3D electron diffraction (3D ED), also known as MicroED, has shown great potential in crystal structure determination in materials, small organic molecules, and macromolecules. In this work, a high-throughput 3D ED method has been implemented to identify six phases in an active pharmaceutical ingredient, griseofulvin (GSF). Batch data collection under low-dose conditions using the widely available commercial software EPU-D (Thermo Fisher Scientific) was combined with semi-automated data processing and clustering to collect and process over 100 datasets over three days. Accurate unit cell parameters obtained from 3D ED data allowed identification of GSF Forms III, I, GSF-and PEG IC-I, as well as three additional phases undetected in powder X-ray diffraction data, namely GSF II, GSF-V and a new phase GSF-PEG IC-II. Their structures were directly determined by 3D ED. Through structure analysis, we discovered polymorphism within inclusion complexes built from the same clinical drug-polymer combination. These results demonstrate the ability of the high-throughput method to accurately reveal the phase information of complex, beam-sensitive crystallisation products, which is significant for drug design where crystal form screening is crucial for the overall efficacy of the drug product.

Background: Respiratory birch pollen allergy and associated oral allergy syndrome affect more than 150 million people. IgE cross-sensitization to major birch pollen allergen Bet v 1 and pathogenesis-related (PR10) plant food allergens is responsible for the pollen-food allergy syndrome. Methods: We designed a recombinant protein, AB-PreS, consisting of non-allergenic peptides derived from the IgE binding sites of Bet v 1 and the cross-reactive apple allergen, Mal d 1, fused to the PreS domain of HBV surface protein as immunological carrier. AB-PreS was expressed in E. coli and purified by chromatography. The allergenic activity of AB-PreS was tested using sera and basophils from birch pollen patients allergic. The protective effect of AB-PreS was assessed by inhibition ELISA test using sera allergic patients and from immunized rabbits. Results: IgE-binding experiments and basophil activation test revealed the hypoallergenic nature of AB-PreS. IgG antibodies induced by 5 injections with AB-PreS inhibited allergic patients’ IgE binding to Bet v 1 and Mal d 1 better than did IgG induced by up to 30 injections of six licensed birch pollen allergen extract-based vaccines. Additionally, immunization with AB-PreS induced HBV-specific antibodies potentially protecting the infection. Conclusion: The recombinant AB-PreS-based vaccine is hypoallergenic, safe and superior to currently registered allergen extract-based vaccines for the treatment of the birch pollen food allergy syndrome.

Millions of people have been vaccinated with Gam-COVID-Vac but fine specificities of induced antibodies have not been fully studied. Plasma from 12 naïve and 10 coronavirus disease 2019 (COVID-19) convalescent subjects was obtained before and after two immunizations with Gam-COVID-Vac. Antibody reactivity in the plasma samples (n = 44) was studied on a panel of micro-arrayed recombinant folded and unfolded severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) proteins and 46 peptides spanning the spike protein (S) and by immunoglobulin G (IgG) subclass enzyme-linked immunosorbent assay (ELISA). The ability of Gam-COVID-Vac-induced antibodies to inhibit binding of the receptor-binding domain (RBD) to its receptor angiotensin converting enzyme 2 (ACE2) was investigated in a molecular interaction assay (MIA). The virus-neutralizing capacity of antibodies was studied by the pseudo-typed virus neutralization test (pVNT) for Wuhan-Hu-1 and Omicron. We found that Gam-COVID-Vac vaccination induced significant increases of IgG1 but not of other IgG subclasses against folded S, spike protein subunit 1 (S1), spike protein subunit 2 (S2), and RBD in a comparable manner in naïve and convalescent subjects. Virus neutralization was highly correlated with vaccination-induced antibodies specific for folded RBD and a novel peptide (i.e., peptide 12). Peptide 12 was located close to RBD in the N-terminal part of S1 and may potentially be involved in the transition of the pre- to post-fusion conformation of the spike protein. In summary, Gam-COVID-Vac vaccination induced S-specific IgG1 antibodies in naive and convalescent subjects in a comparable manner. Besides the antibodies specific for RBD, the antibodies induced against a peptide close to the N-terminus of RBD were also associated with virus-neutralization.

Background The determinants of successful humoral immune response to the severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) are of critical importance for the design of effective vaccines and the evaluation of the degree of protective immunity conferred by exposure to the virus. As novel variants emerge, understanding their likelihood of suppression by population antibody repertoires has become increasingly important. Methods In this study, we analyzed the SARS‐CoV‐2 polyclonal antibody response in a large population of clinically well‐characterized patients after mild and severe COVID‐19 using a panel of microarrayed structurally folded and unfolded SARS‐CoV‐2 proteins, as well as sequential peptides, spanning the surface spike protein (S) and the receptor‐binding domain (RBD) of the virus. Results S‐ and RBD‐specific antibody responses were dominated by immunoglobulin G (IgG), mainly IgG1, and directed against structurally folded S and RBD and three distinct peptide epitopes in S2. The virus neutralization activity of patients´ sera was highly correlated with IgG antibodies specific for conformational but not sequential RBD epitopes and their ability to prevent RBD binding to its human receptor angiotensin‐converting enzyme 2 (ACE2). Twenty percent of patients selectively lacked RBD‐specific IgG. Only immunization with folded, but not with unfolded RBD, induced antibodies against conformational epitopes with high virus‐neutralizing activity. Conformational RBD epitopes required for protection do not seem to be altered in the currently emerging virus variants. Conclusion These results are fundamental for estimating the protective activity of antibody responses after natural infection or vaccination and for the design of vaccines, which can induce high levels of SARS‐CoV‐2–neutralizing antibodies conferring sterilizing immunity., IgG response in convalescent COVID‐19 patients is directed to folded but not to unfolded RBD or RBD peptides. IgGs to folded RBD are required for virus neutralization. Twenty percent of convalescent COVID‐19 patients selectively lack RBD‐specific IgG. Human respiratory syncytial virus (RSV) is one of the most important causes of severe respiratory tract infections in early childhood. The only prophylactic protection is the neutralizing antibody, palivizumab, which targets a conformational epitope of the RSV fusion (F) protein. The F protein is generated as a F0 precursor containing two furin cleavage sites allowing excision of the P27 fragment and then gives rise to a fusion-competent version consisting of the N-terminal F2 subunit and the a C-terminal F1 subunits linked by two disulphide bonds. To investigate natural human F-specific antibody responses, F2 conferring the species-specificity of RSV, was expressed in Escherichia coli. Furthermore, the F0 protein, comprising both subunits F2 and F1, was expressed as palivizumab-reactive glycoprotein in baculovirus-infected insect cells. Six overlapping F2-derived peptides lacking secondary structure were synthesized. The analysis of IgG, IgA and IgM responses of adult subjects to native versions and denatured forms of F2 and F0 and to unfolded F2-derived peptides revealed that mainly non-conformational F epitopes, some of which represented cryptic epitopes which are not exposed on the proteins were recognized. Furthermore, we found a dissociation of IgG, IgA and IgM antibody responses to F epitopes with F2 being a major target for the F-specific IgM response. The scattered and dissociated immune response to F may explain why the natural RSV-specific antibody response is only partially protective underlining the need for vaccines focusing human antibody responses towards neutralizing RSV epitopes.

Three-dimensional electron diffraction (3D ED)/microcrystal electron diffraction (MicroED) techniques are gaining in popularity. However, the data processing often does not fit existing graphical user interface software, instead requiring the use of the terminal or scripting. Scipion-ED, described in this article, provides a graphical user interface and extendable framework for processing of 3D ED/MicroED data. An illustrative project is described, in which multiple 3D ED/MicroED data sets collected on tetragonal lysozyme were processed with DIALS through the Scipion-ED interface. The ability to resolve unmodelled features in the electrostatic potential map was compared between three strategies for merging data sets.

This paper reports on experiments conducted with a cold flow model utilized for the investigation of the particle residence time distribution and mixing characteristics in a bubbling fluidized bed with continuous solids exchange. The investigated system is of a rectangular cross section (0.4 × 0.2 m) with a bed height of 0.17 m. A measurement device based on an alternating current bridge circuitry coupled with lock-in amplifier technology was built in the scope of quantifying the solids residence time distribution, whereby a pulse-injected ferromagnetic tracer creates the input signal. The implementation of a profound mathematical routine ensures the reproducible calculation of the particles mean residence time and characteristic values describing particle mixing phenomena. Therefore, the E-curve was modeled by mathematical convolution of the exit age distributions available for an ideally mixed continuous stirred tank reactor and a plug flow reactor with axial dispersion. It is shown that the in-bed mixing is highly dependent on the fluidization rate as well as on the solids circulation rate. Albeit the lowest superficial gas velocity equals a fluidization number of 4.7, the formation of dead spaces and short-circuit flows was observed under these conditions. Axial dispersion coefficients in the range of 5 · 10 - 3 to 7 · 10 - 1 m2 s−1 were obtained.

Birch pollen allergy is among the most prevalent pollen allergies in Northern and Central Europe. This IgE-mediated disease can be treated with allergen immunotherapy (AIT), which typically gives rise to IgG antibodies inducing tolerance. Although the main mechanisms of allergen immunotherapy (AIT) are known, questions regarding possible Fc-mediated effects of IgG antibodies remain unanswered. This can mainly be attributed to the unavailability of appropriate tools, i.e., well-characterised recombinant antibodies (rAbs). We hereby aimed at providing human rAbs of several classes for mechanistic studies and as possible candidates for passive immunotherapy. We engineered IgE, IgG1, and IgG4 sharing the same variable region against the major birch pollen allergen Bet v 1 using Polymerase Incomplete Primer Extension (PIPE) cloning. We tested IgE functionality and IgG blocking capabilities using appropriate model cell lines. In vitro studies showed IgE engagement with FcεRI and CD23 and Bet v 1-dependent degranulation. Overall, we hereby present fully functional, human IgE, IgG1, and IgG4 sharing the same variable region against Bet v 1 and showcase possible applications in first mechanistic studies. Furthermore, our IgG antibodies might be useful candidates for passive immunotherapy of birch pollen allergy.

Background: N-linked glycans present in venoms, pollen and mites are recognized by IgE antibodies from more than 20% of allergic patients but have low or no allergenic activity. Objectives: To engineer recombinant glycoproteins resembling carbohydrate-specific IgE epitopes from venoms, pollen and mites which can discriminate carbohydrate-specific IgE from allergenic, peptide-specific IgE. Methods: One or two N-glycosylation sites were engineered into the N-terminus of the non-allergenic protein horse heart myoglobin (HHM) using synthetic gene technology. HHM 1 and HHM 2 containing one or two N-glycosylation sites were expressed in baculovirus-infected High-FiveTM insect cells and a non-glycosylated version (HHM 0) was obtained by mutating the glycosylation motif. Recombinant HHM proteins were analyzed regarding fold and aggregation by circular dichroism and gel filtration, respectively. IgE reactivity was assessed by ELISA, immunoblotting and quantitative ImmunoCAP measurements. IgE inhibition assays were performed to study cross-reactivity with venom, plant and mite-derived carbohydrate IgE epitopes. Results: HHM-glycovariants were expressed and purified from insect cells as monomeric and folded proteins. The HHM-glycovariants exhibited strictly carbohydrate-specific IgE reactivity, designed to quantify carbohydrate-specific IgE and resembled IgE epitopes of pollen, venom and mite-derived carbohydrates. IgE-reactivity and inhibition experiments established a hierarchy of plant glcyoallergens (nPhl p 4>nCyn d 1>nPla a 2>nJug r 2>nCup a 1>nCry j 1) indicating a hitherto unknown heterogeneity of carbohydrate IgE epitopes in plants which were completely represented by HHM 2. Conclusion: Defined recombinant HHM-glycoproteins resembling carbohydrate-specific IgE epitopes from plants, venoms and mites were engineered which made it possible to discriminate carbohydrate- from peptide-specific IgE reactivity. Literature offers several excellent contributions in view of hydrodynamics and heat transfer characteristics of fluidized beds. Our investigations deal with the achievable wall-to-bed heat transfer rates at a tube bundle heat exchanger immersed in a bubbling bed of Geldart Type B particles. We confirm the findings of other authors, who described the lateral distribution of bed voidage as well as horizontal and vertical particle velocity and heat transfer. The heat transfer coefficients in the core of the bed are significantly higher than they are at the walls. An adapted gas distributor with a non-uniform nozzle-grid was used to overcome the disadvantage of unevenly distributed heat transfer. While the heat transfer coefficients close to the wall are 25–50% lower than in the core region when a uniform nozzle-grid is used, the effect can be fully balanced with the modified gas distributor. We conclude that a more even lateral distribution of heat transfer rates is possible while the overall heat exchange rate remains unchanged. Nevertheless, an increased uniformity in lateral particle mixing may indicate an improvement in heat surface load and mass transfer.

Heat transfer between bubbling fluidized beds and immersed heat exchanger surfaces is studied in view of continuously operated temperature swing adsorption processes for post-combustion CO2 capture. A novel heat transfer measurement test device was used to measure wall-to-bed heat transfer coefficients. The present work focuses on the comparison of experimentally obtained and calculated heat transfer coefficients. Heat transfer at horizontal single tubes and tube bundles immersed in fluidized particle beds of glass beads with 140 μm and 200 μm in Sauter mean diameter is investigated. It is shown that the experimental results for single tubes are in mediocre agreement to established mathematical models, such as the ones proposed by Natusch et al. (1975) and Molerus et al. (1995), and that heat transfer is significantly influenced by the tube diameter. The model by Petrie et al. (1968) was considered to take the effect of the tube diameter into account, which lead to promising results. Furthermore, measured heat transfer coefficients at tube bundles of different geometries are compared to predictions using the models by Natusch et al. (1975) and Lechner et al. (2013). Some of the tube bundle reduction factors predicted by the model by Lechner et al. (2013) are larger than one, which stands in contrast to the finding that the highest heat transfer coefficients occur at single tubes. However, both models lead to adequate results when calculating heat transfer coefficients for different tube bundle geometries.

As proof-of-concept, 1,3-dihydroxybenzene (resorcinol) was carboxylated in the ortho-position using pressurized CO2 (~30-40 bar) catalyzed by ortho-benzoic acid decarboxylases with up to 68% conversion. Optimization studies revealed tight pH-control and enzyme stability as the most important determinants.

Functionalized solid amine-based temperature swing adsorption (TSA) processes have recently been proposed as a potential way to reduce the energy-penalty of post-combustion carbon capture processes. Thereby, multi-stage fluidized bed contactors with immersed heat exchanger surfaces and counter-current flow of solids and gas phase may solve the heat transfer challenge while maintaining the thermodynamic process requirements. Hence, the present work develops design requirements for TSA stages based on achievable heat transfer rates in bubbling fluidized beds. The considered particles are Geldart Type B. It is shown that the pressure drop of multi-stage fluidized bed TSA units for flue gas CO 2 capture is practically determined by the heat exchange requirement. Scalability, maintainability and durability of different heat exchanger geometries are considered. The net movement and mixing of particles within the bubbling bed stage must be maintained in spite of the immersed heat exchangers concerning possible dead zones in the area of the tube bundles. Comprehensive models are used to predict heat transfer coefficients for tubes immersed in fluidization. A heat transfer measurement test device for optimization of the heat exchanger geometry has been put into operation and heat exchange measurement results are compared to calculated heat transfer coefficients. It is shown that experimentally obtained heat transfer rates for single tubes are in good agreement with modeled values. A model proposed for Geldart A particles is used to estimate heat transfer rates for two particular tube bundles with a tube diameter of 25 mm and horizontal tube spacing of 2.2 and 2.8. It is shown that the calculated results represent heat transfer rates qualitatively and quantitatively for tube bundle heat exchangers immersed in Geldart Type B particle fluidized beds. Although this article has been motivated by heat exchange in TSA, it may be of interest for other applications concerned with heat transfer between bubbling fluidized beds and immersed heat exchanger surfaces.

Arginine kinase (AK) was first identified as an allergen in the Indian-meal moth and subsequently shown to occur as allergen in various invertebrates and shellfish. The cDNA coding for AK from the house dust mite (HDM) species Dermatophagoides pteronyssinus, Der p 20, has been isolated, but no recombinant Der p 20 (rDer p 20) allergen has been produced and characterized so far. We report the expression of Der p 20 as recombinant protein in Escherichia coli. rDer p 20 was purified and shown to be a monomeric, folded protein by size exclusion chromatography and circular dichroism spectroscopy, respectively. Using AK-specific antibodies, Der p 20 was found to occur mainly in HDM bodies, but not in fecal particles. Thirty percent of clinically well-characterized HDM allergic patients (n = 98) whose immunoglobulin E (IgE) reactivity profiles had been determined with an extensive panel of purified HDM allergens (Der f 1, 2; Der p 1, 2, 4, 5, 7, 10, 11, 14, 15, 18, 21, 23 and 37) showed IgE reactivity to Der p 20. IgE reactivity to Der p 20 was more frequently associated with lung symptoms. AKs were detected in several invertebrates with specific antibodies and Der p 20 showed IgE cross-reactivity with AK from shrimp (Litopenaeus vannamei). Thus, Der p 20 is a cross-reactive HDM allergen and may serve as a diagnostic marker for HDM-induced lung symptoms such as asthma.

The molecular basis for the propensity of a small number of environmental proteins to provoke allergic responses is largely unknown. Herein, we report that mite group 13 allergens of the fatty acid-binding protein (FABP) family are sensed by an evolutionarily conserved acute-phase protein, serum amyloid A1 (SAA1), that promotes pulmonary type 2 immunity. Mechanistically, SAA1 interacted directly with allergenic mite FABPs (Der p 13 and Blo t 13). The interaction between mite FABPs and SAA1 activated the SAA1-binding receptor, formyl peptide receptor 2 (FPR2), which drove the epithelial release of the type-2-promoting cytokine interleukin (IL)-33 in a SAA1-dependent manner. Importantly, the SAA1–FPR2–IL-33 axis was upregulated in nasal epithelial cells from patients with chronic rhinosinusitis. These findings identify an unrecognized role for SAA1 as a soluble pattern recognition receptor for conserved FABPs found in common mite allergens that initiate type 2 immunity at mucosal surfaces. Smole and colleagues show that the soluble pattern recognition receptor serum amyloid A (SAA) recognizes several mite allergenic proteins, including Der p 13 and Blo t 13, which are conserved fatty acid-binding proteins. Such FABP–SAA1 binding triggers epithelial cell release of the type-2-promoting cytokine IL-33, which in turn drives IL-13 production and allergic syptoms.

The cross-linking of effector cell-bound IgE antibodies by allergens induces the release of inflammatory mediators which are responsible for the symptoms of allergy. We demonstrate that a recombinant hybrid molecule consisting of the major birch (Bet v 1) and grass (Phl p 5) pollen allergen exhibited reduced allergenic activity as compared to equimolar mixes of the isolated allergens in basophil activation experiments. The reduced allergenic activity of the hybrid was not due to reduced IgE reactivity as demonstrated by IgE binding experiments using sera from allergic patients. Physicochemical characterization of the hybrid by size exclusion chromatography, dynamic light scattering, negative-stain electron microscopy and circular dichroism showed that the hybrid occurred as folded aggregate whereas the isolated allergens were folded monomeric proteins. IgG antibodies raised in rabbits against epitopes of Bet v 1 and Phl p 5 showed reduced reactivity with the hybrid compared to the monomeric allergens. Our results thus demonstrate that aggregation can induce changes in the conformation of allergens and lead to the reduction of allergenic activity. This is a new mechanism for reducing the allergenic activity of allergens which may be important for modifying allergens to exhibit reduced side effects when used for allergen-specific immunotherapy.

Immunoglobulin E (IgE)-associated allergy affects more than 25% of the population. Can f 1 is the major dog allergen associated with respiratory symptoms but the epitopes recognized by allergic patients IgE on Can f 1 are unknown. To characterize IgE epitopes of Can f 1 recognized by dog allergic patients, six overlapping peptides spanning the Can f 1 sequence were synthesized. In direct IgE epitope mapping experiments peptides were analyzed for IgE reactivity by dot blot and Enzyme-linked immunosorbent assay (ELISA) with sera from dog allergic patients. For indirect epitope-mapping, rabbits were immunized with the peptides to generate specific IgG antibodies which were used to inhibit allergic patients’ IgE binding to Can f 1. IgE binding sites were visualized on a model of the Can f 1 three-dimensional structure. We found that Can f 1 does not contain any relevant sequential IgE epitopes. However, IgE inhibition experiments with anti-peptide specific IgGs showed that Can f 1 N- and C-terminal portion assembled a major conformational binding site. In conclusion, our study is the first to identify the major IgE epitope-containing area of the dog allergen Can f 1. This finding is important for the development of allergen-specific treatment strategies.

Ni 45 Co 5 Mn 36.6 In 13.4 ribbons were prepared by the melt spinning technique. They are in the austenitic state at the room temperature with a Cubic L2 1 structure and are composed of columnar grains growing perpendicular to the ribbon plane. A nearly single {001}〈100〉 cube component is formed at low wheel-rotating speed due to the preferential growth of nuclei in the ribbon plane normal and length directions. Appropriate control of the melt spinning condition offers a promising solution to the texturation of polycrystalline ferromagnetic shape memory alloys.

Conjugative transfer through type IV secretion multiprotein complexes is the most important means of spreading antimicrobial resistance. Plasmid pIP501, frequently found in clinicalEnterococcus faecalisandEnterococcus faeciumisolates, is the first Gram-positive (G+) conjugative plasmid for which self-transfer to Gram-negative (G−) bacteria has been demonstrated. The pIP501-encoded type IV secretion system (T4SS) protein TraN localizes to the cytoplasm and shows specific DNA binding. The specific DNA-binding site upstream of the pIP501 origin of transfer (oriT) was identified by a novel footprinting technique based on exonuclease digestion and sequencing, suggesting TraN to be an accessory protein of the pIP501 relaxase TraA. The structure of TraN was determined to 1.35 Å resolution. It revealed an internal dimer fold with antiparallel β-sheets in the centre and a helix–turn–helix (HTH) motif at both ends. Surprisingly, structurally related proteins (excisionases from T4SSs of G+ conjugative transposons and transcriptional regulators of the MerR family) resembling only one half of TraN were found. Thus, TraN may be involved in the early steps of pIP501 transfer, possibly triggering pIP501 TraA relaxase activity by recruiting the relaxosome to the assembled mating pore.

Nitric oxide inhibits growth of glomerular mesangial cells: Role of the transcription factor Egr-1 Background In previous studies, we found a close link of early growth response gene-1 (Egr-1) expression to mesangial cell (MC) proliferation. Antiproliferative agents inhibited mitogen-induced Egr-1 expression. Here we investigated the effect of S-nitrosoglutathione (GSNO) on the proliferation of MCs, specifically asking how GSNO regulates the transcription factor Egr-1, which we have previously shown to be critical for the induction of MC mitogenesis. Methods The proliferation of MCs was measured by thymidine incorporation and cell counting. Egr-1 mRNA and protein levels were detected by Northern and Western blots. Electrophoretic mobility shift assays (EMSAs) and chloramphenicol acetyltransferase (CAT) assays were performed to test whether GSNO modulates DNA binding and transcriptional activation of Egr-1. Results GSNO strongly inhibited serum-induced MC proliferation (-84% at 1 mmol/L). A mild inhibition of serum-induced Egr-1 mRNA was observed at GSNO concentrations from 50 to 200 μmol/L, whereas mRNA levels increased again at concentrations above 500 μmol/L. This increased mRNA expression, however, was not translated into Egr-1 protein. Instead, Egr-1 protein induction was inhibited (-40%). EMSAs indicated that GSNO inhibited specific binding of Egr-1 to its DNA consensus sequence. Moreover, transcriptional activation by Egr-1 in CAT assays using a reporter plasmid bearing three Egr-1 binding sites was strongly suppressed by GSNO. Conclusions Our data identify GSNO as a potent inhibitor of MC growth with potential beneficial effects in proliferative glomerular diseases. This antimitogenic property is mediated at least in part by inhibitory effects of GSNO on Egr-1 protein levels and by reducing the ability of Egr-1 to activate transcription by impairing its DNA binding activity.

Expression of the transcriptional regulator Egr-1 in experimental glomerulonephritis: Requirement for mesangial cell proliferation. The early growth response gene-1 (Egr-1), a zinc finger transcriptional regulator, was induced in a rat model of mesangioproliferative glomerulonephritis (GN). Northern blot analysis revealed a maximal 14.9-fold increase in glomerular Egr-1 mRNA at day 6 of GN. By immunohistochemistry Egr-1 protein expression was demonstrated to be mainly confined to glomerular mesangial cells (MC). To test whether Egr-1 directly regulates MC proliferation, cultured MCs were stimulated with platelet-derived growth factor (PDGF) after preincubation with different Egr-1 antisense oligonucleotides (ASOs). PDGF-induced Egr-1 mRNA levels were inhibited by up to 75% and protein levels by up to 91%. In addition Egr-1-specific ASOs blocked PDGF-induced rise in 3 H-thymidine uptake by 83% and almost completely abrogated increase in MC number. We conclude that Egr-1 induction is of critical importance for PDGF-induced mitogenic signaling in MCs, and inhibition of Egr-1 in vivo may offer an approach to oppose glomerular MC proliferation in glomerular inflammatory disease.

During inflammatory processes of the kidney, lesions of the glomerulus lead to aggregation of thrombocytes and infiltration of macrophages, which can release bioactive mediators. One of these important signalling molecules is lysophosphatidic acid (LPA). Incubation of rat mesangial cells with LPA induced mRNA and protein expression of the early-response genes pghs-2 (for prostaglandin G/H synthase-2/cyclo-oxygenase-2) and egr-1. As shown by antisense experiments, induction of egr-1 was related to the strong mitogenic effect of LPA. LPA-mediated gene expression was inhibited by pertussis toxin, indicating coupling to G-proteins of the Gi family. Specific inhibition of proteins of the small G-protein subfamily Rho with toxin B from Clostridium difficile led to changes in mesangial cell morphology without induction of apoptosis. LPA-mediated expression of pghs-2 and egr-1 was reduced to base-line levels by toxin B, indicating a role for Rho proteins in LPA-mediated gene induction. Of the two mitogen-activated protein kinase (MAPK) pathways investigated, the MAPK kinase-extracellular signal-regulated kinase pathway was involved in the induction of both pghs-2 and egr-1 mRNA expression, as shown by the inhibitory effect of PD98059. Activation of the MAPK p38, however, was only related to pghs-2 expression, whereas egr-1 expression was not affected by treatment of mesangial cells with the specific inhibitor SB203580. Taken together our data provide evidence that LPA-mediated activation of MAPK kinase and Rho proteins leads to the induction of the functionally distinct early-response genes pghs-2 and egr-1, whereas activation of MAPK p38 revealed considerable differences between the regulation of these two genes.

Increase of glomerular mesangial cells (MCs) is a prominent histopathological finding in many types of glomerulonephritis. We have shown previously that expression of the zinc-finger transcription factor, early growth response gene-1 (egr-1), is closely correlated with the proliferation of cultured MCs. To elucidate whether Egr-1 is required for MC proliferation, we inhibited serum-induced Egr-1 expression by phosphothioate-modified antisense oligonucleotides (ODNs). Uptake of antisense ODNs into MCs was demonstrated, and five different egr-1 antisense ODNs were tested for their impact on serum-induced egr-1 mRNA and protein levels and on MC growth. The most potent egr-1 antisense ODN inhibited serum-induced egr-1 mRNA by 68%, protein induction by 58%, and MC replication as measured by [3H]thymidine uptake and cell counts by 78 and 46%, respectively. The effects of antisense ODNs on MC growth correlated closely with their ability to inhibit Egr-1 protein. ODNs acted in a dose-dependent manner, the minimal effective concentration being 1 μM. Control ODNs had no significant effects. In addition, antisense ODNs against egr-1 potently inhibited endothelin-1-induced Egr-1 expression and MC growth. Heparin, a known inhibitor of MC growth, suppressed serum-induced [3H]thymidine uptake by 39% and egr-1 mRNA expression by 44%. We conclude that Egr-1 is an essential part of the mitogenic signal transduction cascade in cultured MCs.

ABSTRACTS Quenched and tempered plates with a yield strength of 420-500 MPa are currently used in offshore platform fabrication. In this paper, the development of a cast steel is reported, which is fully compatible to quenched and tempered plate materials. Mechanical properties as well as fracture mechanics characteristics and extensive welding procedure qualifications are presented. High strength steel castings offer the chance to manufacture complex heavy-lift and fatigue-critical components for largeroffshore structures without increasing the weight of the platforms. INTRODUCTION The use of high integrity steel castings in offshore structural and lifting applications is well established l,2 Deep-water platforms are exposed to severe environmental loads in the form of wind, waves and currents. For these high stressed applications, high strength low alloy (HSLA) steel castings with a yield strength of min. 450 MPa and excellent weldability are essential. An interesting alternative to the normalized steel castings generally used up to now are water quenched and tempered steel casting grades. In this paper, the aim is to emphasize the effect of modern steel production, improvements in casting design, using computer aided simulation of solidification and stress analysis, on weight reduction. By means of optimum designed steel castings, the high-quality standard of the offshore industry can be maintained. CAST STEEL DEVELOPMENT Among the possibilities to improve the toughness and strength of weldable cast steel are:Modifications of the chemical composition of the steelImplementation of modern steel melting techniquesHigher cooling rates during heat treatment Chemical Composition To assure good weldability under shop, as well as field conditions, the carbon equivalent should be as low as possible. Old fashioned weldable cast steel grades, for example GS-20Mn5, generally show a carbon level of 0.20 %. For the new steel grades the carbon level has to be reduced to about 0.10 %. The carbon equivalent CE (IIW) is within the range of 0.400.46 %. Some typical chemical compositions ofweldable steel castings and plates are summarized in table 1(3,4,5,6,7). These steel grades are basically MnNiMoCu-alloyed. Niobium and/or vanadium are added primarily to provide grain refinement. In order to avoid severe precipitation hardening effects, V+Nb should be limited to ? 0.10 %. The chemical composition of the modified "GS-13 MnNi 6 4" had to be adapted (as far as possible) to those of quenched and tempered plate materials, Due to the great differences in the wall thickness of steel castings, specifications concerning the Ni-level have been changed to wider ranges. STEELMAKING The implementation of modern steel melting technology in the foundry was an important decision in achieving high-strength cast steels with adequate toughness as well as sufficient safety against brittle fracture. The concept of modern steel meltingtechnology is illustrated in fig. 1. Due to the significant influence of sulphur on the toughness of structural steel castings, desulphurisation should already be performed on the hot metal.

A method and apparatus for ascertaining internal stresses in a hardened region to be tested for a component made of a predetermined material includes measuring coercive field strength and amplitude of magnetic Barkhausen noise as a function of mechanical stress and hardness in calibration samples of a predetermined material of known hardness and known internal stress. Calibration functions are ascertained from the measured values indicating the dependency of the hardness and the amplitude of the magnetic Barkhausen noise as functions of the hardness and mechanical stress. The coercive field strength and the amplitude of the magnetic Barkhausen noise is measured in a location-dependent manner over the region of the component to be tested. The measured values of the coercive field strength and the amplitude of the magnetic Barkhausen noise are converted in the region to be tested into location-dependent hardness and into location-dependent mechanical stress using the ascertained calibration functions. The location-dependent mechanical stress and the location-dependent hardness in the region to be tested are present independently of one another and are available for further processing.
