Abstract

The homeobox transcription factors HoxA9 and Meis1 are causally involved in the etiology of acute myeloid leukemia. While HoxA9 alone immortalizes cells, cooperation with Meis1 is necessary to induce a full leukemic phenotype. Here, we applied degron techniques to elucidate the leukemogenic contribution of Meis1. Chromatin immunoprecipitation experiments revealed that Meis1 localized mainly to H3K27 acetylated and H3K4 mono-methylated enhancers preactivated by HoxA9. Chromatin association of Meis1 required physical presence of HoxA9 and all Meis1 DNA interactions were rapidly lost after HoxA9 degradation. Meis1 controlled a gene expression pattern dominated by Myc, ribosome biogenesis and ribosomal RNA synthesis genes. While Myc accounted for the cell cycle stimulating effect of Meis1, overexpression of this oncogene alone did not accelerate leukemogenesis. Besides its effect on Myc, Meis1 induced transcription of ribosomal biogenesis genes. This was accompanied by an elevated resistance against inhibition of ribosomal RNA synthesis and translation, but without affecting steady-state protein synthesis. Finally, we demonstrate that HoxA9 and Meis1 proteins are stabilized by post-translational modification. Mutation of HoxA9/Meis1 phosphorylation sites or inhibition of casein kinase 2 lead to rapid protein degradation suggesting a potential pathway for pharmacological intervention.

24. A Monte Carlo density functional theory for the competition between inter and intramolecular association in inhomogeneous fluids

Abstract

A monte carlo density functional theory is developed for chain molecules which both intra and intermolecularly associate. The approach can be applied over a range of chain lengths. The theory is validated for the case of an associating 4-mer fluid in a planar hard slit pore. Once validated the new theory is used to study the effect of chain length and temperature on the competition between intra and intermolecular association near a hard wall. We show that this competition enhances intramolecular association near wall contact and inverts the chain length dependence of the fraction bonded intermolecularly in the inhomogeneous region.

Abstract

Using the framework of Wertheim's thermodynamic perturbation theory we develop the first density functional theory which accounts for intramolecular association in chain molecules. To test the theory new Monte Carlo simulations are performed at a fluid solid interface for a 4 segment chain which can both intra and intermolecularly associate. The theory and simulation results are found to be in excellent agreement. It is shown that the inclusion of intramolecular association can have profound effects on interfacial properties such as interfacial tension and the partition coefficient.

42. A C/ebpα isoform-specific differentiation program in primary myelocytes

Abstract

The transcription factor CCAAT-enhancer binding factor alpha (C/ebpα) is a master controller of myeloid differentiation that is expressed as long (p42) and short (p30) isoform. Mutations within theCEBPAgene selectively deleting p42 are frequent in human acute myeloid leukemia. Here we investigated the individual genomics and transcriptomics of p42 and p30. Both proteins bound to identical sites across the genome. For most targets, they induced a highly similar transcriptional response with the exception of a few isoform-specific genes. Amongst those we identified early growth response 1 (Egr1) and tribbles 1 (Trib1) as key targets selectively induced by p42 that are also underrepresented inCEBPA-mutated AML. Egr1 executed a program of myeloid differentiation and growth arrest. Oppositely, Trib1 established a negative feedback loop through activation of Erk1/2 kinase thus placing differentiation under control of signaling. Unexpectedly, differentiation elicited either by removal of an oncogenic input or by G-CSF did not peruse C/ebpα as mediator but rather directly affected the cell cycle core by upregulation of p21/p27 inhibitors. This points to functions downstream of C/ebpα as intersection point where transforming and differentiation stimuli converge and this finding offers a new perspective for therapeutic intervention.

ilustraciones, diagramas, mapas En el presente estudio se implementaron y validaron metodologías para la determinación simultanea de contaminantes organoclorados (COs) y bifenilos policlorados (PCBs) en leche bovina y leche materna por el método QuEChERS con algunas modificaciones, por cromatografía de gases con detector μECD y por espectrometría de masas con cuadrupolo simple y triple cuadrupolo. La idoneidad de los métodos propuestos se ratificó con dos materiales de referencia certificados (BCR 188 y BCR 450). La aplicabilidad se llevó a cabo en 50 muestras de leche bovina y 56 muestras de leche materna, procedentes de diferentes departamentos de Colombia. A partir de los resultados obtenidos se realizó un análisis de riesgo con un enfoque determinístico y probabilístico para identificar el grado de exposición a los contaminantes orgánicos persistentes (COPs). Los métodos validados demostraron ser altamente selectivos/específicos, lineales, exactos, y sensibles, permitiendo obtener límites de cuantificación menores a los límites máximos de residuos (LMR) establecidos por la Comisión Europea y el Codex Alimentario. Los COPs encontrados en leche bovina y materna no superaron los LMR, sin embargo, en la mayoría de las muestras se evidenció la presencia de DDE, DDT y algunos PCBs. El análisis de riesgo determinístico efectuado en ambas matrices permitió identificar porcentajes de la ingesta diaria admisible (%IDA) inferiores al 50%, indicando que no hay riesgos potenciales para la salud de la población. Sin embargo, al realizar el análisis con un enfoque probabilístico se obtuvo que el DDE representaba una probabilidad de 1% en leche bovina y 5.3% en leche materna de superar el umbral toxicológico seguro para el grupo poblacional más expuesto que es la población infantil. Los hallazgos de este estudio proporcionan información valiosa para la determinación de COPs como una herramienta para el análisis de muestras indicadoras de contaminación ambiental. (Texto tomado de la fuente) In the present study, methodologies were implemented and validated for the simultaneous determination of organochlorine contaminants (COs) and polychlorinated biphenyls (PCBs) in bovine milk and breast milk by the QuEChERS method with some modifications, by gas chromatography with μECD detector and by spectrometry of masses with simple quadrupole and triple quadrupole. The suitability of the proposed methods was confirmed with two certified reference materials (BCR 188 and BCR 450). The applicability was carried out in 50 samples of bovine milk and 56 samples of breast milk, coming from different departments of Colombia. Based on the results obtained, a risk analysis was carried out with a deterministic and probabilistic approach to identify the degree of exposure to persistent organic pollutants (POPs). The validated methods proved to be highly selective/specific, linear, exact (absolute error less than 20% with the reference materials), and sensitive, allowing to obtain limits of quantification lower than the maximum residue limits (MRL) established by the European Commission. and the Alimentary Codex. The POPs found in bovine and breast milk did not exceed the MRLs, however, the presence of DDE, DDT and some PCBs was evidenced in most of the samples. The deterministic risk analysis carried out in both matrices made it possible to identify percentages of the acceptable daily intake (% ADI) (a parameter that was considered as an exposure indicator), lower than 50%, indicating that there are no potential risks for the health of the population. However, when carrying out the analysis with a probabilistic approach, it was obtained that the DDE represented a probability of 1% in bovine milk and 5.3% in breast milk of exceeding the safe toxicological threshold for the most exposed population group, which is the infant population. The findings of this study provide valuable information for the determination of POPs as a tool for the analysis of indicator samples of environmental contamination. Maestría Química analítica
