Detta arbete kommer vara inriktat på personers upplevelser om skrik i ett survival horror-spel. Bakgrunden går igenom vad skräck och rädsla är, ljudets betydelse i spel och skräckspel, och sist om människan och vår inställning till röster och specifikt skrik. Frågeställningen som utgår från bakgrunden är följande: Upplevs skrik som hotfullt eller hotat beroende på om det är kvinnligt eller manligt i ett survival horror-spel? Projektbeskrivningen går igenom artefakt som är tillhörande denna undersökning, som består av en video med skrik och enkel ambiens. Artefakten delades tillsammans med en enkät på olika forum och fick 16 svar. Resultatet från detta visar att både kvinnliga och manliga skrik kan upplevas som både hotfull och som hotade, det verkar mer bero på vad medverkande har för tidigare erfarenheter. Framtida arbete diskuteras mot slutet av denna studie, bland annat att det går att utveckla till att skriva om skrik som specifikt vapen inom spel/media. Det finns övrigt digitalt material (t.ex. film-, bild- eller ljudfiler) eller modeller/artefakter tillhörande examensarbetet som ska skickas till arkivet.

A compound, which is a selective peroxisome proliferator activated receptor (PPAR) agonist, was investigated. The aim of the presented studies was to evaluate the potential of the further development of the compound. Fundamental physicochemical properties and stability of the compound were characterized in solution by liquid chromatography and NMR and in solid-state by various techniques. The drug itself is a lipophilic acid with tendency to form aggregates in solution. The neutral form was only obtained in amorphous form with a glass-transition temperature of approximately 0°C. The intrinsic solubility at room temperature was determined to 0.03mg/mL. Chemical stability studies of the compound in aqueous solutions showed good stability for at least two weeks at room temperature, except at pH1, where a slight degradation was already observed after one day. The chemical stability in the amorphous solid-state was investigated during a period of three months. At 25°C/60% relative humidity (RH) and 40°C/75% RH no significant degradation was observed. At 80°C, however, some degradation was observed after four weeks and approximately 3% after three months. In an accelerated photostability study, degradation of approximately 4% was observed. Attempts to identify a crystalline form of the neutral compound were unsuccessful, however, salt formation with tert-butylamine, resulted in crystalline material. Results from stability tests of the presented crystalline salt form indicated improved chemical stability at conditions whereas the amorphous neutral form degraded. However, the salt form of the drug dissociated under certain conditions. The drug was administered both per oral and intravenously, as amorphous nanoparticles, to conscious dogs. Plasma profiles showed curves with secondary absorption peaks, indicating hepatic recirculation following both administration routes. A similar behavior was observed in rats after oral administration of a pH-adjusted solution. The observed double peaks in plasma exposure and the dissociation tendency of the salt form, were properties that contributed to make further development of the candidate drug challenging. Options for development of solid dosage forms of both amorphous and crystalline material of the compound are discussed.

One central aim of the present work was to find a robust oral formulation approach for Compound A, both to achieve reliable pharmacodynamic read outs but also for long time safety assessment studies. The compound has low aqueous solubility (0.4μM at 37°C), is highly lipophilic and has high Caco-2 permeability, i.e. a typical BCS II compound. A nanocrystal formulation, some oil approaches and a fat diet approach were evaluated in vivo in rats. The two latter strategies resulted in significantly higher in vivo exposures after oral administration compared to the nanocrystal approach. For simplicity, and due to the project development program, a food pellet formulation was selected. In addition, tentative data from a subcutaneous study in mice using nanocrystals of the compound are presented, showing extended profiles on the cost of Cmax. Exposure data in monkeys after administration of nanocrystals both intravenously and per oral are presented. When switched from nanocrystals to an oil formulation, the observed oral exposure behavior was similar as observed in rats.

Bakgrund: Akut postoperativ smärta är en vanlig men också förväntad konsekvens efter kirurgiska ingrepp som ger negativ påverkan på den efterföljande postoperativa återhämtningen. Syfte: Syftet med studien var att undersöka genomsnittlig självskattad smärtintensitet som gav vuxna patienter problem med att vila, sova och röra sig under det första postoperativa dygnet samt om det hos män/kvinnor, yngre/äldre och kirurgi/ortopedpatienter förekom olika hanterbara smärtnivåer i förhållande till vila och aktivitet under tidig postoperativ återhämtning. Metod: En kvantitativ empirisk deskriptiv studie genomfördes av ett datamaterial från 479 elektiva kirurg- och ortopedpatienter. Resultat: Resultatet visade att ortopedpatienterna i genomsnitt skattade högre värden av smärta jämfört med kirurgpatienterna men att kirurgpatienterna redan vid låga smärtnivåer fick problem med vila, sömn och rörelse. Slutsats: Resultatet understryker att smärta är en subjektiv upplevelse och att smärtbehandlingen i hög grad behöver individualiseras för att därigenom minska smärtans negativa effekter, förbättra den tidiga postoperativa återhämtningen och därmed öka patientsäkerheten. Background: Acute postoperative pain is a common but also expected consequence after surgery that gives negative impact on the following postoperative recovery. Study objectives: The purpose was to investigate the average level of self-assessed intensity of pain that gave adult patients problem to rest, sleep or mobilize during the first postoperative day and if men/women, younger/older and surgical or orthopedic patients had different manageable levels of pain related to rest, sleep and mobilization during the early postoperative recovery. Design: A quantitative empirical descriptive study was performed from datasets from 479 elective surgical and orthopedic patients. Results: The result showed that the orthopedic patients self-assessed higher levels of postoperative pain than the surgical patients, but also that the surgical patients earlier got problems with recovery at low pain levels. Conclusions: The result underlines that pain is a subjective experience and that the pain treatment has to be individualized just to prevent the negative impact of pain but also to enhance the postoperative recovery and improve patient safety.

Denna uppsats utgör en explorativ fallstudie om certifieringssystemet BREEAM Communities och dess möjlighet att fungera som ett verktyg för svenska kommuner i plan- och exploateringsprocesser. Sedan ett par år tillbaka pågår de första projekten i Sverige där BREEAM Communities används. Företagen Diligentia och PEAB var först ut att använda verktyget vid utvecklingen av sina stadsdelar Masthusen och Varvsstaden i Västra Hamnen i Malmö. Uppsatsen utgörs i första hand av en kvalitativ intervjustudie med anställda vid Diligentia, PEAB och Malmö Stad, vilken genomfördes våren 2012. Studien kompletterades med två intervjuer med anställda vid Norrköpings kommun, detta eftersom Norrköpings kommun vid tidpunkten för intervjuerna, övervägde en användning av certifieringssystemet vid utvecklingen av stadsdelen Saltängen. Intervjustudien tar utgångspunkt i intervjupersonernas erfarenheter och tankar kring BREEAM Communities’ inverkan på arbetsprocessen och vilka hållbarhetsaspekter som hanteras inom de två projekten i Västra Hamnen. Därefter diskuteras om och hur en kommun skulle kunna använda verktyget i stadsutvecklingsprojekt. I Västra Hamnen kom initiativet att använda BREEAM Communities från de privata markägarna. Dessa uppger att verktyget fungerat som ett stöd i hanteringen av hållbarhetsaspekter i de pågående projekten. Uppsatsen pekar på att BREEAM Communities också kan bidra med fördelar i kommunala plan- och exploateringsprocesser. Vid dessa processer har verktyget potential att fungera som ett stöd och ge inspiration för att integrera hållbarhetsaspekter. Verktygets tillämpbarhet har påverkats av att det är ett relativt nytt verktyg som utvecklas kontinuerligt. Under hösten 2012 lanserades en omarbetad version av verktyget med ett nytt och förenklat upplägg. Utveckling av BREEAM Communities och det faktum att Sweden Green Building Council avser att svenskanpassa och översätta manualen kommer underlätta användningen framöver, oavsett om verktyget används i syfte att certifiera ett stadsutvecklingsprojekt eller endast som stöd och inspiration. The study aims to explore BREEAM Communities’ potential to be used as a tool within municipal planning in Sweden. Since a few years BREEAM Communities is being applied in two development projects in the Western Harbour in Malmö. The use of the tool was initiated by the private landowners Diligentia and PEAB for the development of Masthusen and Varvstaden. By qualitative interviews with employees at Diligentia, PEAB and Malmö Municipality the experiences of using the tool are being explored. The interviews took place during the spring 2012 and focus on BREEAM Communities’ impacts on the process and the sustainability issues being addressed within the projects, followed by a discussion on the possibilities for a municipality to use BREEAM Communities. Additional interviews with civil servants at Norrköping Municipality gave supplementary perspectives on the possibilities to use BREEAM Communities as a tool within urban planning. Likewise the private landowners express that BREEEAM Communities has been a tool for addressing and integrating sustainability aspects in the development of Masthusen and Varvsstaden, this study indicates that BREEAM Communities has potential to benefit in municipal urban planning. The tool has potential to provide support and inspiration to integrate sustainability aspects from an early stage throughout the process. BREEAM Communities is a rather new tool, and it is continuously developing which has impacts on its applicability. In 2012 a new, simplified and more streamlined scheme was launched. The development of the scheme and the fact that Sweden Green Building Council will make a Swedish adjustment and translation of BREEAM Communities the will simplify the use of tool significantly.

Immune responses were assessed at the single-cell level in lymph nodes from children with tuberculous lymphadenitis. Tuberculosis infection was associated with tissue remodeling of lymph nodes as well as altered cellular composition. Granulomas were significantly enriched with CD68+ macrophages expressing the M. tuberculosis complex-specific protein antigen MPT64 and inducible nitric oxide synthase. There was a significant increase in CD8+ cytolytic T cells surrounding the granuloma; however, CD8+ T cells expressed low levels of the cytolytic and antimicrobial effector molecules perforin and granulysin in the granulomatous lesions. Quantitative real-time mRNA analysis revealed that interferon-gamma, tumor necrosis factor-alpha, and interleukin-17 were not up-regulated in infected lymph nodes, but there was a significant induction of both transforming growth factor-beta and interleukin-13. In addition, granulomas contained an increased number of CD4+FoxP3+ T cells co-expressing the immunoregulatory cytotoxic T-lymphocyte antigen-4 and glucocorticoid-induced tumor necrosis factor receptor molecules. Low numbers of CD8+ T cells in the lesions correlated with high levels of transforming growth factor-beta and FoxP3+ regulatory T cells, suggesting active immunosuppression at the local infection site. Compartmentalization and skewing of the immune response toward a regulatory phenotype may result in an uncoordinated effector T-cell response that reduces granule-mediated killing of M. tuberculosis-infected cells and subsequent disease control.
