As diidropirimidinonas (DHPMs) são consideradas estruturas privilegiadas, termo que caracteriza os esqueletos moleculares capazes de interagir com vários alvos biológicos. Diferentes mecanismos de ação para DHPMs com atividade citotóxica têm sido investigados. Um dos alvos amplamente estudados para este grupo de compostos, consiste na inibição da cinesina Eg5, a qual está envolvida na separação dos cromossomos duplicados durante a interfase da mitose. O monastrol foi a primeira DHPM identificada com atividade sobre a Eg5. Outro potencial alvo apontado para as DHPMs é a enzima ecto-5’-nucleotidase (ecto-5’-NT), envolvida no controle dos níveis extracelulares de adenosina, metabólito que pode atuar como um fator para o crescimento tumoral. O papel da ecto-5’-NT sobre a resistência de gliomas a antineoplásicos é reportado na literatura. Este trabalho tem como suporte recentes estudos in silico realizados pelo nosso grupo de pesquisa, os quais apontaram características estruturais de DHPMs capazes de potencializar sua interação com a ecto-5’-NT ou com a Eg5. Nesse sentido, o presente estudo teve por objetivo explorar a geração de diversidade estrutural nessa classe de heterociclos para encontrar DHPMs com potencial atividade sobre a proliferação de linhagens tumorais, atuando sobre a ecto-5’-NT ou a Eg5. Para este objetivo, foram sintetizadas 38 DHPMs com diferentes modificações estruturais, envolvendo principalmente: (i) DHPMs N1-arilas empregando diferentes compostos 1,3-dicarbonílicos na reação de Biginelli e (ii) DPHMs com modificações no anel aromático ligado ao carbono C4. Ainda tendo como alvo a inibição da ecto-5’-NT foram obtidas acil-selenoureias como produtos de simplificação molecular de DHPMs N1-arilas. Além disso, foram realizados estudos usando diferentes experimentos de RMN, HPLC e métodos in sílico para comprovar pela primeira vez a existência de atropoisomerismo nas DHPMs N1-arilas com substituintes na posição orto do anel aromático. A presença de um substituinte na posição orto do anel aromático ligado ao N1 produz limitada rotação na ligação C-N, gerando um eixo de quiralidade. As DHPMs sintetizadas LaSOM 299, LaSOM 302 e LaSOM 307 apresentaram atividade antiproliferativa superior ao monastrol nas linhagens de câncer de bexiga T24 (LaSOM 299: IC50 18,52 μM) e de glioma U138 (LaSOM 299: IC50 63,50 μM; LaSOM 302: IC50 68,03 μM; LaSOM 307: IC50 64,76 μM) e produziram fenótipo monoastral em células de glioma, indicando atividade sobre a Eg5. Estudos realizados in vivo com Caenorhabditis elegans para LaSOM 301, LaSOM 309, LaSOM 299, LaSOM 302 e LaSOM 307 mostraram que são compostos seguros e ainda, que o LaSOM 307 inibiu a produção de espécies reativas de oxigênio. Os resultados reportados aqui permitiram aumentar a diversidade química das DHPMs, explorando modificações em diferentes posições do núcleo da DHPM. Dihydropyrimidinones (DHPMs) are privileged structures, a term that characterizes the molecular scaffolds able to interact with different biological targets. Numerous mechanisms of action for DHPMs derivatives with cytotoxic activity against cancer cell lines have been investigated. Among the potential targets widely investigated for this group of molecules is the kinesin-5 (Eg5), which is involved in the duplicated chromosomes separation during the interphases of mitosis. Monastrol was the first DHPM with an effect on Eg5. Another possible target of DHPMs is the enzyme ecto-5'-nucleotidase (ecto-5'-NT), involved in the control of extracellular adenosine levels, a metabolite that can act as a growth factor in cancer. The role of ecto-5'-NT in the resistance of gliomas against antineoplastic drugs is reported. This work has as supporting recent studies in silico performed by our research group, which showed structural features of DHPMs able to improve its interaction with the ecto-5'-NT or with the Eg5. In this context, the present study aims to explore the structural diversity of this heterocycle class to find DHPMs with biological activity on the cancer cell proliferation acting on ecto-5'-NT or Eg5. For this purpose, were synthesized 38 DHPMs with different structural modifications, involving mainly: (i) DHPMs N1-aryl substituted by using of different carbonyl compounds in the Biginelli reaction; and (ii) DHPMs with modifications in aromatic ring linked to the C4 carbon. Were obtained acyl-selenoureas as N1-aryl substituted DHPMs molecular simplification products targeting the ecto-5’-NT. Also, were performed studies using NMR, HPLC and in silico approaches aiming to demonstrate by the first time the presence of atropoisomerism in the DHPMs N1 ortho aryl-substituted. The presence of a substituent at the ortho position of aromatic ring linked to the N1, produced limited rotation in the C-N bond and a chirality axis. The N1-aryl substituted DHPMs LaSOM 299, LaSOM 302 and LaSOM 307 showed higher antiproliferative effect than monastrol in urinary bladder T24 cancer (LaSOM 299 IC50 18.52 μM) and glioma U138 (LaSOM 299 IC50 63.50 μM; LaSOM 302 IC50 68.03 μM; LaSOM 307 IC50 64.76 μM) and produced monoastral phenotype, suggesting its effect on Eg5. In vivo studies with Caenorhabditis elegans for LaSOM 301, LaSOM 309, LaSOM 299, LaSOM 302 and LaSOM 307 showed that they are safe compounds and that LaSOM 307 inhibited the production of reactive oxygen species. These novel DHPMs were safe in studies using the nematode Caenorhabditis elegans. The results reported here allowed to improve the chemical diversity in DHPMs, exploring structural modifications in different positions of this heterocycle.

An original and focused library of two sets of dihydropyrimidin-2-thiones (DHPMs) substituted with N-1 aryl groups derived from monastrol was designed and synthesized in order to discover a more effective Eg5 ligand than the template. Based on molecular docking studies, four ligands were selected to perform pharmacological investigations against two glioma cell lines. The results led to the discovery of two original compounds, called 20h and 20e, with an anti-proliferative effects, achieving IC(50) values of about half that of the IC(50) of monastrol in both cell lines. As with monastrol, flow cytometry analyses showed that the 20e and 20h compounds induced cell cycle arrest in the G(2)/M phase, and immunocytochemistry essays revealed the formation of monopolar spindles due to Eg5 inhibition without any toxicity to Caenorhabditis elegans.

Introdução: A partir da descoberta da reação de Biginelli, diidropirimidinonas/tionas funcionalizadas têm se destacado como protótipos no desenvolvimento de fármacos, considerando diferentes alvos. O primeiro representante deste grupo de moléculas, com ação inibitória sobre a divisão celular, foi o monastrol, o qual é caracterizado como um inibidor alostérico da cinesina mitótica 5 (Eg5). Este mecanismo de ação representa um alvo promissor para a terapia antineoplásica, considerando que a Eg5 é uma proteína motora envolvida no movimento dos microtúbulos durante a divisão celular. Recentes investigações mostraram que a inserção de um anel aromático no N-1 do núcleo diidrotiopirimidínico seria capaz de otimizar a interação do ligante com a Eg5. Objetivos: Nesse sentido, a presente pesquisa teve por objetivo obter diidropirimidin-2-tionas N-1 aril substituídas, e avaliar seu potencial antiproliferativo em células de glioma. Métodos: Foi sintetizada uma quimioteca de 26 diidropirimidin-2-tionas N-aril substituídas, através da reação de Biginelli promovida por cloreto de trimetilsilano, utilizando uma série de ariltioureias previamente obtidas. A partir da análise do docking destes compostos com a Eg5, foram selecionados 4 deles para avaliação da atividade citotóxica em linhagem celular de glioma, utilizando o ensaio MTS. Resultados: Os derivados do monastrol substituídos em N-1 com 4NO2-Ar (LaSOM 301) e 4OMe-Ar (LaSOM 309) foram mais ativos que o monastrol na linhagem celular investigada. Enquanto o monastrol apresentou valor de IC50 superior à 100 μM, os compostos LaSOM 301 e LaSOM 309 apresentaram valores de IC50 de respectivamente 54,69 ± 4,92 μM e 57,74 ± 2,78 μM. Adicionalmente, o composto LaSOM 308, embora isento da hidroxila em meta, presente no monastrol, e N-1 substituído com 4OMe-Ar apresentou valor de IC50 inferior ao monastrol (78,26 ± 4,18 μM). Conclusões: As condições reacionais utilizadas, permitiram a funcionalização do N-1 de diidropirimidin- 2-tionas com diferentes substituintes. A inserção de um anel aromático substituído no N-1, do núcleo diidrotiopirimidínico consistiu em uma modificação estrutural capaz de gerar compostos com atividade antineoplásica, superior ao monastrol. Introduction: Since the Biginelli reaction discovery, functionalized dihydropyrimidinones/ thiones have been emerged as prototypes for drug design in different targets. Monastrol was the first representative molecule of this group with inhibitory effect on cell division, which produces kinesin-5 (Eg5) inhibition. This action of mechanism is a promising target for anticancer therapy, whereas the motor protein Eg5 is involved in microtubule movement during the cell division. Recent results showed that the aromatic ring at N-1 position of dihydrothiopyrimidinic core would be able to optimize the interaction of the ligand with the Eg5. Objectives: In this context, this study aimed to obtain dihydropyrimidin-2-thiones N-1 aryl substituted, and to evaluate their antiproliferative activity on glioma cells. Methods: A chemical library of 26 dihydropyrimidin-2-thiones N-1 aryl substituted were synthetized employing the Biginelli reaction promoted by trimethylchlorosilane. Considering the results of docking analysis of these compounds with Eg5, four of them were selected for cytotoxic activity assessment on glioma cells, employing MTS assay. Results: The monastrol derivatives N-1 substituted with 4NO2-Ph (LaSOM 301) and 4OMe-Ph (LaSOM 309) showed higher activity than monastrol, in cell line studied. While monastrol showed IC50 value higher than 100 μM, LaSOM 301 and LaSOM 309 compounds showed IC50 values of 54.69 ± 4.92 μM and 57.74 ± 2.78 μM, respectively. Nevertheless, the LaSOM 309 compound, without hydroxyl group present in monastrol, and with N-1 4OMe-Ph substituted also showed IC50 value lower than monastrol (78.26 ± 4.18 μM). Conclusions: Reactional conditions employed in these research, allowed the N-1 functionalization of dihydropyrimidin-2-thiones, with several substituents. The insertion of a substituted aromatic ring at N-1 position, of dihydrothiopyrimidinic core, was a structural change able to generate compounds with antineoplasic activity, higher than monastrol.

Introduction: Candidemia is a bloodstream infection produced by Candida genus yeasts. Objective: The purpose of this study was to characterize the epidemiology and the fluconazole susceptibility in Candida species isolated from patients at a regional hospital in Passo Fundo, RS. Methods: Records from the laboratory were used to identify patients with positive blood cultures for Candida between 2010 and 2011. The in vitro activity of fluconazole was determined using the disk diffusion method. Results: Were analyzed 24 positive blood cultures for Candida and found a 54.16% mortality rate. C. albicans was the most prevalent species, followed by C. parapsilosis and C. krusei. For susceptibility to fluconazole, C. albicans, C. parapsilosis and C. tropicalis showed 100% sensitivity. However, C. krusei was 100% resistant; and C. glabrata, 50% resistant. Conclusion: The high mortality and fluconazole resistance rates emphasize the importance of the diagnosis of candidemia in a hospital environment. RESUMO Introdução: Candidemia é a infecção da corrente sanguínea produzida por leveduras do gênero Candida. Objetivos: Este estudo teve por objetivo caracterizar a epidemiologia e o perfil de suscetibilidade ao fluconazol em espécies de Candida, isoladas em pacientes internados em um hospital regional de Passo Fundo, RS. Métodos: Registros laboratoriais foram utilizados para identificar pacientes com hemocultura positiva para espécies do gênero Candida, entre 2010 e 2011. A atividade in vitro do fluconazol foi determinada por meio do método de difusão em disco. Resultados: Foram analisadas 24 hemoculturas positivas para Candida; a taxa de mortalidade encontrada foi 54,16%. C. albicans foi a espécie mais prevalente, seguida por C. parapsilosis e C. krusei. C. albicans, C. parapsilosis e C. tropicalis apresentaram 100% de sensibilidade ao fluconazol, entretanto C. krusei mostrou-se 100% resistente; e C. glabrata, 50% resistente. Conclusão: A elevada taxa de mortalidade e resistência ao fluconazol enfatiza a importância do diagnóstico de candidemia em ambiente hospitalar.

Nontuberculous mycobacteria are etiologic agents of opportunistic human infections. Although they usually affect superficial tissues, infections in bones and joints have been described. The contamination is associated with increased environmental exposure. With appropriate therapy, the cases usually progress to complete recovery of the patient. This study reports the case of a patient who developed a cutaneous nodule in her left foot acquired when her skin was punctured by a fish. The anatomopathological examination revealed chronic central suppurative granulomatous dermo-hypodermal inflammation. Furthermore, the screening for resistant acid-fast bacilli was positive. Micobactérias não tuberculosas são agentes causadores de infecções oportunistas no homem. Embora, geralmente, afetem tecidos superficiais, infecções em ossos e articulações têm sido descritas. O contágio está associado ao aumento da exposição do homem ao meio ambiente. Diante da terapêutica adequada, os casos normalmente evoluem com recuperação total do indivíduo. Este estudo descreve o caso clínico de um paciente que apresentou nódulo cutâneo no pé esquerdo após traumatismo com um peixe. O exame anatomopatológico revelou inflamação crônica granulomatosa centralmente supurativa dermo-hipodérmica e a pesquisa de bacilos álcool-ácido resistentes (BAAR) foi positiva.
