Abstract

Research was conducted to gain an insight into the novel character of a new approach to laser dynamics. The method goes beyond the usual Maxwell-Bloch (MB) equations in that it takes fully into consideration the phase of the matter quantum field that describes the atomic systems. Results of experiments conducted on the effects of a dephasing of the electromagnetic laser field were in very good agreement with its expectations although they could not be explained within the MB expectations.

Abstract

Flurbiprofen, a nonsteroidal antiinflammatory drug (NSAID), has been recently described to selectively inhibit β-amyloid1-42 (Aβ42) secretion, the most toxic component of the senile plaques present in the brain of Alzheimer patients. The use of this NSAID in Alzheimer's disease (AD) is hampered by a significant gastrointestinal toxicity associated with cyclooxygenase (COX) inhibition. New flurbiprofen analogues were synthesized, with the aim of increasing Aβ42 inhibitory potency while removing anti-COX activity. In vitro ADME developability parameters were taken into account in order to identify optimized compounds at an early stage of the project. Appropriate substitution patterns at the alpha position of flurbiprofen allowed for the complete removal of anti-COX activity, while modifications at the terminal phenyl ring resulted in increased inhibitory potency on Aβ42 secretion. In rats, some of the compounds appeared to be well absorbed after oral administration and to penetrate into the central nervous system. Studies in a transgenic mice model of AD showed that selected compounds significantly decreased plasma Aβ42 concentrations. These new flurbiprofen analogues represent potential drug candidates to be developed for the treatment of AD.

21. Mild Wolf–Hirschhorn phenotype and partial gh deficiency in a patient with a 4p terminal deletion

Abstract

Wolf–Hirschhorn syndrome (WHS) is caused by a variably-sized deletion of chromosome 4 involving band 4p16 whose typical craniofacial features are “Greek warrior helmet appearance” of the nose, microcephaly, and prominent glabella. Almost all patients show mental retardation and pre- and post-natal growth delay. Patient was born at term, after a pregnancy characterized by intra-uterine growth retardation (IUGR). Delivery was uneventful. Developmental delay was evident since the first months of life. At 2 years, he developed generalized tonic-clonic seizures. Because of short stature, low growth velocity and delayed bone age, at 4 years he underwent growth hormone (GH) evaluation. Peak GH after two provocative tests revealed a partial GH deficiency. Clinical observation at 7 years disclosed a distinctive facial appearance, with microcephaly, prominent eyes, and beaked nose. Brain MRI showed left temporal mesial sclerosis. GTG banded karyotype was normal. Because of mental retardation, subtelomeric fluorescence in situ hybridization (FISH) analysis was performed, disclosing a relatively large deletion involving 4p16.2 → pter (about 4.5 Mb), in the proband, not present in the parents. The smallest deletion detected in a WHS patient thus far includes two candidate genes, WHSC1 and WHSC2. Interestingly, that patient did not show shortness of stature, and that could be due to the haploinsufficiency of other genes localized in the flanking regions. Contribution of GH alterations and possible GH therapy should be further considered in WHS patients. © 2004 Wiley-Liss, Inc.

23. Dystrophinopathy in a young boy with Klinefelter's syndrome

Abstract

We report the first case of a child with mild dystrophinopathy associated with Klinefelter's syndrome (karyotype 47, XXY). This 3.5-year-old boy was affected by some symptoms suggestive of Becker's muscular dystrophy. Dystrophin immunostaining and immunoblotting procedures confirmed the diagnosis, but polymerase chain reaction-directed gene analysis failed to reveal any macrodeletion. Methylation-based assay did not show preferential X-inactivation. This confirmed the coexistence of the two active X-chromosomes (one of which was of paternal origin), thus accounting for the mild form of dystrophinopathy in this child affected by Klinefelter's syndrome. © 1998 John Wiley & Sons, Inc. 24. Long-term follow-up of some early-treated aminoacidopathies associated with ocular abnormalities

Abstract

Experience in treating some aminoacidopathies associated with ocular abnormalities is reported. Sixteen out of 18 homocystinuric patients were treated with pyridoxine (300-1200 mg/day): 14 showed complete responsiveness, one was partially responsive, and one was a non-responder. In two early-diagnosed B6-responsive children treatment did not stop the progression of ocular abnormalities already present at the time of diagnosis. In one patient, completely asymptomatic at two years of age, a 5 1/2-year treatment with pyridoxine apparently prevented the appearance of either ectopia lentis or myopia. In a ten-year-old-girl affected by hyperornithinemia with gyrate atrophy of the choroid and retina, dietary treatment with a low-protein diet (0.8 g/kg per day) for more than six years reduced her plasma ornithine levels from 1189 to 774 μmol/1 and stopped the progression of the chorioretinal lesions. A short trial of pyridoxine given during the dietary treatment did not lead to a further reduction of her plasma ornithine concentrations. Four children with Lowe's syndrome underwent early surgical treatment, three for cataracts and glaucoma, and one for cataract only. Unfortunately, the final visual outcome was poor, as a result of the appearance of corneal dystrophy after the age of three to six years. Surgery, however, is still to be recommended for its positive effect on development and behavior.

Abstract

Unlabelled: Obesity is a typical example of a complex multifactorial disease arising from behavioural, environmental and genetic factors that may affect individual responses to dietary intake and physical activity. Observational, longitudinal dietary interventional studies in obese patients present contrasting reports on the predictive value of baseline leptin levels. We report on the effect of a weight reduction programme in three different groups of obese children (82 patients in all) assembled on the basis of their baseline leptin levels adjusted for body mass index (BMI), gender and pubertal development. The effectiveness of this programme was decreased in patients with relative hyperleptinaemia or hypoleptinaemia compared to children with baseline leptin levels appropriate to BMI gender and pubertal development., Conclusion: Information gained from leptin assays could provide predictive insight into an individual's ability to lose body fat and may therefore have important implications for our approach to the treatment and prevention of childhood obesity.
