The rising levels of type 2 diabetes mellitus (T2DM) and the poor medical effects of the commercially available antidiabetic drugs necessitate the development of potent analogs to treat this multifactorial metabolic disorder. It has been demonstrated that targeting two or more biochemical targets associated with the onset and progression of diabetes along with oxidative stress and/or cancer could be a significant strategy for treating complications related to this metabolic disorder. The 3,5-dibromo-4,6-dimethoxychalcones (2a–f) and the corresponding flavone derivatives (3a–f) were synthesized and characterized using spectroscopic (NMR, HR-MS and FT-IR) techniques. The inhibitory effect of both series of compounds against α-glucosidase and α-amylase was evaluated in vitro through enzymatic assays. Selected compounds were also evaluated for potential to activate or inhibit superoxide dismutase. Compound 3c was selected as a representative model for the flavone series and evaluated spectrophotometrically for potential to coordinate Cu(II) and/or Zn(II) ions implicated in the metal-catalyzed free radical generation. A plausible mechanism for metal-chelation of the test compounds is presented. Furthermore, the most active compounds from each series against the test carbohydrate-hydrolyzing enzymes were selected and evaluated for their antigrowth effect on the human breast (MCF-7) and lung (A549) cancer cell lines and for cytotoxicity against the African Green Monkey kidney (Vero) cell line. The parent chalcone 2a and flavone derivatives 3a, 3c and 3e exhibited relatively high inhibitory activity against the MCF-7 cells with IC50 values of 4.12 ± 0.55, 8.50 ± 0.82, 5.10 ± 0.61 and 6.96 ± 0.66 μM, respectively. The chalcones 2a and 2c exhibited significant cytotoxicity against the A549 cells with IC50 values of 7.40 ± 0.67 and 9.68 ± 0.80 μM, respectively. Only flavone 3c exhibited relatively strong and comparable cytotoxicity against the MCF-7 and A549 cell lines with IC50 values of 6.96 ± 0.66 and 6.42 ± 0.79 μM, respectively. Both series of compounds exhibited strong activity against the MCF-7 and A549 cell lines compared to the analogous quercetin (IC50 = 35.40 ± 1.78 and 35.38 ± 1.78 μM, respectively) though moderate compared to nintedanib (IC50 = 0.53 ± 0.11 and 0.74 ± 0.15 μM, respectively). The test compounds generally exhibited reduced cytotoxicity against the Vero cells compared to this anticancer drug. Molecular docking revealed strong alignment of the test compounds with the enzyme backbone to engage in hydrogen bonding interaction/s and hydrophobic contacts with the residues in the active sites of α-glucosidase and α-amylase. The test compounds possess favorable drug-likeness properties, supporting their potential as therapeutic candidates against T2DM.

Series of the 6-bromo/iodo substituted 2-aryl-4-methyl-1,2-dihydroquinazoline-3-oxides and their mixed 6,8-dihalogenated (Br/I and I/Br) derivatives were evaluated for inhibitory properties against α-glucosidase and/or α-amylase activities and for cytotoxicity against breast (MCF-7) and lung (A549) cancer cell lines. The 6-bromo-2-phenyl substituted 3a and its corresponding 6-bromo-8-iodo-2-phenyl-substituted derivative 3i exhibited dual activity against α-glucosidase (IC50 = 1.08 ± 0.02 μM and 1.01 ± 0.05 μM, respectively) and α-amylase (IC50 = 5.33 ± 0.01 μM and 1.18 ± 0.06 μM, respectively) compared to acarbose (IC50 = 4.40 ± 0.05 μM and 2.92 ± 0.02 μM, respectively). The 6-iodo-2-(4-fluorophenyl)-substituted derivative 3f, on the other hand, exhibited strong activity against α-amylase and significant inhibitory effect against α-glucosidase with IC50 values of 0.64 ± 0.01 μM and 9.27 ± 0.02 μM, respectively. Compounds 3c, 3l and 3p exhibited the highest activity against α-glucosidase with IC50 values of 1.04 ± 0.03, 0.92 ± 0.01 and 0.78 ± 0.05 μM, respectively. Moderate cytotoxicity against the MCF-7 and A549 cell lines was observed for these compounds compared to the anticancer drugs doxorubicin (IC50 = 0.25 ± 0.05 μM and 0.36 ± 0.07 μM, respectively) and gefitinib (IC50 = 0.19 ± 0.04 μM and 0.25 ± 0.03 μM, respectively), and their IC50 values are in the range of 10.38 ± 0.08–25.48 ± 0.08 μM and 11.39 ± 0.12–20.00 ± 0.05 μM, respectively. The test compounds generally exhibited moderate to strong antioxidant capabilities, as demonstrated via robust free radical scavenging activity assays, viz., DPPH and NO. The potential of selected derivatives to inhibit superoxide dismutase (SOD) was also investigated via enzymatic assay in vitro. Molecular docking revealed the N-O moiety as essential to facilitate electrostatic interactions of the test compounds with the protein residues in the active site of α-glucosidase and α-amylase. The presence of bromine and/or iodine atoms resulted in increased hydrophobic (alkyl and/or π-alkyl) interactions and therefore increased inhibitory effect against both enzymes.

Pharmacological studies conducted in the past revealed the potential source of medicinal plants in the development of novel medicines. The phenolic contents of medicinal plants containing chlorogenic acids (CGA) have been linked to a variety of therapeutic effects, especially antiviral activity. Helichrysum aureonitens is a medicinal plant which has been reported to contain chlorogenic acids compounds and has also shown antiviral activities against a number of virus species including Herpes Simplex Virus-1 (HSV-1). In this study, the aim was to determine both the influence of seasonal variation and locality on the antiviral properties of H. aureonitens. Since chlorogenic acids have been reported as potent antiviral compounds, these compounds were targeted to determine the effects of locality and seasonal change on the chlorogenic acid profile, and subsequent antiviral activity. The ultra-performance liquid chromatography-quadrupole time-of-flight mass spectroscopy (UPLC-qTOF-MS) was employed to determine the metabolic profile variations of three derivatives of chlorogenic acids-caffeoylquinic acid (CQA), dicaffeoylquinic acid (DCQA) and tricaffeoylquinic acid (TCQA) in the harvested plants growing in two diverse geographical climates and two different seasons (spring and autumn). Using the cytopathic effect (CPE) reduction approach, twenty-six samples of the plants’ leaves and stems collected during spring and autumn at Telperion nature reserve in Mpumalanga and Wakefield farm, Midlands in KwaZulu-Natal region of South Africa were evaluated for anti-HSV activity. The MTT assay was used for the cytotoxicity evaluation of the extracts prior to antiviral determination. Seventeen (mostly spring collections) of the twenty-six extracts examined were found to have considerable anti-HSV activity as measured by a reduction in tissue culture infectious dose (TCID50) of less than 105. The UPLC-qTOF-MS result revealed that dicaffeoylquinic acid (DCQA) is the most abundant, with higher concentrations in both regions and seasons. 3-CQA was also shown to be the most abundant isomer of caffeoylquinic acid in this investigation.

In this study, bimetallic nanoparticles comprising silver and platinum with promising therapeutic activities were synthesized using ethanolic Vernonia mespilifolia plant extract for the first time. The bimetallic silver-platinum nanoparticles (AgPtNPs) were characterized using solid-state techniques including UV–vis spectroscopy, Fourier transform infrared spectroscopy (FTIR), transmission electron microscopy (TEM), and energy-dispersive X-ray spectroscopy (EDX) techniques. The internal morphological structure showed that the AgPtNPs were spherical with a diameter of approximately 35.5 ± 0.8 nm, while FTIR confirmed the effective capping and formation of the nanoparticles by phytoconstituents. The polyphenolic contents of the green synthesized nanoparticles from the ethanolic extract of V. mespilifolia (AgNPs and AgPtNPs) was found to be (28.0 ± 0.8 and 13.6 ± 0.1 mg GAE/g) total phenol, while the flavonoids content was (366.2 ± 17.0 and 126.6 ± 0.2 mg QE/g), and proanthocyanins content was (161.8 ± 0.6 and 70.2 ± 0.6 mg CE/g). The AgPtNPs displayed a greater ability to scavenge free radicals, especially DPPH and ABTS (IC50 19.5 and 21.6 µg/mL) respectively when compared with AgNPs and ascorbic acid. Besides, the AgPtNPs had a higher ferric reducing antioxidant power (FRAP) (44.1 mg GAE/g) when compared to AgNPs (18.5 mg GAE/g). Moreover, the AgPtNPs showed a two-fold antimicrobial activity towards pathogenic microbes compared to AgNPs and a selective cytotoxic potency towards MCF-7 breast cancer cell line compared to HEK 293 normal cell line. In summary, these fascinating bioactivities displayed by the AgPtNPs highlighted their potential in therapeutic biomedical applications.

This work presents the first report on the phytochemical investigation of Harpephyllum caffrum Bernh. gum exudate. A known cardanol, 3-heptadec-12′-Z-enyl phenol (1) and three new alk(en)ylhydroxycyclohexanes, namely, (1R,3R)-1,3-dihydroxy-3-[heptadec-12′(Z)-enyl]cyclohexane (2) (1S,2S,3S,4S,5R)-1,2,3,4,5-pentahydroxy-5-[octadec-13′(Z)-enyl]cyclohexane (3) and (1R,2S,4R)-1,2,4-trihydroxy-4-[heptadec-12′(Z)-enyl]cyclohexane (4) were isolated from the gum. The structures of the compounds were determined by extensive 1D and 2D NMR spectroscopy and HR-ESI-MS data. The ethanolic extract of the gum was found to be the most potent tyrosinase inhibitor with IC50 of 11.32 µg/mL while compounds 2 and 3, with IC50 values of 24.90 and 26.99 µg/mL, respectively, were found to be potential anti-tyrosinase candidates from the gum. Gum exudate may be a potential source for non-destructive harvesting of selective pharmacologically active compounds from plants. The results also provide evidence that H. caffrum gum may find application in cosmetics as a potential anti-tyrosinase agent.

The 5-(styryl)anthranilamides were transformed into the corresponding 5-styryl-2-(p-tolylsulfonamido)benzamide derivatives. These 5-styrylbenzamide derivatives were evaluated through enzymatic assays in vitro for their capability to inhibit acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and β-secretase (BACE-1) activities as well as for antioxidant potential. An in vitro cell-based antioxidant activity assay involving lipopolysaccharides (LPS)-induced reactive oxygen species (ROS) production revealed that compounds 2a and 3b have the capability of scavenging free radicals. The potential of the most active compound, 5-styrylbenzamide (2a), to bind copper (II) or zinc (II) ions has also been evaluated spectrophotometrically. Kinetic studies of the most active derivatives from each series against the AChE, BChE, and β-secretase activities have been performed. The experimental results are complemented with molecular docking studies into the active sites of these enzymes to predict the hypothetical protein–ligand binding modes. Their drug likeness properties have also been predicted.

Rift valley fever virus (RVFV) is a mosquito-borne virus endemic to sub-Saharan African countries, and the first sporadic outbreaks outside Africa were reported in the Asia-Pacific region. There are no approved therapeutic agents available for RVFV; however, finding an effective antiviral agent against RVFV is important. This study aimed to evaluate the antiviral, antioxidant and anti-inflammatory activity of medicinal plant extracts. Twenty medicinal plants were screened for their anti-RVFV activity using the cytopathic effect (CPE) reduction method. The cytotoxicity assessment of the extracts was done before antiviral screening using the MTT assay. Antioxidant and reactive oxygen/nitrogen species’ (ROS/RNS) inhibitory activity by the extracts was investigated using non-cell-based and cell-based assays. Out of twenty plant extracts tested, eight showed significant potency against RVFV indicated by a decrease in tissue culture infectious dose (TCID50) < 105. The cytotoxicity of extracts showed inhibitory concentrations values (IC50) > 200 µg/mL for most of the extracts. The antioxidant activity and anti-inflammatory results revealed that extracts scavenged free radicals exhibiting an IC50 range of 4.12–20.41 µg/mL and suppressed the production of pro-inflammatory mediators by 60–80% in Vero cells. This study demonstrated the ability of the extracts to lower RVFV viral load and their potency to reduce free radicals.

The prevalence of small multi-target drugs containing a fluorinated aromatic moiety among approved drugs in the market is due to the unique properties of this halogen atom. With the aim to develop potent antidiabetic agents, a series of phenylsulfonic esters based on the conjugation of the 5-substituted 2-hydroxy-3-nitroacetophenones 1a–d with phenylsulfonyl chloride derivatives substituted with a fluorine atom or fluorine-containing (-CF3 or -OCF3) group were prepared. Their structures were characterized using a combination of spectroscopic techniques complemented with a single-crystal X-ray diffraction (XRD) analysis on a representative example. The compounds were, in turn, assayed for inhibitory effect against α-glucosidase, α-amylase, protein tyrosine phosphatase 1 B (PTP1B) and the vascular endothelial growth factor receptor-2 (VEGFR-2) all of which are associated with the pathogenesis and progression of type 2 diabetes mellitus (T2DM). The antigrowth effect of selected compounds was evaluated on the human breast (MCF-7) and lung (A549) cancer cell lines. The compounds were also evaluated for cytotoxicity against the African Green Monkey kidney (Vero) cell line. The results of an in vitro enzymatic study were augmented by molecular docking (in silico) analysis. Their ADME (absorption, distribution, metabolism and excretion) properties have been evaluated on the most active compounds against α-glucosidase and/or α-amylase to predict their drug likeness. This study is a quasi-experimental field evaluation of Raise the Bar (RTB), a prevention programme that trains bar staff to intervene when witnessing risk factors for sexual assault. Previous studies have demonstrated effects of similar programmes on attitudes/knowledge among bar staff. Yet, no studies have examined the effects of such programmes on intervention behaviour. Using a waitlist control design, we conducted thirty observations at four trained and four waitlist control bars in a rural U.S. college town, enacting scripts that matched the prompts for intervention that were included in the RTB curriculum implemented at the trained bars. Findings indicated staff at trained bars were significantly more likely to intervene than staff at the waitlisted bars, but this effect attenuated when control variables were included in the model. RTB appears promising, but further research is needed to fully establish its effects on intervention behaviour before recommending wide-spread rollout. Practice Impact Statement: Findings from this study suggest that training bar staff to act as prosocial bystanders has the potential to prevent alcohol-involved sexual assault among college students. Yet, it is important to note that, although staff at trained bars were more likely to intervene than staff at waitlist control bars, intervention was relatively rare and the effect of training attenuated when controlling for contextual factors. Stronger evidence of the effects of such training needs to accumulate before community agencies widely implement these programmes. [ABSTRACT FROM AUTHOR]

The complex bis(acetonitrile)bromido(η6-1-isopropyl-4-methylbenzene)osmium(II) hexafluoroantimonate, [OsBr(C10H14)(CH3CN)2]SbF6 (1) crystallizes as a triclinic system in space group P1 (no. 2), consisting of two cationic osmium(II) complex ions and two anionic hexafluoroantimonate counterions in each cell. Density Functional Theory (DFT) calculations were performed at the B3LYP/LanL2DZ and B3LYP/Def2 − SVP/Def2 − TZVP levels of theory. Reactivity descriptors and charge transfer properties were computed using frontier molecular orbital analysis. The DFT-optimized model systems from both basis sets were comparable to the experimentally determined X-ray diffraction results. Hirshfeld surface analysis shows significant non-covalent interactions including hydrogen bonds and halogen interactions. The MTT technique was utilized to conduct an in vitro cytotoxicity assay of 1 against cancerous cell lines (MCF-7 and A549) and normal cells (HEK293). Complex 1 is potent towards cancer cells as evidenced by the IC50 values of 5.02 ± 0.05 µM for MCF-7 cells and 13.36 ± 0.04 µM for A549 cells. Complex 1 demonstrated lower toxicity to normal cells with an IC50 value of 26.76 ± 0.08 µM for the HEK293 cell line compared to the doxorubicin standard with an IC50 value of 1.2 ± 0.1 µM. Background: Diabetes Mellitus is a chronic metabolic disorder caused by genetic disorders and other factors such as the use of certain medications, pancreatic injury, and autoimmune diseases such as rheumatoid arthritis and high blood pressure. However, several therapeutic agents are currently used to manage blood sugar levels, thus reducing the risk of complications. Despite efforts to treat or manage this disease, current therapeutic agents have detrimental side effects, presenting a considerable need to develop new effective drugs with fewer adverse effects. The aim of the study: This study aimed to evaluate the inhibitory capacity of I. bolusiana and I. crassipe plant extracts on α-amylase enzyme and to assess the antioxidant capacity. Furthermore, the evaluation of the cytotoxicity properties and glucose metabolism of the extracts was carried out in RAW 264.7 macrophages, C3A hepatocytes and L6 myoblast cell lines. Methods: In vitro antidiabetic activity was tested with α-amylase enzyme inhibition method and antioxidant activity was evaluated. Furthermore, their ability to quench free radicals was investigated using the 2,2-diphenyl-1-picryhydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) free radical scavenging method and the superoxide dismutase (SOD) enzyme scavenging method was investigated. Cytotoxicity and glucose metabolism were measured in macrophage (RAW 264.7), hepatocytes (C3A) and myoblast (L6) cells using the colorimetric test 3- (4,5-dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide (MTT). Furthermore, phytochemical characterization and tentative identification was performed using FTIR, 1HNMR and UHPLCOrbitrap HRMS, FTIR, whilst computational studies were also employed to determine the biochemical interaction, antidiabetic and inhibitory potential of the extracts with the α-amylase enzyme. Results: The extracts of I. bolusiana and I. crassipes exhibited moderate inhibitory activity against α-amylase with an inhibitory percentage range of 10.72 – 30.08 %. The extracts did not show cytotoxic effects in the treated cells. The extracts of I. bolusiana and I. crassipes significantly increased glucose uptake in L6 and C3A cell lines. The extracts reduced the DPPH and ABTS radicals, showing a range of IC50 values of 7.35 ± 0.43 - 12.02 ± 0.21 µg/mL. However, the extracts were less toxic to the cells as they showed IC50 values > 20 µg/mL. The phytochemical evaluation demonstrated the presence of phenolics, chlorogenic acids, coumarin glucosides and flavonoids in the extracts, which were docked with the α-amylase enzyme. 5-caffeoylquinic acid exhibited the highest binding energy of -6.829 kcal/mol. Conclusions: This study presents various important phytoconstituents and broad-spectrum activities, including α-amylase inhibitory activity, reduction of oxidative stress, less toxicity, and increased glucose metabolism of methanol extracts of I. bolusiana and I. crassipes as a promising source for the isolation of molecules responsible for antidiabetes activities. The results documented in this study will be valuable leads in the development of therapeutic candidates for diabetes and oxidative stress.

The application of plant extracts in the green synthesis of silver nanoparticles (AgNPs) has attracted significant attention owing to their eco-friendliness and cost-effective features. However, nanoparticles synthesized from plant materials may also display unique physicochemical properties. This study described an improved synthesis process for silver nanoparticles (AgNPs) from the peel of the Citrus unshiu fruit, an agricultural waste product. In addition to evaluating the produced AgNPs' physico-chemical characteristics, the research also examines their antibacterial and cytotoxic/anticancer capabilities. The color change from yellow to dark brown was observed immediately when 20 ml of Citrus unshiu fruit peel extract was added to 0.5 g of silver nitrate. A surface plasmon resonance band at 424 nm revealed by ultra-violet spectroscopy was used as additional confirmation that AgNP had formed. The scanning electron microscope (SEM), and the transmission electron microscope (TEM), revealed spherical-shaped NPs with a mean size of 23 nm. Furthermore, X-ray diffraction (XRD) confirmed the high crystallinity of the obtained NPs while Fourier transform infrared spectroscopy (FTIR) confirmed the bioreducing capacity of the Citrus unshiu fruit peel extract. The minimum inhibitory concentration (MIC) (31.25 μg/mL) of AgNPs significantly revealed a strong dose-dependent antibacterial effect on Staphylococcus aureus and Bacillus cereus. In contrast, their impact on gram-negative bacteria, such as Escherichia coli and Salmonella typhimurium, displayed an MIC of 250 μg/mL. The assessment of cytotoxicity on human embryonic kidney cells (HEK293) and lung cancer cells (A549) through a 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide assay revealed no detectable cytotoxic effects. The IC50 values were determined as 37.73 ± 0.34 μg/mL for HEK293 and 56.37 ± 0.73 μg/mL for A549, indicating the absence of significant adverse impacts on these cell lines. The anticancer drug doxorubicin was used as a reference standard, and had an IC50 value of 1.22 ± 0.09 and 0.76 ± 0.12 μg/mL against HEK293 and A549 cells, respectively. The results show that AgNPs synthesized from Citrus unshiu fruit peel could serve as antibacterial agents that may in the future be optimized for us in for example nanofiltration systems.

Green synthesized nanoparticles are believed to alleviate the detrimental effects of chemical fertilizers and the multidrug resistance of various microbial strains. This study evaluates the use of bush tea-mediated zinc oxide nanoparticles (ZnO NPs) for chickpea fertilization and assesses their cytotoxicity, antioxidant and antibacterial activity. The study demonstrated that the application of ZnO NPs improved the number of leaves, nodules, biomass and yield of chickpea cultivars under glasshouse conditions. The authors suggest that biosynthesized ZnO NPs are a good plant growth stimulant. The control with no nanoparticles application, had higher values of photosynthetic rate and water use efficiency in the Desi genotype whereas for the Kabuli genotype, 25 mg/L showed higher values of stomatal conductance and transpiration efficiency, while 15 mg/L gave the highest water use efficiency and photosynthetic rate. Furthermore, the inhibitory activities of bacteria were reported in which E. coli and S. aureus were the most sensitive bacterial strains. The ZnO NPs also quenched free radicals that demonstrate antioxidant activity. Thus, they can be beneficial to both plants and humans as they reduce oxidative stress that can cause diseases. Furthermore, toxicity to cells was evaluated and ZnO NPs were less toxic to the studied cells with IC50 values of 16.17 µg/mL and 21.30 µg/mL against lung cancer and embryonic cell lines. Thus, the use of ZnO nanoparticles with antimicrobial activity could help fight against several diseases attacking humans and plants.
