The IDEA Challenge 2022 prototyping dataset comprises a total of 240 prototype entries with 1049 edges (connections) and can provide valuable insights into prototyping practices, offering practical knowledge essential for developing prototyping strategies and generating hypotheses for future studies. Data were collected using Pro2booth - an online platform which captured comprehensive information about prototypes and participating teams' development process, including details about the creators, purpose, timing, and methods of creation. It is particularly relevant to design researchers, engineering and design students, educators, and industry professionals seeking to enhance their prototyping skills and strategies. It serves as a robust foundation for subsequent studies, allowing for comparative analyses, hypothesis verification, and trend exploration. A 2B adenosine receptor (A 2B AR) antagonists have therapeutic potential in inflammation-related diseases such as asthma, chronic obstructive pulmonary disease and cancer. However, no drug is currently clinically approved, creating a demand for research on novel antagonists. Over the last decade, the study of target binding kinetics, along with affinity and potency, has been proven valuable in early drug discovery stages, as it is associated with improved in vivo drug efficacy and safety. In this study, we report the synthesis and biological evaluation of a series of xanthine derivatives as A 2B AR antagonists, including an isothiocyanate derivative designed to bind covalently to the receptor. All 28 final compounds were assessed in radioligand binding experiments, to evaluate their affinity and for those qualifying, kinetic binding parameters. Both structure-affinity and structure-kinetic relationships were derived, providing a clear relationship between affinity and dissociation rate constants. Two structurally similar compounds, 17 and 18, were further evaluated in a label-free assay due to their divergent kinetic profiles. An extended cellular response was associated with long A 2B AR residence times. This link between a ligand's A 2B AR residence time and its functional effect highlights the importance of binding kinetics as a selection parameter in the early stages of drug discovery., (Copyright © 2022 The Author(s). COVID-19 and global crises/events are driving governments to rethink their national manufacturing strategies. The drastic change of societal conditions has exposed our reliance on a constrained set of production practices. Furthermore, the future manufacturing landscape indicates - supply chain crises, trade agreements and natural disasters - a high level of volatility which requires a response that is far from being achieved. While these emergent challenges have called the efficacy of established practices into question, new manufacturing technologies, such as Additive Manufacturing (AM), present the capability to provide a solution. One proposal is agent-based brokering of AM which could be a method for tackling local, regional, national, and international production needs. However, to achieve the reality of brokered AM, it is imperative that the diversity of AM capability is considered. Diversity that existing homogeneous modelling of AM and manufacturing systems rarely consider or capture. This paper conceptualizes the reality of AM systems and elucidates parameters that are necessary for successful modelling and subsequent co-ordination. The Electronic Control Device (ECD) will be used by the primary police force in the Netherlands. Hence medical personnel will be confronted with persons that have received ECD shocks more often. In light of these developments, it is important that care providers are aware of potential medical consequences resulting from the use of electric stun guns. The darts usually result in minor injury with small penetration wounds requiring minimal treatment. However, in vulnerable areas, such as the eyes, the darts can cause serious injury and specialist care is indicated. The electric shock causes muscle contractions, potentially resulting in traumatic falls, or fractures. Cardiac problems occur only in exceptional cases; risk factors include long duration of the power surge, short distance from the darts to the heart and underlying heart problems. In rare cases a pneumothorax may occur. Finally, often there are underlying medical problems requiring appropriate treatment such as drug intoxication, excited delirium or psychiatric disorders. Systematic recording of the medical problems caused by anECD is indicated.
