Tracking visual objects while stabilizing the visual background is complicated by the different computational requirements for object tracking and motion vision. In fruit fly, directionally selective motion detectors T4 and T5 cells supply wide-field neurons of the lobula plate, which control smooth gaze stabilization behavior. Here, we hypothesized that an anatomically parallel pathway supplied by T3, which encodes small moving objects and innervates the lobula, drives body saccades toward objects. We combined physiological and behavioral experiments to show that T3 neurons respond omnidirectionally to contrast changes induced by the visual stimuli that elicit tracking saccades, and silencing T3 reduced the frequency of tracking saccades. By contrast, optogenetic manipulation of T3 increased the number of tracking saccades. Our results represent the first evidence that parallel motion detection and feature detection pathways coordinate smooth gaze stabilization and saccadic object tracking behavior during flight.

The mechanism of action selection is a widely shared fundamental process required by animals to interact with the environment and adapt to it. A key step in this process is the filtering of the “distracting” sensory inputs that may disturb action selection. Because it has been suggested that, in principle, action selection may also be processed by shared circuits in vertebrate and invertebrates, we wondered whether invertebrates show the ability to filter out “distracting” stimuli during a goal-directed action, as seen in vertebrates. In this experiment, action selection was studied in wild-type Drosophila melanogaster by investigating their reaction to the abrupt appearance of a visual distractor during an ongoing locomotor action directed to a visual target. We found that when the distractor was present, flies tended to shift the original trajectory toward it, thus acknowledging its presence, but they did not fully commit to it, suggesting that an inhibition process took place to continue the unfolding of the planned goal-directed action. To some extent flies appeared to take into account and represent motorically the distractor, but they did not engage in a complete change of their initial motor program in favor of the distractor. These results provide interesting insights into the selection-for-action mechanism, in a context requiring action-centered attention, that might have appeared rather early in the course of evolution. NEW & NOTEWORTHY Action selection and maintenance of a goal-directed action require animals to ignore irrelevant “distracting” stimuli that might elicit alternative motor programs. In this study we observed, in Drosophila melanogaster, a top-down mechanism inhibiting the response toward salient stimuli, to accomplish a goal-directed action. These data highlight, for the first time in an invertebrate organism, that the action-based attention shown by higher organisms, such as humans and nonhuman primates, might have an ancestral origin.

Individuals vary in how their circadian system synchronizes with the cyclic environment (zeitgeber). Assessing these differences in “phase of entrainment”—often referred to as chronotype—is an important procedure in laboratory experiments and epidemiological studies but is also increasingly applied in circadian medicine, both in diagnosis and therapy. While biochemical measurements (e.g., dim-light melatonin onset [DLMO]) of internal time are still the gold standard, they are laborious, expensive, and mostly rely on special conditions (e.g., dim light). Chronotype estimation in the form of questionnaires is useful in approximating the timing of an individual’s circadian clock. They are simple, inexpensive, and location independent (e.g., administrable on- and offline) and can therefore be easily administered to many individuals. The Munich ChronoType Questionnaire (MCTQ) is an established instrument to assess chronotype by asking subjects about their sleep-wake-behavior. Here we present a shortened version of the MCTQ, the µMCTQ, for use in situations in which instrument length is critical, such as in large cohort studies. The µMCTQ contains only the core chronotype module of the standard MCTQ (stdMCTQ), which was shortened and adapted from 17 to 6 essential questions, allowing for a quick assessment of chronotype and other related parameters such as social jetlag and sleep duration. µMCTQ results correspond well to the ones collected by the stdMCTQ and are externally validated by actimetry and DLMO, assessed at home (no measure of compliance). In the last few years several studies have investigated the neural mechanisms underlying spatial orientation in Drosophila melanogaster. Convergent results suggest that this mechanism is associated with specific neural circuits located within the Central Complex (CC). Furthermore such circuits appear to be associated with visual attention, specifically with selective attention processes implicated in the control of action. Our aim was to understand how wild-type flies react to the abrupt appearance of a visual distractor during an ongoing locomotor action. Thus, we adapted the well-known ‘Buridan paradigm’, used to study walking behaviour in flies, so we could specifically address the mechanisms involved in action selection. We found that flies tended to react in one of two ways when confronted with a visual distractor during ongoing locomotion. Flies either: (i) committed to a new path situated midway between the original target and the distractor, consistent with a novelty effect; or (ii) remained on the original trajectory with a slight deviation in direction of the distractor. We believe that these results provide the first indication of how flies react, from the motor point of view, in a bi-stable context requiring the presence of selection-for-action mechanisms. Some considerations on the neural circuits underlying such behavioural responses are advanced.

Daylight saving time (DST) is a source of circadian disruption impinging on millions of people every year. Our aim was to assess modifications, if any, in the number, type, and outcome of Accident & Emergency (A&E) visits/return visits over the DST months. The study included 366,527 visits and 84,380 return visits to the A&E of Padova hospital (Northern Italy) over 3 periods between the years 2007 and 2016: period 1 (2 weeks prior to DST to 19 weeks after), period 2 (2 weeks prior to the return to “winter time” to 4 weeks after), and period 3 (5 consecutive non-DST weeks). For each A&E visit/return visit, information was obtained on triage severity code, main medical complaint, and outcome. Data were aggregated by day, cumulated over the years, and analyzed by generalized Poisson models. Generalized additive models for Poisson data were then used to include photoperiod as an additional covariate. An increase in A&E visits and return visits (mostly white codes, resulting in discharges) was observed a few weeks after the enforcement of DST and was significant over most weeks of period 1 (increase of ≈30 [2.8%] visits and ≈25 [10%] return visits per week per year). After the return to winter time, a decrease in absolute number of return visits was observed (mostly white codes, resulting in discharges), which was significant at weeks 3 and 4 of period 2 (decrease of ≅25 [10%] return visits per week per year). When photoperiod was taken into account, changes in A&E visits (and related white codes/discharges) were no longer significant, while changes in return visits (and related white codes/discharges) were still significant. In conclusion, changes in A&E visits/return visits were observed in relation to both DST and photoperiod, which are worthy of further study and could lead to modifications in A&E organization/staffing.

In humans, memory consolidation can be aided by the representation of an odor previously associated with target information during sleep. In an elegant study, Zwaka et al. ( Curr Biol 25: 2869–2874, 2015) have demonstrated that the same process occurs in honeybees, suggesting that the relationship between sleep and memory may be similar across different animal species.
