Dengue virus (DENV) is a pathogenic arbovirus that causes human disease. The most severe stage of the disease (severe dengue) is characterized by vascular leakage, hypovolemic shock, and organ failure. Endothelial dysfunction underlies these phenomena, but the causal mechanisms of endothelial dysfunction are poorly characterized. This study investigated the role of c-ABL kinase in DENV-induced endothelial dysfunction. Silencing c-ABL with artificial miRNA or targeting its catalytic activity with imatinib revealed that c-ABL is required for the early steps of DENV infection. DENV-2 infection and conditioned media from DENV-infected cells increased endothelial expression of c-ABL and CRKII phosphorylation, promoted expression of mesenchymal markers, e.g., vimentin and N-cadherin, and decreased the levels of endothelial-specific proteins, e.g., VE-cadherin and ZO-1. These effects were reverted by silencing or inhibiting c-ABL. As part of the acquisition of a mesenchymal phenotype, DENV infection and treatment with conditioned media from DENV-infected cells increased endothelial cell motility in a c-ABL-dependent manner. In conclusion, DENV infection promotes a c-ABL-dependent endothelial phenotypic change that leads to the loss of intercellular junctions and acquisition of motility.

Cet article a pour objet de préciser la conception des systèmes expérimentaux chez Hans-Jörg Rheinberger dans le cadre de la phénoménotechnique telle que conçue par Bachelard afin de montrer son utilité pour l’analyse et la compréhension d’un cas particulier, à savoir l’émergence de la « cellule vivant » en tant qu’objet épistémique dans la biologie cellulaire contemporaine depuis l’origine, avec la théorie cellulaire, la cytologie et la biochimie (Bechtel, 2006), jusqu’à ses développements plus récents fondés sur l’imagerie de cellules vivantes (ICV). La notion de système expérimental inclut des objets épistémiques ou scientifiques, des conditions expérimentales ainsi que des conjonctures, des hybridations et bifurcations. La conception de système expérimental chez Rheinberger est une caractérisation fine de la science contemporaine qui permet de saisir un élément central de l’activité de recherche, l’émergence de l’objet épistémico-scientifique (la cellule vivante) co-généré à l’intérieur d’un espace qui se situe entre la représentation et le graphème (une trace expérimentale qui produit des signifiants) au sein d’un système expérimental appelé ICV. Enfin, dans le contexte des récentes études sociales des sciences et technologies (STS), nous cherchons à montrer qu’il est possible d’intégrer les réflexions philosophiques et l’étude de la pratique scientifique en laboratoire, pratique souvent absente dans les travaux en cours en philosophie des sciences. The present work has the objective to precise the conception of the Experimental Systems (ES) of Rheinberger, within the context of phenomenotechnics of Bachelard by showing after its utility on the analysis and understanding of a particular case, like is the rise of the epistemic-scientific object “living cell” in Contemporary Cell Biology (CCB). In order to get such comprehension this study begins from the origins with Cellular Theory (CT), Cytology and Biochemistry (Bechtel, 2006), until to arrive to the most recent developments based in Live Cell Imaging (LCI). The notion of ES of Rheinberger is a finer characterization of the real science because it articulates a central element to the research activity, specifically with our case study the emerging of epistemic-scientific object (living cell), which is co-generated between a space of representation and the graphemes (experimental traces with signifiers), all together within an ES called Live Cell Imaging. Finally, in the context of the recent STS works, we search showing how is possible to integrate some philosophical reflections with scientific practice, and how it is occurring in the laboratory, which it is absent in many works in philosophy of science. El presente artículo tiene por objetivo precisar la concepción de los sistemas experimentales de Rheinberger, dentro del contexto del concepto de fenomenotecnia de Bachelard, para después mostrar su utilidad en el análisis y comprensión de un caso particular, como es la emergencia del objeto epistémico «célula viviente» en la Biología Celular Contemporánea. En este último caso, se parte de sus orígenes, con la teoría celular, la citología y la bioquímica (Bechtel, 2006), hasta llegar a sus más recientes desarrollos basados en imagenología de células vivas. En relación con la noción de sistema experimental, se plantea que estos incluyen objetos epistémicos o científicos y condiciones experimentales, así como coyunturas, hibridaciones y bifurcaciones (Rheinberger, 2011). La concepción de sistema experimental de Rheinberger, es una caracterización más fina de la ciencia contemporánea que atrapa un elemento central de la actividad investigativa, el surgimiento del objeto científico-epistémico (célula viviente) co-generado entre un espacio de representación y un grafema (un rastro experimental que deja significantes), dentro de un sistema experimental llamado imagenología de células vivas. Finalmente, en el contexto de los recientes trabajos en estudios CTS, también se busca mostrar cómo es posible integrar las reflexiones filosóficas con la práctica científica, como sucede realmente en el laboratorio, que está bastante ausente en los actuales trabajos sobre filosofía de la ciencia.

RESUMEN La quinasa dependiente de ciclina 5 (CDK5) regula diversas funciones en neuronas, células endoteliales y epiteliales, entre ellas la dinámica del citoesqueleto. Así mismo, se ha reportado que componentes del citoesqueleto, tales como, filamentos de actina y microtúbulos juegan un rol importante durante la infección por el virus dengue (DENV). El objetivo del presente trabajo fue evaluar por dos métodos, inhibición química y silenciamiento génico, la participación de CDK5 durante la infección por DENV-2. La actividad antiviral de roscovitina fue evaluada usando ensayos de Unidades Formadoras de Placa (PFU). La eficiencia de transfección y el silenciamiento de CDK5, empleando miARNs artificiales, se determinó por citometría de flujo. El efecto sobre la proteína de envoltura viral y elementos del citoesqueleto se evidenció mediante microscopia avanzada de fluorescencia y análisis de imágenes. Roscovitina mostró actividad antiviral en etapas pre y post-infectivas en una forma dependiente de la dosis. El tratamiento con roscovitina y miRCDK5 mostró ser efectivo reduciendo la cantidad de CDK5 en células no infectadas. En células infectadas y transfectadas con miRCDK5, así como tratadas con el inhibidor, se observó una reducción significativa de la proteína de envoltura viral; sin embargo, no se encontró reducción significativa de CDK5. Además, el tratamiento con roscovitina indujo cambios celulares morfológicos evidentes en células infectadas. Los resultados indican la potencial participación de CDK5 durante la infección por DENV-2, posiblemente mediando la traducción proteica o la replicación del genoma viral a través de la regulación de la dinámica del citoesqueleto. Se requieren datos adicionales para esclarecer la mecanística del fenómeno usando métodos alternativos. ABSTRACT Cyclin-Dependent Kinase 5 (CDK5) regulates several functions in neurons, endothelial, and epithelial cells, including the cytoskeleton dynamics. Likewise, it has been reported that some cytoskeleton elements, such as actin filaments and microtubules, play an essential role during Dengue virus (DENV) infection. This work aimed to evaluate the role of CDK5 during DENV-2 infection by two methods, chemical inhibition, and gene silencing. The antiviral activity of roscovitine was evaluated using Plaque Forming Units (PFU) assay. The transfection efficiency and knockdown of CDK5, using artificial miRNAs, was carried out by flow cytometry. The effect on the viral envelope protein and cytoskeleton elements was evidenced by advanced fluorescence microscopy and image analysis. Roscovitine showed antiviral activity in pre and post-infection stages in a dose-dependent manner. Treatment with roscovitine and miRCDK5 decrease the amount of CDK5 in uninfected cells. In cells infected and transfected with miRCDK5, as well as treated with the inhibitor, a significant reduction of the viral envelope protein was observed; however, no significant reduction of CDK5 was found. Also, evident morphological cellular changes were observed during the treatment with roscovitine in infected cells. The results indicate the potential participation of CDK5 during DENV-2 infection, possibly mediating protein translation or replication of the viral genome through the cytoskeletal dynamics regulation. Additional data are required to clarify the mechanistic of these phenomena using alternative methods.

Objetivo: Describir las manifestaciones clínicas y hallazgos de laboratorio de una serie de casos febriles agudos con diagnóstico presuntivo de infección por el virus dengue. en Quindío (Colombia). Materiales y métodos: Se realizó un estudio de corte transversal, en pacientes con sospecha clínica de dengue en el periodo comprendido entre enero y agosto de 2013, en algunos centros hospitalarios del departamento del Quindío. Se tomaron muestras de sangre para diagnóstico de dengue, leptospira, malaria, hepatitis B, y rickettsiosis. Como pruebas confirmatorias para dengue se realizó aislamiento viral en células C6/36HT y serotipificación para dengue por RTPCR; pruebas de función hepática, cuadro hemático y niveles de citocinas. Resultados: Se caracterizaron 149 casos, de los cuales el 43% presentaron infección por dengue, 4% leptospira, 6,8% rickettsias, un caso de malaria y uno de hepatitis B. En 5 casos se logró el aislamiento del DENV2 y DENV3. Mediante la RT-PCR, se evidenció cocirculación de serotipos 2, 3, 4. Se encontró que las enzimas AST/ALT, el conteo de plaquetas, la erupción y el dolor abdominal fueron marcadores característicos de la infección por dengue, mientras la ictericia y el dolor lumbar se correlacionaron con la leptospirosis. Los valores de citocinas mostraron que la IL-10, TNF α variaron significativamente en casos con dengue frente a otros diagnósticos, y la IL-17 a presentó diferencias significativas en individuos con dengue grave. Conclusiones: El dengue se confirmó como causa etiológica importante de síndrome febril icterohemorrágico en el departamento del Quindío, pero la leptospirosis y la rickettsiosis tienen también una participación importante. Sin embargo, en el 44% de los casos fueron catalogados como síndrome febril indeterminado. Objective: To characterise the clinical and laboratory findings on a series of febrile cases with a presumptive diagnosis of dengue virus infection in Quindío, Colombia. Materials and methods: This study was conducted from January to July 2013. Blood samples were obtained from patients suspected of dengue virus infection from Quindío department hospitals. These samples were tested for dengue, leptospira, malaria, hepatitis B and rickettsiosis. To confirm dengue infection, we performed viral isolation in C6/36HT cells and dengue serotyping by RT-PCR; liver function tests, complete blood counts and cytokine levels. Results: Of 149 cases, 43% were infected by dengue, 4% leptospira, 6.8% rickettsia, one case of malaria and one case of hepatitis b. We obtained 5 clinical isolates of DENV2 and DENV3 that evidenced co-circulation of serotypes 2, 3, and 4. We found that AST/ALT levels, platelet count, rash and abdominal pain were good markers of infection by dengue, while jaundice and lumbar pain suggested leptospirosis. Cytokine levels revealed that IL-10, TNF a varied significantly in dengue compared with other diagnostics and that IL-17 α showed significant differences in individuals with severe dengue. Conclusions: Dengue was confirmed as an important aetiology of acute febrile icterohaemorrhagic syndrome in Quindío, but leptospirosis and rickettsia also play an important role. However, 44% of the cases were classified as undetermined febrile syndrome.

El presente ensayo es una aproximación a la vida revolucionaria e ideas innovadoras de Charles Darwin. El propósito es celebrar los doscientos años de su nacimiento y los 150 de la publicación de su obra "Origen de las especies". Se enfoca la reflexión en la revolución darwiniana desde varias dimensiones: conceptual, filosófica, científica y cultural. En la génesis de esta revolución, se destaca el valor de la observación científica, que siempre tiene una carga teórica, y los casi treinta años que empleó Darwin en el estudio prolongado de las notas de campo, la reflexión sobre sus datos, la contrastación de los mismos con los de otros autores y la extensa red de alianzas estratégicas. Todas estas cualidades que son mucho más valiosas, que los mejores equipos y los laboratorios más dotados, para hacer investigación científica, que apunte a resolver las preguntas esenciales de la ciencia, como "el misterio de los misterios". Finalmente, se mencionan los impactos en la sociedad actual y en la investigación en varios campos, colocando particular interés en esclarecer los buenos usos y abusos de la teoría evolutiva. Para ello se toman como ejemplos las llamadas medicina evolutiva y la psicología evolutiva. This essay is an approach to the revolutionary life This essay is an approach to the revolutionary life and innovative ideas of Charles Darwin. The purpose is to celebrate 200 years of his birth and 150 of the publication of his "Origin of Species". We focus on the Darwin's revolution from its conceptual, philosophical, scientific and cultural points of view. In the genesis of this revolution, we remark the value of scientific observation, which always carries a theoretical load, and so the almost 30 years invested by Darwin in his deep study of field notes, the reflection authors and the wide network of strategic alliances established. All of these qualities are more valuable than the best equipments or the most sophisticated laboratories used in research aiming to answer the essential questions of science, as "the mystery of mysteries". Finally, we mention the impacts on the current society and on research in several fields, emphasizing the good uses and abuses of the evolutional theory. We take as examples the evolutional medicine and the evolutional psychology. O presente ensaio é uma aproximação à vida revolucionária e idéias inovadoras de Charles Darwin. Todas estas qualidades que são bem mais valiosas que os melhores equipes e os laboratórios mais dotados para fazer investigação científica, que anotação a resolver as perguntas essenciais da ciência, como "o mistério dos mistérios". Este artículo es una aproximación multidisciplinar, basada en los fundamentos de la teoría del caos, evolución del cerebro humano y la sistemática filogenética, para intentar contribuir al estudio del cerebro humano en condiciones normales y patológicas, evaluando la posible utilidad de esas disquisiciones en la cotidianidad de la docencia y práctica psiquiátricas. Se expone aquí que el funcionamiento cerebral se puede entender mejor desde la perspectiva de la dinámica no lineal e indeterminista de la teoría del caos, la cual permite una aproximación a fenómenos tan complejos como el origen de la inteligencia, el afecto y la enfermedad mental. Finalmente, se plantea que la metodología de la filogenética puede ser una herramienta útil que aportaría elementos para la nosología de las enfermedades mentales y eventualmente ayudaría a una comprensión más integral de éstas. The following proposal is a multidisciplinary approach using the fundamentals of chaos theory, evolution of the human brain and phylogenetic systematics to try to contribute to the study of the human brain in normal and pathological conditions, evaluating the potential use of these disquisitions in teaching and psychiatric practice. It is exposed here that the functioning of the brain can best be understood from the viewpoint of the non-lineal and undeterministic dynamics of chaos theory, possibly explaining the origins of intelligence, mood and mental disorders. Finally, as a new contribution it is postulated that the methodology of phylogenetics could be a useful tool for the classification of mental disorders also helping to comprehend them better.

The airn of the thesis was to analyze the rnorphogenetic stages and the virusinduced rnorphological changes in infected hurnan cells with three attenuated W rnutants (MVA, M65 and MIOI), in cornparison with cells infected with the wild type virus, strain WR. A detail transrnission electron rnicroscopy (EM) analysis of hurnan HeLa cells showed irnportant differences in fine uitraestructure and arnounts of the viral intermediates cornpared to wild type WR-infected cells. The EM studies revealed that there are distinct changes in the rnorphogenetic process for al1 of the attenuated rnutants. New intermediate viral forms were found between the IV (irnrnature virus) and the IMV (intracellular rnature virus) forms, representing transition stages with the core still in the assernbling process. The rnorphogenesis of MVA was largely blocked at the IV stage, but few rnature forms were obsewed and the IMVs produced acquired a more munded shape than WR. These "IMVs" have an abnormal interna1 structure (referred as "atypical viruses") with potential alterations in the interactions core-envelope, and are unable to significantly acquire the additional double envelope to render intracellular envelope virus (IEVs). In the case of rnutants M65 an MlOl, virus rnorphogenesis was interrupted at later stages, forming lMVs that accurnulate within the cell but do not progress to form extracellular enveloped virus (EEV). Analyses of viral proteins in the rnutants showed alterations in sorne of the structural proteins with roles in the rnorphogenetic pathway. Confocal rnicroscopy revealed that the rnutants produced a srnall plaque-size phenotype with rnicrocornets and absence of cell lysis. While WR induces actin polyrnerization forming characteristic coiled tails, the rnutants do not produce actin tails and do not exit the cell. Moreover, the rnutants produced rnorphological and biochernical changes to the cells, compatible with an epithelial to rnesenchyrnal transition (EMT), possibly to facilitate spreading of the virus between cells. Mice irnrnunized with recornbinant rnutants expressing Plasmodium yoelii circurnsporozoite antigen (CS) are able to induce significant protection against malaria. Our findings are of irnportance in the developrnent of vaccines based on attenuated W rnutants.

A finales de 2019, se inició la pandemia de COVID-19, lo que impulsó a los científicos de todo el mundo a aplicar sus respectivos conocimientos para abordar cómo el SARS-CoV-2 infecta a los humanos. La principal estrategia ha sido la implementación de sistemas convencionales de vigilancia sanitaria para identificar, intervenir y controlar las infecciones virales causadas por estos virus emergentes. Aunque el seguimiento de la evolución genética del virus ha sido de gran importancia, se desconoce hasta qué punto es posible la transmisión zoonótica entre especies animales susceptibles y no susceptibles, así como la eventual funcionalidad de la arquitectura estructural del genoma de RNA de los Betacoronavirus en la fisiopatología, principalmente para SARS-CoV, MERS-CoV y SARS-CoV-2. Para llenar este vacío de conocimiento y facilitar el desarrollo de tratamientos eficaces, se realizó un estudio amplio de los genomas de los Betacoronavirus mediante el análisis de 1,252,952 secuencias virales reportadas en bases de datos que han circulado desde el 2002 pasando de reservorios naturales a huésped intermedio y humanos. Este trabajo considera dos enfoques diferentes de representar la información genómica, como se presentan y discuten en el capítulo 2: Análisis de secuencia. Esta parte del trabajo presenta un análisis evolutivo de transmisión horizontal en las secuencias virales para caracterizar y describir completamente la variación intra-hospedera de los Betacoronavirus. Los resultados revelan que cambios de aminoácidos en la subunidad S1 de la proteína S de SARS-CoV (G > T; A577S), MERS-CoV (C > T; S746R y C > T; N762A) y SARS-CoV-2 (A > G; D614G) con señales de selección positiva son factores fundamentales que subyacen al posible salto de barrera de los murciélagos al huésped intermedio. El capítulo 3: Análisis estructural, es una sección que explora los Betacoronavirus a nivel estructural como propuesta para descubrir si el plegamiento de estructuras secundarias de RNA conservadas podrían actuar como loci putativos para procesar RNAs pequeños virales, con una posible función asociada a la patogénesis en proceso de selección. Más del 87.58% de estas estructuras de RNA indican que 12 regiones portan RNAs pequeños en los Betacoronavirus, sugiriendo la posibilidad de modular la reprogramación transcripcional del nuevo huésped después de la infección. Los hallazgos de este estudio proporcionan una serie de significativos patrones moleculares que contribuyen a expandir las fronteras de la terapéutica humanos en el contexto de la actual crisis sanitaria mundial. Betacoronavirus have caused earlier deadly epidemics, including the 2002 SARS-CoV outbreak and the ongoing prevalence of MERS-CoV, which was first detected in 2012. In late 2019, the emergence of the COVID-19 pandemic encouraged scientists around the globe to apply their respective insights to address how SARS-CoV-2 infects humans. The main strategy has been the implementation of standard health surveillance systems to identify, manage and control viral infections caused by these emerging viruses. Even though monitoring the genetic evolution of the virus has been of high significance, to what extent zoonotic transmission across susceptible and non-susceptible animal species is possible, as well as eventual functionality the structural architecture of the RNA genome of Betacoronavirus in the pathophysiology, mainly for SARS-CoV, MERS-CoV and SARS-CoV-2 is unclear. To fill this knowledge gap and facilitate the development of effective treatments, a comprehensive study of Betacoronavirus genomes was performed by means of the analysis of 1,252,952 viral sequences reported in databases which have circulated since 2002 from natural reservoirs to intermediate hosts and humans. This study includes two different approaches to represent genomic information, as introduced and discussed in Chapter 2: Sequence analyses. This part of the work represents an evolutionary analysis of horizontal transmission in viral sequences to thoroughly characterize and describe the intra-host variation and transmission routes of Betacoronavirus. The results reveal that amino acid changes within S protein S1 subunit of SARS-CoV (G > T; A577S), MERS-CoV (C > T; S746R and C > T; N762A) and SARS-CoV-2 (A > G; D614G) with signals of positive selection are pivotal factors underlying the possible jumping from bats barrier to intermediate host. Chapter 3: Structural analyses, is a section that explores Betacoronavirus at the structural level as a proposal to discover whether the folding of conserved RNA secondary structures may act as putative loci for processing virus-derived small RNAs, with a potential function associated with pathogenesis in the process of selection. Over 87.58% of these RNA structures indicate that 12 regions carry small RNAs in Betacoronavirus, suggesting the possibility of modulation of transcriptional re-programming of the new host upon infection. The findings of this study provide a collection of significant molecular signatures that contribute to pushing the frontiers of human therapeutics in the context of the current global health crisis.
