Start codon targeted polymorphism (SCoT) is a novel gene targeted marker technique. L25 ( 56 ) orthogonal experiment and single factor experiment design were applied to optimize SCoT - PCR system of soybean in five factors as Mg2+, dNTPs, primer, Taq DNA polymerase and template DNA. The results showed that the optimized system was as follows: a total volume of 20 J L including 2.0 mmol · L¹ Mg2+, 0.2 mmol · L¹ dNTPs, 0.250 μmol · L¹ primer, 1.5U Taq polymerase, 30ng DNA template. The most suitable annealing temperature of primers was 50. 4°C. The optimized SCoT - PCR system was tested on eighteen soybeans, and the result was stable and reliable. From the 82 primer combinations tested, 32 were selected with clear band patterns and abundant polymorphism. Amplifications and genetic verification were carried out on Jiyu 75, local black soybean and progenies from Jiyu 75 X local black soybean. The results showed polymorphism bands and common bands from parents and progeny. The system provided a new technology for evaluation of genetic diversity, construction of genetic linkage map and molecular marker assisted breeding for the soybean. [ABSTRACT FROM AUTHOR]

The additive - dominant - maternal model with genotype x environment interaction was analyzed by both unconditional variance and conditional variance separately. The seed oil contents in oil sunflower were estimated at different stages in different environments. The estimations of genetic effect for oil content of seed were determined at the developmental stage (0→t)or the special developmental period (t -1→t). The results showed that the oil content of seed was controlled by both dominant effect and maternal effect. The expression differences of seed nuclear genes and diploid maternal plant genes were observed on oil deposit during different developmental stages. The additive effect and the maternal effect were expressed inconsistently. The additive effect had not been detected at 0 - 10d and 30 - 40d after flowering, but it had been detected at peak during the days of 20 - 25 . There were highly significant dominant effects during 0 - 45d after flowering, especially during the period of 15 - 30d . It was the predominant stage of heterosis performance, while the maternal effect was active from 35 to 45 after flowering, especially during the 35 - 40d . A simulation model for maize spike differentiation and phenophase was established by analysis of temperatures and illumination treatments effects on maize growth and development process based on physiological development time constancy. Specific Bata function was applied in the model to calculate the daily thermal effect, and Gaussian function for photoperiodic effects. Temperature sensitivity, photoperiod sensitivity, intrinsic earliness and filling fractions which are genetic coefficients used to express genetic differences of maize development process. The model was tested at different growth stages and nitrogen factor was corrected by the actual nitrogen content, the critical nitrogen content and the lowest nitrogen content of plant. Results demonstrated that the absolute simulation error on growth stages (including emergence, jointing, silking, filling and maturity) and tassel and ear differentiation stages (including growing tip lengthening, spikelet differentiation, floret differentiation stage and sexual organ development) in different maize (Zea mays L.) cultivars was between 0-5 d, and the root mean square error was found less than 3.5 d. Introduction: To address issues related to shallow soil tillage, low soil nutrient content, and single tillage method in maize production in the Western Inner Mongolia Region, this study implemented various tillage and straw return techniques, including strip cultivation, subsoiling, deep tillage, no-tillage, straw incorporation with strip cultivation, straw incorporation with subsoiling, straw incorporation with deep tillage, and straw incorporation with no tillage, while using conventional shallow spinning by farmers as the control., Methods: We employed Xianyu 696 (XY696) and Ximeng 6 (XM6) as experimental materials to assess maize 100-grains weight, grain filling rate parameters, and grain nutrient quality. This investigation aimed to elucidate how tillage and straw return influence the accumulation of grain material in different maize varieties., Results and Discussion: The results indicated that proper implementation of tillage and straw return had a significant impact on the 100-grains weight of both varieties. In comparison to CK (farmer's rotary rotation), the most notable rise in 100-grains weight was observed under the DPR treatment (straw incorporation with deep tillage), with a maximum increase of 4.84% for XY696 and 6.28% for XM6. The proper implementation of tillage and straw return in the field resulted in discernible differences in the stages of improving the grain filling rates of different maize varieties. Specifically, XY696 showed a predominant increase in the filling rate during the early stage (V1), while XM6 exhibited an increase in the filling rates during the middle and late stages (V2 and V3). In comparison to CK, V1 increased by 1.54% to 27.56% in XY696, and V2 and V3 increased by 0.41% to 10.42% in XM6 under various tillage and straw return practices. The proper implementation of tillage and straw return had a significant impact on the nutritional quality of the grains in each variety. In comparison to CK, the DPR treatment resulted in the most pronounced decrease in the soluble sugar content of grains by 25.43% and the greatest increase in the crude fat content of grains by 9.67%., Conclusion: Ultimately, the proper implementation of soil tillage and straw return facilitated an increase in grain crude fat content and significantly boosted grain weight by improving the grouting rate parameters at all stages for various maize varieties. Additionally, the utilization of DPR treatment proved to be more effective. Aim: To investigate whether continuous veno-venous hemofiltration (CVVH) in different filtration rate to eliminate cytokines would result in different efficiency in acute pancreatitis, whether the saturation time of filter membrane was related to different filtration rate, and whether the onset time of CVVH could influence the survival of acute pancreatitis., Methods: Thirty-seven patients were classified into four groups randomly. Group 1 underwent low-volume CVVH within 48 h of the onset of abdominal pain (early CVVH, n = 9). Group 2 received low-volume CVVH after 96 h of the onset of abdominal pain (late CVVH, n = 10). Group 3 underwent high-volume CVVH within 48 h of the onset of abdominal pain (early CVVH, n = 9). Group 4 received high-volume CVVH after 96 h of the onset of abdominal pain (late CVVH, n = 9). CVVH was sustained for at least 72 h. Blood was taken before hemofiltration, and ultrafiltrate was collected at the start of CVVH and every 12 h during CVVH period for the purpose of measuring the concentrations of TNF-alpha, IL-1beta and IL-6. The concentrations of TNF-alpha, IL-1beta and IL-6 were measured by swine-specific ELISA. The Solartron 1 255 B frequency response analyzer (British) was used to observe the resistance of filter membrane., Results: The survival rate had a significant difference (94.44% vs 68.42%, P<0.01) high-volume and low-volume CVVH patients. The survival rate had also a significant difference (88.89% vs 73.68%, P<0.05) between early and late CVVH patients. The hemodynamic deterioration (MAP, HR, CVP) was less severe in groups 4 and 1 than that in group 2, and in group 3 than in group 4. The adsorptive saturation time of filters membranes was 120-180 min if the filtration rate was 1 000-4 000 mL/h. After the first, second and third new hemofilters were changed, serum TNF-alpha concentrations had a negative correlation with resistance (r: -0.91, -0.89, and -0.86, respectively in group 1; -0.89, -0.85, and -0.76, respectively in group 2; -0.88, -0.92, and -0.82, respectively in group 3; -0.84, -0.87, and -0.79, respectively in group 4). The decreasing extent of TNF-alpha, IL-1beta and IL-6 was significantly different between group 3 and group 1 (TNF-alpha P<0.05, IL-1beta P<0.05, IL-6 P<0.01), between group 4 and group 2 (TNF-alpha P<0.05, IL-1beta P<0.05, IL-6 P<0.01), between group 1 and group 2 (TNF-alpha P<0.05, IL-1beta P<0.05, IL-6 P<0.05), and between group 3 and group 4 (TNF-alpha P<0.01, IL-1beta P<0.01, IL-6 P<0.05), respectively during CVVH period. The decreasing extent of TNF-alpha and IL-1beta was also significantly different between survival patients and dead patients (TNF-alpha P<0.05, IL-1beta P<0.05). In survival patients, serum concentration of TNF-alpha and IL-1beta decreased more significantly than that in dead patients., Conclusion: High-volume and early CVVH improve hemodynamic deterioration and survival in acute pancreatitis patients. High-volume CVVH can eliminate cytokines more efficiently than low-volume CVVH. The survival rate is related to the decrease extent of TNF-alpha and IL-1beta. The adsorptive saturation time of filter membranes are different under different filtration rate condition. The filter should be changed timely once filter membrane adsorption is saturated.
