Herein, a series of novel O-alkyl ferulamide derivatives were designed and synthesised through the multi-target-directed ligands (MTDLs) strategy. The biological activities in vitro showed that compounds 5a, 5d, 5e, 5f, and 5h indicated significantly selective MAO-B inhibitory potency (IC50 = 0.32, 0.56, 0.54, 0.73, and 0.86 µM, respectively) and moderate antioxidant activity. Moreover, compounds 5a, 5d, 5e, 5f, and 5h showed potent anti-inflammatory properties, remarkable effects on self-induced Aβ1-42 aggregation, and potent neuroprotective effect on Aβ1-42-induced PC12 cell injury. Furthermore, compounds 5a, 5d, 5e, 5f, and 5h presented good blood-brain barrier permeation in vitro and drug-like properties. More interesting, the PET/CT images with [11C]5f demonstrated that [11C]5f could penetrate the BBB with a high brain uptake and exhibited good brain clearance kinetic property. Photosynthesis response to carbon dioxide concentration can provide data on a number of important parameters related to leaf physiology. The genetic algorithm (GA), which is a robust stochastic evolutionary computational algorithm inspired by both natural selection and natural genetics, is proposed to simultaneously estimate the parameters [including maximum carboxylation rate allowed by ribulose 1·5-bisphosphate carboxylase/oxygenase (Rubisco) carboxylation rate ( Vcmax), potential light-saturated electron transport rate ( Jmax), triose-phosphate utilization (TPU), leaf dark respiration in the light ( Rd) and mesophyll conductance ( gm)] of the photosynthesis models presented by Farquhar, von Caemmerer and Berry, and Ethier and Livingston. The results show that by properly constraining the parameter bounds the GA-based estimate methods can effectively and efficiently obtain globally (or, at least near globally) optimal solutions, which are as good as or better than those obtained by non-linear curve fitting methods used in previous studies. More complicated problems such as taking the gm variation response to CO2 into account can be easily formulated and solved by using GA. In this study, a series of naringenin-O-alkylamine derivatives were designed and obtained by introducing an alkylamine fragment into the naringenin skeleton. The in vitro biological activity results revealed that compounds 5f and 7k showed good antioxidant activity with ORAC values of 2.3eq and 1.2eq, respectively. Compounds 5f and 7k were reversible and excellent huAChE inhibitors with IC50 values of 0.91 µM and 0.57 µM, respectively. Moreover, compounds 5f and 7k could inhibit self-induced Aβ1-42 aggregation with 62.1% and 43.8% inhibition rate, respectively, and significantly inhibited huAChE-Aβ1-40 aggregation with 51.7% and 43.4% inhibition rate, respectively. In addition, compounds 5f and 7k were selective metal chelators and remarkably inhibited Cu2+-induced Ab1-42 aggregation with 73.5% and 68.7% inhibition rates, respectively. Furthermore, compounds 5f and 7k could cross the blood-brain barrier in vitro and displayed good neuroprotective effects and anti-inflammatory properties. Further investigation showed that compound 5f did not show obvious hepatotoxicity and displayed a good hepatoprotective effect by its antioxidant activity. The in vivo study displayed that compound 5f significantly improved scopolamine-induced mice memory impairment. Riparian ecosystems fundamentally depend on groundwater, and thus accurate estimates of daily groundwater evapotranspiration rate (ET g) is curial for understanding the interactions between groundwater and vegetation. Up to now, various methods (i.e., White, 1932; Hays, 2003; Soylu et al., 2012) have been proposed to estimate daily ET g using the water table fluctuations. The objective of this paper was to evaluate and compare their performances under various water table conditions in field environments. For this purpose, we employed a variety of measurements (i.e., water table levels, latent heat flux and soil water contents) at a riparian forest (T. ramosissima) in Northwest China in 2017. Our results showed that the White method is suitable for use during the water table declining periods. Also, the selected time intervals could have impacts on the estimations of ET g. The Soylu method and the Hays method performed well under various water table conditions. Thus, it seems that the Hays and Soylu methods are more suitable for long-term ET g estimates in field environments. In addition, the percentage of plant transpired water from groundwater varied during the growing seasons. The riparian plants mainly use soil water during the early growing stages, and it tended to use groundwater during the late growing stages. Wetland methane (CH4) emissions are the largest natural source in the global CH4 budget, contributing to roughly one third of total natural and anthropogenic emissions. As the second most important anthropogenic greenhouse gas in the atmosphere after CO2, CH4 is strongly associated with climate feedbacks. However, due to the paucity of data, wetland CH4 feedbacks were not fully assessed in the Intergovernmental Panel on Climate Change Fifth Assessment Report. The degree to which future expansion of wetlands and CH4 emissions will evolve and consequently drive climate feedbacks is thus a question of major concern. Here we present an ensemble estimate of wetland CH4 emissions driven by 38 general circulation models for the 21st century. We find that climate change-induced increases in boreal wetland extent and temperature-driven increases in tropical CH4 emissions will dominate anthropogenic CH4 emissions by 38 to 56% toward the end of the 21st century under the Representative Concentration Pathway (RCP2.6). Depending on scenarios, wetland CH4 feedbacks translate to an increase in additional global mean radiative forcing of 0.04 W⋅m-2 to 0.19 W⋅m-2 by the end of the 21st century. Under the "worst-case" RCP8.5 scenario, with no climate mitigation, boreal CH4 emissions are enhanced by 18.05 Tg to 41.69 Tg, due to thawing of inundated areas during the cold season (December to May) and rising temperature, while tropical CH4 emissions accelerate with a total increment of 48.36 Tg to 87.37 Tg by 2099. Our results suggest that climate mitigation policies must consider mitigation of wetland CH4 feedbacks to maintain average global warming below 2 °C. [ABSTRACT FROM AUTHOR]

Background: Cerebral oedema is closely related to the permeability of blood-brain barrier, vascular endothelial growth factor (VEGF) and its receptor vascular endothelial growth factor receptor 2 (VEGFR2) all of which are important blood-brain barrier (BBB) permeability regulatory factors. Zonula occludens 1 (ZO-1) and claudin-5 are also the key components of BBB. Hypertonic saline is widely used to alleviate cerebral oedema. This study aimed to explore the possible mechanisms underlying hypertonic saline that ameliorates cerebral oedema effectively. Methods: Middle cerebral artery occlusion (MCAO) model in Sprague-Dawley (SD) rats and of oxygen-glucose deprivation model in primary astrocytes were used in this study. The brain water content (BWC) was used to assess the effect of 10 % HS on cerebral oedema. The assessment of Evans blue (EB) extravasation was performed to evaluate the protective effect of 10 % HS on blood-brain barrier. The quantification of VEGF, VEGFR2, ZO-1 and claudin-5 was used to illustrate the mechanism of 10 % HS ameliorating cerebral oedema. Results: BWC was analysed by wet-to-dry ratios in the ischemic hemisphere of SD rats; it was significantly decreased after 10 % HS treatment (P < 0.05). We also investigated the blood-brain barrier protective effect by 10 % HS which reduced EB extravasation effectively in the peri-ischemic brain tissue. In parallel to the above notably at 24 h following MCAO, mRNA and protein expression of VEGF and VEGFR2 in the peri-ischemic brain tissue was down-regulated after 10 % HS treatment (P < 0.05). Along with this, in vitro studies showed increased VEGF and VEGFR2 mRNA and protein expression in primary astrocytes under hypoxic condition (P < 0.05), but it was suppressed after HS treatment (P < 0.05). In addition, HS inhibited the down-regulation of ZO-1, claudin-5 effectively. Conclusions: The results suggest that 10 % HS could alleviate cerebral oedema possibly through reducing the ischemia induced BBB permeability as a consequence of inhibiting VEGF-VEGFR2-mediated down-regulation of ZO-1, claudin-5. A major research plan entitled "Integrated research on the eco-hydrological process of the Heihe River Basin" was launched by the National Natural Science Foundation of China in 2010. One of the key aims of this research plan is to establish a research platform that integrates observation, data management, and model simulation to foster twenty-first-century watershed science in China. Based on the diverse needs of inter-disciplinary studies within this research plan, a program called the Heihe Watershed Allied Telemetry Experimental Research (HiWATER) was implemented. The overall objective of HiWATER is to improve the observability of hydrological and ecological processes, to build a world-class watershed observing system, and to enhance the applicability of remote sensing in integrated eco-hydrological studies and water resource management at the basin scale. This paper introduces the background, scientific objectives, and experimental design of HiWATER. The instrumental setting and airborne mission plans are also outlined. The highlights are the use of a flux observing matrix and an eco-hydrological wireless sensor network to capture multiscale heterogeneities and to address complex problems, such as heterogeneity, scaling, uncertainty, and closing water cycle at the watershed scale. HiWATER was formally initialized in May 2012 and will last four years until 2015. Data will be made available to the scientific community via the Environmental and Ecological Science Data Center for West China.
