2. Using a 'Primer Unit' in an Introductory Biology Course: 'A Soft Landing'

Abstract

This study aimed to facilitate students' entrance to an introductory cell biology course for biology majors. The most prominent difficulty in this introductory course, is students' poor background-knowledge, such as a lack of understanding of very basic concepts and terms, and the huge differences in students' background knowledge. In order to bring all the students to a common and adequate level of comprehension and familiarity of basic terms, thus giving a fair chance to all attending the introductory cell biology course, the chief instructor decided to build a "primer unit". The unit provided an overview of the subject matter of the course, and it was placed on the course website before the beginning of the course. The findings show that students who consulted the primer unit came to the course better prepared. The primer helped to reduce the gap between students who studied advanced biology in high school and those who did not. Even though the differences between students who went over the primer and those who did not was not sustained after formal learning, students reported that due to the primer they started the course less intimidated and with a better idea about what to expect. 3. Using the Concept Map Technique in Teaching Introductory Cell Biology to College Freshmen

Abstract

In our study, we focused on the conceptual understanding of the concepts and processes presented in the first lectures of an introductory course in cellular biology for biology majors. The study topic we considered was, "the structure of DNA and the functions of nucleotides". One hundred and eighteen students were asked to prepare concept maps from a list of twenty given concepts. Analysis of these maps has shown a compartmentalization into two major groups of concepts. The most frequent concepts were from the genetic aspect, while frequencies of concepts from the energetic aspect were comparatively low. Many students did not recognize that molecules like ATP or GTP are simply nucleotides. Other interesting misconceptions concerned the concepts of nucleic acids, purines and pyrimidines. One of the advantages of using the concept map technique was that it encouraged the instructor to start using maps as a graphic instructional tool, summarizing his lectures. In addition, the need to select twenty concepts and arrange them in a map forced him to go over his lectures and reconsider whether or not these concepts should have been selected for instruction. Abstract

The CCR5 gene mutation appears to occur in a high percentage of Ashkenazi Jews. This mutation protects people against HIV infection. The polymerase chain reaction was used to determine the frequency of the CCR5 mutation in DNA samples collected from 520 Israelis. Ten percent of the Ashkenazi Jews tested positive for the mutation compared to 2% of North African Jews, 1.3% of non-Jews, 1% of Middle Eastern Jews and none of the Ethiopian Jews. Other studies have found the frequency of this mutation to be as high as 21% in Ashkenazi Jews.

Abstract

Abstract Filamentous bacteriophages have been used in numerous applications for the display of antibodies and random peptide libraries. Here we describe the introduction of a 13 amino acid sequence LASIFEAQKIEWR (designated BT, which is biotinylated in vivo by E. coli) into the N termini of four of the five structural proteins of the filamentous bacteriophage fd (Proteins 3, 7, 8 and 9). The in vivo and in vitro biotinylation of the various phages were compared. The production of multifunctional phages and their application as affinity reagents are demonstrated.

Abstract

Antibodies are an effective line of defense in preventing infectious diseases. Highly potent neutralizing antibodies can intercept a virus before it attaches to its target cell and, thus, inactivate it. This ability is based on the antibodies’ specific recognition of epitopes, the sites of the antigen to which antibodies bind. Thus, understanding the antibody/epitope interaction provides a basis for the rational design of preventive vaccines. It is assumed that immunization with the precise epitope, corresponding to an effective neutralizing antibody, would elicit the generation of similarly potent antibodies in the vaccinee. Such a vaccine would be a ‘B-cell epitope-based vaccine’, the implementation of which requires the ability to backtrack from a desired antibody to its corresponding epitope. In this article we discuss a range of methods that enable epitope discovery based on a specific antibody. Such a reversed immunological approach is the first step in the rational design of an epitope-based vaccine. Undoubtedly, the gold standard for epitope definition is x-ray analyses of crystals of antigen: antibody complexes. This method provides atomic resolution of the epitope; however, it is not readily applicable to many antigens and antibodies, and requires a very high degree of sophistication and expertise. Most other methods rely on the ability to monitor the binding of the antibody to antigen fragments or mutated variations. In mutagenesis of the antigen, loss of binding due to point modification of an amino acid residue is often considered an indication of an epitope component. In addition, computational combinatorial methods for epitope mapping are also useful. These methods rely on the ability of the antibody of interest to affinity isolate specific short peptides from combinatorial phage display peptide libraries. The peptides are then regarded as leads for the definition of the epitope corresponding to the antibody used to screen the peptide library. For epitope mapping, computational algorithms have been developed, such as Mapitope, which has recently been found to be effective in mapping conformational discontinuous epitopes. The pros and cons of various approaches towards epitope mapping are also discussed.
