Biochar is potentially a functional ingredient in animal nutrition that offers health benefits such as detoxification, while also promoting environmental sustainability through carbon sequestration, emission reduction, and its circular production. However, the heterogeneity of commercially available biochar products requires a detailed assessment of their functional properties for applications in animal feed. This study evaluates chestnut biochar from morphological, chemical, and metabolomic perspectives and assesses its functional properties. Metabolomic analysis of a water extract using QTOF HPLC-MS/MS confirmed the presence of bioactive compounds, such as hydroxybenzoic and succinic acids, highlighting its potential as a functional feed ingredient. The chestnut biochar inhibited the growth of the pathogenic E. coli strains F18+ and F4+, with maximum inhibition rates of 15.8% and 28.6%, respectively, after three hours of incubation. The downregulation of genes associated with quorum sensing (MotA, FliA, FtsE, and HflX, involved in biofilm formation and cellular division) suggests that biochar interferes with several aspects of the pathogenic process. Importantly, biochar was not found to adversely affect beneficial probiotic bacteria, such as Lactiplantibacillus plantarum and Limosilactobacillus reuteri. These findings support the potential of chestnut biochar as a versatile ingredient for sustainable animal nutrition, thus promoting animal welfare while offering environmental benefits. Paramutation, a specific epigenetic phenomenon first identified in Zea mays by Alexander Brink in the 1950s, has since been observed in different plant and animal species. What sets paramutation apart from other gene silencing processes is its ability for one silenced allele (referred to as paramutagenic) to silence another allele (paramutable) in trans. The resultant silenced allele (paramutated) remains stable across generations, even after separating from the paramutagenic allele, and acquires paramutagenic properties itself. This hereditary behavior facilitates the rapid dissemination of specific gene expression patterns or phenotypes within populations, disrupting the Hardy–Weinberg equilibrium even without other evolutionary pressures. Despite extensive research, the molecular mechanisms underlying paramutation are still not fully understood, although numerous studies suggest the involvement of RNA-mediated changes in DNA methylation and in the chromatin structure of silenced genes. In this paper, we report preliminary evidence regarding horizontal paramutation transfer at the pl1 (purple plant1) regulatory gene involved in the accumulation of anthocyanin in several plant tissues such as tassel, kernel, and cob. A paramutated pl1′ allele arose spontaneously in a pl1 population, and in this study, we found evidence of silencing events spatially associated in the field, suggesting a possible horizontal transfer of silencing among nearby plants. Simple Summary: Biochar is a carbon-rich material produced by the thermal decomposition of organic matter (biomass) in an oxygen-limited environment, a process known as pyrolysis. It has attracted significant attention due to its potential environmental benefits, such as carbon sequestration and waste management, promoting a circular economy. The in vivo properties of biochar depend on its physical characteristics, which are influenced by the biomass used as feedstock and the production process. This results in variation among biochar samples. The goal of this study was to assess the functional properties of biochar produced from vine pruning waste, focusing on its chemical composition, antimicrobial activity against Escherichia coli, and prebiotic activity for the Lactobacillus species. The extract was rich in components with antimicrobial and antioxidant properties, showing significant inhibitory activity against E. coli, a causative agent of enteric disease in young pigs. This inhibitory activity led to a downregulation of genes involved in quorum sensing (biofilm formation and cellular division). Additionally, the biochar extract demonstrated potential species-specific prebiotic activity. The aim of this study was to characterize the metabolomic and functional properties of an aqueous biochar extract from vine pruning waste, in order to predict its potential in vivo effects as a functional feed ingredient. A metabolomic analysis of the biochar extracts was conducted using quadrupole time-f-light (QQTOF) high-performance liquid chromatography tandem mass spectrometry (HPLC MS/MS). Antimicrobial activity against E. coli F18+ and E. coli F4+ was assessed using standard growth inhibition assays, while quorum sensing in E. coli exposed to biochar extracts was evaluated using real-time PCR. Prebiotic activity was assessed by exposing selected Lactobacillus strains to the biochar extract, monitoring growth patterns to determine species-specific responses. The metabolomic profile revealed several distinct molecular classes, including multiple peaks for phenolic compounds. The extract significantly inhibited the growth of both E. coli pathotypes, reducing growth by 29% and 16% for the F4+ and F18+, respectively (p < 0.001). The relative expression of the genes involved in quorum sensing (MotA, FliA for biofilm formation, and FtsE, HflX for cell division) indicated that the observed inhibitory effects likely resulted from interference with flagellar synthesis, motility, and reduced cell division. The biochar extract also showed species-specific prebiotic potential. In conclusion, biochar derived from vine pruning waste represents a valuable feed ingredient with functional properties that may help to reduce antibiotic use in livestock production. The optimization of artificial pollination is crucial in breeding programs for allogamous plants. In maize, achieving a balance between the labor-intensive nature of controlled pollinations and the need for large-scale production of hybrid seeds, along with considerations of germinability and pollen production, determines the success of genetic improvement programs. Breeding programs in maize have resulted in a reduction in the number of tassel branches to increase light interception and plant density in production fields. However, despite this genetic improvement, the decreased pollen production per plant has raised critical concerns regarding pollination and subsequent ear filling, especially under adverse environmental conditions. The aim of this work was the analysis of factors that can contribute to increasing the efficiency of controlled pollination in maize. The data obtained showed that pollen diameter, flavonoid, and phenolic acid content do not influence the percentage of germination and therefore the efficiency of pollination. The quantity of pollen is a central factor in ensuring the efficiency of controlled pollinations, and the data obtained by comparing traditional varieties with modern hybrids of maize showed that an increase in pollen production is determined by the increase in branching of the male inflorescence. Maize (Zea mays L.) is a vital crop susceptible to Gibberella ear rot (GER), a disease caused by Fusarium graminearum, resulting in significant yield losses and mycotoxin production. This study aimed to investigate the correlation between ear characteristics and GER resistance in 74 maize inbred lines (42 with non-stiff stalks and 32 stiff stalks) adapted to the northern Italian environment. Mycotoxin analysis was performed to assess the presence of deoxynivalenol (DON) and zearalenone (ZEA). The results showed a positive correlation between the husk traits, like the husk number and husk cover, and GER resistance in both heterotic groups. A positive correlation was also found between the DON and ZEA concentrations. In addition, we conducted a genome-wide association study (GWAS) which identified novel quantitative trait loci (QTLs) associated with the husk number, husk cover, ear attitude, and infection score. These QTLs can be utilized in marker-assisted selection for breeding new GER-resistant maize varieties. Our study provides valuable insights into the genetic basis of ear traits and their relationship with GER resistance, which can contribute to an improvement in the environmental and economical sustainability of the corn growing system. In recent years, the interest in increasingly sustainable agriculture has also turned attention towards new cover crops suitable for use in marginal areas that could enter the food chain as new protein and oil sources or for biodiesel production. In this scenario, Camelina sativa is a perfect crop to study. Camelina is an annual herbaceous plant belonging to the Brassicaceae which is interesting in terms of its oil content, since the seeds contain about 40% oils, with a high level of polyunsaturated fatty acids (30–40% alpha linolenic acid, 15–25% linoleic acid, 15% oleic acid and about 15% eicosenoic acid). It is a hexaploid species (2n = 40, genome size ~782 Mb) characterized by rapid growth, a short life cycle (85–100 days for spring varieties, 190–210 for autumn varieties) and low input cultivation needs. However, its use in feed and food is limited by the presence of glucosinolates (GLS). GLS are sulfur molecules involved in plant defense. In recent years, they have been studied not only as antinutritionals but also for their anti-carcinogenic effects against chronic inflammatory and heart diseases and for their use as natural pesticides. Given the recent interest in camelina and its highly nutritious oil, eight pure lines and a synthetic population were compared in two different growing periods, spring and winter. In this work, the genetic materials were characterized for different phenotypic traits, yields and yield components, and bromatological and glucosinolate content. The results confirmed that in North Italy, camelina has higher yields if cultivated in the autumn–winter period (about 2 t/ha vs. 0.6 t/ha); furthermore, a negative correlation was found between spring and winter yields, indicating that varieties that produce more in winter cultivation produce less in spring cultivation. Moreover, to our knowledge, it is the first work in which a synthetic population of Camelina sativa has been tested and proved to be a valid solution for use in various environments both for its adaptability and for the low content of glucosinolates (about 17 mmol/kg).
