The machinery maintaining fecal continence prevents involuntary loss of stool and is based on the synchronized interplay of multiple voluntary and involuntary mechanisms, dependent on cooperation between motor responses of the musculature of the colon, pelvic floor, and anorectum, and sensory and motor neural pathways. Knowledge of the physiology of fecal continence is key toward understanding the pathophysiology of fecal incontinence. The idea that involuntary contraction of the internal anal sphincter is the primary mechanism of continence and that the external anal sphincter supports continence only by voluntary contraction is outdated. Other mechanisms have come to the forefront, and they have significantly changed viewpoints on the mechanisms of continence and incontinence. For instance, involuntary contractions of the external anal sphincter, the puborectal muscle, and the sphincter of O'Beirne have been proven to play a role in fecal continence. Also, retrograde propagating cyclic motor patterns in the sigmoid and rectum promote retrograde transit to prevent the continuous flow of content into the anal canal. "Asia" is not the end result but a means of intellectual exploration. "Asia" is multivalent; it is not self-sufficient and exclusionary vis-à-vis other cultures. It does not exist as an epistemological abstraction. This unique attribute of "Asia" is, however, where its opportunity lies. Taking "Asia" as a means for intellectual inquiry, this article explores the " fūdo " 風土of humankind and cultural formations in dialogue with historical circumstances. It argues that global integration is not the homogenization of disparate societies but mutual respect for their specificities. Furthermore, this article proposes a new kind of universality and reassesses how the specific relates to the universal. Taking Asia's historical experiences seriously, this article stresses that universality cannot act as an independent and superior imposition vis-à-vis specificities. Rather, specific experiences have to be put into an open dialogue between one another to unleash new possibilities. Background: Serine/threonine kinase is originally identified as an oncogene and the forkhead box transcription factor forkhead box O3a (Foxo3a) has been found to be decreased in various human cancers. In the present study, we explored the expression of Akt and FOXO3a in liver cancer cells.Materials and Methods: Akt level was detected by Western blotting analysis. Cell viability of HepG2, MHCC-97H, Bel7402, and L02 was determined by MTT assay. FoxO3a level was determined by Western blotting analysis.Results: Akt level was significantly higher in liver cancer cell lines HepG2 and MHCC97-H, compared with the immortalized liver cell line L02. MTT assay results demonstrated that LY294002 significantly suppressed cell proliferation of HepG2 and MHCC-97H cells in a dose- and time-dependent manner. The underlying molecular mechanism was that miR-370 inhibited cell proliferation of liver cancer cells by activating FoxO3a.Conclusion: PI3K inhibitor decreased the levels of phosphorylated FOXO3a and increased the levels of nuclear FOXO3a. Fever is a common clinical presentation of a number of diseases. A sustained unexplained fever >38.3°C lasting for >3 weeks without an established diagnosis despite intensive diagnostic evaluation is referred to as fever of unknown origin (FUO). FUO remains a clinical challenge for physicians, as it may be attributed to a wide range of disorders, mainly infections, malignancies, non-infectious inflammatory diseases and miscellaneous diseases. We herein report the case of a 59-year-old male patient who presented with prolonged unexplained fever and was found to have a diffusely enlarged hypermetabolic spleen, as shown on 18F-fluorodeoxyglucose positron emission tomography/computed tomography examination. Following splenectomy, histopathological examination revealed primary splenic lymphoma (PSL) of B-cell origin. The patient received 6 courses of systemic chemotherapy with rituximab, etoposide, cyclophosphamide, doxorubicin, vincristine and prednisone (R-ECHOP regimen) and responded well to treatment. Background: The excretory-secretory (ES) antigens of Trichinella spiralis muscle larvae (ML) are the most commonly used diagnostic antigens for trichinellosis. Their main disadvantage for the detection of anti-Trichinella IgG is false-negative results during the early stage of infection. Additionally, there is an obvious window between clinical symptoms and positive serology. Methods: ELISA with adult worm (AW) ES antigens was used to detect anti-Trichinella IgG in the sera of experimentally infected mice and patients with trichinellosis. The sensitivity and specificity were compared with ELISAs with AW crude antigens and ML ES antigens. Results: In mice infected with 100 ML, anti-Trichinella IgG were first detected by ELISA with the AW ES antigens, crude antigens and ML ES antigens 8, 12 and 12 days post-infection (dpi), respectively. In mice infected with 500 ML, specific antibodies were first detected by ELISA with the three antigen preparations at 10, 8 and 10 dpi, respectively. The sensitivity of the ELISA with the three antigen preparations for the detection of sera from patients with trichinellosis at 35 dpi was 100 %. However, when the patients' sera were collected at 19 dpi, the sensitivities of the ELISAs with the three antigen preparations were 100 % (20/20), 100 % (20/20) and 75 % (15/20), respectively (P < 0.05). The specificities of the ELISAs with the three antigen preparations were 98.11, 95.60 and 89.31 %, respectively (P< 0.05). Conclusions: The sensitivity and specificity of the T. spiralis AW ES antigens were superior to those of the AW crude antigens and ML ES antigens. A new species, Ginkgo huolinhensis sp. nov., with well-preserved cuticle from the Lower Cretaceous, Huolinhe Formation of Huolinhe Coal Field in Inner Mongolia, China, was studied morphologically and anatomically. The main characters of the new species are found in the leaves which are deeply dissected with narrow lobes, trichomes developed on both upper and lower epidermal cells, a media papilla developed on the periclinal walls for each cell in upper cuticle, and no trichomes on the cuticle of the petioles. These characters of G. huolinhensis sp. nov. distinguish from the known species of Ginkgo previously reported. This study of the new species is beneficial for the taxonomic research of fossil Ginkgo, and should be helpful for a better understanding the paleoenvironment of Ginkgo living in northeastern China during the Early Cretaceous. The existence of the Cretaceous-Tertiary (K/T) boundary in the non-marine succession is expected at Jiayin in the Heilongjiang River area, China. Zircons from a tuff sample from the Baishantou Member of Wuyun Formation in Jiayin were analyzed by the laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) U-Pb dating and fission-track dating methods. Ages of 64.1±0.7 Ma (U-Pb) and 61.7±1.8 Ma (fission-track dating) were obtained, which allow re-evaluation of a previously reported late Maastrichian age for the tuff layer that was in conflict with the paleontological evidence. The current molecular systematics of angiosperms recognizes the basal angiosperms and five major angiosperm lineages: the Chloranthaceae, the magnoliids, the monocots, Ceratophyllum and the eudicots, which consist of the basal eudicots and the core eudicots. The eudicots form the majority of the angiosperms in the world today. The flowering plants are of exceptional evolutionary interest because of their diversity of over 250,000 species and their abundance as the dominant vegetation in most terrestrial ecosystems, but little is known of their very early history. In this report we document an early presence of eudicots during the Early Cretaceous Period. Diagnostic characters of the eudicot fossil Leefructus gen. nov. include simple and deeply trilobate leaves clustered at the nodes in threes or fours, basal palinactinodromous primary venation, pinnate secondary venation, and a long axillary reproductive axis terminating in a flattened receptacle bearing five long, narrow pseudo-syncarpous carpels. These morphological characters suggest that its affinities are with the Ranunculaceae, a basal eudicot family. The fossil co-occurs with Archaefructus sinensis and Hyrcantha decussata whereas Archaefructus liaoningensis comes from more ancient sediments. Multiple radiometric dates of the Lower Cretaceous Yixian Formation place the bed yielding this fossil at 122.6-125.8 million years old. The earliest fossil records of eudicots are 127 to 125 million years old, on the basis of pollen. Thus, Leefructus gen. nov. suggests that the basal eudicots were already present and diverse by the latest Barremian and earliest Aptian. [ABSTRACT FROM AUTHOR]

The living fossil Ginkgo, a relict of a once dominant gymnosperm, existed and flourished early in the Mesozoic, but only a vague outline of its evolutionary history in the Tertiary has been unveiled. Here we describe a new species, Ginkgo jiayinensis sp. nov., from the Wuyun Formation of Jiayin, China, which is another well-established Tertiary species based on leaf fossils besides G. adiantoides. The most remarkable feature of the new species is the amphistomatic leaves, likely representing a distinct evolu- tionary line of the genus in this time interval. Ginkgo jiayinensis is similar to the co-occurring G. adiantoides and extant G. biloba in the lower cuticle, but the two latter species clearly differ in having hypostomatic leaves. Ginkgo biloba is used as the nearest living equivalent (NLE) species for both G. adiantoides atid G. jiayinensis in reconstructing paleo-CO2, as it has been used for other fossil species with either hypostomatic or amphistomatic leaves. The two Tertiary species are almost identical in stomatal index in abaxial cuticles (8.4 and 8.5, respectively), showing no strong differences in response to atmospheric CO2. Amphistomatic G. jiayinensis would therefore have used a different ecological strategy from that of G. adiantoides of the same bed. [ABSTRACT FROM AUTHOR]

We demonstrate that activation of nuclear factor κB (NF-κB) in neurons is neuroprotective in response to kainic acid (KA)-induced excitotoxicity. Combination of Western blotting, immunocytochemistry, and electrophoresis mobility shift assay showed that KA exposure induced a fast but transient nuclear translocation of the NF-κB p65 subunit and increased DNA-binding activity of NF-κB in primary cultured cortical neurons. The transient NF-κB activity was associated with upregulation of antiapoptotic Bcl-xL and XIAP gene products revealed by real-time PCR. Knockdown of p65 decreased neuronal viability and antiapoptotic gene expression. In addition, we showed that KA-stimulated DNA-binding activity of NF-κB was associated with reactive oxygen species and calcium signals, using AMPA/KA receptor antagonist, calcium chelator, and antioxidant. These results suggest that the fast and transient activation of NF-κB initiated by calcium signals is one of the important proximal events in response to KA-induced excitotoxicity, which has neuroprotective effect against KA-induced apoptosis. Assessment of long-term impacts of projected changes in climate, population, and land use and land cover on regional water resource is critical to the sustainable development of the southeastern United States. The objective of this study was to fully budget annual water availability for water supply (precipitation − evapotranspiration + groundwater supply + return flow) and demand from commercial, domestic, industrial, irrigation, livestock, mining, and thermoelectric uses. The Water Supply Stress Index and Water Supply Stress Index Ratio were developed to evaluate water stress conditions over time and across the 666 eight-digit Hydrologic Unit Code basins in the 13 southeastern states. Predictions from two Global Circulation Models (CGC1 and HadCM2Sul), one land use change model, and one human population model, were integrated to project future water supply stress in 2020. We found that population increase greatly stressed water supply in metropolitan areas located in the Piedmont region and Florida. Predicted land use and land cover changes will have little effect on water quantity and water supply-water demand relationship. In contrast, climate changes had the most pronounced effects on regional water supply and demand, especially in western Texas where water stress was historically highest in the study region. The simulation system developed by this study is useful for water resource planners to address water shortage problems such as those experienced during 2007 in the study region. Future studies should focus on refining the water supply term to include flow exchanges between watersheds and constraints of water quality and environmental flows to water availability for human use. Natural forests in southern China have been severely logged due to high human demand for timber, food, and fuels during the past century, but are recovering in the past decade. The objective of this study was to investigate how vegetation cover changes in composition and structure affected the water budgets of a 9.6-km2 Dakeng watershed located in a humid subtropical mountainous region in southern China. We analyzed 27 years (i.e., 1967-1993) of streamflow and climate data and associated vegetation cover change in the watershed. Land use/land cover census and Normalized Difference of Vegetation Index (NDVI) data derived from remote sensing were used to construct historic land cover change patterns. We found that over the period of record, annual streamflow ( Q) and runoff/precipitation ratio did not change significantly, nor did the climatic variables, including air temperature, Hamon’s potential evapotranspiration (ET), pan evaporation, sunshine hours, and radiation. However, annual ET estimated as the differences between P and Q showed a statistically significant increasing trend. Overall, the NDVI of the watershed had a significant increasing trend in the peak spring growing season. This study concluded that watershed ecosystem ET increased as the vegetation cover shifted from low stock forests to shrub and grasslands that had higher ET rates. A conceptual model was developed for the study watershed to describe the vegetation cover-streamflow relationships during a 50-year time frame. This paper highlighted the importance of eco-physiologically based studies in understanding transitory, nonstationary effects of deforestation or forestation on watershed water balances. The relationship between forests and streamflows has long been an important research interest in China. The purpose of this paper is to summarize progress and lessons learned from the forest-streamflow studies over the past four decades in China. To better measure the research gaps between China and other parts of the world, a brief global review on the findings from paired watershed studies over the past 100 years was also provided. In China, forest management shifted in the later 1990s from timber harvesting to forest restoration. Forest-streamflow research was accordingly changed from assessing harvesting impacts to evaluating both harvesting and forestation effects. Over the past four decades, Chinese forest hydrology research has grown substantially. Significant progress has been made on measuring individual processes, but little solid, long-term data were available to assess the relationship between forest changes and streamflows because of an absence of standard paired watersheds. In addition, misuse of statistical analyses was often found in the literature. A unique opportunity exists in China to study the forestation effects on streamflow as several large-scale forestation programs are being implemented. Such an opportunity should include a robust paired watershed design under an integrated watershed ecosystem framework to avoid repeating the lessons already learned. It is critical that evapotranspiration (ET) be quantified accurately so that scientists can evaluate the effects of land management and global change on water availability, streamflow, nutrient and sediment loading, and ecosystem productivity in watersheds. The objective of this study was to derive a new semi-empirical ET modeled using a dimension analysis method that could be used to estimate forest ET effectively at multiple temporal scales. The model developed describes ET as a function of water availability for evaporation and transpiration, potential ET demand, air humidity, and land surface characteristics. The model was tested with long-term hydrometeorological data from five research sites with distinct forest hydrology in the United States and China. Averaged simulation error for daily ET was within 0.5 mm/day. The annual ET at each of the five study sites were within 7% of measured values. Results suggest that the model can accurately capture the temporal dynamics of ET in forest ecosystems at daily, monthly, and annual scales. The model is climate-driven and is sensitive to topography and vegetation characteristics and thus has potential to be used to examine the compounding hydrologic responses to land cover and climate changes at multiple temporal scales. Aim: To investigate bilirubin UDP-glucuronosyltransferase ( UGT1A1) gene allele in healthy Chinese neonates, their cord bilirubin level and the subsequent hyperbilirubinemia to determine relationships among them. Methods: Cord blood of 48 neonates was obtained to determine the exon 1 of UGT1A1 gene, total serum bilirubin, albumin, glutamic-pyruvic transaminase (GPT), glutamic-oxalacetic transaminase (GOT) and haemoglobin (Hb) concentration. Neonatal jaundice was assessed by measurement of transcutaneous bilirubin (TCB) and serum bilirubin. Neonates were divided into two groups according to mutant or normal allele to compare the variables. Results: Nineteen infants had the nucleotide 211 G→A allele, 3 had the heterozygous variation (686C→A, 845 A→T, 231G→A). In the 211 A allele group, cord bilirubin was significantly higher than in the 211 G allele group (p = 0.034), but there were no differences in albumin (p = 0.678), GPT (p = 0.460), GOT (p = 0.440) and Hb (p = 0.886). The TCB (at 48, 96 h), the frequency of the hyperbilirubinemia and prolonged jaundice were also significantly higher in the 211 A allele group. Conclusions: The UGT1A1 gene codon G71R allele is a risk factor for neonatal hyperbilirubinemia in the Chinese population. The continuing study of early angiosperms from the Yixian Formation (≈125 Ma) of northeastern China has yielded a second early angiosperm genus. This report is a detailed account of this early flowering plant and recognizes earlier reports of similar fossils from Russia and China. Entire plants, including roots, stems, and branches terminating in fruits are presented and reconstructed. Evidence for a possible aquatic nature of this plant is presented. The relationship of Hyrcantha (‘Sinocarpus’) to the eudicots is discussed. The presence of this second early angiosperm genus, now known as a whole plant, is important in the discussion of its systematics and the ecology of the earliest angiosperms. [ABSTRACT FROM AUTHOR]

Moreover, sustainable afforestation practice is considered for developing forest vegetation.

Most heavy crude oils underwent biodegradation and generated a significant amount of naphthenic acids. Naphthenic acids are polar compounds with the carboxylic group and are considered as a major factor affecting the oil viscosity. However, the relationship between the molecular composition of naphthenic acids and oil viscosity is not well understood. This study examined a "clean" heavy oil with low contents of heteroatoms but had a high content of naphthenic acids. Naphthenic acids were fractionated by distillation and caustic extraction. The molecular composition was characterized by highresolution Orbitrap mass spectrometry. It was found that the 2- and 3-ring naphthenic monoacids with 15e35 carbon atoms are dominant components of the acid fractions; the caustic extraction is capable of isolating naphthenic acids with less than 35 carbons, which is equivalent to the upper limit of the distillable components, but not those in the residue fraction; the total acid number of the heavy distillates is higher than that of the residue fraction; the viscosity of the distillation fraction increases exponentially with an increased boiling point of the distillates. Blending experiments indicates that there is a strong correlation between the oil viscosity and acids content, although the acid content is only a few percent of the total oil. As the theoretical upper bound of evapotranspiration (ET) or water use by ecosystems, potential ET (PET) has always been widely used as a variable linking a variety of disciplines, such as climatology, ecology, hydrology, and agronomy. However, substantial uncertainties exist in the current PET methods (e.g., empiric models and single-layer models) and datasets, because of unrealistic configurations of land surface and unreasonable parameterizations. Therefore, this study comprehensively considered interspecific differences in various vegetation-related parameters (e.g., plant stomatal resistance and CO2 effects on stomatal resistance) to calibrate and parametrize the Shuttleworth-Wallace (SW) model for forests, shrubland, grassland and cropland. We derived the parameters using identified daily ET observations with no water stress (i.e., PET) at 96 eddy covariance (EC) sites across the globe. Model validations suggest that the calibrated model could be transferable from known observations to any location. Based on four popular meteorological datasets, relatively realistic canopy height and time-varying land use/land cover and Leaf Area Index, we generated a global 5 km ensemble mean monthly PET dataset that includes two components of potential transpiration (PT) and soil evaporation (PE) for the 1982-2015 time period. Using this new dataset, the climatological characteristics of PET partitioning and the spatio-temporal changes in PET, PE and PT were investigated. The global mean annual PET was 1200 mm with PT/PET of 40 % and PE/PET of 60 %, and moreover controlled by PT and PE over 43 % and 57 % of the globe, respectively. Globally, the annual PET and PT significantly (p<0.05) increases by 1.25 mm/yr and 1.22 mm/yr over the last 34 years, followed by a slight increase in the annual PE. Overall, the annual PET changes over 53 % of the globe could be attributed to PT, and the rest to PE. The new PET dataset may be used by academic communities and various agencies to conduct climatological analyses, hydrological modelling, drought studies, agricultural water management, and biodiversity conservation. USP14 deubiquitinates ERα to maintain its stability in ECEndometrial cancer (EC) is one of the common gynecological malignancies of which the incidence has been rising for decades. It is considered that continuously unopposed estrogen exposure is the main risk factor for EC initiation. Thus, exploring the modulation of estrogen/estrogen receptor α (ERα) signaling pathway in EC would be helpful to well understand the mechanism of EC development and find the potential target for EC therapy. Ubiquitin-specific peptidase 14 (USP14), a member of the proteasome-associated deubiquitinating enzyme family, plays a crucial role in a series of tumors. However, the function of USP14 in EC is still elusive. Here, our results have demonstrated that USP14 is highly expressed in EC tissues compared with that in normal endometrial tissues, and higher expression of USP14 is positively correlated with poor prognosis. Moreover, USP14 maintains ERα stability through its deubiquitination activity. Our results further demonstrate that USP14 depletion decreases the expression of ERα-regulated genes in EC-derived cell lines. Moreover, knockdown of USP14 or USP14-specific inhibitor treatment significantly suppresses cell growth and migration in EC cell lines or in mice. We further provide the evidence to show that the effect of USP14 on EC cell growth, if not all, at least is partially related to ERα pathway. This study investigated monthly and annual reference evapotranspiration ETo changes over southwestern China (SWC) from 1960 to 2012, using the Food and Agriculture Organization of the United Nations' report 56 (FAO-56) Penman-Monteith equation and routine meteorological observations at 269 weather sites. During 1960-2012, the monthly and annual ETo decreased at most sites. Moreover, the SWC regional average trend in annual ETo was significantly negative (p < 0.05); this trend was the same in most months. A new separation method using several numerical experiments was proposed to quantify each driving factor's contribution to ETo changes and exhibited higher accuracy based on several validation criteria, after which an attribution analysis was performed. Across SWC, the declining annual ETo was mainly due to decreased net radiation (RN). Spatially, the annual ETo changes at most sites in eastern SWC (excluding southeastern West Guangxi) were generally due to RN, whereas wind speed (WND) or vapor pressure deficit (VPD) was the determinant at other sites. Nevertheless, the determinants differed among 12 months. For the whole SWC, increased VPD in February and decreased WND in April, May, and October were the determinant of decreased ETo; however, decreased RN was the determinant in other months. Overall, the determinant of the monthly ETo changes exhibited a complex spatial pattern. A complete analysis of ETo changes and the related physical mechanisms in SWC is necessary to better understand hydroclimatological extremes (e.g., droughts) and to develop appropriate strategies to sustain regional development (e.g., water resources and agriculture). Importantly, this separation method provides new perspective for quantitative attribution analyses and thus may be implemented in various scientific fields (e.g., climatology and hydrology). Background: To investigate the relation between serum leptin levels and cerebral infarction (CI) by meta-analysis. Materials and Methods: Scientific literature databases were searched for studies published in Chinese and English. After retrieving relevant articles through database searches and screening using predefined selection criteria, high-quality studies related to our research topic were selected for inclusion in this meta-analysis. All statistical analyses were conducted using Comprehensive Meta-Analysis 2.0 (CMA 2.0, Biostat Inc., Englewood, New Jersey, USA). Results: The study results revealed that serum leptin levels were significantly higher in CI patients as compared to normal controls. The outcomes of subgroup analysis by ethnicity suggested that the serum leptin levels in CI patients were significantly higher than normal controls in both Asian and Caucasian populations. Further, subgroup analysis based on the detection method indicated that the serum leptin levels in CI patients were significantly higher compared with normal controls when measured by radioimmunoassay (RIA) but enzyme-linked immunosorbent assay (ELISA)-based measurements did not show such statistically significant differences. Conclusion: Our meta-analysis results suggest that serum leptin levels in CI patients may be closely correlated with CI risks. [ABSTRACT FROM AUTHOR]

Background: We have previously reported that a 31 kDa protein was screened from the excretory-secretory (ES) proteins of Tichinella spiralis muscle larvae (ML) by immunoproteomics using early infection sera, and the gene encoding a 31 kDa protein from T. spiralis was cloned and expressed in an E. coli expression system. In this study, the recombinant 31 kDa antigens were used for detection of anti-Trichinella antibodies in serum of experimentally infected mice by ELISA. Methods: Anti-Trichinella IgG antibodies in sera of mice infected with Trichinella were assayed by ELISA with recombinant 31 kDa antigens, and its sensitivity and specificity were compared with ELISA with ES antigen. Results: The sensitivity and specificity of ELISA with recombinant antigens was 96.67% (29/30) and 96.87% (62/64), compared with 100% (30/30) and 98.44% (63/64) of ELISA with ES antigens was (P>0.05). In heavily, moderately and lightly infected mice (500, 300 and 100 larvae/mouse), anti-Trichinella antibodies were firstly detected by ELISA with recombinant antigens at 8, 12 and 14 dpi, respectively; then increased rapidly with a detection rate of 100% respectively at 28, 22 and 30 dpi. While the antibodies were firstly detected by ELISA with ES antigens at 10, 8 and 10 dpi, respectively, the antibody positive rate reached 100% at 14, 12 and 22 dpi, respectively. Conclusion: The recombinant 31 kDa antigens of T. spirali had a good sensitivity and specificity for detecting anti-Trichinella antibodies and might be the potential diagnostic antigen for trichinellosis. Understanding the changes in hydrology of coastal forested wetlands induced by climate change is fundamental for developing strategies to sustain their functions and services. This study examined 60 years of climatic observations and 30 years of hydrological data, collected at the Santee Experimental Forest (SEF) in coastal South Carolina. We also applied a physically-based, distributed hydrological model (MIKE SHE) to better understand the hydrological responses to the observed climate variability. The results from both observation and simulation for the paired forested watershed systems indicated that the forest hydrology was highly susceptible to change due to climate change. The stream flow and water table depth was substantially altered with a change in precipitation. Both flow and water table level decreased with a rise in temperature. The results also showed that hurricanes substantially influenced the forest hydrological patterns for a short time period (several years) as a result of forest damage. [ABSTRACT FROM AUTHOR]

The PRMS_Storm model was built as a storm event, distributed hydrological model for studying the hydrological effects of forest composition and spatial distribution on storm-flow volume and peakflow rates in the Xiangshuixi Watershed in the Three Gorges Reservoir Area, in the Yangtze River Basin in southwestern China. We developed three simulation scenarios based on forest composition and their spatial arrangements across the watershed, including all mixed conifer-evergreen broadleaf forests (Scenario 1), all mixed evergreen broadleaf forests (Scenario 2), and mixed conifer + evergreen broadleaf + shrub forests (Scenario 3). We examined 11 storm events observed during 2002-2005. Compared with the existing forest covers, modeling results suggested that the amount of overland flow was reduced by 21, 23, and 22%, and the interflow increased by 16, 88, and 30%, for Scenarios 1, 2, and 3, respectively. During the same time, peakflow rates were reduced by 20.8, 9.6, and 18.9%, respectively. The reduction of peakflow rates was most significant when rainfall intensity exceeded 0.8 mm/min and events with a short duration and effect was minor when rainfall intensity was below 0.5 mm/min. In general, we found that Scenarios 1 and 3 were preferred for reducing storm-flow volume and peakflow rates due to their higher interception rates, large soil water holding capacity, and higher soil infiltration capacity. The modeled results suggested soil properties are important in affecting the flow processes and thus forest composition and forest spatial distributions will affect storm-flow volume and peakflow rates at the watershed scale. To maximize flood reduction functions of a watershed, high priority should be given to those forest types (Scenarios 1 and 3) in reforestation practices in the study region. The Loess Plateau region in northwestern China has experienced severe water resource shortages due to the combined impacts of climate and land use changes and water resource exploitation during the past decades. This study was designed to examine the impacts of climatic variability on streamflow characteristics of a 12-km2 watershed near Tianshui City, Gansu Province in northwestern China. Statistic analytical methods including Kendall’s trend test and stepwise regression were used to detect trends in relationship between observed streamflow and climatic variables. Sensitivity analysis based on an evapotranspiration model was used to detect quantitative hydrologic sensitivity to climatic variability. We found that precipitation ( P), potential evapotranspiration (PET) and streamflow ( Q) were not statistically significantly different ( p > 0.05) over the study period between 1982 and 2003. Stepwise regression and sensitivity analysis all indicated that P was more influential than PET in affecting annual streamflow, but the similar relationship existed at the monthly scale. The sensitivity of streamflow response to variations of P and PET increased slightly with the increase in watershed dryness (PET/ P) as well as the increase in runoff ratio ( Q/ P). This study concluded that future changes in climate, precipitation in particular, will significantly impact water resources in the Loess Plateau region an area that is already experiencing a decreasing trend in water yield. Quantifying the hydrologic responses to land use/land cover change and climate variability is essential for integrated sustainable watershed management in water limited regions such as the Loess Plateau in Northwestern China where an adaptive watershed management approach is being implemented. Traditional empirical modeling approach to quantifying the accumulated hydrologic effects of watershed management is limited due to its complex nature of soil and water conservation practices (e.g., biological, structural, and agricultural measures) in the region. Therefore, the objective of this study was to evaluate the ability of the distributed hydrologic model, MIKE SHE to simulate basin runoff. Streamflow data measured from an overland flow-dominant watershed (12 km2) in northwestern China were used for model evaluation. Model calibration and validation suggested that the model could capture the dominant runoff process of the small watershed. We found that the physically based model required calibration at appropriate scales and estimated model parameters were influenced by both temporal and spatial scales of input data. We concluded that the model was useful for understanding the rainfall-runoff mechanisms. However, more measured data with higher temporal resolution are needed to further test the model for regional applications. [ABSTRACT FROM AUTHOR]

This study reports the linkage between MIKE SHE and Wetland-DNDC for carbon dynamics and greenhouse gases (GHGs) emissions simulation in forested wetland.Wetland-DNDC was modified by parameterizing management measures, refining anaerobic biogeochemical processes, and was linked to the hydrological model – MIKE SHE. As a preliminary application, we simulated the effect of water table position and forest management practices on GHGs emissions and carbon dynamics to test the capabilities of the models for simulating seasonal and long-term carbon budget. Simulation results show that water table changes had a remarkable effect on GHGs fluxes. Anaerobic conditions in forested wetland soils reduce organic matter decomposition and stimulate CH4 production. Decrease in the water table from the wetland surface decreases methane flux, while CO2 emission was lower with a rise in the water table. When there is a drop in water availability, wetlands can become a net source of atmospheric CO2 as photosynthesis is decreased and respiration loss enhanced. Forest management activities i.e. harvest, fertilization and reforestation practices were parameterized in the model. We predicted carbon fluxes and stores on a pine forest under different forest management scenarios during 160 years. Results show that average long-term carbon storage in ecosystem pools increased with increasing rotation length; Soil carbon showed only minor, long-term responses to harvesting events. In contrast, carbon sequestered in tree biomass and litter fluctuated widely, in concert with the harvest cycle. Application of nitrogen fertilizer increased average carbon storage in all ecosystem pools and wood products. We presented the linkage of MIKE SHE and Wetland-DNDC as a way to use of simulation modeling tools for assessing GHGs mitigation strategies, carbon budgeting and forest management. Background: Glutathione-S-transferase (GST) is a widespread multigene family of detoxification enzymes. The vaccination of mice with recombinant GST of 24 kDa from Trichinella spiralis elicited a low immune protection against challenge infection. The objective of this study was to characterize the T. spiralis putative GST gene (TspGST) encoding a 30.8 kDa protein and to evaluate its potential as a candidate antigen for anti-Trichinella vaccine. Methods: The full-length cDNA sequence of TspGST from T. spiralis muscle larvae (ML) was expressed in E. coli. The enzymatic activity and antigenicity of the rTspGST were identified by spectrophotometry, Western blot, and ELISA. The expression of TspGST at T. spiralis various stages was investigated by RT-PCR and indirect immunofluorescent test (IIFT). Serum level of total IgG, IgG1, and IgG2a antibodies against rTspGST were measured by ELISA. The immune protection produced by vaccination with rTspGST against T. spiralis was evaluated. Results: The sequencing results showed that the cDNA of TspGST was 840 bp, and encoded a protein of 279 amino acids, which had a molecular size of 30.8 kDa and a pI of 5.21. Its amino acid sequence shares 37% similarity with TsGST. The rTspGST protein had enzymatic activity of GST. On Western blot and ELISA analysis, the native TspGST protein with 30. 8 kDa in crude antigens derived from adult worms (AW), newborn larvae (NBL), infective intestinal larvae (IIL) and ML was recognized by anti-rTspGST sera, but the ML ES antigens could be not recognized by anti-rTspGST sera. Expression of TspGST was found in all of T. spiralis various stages (AW, NBL, ML, and IIL). An immunolocalization analysis identified TspGST in different stages (mainly in cuticles) of the nematode. The mice vaccinated with the rTspGST elicited Th2-predominant immune responses, showed a 34.38% reduction of adult worms and a 43.70% reduction of muscle larvae. Conclusions: Immunization with rTspGST produced a partial immune protection, and the rTspGST could be regarded as a potential candidate target for an anti-Trichinella vaccine. [ABSTRACT FROM AUTHOR]

The early Cambrian problematica Xianguangia sinica, Chengjiangopenna wangii, and Galeaplumosus abilus from the Chengjiang biota (Yunnan, China) have caused much controversy in the past and their phylogenetic placements remain unresolved. Here we show, based on exceptionally preserved material (85 new specimens plus type material), that specimens previously assigned to these three species are in fact parts of the same organism and propose that C. wangii and G. abilus are junior synonyms of X. sinica. Our reconstruction of the complete animal reveals an extinct body plan that combines the characteristics of the three described species and is distinct from all known fossil and living taxa. This animal resembled a cnidarian polyp in overall morphology and having a gastric cavity partitioned by septum-like structures. However, it possessed an additional body cavity within its holdfast, an anchoring pit on the basal disk, and feather-like tentacles with densely ciliated pinnules arranged in an alternating pattern, indicating that it was a suspension feeder rather than a predatory actiniarian. Phylogenetic analyses using Bayesian inference and maximum parsimony suggest that X. sinica is a stem-group cnidarian. This relationship implies that the last common ancestor of X. sinica and crown cnidarians was probably a benthic, polypoid animal with a partitioned gastric cavity and a single mouth/anus opening. This extinct body plan suggests that feeding strategies of stem cnidarians may have been drastically different from that of their crown relatives, which are almost exclusively predators, and reveals that the morphological disparity of total-group Cnidaria is greater than previously assumed. Stable isotope ratios (δ13C and δ18O) of tree-ring α-cellulose are important tools in paleoclimatology, ecology, plant physiology and genetics. The Multiple Sample Isolation System for Solids (MSISS) was a major advance in the tree-ring α-cellulose extraction methods, offering greater throughput and reduced labor input compared to traditional alternatives. However, the usability of the method for resinous conifer species may be limited by the need to remove extractives from some conifer species in a separate pretreatment step. Here we test the necessity of pretreatment for α-cellulose extraction in loblolly pine (Pinus taeda L.), and the efficiency of a modified acetone-based ambient-temperature step for the removal of extractives (i) in loblolly pine from five geographic locations representing its natural range in the southeastern USA, and (ii) on five other common coniferous species (black spruce (Picea mariana Mill.), Fraser fir (Abies fraseri (Pursh) Poir.), Douglas fir (Pseudotsuga menziesii (Mirb.) Franco), Norway spruce (Picea abies (L.) Karst) and ponderosa pine (Pinus ponderosa D.)) with contrasting extractive profiles. The differences of δ13C values between the new and traditional pretreatment methods were within the precision of the isotope ratio mass spectrometry method used (±0.2‰), and the differences between δ18O values were not statistically significant. Although some unanticipated results were observed in Fraser fir, the new ambient-temperature technique was deemed as effective as the more labor-consuming and toxic traditional pretreatment protocol. The proposed technique requires a separate acetone-inert multiport system similar to MSISS, and the execution of both pretreatment and main extraction steps allows for simultaneous treatment of up to several hundred microsamples from resinous softwood, while the need of additional labor input remains minimal. Background: The intraperitoneal injection is a common method for establishing the experimental animal model infected with parasites. The aim of this study was to investigate if the intraperitoneal injection was another route of Trichinella spiralis infection. Methods: From June to July 2015, twenty BALB/c mice were intraperitoneally injected with 300 T. spiralis muscle larvae in Department of Parasitology, Medical College, Zhengzhou University, China. The larvae per gr (LPG) muscle from the infected mice and the reproductive capacity index (RCI) of T. spiralis were calculated Results: Sixty percent (12/20) mice injected were successfully infected at 35 day post injection (dpi), but the muscle larval burden (381.53 larvae per gr) and reproductive capacity index (32.33) in infected mice was lower. Conclusion: A mechanical force indicated as a possible mechanism in successful larval invasion of almost all kind of host tissues. Objective: Draining of the chest cavity with two chest tubes after pulmonary lobectomy is a common practice. The objective of this study was to evaluate whether using two tubes after a pulmonary lobectomy is more effective than using a single tube. Patients and Methods: We performed a meta-analysis of five randomized studies that compared the single chest tube with double chest tube application after pulmonary lobectomy. The primary end-point was amount of drainage and duration of chest tube drainage. The secondary end-points were the patient's numbers of new drain insertion after operation, hospital stay after operation, the patient's numbers of subcutaneous emphysema after operation, the patient's numbers of residual pleural air space, pain score, the number of patients who need thoracentesis, and cost. Results: Five randomized controlled trials totaling 502 patients were included. Meta-analysis results are as follows: There were statistically significant differences in amount of drainage (risk ratio [RR] = -0.15; 95% confidence interval [CI] = -3.17, -0.12, P = 0. 03), duration of chest tube drainage (RR = -0.43; 95% CI = -0.57, -0.19, P = 0.02), pain score (P < 0.05). Compared with patients receiving the double chest tube group, there were no statistically significant differences between the two groups with regard to the patient's numbers of new drain insertion after operation. Conclusion: Compared with the double chest tube, the single chest tube significantly decreases amount of drainage, duration of chest tube drainage, pain score, the number of patients who need thoracentesis, and cost. Although there is convincing evidence to confirm the results mentioned herein, they still need to be confirmed by large-sample, multicenter, randomized, controlled trials. [ABSTRACT FROM AUTHOR]

Accurate detection and quantification of vegetation dynamics and drivers of observed climatic and anthropogenic change in space and time is fundamental for our understanding of the atmosphere-biosphere interactions at local and global scales. This case study examined the coupled spatial patterns of vegetation dynamics and climatic variabilities during the past three decades in the Upper Heihe River Basin (UHRB), a complex multiple use watershed in arid northwestern China. We apply empirical orthogonal function (EOF) and singular value decomposition (SVD) analysis to isolate and identify the spatial patterns of satellite-derived leaf area index (LAI) and their close relationship with the variability of an aridity index (AI = Precipitation/Potential Evapotranspiration). Results show that UHRB has become increasingly warm and wet during the past three decades. In general, the rise of air temperature and precipitation had a positive impact on mean LAI at the annual scale. At the monthly scale, LAI variations had a lagged response to climate. Two major coupled spatial change patterns explained 29% and 41% of the LAI dynamics during 1983-2000 and 2001-2010, respectively. The strongest connections between climate and LAI were found in the southwest part of the basin prior to 2000, but they shifted towards the north central area afterwards, suggesting that the sensitivity of LAI to climate varied over time, and that human disturbances might play an important role in altering LAI patterns. At the basin level, the positive effects of regional climate warming and precipitation increase as well as local ecological restoration efforts overwhelmed the negative effects of overgrazing. The study results offer insights about the coupled effects of climatic variability and grazing on ecosystem structure and functions at a watershed scale. Background Previous study showed that Trichinella spiralis proteasome subunit beta type-7 (Tspst) gene is an up-regulated gene in intestinal infective larvae (IIL) compared to muscle larvae (ML), which was screened by using suppression subtractive hybridization (SSH) and confirmed by real-time PCR. Tspst may be related to the larval invasion of intestinal epithelial cells (IECs). The aim of this study was to identify Tspst and to investigate its immune protection against intestinal T. spiralis infection. Methods The Tspst gene encoding a 29 kDa protein from T. spiralis infective larvae was cloned, and recombinant Tspst protein (rTspst) was produced in an Escherichia coli expression system. The rTspst was used to immunize BALB/c mice. Anti-rTspst antibodies were used to determine the immunolocolization of Tspst in the parasite. Transcription and expression of Tspst at T. spiralis different developmental stages were observed by RT-PCR and immunofluorescence test (IFT). The in vitro or in vivo immune protection of anti-rTspst serum or rTspst against intestinal T. spiralis infection in BALB/c mice was evaluated. Results Anti-rTspst serum recognized the native Tspst protein with 29 kDa in ML crude antigens. Transcription and expression of gene was observed at all T. spiralis different developmental stages (IIL, adult worms, newborn larvae, and ML). An immunolocalization analysis identified Tspst in the cuticle and internal organs of the parasite. An in vitro invasion assay showed that, when anti-rTspst serum, serum of mice infected with T. spiralis or normal mouse serum were added to the medium, the invasion rate of the infective larvae in an IEC monolayer was 25.2%, 11.4%, and 79%, respectively (P < 0.05), indicating that anti-rTspst serum partially prevented the larval invasion of IECs. After a challenge infection with T. spiralis muscle larvae, mice immunized with rTspst conferred a 45.7% reduction in adult worm burden in intestines. Conclusions In the present study, Tspst was first identified and characterized. Tspst is an invasion-related protein of T. spiralis IIL and could be considered as a potential vaccine candidate antigen against intestinal T. spiralis infection that merits further study. The exchange of carbon dioxide is a key measure of ecosystem metabolism and a critical intersection between the terrestrial biosphere and the Earth's climate. Despite the general agreement that the terrestrial ecosystems in North America provide a sizeable carbon sink, the size and distribution of the sink remain uncertain. We use a data-driven approach to upscale eddy covariance flux observations from towers to the continental scale by integrating flux observations, meteorology, stand age, aboveground biomass, and a proxy for canopy nitrogen concentrations from AmeriFlux and Fluxnet-Canada Research Network as well as a variety of satellite data streams from the MODIS sensors. We then use the resulting gridded flux estimates from March 2000 to December 2012 to assess the magnitude, distribution, and interannual variability of carbon fluxes for the U.S. and Canada. The mean annual gross primary productivity (GPP), ecosystem respiration (ER), and net ecosystem productivity (NEP) of the U.S. over the period 2001-2012 were 6.84, 5.31, and 1.10 Pg C yr-1, respectively; the mean annual GPP, ER, and NEP of Canada over the same 12-year period were 3.91, 3.26, and 0.60 Pg C yr-1, respectively. The mean nationwide annual NEP of natural ecosystems over the period 2001-2012 was 0.53 Pg C yr-1 for the U.S. and 0.49 Pg C yr-1 for the conterminous U.S. Our estimate of the carbon sink for the conterminous U.S. was almost identical with the estimate of the First State of the Carbon Cycle Report (SOCCR). The carbon fluxes exhibited relatively large interannual variability over the study period. The main sources of the interannual variability in carbon fluxes included drought and disturbance. The annual GPP and NEP were strongly related to annual evapotranspiration (ET) for both the U.S. and Canada, showing that the carbon and water cycles were closely coupled. As one of the key tools for regulating human-ecosystem relations, environmental conservation policies can promote ecological rehabilitation across a variety of spatiotemporal scales. However, quantifying the ecological effects of such policies at the regional level is difficult. A case study was conducted at the regional level in the ecologically vulnerable region of the Loess Plateau, China, through the use of several methods including the Universal Soil Loss Equation (USLE), hydrological modeling and multivariate analysis. An assessment of the changes over the period of 2000-2008 in four key ecosystem services was undertaken to determine the effects of the Chinese government's ecological rehabilitation initiatives implemented in 1999. These ecosystem services included water regulation, soil conservation, carbon sequestration and grain production. Significant conversions of farmland to woodland and grassland were found to have resulted in enhanced soil conservation and carbon sequestration, but decreased regional water yield under a warming and drying climate trend. The total grain production increased in spite of a significant decline in farmland acreage. These trends have been attributed to the strong socioeconomic incentives embedded in the ecological rehabilitation policy. Although some positive policy results have been achieved over the last decade, large uncertainty remains regarding long-term policy effects on the sustainability of ecological rehabilitation performance and ecosystem service enhancement. To reduce such uncertainty, this study calls for an adaptive management approach to regional ecological rehabilitation policy to be adopted, with a focus on the dynamic interactions between people and their environments in a changing world. •Hydraulic redistribution (HR) of water via roots from moist to drier portions of the soil occurs in many ecosystems, potentially influencing both water use and carbon assimilation. •By measuring soil water content, sap flow and eddy covariance, we investigated the temporal variability of HR in a loblolly pine ( Pinus taeda) plantation during months of normal and below-normal precipitation, and examined its effects on tree transpiration, ecosystem water use and carbon exchange. •The occurrence of HR was explained by courses of reverse flow through roots. As the drought progressed, HR maintained soil moisture above 0.15 cm3 cm−3 and increased transpiration by 30–50%. HR accounted for 15–25% of measured total site water depletion seasonally, peaking at 1.05 mm d−1. The understory species depended on water redistributed by the deep-rooted overstory pine trees for their early summer water supply. Modeling carbon flux showed that in the absence of HR, gross ecosystem productivity and net ecosystem exchange could be reduced by 750 and 400 g C m−2 yr−1, respectively. •Hydraulic redistribution mitigated the effects of soil drying on understory and stand evapotranspiration and had important implications for net primary productivity by maintaining this whole ecosystem as a carbon sink. The genus Czekanowskia is an important member of the Mesozoic flora and is widespread in the Northern Hemisphere. In China, it is found mostly in Late Triassic and Early and Middle Jurassic sediments of North China. With the exception of a few specimens, all the reports of this genus from China have been based on gross leaf morphology. Abundant Czekanowskia leaves, preserved as compression fossils, were collected from the Middle Jurassic, Zhaogou Formation of Inner Mongolia, China. In this article, two subgenera of Czekanowskia, Harrisella and Vachrameevia, are recognized from Inner Mongolia, China, on the basis of leaf morphology and epidermal characters. Two new species, Czekanowskia chinensis sp. nov. (subg. Harrisella) and Czekanowskia shiguaiensis sp. nov. (subg. Vachrameevia), are described. The occurrence of these species in the Middle Jurassic of Inner Mongolia significantly extends the stratigraphic and geographic distribution of Czekanowskia in the Northern Hemisphere. Analysis of the epidermal characters of material presented here along with consideration of other associated plants of the flora and sedimentology of the plant-bearing strata suggests that the two species described in this report grew in an area with a warm and humid climate in a warm temperate zone. The study examined the relationships between whole tree hydraulic conductance ( Ktree) and the conductance in roots ( Kroot) and leaves ( Kleaf) in loblolly pine trees. In addition, the role of seasonal variations in Kroot and Kleaf in mediating stomatal control of transpiration and its response to vapour pressure deficit ( D) as soil-dried was studied. Compared to trunk and branches, roots and leaves had the highest loss of conductivity and contributed to more than 75% of the total tree hydraulic resistance. Drought altered the partitioning of the resistance between roots and leaves. As soil moisture dropped below 50%, relative extractable water (REW), Kroot declined faster than Kleaf. Although Ktree depended on soil moisture, its dynamics was tempered by the elongation of current-year needles that significantly increased Kleaf when REW was below 50%. After accounting for the effect of D on gs, the seasonal decline in Ktree caused a 35% decrease in gs and in its sensitivity to D, responses that were mainly driven by Kleaf under high REW and by Kroot under low REW. We conclude that not only water stress but also leaf phenology affects the coordination between Ktree and gs and the acclimation of trees to changing environmental conditions. [ABSTRACT FROM AUTHOR]

During the Mesozoic era, Ginkgo leaves are diverse and widespread, especially in the Northern Hemisphere. Here we present three Ginkgo leaf types collected from the Middle Jurassic Zhaogou Formation of Shiguai coalbearing basin in Inner Mongolia, China. These are described for the first time on the basis of their leaf morphology and well-preserved cuticular characters. One new species, Ginkgo shiguaiensis, is described, and Ginkgo longifolius is recognized, while a third leaf type is designated as Ginkgo sp. The occurrence of these leaf types in the Middle Jurassic of Inner Mongolia significantly extends the stratigraphic and geographic distribution of Ginkgo in China. The associated plants, cuticular characters of the Ginkgo leaves, and the sedimentology of the plant-bearing strata suggest that these plants grew in a warm-temperate and moist climate. Abstract: Native copper mineralization in northeastern Yunnan is related to the Emeishan flood basalts, which have high Cu background concentrations. The deposits are located in the Ludian, Maoling and Yiche synclines between the flood basalts and the overlying Xuanwei Formation. Evidence from REE and HSFE concentrations, oxygen isotope compositions, bitumen reflectance, and mineral paragenesis indicates that the high-grade copper orebodies were deposited from moderate to low temperature hydrothermal solutions (420 to 100°C) in a reducing environment. 40Ar/39Ar spectra for the laumontites that coexist with native copper in the high-grade ores yield consistent plateau and isochron ages (226 to 228Ma). These are interpreted to represent the age of the first stage of hydrothermal mineralization, which took place at temperatures of 200 to350°C, concordant with that of Zn–Pb–Ag–Ge mineralization also occurring in the area. The heulandites yield a stable 40Ar/39Ar plateau age of 134.0±1.7Ma, consistent with isochron ages indicated by U–Th–Pb isotopic systematics, indicating a second stage of native copper mineralization at lower temperatures (100 to 200°C) that took place during the Early Cretaceous.
