Abstract

Caloric Restriction (CR) has established anti-cancer effects, but its clinical relevance and molecular mechanism remain largely undefined. Here, we investigate CR's impact on several mouse models of Acute Myeloid Leukemias, including Acute Promyelocytic Leukemia, a subtype strongly affected by obesity. After an initial marked anti-tumor effect, lethal disease invariably re-emerges. Initially, CR leads to cell-cycle restriction, apoptosis, and inhibition of TOR and insulin/IGF1 signaling. The relapse, instead, is associated with the non-genetic selection of Leukemia Initiating Cells and the downregulation of double-stranded RNA (dsRNA) sensing and Interferon (IFN) signaling genes. The CR-induced adaptive phenotype is highly sensitive to pharmacological or genetic ablation of LSD1, a lysine demethylase regulating both stem cells and dsRNA/ IFN signaling. CR + LSD1 inhibition leads to the re-activation of dsRNA/IFN signaling, massive RNASEL-dependent apoptosis, and complete leukemia eradication in similar to 90% of mice. Importantly, CR-LSD1 interaction can be modeled in vivo and in vitro by combining LSD1 ablation with pharmacological inhibitors of insulin/IGF1 or dual PI3K/MEK blockade. Mechanistically, insulin/IGF1 inhibition sensitizes blasts to LSD1-induced death by inhibiting the anti-apoptotic factor CFLAR. CR and LSD1 inhibition also synergize in patient-derived AML and triple-negative breast cancer xenografts. Our data provide a rationale for epi-metabolic pharmacologic combinations across multiple tumors.

Abstract

A standardized methodology for the ultrasound evaluation of the pelvic sidewall has not been proposed to date. Herein, a collaborative group of gynecologists and gynecological oncologists with extensive ultrasound experience presents a systematic methodology for the ultrasonographic evaluation of structures within the pelvic sidewall. Five categories of anatomical structures are described (muscles, vessels, lymph nodes, nerves and ureters). A step‐by‐step transvaginal ultrasound (or, when this is not feasible, transrectal ultrasound) approach is outlined for the evaluation of each anatomical landmark within these categories. Accurate assessment of the pelvic sidewall using a standardized approach improves the detection and diagnosis of non‐gynecological pathologies that may mimic gynecological tumors, reducing the risk of unnecessary and even harmful intervention. Furthermore, it plays an important role in completing the staging of malignant gynecological conditions. Transvaginal or transrectal ultrasound therefore represents a viable alternative to magnetic resonance imaging in the preoperative evaluation of lesions affecting the pelvic sidewall, if performed by an expert sonographer. A series of videoclips showing normal and abnormal findings within each respective category illustrates how establishing a universally applicable approach for evaluating this crucial region will be helpful for assessing both benign and malignant conditions affecting the pelvic sidewall. © 2024 The Author(s). Abstract

ABSTRACT The lymphatic pathway is an important route of metastasis in gynecological malignancy. Therefore, the examination of lymph nodes is an essential part of the ultrasound evaluation in patients with known or suspected gynecological malignancy. The lymph nodes most frequently involved in gynecological malignancy (apart from vulvar cancer) are parietal (retroperitoneal) and visceral abdominopelvic lymph nodes. In advanced disease, more distant lymph‐node regions, such as the inguinal, axillary and supraclavicular lymph nodes, can also be involved. The standardized description of lymph nodes has been published previously by the Vulvar International Tumor Analysis (VITA) collaborative group. Herein, a collaborative group of gynecologists and gynecological oncologists with extensive ultrasound experience presents a systematic methodology for ultrasonographic lymph‐node assessment performed as part of the locoregional and distant work‐up to assess the extent of gynecological malignancy. The aim of this consensus opinion is also to describe the anatomical classification and drainage pathways of the lymphatic system as relevant to the gynecological organs. © 2024 The Author(s). Abstract

We address nonradiative recombination pathways by leveraging surface passivation and dislocation management in micron-scale arrays of Ge crystals grown on deeply patterned Si substrates. The time decay photoluminescence (PL) at cryogenic temperatures discloses carrier lifetimes approaching 45 ns in band-gap engineered Ge micro-crystals. This investigation provides compelling information about the competitive interplay between the radiative band-edge transitions and the trapping of carriers by dislocations and free surfaces. Furthermore, an in-depth analysis of the temperature dependence of the PL, combined with capacitance data and finite difference time domain modeling, demonstrates the effectiveness of GeO2 in passivating the surface of Ge and thus in enhancing the room temperature PL emission.

Abstract

The circular polarization of direct gap emission of Ge is studied in optically-excited tensile-strained Ge-on-Si heterostructures as a function of doping and temperature. Owing to the spin-dependent optical selection rules, the radiative recombinations involving strain-split light (cG-LH) and heavy hole (cG-HH) bands are unambiguously resolved. The fundamental cG-LH transition is found to have a low temperature circular polarization degree of about 85% despite an off-resonance excitation of more than 300 meV. By photoluminescence (PL) measurements and tight binding calculations we show that this exceptionally high value is due to the peculiar energy dependence of the optically-induced electron spin population. Finally, our observation of the direct gap doublet clarifies that the light hole contribution, previously considered to be negligible, can dominate the room temperature PL even at low tensile strain values of about 0.2%.

Abstract

We present the combined results on electron-pair production in 158 GeV/n {Pb-Au} ($\sqrt{s}$= 17.2 GeV) collisions taken at the CERN SPS in 1995 and 1996, and give a detailed account of the data analysis. The enhancement over the reference of neutral meson decays amounts to a factor of 2.31$\pm0.19 (stat.)\pm0.55 (syst.)\pm0.69 (decays)$ for semi-central collisions (28% $\sigma/\sigma_{geo}$) when yields are integrated over $m>$ 200 MeV/$c^2$ in invariant mass. The measured yield, its stronger-than-linear scaling with $N_{ch}$, and the dominance of low pair $p_t$ strongly suggest an interpretation as {\it thermal radiation} from pion annihilation in the hadronic fireball. The shape of the excess centring at $m\approx$ 500 MeV/$c^2$, however, cannot be described without strong medium modifications of the $\rho$ meson. The results are put into perspective by comparison to predictions from Brown-Rho scaling governed by chiral symmetry restoration, and from the spectral-function many-body treatment in which the approach to the phase boundary is less explicit., Comment: 39 pages, 40 figures, to appear in Eur.Phys.J.C. Abstract

The abandonment of silvicultural activities can lead to changes in species richness and composition of biological communities, when compared to those found in managed forests. The aim of this study was to compare the multi-taxonomical diversity of two mature silver fir-beech-spruce forests in the southern Dolomites (Italy), corresponding to the European Union habitat type 9130. The two sites share similar ecological and structural characteristics, but differ in their recent management histories. In the last 50 years, one site underwent non-intensive management, while the other was left unmanaged and was included in a forest reserve. The species richness and composition of eight taxa were surveyed in the two sites between 2009 and 2011. The difference in mean species richness between the two forest management types was tested through permutation tests, while differences in species composition were tested by principal coordinates analysis and the permutational multivariate analysis of variance. Mean species richness of soil macrofungi, deadwood lichens, bark beetles, and longhorn beetles were significantly higher in the abandoned than in the non-intensively managed forests. Deadwood fungi and epiphytic lichens did not differ in mean species richness between the two study sites, while mean species richness of ground beetles and birds were higher in the non-intensively managed than in the abandoned forest. Significant differences in species composition between the two sites were found for all the taxa, except for longhorn beetles. These results indicate that improving forest landscape heterogeneity through the creation of a mosaic of abandoned and extensively managed forests should better fulfill the requirements of ecologically different taxa.

Abstract

The goal of the present study is to explore the perception of the relationship with parental figures, traumatic experiences, personality traits and psychosocial characteristics of the participant sibling caregivers. The sample was composed of 30 sibling caregivers recruited at psychiatric facilities in Italy, and of 30 control siblings. The battery of instruments administered included Parental Bonding Instrument (PBI), Minnesota Multiphasic Personality Inventory 2 (MMPI-2), and The Inventory of Traumatic Experiences (TEC). This research found that sibling caregivers of patients with severe psychiatric pathologies are distinctly different from the siblings of the control group with respect to the perception of their relationship with parental figures more frequently regarded as dysfunctional, and were also characterized by a higher presence of traumatic experiences. The problematic relationship with parental figures, some traumatic experiences, and the burden of taking care of a sibling with psychiatric disorders are probably important variables with regards to the individual's overall psychological condition.

Abstract

Mineral-hosted melt inclusions have become an important source of information on magmatic processes. As the number of melt inclusion studies increases, so does the need to establish recommended practice guidelines for collecting and reporting melt inclusion data. These guidelines are intended to ensure certain quality criteria are met and to achieve consistency among published melt inclusion data in order to maximize their utility in the future. Indeed, with the improvement of analytical techniques, new processes affecting melt inclusions are identified. It is thus critical to be able to reprocess any previously published data, such that reporting the raw data is one of the first “recommended practices” for authors and a publication-criteria that reviewers should be sensitive to. Our guidelines start with melt inclusion selection, which is a critical first step, and then continue on to melt inclusion preparation and analysis, covering the entire field of methods applicable to melt inclusions. Dedication: In March of 2000, a melt inclusion workshop was held at the Chateau de Sassenage in Grenoble and a companion issue of Chemical Geology entitled “Melt Inclusions at the Millennium” was published. Erik Hauri was heavily involved with the meeting and contributed two landmark papers to the topical issue of Chemical Geology on the use of secondary ion mass spectrometry to analyze volatiles in melt inclusions. When the melt inclusion community re-convened at Woods Hole Oceanographic Institution (WHOI) in August of 2018, we were saddened that Erik was unable to join us due to his failing health. Less than a month later came the devastating news of his passing at only 52 years of age. In recognition of his incredible contributions to science in general and to the in situ analysis of melt inclusions in particular, the participants and organizers of the WHOI melt inclusion workshop dedicate this collegial paper to Erik Hauri, our colleague, mentor and friend. Abstract

La presente ricerca esplora alcune caratteristiche psicologiche presenti nei fratelli con funzione di caregiver di pazienti con gravi patologie psichiche. Gli autori si focalizzano su aspetti quali la percezione della relazione con le figure genitoriali, le esperienze traumatiche, i tratti di personalità e alcune caratteristiche psicosociali. Il campione è costituito da 60 partecipanti: 30 fratelli di persone con gravi patologie psichiatriche (Gruppo A), reperiti all'interno di associazioni di fami liari e servizi psichiatrici territoriali, e 30 partecipanti del gruppo di controllo (Gruppo B) reclutati bilanciandoli al gruppo A in termini di caratteristiche; i partecipanti di entrambi i gruppi risultano residenti al Nord Italia. Sono stati impiegati i seguenti questionari self-report: il Parental Bonding Instrument- PBI, il Minnesota Multiphasic Personality Inventory 2-MMPI-2, l'Inventario delle Esperienze Traumatiche- TEC e un Questionario anamnestico. I risultati mettono in luce come i fratelli caregiver di persone con grave disagio psichico presentino diverse caratteristiche peculiari rispetto al gruppo di controllo; ci si riferisce, in particolare, alle caratteristiche della struttura familiare, alle difficoltà relazionali con le figure genitoriali e alle esperienze traumatiche vissute a livello familiare.

Abstract

To be position sensitive, silicon drift detectors (SDDs) require a trigger signal synchronous with the interaction time (i.e., the start of the electrons' motion) since the interaction position along the drift coordinate is obtained from the measurement of the electrons' drift time. In the case of a photon interaction, the trigger must be provided by the detector itself. One way to obtain a fast trigger signal exploits the induction signal given by the holes collected by the [p.sup.+] strips close to the interaction point. We have designed and instrumented a multilinear SDD especially intended to study the self-triggering technique and its application to two-dimensional (2-D) X-ray position sensing. The measured position resolution along the drift coordinate is of the order of 10 ns with 10 keV photons, leading to a position resolution of 30 [micro]m rms at typical drift velocities. A comparable position resolution can be obtained on the lateral coordinate. By engineering the electron trajectory in the collection region, the anode capacitance can be minimized ( Index Terms--High-rate spectroscopy, position-sensing detector, silicon drift detectors, X-ray imaging.

Abstract

This article provides a systematic methodology for the ultrasound evaluation of the pelvic sidewall, which is important for assessing the spread of gynecological malignancies and diagnosing benign gynecological conditions causing pelvic pain. The article describes the anatomical structures within the pelvic sidewall and provides step-by-step instructions for their evaluation using transvaginal ultrasound. The authors conclude that accurate assessment of the pelvic sidewall using ultrasound can improve the detection and diagnosis of non-gynecological pathologies, reduce unnecessary interventions, and aid in the staging of malignant gynecologic conditions. Abstract

In this paper we present a new VLSI shaper amplifier suitable for multi-anode Silicon Drift Detectors, or other multi-element solid-state X-ray detection systems. The readout scheme has been conceived and developed for demanding applications with synchrotron light sources, such as X-ray holography or EXAFS experiments, where both high count-rates and high energy resolutions are required. The circuit is of time-variant nature, accepts randomly distributed events and features a finite-width (1.1 [[micro]second]) weight function, a low-level energy discrimination, a low power consumption (a few mW per channel) and a full compatibility with monolithic integration in CMOS technology. Its impulse response has a staircase-like shape, but the implemented weight function (which is in general different from the impulse response in time-variant systems) is quasi trapezoidal. A first prototype of the circuit is here described and the first experimental results are also reported.

Abstract

Obesity correlates with hematological malignances including leukemias, but risk of specific leukemia subtypes like Acute Promyelocytic Leukemia and underlying molecular mechanisms are poorly understood. We explored multiple datasets for correlation between leukemia, Body Mass Index and molecular features. In a population-based study (n=5.2 million), we correlated Body Mass Index with promyelocytic, other acute myeloid, lymphoid or other leukemias. In cross-sectional studies, we tested body mass index deviation in promyelocytic leukemia trial cohorts from what expected based on national surveys. We interrogated The Cancer Genome Atlas for transcriptional signatures and mutations enriched in promyelocytic leukemia and/or obesity and confirmed correlation between body mass and FLT3 mutations in promyelocytic leukemia cohorts by logistic regression. In the population-based study, Hazard Ratio per 5 kg/m2 increase was: promyelocytic leukemia 1.44 (95% CI 1.0-2.08); non-promyelocytic acute myeloid leukemias 1.17 (1.10-1.26); lymphoid leukemias 1.04 (1.0-1.09); other 1.10 (1.04-1.15). Abstract

In this paper the authors present a novel JFET/CMOS signal processor, ROtational TrapezOidal Readout (ROTOR), conceived for high-rate high-resolution X-ray spectroscopy with asynchronous event occurrence. The circuit is of time-variant nature, uses no fast channel as a trigger, and displays a quasitrapezoidal weight function with virtually no exponential tail. Such a finite-width weight function is typically nearly optimal for submicrosecond processing times and is adequate when intense radiation fluxes must be handled. The filter also permits us to suppress possible ballistic deficits. Abstract

In this paper we present the working principle and the design criteria of the Controlled-Drift Detector, a novel device suitable for the simultaneous measurement of energy and two-dimensional position of X rays. In the Controlled-Drift Detector the pixel structure typical of a charge-coupled device and the fast readout typical of a silicon drift detector are joined. When the radiation is to be detected, suitable potential barriers are generated to prevent the drift of the signal electrons that are confined within a matrix of integration wells. The potential barrier which prevents the drift is selectively removable to allow the fast transport of the confined signal electrons to the readout electrodes by means of a static drift field. In principle such a device can achieve excellent energy resolution and readout times of some tens of microseconds. Three different confining mechanisms that allow control of the drift of the stored electrons are studied, and their advantages and drawbacks are discussed. First prototypes of the Controlled-Drift Detector are currently in production. Index Terms - Fast read-out, high-energy resolution, silicon detector, X-ray imaging.
