Abstract

Abstract Glioblastoma is one of the most aggressive types of cancer with success of therapy being hampered by the existence of treatment resistant populations of stem-like Tumour Initiating Cells (TICs) and poor blood–brain barrier drug penetration. Therapies capable of effectively targeting the TIC population are in high demand. Here, we synthesize spherical diketopyrrolopyrrole-based Conjugated Polymer Nanoparticles (CPNs) with an average diameter of 109 nm. CPNs were designed to include fluorescein-conjugated Hyaluronic Acid (HA), a ligand for the CD44 receptor present on one population of TICs. We demonstrate blood–brain barrier permeability of this system and concentration and cell cycle phase-dependent selective uptake of HA-CPNs in CD44 positive GBM-patient derived cultures. Interestingly, we found that uptake alone regulated the levels and signaling activity of the CD44 receptor, decreasing stemness, invasive properties and proliferation of the CD44-TIC populations in vitro and in a patient-derived xenograft zebrafish model. This work proposes a novel, CPN- based, and surface moiety-driven selective way of targeting of TIC populations in brain cancer.

Abstract

Organic electroactive materials, particularly semiconducting polymers, are at the forefront of emerging organic electronics. Among the plethora of unique features, the possibility to formulate inks out of these materials is particularly promising for the large‐scale manufacturing of electronics at lower cost on a variety of soft substrates. While solution deposition of semiconducting materials is promising for developing printed electronics, the environmental footprint of the materials and related devices needs to be considered to achieve sustainable manufacturing. Towards the development of greener printed electronics, this work investigates the utilization of a non‐toxic, environmentally‐friendly solvent, namely branched polyethylene (BPE), to formulate semiconducting inks. Focusing on a diketopyrrolopyrrole‐based (DPP) semiconducting polymer, shellac as dielectric, and BPE as the solvent, solutions were prepared in different concentrations and their rheological properties were characterized. Then, printing on polyethylene terephthalate (PET) substrates using two different techniques was performed to fabricate organic field‐effect transistors (OFETs). Both printing techniques yielded OFETs with good performance and device characteristics, averaging approximately 10−2 and 10−4 cm2 V−1 s−1, respectively, for slot‐die coating and direct‐ink writing deposition. Notably, despite some difference in threshold voltages, OFETs produced via slot‐die coating and direct‐ink writing showed comparable charge mobilities to previously reported OFETs prepared from similar materials, particularly those prepared on silicon dioxide wafers. Overall, this work confirms the suitability of BPE to formulate semiconducting inks to develop printed electronics in a greener manner. Abstract

The optoelectronic properties of semiconducting polymers and device performance rely on a delicate interplay of design and processing conditions. However, screening and optimizing the relationships between these parameters for reliably fabricating organic electronics can be an arduous task requiring significant time and resources. To overcome this challenge, Polybot is developed—a robotic platform within a self‐driving lab that can efficiently produce organic field‐effect transistors (OFETs) from various semiconducting polymers via high‐throughput blade coating deposition. Polybot not only handles the fabrication process but also can conduct characterization tests on the devices and autonomously analyze the data gathered, thus facilitating the rapid acquisition of data on a large scale. This work leverages the capabilities of this platform to investigate the fabrication of OFETs using hydrogen bonding‐containing semiconducting polymers. Through high‐throughput fabrication and characterization, various data trends are analyzed, and large extents of anisotropic charge mobility are observed in devices. The materials are thoroughly characterized to understand the role of processing conditions in solid state and electronic properties of these organic semiconductors. The findings demonstrate the effectiveness of automated fabrication and characterization platforms in uncovering novel structure–property relationships, facilitating refinement of rational chemical design, and processing conditions, ultimately leading to new semiconducting materials. Abstract

Relapse susceptibility in women with substance use disorders (SUDs) has been linked to the estrogen, 17β-estradiol (E2). Our previous findings in female rats suggest that the influence of E2 on cocaine seeking can be localized to the prelimbic prefrontal cortex (PrL-PFC). Here, we investigated the receptor mechanisms through which E2 regulates the reinstatement of extinguished cocaine seeking. Sexually mature female rats underwent intravenous cocaine self-administration (0.5 mg/inf; 14 × 2 h daily) and extinction, and then were ovariectomized before reinstatement testing. E2 (10 µg/kg, i.p.) alone did not reinstate cocaine seeking, but it potentiated reinstatement when combined with an otherwise subthreshold priming dose of cocaine. A similar effect was observed following intra-PrL-PFC microinfusions of E2 and by systemic or intra-PrL-PFC administration of the estrogen receptor (ER)β agonist, DPN, but not agonists at ERα or the G-protein-coupled ER1 (GPER1). By contrast, E2-potentiated reinstatement was prevented by intra-PrL-PFC microinfusions of the ERβ antagonist, MPP, or the GPER1 antagonist, G15, but not an ERα antagonist. Whole-cell recordings in PrL-PFC layer (L)5/6 pyramidal neurons revealed that E2 decreases the frequency, but not amplitude, of GABA(A)-dependent miniature IPSCs (mIPSC). As was the case with E2-potentiated reinstatement, E2 reductions in mIPSC frequency were prevented by ERβ and GPER1, but not ERα, antagonists and mimicked by ERβ, but not GPER1, agonists. Altogether, the findings suggest that E2 activates ERβ and GPER1 in the PrL-PFC to attenuate the GABA-mediated constraint of key outputs that mediate cocaine seeking.

Abstract

To explore the role of peptide conformation on catalytic activity in the context of ester hydrolysis catalysts, pairs of sequences were designed that contained or lacked β-hairpin character. For the hydrolysis of para-nitrophenylacetate in aqueous media, we found small but consistent trends wherein His-containing sequences based on a TrpZip scaffold showed higher catalytic activity without β-hairpin character., Peptide catalysts based on TrpZip scaffolds for the hydrolysis of para-nitrophenylacetate in aqueous media were found to have higher catalytic activity in sequences without β-hairpin character.

25. Impact of Sequential Chemical Doping on the Thin Film Mechanical Properties of Conjugated Polymers

Abstract

Conjugated polymer (CP) films with nanometer-scale thickness exhibit unique properties distinct from their bulk counterparts, which is an important consideration for their end application as thin film devices. In the realm of organic electronic devices, enabling high electrical conductance properties of CPs often necessitates doping. However, the impact of doping on intrinsic polymer mechanical properties, such as the elastic modulus, in ultrathin films at device-relevant thicknesses is not well understood and has not been directly measured. In this study, we quantified the effect of doping on the mechanical properties of poly(3-alkylthiophenes) (P3ATs) using pseudofree-standing tensile testing. We observed modulation of the mechanical properties of ultrathin CP films through sequential doping of P3ATs thin films (60–80 nm thick) with the molecular dopant F4TCNQ. Our findings reveal that, despite the ease of doping all P3ATs with F4TCNQ, the resulting changes in mechanical properties are highly dependent on the side-chain lengths of the P3ATs. Specifically, the elastic modulus of rubbery P3ATs with side-chain lengths of six carbons or more (e.g., P3HT and P3OT) increases significantly─by one to two times─upon F4TCNQ doping, while the modulus of the glassy poly(3-butylthiophene-2,5-diyl) (P3BT) remains nearly unchanged. Such a phenomenon is linked to the changes in the glass transition temperature (Tg) of the doped film, where the rise of Tgresults in a large change in the modulus for P3HT samples. However, the P3BT remained in a glassy state before and after doping, exhibiting a minimal change in its mechanical properties. These insights into the mechanical behavior of doped ultrathin CP films are crucial for the design and optimization of flexible electronic devices.
