1. Dopamine pathway characterization during the reproductive mode switch in the pea aphid

Abstract

Aphids are major pests of most of the crops worldwide. Such a success is largely explained by the remarkable plasticity of their reproductive mode. They reproduce efficiently by viviparous parthenogenesis during spring and summer generating important damage on crops. At the end of the summer, viviparous parthenogenetic females perceive the photoperiod shortening and transduce this signal to their embryos that change their reproductive fate to produce sexual individuals: oviparous females and males. After mating, those females lay cold-resistant eggs. Earlier studies showed that some transcripts coding for key components of dopamine pathway were regulated between long days and short days conditions suggesting that dopamine might be involved in the transduction of seasonal cues prior to reproductive mode switch. In this study, we aimed at going deeper into the characterization of the expression dynamics of this pathway but also in the analysis of its functional role in this context in the pea aphid Acyrthosiphon pisum. We first analysed the level of expression of ten genes of this pathway in embryos and larval heads of aphids reared under long days (asexual producers) or short days (sexual producers) conditions. We then performed in situ hybridization experiments to localize in embryos the ddc and pale transcripts that are coding for two key enzymes in dopamine synthesis. Finally, Using CRISPR-Cas9 mutagenesis in eggs produced after the mating of sexual individuals, we targeted the ddc gene. We could observe strong melanization defaults in ddc mutated eggs, which confidently mimicked the Drosophila ddc phenotype. Nevertheless, such a lethal phenotype did not allow us to validate the involvement of dopamine as a signaling pathway necessary to trigger the reproductive mode switch in embryos.

Abstract

The Sound Settler is a utopian project which combines engineering and the arts. The utopian vision is to deploy an interactive music system on Mars before the first humans arrive. Two real robotic arms are controlled in a gravity compensated master-slave mode resembling the human part on Earth, and the robotic art piece which is envisioned to be located on Mars. The movements generate electronic music by charging physical capacitors and on screen a simulated third arm is deployed, visualizing the movements on the red planet. This report presents the artistic design, the implemented infrastructure and first subjective insights about human interactions with the art piece in public spaces. It motivates a more formal analysis and opens several research avenues for the future.

Abstract

Arctic ecosystems are subjected to strong environmental constraints that prevent both the colonization and development of many organisms. In Svalbard, few aphid species have established permanent populations. These high arctic aphid species have developed peculiar life-history traits such as shortened life cycles and reduced dispersal capacities. Here, we present data on the distribution and population genetics of Acyrthosiphon svalbardicum in Spitsbergen, the main island of the Svalbard archipelago, and compared its genetic structure with that of its close relative Acyrthosiphon brevicorne, sampled in the top of Scandinavian mainland. We found that A. svalbardicum is common but heterogeneously distributed along the west coast of Spitsbergen. We recorded this species up to 79&deg, 12&rsquo, which constitutes the northernmost location for any aphid. Genetic structure examined using microsatellite markers showed more pronounced spatial differentiation in A. svalbardicum than in A. brevicorne populations, presumably due to reduced dispersal capacities in the former species. Although populations of A. brevicorne and A. svalbardicum were well-delineated at nuclear loci, they shared similar cytoplasmic DNA haplotypes as revealed by sequence analysis of two DNA barcodes. These results raise questions about whether these two taxa are different species, and the colonization sources and history of the Svalbard archipelago by A. svalbardicum.

Abstract

Two important race groups of the stem nematode, Ditylenchus dipsaci, an obligatory polyphagous endoparasite, attack the bean, Vicia faba. Morphological differences have been found in the two groups and there were corresponding differences in faba bean disease symptoms. Agreement has been found between RAPD and phenotypic characteristics in the nematodes. Variation between D. dipsaci species in populations involved with V. faba was low. No strong correlation was found between geographical origin and morphological or molecular characteristics.

Abstract

Thirty-seven pearls originating from bivalves of the Tridacnidae family were analyzed to determine their gemological properties and to characterize the optical features of their distinctive flame structure, terminating at the apex and base with patterns like those of blade impellers. All but two of the samples exhibited cylindrical symmetry around a fixed axis, suggesting a rotating movement during growth, as with numerous pearls from Pinctada margaritifera. One quite unusual feature, which is difficult to detect and to our knowledge has never before been described, consisted of spiral patterns at the apex and base of the pearl, and one of these patterns suggested the possibility of rotation. Other details that supported the conclusion of rotation during growth included comets associated with spot defects and pseudo-chatoyancy that appeared shifted to one side.

Abstract

This article documents an optical illusion displayed by the asterism of one of the largest-known star garnets. This dark brownish purple gem weighs 5,737 ct and exhibits a sharp, classical four-rayed star. The three-dimensional phenomenon—in which some or all branches of the asterism appear to float above the stone—is mainly visible due to the stone’s large size. Such an illusion was partially documented for star and cat’s-eye gemstones by Killingback (2005), who invoked the concept of stereoscopy to explain the effect. We report new observations of the phenomenon displayed by this garnet, as well as a star rose quartz, through stereoscopic images and anaglyphs. These show that the amplitude of the floating effect depends on the orientation of the star with respect to the observation plane. [ABSTRACT FROM AUTHOR]

Abstract

A large (112.64 ct) black star sapphire of probable Sri Lankan origin that exhibits 12-rayed asterism was studied for this report. As in other 12-rayed star sapphires, its phenomenal appearance is due to the presence of oriented needle-like inclusions that produce two superimposed six-rayed stars that are rotated 30° relative to one another. However, Raman spectroscopy combined with optical microscopy unexpectedly revealed that the inclusions responsible for both stars consist of the same mineral, ilmenite. By contrast, 12-rayed star sapphires from Thailand typically contain separate inclusion sets of rutile and hematite-ilmenite series minerals corresponding to the two six-rayed stars.

Abstract

International audience; Cultured pearls are human creations formed by inserting a nucleus and a small piece of mantle tissue into a living shelled mollusc, usually a pearl oyster. Although many pearl observations intuitively suggest a possible rotation of the nucleated pearl inside the oyster, no experimental demonstration of such a movement has ever been done. This can be explained by the difficulty of observation of such a phenomenon in the tissues of a living animal. To investigate this question of pearl rotation, a magnetometer system was specifically engineered to register magnetic field variations with magnetic sensors from movements of a magnetic nucleus inserted in the pearl oyster. We demonstrated that a continuous movement of the nucleus inside the oyster starts after a minimum of 40 days post-grafting and continues until the pearl harvest. We measured a mean angular speed of 1.27. min(-1) calculated for four different oysters. Rotation variability was observed among oysters and may be correlated to pearl shape and defects. Nature's ability to generate so amazingly complex structures like a pearl has delivered one of its secrets.

Abstract

SPE ECOLDUR GENOSOL IPMSPEECOLDURGENOSOLIPM; The growing need to survey the tremendous microbial diversity in a culture independent manner, has led to the development of molecular methods through sequence profiling of conserved genes such as 16S rDNA. Next-generation sequencing technologies are now used routinely to assess bacterial communities composition in complex environmental samples. Recently, the improvement of the Illumina MiSeq platform to a 2×300 bases paired-end version made it much more attractive for 16S rDNA amplicons sequencing enabling the analysis of a longer region thus facilitating taxonomic assignation. Additionally, the Illumina V3 chemistry enables high-throughput metagenomics analysis at the greatest coverage yet possible with a lower cost per sequence. Nevertheless, known limitations associated with the sequencing of low sequence diversity samples have hampered harnessing its true potential to sequence 16S rDNA gene amplicons. Moreover, validated Illumina 16S rDNA amplicon sequencing protocols do not support more than 96 samples per run, which underutilized the overall capacity of a sequencing run. We therefore worked on the development of a standardized and optimized high-multiplexed metagenomic solution for the exploration of complex environments. We developed an integrated solution combining several technical and bioinformatical strategies that are (i) a custom and standardized amplification protocol for 16S rDNA library preparation to minimize as full as possible technical biases; (ii) a custom 192-index strategy within a single run; (iii) the inclusion of a nucleotide sequence variability at the first sequencing cycles; (iv) an accurate paired-end reads assembly process for full length amplicon analysis and (v) a dedicated pipeline for taxonomic affiliation of 16S rDNA sequences and microbial communities analysis. Our solution validated within soil samples provides high sequencing accuracy and technical reproducibility with no index biases based on taxonomic distribution analysis. The association of technical and bioinformatical improvements yields substantial cost reductions and provides greater target flexibility to assess soil biodiversity as well as functions of microbial communities through carbon, phosphorus or nitrogen cycles in future developments.

Symbiotic associations between microorganisms and eukaryotes are common in nature. These microorganisms can play important role of the evolution and ecology of their hosts by modifying their phenotype like conferring protection against parasitic wasps. This symbiotic protection is encountered in the pea aphid, Acyrthosiphon pisum, where the bacterial symbiont Hamiltonella defensa confers protection against parasitoid wasp, Aphidius ervi. Despite this protective benefit, this symbiont occurs only at intermediate frequencies in natural populations. To identify factors influencing the frequency of H. defensa in natural population, we measured pea aphid life history traits and the protective effect of this symbiont. We found that protective symbiont doesn’t influence the host life history traits and the protection conferred by this symbiont varies according the biotype of pea aphid and the bacterial strain. However, the multiple infections with facultative symbionts reduce the fitness of the host while increasing protection against parasitoids. We also observed the positive correlation between aphid reproduction in the absence of parasitoid and the level of resistance: higher the symbiotic protection higher the aphid fecundity. This finding has important implications for the evolution of defensive symbiosis and allows evidence the need for better understanding of how these protective symbionts is maintained in the population.

Abstract

A majority of species interacts, closely and durably, with symbiotic microorganisms. These latter can modify the ecology and evolution of their hosts by inducing a variety of effects on their phenotype. However, the extended phenotype associated with a symbiotic organism can vary largely depending on the host species. For example, the symbiont Wolbachia can manipulate the reproduction of its host, protect it against natural enemies or enhance its nutrition. All these effects induced by Wolbachia, which can be deleterious or beneficial, depend on the host species. The aim of our study is to measure the variability of phenotypes that can be associated with a bacterial symbiont within a single host species. Our biological model is the pea aphid (Acyrthosiphon pisum). This particular aphid presents populations that have specialized on different legumes species (ie, biotypes). Among the symbiotic bacteria hosted by this insect, Hamiltonella defensa is highly prevalent in different biotypes but its phenotypic effects – altered behavior, reduced fitness and parasitic resistance - are known only in two of these biotypes. By analyzing the life history traits and the level of resistance of individuals carrying this symbiotic bacterium from four A. pisum biotypes, our study reveals that there is a high variability associated with this bacterium amongst inter and intrabiotype phenotypes. Therefore, a symbiotic microorganism/eukaryote relationship cannot be summarized by a single phenotypic effect.

International audience; Natural-color saltwater cultured pearls from Pinctada margaritifera were studied by diffuse reflectance UV-Vis-NIR spectroscopy to identify the absorption features associated with their various colors. Nine patterns observed in the visible range demonstrated that individual colors are caused not by one pigment but by a mixture of pigments.

Abstract

AphidBase is a centralized bioinformatic resource that was developed to facilitate community annotation of the pea aphid genome by the International Aphid Genomics Consortium (IAGC). The AphidBase Information System designed to organize and distribute genomic data and annotations for a large international community was constructed using open source software tools from the Generic Model Organism Database (GMOD). The system includes Apollo and GBrowse utilities as well as a wiki, blast search capabilities and a full text search engine. AphidBase strongly supported community cooperation and coordination in the curation of gene models during community annotation of the pea aphid genome. AphidBase can be accessed at http://www.aphidbase.com.

International audience; Genome annotation includes any features mapped on the genome, including protein coding genes and their multiples mRNA, pseudogenes, transposons and repeats, non-coding RNAs, SNPs, or similarities to other genomes. While many of these features can be automatically locally predicted by sophisticated software packages based on sequence or structure comparisons, setting up centralized a boinformatic warehouse is crucial to organize, display, and distribute all these data, and facilitates their handling by non skilled bio-analyst. It also provides the opportunity to unite a broad community for curating gene models and gathering manual and functional annotations, which is an essential step to attain a basal data quality. AphidBase (http://www.aphidbase.org) is a comprehensive information system dedicated to aphids. It has been set up to safely centralize, manage, mine, disseminate and promulgate data generated by International Aphid Genomics Consortium (IAGC). It allowed an international community dispersed at many sites to produce a robust and comprehensive annotation of the pea aphid genome. Lepido-DB (http://www.inra.fr/lepidodb) is a centralized bioinformatic resource that was first developed to facilitate the comparative genomics of two major lepidopteran pests, the noctuid moths Helicoverpa armigera and Spodoptera frugiperda, by the analyzis of syntenic relationships and genome rearrangements of 15 pairs of BACs sequences and their corresponding colinear regions extracted from 10 Bombyx mori chromosomes. These systems were constructed using open source software tools from the Generic Model Organism Database (GMOD, http://www.gmod.org) including several Chado instances and genome browsers (Gbrowse and Cmap), gene and protein reports, Apollo, an application for the manual curation, and various other tools such as a a blast search and a full text search facilities. At a a larger scope, ensuing the AphidBase and LepidoDB experience, this system will be extended to definitely support complete genomes sequencing or resequencing, annotation projects, or any other project based on deep sequencing strategies (expression profiling by RNA-Seq, variability studies, ChIP-Seq, ...) for various Insect pest species. And, to ensure that all these resources were fully exploited by the community, this expansion would be accompanied by gathering many new biological data such as phenotypes, ontologies and atlas.

Abstract

International audience; A new opal deposit has been recently discovered in the Welo province of Ethiopia. Unlike the opals previously found in the Shewa province, the new material is essentially white body color, resembling Australian opals. Also, in this new find, opal does not appear to form nodules. Otherwise, all its properties are consistent with volcanic opal-CT, as other Ethiopian opals.
