Objective: To systematically evaluate the incidence and risk factors of immune checkpoint inhibitor-associated pneumonia (CIP) in patients with lung cancer. Methods: The literature evaluating the incidence of CIP and its influencing factors in lung cancer patients were searched, in Chinese Biomedical Literature Database (CBM), CNKI Database, WanFang Database, VIP Chinese Biomedical Journal Database, PubMed, Cochrane Library, Web of Science, EBSCOhost, Medline and CINAHL Database from the construction of the library to June 30, 2022. Stata 17.0 software was applied to perform a Meta-analysis of the incidence and risk factors of CIP. Results: A total of 19 articles were included, involving 3 101 patients with lung cancer. The results of Meta-analysis showed that the total incidence of CIP in lung cancer patients was 18% (95%CI:0.14-0.21). Subgroup analysis showed that the incidence of CIP was higher in patients with lung cancer reported in 2021 and 2022, Asian countries and case-control studies. Risk factors included prior chronic obstructive pulmonary disease, prior interstitial pulmonary disease, performance status (PS) score≥2 points and combination therapy. Conclusion: The incidence of CIP is high in lung cancer patients and the occurrence of CIP is affected by many factors. Clinical medical staff should strengthen the identification and screening of risk factors, and intervene early to avoid serious consequences.

Summary: Artificial intelligence, which simulates the behavior of the human brain with a computer, including learning, reasoning, decision‐making and prediction, association, and other aspects, with the help of artificial intelligence technology to help visual communication designers complete high‐quality, efficient, and high‐volume design output, has become a trend. However, there are many defects in the existing visual design and creation system under the intelligent human–computer cooperation. Therefore, based on 5G virtual reality, this paper proposes the framework of visual design and creation system under intelligent human–computer cooperation research that has been carried out. This paper uses the method of literature analysis and other methods to study. In the research process, based on 5G virtual reality, this paper builds the framework of visual design and creation system under the intelligent human–computer cooperation of virtual reality and tests the visual design creation system based on 5G virtual reality from the aspects of response time, visual design completion time, and visual presentation effect; the system is compared with other intelligent human–computer cooperation visual design creation systems. The results show that the clarity score of this system is 96.5, the visual presentation quality score is 95.8, the clarity score of other systems is 87.6, and the visual presentation quality score is 86.3. We aimed to detect the effects of miR-145-5p on the cell proliferation, apoptosis, migration, and invasion in NRAS-mutant, BRAF-mutant, and wild-type melanoma cells, in order to figure out the potential mechanisms and provide a novel therapeutic target of melanoma. RT-qPCR and western blot were used to detect the expression of miR-145-5p and NRAS in melanoma tumor tissues and cells, respectively. Luciferase assay was performed to determine whether miR-145-5p directly targeted NRAS. After transfecting miR-145-5p mimics, miR-145-5p inhibitors, NRAS cDNA and NRAS siRNA into CHL-1, VMM917 and SK-mel-28 cells, functional assays were used to detect the proliferation, apoptosis, invasion and migration, including MTT, flow cytometry, Transwell and wound healing assays. In addition, xenograft models in nude mice were also conducted to verify the role of miR-145-5p in vivo. MiR-145-5p was able to suppress proliferation, invasion, and migration of VMM917 and CHL-1 cells and induce apoptosis by inhibiting MAPK and PI3K/AKT pathways. However, aberrant expression of miR-145-5p and NRAS has little impact on the viability and metastasis of BRAF-mutant melanoma. The higher expression of miR-145-5p in xenograft models repressed the VMM917-induced and CHL-1-induced tumor growth observably and has little effect on SK-mel-28-induced tumor growth which was consistent with the results in vitro. Through targeting NRAS, miR-145-5p could suppress cell proliferation, invasion, and migration and induce apoptosis of CHL-1 and VMM917 melanoma cells by inhibiting MAPK and PI3K/AKT pathways. Increasing evidences demonstrated that long noncoding RNAs (lncRNAs) are frequently dysregulated and have critical roles in many tumors. However, the roles and functional mechanisms of lncRNAs in melanoma remain largely unknown. In this study, we identified a novel lncRNA MHENCR which was upregulated in melanoma tissues and further upregulated in metastatic melanoma. Increased expression of MHENCR indicted poor survival of melanoma patients. Functional experiments revealed that MHENCR knockdown significantly inhibited melanoma cells proliferation, induced cell cycle arrest and apoptosis, and also attenuated melanoma cells migration in vitro . Furthermore, we identified MHENCR as a competitively endogenous RNA, which specifically bound to miR-425 and miR-489, upregulated their target genes IGF1 and SPIN1 expression, and further activated PI3K-Akt pathway. Statistically significant correlations were observed between MHENCR expression and IGF1 and SPIN1 in melanoma tissues. In vivo functional experiments further confirmed the pro-growth and pro-metastasis roles of MHENCR. Collectively, our findings revealed that MHENCR functions as an oncogene in melanoma via activating miR-425/489-mediated PI3K-Akt pathway, and may be a therapeutic target for melanoma.
