The National Environmental Isotope Facility (NEIF) Radiocarbon Laboratory at the Scottish Universities Environmental Research Centre (SUERC) performs radiocarbon measurement of a wide range of sample matrices for applications in environmental research. Radiocarbon is applied to palaeoenvironmental, palaeoceanographic, and palaeoclimatic investigations, as well as work to understand the source, fate, turnover, and age of carbon in the modern carbon cycle. The NEIF Radiocarbon Laboratory supports users in the development and deployment of novel sampling techniques and laboratory approaches. Here, we give an overview of methods and procedures used by the laboratory to support the field collection, laboratory processing, and measurement of samples. This includes in-house development of novel and/or specialized methods and approaches, such as field collection of CO
2 and CH4 , hydropyrolysis, and ramped oxidation. Radiocarbon (
14 C) measurements undertaken by the NERC Radiocarbon Laboratory using accelerator mass spectrometry (AMS) are now freely available on a new online database. The data presented covers measurement of the wide range of sample types that are processed for research projects in the fields of Earth and environmental science, supported by the United Kingdom's Natural Environment Research Council. Sample types within the database include organic remains, soils, sediments, carbonates, dissolved organic and inorganic carbon, and carbon dioxide. Currently, the database contains14 C data for over 2400 individual samples that were measured and reported between 2005 and 2013, but it is envisaged that this will expand considerably as more data are made available. Contextual information such as sampling location and associated publications are provided where available, and searches can be performed on sample location, sample type, project number, and publication code. This new database compliments an existing, publicly available database of measurements performed using radiometric methods by the laboratory which has recently been expanded to present over 2000 measurements. It is hoped that this archive will prove useful to workers in the community who would benefit from greater availability of measurements for particular locations or sample types, and for the purposes of performing meta-analyses, and/or synthesis of larger datasets. 8. Citizenship and ideology in David Cameron's 'Big Society'

- Abstract
As the main idea of the governments of David Cameron after 2010, the Big Society intended to place civil society at the heart of the Conservative agenda while also serving as a rhetorical tool to distance the ‘modern’ Conservative party from the Thatcherite legacy. In comparing Margaret Thatcher’s view on citizenship to the one upheld by David Cameron, this article argues that despite many similarities, Cameron broke with Thatcher in the way he reinterpreted the nature of the state, as well as how he planned to remake civil society and to inspire ‘active citizenship’. Unlike Thatcher, who believed that a dynamic civil society would spontaneously flourish once the state was reduced, Cameron believed that society could be rebuilt only through the work of the state. From this perspective, it is possible to reassess the academic debate about the Big Society and to regard it not as a means of justifying a smaller state at a time of economic austerity, but rather as an initiative which failed at least in part because of austerity.

The way that mothers talk about the past (reminisce) with young children is linked to key memory, language, and socioemotional outcomes. The present research explored the role of a range of child, maternal, socioeconomic, and cultural factors that predict maternal reminiscing style, with a particular focus on maternal personality and child temperament. A total of 1,404 mother-child dyads from the prebirth longitudinal cohort study Growing Up in New Zealand (https://www.growingup.co.nz) participated in a reminiscing task about a negative event when children were 8 years old. This broader cohort is broadly representative of the New Zealand population in terms of maternal ethnicity and socioeconomic status. Conversations were scored using a revised version of the Elaborative Reminiscing Scale. Child temperament during infancy, but not childhood, uniquely predicted maternal reminiscing style. Maternal extraversion also predicted a more elaborative reminiscing style. Other maternal factors, including education, ethnicity, and age, were also identified as unique predictors of maternal reminiscing style. Sample materials such as sediments and soils contain complex mixtures of different carbon-containing compounds. These bulk samples can be split into individual fractions, based on the temperature of thermal decomposition of their components. When coupled with radiocarbon (
14 C) measurement of the isolated fractions, this approach offers the advantage of directly investigating the residence time, turnover time, source, or age of the different components within a mixed sample, providing important insights to better understand the cycling of carbon in the environment. Several laboratories have previously reported different approaches to separate radiocarbon samples based on temperature in what is a growing area of interest within the research community. Here, we report the design and operation of a new ramped oxidation facility for separation of sample carbon on the basis of thermal resistance at the NEIF Radiocarbon Laboratory in East Kilbride, UK. Our new instrumentation shares some characteristics with the previously-reported systems applying ramped oxidation and/or ramped pyrolysis for radiocarbon measurement, but also has several differences which we describe and discuss. We also present the results of a thorough program of testing of the new system, which demonstrates both the reproducibility of the thermograms generated during sample combustion, and the reliability of the radiocarbon measurements obtained on individual sample fractions. This is achieved through quantification of the radiocarbon background and analysis of multiple standards of known14 C content during standard operation of the instrumentation. Rates of global and regional sea‐level rise between ~1850 and 1950 were high compared to those in preceding centuries. The cause of this sea‐level acceleration remains uncertain, but it appears to be pronounced in a small set of relative sea‐level proxy records from the Southern Hemisphere. Here we generate three new proxy‐based relative sea‐level reconstructions for southeastern Australia to investigate spatial patterns and causes of historical sea‐level changes in the Tasman Sea. Palaeo sea‐level estimates were determined using salt‐marsh foraminifera as sea‐level indicators. Records are underpinned by chronologies based on accelerator mass spectrometry 14C, radiogenic lead (210Pb), stable lead isotopes and palynological analyses. Our reconstructions show that relative sea level rose by ~0.2–0.3 m over the last 200 years in southeastern Australia, and rates of sea‐level rise were especially high over the first half of the 20th century. Based on modelled estimates of the contributing components to sea‐level rise, we suggest that the episode of rapid sea‐level rise was driven by barystatic contributions, but sterodynamic contributions were dominant by the mid‐20th century. Significant spatial variability in relative sea level indicates that local to sub‐regional drivers of sea level are also prominent. Our reconstructions significantly enhance our understanding of the spatiotemporal pattern of early 20th century sea‐level rise in the region. [ABSTRACT FROM AUTHOR]

PubMed: 31209238, The effectiveness of most cancer targeted therapies is short-lived. Tumors often develop resistance that might be overcome with drug combinations. However, the number of possible combinations is vast, necessitating data-driven approaches to find optimal patient-specific treatments. Here we report AstraZeneca’s large drug combination dataset, consisting of 11,576 experiments from 910 combinations across 85 molecularly characterized cancer cell lines, and results of a DREAM Challenge to evaluate computational strategies for predicting synergistic drug pairs and biomarkers. 160 teams participated to provide a comprehensive methodological development and benchmarking. Winning methods incorporate prior knowledge of drug-target interactions. Synergy is predicted with an accuracy matching biological replicates for >60% of combinations. However, 20% of drug combinations are poorly predicted by all methods. - Abstract
The effectiveness of most cancer targeted therapies is short-lived. Tumors often develop resistance that might be overcome with drug combinations. However, the number of possible combinations is vast, necessitating data-driven approaches to find optimal patient-specific treatments. Here we report AstraZeneca’s large drug combination dataset, consisting of 11,576 experiments from 910 combinations across 85 molecularly characterized cancer cell lines, and results of a DREAM Challenge to evaluate computational strategies for predicting synergistic drug pairs and biomarkers. 160 teams participated to provide a comprehensive methodological development and benchmarking. Winning methods incorporate prior knowledge of drug-target interactions. Synergy is predicted with an accuracy matching biological replicates for >60% of combinations. However, 20% of drug combinations are poorly predicted by all methods. Genomic rationale for synergy predictions are identified, including ADAM17 inhibitor antagonism when combined with PIK3CB/D inhibition contrasting to synergy when combined with other PI3K-pathway inhibitors in PIK3CA mutant cells.

Esophageal adenocarcinoma (EAC) incidence is increasing while 5-year survival rates remain less than 15%. A lack of experimental models has hampered progress. We have generated clinically annotated EAC organoid cultures that recapitulate the morphology, genomic, and transcriptomic landscape of the primary tumor including point mutations, copy number alterations, and mutational signatures. Karyotyping of organoid cultures has confirmed polyclonality reflecting the clonal architecture of the primary tumor. Furthermore, subclones underwent clonal selection associated with driver gene status. Medium throughput drug sensitivity testing demonstrates the potential of targeting receptor tyrosine kinases and downstream mediators. EAC organoid cultures provide a pre-clinical tool for studies of clonal evolution and precision therapeutics., Esophageal adenocarcinoma (EAC) has a poor 5-year survival rate and lacks robust preclinical models for use in research. Here, the authors show that newly derived organoids recapitulate the transcriptomic, genetic, and morphological landscape of the primary EAC tumors and provide a platform to test drug sensitivity and study tumor clonality.

The primary objective of the work described in this thesis was to investigate some of the effects of chronic oxygen toxicity in mammals. This thesis describes in vivo changes in certain metabolites and enzymes following the exposure of mice and rats to increased concentrations of oxygen at atmospheric pressure. The introductory section reviews the historical background to oxygen toxicity, delineates its two forms and describes their aetiology. The physical and chemical properties of oxygen which contribute to its unusual reactivity are discussed, together with the reactive species of oxygen which may be responsible for its toxicity. To complete the introduction, the known effects of oxygen on metabolism are reviewed. In order to pursue these studies it was necessary to design an apparatus capable of providing a controlled high oxygen atmosphere. Its construction and operation is described. Results obtained indicated that the levels of the protective enzymes superoxide dismutase and catalase varied in lung, liver and brain of mice following exposure to increased oxygen levels. Overall protein levels were also found to vary, so an experiment involving a polyacrylamide gel separation was performed to reveal any qualitative differences. Another experiment monitored the changes in free amino acids in brain, liver, lung and muscle. A few possible deviations from normal were detected but these changes were not consistent between the tissues studied. Results are presented of the changes in some gross physical parameters and the effect of 3 prostaglandin inhibitors on these in vivo changes. Other experiments revealed the changes in haematocrit, in retinol and in haemoglobin levels. For comparative purposes some of the above measurements were also applied to samples of body fluids from men suffering a mild experimental oxygen toxicity. The relevance of these results to the toxic effects of oxygen in mammals is discussed.
