The antioxidant properties and bioactive compound contents of fresh new tomato hybrids before and after in vitro digestion were investigated. To this aim, the antioxidant activities of lipophilic, hydrophilic and polyphenolic extracts of tomato hybrids were determined by ABTS (2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)), DMPD (N,N-dimethyl-p-phenylenediamine dihydrochloride) and DPPH (2,2-diphenyl-1-picrylhydrazyl) methods respectively, while the bioactive compound contents were estimated via Folin-Ciocalteu (polyphenols), pH differential (anthocyanins) and high-performance liquid chromatography (lycopene and β-carotene) methods.; Results: After the digestion process, a marked loss (ranging from 37 to 77%) of antioxidant capacity linked to the hydrophilic fraction was observed. In contrast, the lipophilic and methanolic fractions showed an increase in antioxidant activity (ranging from 9 to 40%) after gastric digestion, and a rapid decrease was observed after total digestion. Moreover, the presence of anthocyanins and carotenoids after simulated digestion was a notable result.; Conclusion: The bioavailability of bioactive metabolites from nutraceutical food and their healthful properties in humans are strictly dependent on the digestion process. © 2017 Society of Chemical Industry.; © 2017 Society of Chemical Industry.

The development of antimicrobial active packaging constitutes a powerful tool to reduce waste and increase quality standards of perishable goods. Among numerous available antimicrobial agents, essential oils stand out for their renowned efficiency, and their use is beneficial due to their sustainability compared to other oil-based antimicrobials. In this work, we report on the use of photo-responsive nanocapsules containing thyme essential oil as functional coatings for polyethylene and polylactic acid films to obtain antimicrobial active packaging. Polymer surface activation treatment enhanced compatibility with nanocapsules solution. The films were analyzed to assess the structural and functional properties of the coating, evaluate morphological changes due to their photo-responsive behavior, and monitor the light-induced release of volatile thyme oil. It was found that 24 h after a 15-min UV exposure of the coated films, the concentration of thyme oil in the headspace was eight times higher with respect to un-irradiated films, thus confirming the efficiency of the light-triggered release system. Therefore, the manufactured films are proposed as on-demand release devices for application in non-contact antimicrobial active packaging.

Extremophiles represent a heterogeneous class of organisms that inhabit environments characterized by atypical parameters (high or low temperatures/pH/salinity, etc.). These organisms not only adapt to these habitats, but the exceptional environmental characteristics ensure their optimal growth. The environmental adaptation of extremophiles has been of particular interest for researcher. In particular, special attention has been addressed to the potential use of extreme biomolecules for industrial applications, because of their unique resistance properties, which represent a clear advantage for the development of industrial processes. Quorum sensing (QS) is a kind of molecular communication system that regulates gene expression in a range of organisms. It regulates many important microbial processes, such as motility, protease expression, antibiotic and exopolysaccharide production, and so on. It could contribute to environmental adaptation of extremophiles, but its role needs to be further elucidated. This chapter presents the recent advances in the understanding of QS in extremophilic microorganisms.

Silver nanoparticles hold great promise as effective anti-microbial compounds in a myriad of applications but may also pose a threat to non-target bacteria and fungi in the environment. Because microorganisms are involved in extensive interactions with many other organisms, these partner species are also prone to indirect negative effects from silver nanoparticles. Here, we focus on the effects of nanosilver exposure in the rhizosphere. Specifically, we evaluate the effect of 100 mg kg−1 silver nanoparticles on maize plants, as well as on the bacteria and fungi in the plant's rhizosphere and the surrounding bulk soil. Maize biomass measurements, microbial community fingerprints, an indicator of microbial enzymatic activity, and carbon use diversity profiles are used. Hereby, it is shown that 100 mg kg−1 silver nanoparticles in soil increases maize biomass, and that this effect coincides with significant alterations of the bacterial communities in the rhizosphere. The bacterial community in nanosilver exposed rhizosphere shows less enzymatic activity and significantly altered carbon use and community composition profiles. Fungal communities are less affected by silver nanoparticles, as their composition is only slightly modified by nanosilver exposure. In addition, the microbial changes noted in the rhizosphere were significantly different from those noted in the bulk soil, indicated by different nanosilver-induced alterations of carbon use and community composition profiles in bulk and rhizosphere soil. Overall, microorganisms in the rhizosphere seem to play an important role when evaluating the fate and effects of silver nanoparticle exposure in soil, and not only is the nanosilver response different for bacteria and fungi, but also for bulk and rhizosphere soil. Consequently, assessment of microbial populations should be considered an essential parameter when investigating the impacts of nanoparticle exposure.

This chapter will discuss some remarkable examples of extremophilic bacteria, isolated from different ecosystems, which produce exopolysaccharides (EPSs). The chapter will also cover the properties of EPSs produced by extremophiles and their possible commercial applications ranging from pharmaceutical to food processing, detoxification and bioremediation. This chapter will also cover the techniques that are used to purify, analyse and structurally characterise the bacterial EPSs; the state of the art in the field of bacterial EPSs research, with mention to the main examples of well-studied and commercially exploited EPSs; an overview of the main EPSs’ producing extremophiles that have been isolated from both aquatic and terrestrial environments; the description of the main biosynthetic routes leading to the EPSs production in archaeal and bacterial extremophiles, with reference to the enzymes involved and to the genetic manipulations for biosynthesis’s tailoring; the analysis of the most interesting biological properties of extremophiles’ EPSs (that can act as anti-inflammatory, immunomodulating and antiviral agents) and of their biotechnological applications in drug delivery systems; and the compendium of the most used techniques to perform the purification, the analysis of chemical composition, the identification of glycoside linkage position and substitution pattern, the determination of the molecular-weight distribution and the structural analysis of EPSs.

Microbial EPSs are ubiquitous in the extreme environments where they are crucial for microbial survival. Most of the functions attributed to EPSs are related to a protective role, which are dependent on the ecological niches in which the producer microorganisms live. They could support the microbial communities to tolerate extremes of temperature, salt concentration and nutrient supply, building an interface between the bacterial cell and its surrounding environment. Several EPSs produced by microorganisms from extreme habitats show biotechnological promise. By examining their structure and chemical/physical characteristics, it is possible to gain insight into their commercial application; they are employed in several fields ranging from food-processing to pharmaceutical industries, through to the bioremediation ability of polluted areas. Considering that most of extreme ecosystems and, therefore, the respective microbial communities are still unexplored, it is reasonable to hypothesize that the isolation of new microorganisms, together with their biomolecules, in particular exopolysaccharides, will provide interesting perspectives for new industrial processes in several fields.

The antioxidant and cytotoxic activities and the polyphenolic and anthocyanin contents of tomato hybrid powders were studied. Tomato powders were obtained, starting from the fresh fruits that had undergone an industrial process of drying and pulverization at two different temperatures. Antioxidant activities were evaluated in different extracts by using spectrophotometric assays: 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid and N,N-dimethyl-p-phenylenediamine dihydrochloride cation radical inhibition for lipophilic and hydrophilic extracts, respectively, and 2,2-diphenyl-1-picrylhydrazyl radical scavenging assay for polyphenolic extracts. Moreover, polyphenolic and anthocyanin contents were also carried out to detect the presence of these bioactive compounds. The effect of cytotoxic activity in vitro of tomato hybrid powder extracts on T47D (human breast carcinoma) cells was also evaluated. Results showed good antioxidant activities in lipophilic, polyphenolic, and hydrophilic extracts of samples that were obtained at a lower temperature. Extracts of the sample obtained at a higher temperature presented moderate antioxidant activity, lower than the extracts of other samples, which was probably due to the loss of labile antioxidant compounds during the industrial process. Very interesting was the presence of anthocyanins in both samples, even if in traces, and also a moderate cytotoxicity of a lipophilic extract on T47D cells. Therefore, tomato hybrid powders, on the basis of their multifunctional properties, could have a biotechnological application in agri-food or cosmetic industries as an additive for improving nutritional and/or bioactive qualities of commercial products used in daily nutrition and cosmetics.

To increase the knowledge about the biological properties of tomatoes, the antioxidant and antiproliferative activities of extracts of nine cultivars of Lycopersicon esculentum, as well as their chemical composition, were studied. The highest antioxidant capacity has been revealed in San Marzano Cirio 3 and Pomodoro Giallo cultivars, both in hydrophilic and lipophilic fractions, while San Marzano, Corbarino di Corbara, and Pomodoro Giallo exhibited the best radical scavenger activity in methanolic fraction. As regards the antiproliferative activity, the median inhibition concentrations of the lipophilic extracts ranged from 272.3 (Pomodoro Giallo) to 364.4 (Corbarino di Corbara) mg dried extract L-1 on Hep-G2 and from 324.6 (San Marzano Cirio 3) to 455.4 (Nero di Sicilia) on Caco-2. The methanolic extracts were more active on Caco-2 than Hep-G2, while the hydrophilic extracts were not active. These biological properties could be ascribed to the identified carotenoids and phenolic acids as well as to a pull of minor compounds exerting their synergistic effect.

The objective of the present work was to establish the antioxidative ability linked to lipophilic, hydrophilic, and polyphenolic fractions of new tomato hybrids of industrial interest, grown in an outdoor field, named "Medugno", situated in the Agro-Nocerino Sarnese area (Province of Salerno, Campania Region, Italy). Antioxidant activities of lipophilic, hydrophilic, and polyphenolic extracts of tomato hybrids determined by the 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid), N,N-dimethyl-p-phenylenediamine dihydrochloride, and 2,2-diphenyl-1-picrylhydrazyl methods, respectively, showed the best results in hybrids obtained between the pure San Marzano and Black Tomato lines. Antioxidant activity tests, performed also on the San Marzano × Black Tomato hybrid (pulped tomatoes), indicated that the industrial transformation process of this new tomato hybrid did not cause a significant loss of antioxidant activity. The in vitro production of nitrite by lipopolysaccharide-stimulated macrophages J774A.1 performed on lipophilic extracts showed that only two hybrids (San Marzano × Black Tomato and Marmande × Black Tomato) inhibited, in a concentration-related manner, nitric oxide release. Results suggested that genotypic factors could determine the nutritional quality of tomato because of the content of biologically active compounds and their biosynthesis. Moreover, the new tomato hybrid achieved could have a potential for the agri-food industry because of its nutritional quality and because it lends itself in processes of industrial transformation.

The generic term "quorum sensing" has been adopted to describe the bacterial cell-to-cell communication mechanism which coordinates gene expression when the population has reached a high cell density. Quorum sensing depends on the synthesis of small molecules that diffuse in and out of bacterial cells. There are few reports about this mechanism in Archaea. We report the isolation and chemical characterization of small molecules belonging to class of diketopiperazines (DKPs) in Haloterrigena hispanica, an extremely halophilic archaeon. One of the DKPs isolated, the compound cyclo-(L-prolyl-L-valine) activated N-acyl homoserine lactone (AHL) bioreporters, indicating that Archaea may have the ability to interact with AHL-producing bacteria within mixed communities.

Sea environments harbor a wide variety of life forms that have adapted to live in hard and sometimes extreme conditions. Among the marine living organisms, extremophiles represent a group of microorganisms that attract increasing interest in relation to their ability to produce an array of molecules that enable them to thrive in almost every marine environment. Extremophiles can be found in virtually every extreme environment on Earth, since they can tolerate very harsh environmental conditions in terms of temperature, pH, pressure, radiation, etc. Marine extremophiles are the focus of growing interest in relation to their ability to produce biotechnologically useful enzymes, the so-called extremozymes. Thanks to their resistance to temperature, pH, salt, and pollutants, marine extremozymes are promising biocatalysts for new and sustainable industrial processes, thus representing an opportunity for several biotechnological applications. Since the marine microbioma, i.e., the complex of microorganisms living in sea environments, is still largely unexplored finding new species is a central issue for green biotechnology. Here we described the main marine environments where extremophiles can be found, some existing or potential biotechnological applications of marine extremozymes for biofuels production and bioremediation, and some possible approaches for the search of new biotechnologically useful species from marine environments.

Quorum sensing (QS) mechanisms regulate a variety of vital functions such as motility, growth inhibition, biofilm development, virulence expression and plasmid conjugation. Many of these activities are essential for the well-being of organisms, in particular in extreme conditions such as hypersaline environments. Halophiles are able to synthesize products that are stable, and to exploit their activities under such “extreme” conditions, namely saline environments such as marine habitat, salt lakes, brines and saline soils. The versatility of these products leads researchers to address their interest in biotechnological applications. In this study, the QS activity of Halomonas smyrnensis AAD6—a moderately halophilic and exopolysaccharide-producing bacterium—was investigated. Dichloromethane extracts from stationary phase cultures of Halomonas smyrnensis AAD6 were partially purified, and C16-AHL was identified by LC-MS analysis. The synthesis of autoinducers was assayed and their production was detected starting from 48 h of growth. The growth-phase dependent production of exopolysaccharide (EPS) and signal molecules suggested that these activities could be linked.

The 3.94-Mb draft genome of Acinetobacter calcoaceticus GK1, a hydrocarbonoclastic plant growth-promoting Gram-negative rhizospheric bacterium, is presented here. Isolated at the Ford Motor Company site in Genk, Belgium, from poplar trees planted on a diesel-contaminated plume, GK1 is useful for enhancing hydrocarbon phytoremediation.

Biological activities of different varieties of tomato seed extracts were evaluated to verify the potential antioxidant and/or antiproliferative activity of the bioactive metabolites present in them. Findings demonstrated that among all the varieties investigated (San Marzano Rosso, San Marzano Giallo, Corbarino, Black Tomato and San Marzano/Black Tomato hybrid) San Marzano Rosso seed extract exhibited the highest free radical-scavenging activity with 68% of 2,2-Diphenyl-1-picrylhydrazyl radical inhibition, and the best cytotoxic activity evaluated by using the brine shrimp test (LD50: 23,198 ppm) and 3-(4,5-dimethylthiazol-2-yl)-2,5-phenyl-2H-tetrazolium bromide assay on A375 cell line (IC50: 137.7 μg/mL).

Four bacterial strains belonging to the genera Vibrio, Pseudoalteromonas and Photobacterium were isolated from the marine sponges Dysidea avara and Geodia cynodium. A Bacillus strain was isolated from Ircinia variabilis. A screening of molecules involved in quorum sensing (QS) was carried out by TLC-overlay and a new “plate T-streak” test. To analyze quorum quenching (QQ), a plate T-streak was performed with Chromobacterium violaceum. Strains of Vibrio isolated from both marine sponges and a strain of Photobacterium isolated from G. cynodium, activated QS bioreporters. A strain of Pseudoalteromonas isolated from D. avara showed QQ activity. Finally, it is reported that cyclic dipeptides isolated from strains of Vibrio sp. and Bacillus sp. (isolated from D. avara and I. variabilis, respectively) were involved in the QS mechanism. The simultaneous presence of bacteria that showed contrasting responses in bioassays for QS signal molecule synthesis in marine sponges could add an interesting dimension to the signalling interactions which may be happening in sponges.

Extreme environments, generally characterized by atypical temperatures, pH, pressure, salinity, toxicity, and radiation levels, are inhabited by various microorganisms specifically adapted to these particular conditions, called extremophiles. Among these, the microorganisms belonging to the Archaea domain are of significant biotechnological importance as their biopolymers possess unique properties that offer insights into their biology and evolution. Particular attention has been devoted to two main types of biopolymers produced by such peculiar microorganisms, that is, the extracellular polysaccharides (EPSs), considered as a protection against desiccation and predation, and the endocellular polyhydroxyalkanoates (PHAs) that provide an internal reserve of carbon and energy. Here, we report the composition, biosynthesis, and production of EPSs and PHAs by different archaeal species.

Background Conventional agriculture relies on chemical pesticides and fertilizers, which can degrade ecosystems. A reduction of these harmful practices is required, replacing (or integrating) them with more eco-friendly approaches, such as microbial inoculation. Tomato is an important agricultural product, with a high content of bioactive compounds (folate, ascorbate, polyphenols, and carotenoids). The focus of this research was to investigate the plant growth-promoting (PGP) abilities of bacterial strains isolated from different tomato cultivars, with the aim to develop systems to improve plant health and crop productivity based on microbial inoculation. Methods A pool of different tomato cultivars already available on the market and new tomato hybrids were selected based on their nutritional quality (high content of biologically active compounds). A total of 23 strains were isolated from tomato roots (11 rhizospheric strains and 12 root endophytes). The cultivable isolates were analyzed for a number of different PGP traits: organic acids (OA), indole acetic acid (IAA), ACC deaminase, and siderophore production. The effects of microbial inoculation on root growth of Arabidopsis thaliana were also evaluated using a Vertical Agar Plate assay. Results A high percentage of the isolated strains tested positive for the following PGP traits: 73 % were able to produce OA, 89 % IAA, 83 % ACC deaminase, and 87 % siderophores. The most striking result were remarkable increases in the formation of root hairs for most of the inoculated plants. This effect was obvious for all A. thaliana seedlings inoculated with the isolated endophytes, and for the 50 % of the seedlings inoculated with the rhizospheric strains. Conclusions A better knowledge of the plant growth-promotion activity of these strains can provide an important contribution to increase environmental sustainability in agriculture.
