2. Anti-CD44-Conjugated Olive Oil Liquid Nanocapsules for Targeting Pancreatic Cancer Stem Cells

Abstract

The latest trends in cancer research and nanomedicine focus on using nanocarriers to target cancer stem cells (CSCs). Specifically, lipid liquid nanocapsules are usually developed as nanocarriers for lipophilic drug delivery. Here, we developed olive oil liquid NCs (O2LNCs) functionalized by covalent coupling of an anti-CD44-fluorescein isothiocyanate antibody ({\alpha}CD44). First, O2LNCs are formed by a core of olive oil surrounded by a shell containing phospholipids, a nonionic surfactant, and deoxycholic acid molecules. Then, O2LNCs were coated with an {\alpha}CD44 antibody ({\alpha}CD44-O2LNC). The optimization of an {\alpha}CD44 coating procedure, a complete physicochemical characterization, as well as clear evidence of their efficacy in vitro and in vivo were demonstrated. Our results indicate the high targeted uptake of these {\alpha}CD44-O2LNCs, and the increased antitumor efficacy (up to four times) of paclitaxel-loaded-{\alpha}CD44-O2LNC compared to free paclitaxel in pancreatic CSCs (PCSCs). Abstract

Despite all the advances achieved in the field of tumor-biology research, in most cases conventional therapies including chemotherapy are still the leading choices. The main disadvantage of these treatments, in addition to the low solubility of many antitumor drugs, is their lack of specificity, which explains the frequent occurrence of serious side effects due to nonspecific drug uptake by healthy cells. Progress in nanotechnology and its application in medicine have provided new opportunities and different smart systems. Such systems can improve the intracellular delivery of the drugs due to their multifunctionality and targeting potential. The purpose of this manuscript is to review and analyze the recent progress made in nanotherapy applied to cancer treatment. First, we provide a global overview of cancer and different smart nanoparticles currently used in oncology. Then, we analyze in detail the development of drug-delivery strategies in cancer therapy, focusing mainly on the intravenously administered smart nanoparticles with protein corona to avoid immune-system clearance. Abstract

Nanoparticles (NPs) based on the polymer poly (lactide-co-glycolide) acid (PLGA) have been widely studied in developing delivery systems for drugs and therapeutic biomolecules, due to the biocompatible and biodegradable properties of the PLGA. In this work, a synthesis method for bone morphogenetic protein (BMP-2)-loaded PLGA NPs was developed and optimized, in order to carry out and control the release of BMP-2, based on the double-emulsion (water/oil/water,W/O/W) solvent evaporation technique. The polymeric surfactant Pluronic F68 was used in the synthesis procedure, as it is known to have an effect on the reduction of the size of the NPs, the enhancement of their stability, and the protection of the encapsulated biomolecule. Spherical solid polymeric NPs were synthesized, showing a reproducible multimodal size distribution, with diameters between 100 and 500 nm. This size range appears to allow the protein to act on the cell surface and at the cytoplasm level. The effect of carrying BMP-2 co-adsorbed with bovine serum albumin on the NP surface was analyzed. The colloidal properties of these systems (morphology by SEM, hydrodynamic size, electrophoretic mobility, temporal stability, protein encapsulation, and short-term release profile) were studied. Abstract

The main aim of this study is to report basic knowledge on how a protein corona (PC) could affect or modify the way in which multifunctionalized nanoparticles interact with cells. With this purpose, we have firstly optimized the development of a target-specific nanocarrier by coupling a specific fluorescent antibody on the surface of functionalized lipid liquid nanocapsules (LLNCs). Thus, an anti-HER2-FITC antibody ({\alpha}HER2) has been used, HER2 being a surface receptor that is overexpressed in several tumor cells. Subsequently, the in vitro formation of a PC has been developed using fetal bovine serum supplemented with human fibrinogen. Dynamic Light Scattering (DLS), Nanoparticle Tracking Analysis (NTA), Laser Doppler Electrophoresis (LDE), and Gel Chromatography techniques have been used to assure a complete physico-chemical characterization of the nano-complexes with (LLNCs-{\alpha}HER2-PC) and without (LLNCs-{\alpha}HER2) the surrounding PC. In addition, cellular assays were performed to study the cellular uptake and the specific cellular-nanocarrier interactions using the SKBR3 (high expression of HER2) breast cancer cell line and human dermal fibroblasts (HDFa) (healthy cell line without expression of HER2 receptors as control), showing that the SKBR3 cell line had a higher transport rate (50-fold) than HDFa at 60 min with LLNCs-{\alpha}HER2. Moreover, the SKBR3 cell line incubated with LLNCs-{\alpha}HER2-PC suffered a significant reduction (40%) in the uptake. These results suggest that the formation of a PC onto LLNCs does not prevent specific cell targeting, although it does have an important influence on cell uptake., Comment: 19 pages, 7 figures

10. Code Detection for Hardware Acceleration Using Large Language Models

Abstract

Large language models (LLMs) have been massively applied to many tasks, often surpassing state-of-the-art approaches. While their effectiveness in code generation has been extensively studied (e.g., AlphaCode), their potential for code detection remains unexplored. This work presents the first analysis of code detection using LLMs. Our study examines essential kernels, including matrix multiplication, convolution, and fast-fourier transform, implemented in C/C++. We propose both a preliminary, naive prompt and a novel prompting strategy for code detection. Results reveal that conventional prompting achieves great precision but poor accuracy (68.8%, 22.3%, and 79.2% for GEMM, convolution, and FFT, respectively) due to a high number of false positives. Our novel prompting strategy substantially reduces false positives, resulting in excellent overall accuracy (91.1%, 97.9%, and 99.7%, respectively). These results pose a considerable challenge to existing state-of-the-art code detection methods.

13. Matching Linear Algebra and Tensor Code to Specialized Hardware Accelerators

Abstract

Dedicated tensor accelerators demonstrate the importance of linear algebra in modern applications. Such accelerators have the potential for impressive performance gains, but require programmers to rewrite code using vendor APIs - a barrier to wider scale adoption. Recent work overcomes this by matching and replacing patterns within code, but such approaches are fragile and fail to cope with the diversity of real-world codes. We develop ATC, a compiler that uses program synthesis to map regions of code to specific APIs. The mapping space that ATC explores is combinatorially large, requiring the development of program classification, dynamic analysis, variable constraint generation and lexical distance matching techniques to make it tractable. We apply ATC to real-world tensor and linear algebra codes and evaluate them against four state-of-the-art approaches. We accelerate between 2.6x and 7x more programs, leading to over an order of magnitude performance improvement., Comment: This is the author's version of the work. 14. Teacher Training for Educational Change: The View of International Experts

Abstract

Digital competence has become a new type of literacy that significantly impacts on people's lives, as it is paramount to their social and occupational integration. In educational institutions, teachers play a major role in developing the digital skills of their students. This study aims to determine the key elements of teacher training in the use of innovative technologies and methodologies with a view to providing alternatives to improve the educational process. The objective is to overcome deficiencies in the training of teachers in new technologies and innovative methodologies. A qualitative study based on interview data is used in two phases. For the first phase research, semi-structured interviews were conducted with eight international education experts from various fields on how best to train teachers. The second phase research involved a text mining analysis of the interview transcripts based on sentiment analysis and word counts using the advanced R statistical programming language. The second phase research was conducted using the R statistical programming language. The interview transcripts were analysed using sentiment analysis and a word cloud. The results reveal that teacher education should be practice-focused and supervised by senior teachers with the aid of students and heads of schools. Moreover, standards should be set to monitor the digital competence of teachers, who should be trained on an ongoing basis to keep them up to date in the use of digital technologies.

16. POAS: A high-performance scheduling framework for exploiting Accelerator Level Parallelism

Abstract

Heterogeneous computing is becoming mainstream in all scopes. This new era in computer architecture brings a new paradigm called Accelerator Level Parallelism (ALP). In ALP, accelerators are used concurrently to provide unprecedented levels of performance and energy efficiency. To reach that, there are many problems to be solved, one of the most challenging being co-execution. This paper develops a scheduling framework called POAS, a general method for providing co-execution to generic applications. Unlike other scheduling approaches, POAS does not directly schedule applications. Instead, it is a generic model that transforms any application to make it suitable for co-execution, so that it can be executed in ALP environments. Our proposal is composed of four differentiated steps: predict, optimize, adapt and schedule. During these phases, different modifications are implemented in the application to make it suitable to be executed in ALP environments. In this work we also apply our framework to a matrix multiplication case study, outlining the critical and most important steps to port the application with POAS. We evaluate our POAS-based implementation for matrix multiplication on a CPU/GPU/XPU environment using CPU cores, CUDA cores and tensor cores (XPU). Our experiments prove that co-execution in the studied scenario can benefit from ALP, yielding speedups of up to 45% with respect to using only one accelerator. The proven flexibility and potential of POAS make it an excellent candidate to reach ALP in future computer systems.

17. Analysis of Digital Competence of Educators (DigCompEdu) in Teacher Trainees: The Context of Melilla, Spain

Abstract

The Spanish autonomous city of Melilla, located in northwest Africa, has one of the highest academic failure and abandonment rates in Europe. An effective way to improve this situation would be to improve students' digital competence. In order to do so, teachers must have competent digital skills themselves and also be able to teach them. To determine teachers' level of digital competence, the Spanish adaptation of the European Framework for Digital Competence of Educators was used to analyse the self-assessment responses of teachers in training at the Faculty of Education and Sport Sciences in Melilla, Spain. Several quantitative techniques were used to analyse data collected from a questionnaire based on the items in the framework. Indicators were given to each competence using a factor analysis to contrast differences between undergraduate and postgraduate students. Correlations between some of the students' characteristics and the competences were estimated using OLS. The results show students' self-assessment level of digital competence in different areas and differences between the bachelor's and master's programmes. Digital competence gaps were also detected in teacher training, especially in security. The conclusions highlight the need to improve digital security and facilitate a higher level of digital skills in line with the framework. Indeed, more hours of training in digital competence are required while taking into account the educational context and the technological, pedagogical and content knowledge needed to teach. Equally, the same skills must be developed by educators in order for them to transmit digital competence to their students and support them in educational centres.

Abstract

Objective: To conduct a proof-of-concept study of the detection of two synthetic models of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) using polarimetric imaging. Methods: Two SARS-CoV-2 models were prepared as engineered lentiviruses pseudotyped with the G protein of the vesicular stomatitis virus, and with the characteristic Spike protein of SARS-CoV-2. Samples were preparations in two biofluids (saline solution and artificial saliva), in four concentrations, and deposited as 5-{\mu}L droplets on a supporting plate. The angles of maximal degree of linear polarization (DLP) of light diffusely scattered from dry residues were determined using Mueller polarimetry of 87 samples at 405 nm and 514 nm. A polarimetric camera was used for simultaneous imaging of several samples under 380-420 nm illumination at angles similar to those of maximal DLP. A per-pixel image analysis included quantification and combination of polarization feature descriptors in other 475 samples. Results: The angles (from sample surface) of maximal DLP were 3 degrees for 405 nm and 6 degrees for 514 nm. Similar viral particles that differ only in the characteristic spike protein of the SARS-CoV-2, their corresponding negative controls, fluids, and the sample holder were discerned from polarimetric image analysis at 10-degree and 15-degree configurations. Conclusion: Polarimetric imaging in the visible spectrum has the potential for non-contact, reagent-free detection of viruses in multiple dry fluid residues simultaneously. Further analysis including real SARS-CoV-2 in human samples -- particularly, fresh saliva -- are required. Significance: Polarimetric imaging under visible light could contribute to fast, cost-effective screening of SARS-CoV-2 and other pathogens.

Methods: In this research, eight international experts in education are interviewed on what they consider to be the best way of implementing new technologies in disadvantaged contexts to achieve educational improvement. A mixed methodology has been used, qualitative through semi-structured interviews and quantitative through a text mining analysis of the interviews' transcripts with the R programming language. Results and Conclusions: The results show that the teaching staff is the key element. The curriculum, objectives and school director's help are also highlighted, as well as the collaboration of the students who have to be provided with the necessary digital resources. At the state level, greater funding and support from universities are needed to support research and the quality of teacher training.

Abstract

Real-Time Bidding is a new Internet advertising system that has become very popular in recent years. This system works like a global auction where advertisers bid to display their impressions in the publishers' ad slots. The most popular system to select which advertiser wins each auction is the Generalized second-price auction in which the advertiser that offers the most wins the bet and is charged with the price of the second largest bet. In this paper, we propose an alternative betting system with a new approach that not only considers the economic aspect but also other relevant factors for the functioning of the advertising system. The factors that we consider are, among others, the benefit that can be given to each advertiser, the probability of conversion from the advertisement, the probability that the visit is fraudulent, how balanced are the networks participating in RTB and if the advertisers are not paying over the market price. In addition, we propose a methodology based on genetic algorithms to optimize the selection of each advertiser. We also conducted some experiments to compare the performance of the proposed model with the famous Generalized Second-Price method. We think that this new approach, which considers more relevant aspects besides the price, offers greater benefits for RTB networks in the medium and long-term.

Abstract

Performance has always been a hot topic in computing. However, the viable ways to achieve it have taken many forms in the different moments of computing history. Today, technological limits have pushed the adoption of increasingly parallel multi-core and many-core architectures and even the use of highly specific hardware (aka Domain-Specific Architectures, or DSAs) to solve very specific problems. In this new context, one major problem is how to develop software once, and be able to run it on multiple accelerator architectures, seamlessly. Ideally aiming at a single programming model that can automatically target the code to different kinds of parallel architectures, allowing specific tuning with minimal, if any, changes to the source-code in order to seek performance portability. A comprehensive solution to this is still lacking. In this work, we present the use of the PHAST Library, which allows users to code once, at a high level of abstraction and thus with high productivity, and automatically targeting different parallel devices by changing the compilation process. As a case study, we have worked on the porting of the well-known deep-learning Caffe framework. The framework has been split into different parts and some of them have been ported, obtaining a working straightforward implementation that can be run on both CPUs and GPUs. Abstract

In this article, we report on the efficiency and effectiveness of multiagent reinforcement learning methods (MARL) for the computation of flight delays to resolve congestion problems in the Air Traffic Management (ATM) domain. Specifically, we aim to resolve cases where demand of airspace use exceeds capacity (demand-capacity problems), via imposing ground delays to flights at the pre-tactical stage of operations (i.e. few days to few hours before operation). Casting this into the multiagent domain, agents, representing flights, need to decide on own delays w.r.t. own preferences, having no information about others' payoffs, preferences and constraints, while they plan to execute their trajectories jointly with others, adhering to operational constraints. Specifically, we formalize the problem as a multiagent Markov Decision Process (MA-MDP) and we show that it can be considered as a Markov game in which interacting agents need to reach an equilibrium: What makes the problem more interesting is the dynamic setting in which agents operate, which is also due to the unforeseen, emergent effects of their decisions in the whole system. We propose collaborative multiagent reinforcement learning methods to resolve demand-capacity imbalances: Extensive experimental study on real-world cases, shows the potential of the proposed approaches in resolving problems, while advanced visualizations provide detailed views towards understanding the quality of solutions provided.
