24. Evaluation of an elastic decellularized tendon‐derived scaffold for the vascular tissue engineering application

Abstract

Due to the limited success rate of currently available vascular replacements, tissue engineering has received tremendous attention in recent years. A main challenge in the field of regenerative medicine is creating a mechanically functional tissue with a well-organized extracellular matrix, particularly of collagen and elastin. In this study, the native collagen scaffold derived from decellularized tendon sections, as a scaffold having the potential to be used for vascular tissue engineering applications, was studied. We showed that the elasticity of the scaffolds was improved when crosslinked with the bovine elastin. The effect of different concentrations of elastin on mechanical properties of the collagen scaffolds was evaluated of which 15% elastin concentration was selected for further analysis based on the results. Addition of 15% elastin to collagen scaffolds significantly decreased Young's modulus and the tensile stress at the maximum load and increased the tensile strain at the maximum load of the constructs as compared to those of the collagen scaffolds or control samples. Moreover, tubular elastin modified collagen scaffolds showed significantly higher burst pressure compared to the control samples. Smooth muscle cells and endothelial cells cultured on the elastin modified collagen scaffolds showed high viability (>80%) after 1, 3, and 7 days. Furthermore, the cells showed a high tendency to align with the collagen fibers within the scaffold and produced their own extracellular matrix over time. In conclusion, the results show that the decellularized tendon sections have a great potential to be used as scaffolds for vascular tissue engineering applications. (c) 2019 Wiley Periodicals, Inc. Abstract

Resin-based composites are used for bone repair applications and comprise resin matrix and different sized filler particles. Nanometer-sized filler particles improve composite's mechanical properties compared with micrometer-sized filler particles, but whether differences exist in the biological response to these composites is unknown. Natural bone comprises a nanocomposite structure, and nanoscale interactions with extracellular matrix components influence stem cell differentiation. Therefore we hypothesized that nanometer-sized filler particles in resin-based composites enhance osteogenic differentiation of stem cells showing a more bone cell-like response to mechanical loading compared with micrometer-sized filler particles. Pulsating fluid flow (PFF; 5 Hz, mean shear stress: 0.7 Pa; 1 h) rapidly, within 5 min, increased nitric oxide production in human adipose stem cells (hASCs) on nanocomposite, but not on micro-hybrid composite. PFF increased RUNX2 expression in hASCs on micro-hybrid composite, but not on nanocomposite after 2 h post-incubation. PFF did not affect mean cell orientation and shape index of hASCs on both composites. In conclusion, the PFF-increased nitric oxide production in hASCs on nanocomposite, and increased osteogenic differentiation of hASCs on micro-hybrid composite suggest different responses to mechanical loading of hASCs on composite with nanometer-sized and micrometer-sized filler particles. Abstract

Background:Decellularized tissue-engineered heart valves (TEHVs) are under investigation as alternative for current heart valve prostheses with the potential to rapidly repopulate with cells within the body. Ideally, these valves are stented for transapical or minimally invasive delivery. It is unclear if and how the matrix of these valves remodels under in vivohemodynamic loading conditions and in the presence of a stent. Here, we study the evolution of collagen orientation and tissue maturation in the wall of stented decellularized TEHVs with time after implantation.Methods and Results:In a previous study, stented TEHVs based on rapidly degrading scaffolds were cultured in bioreactors, decellularized, and transapically implanted as pulmonary valve replacement in sheep. In the present study, collagen (re)orientation in the initially isotropic valvular wall was assessed using a fluorescent collagen probe combined with confocal imaging and image analysis of explanted tissue at 8, 16, and 24 weeks following implantation. Collagen tortuosity or waviness in the explants, as a measure of matrix maturity, was quantified using a Gabor wavelet method and compared with tortuosity in native sheep vascular wall tissue. Results indicate that on the luminal side of the valvular wall, fibers became aligned in circumferential direction, while tortuosity increased with implantation time, showing striking similarities with the native collagen structure after 24 weeks. On the outside of the wall, where the engineered tissue touches the stent, collagen fibers in the vicinity of the struts aligned along the struts, whereas collagen fibers in between struts were randomly oriented. Immunohistochemistry was performed to evaluate the presence of elastin and collagen type I and III. After 8 weeks, collagen types I and III were mostly present at the luminal side of the wall, whereas at 16 and 24 weeks, a homogenous distribution of collagen I and III was observed throughout the wall. Elastin was mostly expressed at the luminal side after 24 weeks. Biochemical assays showed that the amount of DNA (as a measure of cell number) increased significantly after 8 and 24 weeks, glycosaminoglycans increased significantly after 8, 16, and 24 weeks, and hydroxyproline, as a measure of collagen amount, increased significantly after 24 weeks compared to the controls.Conclusions:The collagen matrix in the wall of decellularized TEHVs shows clear structural remodeling and maturation with time. While collagen orientation rapidly remodels toward a native anisotropic architecture on the luminal side of the engineered valvular wall, it is dominated and guided by stent geometry on the outer side of the wall. Collagen tortuosity was increased with implantation time and was accompanied by an increase in elastin, especially on the luminal side of the vessel.

Abstract

Objective Aging is accompanied by a series of changes in mature tissues that influence their properties and functions. Collagen, as one of the main extracellular components of cartilage, becomes highly crosslinked during aging. In this study, the aim was to examine whether a correlation exists between collagen crosslinking induced by artificial aging and mechanical properties of the temporomandibular joint (TMJ) condyle. To evaluate this hypothesis, collagen crosslinks were induced using ribose incubation. Methods Porcine TMJ condyles were incubated for 7 days with different concentrations of ribose. The compressive modulus and stiffness ratio (incubated versus control) was determined after loading. Glycosaminoglycan and collagen content, and the number of crosslinks were analyzed. Tissue structure was visualized by microscopy using different staining methods. Results Concomitant with an increasing concentration of ribose, an increase of collagen crosslinks was found. The number of crosslinks increased almost 50 fold after incubation with the highest concentration of ribose. Simultaneously, the stiffness ratio of the samples showed a significant increase after incubation with the ribose. Pearson correlation analyses showed a significant positive correlation between the overall stiffness ratio and the crosslink level; the higher the number of crosslinks the higher the stiffness. Conclusion The present model, in which ribose was used to mimic certain aspects of age-related changes, can be employed as an in vitro model to study age-related mechanical changes in the TMJ condyle. Abstract

Some wounds are non-healing and can be life-threatening particularly in older patients. Only in EU annually around 4 million individuals suffer from different types of wounds, requiring further medical treatment, such as wound dressings. In this PhD project, the fabrication of cellulose aerogel fibers and their potential for biomedical application of drug delivery and wound dressing has been investigated. Cellulose aerogel fibers are highly porous and nanostructured material made from the most abundant biobased polymer, “cellulose’’. This research has proven that cellulose aerogel fibers can provide a moist environment and absorbs wound exudate for optimal wound healing. It is possible to deposit drug in the porous structure of the fibers and tune their release time; They are biocompatible and can be removed painlessly from the wound site. Therefore, cellulose aerogel fibers have a great potential to be used as wound dressing material.
