-inflammatory marker CD163L1 in skin lesions from patients with bullous pemphigoid (BP), a chronic inflammatory autoimmune blistering disorder. These results suggest that Nurr1 expression is linked with the pro-inflammatory phenotype of human macrophages, both in vivo and in vitro , where it may constitute a brake to attenuate the synthesis of inflammatory mediators. [Display omitted] Degradation of polysaccharides based on lytic polysaccharide monooxygenases (LPMOs) has received considerably interest in the environment and energy fields since 2010. With the rapid development of nanozymes in various fields, it is highly desirable but challenging to develop LPMO-like nanozymes with high specificity and satisfied activity. Here, a defective copper-cobalt binuclear Prussian blue analogue (CuCoPBA) nanozyme was developed via a facile and ingenious methodology based on single histidine (His). For the first time, His-CuCoPBA nanozyme was found to exhibit LPMO-like activity with H 2 O 2 as a cosubstrate at room temperature and neutral pH, which can efficiently catalyze the degradation of galactomannans selectively. Significantly, the high degradation activity at pH 10 expands the application of Fenton-like nanozymes in alkaline condition. Singlet oxygen (1O 2), as a main reactive intermediate, plays a crucial role in the galactomannan degradation catalyzed by His-CuCoPBA nanozyme. Both control experimental and density functional theory (DFT) results indicate Cu - N x His contributes to the efficiently and selectively catalytic activity of His-CuCoPBA nanozymes by emulating the binding and catalytic sites of LPMOs. The present work not only represents a fundamental breakthrough toward degradation of polysaccharide based on nanozyme, but also contributes to understanding the catalytic mechanism of natural Cu-dependent LPMOs. α-Galactosidase is an important exoglycosidase belonging to the hydrolase class of enzymes, which has therapeutic and industrial potential. It plays a crucial role in hydrolyzing α-1,6 linked terminal galacto-oligosaccharide residues such as melibiose, raffinose, and branched polysaccharides such as galacto-glucomannans and galactomannans. In this study, Actinoplanes utahensis B1 was explored for α-galactosidase production, yield improvement, and activity enhancement by purification. Initially, nine media components were screened using the Plackett–Burman design (PBD). Among these components, sucrose, soya bean flour, and sodium glutamate were identified as the best-supporting nutrients for the highest enzyme secretion by A. Utahensis B1. Later, the Central Composite Design (CCD) was implemented to fine-tune the optimization of these components. Based on sequential statistical optimization methodologies, a significant, 3.64-fold increase in α-galactosidase production, from 16 to 58.37 U/mL was achieved. The enzyme was purified by ultrafiltration-I followed by multimode chromatography and ultrafiltration-II. The purity of the enzyme was confirmed by Sodium Dodecyl Sulphate–Polyacrylamide Agarose Gel Electrophoresis (SDS-PAGE) which revealed a single distinctive band with a molecular weight of approximately 72 kDa. Additionally, it was determined that this process resulted in a 2.03-fold increase in purity. The purified α-galactosidase showed an activity of 2304 U/mL with a specific activity of 288 U/mg. Background: Chitin, the main form of aminated polysaccharide in nature, is a biocompatible, polycationic, and antimicrobial biopolymer used extensively in industrial processes. Despite the abundance of chitin, applications thereof are hampered by difficulties in feedstock harvesting and limited structural versatility. To address these problems, we proposed a two-step cascade employing carbohydrate oxidoreductases and amine transaminases for plant polysaccharide aminations via one-pot reactions. Using a galactose oxidase from Fusarium graminearum for oxidation, this study compared the performance of CvATA (from Chromobacterium violaceum) and SpATA (from Silicibacter pomeroyi) on a range of oxidized carbohydrates with various structures and sizes. Using a rational enzyme engineering approach, four point mutations were introduced on the SpATA surface, and their effects on enzyme activity were evaluated. Results: Herein, a quantitative colorimetric assay was developed to enable simple and accurate time-course measurement of the yield of transamination reactions. With higher operational stability, SpATA produced higher product yields in 36 h reactions despite its lower initial activity. Successful amination of oxidized galactomannan by SpATA was confirmed using a deuterium labeling method; higher aminated carbohydrate yields achieved with SpATA compared to CvATA were verified using HPLC and XPS. By balancing the oxidase and transaminase loadings, improved operating conditions were identified where the side product formation was largely suppressed without negatively impacting the product yield. SpATA mutants with multiple alanine substitutions besides E407A showed improved product yield. The E407A mutation reduced SpATA activity substantially, supporting its predicted role in maintaining the dimeric enzyme structure. Conclusions: Using oxidase–amine transaminase cascades, the study demonstrated a fully enzymatic route to polysaccharide amination. Although the activity of SpATA may be further improved via enzyme engineering, the low operational stability of characterized amine transaminases, as a result of low retention of PMP cofactors, was identified as a key factor limiting the yield of the designed cascade. To increase the process feasibility, future efforts to engineer improved SpATA variants should focus on improving the cofactor affinity, and thus the operational stability of the enzyme. Background: Histoplasmosis is a fungus disease caused by a dimorphic organism called Histoplasma capsulatum, it is endemic in United States, Asia, and Africa. Inoculation is through inhalation of infectious spores. Histoplasmosis is missed in diagnosis because it mimics tuberculosis in clinical presentations and not all patients suffering from tuberculosis are tuberculosis patients. The aim is to determine the prevalence of disseminated histoplasmosis among patients with pulmonary infections. Method: This is cross sectional and hospital based. Samples were collected from patients and questionnaires were administered. Three hundred and twenty-two (322) urine samples were aseptically collected in urine tubes and analyzed by enzyme linked immunosorbent assay to detect the presence of galactomannan in the urine samples. Results: Out of 322 samples analyzed, 12 (3.74 %) were positive for the presence of galactomannan while 310 (96.27 %) were negative. Participants of the geriatric age group had the highest prevalence of 12.5 %. Male 8.07 % were more prone to histoplasmosis disease than their female 3.10 % counterpart. The married displayed a high prevalence value of 6.25 % than singles 2.25 % and the divorced 3.57 % and framer were demonstrated a high value of 7.69 % prevalence. Among the risk factors trader had an odd ratio of 3.800 and chest discomfort 1.037 % is a disease symptom associated with histoplasmosis. Conclusion: Disseminated histoplasmosis occur among the study participants through low in prevalence, awareness of its presence should be known to the general public and clinician, diagnosis of histoplasmosis is be done along that of Tuberculosis because it can be co-infection with Tuberculosis for proper treatment regime. Natural polysaccharides are among the renewable sources with great potential for replacing petroleum-derived chemicals as precursors to produce biodegradable films. This study aimed to prepare biopolymeric films using starch extracted from the periderm and cortex of cassava roots (waste from cassava root processing), locust bean galactomannan, and cellulose nanofibers also obtained from cassava waste. The films were prepared by casting, and their physicochemical, mechanical, and biodegradability properties were evaluated. The content of cellulose nanofibers varied from 0.5 to 2.5%. Although the addition of cellulose nanofibers did not alter the mechanical properties of the films, it significantly enhanced the vapor barrier of the films (0.055 g mm/m2 h kPa–2.5% nanofibers) and their respective stabilities in aqueous acidic and alkaline media. All prepared films were biodegradable, with complete degradation occurring within five days. It was found by the photocolorimetry method that binary compositions of protocatechic acid with monosaccharides (galactose, mannose) exhibit a pronounced antiradical synergistic effect in reaction with 2,2ʹ-diphenyl-1-picrylhydrazyl in acidic media. The synergistic properties of phenol-saccharide mixtures decrease with increasing pH of the medium. Using NMR spectroscopy and density functional theory, it was shown that the mechanism of synergism consists in the formation of intermolecular hydrogen–bonded phenol-monosaccharide complexes. The ionization energies of the donor and donor-acceptor complexes are lower than those of protocatechuic acid and hence the complexes react more actively with the radical compared to the initial antioxidant. Activation of the lectin pathway of the complement system, as demonstrated by elevated levels of mannan-binding lectin proteins (MBL), contributes to vascular pathology in type 1 diabetes (T1D). Vascular complications are greatest in T1D individuals with concomitant insulin resistance (IR), however, whether IR amplifies activiation of the lectin pathway in T1D is unknown. We pooled pretreatment data from two RCTs and performed a cross-sectional analysis on 46 T1D individuals. We employed estimated glucose disposal rate (eGDR), a validated IR surrogate with cut-points of: <5.1, 5.1–8.7, and > 8.7 mg/kg/min to determine IR status, with lower eGDR values conferring higher degrees of IR. Plasma levels of MBL-associated proteases (MASP-1, MASP-2, and MASP-3) and their regulatory protein MAp44 were compared among eGDR classifications. In a subset of 14 individuals, we assessed change in MASPs and MAp44 following improvement in IR. We found that MASP-1, MASP-2, MASP-3, and MAp44 levels increased in a stepwise fashion across eGDR thresholds with elevated MASPs and MAp44 levels conferring greater degrees of IR. In a subset of 14 patients, improvement in IR was associated with significant reductions in MASPs, but not MAp44, levels. In conclusion, IR in T1D amplifies levels of MASP-1/2/3 and their regulator MAp44, and improvement of IR normalizes MASP-1/2/3 levels. Given that elevated levels of these proteins contribute to vascular pathology, amplification of the lectin pathway of the complement system may offer mechanistic insight into the relationship between IR and vascular complications in T1D. Ultrasound-assisted extraction (UAE) of galactomannan from the endosperm of ripe carob seeds (LBG.S3) was investigated through response surface methodology (RSM). The optimized UAE conditions were an extraction time of 40 minutes, a temperature of 45°C, and a solid-liquid ratio of 1:50 g/mL. Under these settings, the experimental yield of galactomannan was 47.53 ± 2.55%, which is closely aligning with the expected value of 49.78 ± 3.15%. The yield of galactomannan extracted from unripe carob seeds (LBG.S2) under the same optimal conditions was significantly higher 56.73 ± 2.76% (p < 0.05) than the yield of LBG.S3. The infrared spectroscopy (FT-IR) characterization revealed the existence of various functional groups in the spectrum of galactomannan extracted from unripe seeds, not found in LBG.S3. The galactomannan from ripe carob seeds exhibited superior functional properties, including solubility, emulsion and stability capacity, water and oil holding capacity, and viscosity, compared to LBG.S2. In vitro, tests demonstrated notable anti-inflammatory activity for both LBG.S2 and LBG.S3, as observed through the inhibition of bovine serum albumin (BSA) protein denaturation by heat and the membrane stabilization test. Carob galactomannan is a potential source of bioactive polysaccharides with anti-inflammatory and functional properties that can be used in the food and pharmaceutical industries. Pyrotechnic‐based firefighting systems have been found to be effective in a wide variety of firefighting applications. Binders in pyrotechnic mixtures play a significant role as they consolidate the ingredients, provide excellent mechanical strength and increase the storage life. The rise in health and environmental safety concerns in recent years has directed our research towards natural binder systems of biological origins. Therefore, in this work, galactomannan, also called guar gum, was comparatively investigated against four different types of conventional binders, phenol–formaldehyde resin, ethyl cellulose, hydroxyethyl cellulose and carboxymethyl cellulose sodium salt, which are being used in aerosol‐forming pyrotechnic mixtures for firefighting application. It was found that the nature of the binder in an amount of 3% by weight significantly influences the combustion behaviour, fire suppression performance, mechanical and thermal characteristics of gallic acid–KNO3–KClO3 tertiary aerosol‐forming pyrotechnic mixture for fire fighting application. The safety and performance characteristics of these formulations were evaluated by conducting analyses specific to pyrotechnic formulations, that is, burn rate measurement, heat of combustion, TGA/DSC thermal analysis, hygroscopic study, compression test and n‐heptane pool fire suppression test. It can be inferred from the values obtained from these analyses that guar gum is a preferred binder to the other four‐binder systems for the gallic acid–KNO3–KClO3 pyrotechnic mixture. Most polymeric food packaging materials are non-biodegradable and derived from petroleum, thus recent studies have focused on evaluating alternative biodegradable materials from renewable sources, with polysaccharides and proteins as the main types of employed biopolymers. Therefore, this study aimed to develop biopolymeric films based on sunflower proteins and galactomannans from locust bean gum. The influence of the galactomannan amount (0.10%, 0.30%, 0.50%, and 0.75% w/v) on the physicochemical, thermal, and mechanical properties of cast sunflower protein-based films was studied. Sunflower proteins gave rise to yellowish, shining, and translucid films. With the incorporation of locust bean gum-derived galactomannans, the films became more brown and opaque, although they still maintained some translucency. Galactomannans significantly changed the proteins’ secondary structures, giving rise to films with increased tensile resistance and stretchability. Nevertheless, the increase in the galactomannan amount did not have a significant effect on the film’s thermal stability. The protein/galactomannan-based films showed values of water vapor and oxygen permeability that were slightly higher than those of the pristine materials. Overall, blending locust bean gum galactomannans with sunflower proteins was revealed to be a promising strategy to develop naturally colored and translucid films with enhanced mechanical resistance while maintaining flexibility, fitting the desired properties for biodegradable food packaging materials.

Aims: This study aimed to explore associations of mannan-binding lectin-associated serine protease (MASP) levels in early pregnancy with gestational diabetes mellitus (GDM). We also examined interactions of MASPs and deoxycholic acid (DCA)/glycoursodeoxycholic acid (GUDCA) for the GDM risk and whether the interactive effects if any on the GDM risk were mediated via lysophosphatidylcholine (LPC) 18:0. Materials and methods: A 1:1 case-control study (n = 414) nested in a prospective cohort of pregnant women was conducted in Tianjin, China. Binary conditional logistic regressions were performed to examine associations of MASPs with the GDM risk. Additive interaction measures were used to examine interactions between MASPs and DCA/GUDCA for the GDM risk. Mediation analyses and Sobel tests were used to examine mediation effects of LPC18:0 between the copresence of MASPs and DCA/GUDCA on the GDM risk. Results: High MASP-2 was independently associated with GDM [odds ratio (OR): 2.62, 95% confidence interval (CI): 1.44-4.77], while the effect of high MASP-1 on GDM was attributable to high MASP-2 (P for Sobel test: 0.003). Low DCA markedly increased the OR of high MASP-2 alone from 2.53 (1.10-5.85) up to 10.6 (4.22-26.4), with a significant additive interaction. In addition, high LPC18:0 played a significant mediating role in the links from low DCA to GDM and from the copresence of high MASP-2 and low DCA to GDM (P for Sobel test <0.001) but not in the link from high MASP-2 to GDM. Conclusions: High MASP-1 and MASP-2 in early pregnancy were associated with GDM in Chinese pregnant women. MASP-2 amplifies the risk of low DCA for GDM, which is mediated via LPC18:0. The review presents the main results obtained at the Favorsky Irkutsk Institute of Chemistry on the use of natural galactose-containing polysaccharides, such as arabinogalactan, galactomannan, and κ-carrageenan, for the synthesis of new water-soluble aggregation-stable nanocomposites, which are various inorganic nanoparticles (noble metals, metal oxides or elemental chalcogens) stabilized by polysaccharide macromolecules. Original synthetic approaches to nanocomposites and their structural characterization by spectral and microscopic methods, including X-ray phase analysis, are considered. Experimental evidence for the pronounced biological activity of a number of nanocomposites is provided, which in turn determines the prospects for developing on their basis of products for biomedicine and agriculture. A previous study found that a crude Perilla seed polysaccharide (PFSP) fraction exhibited obviously antitumor activity; however, the structural characterization and antitumor properties of this polysaccharide remain unclear. In this study, the PFSP was extracted and purified via combined column chromatography, and the structure of a single polysaccharide fraction was characterized by methylation, IC, GC-MS, NMR, and AFM. The results demonstrated that the efficient antitumor polysaccharide fraction PFSP-2-1 was screened from PFSP with a relative molecular weight of 8.81 × 106 Da. The primary structure of the PFSP main chain was →1)-Araf-(5→, →1,3)-Galp-(6→, →1)-Galp-(6→, →1,3)-Araf-(5→ and →1)-Xylp-(4→, and that of the side chains was →1)-Arap, →1,3)-Galp-(6→, →1)-Araf and →1)-Glcp-(4→, →1)-Galp-(3→ and →1)-Glcp, leading to a three-dimensional helical structure. CCK-8 experiments revealed that PFSP-2-1 significantly inhibited the growth of human hepatocellular carcinoma cells in vitro (p < 0.05), and its inhibitory effect positively correlation with the concentration of PFSP-2-1, and when the concentration of PFSP-2-1 was 1600 µg/mL, it showed the highest inhabitation rate on three hepatocellular carcinoma cells (HepG-2, Hep3b, and SK-Hep-1), for which the survival rates of HepG-2, Hep3b, and SK-Hep-1 were 53.34%, 70.33%, and 71.06%. This study clearly elucidated the structure and antitumor activity of PFSP-2-1, which lays a theoretical foundation for revealing the molecular mechanism of antitumor activity of Perilla seed polysaccharides and provides an important theoretical basis for the development of high-value Perilla. Galacto-oligosaccharides (GOS) are the prebiotic constituents with various functional qualities that promote the optimal proliferation of gut microbiota and are consequently beneficial to human health. GOS are not absorbed by humans and animals, but they play numerous health-related positive roles in the human body. Currently, their production has occupied much attention due to the increasing demand for functional foods. It could be produced by enzymatic and chemical approaches. Many microbial enzymes have been utilized for GOS production but the main source of these enzymes is lactose. Moreover, characterization of GOS structures is also a necessary and critical effort for understanding their mechanism of action. Hence, this review is written to understand the extensive but related features for the synthesis of GOS which includes: the enzymatic production of GOS through various enzymes and the types of enzymes such as free, immobilized, recombinant, crude, and purified enzymes that are involved in their synthesis, the techniques used in the purification of GOS, the analysis and quantification of GOS through various practices, and the functional properties of GOS are also discussed in this review. Nanoparticles (NPs) for allergen immunotherapy have garnered attention for their high efficiency and safety compared with naked antigen proteins. In this work, we present mannan-coated protein NPs, incorporating antigen proteins for antigen-specific tolerance induction. The heat-induced formation of protein NPs is a one-pot preparation method and can be applied to various proteins. Here, the NPs were formed spontaneously via heat denaturation of three component proteins: an antigen protein, human serum albumin (HSA) as a matrix protein, and mannoprotein (MAN) as a targeting ligand for dendritic cells (DCs). HSA is non-immunogenic, therefore suitable as a matrix protein, while MAN coats the surface of the NP. We applied this method to various antigen proteins and found that the self-disperse after heat denaturation was a requirement for incorporation into the NPs. We also established that the NPs could target DCs, and the incorporation of rapamycin into the NPs enhanced the induction of a tolerogenic phenotype of DC. The MAN coating provided steric hindrance and heat denaturation destroyed recognition structures, successfully preventing anti-antigen antibody binding, indicating the NPs may avoid anaphylaxis induction. During the perinatal period, unique metabolic adaptations support energetic requirements for rapid growth. To gain insight into perinatal adaptations, quantitative proteomics was performed comparing the livers of Yorkshire pigs at postnatal day 7 and adult. These data revealed differences in the metabolic control of liver function including significant changes in lipid and carbohydrate metabolic pathways. Newborn livers showed an enrichment of proteins in lipid catabolism and gluconeogenesis concomitant with elevated liver carnitine and acylcarnitines levels. Sugar kinases were some of the most dramatically differentially enriched proteins compared with neonatal and adult pigs including galactokinase 1 (Galk1), ketohexokinase (KHK), hexokinase 1 (HK1), and hexokinase 4 (GCK). Interestingly, hexokinase domain containing 1 (HKDC1), a newly identified fifth hexokinase associated with glucose disturbances in pregnant women, was highly enriched in the liver during the prenatal and perinatal periods and continuously declined throughout postnatal development in pigs and mice. These changes were confirmed via Western blot and mRNA expression. These data provide new insights into the developmental and metabolic adaptations in the liver during the transition from the perinatal period to adulthood in multiple mammalian species. The development of bacterial cellulose (BC) industrialization has been seriously affected by its production. Mannose/mannan is an essential component in many biomass resources, but Komagataeibacter xylinus uses mannose in an ineffective way, resulting in waste. The aim of this study was to construct recombinant bacteria to use mannose-rich biomass efficiently as an alternative and inexpensive carbon source in place of the more commonly used glucose. This strategy aimed at modification of the mannose catabolic pathway via genetic engineering of K. xylinus ATCC 23770 strain through expression of mannose kinase and phosphomannose isomerase genes from the Escherichia coli K-12 strain. Recombinant and wild-type strains were cultured under conditions of glucose and mannose respectively as sole carbon sources. The fermentation process and physicochemical properties of BC were investigated in detail in the strains cultured in mannose media. The comparison showed that with mannose as the sole carbon source, the BC yield from the recombinant strain increased by 84%, and its tensile strength and elongation were increased 1.7 fold, while Young's modulus was increased 1.3 fold. The results demonstrated a successful improvement in BC yield and properties on mannose-based medium compared with the wild-type strain. Thus, the strategy of modifying the mannose catabolic pathway of K. xylinus is feasible and has significant potential in reducing the production costs for industrial production of BC from mannose-rich biomass.
