The emerald ash borer (EAB), Agrilus planipennis (Fairmaire), is the most destructive invasive insect species of ash (Fraxinus spp.) in North America. An accurate method for early detection of this noxious insect pest is indispensable to providing adequate warning of A. planipennis infestation. A loop-mediated isothermal amplification (LAMP) assay (EAB-LAMP) was developed based on mitochondrial cytochrome c oxidase subunit I (COI) gene. The EAB-LAMP required only 30 min at 65°C to amplify A. planipennis DNA from specimens collected from geographically distinct locations. There was no cross-reactivity with other Agrilus and insect species. The developed EAB-LAMP differentially detected traces of A. planipennis genome (COI) within frass from various Fraxinus species. EAB-LAMP was also able to distinguish among A. planipennis DNA and other Agrilus species and nontarget insect species in trap captures. By detecting A. planipennis DNA in two additional trap captures (in situ), the EAB-LAMP was more sensitive and reliable than visual inspection. We tested the quantitative nature of the assay by evaluating pooled trap samples and demonstrated that the EAB-LAMP was capable of functioning optimally using a pool size of at least five individual trap samples. This potentially circumvents the need to perform large-scale individual analysis for processing trap samples. Considering its performance, specificity, sensitivity, and repeatability, the developed EAB-LAMP could be a valuable tool to support strategy and operation of large-scale surveillance for A. planipennis and could profitably be used in routine monitoring programs for effective management of A. planipennis.

Annual monitoring of mortality agents in the course of a spruce budworm (Choristoneura fumiferana (Clemens) (Lepidoptera: Tortricidae)) population cycle is essential to understanding the factors governing the rise and collapse of outbreaks. To date, assessments of causes of budworm mortality have relied on laboratory rearing of field-collected larvae, followed by visual identification of emerging parasitoids and/or microscopic analysis of pathogens in larval carcasses. Although this approach has provided vital information on the abundance and identity of mortality agents, the procedure is labor-intensive and has limits in terms of accuracy. To overcome these shortcomings, we developed a molecular identification tool that makes use of real-time quantitative PCR (qPCR) and TaqMan&reg, technologies. The tool relies on taxon-specific molecular variants (single nucleotide polymorphism [SNP] markers) found in mitochondrial (COI) and nuclear (28S rDNA) genes, for parasitoids, and in the nuclear SSU rDNA gene for microsporidian pathogens, these are then used as molecular signatures targeted by qPCR primers and TaqMan probes. Thus, the design of several sets of primers and probes deployed in multiplex format enables the identification of natural enemies via a molecular sorting process, bypassing barcode sequencing. Crude budworm DNA extracts are processed through a first module that detects dipteran and hymenopteran parasitoids, and microsporidian infections. Positive samples are then processed for species determination using three additional modules, enabling the identification of 20 common natural enemies of the spruce budworm. The tool has been fully validated using DNA samples from all comprised taxa, and both its sensitivity and accuracy compared favorably with the rearing-based method in an analysis of field-collected budworms. Using this tool, sample processing can be completed within two days, does not require larval rearing, provides accurate species identification, and can be conducted by technical staff without extensive molecular biology or insect taxonomy training.

Diapause is an important strategy for certain insect species to survive unfavorable environmental conditions, including low temperatures experienced when they overwinter in cold climate. Many studies have indicated that the increased expression of heat shock proteins during diapause improves the thermal tolerance of insects. However, the relationship between small heat shock proteins (sHSPs) and diapause is not clear or well-researched. In this study, we investigated the transcript levels of 14 sHSP genes in the spruce budworm, Choristoneura fumiferana, a major pest of spruce and fir in Canada, during pre-diapause, diapause, and post-diapause under normal rearing conditions and in response to a heat shock treatment. We found that sHSP expression profiles could be classified into five patterns under normal laboratory conditions: pattern I was upregulated only during pre-diapause, pattern II was upregulated only during diapause, pattern III was constantly expressed throughout diapause, pattern IV was upregulated in both pre-diapause and diapause, and pattern V was upregulated only during post-diapause. After heat shock, five different expression patterns were observed: pattern I responded weakly or not at all throughout diapause, pattern II responded weakly during the diapause stage but strongly at the onset of diapause and in the post-diapause period, pattern III was upregulated only during post-diapause, pattern IV was strongest during diapause, and pattern V was strongest only in early diapause. These complex expression profiles lead us to suggest that most of the sHSP genes are involved in the diapause process and that they may have multiple and important roles in different phases of this process.

International audience; We evaluated the use of fluorescent powders for tracking dispersal by the emerald ash borer, Agrilus planipennis Fairmaire (Coleoptera: Buprestidae), of Beauveria bassiana isolates from an autocontamination device. Neither of the two DayGlow powders tested (Arc Yellow and Aurora Pink) interfered with fungal germination or growth, nor did they affect survival of beetles in the laboratory, or affect virulence of the fungus. The powders persisted at least 10 days out-of-doors on dead beetles in sticky band traps, and at least 14 days on pouches inside autocontamination traps. During field trials of autocontamination traps with powder-dusted fungal pouches in southwestern Ontario, 8.0% of the 4010 beetles captured in green prism and sticky-band traps were positive for fluorescent powders. Only half (46.2–57.8%) of the powder-positive beetles actually carried viable fungal conidia, as determined by plating of beetle rinses, possibly as a result of patchy growth of fungal isolates and reduced conidia production on pouch surfaces during the 16-day trapping experiment. The presence of viable conidia (either one or both isolates) on about 10% of beetles that did not carry any visible powder particles may be an indication of horizontal transmission of the fungus by beetles that had visited the autocontamination traps.

A microsporidium Nosema disstriae (Thomson) is a parasite of the forest tent caterpillar Malacasoma disstria (Lepidoptera: Lasiocampidae), a notable defoliator of deciduous trees in North America. The goal of this paper was to demonstrate the ultrastructure of N . disstriae and to determine the position of this microsporidium within the N . bombycis clade (NBC) using comparative morphology and multiple molecular phylogenetic markers: RPB1, LSU-, ITS- and SSU-rDNA. As a part of this goal, the revision of the described members of the NBC has been performed. The ultrastructure of proliferating stages and spores of N . disstriae were similar to previously described Nosema spp. parasitizing lepidopteran species. Meronts produced tubular-like structures on their surfaces and exhibited a tight association with host mitochondria. All stages were diplokaryotic and developed without interfacial envelopes. Disporoblastic sporogony produced typical Nosema -type spores with 9–12 polar filament coils. A vesicle with immature spores was once recognized on sections, concordant with the previous record of octosporous sporogony in the N . disstriae life cycle. Rarely, spores with thinner envelopes and large posterior vacuoles were seen in the midgut. Tracheae were most heavily infected. Midgut, surrounding muscles, haemocytes and fat body also contained microsporidia. SSUrRNA-inferred phylogenies were consistent with previously published articles and did not resolve the relation within the NBC clade. The RPB1-inferred trees and concatenated RPB1 and LSU-ITS-SSUrDNA-based trees demonstrated clustering of N . disstriae with N . antheraeae as early divergent species within the NBC.

Entomopathogenic fungi of the genera Isaria and Purpureocillium were recovered from infestation sites of emerald ash borer (EAB) in Southern Ontario, Canada. Isolates were identified using morphological characters and by sequencing the ITS1-5.8S-ITS2 ribosomal DNA gene and partial β-tubulin gene. Phylogenetic analysis and constructed trees based on the ITS and β-tubulin gene explicitly confirm isolates L66B, SY17-a and LHY46-a as Isaria farinosa and B3A, B59A and SY45B-a as Purpureocillium lilacinum. Pathogenicity was tested in the laboratory against adult EAB using a single concentration (2.0×107 conidia/ml) applied topically to adults. Controls included three commercial isolates: Isaria fumosorosea LRC176, Metarhizium brunneum LRC187 and Beauveria bassiana strain GHA. Nosema isolates from five lepidopteran forest defoliators, Nosema fumiferanae from spruce budworm, Choristoneura fumiferana; a Nosema sp. from jack pine budworm, Choristoneura pinus pinus and western spruce budworm, Choristoneura occidentalis (Nosema sp. CPP and Nosema sp. CO, respectively); Nosema thomsoni from large aspen tortrix, Choristoneura conflictana; and Nosema disstriae, from the forest tent caterpillar, Malacosoma disstria were compared based on their small subunit (SSU) ribosomal RNA (rRNA) gene sequences. Four of the species sequenced, N. fumiferanae, Nosema sp. CPP, Nosema sp. CO, and N. disstriae have a high SSU rDNA sequence identity (0.6%-1.5%) and are members of the "true Nosema" clade. They all showed the reverse arrangement of the (large subunit [LSU]-internal transcribed spacer [ITS]-SSU) of the rRNA gene. The fifth species, N. thomsoni has the usual (SSU-ITS-LSU) arrangement and is not a member of this clade showing only an 82% sequence similarity. We speculate, therefore, that a genetic reversal may have occurred in the common ancestor to the "true Nosema" clade. Although, the mechanism for rearrangement of the rRNA gene subunits is not known we provide a possible explanation for the localization. N. fumiferanae, Nosema sp. CPP, and Nosema sp. CO clustered together on the inferred phylogenetic tree. The high sequence similarities, the reverse arrangement in the rRNA gene subunits, and the phylogenetic clustering suggest that these three species are closely related but separate species.

We examined isolates from 4 commercial bioinsecticides based on different strains of Bacillus thuringiensis subspecies ( kurstaki , israelensis , aizawai , and tenebrionis ) for the presence of genes encoding proteins with known enterotoxigenicity (nhe, hbl, cytk, ces) and various other putative virulence genes (piplc, sph, bceT, entFM, entS, entT). The piplc and bceT sequences were present in all the isolates; sph was found in aizawai and israelensis; entFM only in israelensis; and entS in kurstaki, israelensis, and tenebrionis. Our results corroborate previous findings that isolates used in commercial products contain all nhe and hbl component genes but not the ces gene. We ascertained that the cytK gene present in the kurstaki-, israelensis-, and aizawai-based products belongs to the cytK-2 type and not the more toxigenic cytK-1 variant originally isolated from enterotoxic Bacillus cereus. We provide the first evidence that hemolytic (hblA) and nonhemolytic (nheA, nheB, nheC) enterotoxin genes are expressed during septicemia in a target insect. This opens the door for their possible participation in pathogenesis in target insects. If enterotoxins do not contribute to bacterial pathogenesis in target insects, their genes could be deleted from commercial production strains to pre-empt perceptions of public health risks.

The house fly, Musca domestica L., and the stable fly, Stomoxys calcitrans (L.) (Diptera: Muscidae), are cosmopolitan pests parasitized by a guild of more than two dozen species of wasps. Several species of these wasps have been commercialized as biocontrol agents or are being studied for this purpose. Wolbachia bacteria are known to infect at least some of these wasps and are of interest because infections can dramatically affect insect reproduction. A survey in this parasitoid-fly system detected Wolbachia in 15 of 21 species of wasps and in three of nine species of flies parasitized by these wasps. Phylogenetic analyses using wsp gene sequences identified single isolate infections in most cases. Infections of two and four isolates were detected in Nasonia vitripennis (Walker) and Spalangia cameroni Perkins (Hymenoptera: Pteromalidae), respectively. Laboratory experiments showed infections in S. cameroni to cause an incomplete form of female-mortality (FM) type cytoplasmic incompatibility (CI). Crosses between uninfected female and infected male partners (♀ x ♂ w ) produced fewer progeny, which had a strong male-biased sex ratio. Crosses between 9 x ♂,♀ w x ♂ w , and ♀ w x ♂ produced more progeny, which had a female-biased sex ratio. Developmental times of progeny were increased when the paternal parent was infected with Wolbachia, regardless of whether the maternal parent was infected or whether offspring developed from fertilized eggs. This result may reflect the action of Wolbachia on components of the seminal fluid that then affect the development of offspring from inseminated females. It is hoped that future studies of Wolbachia in this guild will facilitate the rearing and application of these wasps as biocontrol agents of house fly and stable fly.

Wolbachia bacteria are an unsuspected, but potentially important, component of many biocontrol programs. To heighten awareness of these bacteria, we review current knowledge of Wolbachia and their possible application in biocontrol research. Wolbachia promote their spread by altering the reproductive success of their arthropod hosts. This ability frequently is identified as having potential either to reduce populations of pest species, or to increase populations of beneficial species. However, only 19 and 1% of peer-reviewed research articles (n=844) on Wolbachia appear in arthropod- and biocontrol-specific journals, respectively. Although Wolbachia will not have application for all programs, their prevalence cannot be denied. We screened for Wolbachia in populations of arthropods of current interest to biocontrol programs in Canada. Wolbachia are intracellular bacteria that occur in an estimated 20% of arthropod species. They are of broad interest because they profoundly affect the reproductive fitness of diverse host taxa. Here we document the apparent ubiquity and diversity of Wolbachia in the insect orders Anoplura (sucking lice) and Mallophaga (chewing lice), by detecting single or multiple infections in each of 25 tested populations of lice, representing 19 species from 15 genera spanning eight taxonomic families. Phylogenetic analyses indicate a high diversity of Wolbachia in lice, as evidenced by the identification of 39 unique strains. Some of these strains are apparently unique to lice, whereas others are similar to strains that infect other insect taxa. Wolbachia are transmitted from infected females to their offspring via egg cytoplasm, such that similar species of lice are predicted to have similar strains of Wolbachia. This predicted pattern is not supported in the current study and may reflect multiple events of recent horizontal transmission between host species. At present, there is no known mechanism that would allow for this latter mode of transmission to and within species of lice.

Emerald ash borer is an invasive species from Asia. Beauveria bassiana strain L49-1AA is being tested for the control of emerald ash borer in Canada, using an autocontamination trapping system. We have developed a simplified allele discrimination polymerase chain reaction (PCR) assay to screen B. bassiana strain, L49-1AA from other Beauveria species by targeting the inter-strain genetic differences in 5' end of EF1-α gene of the genus Beauveria. A single nucleotide polymorphism (SNP) site, T→C was identified only in L49-1AA and was used to develop a simplified allele discrimination polymerase chain reaction (PCR) assay based on a modified allelic inhibition of displacement activity (AIDA) approach for distinguishing B. bassiana L49-1AA from all background Beauveria isolates. The SNP site was employed to design inner primers but with a deliberate mismatch introduced at the 3' antepenultimate from the mutation site in order to maximize specificity and detection efficiency. Amplification was specific to L49-1AA without cross-reaction with DNA from other Beauveria strains. In addition, the designed primers were also tested against environmental samples in L49-1AA released plots and observed to be highly efficient in detecting and discriminating the target strain, L49-1AA from both pure and crude DNA samples. This new method can potentially allow for more discriminatory tracking and monitoring of released L49-1AA in our autocontamination and dissemination projects for managing EAB populations. Additionally, the modified-AIDA format has potential as a tool for simultaneously identifying and differentiating closely related Beauveria species, strains/isolates as well as general classification of other pathogens or organisms.

We provide molecular systematics of a microporidian species, Nosema fumiferanae, one of the most common natural enemies of spruce budworm, Choristoneura fumiferana. The uncharacterized flanking region upstream of the large subunit (LSU) rRNA and the complete rRNA cistron of N. fumiferanae was 4,769 bp long. The organization of the rRNA gene was 5′-LSU rRNA-ITS-SSU rRNA-IGS-5S-3′ and corresponded primarily to most insect (i.e. lepidopteran) Nosema species identified and classified to date. Phylogenetic analysis based on the complete rRNA cistron indicated that N. fumiferanae is closely related to Nosema plutellae and is correctly assigned to the “true” Nosema group. Suggestions were provided on a criterion to delineate the “true” Nosema from other microsporidian species.

A microsporidium was isolated from the bronze birch borer, Agrilus anxius Gory (Coleoptera: Buprestidae), collected near Sudbury and Sault Ste Marie, Canada. Light and electron microscopic investigations showed that gross pathology and ultrastructure of the investigated Cystosporogenes species was similar to those characterized and described for other Cystosporogenes species. Small subunit rRNA gene sequence data and comparative phylogenetic analysis confirmed that the microsporidian species from A. anxius is most closely related to the genus Cystosporogenes clade of microsporidia. Infection average in the Sudbury and Sault Ste Marie beetle populations was >80% and relatively stable in 2006-2007 but declined in 2008. Field prevalence of the A. anxius isolate, mechanisms that may potentially be involved in its horizontal (autoinfection) and vertical (transovarial) transmission, and disease dynamics are discussed. The congeneric relationship between Agrilus planipennis and A. anxius makes it imperative to study the virulence of this Cystosporogenes species in A. planipennis.

Conclusions about the nontarget effects of putatively invasive pathogens should be based on biologically relevant data. We disagree that the research experiments on a microsporidium isolated from Harmonia axyridis conducted by Vilcinskas et al . (Reports, 17 May 2013, p. 862) can explain the decline of native coccinellid species in the absence of such data.
