Abstract

Abstract Drought stress is a prevalent environmental factor that results in significant yield losses for maize (Zea mays L.). Genetic improvements through modern breeding methods and the introduction of biotechnology‐derived traits have been used to improve tolerance to drought stress. Conducting yield efficacy trials in the field for plants that exhibit tolerance to abiotic stress poses several challenges, including the inability to control multiple unpredictable environmental stressors. Controlled environment conditions were tested to evaluate yield parameters commonly observed in the field for MON 87460 maize expressing the cold shock protein B (CspB) protein. Maize plants subjected to water‐limitation treatment in the controlled environment exhibited phenotypic characteristics of drought stress consistent with those observed under field conditions. MON 87460 exhibited significant relative increases in ear length, kernel number, and grain weight following water‐limitation treatment. These results demonstrate that the controlled environment is an alternate option for evaluating drought tolerance in maize.

Abstract

Task-free functional connectivity in animal models provides an experimental framework to examine connectivity phenomena under controlled conditions and allows for comparisons with data modalities collected under invasive or terminal procedures. Currently, animal acquisitions are performed with varying protocols and analyses that hamper result comparison and integration. Here we introduce StandardRat, a consensus rat functional magnetic resonance imaging acquisition protocol tested across 20 centers. To develop this protocol with optimized acquisition and processing parameters, we initially aggregated 65 functional imaging datasets acquired from rats across 46 centers. We developed a reproducible pipeline for analyzing rat data acquired with diverse protocols and determined experimental and processing parameters associated with the robust detection of functional connectivity across centers. We show that the standardized protocol enhances biologically plausible functional connectivity patterns relative to previous acquisitions. The protocol and processing pipeline described here is openly shared with the neuroimaging community to promote interoperability and cooperation toward tackling the most important challenges in neuroscience. The authors pooled resources to identify best practices and develop a new standardized protocol for estimating functional connectivity in rats with magnetic resonance imaging.

Contains fulltext : 291641.pdf (Publisher’s version ) (Open Access), Tourette Syndrome (TS) is a complex neurodevelopmental disorder characterized by vocal and motor tics lasting more than a year. It is highly polygenic in nature with both rare and common previously associated variants. Epidemiological studies have shown TS to be correlated with other phenotypes, but large-scale phenome wide analyses in biobank level data have not been performed to date. In this study, we used the summary statistics from the latest meta-analysis of TS to calculate the polygenic risk score (PRS) of individuals in the UK Biobank data and applied a Phenome Wide Association Study (PheWAS) approach to determine the association of disease risk with a wide range of phenotypes. A total of 57 traits were found to be significantly associated with TS polygenic risk, including multiple psychosocial factors and mental health conditions such as anxiety disorder and depression. Additional associations were observed with complex non-psychiatric disorders such as Type 2 diabetes, heart palpitations, and respiratory conditions. Cross-disorder comparisons of phenotypic associations with genetic risk for other childhood-onset disorders (e.g.: attention deficit hyperactivity disorder [ADHD], autism spectrum disorder [ASD], and obsessive-compulsive disorder [OCD]) indicated an overlap in associations between TS and these disorders. ADHD and ASD had a similar direction of effect with TS while OCD had an opposite direction of effect for all traits except mental health factors. Sex-specific PheWAS analysis identified differences in the associations with TS genetic risk between males and females. Type 2 diabetes and heart palpitations were significantly associated with TS risk in males but not in females, whereas diseases of the respiratory system were associated with TS risk in females but not in males. This analysis provides further evidence of shared genetic and phenotypic architecture of different complex disorders.

Abstract

The commercialization of RNA-based agricultural products requires robust ecological risk assessments. Ecological risk is operationally defined as a function of exposure and adverse effects. Information on the environmental fate of RNA-based plant-incorporated protectants is essential to define routes and duration of exposure to potentially sensitive nontarget organisms. Providing these details in problem formulation helps focus the ecological risk assessment on the relevant species of concern. Postharvest plant residue is often considered to be the most significant route of exposure for genetically modified crops to adjacent aquatic environments. Previous studies have shown that DvSnf7 RNA from SmartStax PRO maize dissipates rapidly in both terrestrial and aquatic environments. Although these studies suggest that direct exposure to DvSnf7 RNA is likely to be low, little is known regarding the fate of DvSnf7 RNA produced in plants after entering an aquatic environment. This exposure scenario is relevant to detritivorous aquatic invertebrates that process conditioned maize tissues that enter aquatic environments. To assess potential exposure to shredders, dissipation of DvSnf7 RNA expressed maize tissue was evaluated following immersion in microcosms containing sediment and water. Concentrations of DvSnf7 RNA in the tissue were measured over a duration of 21 d. The DvSnf7 RNA dissipated rapidly from immersed maize tissue and was undetectable in the tissues after 3 d. Concentrations of DvSnf7 RNA found in tissue as well as calculated water column concentrations were below levels known to elicit effects in a highly sensitive surrogate species, supporting the conclusion of minimal risk to aquatic nontarget organisms. Abstract

Carnosine and related β-alanine–containing peptides are believed to be important antioxidants, pH buffers, and neuromodulators. However, their biosynthetic routes and therapeutic potential are still being debated. This study describes the first animal model lacking the enzyme glutamic acid decarboxylase–like 1 (GADL1). We show that Gadl1−/− mice are deficient in β-alanine, carnosine, and anserine, particularly in the olfactory bulb, cerebral cortex, and skeletal muscle. Gadl1−/− mice also exhibited decreased anxiety, increased levels of oxidative stress markers, alterations in energy and lipid metabolism, and age-related changes. Examination of the GADL1 active site indicated that the enzyme may have multiple physiological substrates, including aspartate and cysteine sulfinic acid. Human genetic studies show strong associations of the GADL1 locus with plasma levels of carnosine, subjective well-being, and muscle strength. Together, this shows the multifaceted and organ-specific roles of carnosine peptides and establishes Gadl1 knockout mice as a versatile model to explore carnosine biology and its therapeutic potential.

Abstract

The mismatch repair gene MSH3 has been implicated as a genetic modifier of the CAG·CTG repeat expansion disorders Huntington's disease and myotonic dystrophy type 1. A recent Huntington's disease genome-wide association study found rs557874766, an imputed single nucleotide polymorphism located within a polymorphic 9 bp tandem repeat in MSH3/DHFR, as the variant most significantly associated with progression in Huntington's disease. Using Illumina sequencing in Huntington's disease and myotonic dystrophy type 1 subjects, we show that rs557874766 is an alignment artefact, the minor allele for which corresponds to a three-repeat allele in MSH3 exon 1 that is associated with a reduced rate of somatic CAG·CTG expansion (P = 0.004) and delayed disease onset (P = 0.003) in both Huntington's disease and myotonic dystrophy type 1, and slower progression (P = 3.86 × 10-7) in Huntington's disease. RNA-Seq of whole blood in the Huntington's disease subjects found that repeat variants are associated with MSH3 and DHFR expression. A transcriptome-wide association study in the Huntington's disease cohort found increased MSH3 and DHFR expression are associated with disease progression. These results suggest that variation in the MSH3 exon 1 repeat region influences somatic expansion and disease phenotype in Huntington's disease and myotonic dystrophy type 1, and suggests a common DNA repair mechanism operates in both repeat expansion diseases.

Abstract

Previous research has suggested that robot-mediated therapy is effective in the treatment of children with Autism Spectrum Disorder (ASD), but not all robots seem equally appropriate for this purpose. We investigated in an exploratory study whether a robot’s intonation (monotonous vs. normal) and bodily appearance (mechanical vs. humanized) influence the treatment outcomes of Pivotal Response Treatment (PRT) sessions for children with ASD. The children (age range 4–8 years) played puzzle games with a robot which required communication with the robot. The treatment outcomes were measured in terms of both task performance and affective states. We have found that intonation and bodily appearance have an effect on children’s affective states but not on task performance. Specifically, humanized bodily appearance leads to more positive affective states in general and a higher degree of interest in the interaction than mechanical bodily appearance. Congruence between bodily appearance and intonation triggers a higher degree of happiness in children with ASD than incongruence between these two factors.

Abstract

As the leading cause of morbidity and mortality of cystic fibrosis (CF) patients, early detection of Pseudomonas aeruginosa (PA) is critical in the clinical management of this pathogen. Herein, we describe rapid and sensitive electroanalytical methods using differential pulse voltammetry (DPV) at a boron-doped diamond (BDD) electrode for the detection of PA signaling biomolecules. Monitoring the production of key signaling molecules in bacterial cultures of P. aeruginosa PA14 over 8 h is described, involving sample pretreatment by liquid-liquid and solid-phase extraction. In addition, direct electrochemical detection approach of PA signaling molecules is also reported in conjunction with hexadecyltrimethylammonium bromide (CTAB) to disrupt the bacterial membrane.

Abstract

A simple and robust chemosensing approach using a boron-doped diamond (BDD) electrode has been developed and applied to analyze tryptophan (TRP) and indole during the growth of Escherichia coli in a complex growth medium. The bacterial enzyme tryptophanase catalyzes TRP to indole, an emerging signaling molecule. The process can now be monitored using electrochemistry, in a method far beyond the traditional identification protocols. Electroanalysis in a non-aqueous medium comprising acetonitrile (ACN) and tetrabutylammonium hexafluorophosphate (TBAH) is capable of separating the oxidation peak of TRP from that of indole. Mechanisms are postulated for the electrochemical oxidation of indole and TRP in ACN chosen because of its wider potential range, proton acceptor property, and solubilization of analytes. The electrochemical oxidation of TRP involves the elimination of two electrons. With a detection limit of 0.5 µM for both indole and TRP, this chemosensing approach is sufficient to monitor the level of these two biomolecules during the bacterial growth period.

Abstract

Rapid detection of pathogenic bacteria present in patient samples is of utmost importance for the clinical management of bacterial-induced diseases. Herein, we describe an efficient and direct electrochemical approach for the detection of 2-heptyl-3-hydroxy-4-quinolone (PQS), 2-heptyl-4-hydroxyquinoline (HHQ), and pyocyanin (PYO) as a molecular signature of Pseudomonas aeruginosa (PA), a frequently infecting pathogen with high antibiotic resistance. The cationic surfactant hexadecyltrimethylammonium bromide (CTAB) enhances the effectiveness of an unmodified thin-film boron-doped diamond (BDD) electrode for the direct detection of PYO, HHQ, and PQS in bacterial cultures of PAO1 and PA14. Differential pulse voltammetry (DPV) is then used to monitor the production of these microbial metabolites in bacterial cultures of PAO1 over 10 h without any sample pretreatment. A proposed mechanism for the interaction of CTAB with bacteria cells is examined by zeta (?) potential measurements. Furthermore, the detection method is successfully extended to a clinical fluid matrix and applied to PA spiked cystic fibrosis (CF) sputum samples.
