While it is well accepted that climate change and growing water needs affect long-term sustainable water resources management, performing accurate simulations of water cycle and energy balance dynamics at regional scale remains a challenging task.Traditional Soil-Vegetation-Atmosphere-Transfer (SVAT) models are used for numerical surface water and energy simulations. These models, by conception, do not account for the groundwater lower boundary that permits a full hydrosystem representation. Conversely, while addressing important features such as subsurface heterogeneity and river–aquifer exchanges, groundwater models often integrate overly simplified upper boundary conditions ignoring soil heating and the impacts of vegetation processes on radiation fluxes and root-zone uptakes. In this paper, one of the first attempts to jointly model water and energy fluxes with a special focus on both surface and groundwater at the regional scale is proposed on the Seine hydrosystem (78,650 km$^{2}$), which overlays one of the main multi-aquifer systems of Europe.This study couples the SVAT model ORCHIDEE and the process-based hydrological–hydrogeological model CaWaQS, which describes water fluxes, via a one-way coupling approach from ORCHIDEE toward CaWaQS based on the blueprint published by [de Marsily et al., 1978]. An original transport library based on the resolution of the diffusion/advection transport equation was developed in order to simulate heat transfer in both 1D-river networks and pseudo-3D aquifer systems. In addition, an analytical solution is used to simulate heat transport through aquitards and streambeds. Simulated ORCHIDEE surface water and energy fluxes feed fast surface runoff and slow recharge respectively and then is used as CaWaQS forcings to compute river discharges, hydraulic heads and temperature dynamics through space and time, within each of the hydrosystem compartments. The tool makes it possible to establish a fully consistent water and energy budget over a period of 17 years. It also simulates temperature evolution in each aquifer and evaluates that river thermal regulation mostly relies by order of importance on short wave radiations (109.3 W${\cdot }$m$^{-2}$), groundwater fluxes (48.1 W${\cdot }$m$^{-2}$) and surface runoff (22.7 W${\cdot }$m$^{-2}$).

Abstract In small‐scale societies, people learn to identify plant species during childhood. Plant recognition is an important baseline knowledge, immediately useful to avoid intoxication risk due to wrong identification. Plant recognition is the basis of other ethnobotanical knowledge essential for safeguarding biocultural diversity. However, despite many studies on folk classification, we still have a narrow understanding of the criteria locally used for species identification; the gap being even larger regarding children's plant identification criteria. Here, we study the criteria used by Betsileo children and adolescents to identify wild edible plant (WEP) species using a child‐adapted method including drawings and follow‐up interviews. We worked with 80 teenagers (from 12 to 17 years old; 51 girls, and 29 boys). Our results suggest that teenagers use a large spectrum of visual criteria to identify plants and that these criteria match with botanical and ecological knowledge documented in the literature and herbarium vouchers. We found that 35% of the identification criteria used were non‐morphological (e.g. phenology, biotic interactions), suggesting deep ecological knowledge. On average, teenagers use more than nine distinct criteria per plant, which allows them to identify most plant species with a very high level of precision. The precision level of plant representation increases with age for boys, but remains constant for girls, suggesting different dynamics in plant identification knowledge acquisition. We also found that boys and girls use different identification criteria: girls focus on morphological criteria while boys also incorporate ecological criteria, such as landscape features and biotic interactions, in their spectrum of identification keys. Our results highlight the complexity of teenagers' plant knowledge and the importance of the ecological context and gender in plant identification's knowledge acquisition. This knowledge acquired very early in childhood, constitutes the foundation of future interactions with nature and should be at the heart of environmental humanities studies and knowledge co‐production projects to tackle socio‐ecological concerns. Hence, we urge further research to explore innovative methods that complement traditional ethnoecological tools and capture complex sensory aspects of folk children's taxonomy to better understand human‐plant interactions and knowledge. Abstract Background The mesenchymal subtype of colorectal cancer (CRC), associated with poor prognosis, is characterized by abundant expression of the cellular prion protein PrPC, which represents a candidate therapeutic target. How PrPC is induced in CRC remains elusive. This study aims to elucidate the signaling pathways governing PrPC expression and to shed light on the gene regulatory networks linked to PrPC. Methods We performed in silico analyses on diverse datasets of in vitro, ex vivo and in vivo models of mouse CRC and patient cohorts. We mined ChIPseq studies and performed promoter analysis. CRC cell lines were manipulated through genetic and pharmacological approaches. We created mice combining conditional inactivation of Apc in intestinal epithelial cells and overexpression of the human prion protein gene PRNP. Bio-informatic analyses were carried out in two randomized control trials totalizing over 3000 CRC patients. Results In silico analyses combined with cell-based assays identified the Wnt-β-catenin and glucocorticoid pathways as upstream regulators of PRNP expression, with subtle differences between mouse and human. We uncover multiple feedback loops between PrPC and these two pathways, which translate into an aggravation of CRC pathogenesis in mouse. In stage III CRC patients, the signature defined by PRNP-CTNNB1-NR3C1, encoding PrPC, β-catenin and the glucocorticoid receptor respectively, is overrepresented in the poor-prognosis, mesenchymal subtype and associates with reduced time to recurrence. Conclusions An unleashed PrPC-dependent vicious circle is pathognomonic of poor prognosis, mesenchymal CRC. Patients from this aggressive subtype of CRC may benefit from therapies targeting the PRNP-CTNNB1-NR3C1 axis.

Circulating tumor DNA (ctDNA) has been studied as a non-invasive tool for disease monitoring in different cancer types. Despite advances in colorectal cancer (CRC) management, it remains a leading cause of mortality and there is an unmet need for new biomarkers to guide therapeutic approaches and improve patient’s outcome after surgical resection of the primary tumor or its metastatic sites. This review summarizes the different clinical results and ongoing studies on the performances of ctDNA as a prognostic marker for disease recurrence, both in non-metastatic patients with resection of the primary tumor and in those with full resection of metastatic disease.

Background: Bacterial peritonitis (BP) in patients with gastrointestinal (GI) cancer has been poorly described, and its prevalence is unknown. Objectives: This study aimed to evaluate in patients with both GI cancer and ascites the prevalence of BP, associated features, mechanisms, prognosis, and the diagnostic performance of neutrophil count in ascites. Design: A retrospective, multicenter, observational study. Methods: All patients with GI cancer and ascites who underwent at least one paracentesis sample analyzed for bacteriology over a 1-year period were included. BP was defined by a positive ascites culture combined with clinical and/or biological signs compatible with infection. Secondary BP was defined as BP related to a direct intra-abdominal infectious source. Results: Five hundred fifty-seven ascites from 208 patients included were analyzed. Twenty-eight patients had at least one episode of BP and the annual prevalence rate of BP was 14%. Among the 28 patients with BP, 19 (65%) patients had proven secondary BP and 17 (59%) patients had multi-microbial BP, mainly due to Enterobacterales . A neutrophil count greater than 110/mm 3 in ascites had negative and positive predictive values of 96% and 39%, respectively, for the diagnosis of BP. The median survival of patients with BP was 10 days (interquartile range 6–40) after the diagnosis. Conclusion: BP is not rare in patients with GI cancer and is associated with a poor short-term prognosis. When a patient with GI cancer is diagnosed with BP, a secondary cause should be sought. Further studies are needed to better define the best management of these patients.

Abstract The Plastid Terminal Oxidase (PTOX) is a chloroplast localized plastoquinone oxygen oxidoreductase suggested to have the potential to act as a photoprotective safety valve for photosynthesis. However, PTOX overexpression in plants has been unsuccessful at inducing photoprotection, and the factors that control its activity remain elusive. Here, we show that significant PTOX activity is induced in response to high light in the model species Eutrema salsugineum and Arabidopsis thaliana. This activation correlates with structural reorganization of the thylakoid membrane. Over-expression of PTOX in mutants of Arabidopsis thaliana perturbed in thylakoid stacking also results in such activity, in contrast to wild type plants with normal granal structure. Further, PTOX activation protects against photoinhibition of Photosystem II and reduces reactive oxygen production under stress conditions. We conclude that structural re-arrangements of the thylakoid membranes, bringing Photosystem II and PTOX into proximity, are both required and sufficient for PTOX to act as a Photosystem II sink and play a role in photoprotection.

Abstract Considerable progress has been made in understanding the molecular host-virus battlefield during SARS-CoV-2 infection. Nevertheless, the assembly and egress of newly formed virions are less understood. To identify host proteins involved in viral morphogenesis, we characterize the proteome of SARS-CoV-2 virions produced from A549-ACE2 and Calu-3 cells, isolated via ultracentrifugation on sucrose cushion or by ACE-2 affinity capture. Bioinformatic analysis unveils 92 SARS-CoV-2 virion-associated host factors, providing a valuable resource to better understand the molecular environment of virion production. We reveal that G3BP1 and G3BP2 (G3BP1/2), two major stress granule nucleators, are embedded within virions and unexpectedly favor virion production. Furthermore, we show that G3BP1/2 participate in the formation of cytoplasmic membrane vesicles, that are likely virion assembly sites, consistent with a proviral role of G3BP1/2 in SARS-CoV-2 dissemination. Altogether, these findings provide new insights into host factors required for SARS-CoV-2 assembly with potential implications for future therapeutic targeting.

Among the many sorts of policies used to fight obesity and overweight, behavioral public policies are currently expanding. However, by their very nature, behavioral public policies may threaten the respect of persons’ identity. In particular, the normative status of behavioral interventions is questionable because of the way they interfere with people’s choices. Indeed, behavioral interventions may sometimes affect individuals’ attitudes and choices without the targets being aware of this influence. This article discusses whether and how dietary guidelines and their related tools, as cases of behavioral public policies, deal with the respect of persons’ identity. This issue is central to assess the normative status of these public policies. Identity is a recent issue in economics but some works show that identity plays a role in economic behavior. We thus develop a method to characterize identity. On this basis, we examine how behavioral public food policies take identity into account. We analyze dietary food guides of four countries and some of their related tools. We show that dietary food guides give a strong attention to (some) cultural and personal identity and aim at respecting the persons’ autonomy. However, their declination through tools appears more problematic and includes choice restriction and eventually the exploitation of cognitive biases. Such a difference between the claim of respecting identity and actions calls for a discussion of the normative status of those policies. From a philosophical account of autonomy, we argue that the major issue associated with behavioral public policies is not that they interfere with one’s behavior but rather that they must give sufficient attention to the respect of a person’s values, i.e., to respect her personal and social identity.
