Parkinson's Disease (PD) is associated with motor and non-motor symptoms. Among the latter are deficits in matching, identification, and recognition of emotional facial expressions. On one hand, this deficit has been attributed to a dysfunction in emotion processing. Another explanation (which does not exclude the former) links this deficit with reduced facial expressiveness in these patients, which prevents them from properly understanding or embodying emotions. To disentangle the specific contribution of emotion comprehension and that of facial expression processing in PD's observed deficit with emotions we performed two experiments on non-emotional facial expressions. In Experiment 1, a group of PD patients and a group of Healthy Controls (HC) underwent a task of non-emotional expression recognition in faces of different identity and a task of identity recognition in faces with different expression. No differences were observed between the two groups in accuracies. In Experiment 2, PD patients and Healthy Controls underwent a task where they had to recognize the identity of faces encoded through a non-emotional facial expression, through a rigid head movement, or as neutral. Again, no group differences were observed. In none of the two experiments hypomimia scores had a specific effect on expression processing. We conclude that in PD patients the observed impairment with emotional expressions is likely due to a specific deficit for emotions to a greater extent than for facial expressivity processing.

Objectives: The high incidence of prostate cancer causes prostatic samples to significantly affect pathology laboratories workflow and turnaround times (TATs). Whole-slide imaging (WSI) and artificial intelligence (AI) have both gained approval for primary diagnosis in prostate pathology, providing physicians with novel tools for their daily routine. Methods: A systematic review according to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines was carried out in electronic databases to gather the available evidence on the application of AI-based algorithms to prostate cancer. Results: Of 6290 articles, 80 were included, mostly (59%) dealing with biopsy specimens. Glass slides were digitized to WSI in most studies (89%), roughly two-thirds of which (66%) exploited convolutional neural networks for computational analysis. The algorithms achieved good to excellent results about cancer detection and grading, along with significantly reduced TATs. Furthermore, several studies showed a relevant correlation between AI-identified histologic features and prognostic predictive variables such as biochemical recurrence, extraprostatic extension, perineural invasion, and disease-free survival. Conclusions: The published evidence suggests that AI can be reliably used for prostate cancer detection and grading, assisting pathologists in the time-consuming screening of slides. Further technologic improvement would help widening AI's adoption in prostate pathology, as well as expanding its prognostic predictive potential.

Current models propose that facial recognition is mediated by two independent yet interacting anatomo-functional systems: one processing facial features mainly mediated by the Fusiform Face Area and the other involved in the extraction of dynamic information from faces, subserved by Superior Temporal Sulcus (STS). Also, the pre-Supplementary Motor Area (pre-SMA) is implicated in facial expression processing as it is involved in its motor mimicry. However, the literature only shows evidence of the implication of STS and preSMA for facial expression recognition, without relating it to face recognition. In addition, the literature shows a facilitatory role of facial motion in the recognition of unfamiliar faces, particularly for poor recognizers. The present study aimed at studying the role of STS and preSMA in unfamiliar face recognition in people with different face recognition skills. 34 healthy participants received repetitive transcranial magnetic stimulation over the right posterior STS, pre-SMA and as sham during a task of matching of faces encoded through: facial expression, rigid head movement or as static (i.e., absence of any facial or head motion). All faces were represented without emotional content. Results indicate that STS has a direct role in recognizing identities through rigid head movement and an indirect role in facial expression processing. This dissociation represents a step forward with respect to current face processing models suggesting that different types of motion involve separate brain and cognitive processes. PreSMA interacts with face recognition skills, increasing the performance of poor recognizers and decreasing that of good recognizers in all presentation conditions. Together, the results suggest the use of at least partially different mechanisms for face recognition in poor and good recognizers and a different role of STS and preSMA in face recognition.

The goal of this preregistered scoping review is to create an overview of the research on developmental prosopagnosia (DP). Through analysis of all empirical studies of DP in adults, we investigate 1) how DP is conceptualized and defined, 2) how individuals are classified with DP and 3) which aspects of DP are investigated in the literature. We reviewed 224 peer-reviewed studies of DP. Our analysis of the literature reveals that while DP is predominantly defined as a lifelong face recognition impairment in the absence of acquired brain injury and intellectual/cognitive problems, there is far from consensus on the specifics of the definition with some studies emphasizing e.g., deficits in face perception, discrimination and/or matching as core characteristics of DP. These differences in DP definitions is further reflected in the vast heterogeneity in classification procedures. Only about half of the included studies explicitly state how they classify individuals with DP, and these studies adopt 40 different assessment tools. The two most frequently studied aspects of DP are the role of holistic processing and the specificity of face processing, and alongside a substantial body of neuroimaging studies of DP, this paints a picture of a research field whose scientific interests and aims are rooted in cognitive neuropsychology and neuroscience. We argue that these roots – alongside the heterogeneity in DP definition and classification – may have limited the scope and interest of DP research unnecessarily, and we point to new avenues of research for the field.

Introduction. The surface protein TROP-2/TACSTD2 and the cell adhesion protein NECTIN-4/NECTIN4 are responsible for the efficacy of anticancer therapies based on antibody- drug conjugates (ADC) targeting intracellular microtubules. In contrast with common histologic subtypes of bladder urothelial carcinoma (BUC), little is known of TROP-2 and NECTIN-4 expression in sarcomatoid and rhabdoid BUC. Aims. In this study, we aimed to analyze TROP-2 and NECTIN-4 expression and additional predictive biomarkers by immunohistochemistry and fluorescence in situ hybridization (FISH) on 35 undifferentiated BUC (28 sarcomatoid and 7 rhabdoid). Wide genomic investigation was also performed on 411 BUC cases of the PanCancer Atlas, focusing on genes related to the microtubule pathways. Results. Seven of 35 (20%) undifferentiated BUC showed expression of TROP-2. NECTIN-4 was expressed in 10 cases (29%). Seven cases (20%) co-expressed TROP-2 and NECTIN-4. HER-2 FISH was amplified in 5 cases (14%) while HER-2 immunoexpression was observed in 14 cases (40%). PD-L1 scored positive for combined proportion score (CPS) in 66% of cases and for tumor proportion score (TPS) in 51% of cases. Pan-NTRK1-2/3 was elevated in 9 cases (26%) and FGFR-2/3 was broken in 7 of 35 cases (20%). Of 28 sarcomatoid BUC, 9 (32%) were negative for all (TROP-2, NECTIN-4, PD-L1, HER-2, FGFR and pan-NTRK) biomarkers and 3 (11%) expressed all five biomarkers. Among cases with rhabdoid dedifferentiation, 1 of 7 (14%) showed activation of all biomarkers, whereas 2 of 7 (28%) showed none. The mRNA analysis identified microtubulerelated genes and pathways suitable for combined ADC treatments in BUC. Conclusion. Sarcomatoid and rhabdoid BUC do harbor positive expression of the ADC targets TROP-2 or NECTIN-4 in a relatively modest subset of cases, whereas the majority do not. La prosopagnosia è un disturbo diffuso sia nella sua forma congenita che acquisita. Tuttavia, non esistono ancora protocolli unitari per la sua riabilitazione. Di consequenza, nello studio 1 è stata svolta una revisione sistematica della letteratura per avere una panoramica sugli studi esistenti sulla riabilitazione dell’agnosia per gli oggetti e della prosopagnosia. I risultati riguardanti la prosopagnosia mostrano che trattamenti basati su un’elaborazione olistica dei volti sono più efficaci e vengono generalizzati a nuove prospettive dei volti. Tuttavia, esistono pochi articoli sull’argomento ed è necessario sviluppare nuovi studi. Questo potrebbe essere dovuto alla poca chiarezza sui meccanismi di elaborazione dei volti. Per questo motivo, abbiamo deciso di approfondirli. Innanzitutto, abbiamo indagato la relazione tra espressioni facciali e riconoscimento di identità. L’obiettivo dello studio 2 era quello di indagare se c’è una relazione tra le abilità di riconoscimento individuali e l’elaborazione di espressioni facciali. Gli attuali modelli di riconoscimento volti postulano che elaboriamo i volti tramite due sistemi separati in interazione: uno elabora le caratteristiche del volto e l’altro le espressioni facciali. Di conseguenza, nello studio 2 ci aspettavamo di osservare una facilitazione delle espressioni sul riconoscimento di identità. I risultati confermano che espressioni facciali dinamiche non emotive aiutano nel riconoscimento di identità, ma solamente in chi ha delle scarse abilità di riconoscimento volti. L’obiettivo dello studio 3 era quello di approfondire le basi neurali di questa relazione. Per farlo, abbiamo stimolato STS, preSMA e un’area Sham con la TMS mentre i partecipanti dovevano riconoscere volti codificati tramite espressioni facciali dinamiche non emotive, un movimento rigido o come neutri. I risultati ottenuti nella condizione sham replicano quelli ottenuti nello studio 2: espressioni facciali non emotive aiutano il riconosc, Prosopagnosia is a diffuse deficit both in its acquired and developmental forms. However, no unitary protocols for its rehabilitation have been developed yet. Thus, a systematic review of the literature on the topic (Study 1) has been performed to understand the state of the art of rehabilitation of object agnosia and prosopagnosia. Results regarding developmental and acquired prosopagnosia, reveal that treatments involving holistic perceptual processing of faces are the most effective and can be generalized to new views and perspectives of faces. However, few papers were obtained and new studies addressing the topic are required. This might be due to the scarce clarity about the mechanisms subserving face processing. Thus, we decided to deepen our understanding of the mechanisms subserving face recognition. First, we wanted to shed light on the relationship between facial expression and identity processing. The aim of study 2 was that of assessing whether there is a relation between individual face recognition abilities and facial expression processing. Indeed, according to models of face recognition, we recognize faces through two partially independent systems which process respectively, facial features and facial expressions. Based on that, we expected expressions, even when non-emotional to aid identity recognition. Results of study 2 showed that when the system used to elaborate facial identity is deficient, dynamic facial expressions, even when non-emotional, help recognition. Study 3 aimed at deepening our understanding of the neural bases subserving the relation between facial expression and identity processing. To do so, we stimulated STS, preSMA and a Sham area with repetitive TMS while administering a task of identity matching of faces encoded through non-emotional facial expressions, rigid head movement or as neutral. In the spectrum of oncocytic renal neoplasms, a subset of tumors with high-grade-appearing histologic features harboring pathogenic mutations in mammalian target of rapamycin (mTOR) and hitherto clinical indolent behavior has been described. Three cases (2F,1 M) with histologically documented metastases (lymph node, skull, and liver) were retrieved and extensively investigated by immunohistochemistry, FISH, and next-generation sequencing. Tumors were composed of eosinophilic cells with prominent nucleoli (G3 by ISUP/WHO) arranged in solid to nested architecture. Additionally, there were larger cells with perinuclear cytoplasmic shrinkage and sparse basophilic Nissl-like granules, superficially resembling the so-called spider cells of cardiac rhabdomyomas. The renal tumors, including the skull and liver metastases, showed immunoexpression PAX8, CK8-18, and cathepsin-K, and negativity for vimentin. NGS identified mTOR genetic alterations in the three cases, including the skull and liver metastases. One patient was then treated with Everolimus (mTOR inhibitors) with clinical response (metastatic tumor shrinkage). We present a distinct renal tumor characterized by high-grade eosinophilic cells, cathepsin-K immunohistochemical expression, and harboring mTOR gene mutations demonstrating a malignant potential and showing responsiveness to mTOR inhibitors.

Several neuroimaging studies have shown that a distributed network of brain regions is involved in our ability to appraise the emotions we experience in daily life. In particular, scholars suggested that the dorsal anterior cingulate cortex (dACC) may play a role in the appraisal of emotional stimuli together with subcortical regions, especially when stimuli are negatively valenced, and the dorsolateral prefrontal cortex (dlPFC) may play a role in regulating emotions. However, proofs of the causal role of these regions are lacking. In the present study, we aim at testing this model by stimulating both the dACC and the left dlPFC via cathodal tDCS. Twenty-four participants were asked to attend and rate the arousal and valence of negative and neutral emotional stimuli (pictures and words) in three different experimental sessions: cathodal stimulation of dACC, left dlPFC, or sham. In addition to the experimental task, the baseline affective state was measured before and after the stimulation to further assess the effect of stimulation over the baseline affective state after the experimental session. Results showed that cathodal stimulation of dACC, but not the left dlPFC, was associated with reduced arousal ratings of emotional stimuli, both compared with the sham condition. Moreover, cathodal stimulation of left dlPFC decreased participant’s positive affective state after the session. These findings suggest for the first time, a dissociation between the dACC and dlPFC, with the former more involved in emotion appraisal, and the latter more involved in mood modulation.

Background:Agnosia for objects is often overlooked in neuropsychology, especially with respect to rehabilitation. Prosopagnosia has been studied more extensively, yet there have been few attempts at training it. The lack of training protocols may partially be accounted for by their relatively low incidence and specificity to sensory modality. However, finding effective rehabilitations for such deficits may help to reduce their impact on the social and psychological functioning of individuals. Objective:Our aim in this study was to provide clinicians and researchers with useful information with which to conduct new studies on the rehabilitation of object agnosia and prosopagnosia. To accomplish this, we performed a systematic and comprehensive review of the effect of neuropsychological rehabilitation on visual object and prosopagnosia. Methods:The Preferred Reporting Items for Systematic reviews and Meta-Analyses guidelines were followed. In addition, the Single-Case Experimental Design (SCED) and the Critical Appraisal Skills Programme (CASP) scales were used to assess the quality of reporting. Results:Seven articles regarding object agnosia, eight articles describing treatments for prosopagnosia, and two articles describing treatments for both deficits were included. Conclusions:In the light of the studies reviewed, treatments based on analysis of parts seem effective for object agnosia, while prosopagnosia appears to benefit most from treatments relying on holistic/configural processing. However, more attempts at rehabilitation of face and object agnosia are needed to clarify the mechanisms of these processes and possible rehabilitations. Moreover, a publication bias could mask a broader attempt to find effective treatments for visual agnosia and leaving out studies that are potentially more informative.
