16. Investigation and characterization of Aliarcobacterspp. isolated from cattle slaughterhouse in Türkiye

Abstract

Aliarcobacterspp. have been isolated from numerous food products at retail and from animal carcasses and feces at slaughter. The objectives of this study were as follows: (i) to isolate Aliarcobacter species from different slaughterhouses’ samples and (ii) to detect genetic diversity, antibiotic resistance, biofilm ability, and putative virulence gene profiles of the isolates. A molecular investigation of antibiotic resistance and virulence factors was also conducted using polymerase chain reaction (PCR). Among 150 samples, a total of 22 (14.6%) Aliarcobacterspp. isolates were obtained, with varying levels of antibiotic resistance observed. The genes tetO, tetW, and gyrAwere detected in 0%, 31.8%, and 27.2% of the isolates, respectively. All isolates were resistant to ampicillin, rifampin, and erythromycin, while tetracycline was found to be the most effective antibiotic, with 81.8% of the isolates showing susceptibility to it. All isolates (100%) harbored more than one of the nine putative virulence genes tested, with 18.1% of isolates carrying more than three. Regarding biofilm formation, 7 (31.8%) and 4 (18.1%) isolates were found to form strong and moderate biofilms, respectively, while one (4.5%) isolate was classified as a weak biofilm producer. ERIC-PCR band patterns suggested that the isolated Aliarcobacterspp. from slaughterhouses had different sources of contamination. These findings highlight the potential risk posed by pathogenic and multidrug-resistant Aliarcobacterspp. in food and the need for control measures throughout the food chain to prevent the spread of these strains. The results indicate that foods of animal origin and cattle slaughterhouses are significant sources of antimicrobial resistant Aliarcobacter.

17. Biogenic synthesis of silver nanoparticles using Sambucus nigraleaves: elucidation, antimicrobial, antioxidant activities and quantification of phenolics

Abstract

Biogenic production of silver nanoparticles (sn-AgNPs) was accomplished by Sambucus nigra. The identification of structures was performed by a wide spectroscopic study. Ultraviolet–visible (UV–Vis) spectroscopic study indicated the maximum absorption of nanoparticles at 480 nm. Scanning electron microscopy (SEM) presented the nanoparticles as spherical shapes. X-ray diffraction (XRD) signals (2θ) at the degrees of 38.20°, 44.40°, 64.61°, 77.61°, and 81.76° corresponded to [111, 020, 202, 131, and 222] facet, respectively, confirming the nanostructure as face-centered cubic structure. The size of the nanoparticles was determined as 28.6 nm and 27.4 nm by SEM and TEM images, respectively. Zeta potential presented the stability of the product (− 51.3 mV). Furthermore, the histogram of dynamic light scattering (DLS) displayed the particle size of the nanostructure as 31.01 nm. HPLC analysis was used to determine the phenolics in S. nigraaerial part quantitatively and chlorogenic acid was the main product (10.76 mg/g extract). sn-AgNPs showed the substantial antioxidant effect as 12.1 ± 0.19 (µg/mL, lC50) and 3.1 ± 0.1 (lC50) for DPPH•and ABTS•+assays, respectively. Antibacterial activity of extract and sn-AgNPs was executed. Both extract and Sn-AgNPs exhibited considerable activity.

24. Meta-Analysis of the Global Prevalence of Enterococcusspp. in Foods: Antibiotic Resistance Profile of Enterococcus faecalisand Enterococcus faecium

Abstract

The objective of this study was to evaluate the prevalence and diversity of Enterococcusspp. and antibiotic-resistant Enterococcus faecalisand Enterococcus faeciumisolates in different foods worldwide. This study used meta-analytical methods. Besides, Web of Science (n= 705), Medline (n= 6), and Scopus (n= 1338) were searched for studies in the years 1995–2021 using related keywords. Results showed that the pooled prevalence for Enterococcusspp. and antibiotic-resistance of E. faecalisand E. faeciumisolates were 0.41 (95% C.I. 0.34–0.47), 0.25 (95% C.I. 0.13–0.38), respectively. According to the results of the subgroup analysis, the lowest and highest prevalence of Enterococcusspp. in food types were calculated for red meat (0.56), and fermented foods (0.29). Also, as a result of subgroup analyses by country the highest prevalence of Enterococcusspp. was calculated in studies conducted in Slovakia (0.74). In contrast, the lowest prevalence was calculated in studies conducted in Georgia (0.07). The meta-analyses improved our understanding of the prevalence of Enterococcusspp. and the antibiotic resistance of E. faecalisand E. faeciumisolates in different foods and provided results that can be useful as input for quantitative microbiological risk evaluation modeling.

28. Prevalence and molecular characterization of Listeria monocytogenes isolated from wastewater of cattle slaughterhouses in Turkey.

Abstract

Aim: The study aimed to investigate the role of cattle slaughterhouse wastewater as a possible source for the environmental distribution of Listeria monocytogenes. Methods and Results: Listeria spp. isolation was performed by collecting 117 wastewater samples from four different cattle slaughterhouses in Turkey. Species‐specific identification was performed biochemically, and L. monocytogenes isolates were confirmed with polymerase chain reaction (PCR). In all, 71 (62.2%) of the wastewater samples were found to be positive for Listeria spp., and 17 (14.9%) of these samples were contaminated with L. monocytogenes. Pulsed field gel electrophoresis (PFGE) analysis revealed that all L. monocytogenes isolates were of different pulsotypes and isolates belonged to seven different phylogenetic clusters. Multiplex PCR analysis for genoserotypes and lineage determination showed that the isolates were divided into genoserotypes IVb and IIc in Lineages I and II. Also, it has been investigated with SYBR‐Green Real‐time PCR whether the L. monocytogenes isolates harboured virulence genes (hly, sigB, plcA, plcB, inlA, inlB, inlC and inlJ), and it was found that all isolates were substantially positive. Antibiotic resistance profiles and MIC values of the isolates were determined, and all L. monocytogenes isolates were found susceptible to ampicillin. In contrast, two isolates were resistant to meropenem and erythromycin, and three isolates were resistant to trimethoprim/sulfamethoxazole. Conclusion: L. monocytogenes, which pose a threat to public health and resists to antibiotics effectively used in treatments, can environmentally spread via wastewater of cattle slaughterhouses. The wastewater of the food industry, which has rich microbiota, should be treated carefully, and possible environmental contamination should be prevented. Significance and Impact of Study: This is the first study that investigates the molecular characterization of L. monocytogenes isolated from cattle slaughterhouse wastewater and the findings represent the importance of cattle wastewater in the epidemiology of L. monocytogenes in Turkey. Abstract

Bu çalışma, kımız’dan elde edilen Lactobacillus paraplantarum ve Leuconostoc mesenteroides subs. cremorisbakterilerinin yoğurt üretiminde starter kültür olarak kullanımını incelemek için gerçekleştirilmiştir. Kırgızistan’ın farlıyerlerinden temin edilen 23 adet kımız örneğinden Lactobacillus paraplantarum ve Leuconostoc mesenteroides subs.cremoris standart kültür metotları kullanılarak izole edildi. İdentifikasyon için, matriks ile desteklenmiş lazer desorbsiyon/iyonizasyon kütle spektrometresi (MALDI-TOF MS) yöntemi kullanıldı. Elde edilen kültürler geleneksel yöntemleyoğurt yapımında starter kültür olarak kullanıldı. Üretimi yapılan yoğurt örnekleri ile kontrol olarak kullanılanyoğurt örneği duyusal analiz ile değerlendirildi. İstatistiksel analizde yoğurt örnekleri, kontrol grubu arasında önemli birfark tespit edilemedi. Sonuç olarak, kımızdan elde edilen starter kültürler ile yoğurt üretilebileceği, aynı zamanda eldeedilen kültürlerin farklı süt ürünlerinin üretiminde kullanılmasının mümkün olabileceği ifade edilebilir., The objective of this study is to use Lactobacillus paraplantarum and Leuconostoc mesenteroides subs.cremoris to obtain koumiss and practice them in preparation of yoghurt. Twenty five samples of koumiss bought fromdifferent places of Kyrgyzstan were used to obtain Lactobacillus paraplantarum and Leuconostoc mesenteroides subs.cremoris. Standard microbiological methods were conducted for isolation of starter culture microorganisms. Identificationprocess was performed with characterizing by Matrix-assisted laser desorption/ionization mass spectroscopy(MALDI-TOF MS). The obtained starter culture microorganisms were used in preparation of yoghurt according to thetraditional method of yoghurt production. Produced yoghurt samples and control group were exposed to sensorialanalysis. Sensorial and physical properties of yoghurt prepared by using the isolated strains from koumiss were notfound to be significantly different from commercial yogurt in statistical analyses. In conclusion, it was seen that starterculture obtained from koumiss can be used in production of yoghurt and also received results can be used as a basefor investigations on using culture microorganisms obtained from koumiss in production of different types of dairy products.

42. Prevalence Of ArcobacterSpecies In Drinking Water, Spring Water, And Raw Milk As Determined By Multiplex PCR

Abstract

The aim of this study was to investigate the incidence of Arcobacterspecies in water sources and raw milk from healthy animals in Kayseri, Turkey. A total of 175 samples of drinking water (n= 100), spring water (n= 25), and raw milk (n= 50) were examined. Arcobacterspecies were isolated using the membrane filtration technique. Overall, 7 (4%) of the 175 samples yielded Arcobacterspp.: 3 (3%) drinking water samples, 1 (4%) spring water sample, and 3 (6%) raw milk samples. Two species of Arcobacterwere recovered from the seven positive samples: Arcobacter butzleri, Arcobacter skirrowii,and A. butzleriplus A. skirrowiifound in 3 (1.7%), 2 (1.1%), and 2 (1.1%) samples, respectively. Our study is the first to report the isolation of both A. butzleriand A. skirrowiitogether from drinking water and is the first report of Arcobacterin milk from healthy cows in Turkey. Based on these findings, the presence of Arcobacterspecies in environmental waters and raw milk may pose a potential hazard for human health.

Abstract

WOS: 000399994700021, PubMed ID: 28216007, Objective: The aim of this study is to trace the possible relations between the hygienic status of slicing utensils and the microbiological quality of pastrami. Study Design: A total of 75 pastrami retail markets were visited in Kayseri, Turkey, where the pastrami (a ready-to-eat meat product) is commonly produced and consumed. Sliced pastrami, the cutting board and knife surface swabs were collected from each pastrami retail point to trace possible sources of contamination. Methods: Samples were analysed for the presence of total viable counts (TVC), total coliforms, Escherichia coli, members of Enterobacteriaceae, Staphylococcus aureus and Listeria spp. In addition, pastrami samples were analysed for sulphite-reducing Clostridium spp. and Toxoplasma gondii. Results: When compared with the target values of related literatures, a total of 6 (8%) pastrami samples were found unsatisfactory as a result of TVC (5.3%), Enterobacteriaceae (5.3%), E. coli (2.6%), S. aureus (2.6%), Listeria spp. (2.6%) and Listeria monocytogenes (1.3%) contaminations. No T. gondii positivity was observed among the pastrami samples. None of the cutting board and knife surface swabs were found to harbour TVC level >10(3) cfu/cm(2), E. coli and L. monocytogenes. Abstract

WOS: 000325034500018, PubMed ID: 23809061, The present study was conducted to investigate the presence of Escherichia coli O157:H7 in food samples of animal origin and to detect its virulence genes by immunomagnetic separation technique and multiplex PCR (mPCR). A total of 500 samples (consisting of diced meat, minced meat, burger, raw cow's milk and raw cow's milk cheese) were analysed. Escherichia coli O157:H7 was detected in 5 (1%) of 500 analysed samples including two diced meat, one minced meat and two raw-milk cheese. None of the burger samples tested contained E.coli O157:H7. Three isolates obtained from minced and diced meat were found to carry stx(1), stx(2), hlyA and eaeA genes whereas two isolates from raw-milk cheese were found to harbour the stx(1), eaeA and hlyA genes. The results of this study suggest that raw meat and raw-milk cheese tested could pose public health problems in consumers with regard to their virulence factors.

45. Investigation of Staphylococcus Spp and Coliform Bacteria Contamination Sources after Cleaning-in-Place in Production Lines of Dairy Factories in Türkiye.

Abstract

Microorganisms detected on dairy factory surfaces after disinfection can cause product contamination, leading to economic losses and health hazards for consumers. In this study, the presence of Staphylococcus spp. and Coliform in a total of 450 samples taken from food-contact and non-contact surfaces (stainless steel, plastic, cloth, and tiles surfaces), raw milk and final product (white cheese, kashar cheese, butter, yogurt, and cream) samples in five dairy factories was investigated by conventional techniques. The isolates obtained were identified by matrix-assisted laser desorption/ionization-time of flight mass spectrometry. In this study, a total of 54 Staphylococci (16.7% S. aureus and 81.5% coagulase-negative Staphylococci [CNS]) and 44 coliform isolates were identified at the species level. The most common CNS isolated by samples was S. epidermidis followed by S. saprophyticus, S. capitis, S. succinus S. carnosus . S. xylosus, S. sciuri, S. equorum, S. warneri, and S. hominis . Eighteen of the coliform isolates (41%) were identified as E. coli ; 13 (29.5%) as E. cloacae ; 3 (6.9%) as E. kobei, C. freundii, and K. oxytoca ; 2 (4.5%) as K. pneumoniae ; 1 (2.3%) as E. ludwigii and C. farmeri . The contamination rate of non-food contact surfaces (71.3%) was found to be higher than food contact surfaces (10.4%), and contaminated surfaces were found to be effective in product contamination. Study results show that some bacterial species obtained from raw milk, surfaces, and final products are factory specific and surface-associated bacteria are prominent in the product microbial profile.
