1. A Giant Disk Galaxy Two Billion Years After The Big Bang

Abstract

Observational studies showed that galaxy disks are already in place in the first few billion years of the universe. The early disks detected so far, with typical half-light radii of 3 kiloparsecs at stellar masses around 10^11 M_sun for redshift z~3, are significantly smaller than today's disks with similar masses, in agreement with expectations from current galaxy models. Here, we report observations of a giant disk at z=3.25, when the universe was only 2 billion years old, with a half-light radius of 9.6 kiloparsecs and stellar mass of 3.7^+2.6_-2.2x10^11 M_sun. This galaxy is larger than any other kinematically-confirmed disks at similar epochs and surprisingly similar to today's largest disks regarding size and mass. JWST imaging and spectroscopy reveal its spiral morphology and a rotational velocity consistent with local Tully-Fisher relation. Multi-wavelength observations show that it lies in an exceptionally dense environment, where the galaxy number density is over ten times higher than the cosmic average and mergers are frequent. The discovery of such a giant disk suggests the presence of favorable physical conditions for large-disk formation in dense environments in the early universe, which may include efficient accretion of gas carrying coherent angular momentum and non-destructive mergers between exceptionally gas-rich progenitor galaxies., Comment: 22 pages, 11 figures; submitted

2. 3D{\pi}: Three-Dimensional Positron Imaging, A Novel Total-Body PET Scanner Using Xenon-Doped Liquid Argon Scintillator

Abstract

Objective: This paper introduces a novel PET imaging methodology called 3-dimensional positron imaging (3D{\pi}), which integrates total-body (TB) coverage, time-of-flight (TOF) technology, ultra-low dose imaging capabilities, and ultra-fast readout electronics inspired by emerging technology from the DarkSide collaboration. Approach: The study evaluates the performance of 3D{\pi} using Monte Carlo simulations based on NEMA NU 2-2018 protocols. The methodology employs a homogenous, monolithic scintillator composed of liquid argon (LAr) doped with xenon (Xe) with silicon photomultipliers (SiPM) operating at cryogenic temperatures. Main results: Significant enhancements in system performance are observed, with the 3D{\pi} system achieving a noise equivalent count rate (NECR) of 3.2 Mcps which is approximately two times higher than uEXPLORER's peak NECR (1.5 Mcps) at 17.3 (kBq/mL). Spatial resolution measurements show an average FWHM of 2.7 mm across both axial positions. The system exhibits superior sensitivity, with values reaching 373 kcps/MBq with a line source at the center of the field of view. Additionally, 3D{\pi} achieves a TOF resolution of 151 ps at 5.3 kBq/mL, highlighting its potential to produce high-quality images with reduced noise levels. Significance: The study underscores the potential of 3D{\pi} in improving PET imaging performance, offering the potential for shorter scan times and reduced radiation exposure for patients. The Xe-doped LAr offers advantages such as fast scintillation, enhanced light yield, and cost-effectiveness. Abstract

DarkSide-20k (DS-20k) is a dark matter detection experiment under construction at the Laboratori Nazionali del Gran Sasso (LNGS) in Italy. It utilises ~100 t of low radioactivity argon from an underground source (UAr) in its inner detector, with half serving as target in a dual-phase time projection chamber (TPC). The UAr cryogenics system must maintain stable thermodynamic conditions throughout the experiment's lifetime of >10 years. Continuous removal of impurities and radon from the UAr is essential for maximising signal yield and mitigating background. We are developing an efficient and powerful cryogenics system with a gas purification loop with a target circulation rate of 1000 slpm. Central to its design is a condenser operated with liquid nitrogen which is paired with a gas heat exchanger cascade, delivering a combined cooling power of >8 kW. Here we present the design choices in view of the DS-20k requirements, in particular the condenser's working principle and the cooling control, and we show test results obtained with a dedicated benchmarking platform at CERN and LNGS. We find that the thermal efficiency of the recirculation loop, defined in terms of nitrogen consumption per argon flow rate, is 95 % and the pressure in the test cryostat can be maintained within $\pm$(0.1-0.2) mbar. We further detail a 5-day cool-down procedure of the test cryostat, maintaining a cooling rate typically within -2 K/h, as required for the DS-20k inner detector. Additionally, we assess the circuit's flow resistance, and the heat transfer capabilities of two heat exchanger geometries for argon phase change, used to provide gas for recirculation. We conclude by discussing how our findings influence the finalisation of the system design, including necessary modifications to meet requirements and ongoing testing activities., Comment: 45 pages, 24 figures

4. The MUSE Ultra Deep Field (MUDF). VI. The relationship between galaxy properties and metals in the circumgalactic medium

Abstract

We present intial results associating galaxies in the MUSE Ultra Deep Field (MUDF) with gas seen in absorption along the line-of-sight to two bright quasars in this field, to explore the dependence of metals in the circumgalactic medium (CGM) on galaxy properties. The MUDF includes $\sim$140h of VLT/MUSE data and 90 orbits of HST/G141M grism observations alongside VLT/UVES spectroscopy of the two quasars and several bands of HST imaging. We compare the metal absorption around galaxies in this field as a function of impact parameter, azimuthal angle and galaxy metallicity across redshifts 0.5 $<$ z $<$ 3.2. Due to the depth of our data and a large field-of-view, our analysis extends to low stellar masses ($<$ $10^{7}$ M$_{\odot}$) and high impact parameters ($>$ 600 kpc). We find a correlation between absorber equivalent width and number of nearby galaxies, but do not detect a significant anti-correlation with impact parameter. Our full sample does not show any significant change in absorber incidence as a function of azimuthal angle. However, we do find a bimodality in the azimuthal angle distribution of absorption at small impact parameters ($<$2 r$_{vir}$) and around highly-star-forming galaxies, possibly indicating disk-like accretion and biconical outflows. Finally, we do not detect any systematic deviation from the fundamental metallicity relation (FMR) among galaxies with detected absorption. This work is limited by gaps in the wavelength coverage of our current data; broader-wavelength observations with JWST will allow us to unlock the full potential of the MUDF for studying the CGM., Comment: Accepted for publication in The Astrophysical Journal. 31 pages, 17 figures

Abstract

The dual-phase liquid argon time projection chamber is presently one of the leading technologies to search for dark matter particles with masses below 10 GeV/c$^2$. This was demonstrated by the DarkSide-50 experiment with approximately 50 kg of low-radioactivity liquid argon as target material. The next generation experiment DarkSide-20k, currently under construction, will use 1,000 times more argon and is expected to start operation in 2027. Based on the DarkSide-50 experience, here we assess the DarkSide-20k sensitivity to models predicting light dark matter particles, including Weakly Interacting Massive Particles (WIMPs) and sub-GeV/c$^2$ particles interacting with electrons in argon atoms. With one year of data, a sensitivity improvement to dark matter interaction cross-sections by at least one order of magnitude with respect to DarkSide-50 is expected for all these models. A sensitivity to WIMP--nucleon interaction cross-sections below $1\times10^{-42}$ cm$^2$ is achievable for WIMP masses above 800 MeV/c$^2$. 6. MUSE Analysis of Gas around Galaxies (MAGG) -- VI. The cool and enriched gas environment of z$\gtrsim$3 Ly$\alpha$ emitters

Abstract

We present a novel dataset that extends our view of the cosmic gas around z$\approx$3-4 Ly$\alpha$ emitting galaxies (LAEs) in the Muse Analysis of Gas around Galaxies (MAGG) survey by tracing a cool and enriched gas phase through 47 MgII absorbers identified in newly-obtained VLT/XSHOOTER near-infrared quasar spectra. Jointly with the more ionized gas traced by CIV systems and the neutral HI from previous work, we find that LAEs are distributed inside cosmic structures that contain multiphase gas in composition and temperature. All gas phases are a strong function of the large-scale galaxy environment: the MgII and the CIV strength and kinematics positively correlate with the number of associated galaxies, and it is $\approx$3-4 times more likely to detect metal absorbers around group than isolated LAEs. Exploring the redshift evolution, the covering factor of MgII around group and isolated LAEs remains approximately constant from z$\approx$3-4 to z<2, but the one of CIV around group galaxies drops by z<2. Adding the cool enriched gas traced by the MgII absorbers to the results we obtained for the HI and CIV gas, we put forward a picture in which LAEs lie along gas filaments that contain high column-density HI systems and are enriched by strong CIV and MgII absorbers. While the MgII gas appears to be more centrally concentrated near LAEs, weaker CIV systems trace instead a more diffuse gas phase extended up to larger distances around the galaxies., Comment: Accepted for publication on A&A

Abstract

Experiments aimed at direct searches for WIMP dark matter require highly effective reduction of backgrounds and control of any residual radioactive contamination. In particular, neutrons interacting with atomic nuclei represent an important class of backgrounds due to the expected similarity of a WIMP-nucleon interaction, so that such experiments often feature a dedicated neutron detector surrounding the active target volume. In the context of the development of DarkSide-20k detector at INFN Gran Sasso National Laboratory (LNGS), several R&D projects were conceived and developed for the creation of a new hybrid material rich in both hydrogen and gadolinium nuclei to be employed as an essential element of the neutron detector. Thanks to its very high cross-section for neutron capture, gadolinium is one of the most widely used elements in neutron detectors, while the hydrogen-rich material is instrumental in efficiently moderating the neutrons. In this paper results from one of the R&Ds are presented. In this effort the new hybrid material was obtained as a poly(methyl methacrylate) (PMMA) matrix, loaded with gadolinium oxide in the form of nanoparticles. We describe its realization, including all phases of design, purification, construction, characterization, and determination of mechanical properties of the new material.

Abstract

Sub-mm surveys toward overdense regions in the early Universe are essential to uncover the obscured star-formation and the cold gas content of assembling galaxies within massive dark matter halos. In this work, we present deep ALMA mosaic observations covering an area of $\sim 2'\times2'$ around MQN01 (MUSE Quasar Nebula 01), one of the largest and brightest Ly-$\alpha$ emitting nebulae discovered thus far surrounding a radio-quiet quasar at $z\simeq3.25$. Our observations target the 1.2- and the 3-mm dust continuum, as well as the carbon monoxide CO(4-3) transition in galaxies in the vicinity of the quasar. We identify a robust sample of eleven CO line-emitting galaxies (including a closely-separated quasar companion) which lie within $\pm 4000\,{\rm km\,s^{-1}}$ relatively to the quasar systemic redshift. A fraction of these objects are missed in previous deep rest-frame optical/UV surveys thus highlighting the critical role of (sub-)mm imaging. We also detect a total of eleven sources revealed in their 1.2-mm dust continuum with six of them having either high-fidelity spectroscopic redshift information from rest-frame UV metal absorptions, or CO line which place them in the same narrow redshift range. A comparison of the CO luminosity function (LF) and 1.2-mm number count density with that of the general fields points to a galaxy overdensity of $\delta > 10$. We find evidence of a systematic flattening at the bright-end of the CO LF with respect to the trend measured in blank fields. Our findings reveal that galaxies in dense regions at $z\sim3$ are more massive and significantly richer in molecular gas than galaxies in fields, hence enabling a faster and accelerated assembly. Abstract

This research work aims to develop an analytical approach for optimizing team formation and predicting team performance in a competitive environment based on data on the competitors' skills prior to the team formation. There are several approaches in scientific literature to optimize and predict a team's performance. However, most studies employ fine-grained skill statistics of the individual members or constraints such as teams with a set group of members. Currently, no research tackles the highly constrained domain of the FIRST Robotics Competition. This research effort aims to fill this gap by providing an analytical method for optimizing and predicting team performance in a competitive environment while allowing these constraints and only using metrics on previous team performance, not on each individual member's performance. We apply our method to the drafting process of the FIRST Robotics competition, a domain in which the skills change year-over-year, team members change throughout the season, each match only has a superficial set of statistics, and alliance formation is key to competitive success. First, we develop a method that could extrapolate individual members' performance based on overall team performance. An alliance optimization algorithm is developed to optimize team formation and a deep neural network model is trained to predict the winning team, both using highly post-processed real-world data. Our method is able to successfully extract individual members' metrics from overall team statistics, form competitive teams, and predict the winning team with 84.08% accuracy.

Abstract

The stability of a dark matter detector on the timescale of a few years is a key requirement due to the large exposure needed to achieve a competitive sensitivity. It is especially crucial to enable the detector to potentially detect any annual event rate modulation, an expected dark matter signature. In this work, we present the performance history of the DarkSide-50 dual-phase argon time projection chamber over its almost three-year low-radioactivity argon run. In particular, we focus on the electroluminescence signal that enables sensitivity to sub-keV energy depositions. The stability of the electroluminescence yield is found to be better than 0.5%. Finally, we show the temporal evolution of the observed event rate around the sub-keV region being consistent to the background prediction., Comment: 13 pages, 5 figures

Abstract

Borexino could efficiently distinguish between alpha and beta radiation in its liquid scintillator by the characteristic time profile of their scintillation pulse. This alpha/beta discrimination, first demonstrated at the tonne scale in the Counting Test Facility prototype, was used throughout the lifetime of the experiment between 2007 and 2021. With this method, alpha events are identified and subtracted from the beta-like solar neutrino events. This is particularly important in liquid scintillator as alpha scintillation is quenched many-fold. In Borexino, the prominent Po-210 decay peak was a background in the energy range of electrons scattered from Be-7 solar neutrinos. Optimal alpha-beta discrimination was achieved with a "multi-layer perceptron neural network", which its higher ability to leverage the timing information of the scintillation photons detected by the photomultiplier tubes. An event-by-event, high efficiency, stable, and uniform pulse shape discrimination was essential in characterising the spatial distribution of background in the detector. This benefited most Borexino measurements, including solar neutrinos in the \pp chain and the first direct observation of the CNO cycle in the Sun. Abstract

Ly$\alpha$ nebulae ubiquitously found around z>2 quasars can supply unique constraints on the properties of the Circumgalactic Medium, such as its density distribution, provided the quasar halo mass is known. We present a new method to constrain quasar halo masses based on the line-of-sight velocity dispersion maps of Ly$\alpha$ nebulae. By using MUSE-like mock observations obtained from cosmological hydrodynamic simulations under the assumption of maximal quasar fluorescence, we show that the velocity dispersion radial profiles of Ly$\alpha$-emitting gas are strongly determined by gravity and that they are thus self-similar with respect to halo mass when rescaled by the virial radius. Through simple analytical arguments and by exploiting the kinematics of HeII1640\.A emission for a set of observed nebulae, we show that Ly$\alpha$ radiative transfer effects plausibly do not change the shape of the velocity dispersion profiles but only their normalisation without breaking their self-similarity. Taking advantage of these results, we define the variable $\eta^{140-200}_{40-100}$ as the ratio of the median velocity dispersion in two specifically selected annuli and derive an analytical relation between $\eta^{140-200}_{40-100}$ and the halo mass which can be directly applied to observations. Abstract

Abstract Mutations targeting distinct domains of the neuron-specific kinesin KIF5A associate with different neurodegenerative/neurodevelopmental disorders, but the molecular bases of this clinical heterogeneity are unknown. We characterised five key mutants covering the whole spectrum of KIF5A-related phenotypes: spastic paraplegia (SPG, R17Q and R280C), Charcot-Marie-Tooth disease (CMT, R864*), amyotrophic lateral sclerosis (ALS, N999Vfs*40), and neonatal intractable myoclonus (NEIMY, C975Vfs*73) KIF5A mutants. CMT-R864*-KIF5A and ALS-N999Vfs*40-KIF5A showed impaired autoinhibition and peripheral localisation accompanied by altered mitochondrial distribution, suggesting transport competence disruption. ALS-N999Vfs*40-KIF5A formed SQSTM1/p62-positive inclusions sequestering WT-KIF5A, indicating a gain of toxic function. SPG-R17Q-KIF5A and ALS-N999Vfs*40-KIF5A evidenced a shorter half-life compared to WT-KIF5A, and proteasomal blockage determined their accumulation into detergent-insoluble inclusions. Interestingly, SPG-R280C-KIF5A and ALS-N999Vfs*40-KIF5A both competed for degradation with proteasomal substrates. Finally, NEIMY-C975Vfs*73-KIF5A displayed a similar, but more severe aberrant behaviour compared to ALS-N999Vfs*40-KIF5A; these two mutants share an abnormal tail but cause disorders on the opposite end of KIF5A-linked phenotypic spectrum. Thus, our observations support the pathogenicity of novel KIF5A mutants, highlight abnormalities of recurrent variants, and demonstrate that both unique and shared mechanisms underpin KIF5A-related diseases.

Abstract

Abstract Lysosomes are acidic organelles involved in crucial intracellular functions, including the degradation of organelles and protein, membrane repair, phagocytosis, endocytosis, and nutrient sensing. Given these key roles of lysosomes, maintaining their homeostasis is essential for cell viability. Thus, to preserve lysosome integrity and functionality, cells have developed a complex intracellular system, called lysosome quality control (LQC). Several stressors may affect the integrity of lysosomes, causing Lysosomal membrane permeabilization (LMP), in which membrane rupture results in the leakage of luminal hydrolase enzymes into the cytosol. After sensing the damage, LQC either activates lysosome repair, or induces the degradation of the ruptured lysosomes through autophagy. In addition, LQC stimulates the de novo biogenesis of functional lysosomes and lysosome exocytosis. Alterations in LQC give rise to deleterious consequences for cellular homeostasis. Specifically, the persistence of impaired lysosomes or the malfunctioning of lysosomal processes leads to cellular toxicity and death, thereby contributing to the pathogenesis of different disorders, including neurodegenerative diseases (NDs). Recently, several pieces of evidence have underlined the importance of the role of lysosomes in NDs. In this review, we describe the elements of the LQC system, how they cooperate to maintain lysosome homeostasis, and their implication in the pathogenesis of different NDs. Graphical Abstract

Abstract

Abstract Background Common bean (Phaseolus vulgaris) is one of the main nutritional resources in the world, and a low environmental impact source of protein. However, the majority of its cultivation areas are affected by drought and this scenario is only expected to worsen with climate change. Stomatal closure is one of the most important plant responses to drought and the MYB60 transcription factor is among the key elements regulating stomatal aperture. If targeting and mutating the MYB60 gene of common bean would be a valuable strategy to establish more drought-tolerant beans was therefore investigated. Results The MYB60 gene of common bean, with orthology to the Arabidopsis AtMYB60 gene, was found to have conserved regions with MYB60 typical motifs and architecture. Stomata-specific expression of PvMYB60 was further confirmed by q-RT PCR on organs containing stomata, and stomata-enriched leaf fractions. Further, function of PvMYB60 in promoting stomata aperture was confirmed by complementing the defective phenotype of a previously described Arabidopsis myb60-1 mutant. Conclusions Our study finally points PvMYB60 as a potential target for obtaining more drought-tolerant common beans in the present context of climate change which would further greatly contribute to food security particularly in drought-prone countries.

Abstract

The direct search for dark matter in the form of weakly interacting massive particles (WIMP) is performed by detecting nuclear recoils (NR) produced in a target material from the WIMP elastic scattering. A promising experimental strategy for direct dark matter search employs argon dual-phase time projection chambers (TPC). One of the advantages of the TPC is the capability to detect both the scintillation and charge signals produced by NRs. Furthermore, the existence of a drift electric field in the TPC breaks the rotational symmetry: the angle between the drift field and the momentum of the recoiling nucleus can potentially affect the charge recombination probability in liquid argon and then the relative balance between the two signal channels. This fact could make the detector sensitive to the directionality of the WIMP-induced signal, enabling unmistakable annual and daily modulation signatures for future searches aiming for discovery. The Recoil Directionality (ReD) experiment was designed to probe for such directional sensitivity. The TPC of ReD was irradiated with neutrons at the INFN Laboratori Nazionali del Sud, and data were taken with 72 keV NRs of known recoil directions. The direction-dependent liquid argon charge recombination model by Cataudella et al. was adopted and a likelihood statistical analysis was performed, which gave no indications of significant dependence of the detector response to the recoil direction. The aspect ratio R of the initial ionization cloud is estimated to be 1.037 +/- 0.027 and the upper limit is R < 1.072 with 90% confidence level, Comment: 20 pages, 10 figures, submitted to Eur. Abstract

We report the first measurement of CNO solar neutrinos by Borexino that uses the Correlated Integrated Directionality (CID) method, exploiting the sub-dominant Cherenkov light in the liquid scintillator detector. The directional information of the solar origin of the neutrinos is preserved by the fast Cherenkov photons from the neutrino scattered electrons, and is used to discriminate between signal and background. The directional information is independent from the spectral information on which the previous CNO solar neutrino measurements by Borexino were based. While the CNO spectral analysis could only be applied on the Phase-III dataset, the directional analysis can use the complete Borexino data taking period from 2007 to 2021. The absence of CNO neutrinos has been rejected with >5{\sigma} credible level using the Bayesian statistics. The directional CNO measurement is obtained without an external constraint on the $^{210}$Bi contamination of the liquid scintillator, which was applied in the spectral analysis approach. The final and the most precise CNO measurement of Borexino is then obtained by combining the new CID-based CNO result with an improved spectral fit of the Phase-III dataset. Including the statistical and the systematic errors, the extracted CNO interaction rate is $R(\mathrm{CNO})=6.7^{+1.2}_{-0.8} \, \mathrm{cpd/100 \, tonnes}$. Taking into account the neutrino flavor conversion, the resulting CNO neutrino flux at Earth is $\Phi_\mathrm{CNO}=6.7 ^{+1.2}_{-0.8} \times 10^8 \, \mathrm{cm^{-2} s^{-1}}$, in agreement with the high metallicity Standard Solar Models. The results described in this work reinforce the role of the event directional information in large-scale liquid scintillator detectors and open up new avenues for the next-generation liquid scintillator or hybrid neutrino experiments.

Abstract

To understand how well a proposed augmented reality (AR) solution works, existing papers often conducted tailored and isolated evaluations for specific AR tasks, e.g., depth or lighting estimation, and compared them to easy-to-setup baselines, either using datasets or resorting to time-consuming data capturing. Conceptually simple, it can be extremely difficult to evaluate an AR system fairly and in scale to understand its real-world performance. The difficulties arise for three key reasons: lack of control of the physical environment, the time-consuming data capturing, and the difficulties to reproduce baseline results. This paper presents our design of an AR experimentation platform, ExpAR, aiming to provide scalable and controllable AR experimentation. ExpAR is envisioned to operate as a standalone deployment or a federated platform; in the latter case, AR researchers can contribute physical resources, including scene setup and capturing devices, and allow others to time share these resources. Our design centers around the generic sensing-understanding-rendering pipeline and is driven by the evaluation limitations observed in recent AR systems papers. We demonstrate the feasibility of this vision with a preliminary prototype and our preliminary evaluations suggest the importance of further investigating different device capabilities to stream in 30 FPS. Dark matter induced event rate in an Earth-based detector is predicted to show an annual modulation as a result of the Earth's orbital motion around the Sun. We searched for this modulation signature using the ionization signal of the DarkSide-50 liquid argon time projection chamber. Abstract

We present two new tools for studying and modelling metal absorption lines in the circumgalactic medium. The first tool, dubbed ``NMF Profile Maker'' (NMF$-$PM), uses a non-negative matrix factorization (NMF) method and provides a robust means to generate large libraries of realistic metal absorption profiles. The method is trained and tested on 650 unsaturated metal absorbers in the redshift interval $z=0.9-4.2$ with column densities between $11.2 \le \log{(\mathrm{N/cm^{-2}})} \le 16.3$, obtained from high-resolution ($R> 4000$) and high signal-to-noise ratio ($S/N \ge 10$) quasar spectroscopy. To avoid spurious features, we train on infinite $S/N$ Voigt models of the observed line profiles derived using the code ``Monte-Carlo Absorption Line Fitter'' (MC$-$ALF), a novel automatic Bayesian fitting code that is the second tool we present in this work. MC$-$ALF is a Monte Carlo code based on nested sampling that, without the need for any prior guess or human intervention, can decompose metal lines into individual Voigt components. Both MC$-$ALF and NMF$-$PM are made publicly available to allow the community to produce large libraries of synthetic metal profiles and to reconstruct Voigt models of absorption lines in an automatic fashion. Both tools contribute to the scientific effort of simulating and analysing metal absorbers in very large spectroscopic surveys of quasars like the ongoing Dark Energy Spectroscopic Instrument (DESI), the 4-meter Multi-Object Spectroscopic Telescope (4MOST), and the WHT Enhanced Area Velocity Explorer (WEAVE) surveys., Comment: 22 pages, 19 figures. Abstract

Chaperone-assisted selective autophagy (CASA) is a highly selective pathway for the disposal of misfolding and aggregating proteins. In muscle, CASA assures muscle integrity by favoring the turnover of structural components damaged by mechanical strain. In neurons, CASA promotes the removal of aggregating substrates. A crucial player of CASA is HSPB8 (heat shock protein family B (small) member 8), which acts in a complex with HSPA, their cochaperone BAG3, and the E3 ubiquitin ligase STUB1. Recently, four novel HSPB8 frameshift (fs) gene mutations have been linked to neuromyopathies, and encode carboxy-terminally mutated HSPB8, sharing a common C-terminal extension. Here, we analyzed the biochemical and functional alterations associated with the HSPB8_fs mutant proteins. We demonstrated that HSPB8_fs mutants are highly insoluble and tend to form proteinaceous aggregates in the cytoplasm. Notably, all HSPB8 frameshift mutants retain their ability to interact with CASA members but sequester them into the HSPB8-positive aggregates together with two autophagy receptors SQSTM1/p62 and TAX1BP1. This copartitioning process negatively affects the CASA capability to remove its clients and causes a general failure in proteostasis response. Further analyses revealed that the aggregation of the HSPB8_fs mutants occurs independently of the other CASA members or from the autophagy receptors interaction, but it is an intrinsic feature of the mutated amino acid sequence. HSPB8_fs mutants aggregation alters the differentiation capacity of muscle cells and impairs sarcomere organization. Abstract

The search for neutrino events in correlation with gravitational wave (GW) events for three observing runs (O1, O2 and O3) from 09/2015 to 03/2020 has been performed using the Borexino data-set of the same period. We have searched for signals of neutrino-electron scattering with visible energies above 250 keV within a time window of 1000 s centered at the detection moment of a particular GW event. The search was done with three visible energy thresholds of 0.25, 0.8 and 3.0 MeV.Two types of incoming neutrino spectra were considered: the mono-energetic line and the spectrum expected from supernovae. The same spectra were considered for electron antineutrinos detected through inverse beta-decay (IBD) reaction. GW candidates originated by merging binaries of black holes (BHBH), neutron stars (NSNS) and neutron star and black hole (NSBH) were analysed separately. Additionally, the subset of most intensive BHBH mergers at closer distances and with larger radiative mass than the rest was considered. In total, follow-ups of 74 out of 93 gravitational waves reported in the GWTC-3 catalog were analyzed and no statistically significant excess over the background was observed. As a result, the strongest upper limits on GW-associated neutrino and antineutrino fluences for all flavors (\nu_e, \nu_\mu, \nu_\tau) have been obtained in the (0.5 - 5.0) MeV neutrino energy range., Comment: 13 pages, 8 figures

Abstract

We present a novel approach for the search of dark matter in the DarkSide-50 experiment, relying on Bayesian Networks. This method incorporates the detector response model into the likelihood function, explicitly maintaining the connection with the quantity of interest. No assumptions about the linearity of the problem or the shape of the probability distribution functions are required, and there is no need to morph signal and background spectra as a function of nuisance parameters. By expressing the problem in terms of Bayesian Networks, we have developed an inference algorithm based on a Markov Chain Monte Carlo to calculate the posterior probability. A clever description of the detector response model in terms of parametric matrices allows us to study the impact of systematic variations of any parameter on the final results. Our approach not only provides the desired information on the parameter of interest, but also potential constraints on the response model. Abstract

We use 28 quasar fields with high-resolution (HIRES and UVES) spectroscopy from the MUSE Analysis of Gas Around Galaxies survey to study the connection between Ly${\alpha}$ emitters (LAEs) and metal-enriched ionized gas traced by CIV in absorption at redshift $z\approx3-4$. In a sample of 220 CIV absorbers, we identify 143 LAEs connected to CIV gas within a line-of-sight separation ${\rm \pm 500\,km\,s^{-1}}$, equal to a detection rate of $36\pm 5$ per cent once we account for multiple LAEs connected to the same CIV absorber. The luminosity function of LAEs associated with CIV absorbers shows a $\approx 2.4$ higher normalization factor compared to the field. CIV with higher equivalent width and velocity width are associated with brighter LAEs or multiple galaxies, while weaker systems are less often identified near LAEs. The covering fraction in groups is up to $\approx 3$ times larger than for isolated galaxies. Compared to the correlation between optically-thick HI absorbers and LAEs, CIV systems are twice less likely to be found near LAEs especially at lower equivalent width. Similar results are found using SiIV as tracer of ionized gas. We propose three components to model the gas environment of LAEs: i) the circumgalactic medium of galaxies, accounting for the strongest correlations between absorption and emission; ii) overdense gas filaments connecting galaxies, driving the excess of LAEs at a few times the virial radius and the modulation of the luminosity and cross-correlation functions for strong absorbers; iii) an enriched and more diffuse medium, accounting for weaker CIV absorbers farther from galaxies., Comment: 28 pages, 21 figures, 10 tables. Abstract

Magnetic 2D materials hold promise to change the miniaturization paradigm of unidirectional photonic components. However, the integration of these materials in devices hinges on the accurate determination of the optical properties down to the monolayer limit, which is still missing. By using hyperspectral wide-field imaging we reveal a non-monotonic thickness dependence of the complex optical dielectric function in the archetypal magnetic 2D material CrI3 extending across different length scales: onsetting at the mesoscale, peaking at the nanoscale and decreasing again down to the single layer. These results portray a modification of the electronic properties of the material and align with the layer-dependent magnetism in CrI3, shedding light into the long-standing structural conundrum in this material. The unique modulation of the complex dielectric function from the monolayer up to more than 100 layers will be instrumental for understanding and manipulating the magneto-optical effects of magnetic 2D materials.

Abstract

Variability in disease onset and progression is a hallmark of amyotrophic lateral sclerosis (ALS), both in sporadic and genetic forms. Recently, we found that SOD1-G93A transgenic mice expressing the same amount of mutant SOD1 but with different genetic backgrounds, C57BL/6JOlaHsd and 129S2/SvHsd, show slow and rapid muscle wasting and disease progression, respectively. Here, we investigated the different molecular mechanisms underlying muscle atrophy. Although both strains showed similar denervation-induced degradation of muscle proteins, only the rapidly progressing mice exhibited early and sustained STAT3 activation that preceded atrophy in gastrocnemius muscle. We therefore investigated the therapeutic potential of sunitinib, a tyrosine kinase inhibitor known to inhibit STAT3 and prevent cancer-induced muscle wasting. Although sunitinib treatment reduced STAT3 activation in the gastrocnemius muscle and lumbar spinal cord, it did not preserve spinal motor neurons, improve neuromuscular impairment, muscle atrophy and disease progression in the rapidly progressing SOD1-G93A mice. Thus, the effect of sunitinib is not equally positive in different diseases associated with muscle wasting. Moreover, given the complex role of STAT3 in the peripheral and central compartments of the neuromuscular system, the present study suggests that its broad inhibition may lead to opposing effects, ultimately preventing a potential positive therapeutic action in ALS.

Abstract

Purpose: Sleep State Misperception (SSM) is described as the tendency of Insomnia Disorder (ID) patients to overestimate Sleep Latency (SL) and underestimate Total Sleep Time (TST). Literature exploring topographical components in ID with SSM is scarce and does not allow us to fully understand the potential mechanisms underlying this phenomenon. This study aims to evaluate the existence of sleep EEG topography alterations in ID patients associated with SSM compared to Healthy Controls (HC), focusing on two distinct periods: the Sleep Onset (SO) and the whole night. Methods: Twenty ID patients (mean age: 43.5 ± 12.7; 7 M/13F) and 18 HCs (mean age: 41.6 ± 11.9; 8 M/10F) underwent a night of Polysomnography (PSG) and completed sleep diaries the following morning upon awakening. Two SSM indices, referring to the misperception of SL (SLm) and TST (TSTm), were calculated by comparing objective and subjective sleep indices extracted by PSG and sleep diary. According to these indices, the entire sample was split into 4 sub-groups: ID +SLm vs HC –SLm; ID +TSTm vs HC –TSTm. Results: Considering the SO, the two-way mixed-design ANOVA showed a significant main effect of Groups pointing to a decreased delta/beta ratio in the whole scalp topography. Moreover, we found a significant interaction effect for the sigma and beta bands. Post Hoc tests showed higher sigma and beta power in anterior and temporo-parietal sites during the SO period in IDs +SLm compared to HC –SLm.Considering the whole night, the unpaired t-test revealed in IDs +TSTm significantly lower delta power during NREM, and lower delta/beta ratio index during NREM and REM sleep compared to HCs –TSTm.Finally, we found diffuse significant negative correlations between SSM indices and the delta/beta ratio during SO, NREM, and REM sleep. Conclusion: The main finding of the present study suggests that higher SL overestimation and TST underestimation are both phenomena related to diffuse cortical hyperarousal interpreted as a sleep state-independent electrophysiological correlate of the SSM, both during the SO and the whole night.

Abstract

We study the link between galaxies and HI-selected absorption systems at z~3-4 in the MUSE Analysis of Gas around Galaxies (MAGG) survey, an ESO large programme consisting of integral field pectroscopic observations of 28 quasar fields hosting 61 strong absorbers with $\rm N_{\rm HI}\gtrsim 10^{16.5}~\rm cm^{-2}$. We identify 127 Ly$\alpha$ emitting galaxies (LAEs) around the absorbers, corresponding to a detection rate of 82$\pm$16 per cent. The luminosity function of these LAEs is approximately 5 times higher in normalization than the field population and we detect a significant clustering of galaxies with respect to the gas, confirming that high column density absorbers and LAEs trace each other. Between 30 and 40 per cent of the absorbers are associated with multiple LAEs, which lie preferentially along filaments. Galaxies in groups also exhibit a three times higher covering factor of optically-thick gas compared to isolated systems. No significant correlations are identified between the emission properties of LAEs and the absorption properties of optically-thick gas clouds, except for a weak preference of brighter and multiple galaxies to reside near broad absorbers. Based on the measured impact parameters and the covering factor, we conclude that the near totality of optically-thick gas in the Universe can be found in the outer circumgalactic medium (CGM) of LAEs or in the intergalactic medium (IGM) in proximity to these galaxies. Thus, LAEs act as tracers of larger scale structures within which both galaxies and optically-thick clouds are embedded. The patchy and inhomogeneous nature of the CGM and IGM explains the lack of correlations between absorption and emission properties. Abstract

This report summarizes the findings of the CF1 Topical Subgroup to Snowmass 2021, which was focused on particle dark matter. One of the most important scientific goals of the next decade is to reveal the nature of dark matter (DM). To accomplish this goal, we must delve deep, to cover high priority targets including weakly-interacting massive particles (WIMPs), and search wide, to explore as much motivated DM parameter space as possible. A diverse, continuous portfolio of experiments at large, medium, and small scales that includes both direct and indirect detection techniques maximizes the probability of discovering particle DM. Detailed calibrations and modeling of signal and background processes are required to make a convincing discovery. In the event that a candidate particle is found through different means, for example at a particle collider, the program described in this report is also essential to show that it is consistent with the actual cosmological DM. Abstract

Dark matter lighter than 10 GeV/c$^2$ encompasses a promising range of candidates. A conceptual design for a new detector, DarkSide-LowMass, is presented, based on the DarkSide-50 detector and progress toward DarkSide-20k, optimized for a low-threshold electron-counting measurement. Sensitivity to light dark matter is explored for various potential energy thresholds and background rates. These studies show that DarkSide-LowMass can achieve sensitivity to light dark matter down to the solar neutrino floor for GeV-scale masses and significant sensitivity down to 10 MeV/c$^2$ considering the Migdal effect or interactions with electrons. Requirements for optimizing the detector's sensitivity are explored, as are potential sensitivity gains from modeling and mitigating spurious electron backgrounds that may dominate the signal at the lowest energies.

Abstract

Abstract Idiopathic REM sleep Behavior Disorder (iRBD) is a condition at high risk of developing Parkinson’s disease (PD) and other alpha-synucleinopathies. The aim of the study was to evaluate subtle turning alterations by using Mobile health technology in iRBD individuals without subthreshold parkinsonism. A total of 148 participants (23 persons with polysomnography-confirmed iRBD without subthreshold parkinsonism, 60 drug-naïve PD patients, and 65 age-matched controls were included in this prospective cross-sectional study. All underwent a multidimensional assessment including cognitive and non-motor symptoms assessment. Then a Timed-Up-and-Go test (TUG) at normal and fast speed was performed using mobile health technology on the lower back (Rehagait®, Hasomed, Germany). Duration, mean, and peak angular velocities of the turns were compared using a multivariate model correcting for age and sex. Abstract

Abstract Action observation plus motor imagery (AOMI) is a rehabilitative approach to improve gait and balance performance. However, limited benefits have been reported in older adults. Early sleep after motor practice represents a strategy to enhance the consolidation of trained skills. Here, we investigated the effects of AOMI followed by early sleep on gait and balance performance in older adults. Forty-five older adults (mean age: 70.4 ± 5.2 years) were randomized into three groups performing a 3-week training. Specifically, AOMI-sleep and AOMI-control groups underwent observation and motor imagery of gait and balance tasks between 8:00 and 10:00 p.m. or between 8:00 and 10:00 a.m. respectively, whereas Control group observed landscape video-clips. Participants were assessed for gait performance, static and dynamic balance and fear of falling before and after training and at 1-month follow-up. The results revealed that early sleep after AOMI training sessions improved gait and balance abilities in older adults compared to AOMI-control and Control groups. Furthermore, these benefits were retained at 1-month after the training end. These findings suggested that early sleep after AOMI may represent a safe and easy-applicable intervention to minimize the functional decay in older adults.

Abstract

Dark matter elastic scattering off nuclei can result in the excitation and ionization of the recoiling atom through the so-called Migdal effect. The energy deposition from the ionization electron adds to the energy deposited by the recoiling nuclear system and allows for the detection of interactions of sub-GeV/c$^2$ mass dark matter. We present new constraints for sub-GeV/c$^2$ dark matter using the dual-phase liquid argon time projection chamber of the DarkSide-50 experiment with an exposure of (12306 $\pm$ 184) kg d. The analysis is based on the ionization signal alone and significantly enhances the sensitivity of DarkSide-50, enabling sensitivity to dark matter with masses down to 40 MeV/c$^2$. Furthermore, it sets the most stringent upper limit on the spin independent dark matter nucleon cross section for masses below $3.6$ GeV/c$^2$., Comment: 7 pages, 3 figures

Abstract

We present a search for dark matter particles with sub-GeV/$c^2$ masses whose interactions have final state electrons using the DarkSide-50 experiment's (12306 $\pm$ 184) kg d low-radioactivity liquid argon exposure. By analyzing the ionization signals, we exclude new parameter space for the dark matter-electron cross section $\bar{\sigma}_e$, the axioelectric coupling constant $g_{Ae}$, and the dark photon kinetic mixing parameter $\kappa$. Abstract

We report on the search for dark matter WIMPs in the mass range below 10 GeV/c$^2$, from the analysis of the entire dataset acquired with a low-radioactivity argon target by the DarkSide-50 experiment at LNGS. The new analysis benefits from more accurate calibration of the detector response, improved background model, and better determination of systematic uncertainties, allowing us to accurately model the background rate and spectra down to 0.06 keV$_{er}$. A 90% C.L. exclusion limit for the spin-independent cross section of 3 GeV/c$^2$ mass WIMP on nucleons is set at 6$\times$10$^{-43}$ cm$^2$, about a factor 10 better than the previous DarkSide-50 limit. This analysis extends the exclusion region for spin-independent dark matter interactions below the current experimental constraints in the $[1.2, 3.6]$ GeV/c$^2$ WIMP mass range., Comment: 11 pages, 12 figures
