The study of groundwater evolution is of great significance for water resource protection and management, groundwater pollution control, and ecological environment protection. Experts and scholars have found that the hydrochemical processes and evolutionary patterns of groundwater are determined by both natural processes and human activities. However, there is relatively little research on the evolution of groundwater in mining areas where human activities have a significant impact. Therefore, to study the main controlling factors affecting the hydrogeochemical evolution of groundwater in mining areas, this paper proposes a method combining mixed ratio calculation and multivariate statistical analysis. Firstly, a total of 40 groundwater samples are classified into six clusters via hierarchical cluster analysis. By comprehensively analyzing the spatial location of the samples, it was found that there was no obvious distribution pattern of groundwater in space. Furthermore, the rationality of the cluster analysis is evaluated via principal component analysis. Next, hydrochemical and isotopic analyses were conducted to determine the source of groundwater in the mining area, and a three terminal element mixing model was established to identify the source of pollutants and calculate the terminal element mixing ratio. The research results indicate that groundwater in mining areas is formed by a mixture of shallow bedrock fissure water, deep bedrock fissure water, and rainwater, and the mixing effect is the main factor affecting the evolution of groundwater in mining areas, with a more significant impact than the depth of groundwater circulation. In addition, different types and degrees of water–rock interaction in different regions have altered the hydrochemical characteristics of groundwater in mining areas, such as the dissolution of multiple minerals, cation exchange, and common ion effects. Based on the above analysis results, a water circulation model for the mining area has been established. The findings of this study not only contribute to the protection of shallow fissure groundwater in the study area, but also provide a basis for investigating the groundwater evolution patterns in other metal mines.

Type 2 diabetes mellitus (T2DM) is a serious metabolic disease characterized by insulin resistance and reduced insulin production, which causes abnormally elevated blood glucose. It has been reported that T2DM can enhance oxidative stress and inflammatory responses, and stimulate a variety of complications including liver injury. Studies have shown that taurine has antioxidant and anti-inflammatory effects that can not only ameliorate diabetes but also alleviate liver injury caused by various diseases. However, its effect on liver injury in T2DM is not clear. In our study, a high-fat diet and intraperitoneal injection of streptozotocin (STZ) was used to induce liver injury in T2DM rats, and taurine was given as a treatment. Through the use of HE staining on paraffin sections, ELISA, and qRT-PCR, the effects of taurine on liver pathological alterations, antioxidant capacity, and inflammatory response were investigated. We found that: hepatic transaminase levels of rats were reduced significantly following taurine administration; histopathological observations revealed that the morphology of rat hepatocytes was close to normal, and the number of inflammatory cells around liver vessels was significantly reduced; antioxidant-related indicators were significantly increased, including SOD, CAT, GSH-Px and T-AOC, while related factors of the Nrf2 signalling pathway and its downstream HO-1, NQO1 and γ-GCS were significantly increased; the expression of the JAK2-STAT1 signalling pathway, TLR4/NF-κB signalling pathway and NLRP3 inflammatory vesicle-related factors were significantly reduced. Our results suggest taurine can alleviate T2DM-induced liver injury by improving the antioxidant capacity of the liver and inhibiting macrophage M1-type polarization and the inflammatory response mediated thereby.

Abstract Background Hypertrophic scars (HS) affect millions of people each year and require better treatment strategies. Bacterial extracellular vesicles (EVs) are advantaged by low cost and high yield which was commonly used in the treatment of diseases. Here, we investigated the therapeutic efficacy of EVs obtained from Lactobacillus druckerii in hypertrophic scar. In vitro, the effects of Lactobacillus druckerii-derived EVs (LDEVs) on Collagen I/III and α-SMA in fibroblasts obtained from HS. In vivo, a scleroderma mouse model was used to investigate the effects of LDEVs on fibrosis. The impact of LDEVs on excisional wound healing was explored. The different proteins between PBS and LDEVs treated fibroblasts derived from hypertrophic scar were studied by untargeted proteomic analysis. Results In vitro, LDEVs treatment significantly inhibited the expression of Collagen I/III and α-SMA and cell proliferation of fibroblasts derived from HS. In vivo, LDEVs withdrawn the hypertrophic scar formation in scleroderma mouse model and decreased the expression of α-SMA. LDEVs promoted the proliferation of skin cells, new blood vessel formation and wound healing in excisional wound healing mice model. Moreover, proteomics has shown that LDEVs inhibit hypertrophic scar fibrosis through multiple pathways. Conclusions Our results indicated that Lactobacillus druckerii-derived EVs has the potential application in the treatment of hypertrophic scars and any other fibrosis diseases. Graphical Abstract

To achieve automatic recognition of lightning images, which cannot easily be handled using the existing methods and still requires significant human resources, we propose a lightning image dataset and a preprocessing method. The lightning image data over five months were collected using a camera based on two optical observation stations, and then a series of batch labeling methods were applied, which greatly reduced the workload of subsequent manual labeling, and a dataset containing more than 30,000 labeled samples was established. Considering that lightning varies rapidly over time, we propose a time sequence composite (TSC) preprocessing method that inputs lightning’s time-varying characteristics into a model for better recognition of lightning images. The TSC method was evaluated through an experiment on four backbones, and it was found that this preprocessing method enhances the classification performance by 40%. The final trained model could successfully distinguish between the “lightning” and “non-lightning” samples, and a recall rate of 86.5% and a false detection rate of 0.2% were achieved.

The probability density analysis formula of the single orbital angular momentum (OAM) mode of an annular vortex beam with a focusing mirror in a turbulent atmosphere is derived theoretically, and the effects of different parameters on the OAM spectrum are investigated numerically, and the results show that the OAM diffusion of the annular vortex beam is weaker than the Gaussian vortex beam in a turbulent atmosphere under the same conditions, and the annular vortex beam with a focusing mirror can more effectively reduce the crosstalk of OAM modes. Our findings will be useful for improving FSO system performance.

Summary: The clinical correlation between adiponectin (APN) signal and hypertrophic scar (HS) remains unclear. Here, we found significantly reduced expression of APN receptors (AdipoR1/2) in HS tissues and derived fibroblasts (HFs), suggesting that HS formation may be associated with APN/AdipoR1/2 decline. RNA sequencing and RT-PCR validation revealed that APN significantly elevated the expression of SIRT1. Both in vitro and in vivo experiments confirmed that SIRT1 plays important role in APN inhibiting the fibrotic phenotype transformation and proliferation of scar fibroblasts and improving skin fibrosis. Mechanistically, SIRT1 inhibited the acetylation of C/EBPβ K39, histone H3K27, and H3K9, resulting in impaired transcription activity of C/EBPβ and compact chromatin conformation, thus preventing C/EBPβ from activating the transcription of YAP. Moreover, we found that YAP was critical for the transcriptional regulation of CTGF, CCND1, and CCNE1 by TEAD4. In conclusion, our study revealed the role of APN in antagonizing HS fibrosis by regulating the SIRT1/C/EBPβ/YAP pathway.

In this article, we analyze the impact of the off-axis noncanonical vortex on the dynamics of 3D energy flux. The results reveal that the properties of energy flux are significantly influenced by the anisotropy parameter σc of the off-axis noncanonical vortex. It is demonstrated that by adjusting the anisotropy parameter, we can control the transverse energy flux, from the position of the transverse energy tornado and the distribution of the transverse energy strength to the rotation direction of the transverse energy flux as the beam propagates. The interesting phenomenon, the reversed energy flux, is also closely related to the anisotropy parameter. The position and size of the reversed energy flux region, as well as the production of two energy tornadoes with opposite rotating directions, resembling a Chinese “Taiji” pattern, can be controlled by varying the anisotropy parameter. This result will potentially provide a new freedom for tailoring the 3D optical field and be useful in optical manipulation and processing.

ABSTRACT: Meat quality characteristics, including juiciness, flavor, and tenderness, can be mostly attributed to the total muscle fat content, intramuscular fat (IMF), and the composition of its fatty acids, which are regulated by the balance between lipid uptake, transport, synthesis, and subsequent metabolism, involving many genes and pathways. However, the detailed molecular mechanisms remain unclear. The purpose of this study was to identify the key signaling pathways related to chicken meat quality, and to provide help for improving chicken meat quality. The present study reports the RNA-sequencing analysis of pectorales and crureus of the Zhuanghe dagu chicken and the Arbor Acres Broiler chicken (AA chicken). We identified certain differentially expressed genes that affect IMF deposition, such as EHHADH, TECRL, NDUFAB1, PCCB, and HIBCH, which were upregulated in Zhuanghe dagu chicken , and GCDH, TPI1, ABHD13, PSMC1, MYST2, and FBXO11, which were upregulated in AA chickens. Pathway analysis using the Kyoto Encyclopedia of Genes and Genomes indicated that the extracellular matrix (ECM)–receptor interaction pathway is co-enriched in both tissues, and forms a sub-pathway of other enriched pathways. Intriguingly, the ECM–receptor interaction pathway genes are regulated differently in different gene pools. Collagens, which are main ECM constituents, and laminin and integrin β1 transmembrane receptors were significantly downregulated in both tissues of the AA chicken. The results showed that the ECM-receptor interaction pathway affect the quality of chicken meat by affecting the metabolism of intramuscular adipocytes. Further investigation of this signaling pathway will be helpful to the improvement of chicken meat quality.

Ca and P metabolic disorders during the egg-laying period can reduce egg production, impair eggshell quality, and even cause bone problems in hens; however, little is known regarding the capacity of duodenal and nephritic Ca and P absorption. Here, the levels of serum Ca and P metabolic indices and the expression of duodenal and renal Ca and P transporter genes were measured in hens at different egg-laying stages. The Ca, 25-(OH)-VD3, and 1,25-(OH)2-VD3 content increased during the peak (43 weeks of age) and late (72 weeks of age) egg-laying periods compared to that during the early (23 weeks of age) egg-laying period; however, there were no differences in Pi levels among the three egg-laying periods. Moreover, duodenal VDR and CaBP-D28k mRNA expression was markedly higher but NPt2b mRNA expression was markedly lower during the peak and late egg-laying periods than during the early egg-laying period. Furthermore, nephritic CaBP-D28k, PMCA1b, and FGFR1 mRNA expression was markedly higher but NPt2a and Cyp24a1 mRNA expression was markedly lower during the peak and late egg-laying periods than during the early egg-laying period. In conclusion, the present study indicated that the increased duodenal and nephritic Ca absorption during the peak and late egg-laying periods may be associated with the VD–VDR pathway, while the decreased P absorption despite relatively stable serum P levels in all three egg-laying stages may associated with osteolysis.

Sepsis is a life-threatening organ dysfunction condition caused by a dysregulated host response to infection and lack of effective treatment method. Supplementation of probiotics has emerged as a potential biotherapy for inflammatory diseases in recent years, but its role in protecting viscera against the damage caused by sepsis and the underlying mechanism is poorly understood. Streptococcus thermophilus 19 is one of the most well-studied probiotics, which is selected in this study among seven strains isolated from homemade yogurt due to its optimal ability of suppressing the inflammation response in vitro. It showed significant decrease in the expression of TNF-α, IL-1β, and IL-6 in the co-culture of S. thermophilus 19 and LPS-treated mouse macrophage. The effect of S. thermophilus 19 in mice and the response of mice gut microbiota were subsequently investigated. In LPS-induced septic mouse model, S. thermophilus 19 was highly resistant to LPS and exhibited significantly decreased expressions of inflammatory factors compared to LPS-treated mice. A MiSeq-based 16S rDNA sequence analysis revealed that the decrease of gut microbial diversity in mice intraperitoneally injected with 1 mg/ml LPS were mitigated by the administration of S. thermophilus 19. Fusobacterium significantly decreased during the development of sepsis and rose again after supplement strain 19, while Flavonifractor showed the opposite trend, which demonstrated these two genera were the key bacteria that may function in the mice gut microbiota for alleviation of LPS-induced inflammation reaction. To conclude, S. thermophilus 19 may be a potential candidate for novel biotherapeutic interventions against inflammation caused by sepsis.

Hypertrophic scar (HS) is a serious fibrotic skin disease and often considered as a kind of benign skin tumor. Tetramethylpyrazine (TMP), the main chemical composition of the traditional Chinese medicine Chuanxiong Rhizoma, has shown significant clinical benefits in the treatment of fibrosis disease and tumor, while the role in HS and the concrete mechanisms remain elusive. Herein, the protective effects of TMP in the treatment of HS was investigated and the results showed that the protein expression levels of type I collagen (Col I), type III collagen (Col III), and α-smooth muscle actin (α-SMA) were all inhibited remarkably after addition of TMP in HS-derived fibroblasts (HFs). Moreover, TMP also suppressed fibroblast proliferative and induced cell apoptosis. The protein expression levels of Caspase-3 and Bcl-2 were all decreased comparing with the control group while proapoptotic proteins Bax and Cleaved Caspase-3 were increased. In addition, TMP treatment markedly reduced the phosphorylation levels of AKT. Taken together, our investigations demonstrated that TMP could down-regulate the expression of fibrosis-related molecules, inhibit scar fibroblast proliferation and activate cell apoptosis, during which AKT pathway was involved. Thus, this study shed more light on the pharmacological mechanisms of TMP, and provided a novel therapeutic alternative for prevention and treatment of HS.

Abstract Remarkably little information is available about the interaction between the gut microbiota and intestinal immunity in fish and crustaceans. In our study, we used Illumina MiSeq sequencing and real-time quantitative PCR to compare the microbial community and immunity genes expression in the foregut, midgut and hindgut of Chinese mitten crab (Eriocheir sinensis). Our results indicated that the community richness of the midgut is higher than in the foregut or the hindgut, although the bacterial diversity in the hindgut is higher. The predominant phyla were Tenericutes and Firmicutes in the foregut, Tenericutes and Proteobacteria in the midgut and Proteobacteria, Tenericutes and Bacteroidetes in the hindgut. Convolution expression for partially coherent beams that are partially blocked by an opaque obstacle is derived in detail in a paraxial $ABCD$ optical system. The examples are showed to how the formula can be applied to study the self-healing for different beams and different obstructions. In particular, this approach can be used to conveniently deal with any partially coherent Schell-model beams partially blocked by any shape obstacle. Furthermore, the self-healing ability characterization based on the similarity degree of intensity to intensity is proposed. Our method not only can be used to estimate the self-healing ability of beams in theory, but also can be applied to characterize the self-healing ability of beams in experiment. It offers a good and convenient method for investigating or comparing the self-healing ability of different beams, including fully coherent beams and partially coherent beams.

In recent years, unmanned aerial vehicles (UAVs) have been considered an ideal relay platform for enhancing the communication between ground agents, because they fly at high altitudes and are easy to deploy with strong adaptabilities. Their maneuvering allows them to adjust their location to optimize the performance of links, which brings out the relay UAV autonomous mobility control problem. This work addressed the problem in a novel scene with mobile agents and completely unknown wireless channel properties, using only online measured information of received signal strength (RSS) and agent positions. The problem is challenging because of the unknown and dynamic radio frequency (RF) environment cause by agents and UAV maneuvering. We present a framework for both end-to-end communication and multi-agent-inter communication applications, and focus on proposing: (1) least square estimation-based channel approximation with consideration of environment effects and, (2) gradient-based optimal relay position seeking. Simulation results show that considering the environmental effects on channel parameters is meaningful and beneficial in using UAV as relays for the communication of multiple ground agents, and validate that the proposed algorithms optimizes the network performance by controlling the heading of the UAV.

In this paper, a novel deep reinforcement learning (DRL) method, and robust deep deterministic policy gradient (Robust-DDPG), is proposed for developing a controller that allows robust flying of an unmanned aerial vehicle (UAV) in dynamic uncertain environments. This technique is applicable in many fields, such as penetration and remote surveillance. The learning-based controller is constructed with an actor-critic framework, and can perform a dual-channel continuous control (roll and speed) of the UAV. To overcome the fragility and volatility of original DDPG, three critical learning tricks are introduced in Robust-DDPG: (1) Delayed-learning trick, providing stable learnings, while facing dynamic environments; (2) adversarial attack trick, improving policy’s adaptability to uncertain environments; (3) mixed exploration trick, enabling faster convergence of the model. The training experiments show great improvement in its convergence speed, convergence effect, and stability. The exploiting experiments demonstrate high efficiency in providing the UAV a shorter and smoother path. While, the generalization experiments verify its better adaptability to complicated, dynamic and uncertain environments, comparing to Deep Q Network (DQN) and DDPG algorithms.

Previous work in our laboratory demonstrated that exosomes derived from human amniotic epithelial cells (hAECs) accelerated wound healing by promoting the proliferation and migration of fibroblasts. It is reported that exosomes, which are carriers of the microRNAs (miRNAs) and proteins, play an important role in the regulation of cell-to-cell communication. However, it is still unclear precisely which molecule or which group of molecules carried within hAEC-derived exosomes (hAEC-Exos) mediated wound healing. Here, we explored purified hAEC-Exos together with either proteinase K (PROse) or RNase A on the effect of fibroblasts and cutaneous wound healing. Our experiments demonstrated that hAEC-Exos were positive for exosomal markers CD9, CD63, and CD81. Also, we found that hAEC-Exos could be internalized by fibroblasts and then stimulated cell migration and proliferation. However, the promotive effect of hAEC-Exos was abolished by pretreating hAEC-Exos with RNase A, not PROse. Importantly, in vivo wound healing assay showed that local injection of hAEC-Exos or PROse pretreated hAEC-Exos at skin wounds significantly accelerated wound healing. Our findings revealed an important role of exosomal miRNAs in wound healing.

This paper puts forward some thoughts on the fine management of power transmission and transformation engineering construction with the help of three-dimensional technology, and discusses the installation, quality acceptance and construction organization deepening application of steel components in power transmission and transformation engineering construction.The physical 3D model is established by scanning technology, and the accuracy of machining components is checked by comparing the design model with the physical model. The construction process is simulated by the three-dimensional engineering management platform, and the problems existing in the actual construction are found, and the optimization of the construction organization design is carried out through the platform.

Numerical simulation of random optical field with special autocorrelation is performed. It can be seen that the speckle pattern with special autocorrelation is different from the general speckle pattern whose autocorrelation is a Gaussian function. Furthermore, we study the propagation property of the special speckle in free space after blocking by an opaque obstruction and find the self-healing effect of speckle. In particular, we investigate the influence of size and shape of obstruction and the coherence of speckle on the speckle’s reconstruction process. We find that it is affected by the size of obstruction and the coherence of the speckle but is almost not affected by the shape of obstruction. Our results can be applied to the imaging, optical communication, and so on.
