Abstract Anthocidaris crassispina is a very popular edible sea urchin distributed along the coast of the South China Sea. In this study, we performed whole-genome sequencing and generated a chromosome-level assembly of this species. The total length of the genomic contig sequence was 891.02 Mb, and contig N50 was 808.15 kb when Hifiasm was used for assembly. The Hi-C library was constructed and sequenced, yielding approximately 68.61 Gb of data. After Hi-C assembly, approximately 886.72 Mb of sequence was able to be mapped onto 21 chromosomes, accounting for 99.52% of the total genome length. Among the sequences located on the chromosomes, those for which the order and direction could be determined accounted for approximately 826.82 Mb, or 93.24% of the total length. These results provide valuable resources for further study of A. crassispina at the genetic level.

Detection of small, slow-moving targets, such as drones using Unmanned Aerial Vehicles (UAVs) poses considerable challenges to radar target detection and recognition technology. There is an urgent need to establish relevant datasets to support the development and application of techniques for detecting small, slow-moving targets. This paper presents a dataset for detecting low-speed and small-size targets using a multiband Frequency Modulated Continuous Wave (FMCW) radar. The dataset utilizes Ku-band and L-band FMCW radar to collect echo data from six UAV types and exhibits diverse temporal and frequency domain resolutions and measurement capabilities by modulating radar cycles and bandwidth, generating an LSS-FMCWR-1.0 dataset (Low Slow Small, LSS). To further enhance the capability for extracting micro-Doppler features from UAVs, this paper proposes a method for UAV micro-Doppler extraction and parameter estimation based on the local maximum synchroextracting transform. Based on the Short Time Fourier Transform (STFT), this method extracts values at the maximum energy point in the time-frequency domain to retain useful signals and refine the time-frequency energy representation. Validation and analysis using the LSS-FMCWR-1.0 dataset demonstrate that this approach reduces entropy on an average by 5.3 dB and decreases estimation errors in rotor blade length by 27.7% compared with traditional time-frequency methods. Moreover, the proposed method provides the foundation for subsequent target recognition efforts because it balances high time-frequency resolution and parameter estimation capabilities.

Abstract Background HIV-tuberculosis (HIV-TB) co-infection is a significant public health concern worldwide. TB delay, consisting of patient delay, diagnostic delay, treatment delay, increases the risk of adverse anti-TB treatment (ATT) outcomes. Except for individual level variables, differences in regional levels have been shown to impact the ATT outcomes. However, few studies appropriately considered possible individual and regional level confounding variables. In this study, we aimed to assess the association of TB delay on treatment outcomes in HIV-TB co-infected patients in Liangshan Yi Autonomous Prefecture (Liangshan Prefecture) of China, using a causal inference framework while taking into account individual and regional level factors. Methods We conducted a study to analyze data from 2068 patients with HIV-TB co-infection in Liangshan Prefecture from 2019 to 2022. To address potential confounding bias, we used a causal directed acyclic graph (DAG) to select appropriate confounding variables. Further, we controlled for these confounders through multilevel propensity score and inverse probability weighting (IPW). Results The successful rate of ATT for patients with HIV-TB co-infection in Liangshan Prefecture was 91.2%. Total delay (OR = 1.411, 95% CI: 1.015, 1.962), diagnostic delay (OR = 1.778, 95% CI: 1.261, 2.508), treatment delay (OR = 1.749, 95% CI: 1.146, 2.668) and health system delay (OR = 1.480 95% CI: (1.035, 2.118) were identified as risk factors for successful ATT outcome. Sensitivity analysis demonstrated the robustness of these findings. Conclusions HIV-TB co-infection prevention and control policy in Liangshan Prefecture should prioritize early treatment for diagnosed HIV-TB co-infected patients. It is urgent to improve the health system in Liangshan Prefecture to reduce delays in diagnosis and treatment.

ABSTRACTThe influence of the powered effect on the aerodynamic performance of the blended-wing-body (BWB) takeoff configuration is analysed in this study by solving the three-dimensional Reynolds-averaged Navier–Stokes equation on a multiblock structured grid. The powered effect on the aerodynamic performance of the BWB300 configuration was investigated with through-flow nacelle (TFN) and powered nacelle (PN) using this method. The results showed that although the influence of the powered effect caused a slight change in the aerodynamic performance of the BWB takeoff configuration, it did not change the development trend of the aerodynamic performance of the BWB takeoff configuration. Therefore, the TFN model can be used to obtain the general development trend of the aerodynamic performance of the BWB takeoff configuration in the early stages of the BWB design. However, when more accurate aerodynamic values are required, the powered effect should be considered.

In advancing the practical application of photocatalytic degradation for pollutant removal, batch preparation of environmentally friendly photocatalysts plays a crucial role. Herein, we prepared C/P co-doped urchin-like TiO2 (C/P–TiO2) through a straightforward room temperature impregnation and atmospheric annealing process. Phytic acid served as the dopant precursor for introducing C and P onto the TiO2 surface. This doping process was verified through energy dispersive X-ray spectroscopy (EDX), electron spin resonance (ESR), and X-ray photoelectron spectroscopy (XPS) analysis. The C/P–TiO2 photocatalyst exhibited exceptional efficiency in degrading the target compound trimethoprim (TMP), achieving a degradation of 98% TMP within 60 min and a mineralization of 40% TMP within 120 min, which were respectively 2.4 and 1.6 times higher than that of pristine TiO2. The intermediate products of TMP were detected, and a plausible degradation pathway was proposed. Photoelectric characterization results also confirmed that C/P co-doping could effectively enhance the carrier separation efficiency of C/P–TiO2. The catalysts prepared using the scaled-up impregnation system and reusing the phytic acid solution consistently demonstrated stable catalytic performance, which confirmed the viability of the method for green and batch preparation of C/P co-doped TiO2. Notably, the obtained catalyst consistently exhibited high degradation efficacy in various real water bodies, highlighting its substantial potential for practical application. The green and batch preparation method of C/P–TiO2 introduces a novel approach to the practical application of photocatalysis technology in treatment of water contaminants.

Summary: Plant pathogens manipulate host development, facilitating colonization and proliferation. Ralstonia solanacearum is a soil-borne bacterial pathogen that penetrates roots and colonizes plants through the vascular system, causing wilting and death. Here, we find that RipAC, an effector protein from R. solanacearum, alters root development in Arabidopsis, promoting the formation of lateral roots and root hairs. RipAC interacts with CELLULOSE SYNTHASE (CESA)-INTERACTIVE PROTEIN 1 (CSI1), which regulates the activity of CESA complexes at the plasma membrane. RipAC disrupts CESA-CSI1 interaction, leading to a reduction in cellulose content, root developmental alterations, and a promotion of bacterial pathogenicity. We find that CSI1 also associates with the receptor kinase FERONIA, forming a complex that negatively regulates immunity in roots; this interaction, however, is not affected by RipAC. Our work reveals a bacterial virulence strategy that selectively affects the activities of a host target, promoting anatomical alterations that facilitate infection without causing activation of immunity.

Introduction: Neuromyelitis optica spectrum disorder (NMOSD) is a central nervous system demyelinating disease. Current therapy methods, however, have limited effect on acute attacks except for intravenous methylprednisolone (IVMP). Efgartigimod is a first-in-class novel human immunoglobulin G1 (IgG1) Fc fragment approved for the treatment of generalized myasthenia gravis. Its capacity to rapidly decrease serum IgG levels, including pathogenic autoantibodies, positions it as a potentially effective option for managing the acute phase of NMOSD. Case presentation: We report the case of a 59-year-old female patient with acute NMOSD, presenting with vision loss and numbness in all four limbs. Despite an initial inadequate response to intravenous methylprednisolone (IVMP), the addition of Efgartigimod to her treatment regimen led to rapid improvement, notably including a significant reduction in serum aquaporin-4 antibody titers, total IgG levels, and inflammation cytokine levels. Furthermore, no adverse events were reported during a four-month follow-up period. Conclusion: As an adjunct to glucocorticoid therapy, Efgartigimod has proven effective and safe for this patient. However, to ascertain its potential as a novel therapeutic option for acute NMOSD, larger-scale prospective clinical trials are required.

Spotted babylon were exposed to three different pH levels (7.0, 8.0 and 9.0) and four different concentrations of ammonia nitrogen (0.02, 1.02, 5.10 and 10.20 mg/L) in seawater to determine their acute toxicity and physiological responses to environmental fluctuation. The study evaluated four antioxidant enzymes: catalase (CAT), alkaline, superoxide dismutase (SOD), peroxidase (POD) and glutathione peroxidase (GSH-PX), and two immunoenzymes: acid phosphatase (ACP) and phosphatase (AKP). Over time, the immunoenzyme activity was significantly affected by pH and ammonia nitrogen concentration. After being exposed to pH and ammonia nitrogen, the spotted babylon showed signs of unresponsiveness to external stimuli, reduced vitality, slow movement, and an inability to maintain an upright position. Over time, the spotted babylon exhibited a trend of increasing and then decreasing GSH-PX, CAT, and SOD activities to adapt to the changing environment and enhance its immunity. On the contrary, the POD and ACP activities exhibited a decreasing trend initially, followed by an increasing trend over time and the AKP activity showed a gradual increase with time. The combined effect of pH and ammonia was found to be stronger than the effect of either factor alone. The interaction between pH and ammonia increased the activity of the spotted babylon antioxidant enzymes, induced oxidative stress, and reduced the ability of the spotted babylon's non-specific immune system to reverse it. Thus, the reverse-back of the spotted babylon was higher when pH and ammonia stress were dual than when pH or ammonia were single-factor stresses. The study results will establish a theoretical basis for analyzing the risk of multiple factors to the spotted babylon, and also enrich the basic information about the shellfish immune system.

This study investigated the impacts of light irradiation and polymer types on the leaching behavior of dissolved organic matter (DOM) from microplastics (MPs) in freshwater. Polypropylene had the highest leaching capacity of DOM after photoaging, followed by polystyrene (PS), polyamide (PA) and polyethylene terephthalate (PET). While similarly low levels of DOM were observed in the remaining 5 MP suspensions under UV irradiation and in almost all MP suspensions (except PA) under darkness. These suggest that the photooxidation of some buoyant plastics may influence the carbon cycling of nature waters. Among 9 MP-derived leachates, PET leachates had the highest chromophoric DOM concentration and aromaticity, probably owing to the special benzene rings and carbonyl groups in PET structures and its fast degradation rate. Protein-like substances were the primary fluorescent DOM in MP suspensions (except PS), especially in darkness no other fluorescent substances were found. Considering the bio-labile properties of proteins together, MPs regardless of floating or suspended in an aquatic environment may have prevalent long-term effects on microbial activities. Besides, from monomers to hexamers with newly formed chemical bonds were identified in UV-irradiated MP suspensions. These results will contribute to a deep insight into the potential ecological effects related to MP degradation.

Objective: Chondrocyte death is the hallmark of cartilage degeneration during osteoarthritis (OA). However, the specific pathogenesis of cell death in OA chondrocytes has not been elucidated. This study aims to validate the role of CDKN1A, a key programmed cell death (PCD)-related gene, in chondrogenic differentiation using a combination of single-cell and bulk sequencing approaches. Design: OA-related RNA-seq data (GSE114007, GSE55235, GSE152805) were downloaded from Gene Expression Omnibus database. PCD-related genes were obtained from GeneCards database. RNA-seq was performed to annotate the cell types in OA and control samples. Differentially expressed genes (DEGs) among those cell types (scRNA-DEGs) were screened. A nomogram of OA was constructed based on the featured genes, and potential drugs targeting the featured genes were predicted. The presence of key genes was confirmed using Real-Time Quantitative Polymerase Chain Reaction (RT-qPCR), Western blot (WB), and immunohistochemistry (IHC). Micromass culture and Alcian blue staining were used to determine the effect of CDKN1A on chondrogenesis. Results: Six cell types, namely HomC, HTC, RepC, preFC, FC, and RegC, were annotated in scRNA-seq data. Five featured genes (JUN, CDKN1A, HMGB2, DDIT3, and DDIT4) were screened by multiple biological information analysis methods. TAXOTERE had the highest ability to dock with DDIT3. Functional analysis indicated that CDKN1A was enriched in processes related to collagen catabolism and acts as a positive regulator of autophagy. Additionally, CDKN1A was found to be associated with several KEGG pathways, including those involved in acute myeloid leukemia and autoimmune thyroid disease. CDKN1A was confirmed down-regulated in the joint tissues of OA mouse model and OA model cell. Inhibiting the expression of CDKN1A can significantly suppress the differentiation of OA chondrocytes. Conclusion: Our findings highlight the critical role of CDKN1A in promoting cartilage formation in both in vivo and in vitro and suggest its potential as a therapeutic target for OA treatment.

In recent years, the fishery resources in China's coastal and inland waters have declined seriously, along with a significant decrease in the number of economic fish stocks in many natural waters. Enhancement and release are important measures to conserve fishery resources, protect biodiversity and replenish the fishery environment. Since the 1980s, enhancement and release have gradually become an important way to increase fishery resources in China. With the yearly increase in variety, quantity, and scale of enhancement and release, the problem with the low efficiency of enhancement and release has become increasingly prominent. Therefore, a scientific and reasonable variant of the enhancement and release technique is the key to ensure its successful role in resource proliferation, including prior scientific planning as well as an accurate evaluation post release. At present, there are many studies on release assessment methods. Ecopath model method and empirical assessment method are often used to carry out pre-release planning research. Ecopath model method determines the release quantity in the water area by calculating the ecological capacity of the release water area. However, this method, like the empirical assessment method has the disadvantage of an extensive assessment of the capacity limiting its accuracy in planning the release quantity. The commonly used methods for evaluating the effect of post-release include external tagging method and statistical evaluation method. The external tagging method causes certain damage to fish and has poor stability, making it unsuitable for large-scale enhancement and releasing. The statistical evaluation method needs regular follow-up post release, making it manpower-, resources- as well as cost-intensive. In order to formulate scientific enhancement and release strategies and improve the effect of breed enhancement and release, the EnhanceFish model was introduced in this study using Acanthopagrus latus as the research object, Nanlang water area as the research area, and considering the life cycle, growth rate, mortality, sexual maturity, heritability, recruitment, economic cost and effort as the study parameters. The enhancement and release model of A. latus in Nanlang water was constructed, and the optimal release size, ecological capacity, population structure, and economic benefits of A. latus were scientifically evaluated in order to provide scientific guidance for the rational and standardized formulation of enhancement and release strategies.In this study, the effects of spawning stock biomass, profit, and fishing effort on the ecological structure of A. latus were comprehensively considered, and an intuitive data model was constructed to determine the range of release quantity and fishing effort. The results showed that the natural recruitment amount increased with the increase in releasing quantity. When the release quantity was 2 million, the natural recruitment amount reached the progressive value of 1.6 million. Therefore, the number of A. latus released in Nanlang water area was 0-1.6 million. While evaluating the relationship between fishing effort and economic benefit before release, the profit was 1 000 CNY assuming the current fishing effort E=1. When the fishing effort was E < 1, the profit increased with the increase in fishing effort. When it was E > 1, the profit decreased with the increase in fishing effort. When it was E > 2, the profit was 2 000 CNY, which has no research value. Therefore, the limit of fishing effort E is 0-2.

Abstract Kidney renal clear cell carcinoma (KIRC) is the most prevalent type of kidney cancer and causes thousands of deaths each year. The prognosis for KIRC is poor. One critical factor is that the mechanism beneath KIRC is unclear. ORM1 is a reactant to acute inflammation. In this study, we demonstrated that methylation of ORM1 promoter was low and ORM1 was expressed significantly higher in KIRC. KIRC with higher ORM1 expression exhibited worse survival probability. Meanwhile, ORM1 was expressed higher in KIRC cell lines. When ORM1 was knocked down, cell proliferation ability was inhibited potently compared to the NC control. Cell migration as well as invasion ability were also suppressed dramatically. At molecular level, the expression of active caspase-3 and Bax was upregulated in ORM1-KD group while Bcl-2 downregulated. Moreover, CALR decreased following ORM1-KD and rescued expression of CALR increased Bcl-2 level but reduced the level of cleaved caspase-3 and Bax. Consistently, the apoptotic rate of 786-O and Caki-2 cells was upregulated in ORM1-KD but downregulated after CALR overexpression. The activity of caspase-3 was also regulated by ORM1-KD. In addition, the inhibition rate of sorafenib was enhanced in ORM1 KD group but reduced after overexpression of ORM1. Conclusively, ORM1 is clinically associated with progression of KIRC and regulates cell proliferation, migration, invasion, and apoptosis in KIRC. Moreover, ORM1 affects the efficiency of sorafenib in KIRC and regulates caspase-3 mediated cascades response through CALR molecule. This study provides us a new way to recognize the development and progression in KIRC.

The monitoring of pharmaceuticals, personal care products (PCPs), pesticides, and their metabolites through wastewater-based epidemiology (WBE) provides timely information on pharmaceutical consumption patterns, chronic disease treatment rates, antibiotic usage, and exposure to harmful chemicals. However, before applying them for quantitative WBE back-estimation, it is necessary to understand their stability in the sewer system to screen suitable WBE biomarkers thereby reducing research uncertainty. This study investigated the in-sewer stability of 140 typical pharmaceuticals, PCPs, pesticides, and their metabolites across 15 subcategories, using a series of laboratory sewer sediment and biofilm reactors. For the first time, stability results for 89 of these compounds were reported. Among the 140 target compounds, 61 biomarkers demonstrated high stability in all sewer reactors, while 41 biomarkers were significantly removed merely by sediment processes. For biomarkers exhibiting notable attenuation, the influence of sediment processes was generally more pronounced than biofilm, due to its stronger microbial activities and more pronounced diffusion or adsorption processes. Adsorption emerged as the predominant factor causing biomarker removal compared to biodegradation and diffusion. Significantly different organic carbon–water partitioning coefficient (Koc) and distribution coefficient at pH = 7 (logD) values were observed between highly stable and unstable biomarkers, with most hydrophobic substances (Koc > 100 or logD > 2) displaying instability. In light of these findings, we introduced a primary biomarker screening process to efficiently exclude inappropriate candidates, achieving a commendable 77 % accuracy. Overall, this study represents the first comprehensive report on the in-sewer stability of 89 pharmaceuticals, PCPs, pesticides, and their metabolites, and provided crucial reference points for understanding the intricate sewer sediment processes.

The purple sea urchin (Heliocidaris crassispina) is one of the main drivers of ecosystem dynamics, and its reproductive cycle plays an important role in population structure and size variability. In this study, we analyzed the population structure, gonadal developmental characteristics, reproductive cycle and other factors influencing wild purple sea urchins from December 2021 to November 2022 in Daya Bay, Guangdong, China. Abstract White organic light-emitting diodes (WOLEDs) is a new generation of lighting technology and has stimulated wide-ranging studies. Despite the advantage of simple device structure, single-emitting-layer WOLEDs (SEL-WOLEDs) still face the challenges of difficult material screening and fine energy level regulation. Herein, we report efficient SEL-WOLEDs with a sky-blue emitting cerium(III) complex Ce-TBO2Et and an orange-red emitting europium(II) complex Eu(Tp2Et)2 as the emitters, showing a maximum external quantum efficiency of 15.9% and Commission Internationale de l’Eclairage coordinates of (0.33, 0.39) at various luminances. To better understand the habitat preferences and behavioural ecology of Sparus latus, we performed an experiment using box-shaped reefs as habitat enrichment materials, allowing us to determine the behavioural strategies and drivers involved in the response to different enrichment structures. The results showed that the first contact time of S. latus was negatively correlated (Pearson’s correlation, p < 0.005) with the distribution rate in the artificial reef area. Enrichment structures affected the habitat preferences of S. latus, and there was a significant difference in the average distribution rate between the control and treatment groups (Adonis, p < 0.001). The opening ratio (Adonis, R2 = 0.36) explained the distribution difference of S. latus better than the opening shape (Adonis, R2 = 0.12). In the absence of an enrichment structure, S. latus remained more active during the daytime, exhibiting poor clustering, while in the presence of an enrichment structure, S. latus exhibited clustered movement at night. The opening ratio was negatively correlated with the average interindividual distance (Spearman’s correlation, p < 0.01) and showed a significant positive correlation with the average distribution rate in the reef area (Spearman’s correlation, p < 0.001), indicating that the reef opening ratio significantly affected the reef-tropism and clustering behaviours of S. latus. The light intensity was negatively correlated with the average distance moved, and the average speed (Spearman’s correlation, p < 0.05) was significantly positively correlated with the reef first contact time (Spearman’s correlation, p < 0.001), indicating that the light intensity affected the exploration and activity patterns of S. latus. These results provide a research basis for analysing the pattern and process of fish proliferation induced by artificial reef habitats.

As one of the most important components in rotating machinery, if bearings fail, serious disasters may occur. Therefore, the remaining useful life (RUL) prediction of bearings is of great significance. Health indicator (HI) construction and early fault detection play a crucial role in data-driven RUL prediction. Unfortunately, most existing HI construction methods require prior knowledge and preset trends, making it difficult to reflect the actual degradation trend of bearings. And the existing early fault detection methods rely on massive historical data, yet manual annotation is time-consuming and laborious. To address the above issues, a novel deep convolutional auto-encoder (CAE) based on envelope spectral feature extraction is developed in this work. A sliding value window is defined in the envelope spectrum to obtain initial health indicators, which are used as preliminary labels for model training. Subsequently, CAE is trained by minimizing the composite loss function. The proposed construction method can reflect the actual degradation trend of bearings. Afterwards, the autoencoder is pre-trained through contrast learning (CL) to improve its discriminative ability. The model that has undergone offline pre-training is more sensitive to early faults. Finally, the HI construction method is combined with the early fault detection method to obtain a comprehensive network for online health assessment and fault detection, thus laying a solid foundation for subsequent RUL prediction. The superiority of the proposed method has been verified through experiments.

With the maturation of digital radio frequency memory (DRFM) technology, various intra-pulse retransmission interference methods have emerged. These flexible and changeable retransmission interference methods pose significant challenges to radar detection tasks, particularly in modern battlefields. This paper proposes an attention-guided complex-valued transformer (AGCT) as a solution. First, the encoder maps the received signal contaminated by interference and noise into a high-dimensional space. Then, the dilated convolution block (DCB) group and attention block (AB) group in the mask estimator extract the delicate multi-scale features and large-scale features of the interference, respectively, to obtain a multidimensional space mask. Finally, the decoder restores interference to the time domain and outputs the estimated target echo using residual learning. Considering the characteristics of intra-pulse interference, we introduced the energy attention block (EAB) at the end of the DCBs and the ABs within our network. This addition ensures a heightened focus on extracting interference features. Furthermore, we implemented a curriculum learning strategy during the network training. This approach gradually acclimates the network to fit different types of retransmission interference, starting from simpler to more complex scenarios. Our extensive experiments, conducted under various conditions, have provided compelling evidence of the AGCT’s superior performance. Compared to the comparative network, the AGCT’s advantages are particularly pronounced under more harsh conditions, demonstrating its robustness and effectiveness.

Abstract In the past period, due to the rapid development of next-generation sequencing technology, accumulating evidence has clarified the complex role of the human microbiota in the development of cancer and the therapeutic response. More importantly, available evidence seems to indicate that modulating the composition of the gut microbiota to improve the efficacy of anti-cancer drugs may be feasible. However, intricate complexities exist, and a deep and comprehensive understanding of how the human microbiota interacts with cancer is critical to realize its full potential in cancer treatment. The purpose of this review is to summarize the initial clues on molecular mechanisms regarding the mutual effects between the gut microbiota and cancer development, and to highlight the relationship between gut microbes and the efficacy of immunotherapy, chemotherapy, radiation therapy and cancer surgery, which may provide insights into the formulation of individualized therapeutic strategies for cancer management. In addition, the current and emerging microbial interventions for cancer therapy as well as their clinical applications are summarized. Although many challenges remain for now, the great importance and full potential of the gut microbiota cannot be overstated for the development of individualized anti-cancer strategies, and it is necessary to explore a holistic approach that incorporates microbial modulation therapy in cancer.

Abstract Background Teriflunomide, the active metabolite of leflunomide, is a disease-modifying therapy drug used for the treatment of multiple sclerosis (MS), yet the complications associated with this drug remain not fully understood. Here we present the rare case of a 28-year-old female MS patient who developed subacute cutaneous lupus erythematosus (SCLE) following teriflunomide treatment. Though SCLE has been reported to be associated with leflunomide, the current report represents the first documented evidence demonstrating SCLE as a potential teriflunomide treatment-related complication. Additionally, a literature review on the leflunomide-induced SCLE was conducted to emphasize the association of SCLE with teriflunomide, specifically amongst the female demographic with a preexisting autoimmune diathesis. Case presentation A 28-year-old female first presented with MS symptoms in the left upper limb along with blurred vision in the left eye. Medical and family histories were unremarkable. The patient exhibited positive serum biomarkers including ANA, Ro/SSA, La/SSB, and Ro-52 antibodies. Relapsing–remitting MS was diagnosed according to the 2017 McDonald’s diagnostic criteria, and remission was achieved upon intravenous administration of methylprednisolone followed by teriflunomide sequential therapy. Three months post-teriflunomide treatment, the patient developed multiple facial cutaneous lesions. SCLE was subsequently diagnosed and was attributed to treatment-related complication. Interventions include oral administration of hydroxychloroquine and tofacitinib citrate effectively resolved cutaneous lesions. Discontinuation of hydroxychloroquine and tofacitinib citrate treatment led to recurring SCLE symptoms under continuous teriflunomide treatment. Full remission of facial annular plaques was achieved after re-treatment with hydroxychloroquine and tofacitinib citrate. The patient’s clinical condition remained stable in long-term outpatient follow-ups. Conclusions As teriflunomide has become a standard disease-modifying therapy for MS, the current case report highlights the importance of monitoring treatment-related complications, specifically in relation to SCLE symptoms.

BackgroundThe COVID-19 pandemic has significantly altered the global health and medical landscape. In response to the outbreak, Chinese hospitals have established 24-hour fever clinics to serve patients with COVID-19. The emergence of these clinics and the impact of successive epidemics have led to a surge in visits, placing pressure on hospital resource allocation and scheduling. Therefore, accurate prediction of outpatient visits is essential for informed decision-making in hospital management. ObjectiveHourly visits to fever clinics can be characterized as a long-sequence time series in high frequency, which also exhibits distinct patterns due to the particularity of pediatric treatment behavior in an epidemic context. This study aimed to build models to forecast fever clinic visit with outstanding prediction accuracy and robust generalization in forecast horizons. In addition, this study hopes to provide a research paradigm for time-series forecasting problems, which involves an exploratory analysis revealing data patterns before model development. MethodsAn exploratory analysis, including graphical analysis, autocorrelation analysis, and seasonal-trend decomposition, was conducted to reveal the seasonality and structural patterns of the retrospective fever clinic visit data. The data were found to exhibit multiseasonality and nonlinearity. On the basis of these results, an ensemble of time-series analysis methods, including individual models and their combinations, was validated on the data set. Root mean square error and mean absolute error were used as accuracy metrics, with the cross-validation of rolling forecasting origin conducted across different forecast horizons. ResultsHybrid models generally outperformed individual models across most forecast horizons. A novel model combination, the hybrid neural network autoregressive (NNAR)-seasonal and trend decomposition using Loess forecasting (STLF), was identified as the optimal model for our forecasting task, with the best performance in all accuracy metrics (root mean square error=20.1, mean absolute error=14.3) for the 15-days-ahead forecasts and an overall advantage for forecast horizons that were 1 to 30 days ahead. ConclusionsAlthough forecast accuracy tends to decline with an increasing forecast horizon, the hybrid NNAR-STLF model is applicable for short-, medium-, and long-term forecasts owing to its ability to fit multiseasonality (captured by the STLF component) and nonlinearity (captured by the NNAR component). The model identified in this study is also applicable to hospitals in other regions with similar epidemic outpatient configurations or forecasting tasks whose data conform to long-sequence time series in high frequency exhibiting multiseasonal and nonlinear patterns. However, as external variables and disruptive events were not accounted for, the model performance declined slightly following changes in the COVID-19 containment policy in China. Future work may seek to improve accuracy by incorporating external variables that characterize moving events or other factors as well as by adding data from different organizations to enhance algorithm generalization.

In order to understand the effects of fermented Astragalus membranaceus (FAM) on the liver and intestinal health of tiger grouper (Epinephelus fuscoguttatus), this study was conducted. This study evaluates the effects of different levels of FAM on liver and intestinal tissue structure, serum biochemical parameters, intestinal digestive enzyme, and microbiota structure of tiger grouper. Fish were fed with diets (crude protein ≥ 48.0%, crude fat ≥ 10.0%) with five levels of FAM (L1:0.25%, L2: 0.5%, L3: 1%, L4: 2% and L5: 4%) in the experimental groups and a regular diet was used as the control (L0: 0%) for 8 weeks. Compared with AM, the protein content of FAM was significantly changed by 34.70%, indicating that a large amount of bacterial protein was produced after AM fermentation, and its nutritional value was improved. FAM had significant effects on the growth performance of tiger grouper (p < 0.05). The high-density lipoprotein cholesterol (HDL-C) was highest in L4 group, being significantly different from L0 group. The area and diameter of hepatocytes were lowest in L3 and L4, and the density of hepatocyte was highest in L4 group and relatively decreased in L5 group. The mucosal height and muscular thickness were highest in L3 group. The intestinal microbiota structure of tiger grouper was changed under the intervention of FAM. The lower abundance of potential pathogenic bacteria and higher abundance of probiotics colonization in the L4 group showed that the dose of FAM had the best effect on improving the health of intestinal microbiota. This study indicates that the addition of FAM in the feed contributes to liver health, improves intestinal morphology, and regulates the intestinal microbiota of tiger grouper. The addition ratio of 1%–2% is better for intestinal and liver health, and a high addition ratio will cause liver damage. Our work will provide a reference for the addition and management of FAM in the aquaculture industry.

Purpose: Variations of cytokines and gut microbiota diversity with improved cognitive function in patients with obesity following bariatric surgery were poorly understood. The aim of this study was to testify the relationship among gut microbiota, cytokines and cognitive function in patients with obesity before and after laparoscopic sleeve gastrectomy (LSG). Methods: Forty patients were enrolled in this study. Demographics, and serum and stool specimens were collected from all patients before and 3 months after LSG. The Montreal Cognitive Assessment (MoCA) scale, as well as assessment of immediate and delayed memory were used to evaluate self-perceived cognitive improvement after LSG. Results: LSG resulted in significant weight loss and improvement in cognitive functions, as measured by questionnaires. Bariatric surgery tended to increase gut microbiota relative abundance and diversity. The intestinal flora increased in the proportion of Bacteroidetes and Fusobacteria phyla, and decreased in the proportion of Firmicutes, Proteobacteria, and Actinobacteria phyla after LSG. Plasma IL-1β and TNF-α levels were significantly decreased following LSG, while IL-4 was significantly increased. MoCA test scores were significant correlated with IL-4, TNF-α and IL-1β. In addition, Firmicutes had a positive correlation with TNF-α, while Fuscobacteria had a negative correlation with IL-1β. Bacteroidetes was negatively correlated with IL-4. Conclusion: Changes in gut microbiota were positive relationship with cognitive function improvement following LSG. Inflammation cytokines maybe played as a mediator between gut microbiota and cognitive function through gut-microbiota-brain axis.

Summary: Ralstonia solanacearum invades plants through their roots and causes devastating bacterial diseases in multiple crops. Here, we present a versatile inoculation assay in Arabidopsis thaliana seedlings grown in sterile agar plates. We describe steps for plant preparation, bacterial inoculation, tissue sampling, and bacterial quantification. Abstract Background Plant growth regulators are chemicals that regulate plant growth and development, which can regulate hormonal balance and affect plant growth, thereby increasing crop yield and improving crop quality. Our studies have revealed a new compound, GZU001, which could be used as a plant growth regulator. This compound has been observed to affect root elongation in maize significantly. However, the exact mechanism of this phenomenon is still being investigated. Results Metabolomics and proteomics were used in unison in this study to explore the response pathway and regulation mechanism of GZU001 in promoting maize root elongation. From the appearance, we can see that both roots and plants of maize treated with GZU001 are significantly improved. Maize root metabolism revealed 101 differentially abundant proteins and 79 differentially expressed metabolites. The current study identified altered proteins and metabolites associated with physiological and biochemical processes. GZU001 treatment has been demonstrated to promote primary metabolism, essential for carbohydrates, amino acids, energy, and secondary metabolism. The result suggests that the stimulation of primary metabolism is beneficial for the growth and development of maize and plays a significant role in sustaining metabolism and growth. Conclusions This study recorded the changes of related proteins and metabolites in maize roots after GZU001 treatment and provided evidence for this compound’s action mode and mechanism in plants.

Pedestrian re-identification is a crucial task in various safety applications, such as traffic management, collision avoidance, and emergency response. One of the challenging issues in pedestrian re-identification is how to identify the same object when the collection perspective changes or carrying different items. To address the above issues, this paper proposes a new gait based pedestrian re-identification method from the perspective of improving the representation ability of different pedestrians. The proposed method extracts spatial and temporal features separately from the gait data and designs a spatiotemporal feature fusion module at the top of the network to prevent features from interfering with each other in different domains. The temporal scoring strategy based on gated recurrent unit (GRU) is then used to evaluate the image quality of each frame in the network branch where temporal gait features are extracted. Finally, the proposed method is evaluated on two open datasets, CASIA-B and OU-ISIR-LP. Experimental results show that, compared with existing methods, the proposed method has a higher recognition rate under cross-view and scene conditions, such as carrying a bag and heavily clothed, in which the proposed method demonstrates better performance in pedestrian recognition. These results highlight the effectiveness and robustness of the proposed method and demonstrate its potential as a future research direction in pedestrian re-identification.

We propose to use a Few-Shot Learning (FSL) method for the pre-stack seismic inversion problem in obtaining a high resolution reservoir model from recorded seismic data. Recently, artificial neural network (ANN) demonstrates great advantages for seismic inversion because of its powerful feature extraction and parameter learning ability. Hence, ANN method could provide a high resolution inversion result that are critical for reservoir characterization. However, the ANN approach requires plenty of labeled samples for training in order to obtain a satisfactory result. For the common problem of scarce samples in the ANN seismic inversion, we create a novel pre-stack seismic inversion method that takes advantage of the FSL. The results of conventional inversion are used as the auxiliary dataset for ANN based on FSL, while the well log is regarded the scarce training dataset. According to the characteristics of seismic inversion (large amount and high dimensional), we construct an arch network (A-Net) architecture to implement this method. An example shows that this method can improve the accuracy and resolution of inversion results.

Abstract The combined effects of temperature and salinity on the digestion and respiration metabolism of Pinctada fucata were evaluated via response surface methodology and box-benhnken design under laboratory condition. Results indicated that the primary and secondary effects of salinity and temperature had significant effects on amylase (AMS) of P. fucata (P 0.05), but had a significant effect on TRYP as a secondary effect (P 0.05). To study the effects of light color on sea urchin (Tripneustes gratilla), blue light (B, λ450nm), yellow light (Y, λ585–590nm), red light (R, λ640nm), green light (G, λ510nm), white light (W, λ400–780nm), and darkness (H) groups were established in a recirculating seawater aquaculture system. Six different LED light color treatment groups with a photoperiod of 12 L:12 D were tested for 30 days to investigate the effects of different light colors on the feeding, growth, and enzyme activities of T. gratilla (142.45 ± 4.36 g). We found that using different LED light colors caused significantly different impacts on the feeding, growth, and enzyme activity of T. gratilla. Notably, the sea urchins in group B exhibited better growth, with a weight gain rate of 39.26%, while those in group R demonstrated poorer growth, with a weight gain rate of only 26%. The feeding status differed significantly (p < 0.05) between groups B and R, with group B consuming the highest daily intake (6.03 ± 1.69 g) and group R consuming the lowest (4.54 ± 1.26 g). Throughout the three phases, there was no significant change in the viability of the α-amylase (p > 0.05). Conversely, the pepsin viability significantly increased (p < 0.05) in group B. The lipase viability consistently remained at the lowest level, with no notable differences between group W and group B. In group R, both the α-amylase and pepsin viabilities remained lower, whereas the lipase viability was noticeably greater in each phase than in group B (p < 0.05). Among the antioxidant enzymes, group R exhibited a trend of initial increase followed by decreases in catalase, superoxide dismutase, and glutathione peroxidase activities, particularly during the third stage (15–30 days), during which a significant decrease in antioxidant enzyme activity was observed (p < 0.05). Taken together, these findings suggest that blue light positively affects the growth, feeding, digestion, and antioxidant capacity of T. gratilla in comparison with those in other light environments, whereas red light had an inhibitory effect. Furthermore, T. gratilla is a benthic organism that lives on shallow sandy sea beds. Thus, as short wavelengths of blue and green light are more widely distributed on the seafloor, and long wavelengths of red light are more severely attenuated on the seafloor, shorter wavelengths of light promote the growth of bait organisms of sea urchins, which provide better habitats for T. gratilla.

Laser cladding is one of the emerging additive manufacturing technologies and has been adopted in various industrial fields. In this study, the morphological characteristics of a single clad of Inconel 718 manufactured by coaxial laser cladding with high laser power from 4200 W to 5400 W, powder feeding rate from 25 g/min to 50 g/min, and cladding speed from 20 mm/s to 50 mm/s are studied. The cross-section of the melt pool is analyzed and classified by type into three types: shallow dilution, flat dilution, and fluctuating dilution. Nine parameters are designed to describe the morphological characteristics of the clad, and the corresponding linear regression models are developed to establish a quantitative relationship between the combined process parameters and morphological characteristics. The results indicate that the total area of the cross-section A, the clad area above the substrate Ac, the area of the molten substrate Am, the total height of the cross-section H, the height of the clad above the substrate hc, the penetration depth hm, the clad width W, the dilution ratio D, and the wetting angle θ are determined by complex coupling of energy input and mass accumulation, and they are proportional to PF0.4/V, P0.5F/V, P/F0.2/V0.4, P2F0.6/V, PF0.7/V, P2/F/V0.3, P/V0.8, P/FV0.2, and PF7/V0.8, respectively. The large linear regression coefficients and the analysis residuals indicate the high reliability of the statistical linear regression models. This work aims to provide a comprehensive understanding of the influence of the main processing parameters on the morphological characteristics of the clad, which is of great value in providing a reference and laying a basis for the practical application of laser cladding technology at a high deposition rate.

LR004 is a novel chimeric (human/mouse) monoclonal antibody developed for the treatment of advanced colorectal carcinoma with detectable epidermal growth factor receptor (EGFR) expression. We aimed to investigate the preclinical pharmacokinetics (PK) and in vivo biodistribution of LR004. The PK profiles of LR004 were initially established in rhesus monkeys. Subsequently, 125I radionuclide-labeled LR004 was developed and the biodistribution, autoradiography, and NanoSPECT/CT of 125I-LR004 in xenograft mice bearing A431 tumors were examined. The PK data revealed a prolonged half-life and nonlinear PK characteristics of LR004 within the dose range of 6–54 mg/kg. The radiochemical purity of 125I-LR004 was approximately 98.54%, and iodination of LR004 did not affect its specific binding activity to the EGFR antigen. In a classical biodistribution study, 125I-LR004 exhibited higher uptake in highly perfused organs than in poorly perfused organs. Prolonged retention properties of 125I-LR004 in tumors were observed at 4 and 10 days. Autoradiography and NanoSPECT/CT confirmed the sustained retention of 125I-LR004 at the tumor site in xenograft mice. These findings demonstrated the adequate tumor targeting capabilities of 125I-LR004 in EGFR-positive tumors, which may improve dosing strategies and future drug development.

In this study, we sought to characterize the effect of water vibration frequency stress on crimson snapper (Lutjanus erythropterus) survival to determine an optimal transportation speed. To achieve this, we used a transport tank (25 cm × 17 cm × 16 cm) to simulate the transport process. After 8 h at five different vibration frequencies (D1 = 75 rpm, D2 = 105 rpm, D3 = 135 rpm, D4 = 165 rpm, and D5 = 195 rpm), the pH and dissolved oxygen (DO) levels in the tanks decreased; ammonia nitrogen levels (NH4-N) and temperature (T) increased with increasing density; and significant changes in oxidative stress biomarkers, digestive enzymes, and ATPase levels were observed in crimson snapper fry. The enzyme activity increased and reached the maximum value at 195 rpm. The experimental results suggested that during the actual transport, when using transport tanks, the length of the transport time was less than 8 h, and setting the vibration frequency for transportation at 135 rpm was more appropriate, that is, a speed of 50 km/h for transporting crimson snapper fry.

Abstract To investigate the differential expression of genes in whole transcripts of congenital pulmonary airway malformation (CPAM) using second-generation sequencing (also known as next-generation sequencing, NGS) technology. Children with CPAM were strictly screened after setting the criteria, and grouped by taking CPAM parietal tissue and CPAM lesion tissue respectively, and RNA-Seq libraries were established separately using second-generation sequencing technology, followed by differential expression analysis and GO (gene ontology) functional enrichment analysis, KEGG (Kyoto encyclopedia of genes and genomes, a database) pathway analysis and GSEA (Gene Set Enrichment Analysis) analysis. Five cases were screened from 36 children with CPAM, and high-throughput sequencing was performed to obtain 10 whole transcripts of samples with acceptable sequence quality and balanced gene coverage. One aberrantly expressed sample (3b) was found by analysis of principal components, which was excluded and then subjected to differential expression analysis, and 860 up-regulated genes and 203 down-regulated genes. GO functional enrichment analysis of differentially expressed genes demonstrates the functional class and cellular localization of target genes. The whole transcript of CPAM shows obvious gene up and down-regulation, differentially expressed genes are located in specific cells and belong to different functional categories, and NGS can provide an effective means to study the transcriptional regulation of CPAM from the overall transcriptional level.

Abstract The types and numbers of components in end-of-life (EOL) products are often uncertain during remanufacturing, leading to low disassembly efficiencies for traditional remanufacturing disassembly lines. To address this problem, a parallel mixed-flow workstation layout was designed, and a novel parallel mixed remanufacturing disassembly line balancing optimization method for multi-variety products was proposed. A mixed-flow product disassembly task hierarchical assignment matrix was constructed to perform disassembly task allocations for similar components. Furthermore, a parallel mixed-flow remanufacturing disassembly line balancing (PMRDLB) optimization model was developed with the optimization objectives of minimizing the number of workstations, the disassembly line balancing rate, and the remanufacturing value indexes of the components. Furthermore, the multi-objective non-dominated genetic optimization method (NSGA-III) was improved, in which a chromosome construction method, based on the parallel mixed-flow disassembly task allocation matrix, was proposed to conduct mapping between the chromosomes and the PMRDLB model. In addition, non-dominated solution sorting was performed based on a Pareto hierarchy, which increased the searching rate of the algorithm during optimization. Finally, a case study verified the effectiveness and feasibility of the proposed method.

Abstract Magnetic solid‐phase extraction (MSPE) strategy based on the Fe3O4@PDA/MIL‐101(Cr) has been proposed to separate and purify five common mycotoxins in licorice, including aflatoxin B1, aflatoxin G1, sterigmatocystin, zearalenone, and ochratoxin A. Integrating the MSPE and solid–liquid extraction/partitioning, a modified QuEChERS was established to adapt to the complex licorice samples. The Fe3O4@PDA/MIL‐101(Cr) was successfully synthesized and characterized by Fourier transform infrared spectroscopy (FT‐IR), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and nitrogen adsorption–desorption isotherms. Sorbents with superior advantages for exclusion of matrix interference and extraction of target analytes in a short time were obtained, according to their ability of magnetic separation, high surface area (287.75 m2/g), large pore volume (0.61 cm3/g), and nanosized structure with mesopores. Prior to analysis with ultrahigh‐performance liquid chromatography‐tandem mass spectrometry (UHPLC‐MS/MS), several key parameters that would affect the sorbents’ extraction efficiency were extensively investigated. Under the optimized conditions, the practicality of the developed method for analysis of mycotoxins in licorice samples was confirmed by adequate linearity (R2 ≥ 0.9967), high sensitivity (LODs and LOQs, respectively, in the ranges 0.01–0.09 and 0.02–0.30 μg/kg), acceptable recovery (78.53%–116.28%), satisfactory reusability, and good interbatch precision of the sorbents (RSDs in the ranges 6.70%–11.20% and 6.02%–10.35%, respectively). The results indicated that the established method was feasible and reliable for the environment‐friendly and rapid screening of mycotoxins in complex licorice samples.

Abstract Background Little is known about how the obesogenic environment influences emotional states associated with glial responses and neuronal function. Here, we investigated glial reactivation and neuronal electrophysiological properties in emotion-related brain regions of high-fat diet (HFD) and ob/ob mice under chronic stress. Methods The glial reactivation and neuronal activities in emotion-related brain regions were analyzed among normal diet mice (ND), HFD mice, wild-type mice, and ob/ob mice. To further activate or inhibit astrocytes in medial prefrontal cortex (mPFC), we injected astrocytes specific Gq-AAV or Gi-AAV into mPFC and ongoing treated mice with CNO. Results The results showed that obesogenic factors per se had no significant effect on neuronal activities in emotion-related brain regions, or on behavioral performance. However, exposure to a chronic stressor profoundly reduced the frequency of spontaneous inhibitory postsynaptic currents (sIPSCs) and spontaneous excitatory postsynaptic currents (sEPSCs) in the mPFC; depressive-like behaviors were seen, accompanied by significant upregulation of astrocyte reactivation. We identified resilient and susceptible mice among chronic social defeat stress-exposed HFD mice. As expected, astrocyte reactivity was upregulated, while neuronal activity was depressed, in the mPFC of susceptible compared to resilient mice. Furthermore, activating astrocytes resulted in similar levels of neuronal activity and depressive-like behaviors between resilient and susceptible mice. Additionally, inhibiting astrocyte reactivation in the mPFC of HFD mice upregulated neuronal activities and inhibited depressive-like behaviors. Conclusions These observations indicate that obesogenic factors increase the risk of depression, and improve our understanding of the pathological relationship between obesity and depression. The fatty acid composition in organisms can reflect the trophic level, feeding habits, and utilization of local resources. In the present study, the living resources of different functional areas (artificial reef area, shellfish area, macroalgae area) in the Zhelin Bay Marine Ranch were investigated, and fatty acid analysis was used to elucidate the trophic relations across the food web. The results showed that 22:6ω3 FA (docosahexaenoic acid, DHA), 20:5ω3 FA (eicosapentaenoic acid, EPA), 16:1ω7 FA, and 16:0 FA are fatty acid biomarkers that distinguish carnivorous, omnivorous, and herbivorous organisms. The ratios of DHA/EPA, polyunsaturated fatty acids/saturated fatty acids (PUFA/SFA), and sum of ω3 fatty acids/sum of ω6 fatty acids (Σω3/Σω6) can be used as an important basis to assess the trophic level and feeding habits of organisms. In the comparison of the food web structure of different functional areas, the DHA/EPA ratio of carnivorous organisms in the Artificial Reef area was higher than that in other functional areas due to the abundant living resources, indicating that the food web of the Artificial Reef area presents lower stability to cope with perturbations. Furthermore, MixSIAR was used to quantitatively estimate the diet composition of consumers in the Zhelin Bay Marine Ranch based on their fatty acids. The results of the present study are a valuable contribution to understanding the trophic relationships in the Zhelin Bay Marine Ranch and provide theoretical support for future planning and construction of marine ranches.

Abstract It is anticipated that perovskite solar cells (PSCs) will overtake other products in the market for next‐generation photovoltaics. The optical loss, however, continues to be a flaw that restricts the photocurrent (Jph) of PSCs. Mesoporous antireflection coatings (ARCs), both monolayer and multilayer, are designed using a combination of the finite element method and equivalent medium theory, and ARCs models are merged with PSCs. In the current work, mesoporous ARCs are made, the optical performance of the device is evaluated using optical modeling, and then the ARCs are integrated into solar cells. The simulation results show that the Jph of planar inverted PSCs can increase to 24.00 mA cm−2 when the front surface of PSCs adopts mesoporous ARC (via parameter optimization and sensible arrangement and combination). An increase of 0.98 mA cm−2 in Jph of PSCs is observed in comparison with flat ARC (23.02 mA cm−2). The strong light transmission and low reflection properties of the mesoporous ARCs are confirmed by the optimized solution. It is important to note that the first fusion of mesoporous and multilayer ARC offers a fresh approach to the development of perovskite and perovskite/silicon tandem solar cells with extremely high efficiency.

Traffic congestion has been widely perceived as an inevitable byproduct in the process of global urbanization, leading to significant negative economic and environmental impacts. Existing studies have attached significant importance to revealing the interaction between traffic congestion patterns and built environment features in many metropolises with little attention, if any, paid to their counterparts of satellite cities (SC). For being aware that developing SCs has been a main trend to address many urban issues of metropolises in the rapid urbanization process of emerging economies, this study makes one of the first research attempts to investigate how traffic congestion temporally and spatially evolves with the built environment characterized by land use and transportation network features in Foshan, a typical SC of Guangzhou in China by using valuable hyperlocal travel data. The results show that while weekdays experience traffic peaks in both morning and evening, weekends or holidays generally only have the evening peak. The traffic congestion in Foshan during the pre-weekend is found much heavier compared with normal weekdays and weekends with a long-lasting effect possibly due to Foshan's role as a SC of Guangzhou. Additionally, the rising traffic congestion in a SC associated with company land use during the peak hours can be partly offset by the increasing supply of public transit, suggesting urban planners increase the density of public transit where there is a denser distribution of companies. This study provides new knowledge on travel behaviors in SCs in emerging economies, supplying urban governors with new insights for improving their traffic conditions generally.

BackgroundChest x-ray (CXR) is widely applied for the detection and diagnosis of children's lung diseases. Lung field segmentation in digital CXR images is a key section of many computer-aided diagnosis systems.ObjectiveIn this study, we propose a method based on deep learning to improve the lung segmentation quality and accuracy of children's multi-center CXR images.MethodsThe novelty of the proposed method is the combination of merits of TransUNet and ResUNet. The former can provide a self-attention module improving the feature learning ability of the model, while the latter can avoid the problem of network degradation.ResultsApplied on the test set containing multi-center data, our model achieved a Dice score of 0.9822.ConclusionsThis novel lung segmentation method proposed in this work based on TransResUNet is better than other existing medical image segmentation networks.

The direct biological effects of radiofrequency electromagnetic radiation (RF-EMR) from wireless communication equipment on the testes are still unclear. Our previous study proved that long-term exposure to 2605 MHz RF-EMR gradually damage spermatogenesis and resulted in time-dependent reproductive toxicity by directly disrupting blood-testis barrier circulation. Although short-term exposure did not cause readily observable damage to fertility, whether it caused specific biological effects and how these effects contributed to the time-dependent reproductive toxicity of RF-EMR were currently unknown. Studies on this issue are important for elucidating the time-dependent reproductive toxicity of RF-EMR. The present study established a 2605 MHz RF-EMR (SAR=1.05 W/Kg) scrotal exposure model with rats and extracted primary Sertoli cells for exposure to investigate the direct biological effects of short-term RF-EMR exposure on the testis. The results showed that short-term RF-EMR exposure did not decrease sperm quality and spermatogenesis, but it increased the levels of testicular testosterone (T) and zinc transporter 9 (ZIP9) in Sertoli cells of rats. In vitro, 2605 MHz RF-EMR exposure did not increase the apoptosis rate of Sertoli cells, but it increased the apoptosis rate and MDA of Sertoli cells exposed to H2O2. T reversed these changes and increased ZIP9 level in Sertoli cells, whereas inhibiting ZIP9 expression significantly suppressed these T-mediated protective effects. Moreover, T increased the levels of phosphorylated inositol-requiring enzyme 1 (P-IRE1), phosphorylated protein kinase R (PKR)-like endoplasmic reticulum kinase (P-PERK), phosphorylated eukaryotic initiation factor 2a (P-eIF2a) and phosphorylated activating transcription factor 6 (P-ATF6) in Sertoli cells, and these effects were reversed by ZIP9 inhibition. With prolonged exposure time, testicular ZIP9 was gradually downregulated, and testicular MDA increased. ZIP9 level was negatively correlated with MDA level in the testes of exposed rats. Thus, although short-term exposure to 2605 MHz RF-EMR (SAR=1.05 W/kg) did not significantly disturb spermatogenesis, it suppressed the ability of Sertoli cells to resist external insults, which was rescued by enhancing the ZIP9-centered androgen pathway in the short term. Increasing the unfolded protein response might be an important downstream mechanism involved. These results promote a better understanding of the time-dependent reproductive toxicity of 2605 MHz RF-EMR.

Free-text data with long duration and continuity have great potential for studying environmental concerns and emotional expressions in child development. Based on ecosystem theory, using topic modeling and sentiment analysis in text mining to mine 4556 free-text writing data from first to sixth grade in elementary school, this study aims to reveal concerned topics’ evolutionary trends and sentiment expression differences in topics during elementary school children’s development. The results show the following: (1) Children prefer to focus on the topics of school and family in elementary school; (2) With the growth of grades, the proportion of family topics continues to decline, while that of social culture topics keeps rising; (3) When describing school, family, social culture, and interest, children mostly express negative emotions, and when describing peers and ability they mostly express positive emotions; (4) As the grade increases, the emotional expression on social culture topics become negative, while that on ability and interest topics become positive, and there are more differences in emotion expression between topics in junior and senior elementary grades. Discussion and conclusion are discussed at the end.

This study investigates the effect of a sudden change in salinity for 48 h on the digestive enzyme activity of juvenile yellowfin tuna. The treatment included a control salinity of 32‰ in natural seawater and an experimental salinity of 29‰. Acute stress experiments were carried out on 72 juvenile yellowfin tuna (646.52 ± 66.32 g) for 48 h to determine changes in digestive enzyme activity in different intestinal sections over time (0 h, 12 h, 24 h, 48 h). The activities of pepsin, trypsin, α-amylase, lipase, and chymotrypsin in the digestive organs (stomach, foregut, and pyloric ceca) of juvenile yellowfin tuna were measured. Pepsin and pancreatic protease in the experimental group were significantly lower than in the control group (p < 0.05). α-amylase showed a fluctuating trend of decreasing and then increasing, and its activity trend was pyloric ceca > foregut > stomach. The lipase activity of gastric tissues decreased at the beginning and then increased, reaching a minimum at 24 h (2.74 ± 1.99 U·g protein−1). The change of lipase in the pyloric ceca and foregut was increasing and then decreasing. The lipase activity trend was pyloric ceca > foregut > stomach. The chymotrypsin showed a decreasing and increasing trend and then stabilized at 48 h with a pattern of pyloric ceca > foregut > stomach. Similarly, the gut villi morphology was not significantly altered in the acutely salinity-stressed compared to the non-salinity-stressed. This study suggests that salinity may change the digestive function of juvenile yellowfin tuna, thereby affecting fish feeding, growth, and development. On the contrary, yellowfin tuna is highly adapted to 29‰ salinity. However, excessive stress may negatively affect digestive enzyme activity and reduce fish digestibility. This study may provide a scientific basis for a coastal aquaculture water environment for yellowfin tuna farming, which may guide the development and cultivation of aquaculture.

The pathology is decisive for disease diagnosis but relies heavily on experienced pathologists. In recent years, there has been growing interest in the use of artificial intelligence in pathology (AIP) to enhance diagnostic accuracy and efficiency. However, the impressive performance of deep learning-based AIP in laboratory settings often proves challenging to replicate in clinical practice. As the data preparation is important for AIP, the paper has reviewed AIP-related studies in the PubMed database published from January 2017 to February 2022, and 118 studies were included. An in-depth analysis of data preparation methods is conducted, encompassing the acquisition of pathological tissue slides, data cleaning, screening, and subsequent digitization. Expert review, image annotation, dataset division for model training and validation are also discussed. Furthermore, we delve into the reasons behind the challenges in reproducing the high performance of AIP in clinical settings and present effective strategies to enhance AIP’s clinical performance. The robustness of AIP depends on a randomized collection of representative disease slides, incorporating rigorous quality control and screening, correction of digital discrepancies, reasonable annotation, and sufficient data volume. Digital pathology is fundamental in clinical-grade AIP, and the techniques of data standardization and weakly supervised learning methods based on whole slide image (WSI) are effective ways to overcome obstacles of performance reproduction. The key to performance reproducibility lies in having representative data, an adequate amount of labeling, and ensuring consistency across multiple centers. Digital pathology for clinical diagnosis, data standardization and the technique of WSI-based weakly supervised learning will hopefully build clinical-grade AIP.

The aim of this study was to investigate the circadian rhythm of muscle-related gene expression in mackerel tuna under different weather conditions. The experiment was carried out under two weather conditions at four sampling times (6:00, 12:00, 18:00, and 24:00) to determine the expression of growth, function, and rhythm genes: white muscle rhythm genes were rhythmic on sunny and cloudy days, except for PER3 and RORA; all functional genes had daily rhythmicity. Red muscle had daily rhythmicity on both sunny and cloudy days; functional genes had daily rhythmicity except for MBNL. The expression levels of the rhythm gene PER1 were determined to be significantly different by independent t-test samples in white muscle at 6:00, 12:00, 18:00, and 24:00 under different weather conditions; the expression levels of the functional genes MBNL and MSTN were both significantly different. In the red muscle, the expression of the rhythm genes PER3, REVERBA, and BMAL1 was determined by independent t-test samples at 6:00, 12:00, 18:00, and 24:00 on cloudy and sunny days; the functional gene MBNL was significantly different. The present study showed that mackerel tuna muscle rhythm genes and functional genes varied significantly in expression levels depending on weather, time of day, and light intensity and that the expression levels of myogenic genes were closely related to clock gene expression. The fish were also able to adapt to changes in light intensity in different weather conditions through positive physiological regulation.
