Abstract The common bean (Phaseolus vulgaris L.) is a crucial legume crop and an ideal evolutionary model to study adaptive diversity in wild and domesticated populations. Here, we present a common bean pan-genome based on five high-quality genomes and whole-genome reads representing 339 genotypes. It reveals ~234 Mb of additional sequences containing 6,905 protein-coding genes missing from the reference, constituting 49% of all presence/absence variants (PAVs). More non-synonymous mutations are found in PAVs than core genes, probably reflecting the lower effective population size of PAVs and fitness advantages due to the purging effect of gene loss. Our results suggest pan-genome shrinkage occurred during wild range expansion. Selection signatures provide evidence that partial or complete gene loss was a key adaptive genetic change in common bean populations with major implications for plant adaptation. The pan-genome is a valuable resource for food legume research and breeding for climate change mitigation and sustainable agriculture.

Sea turtles are threatened by factors associated with overfishing. To identify these factors, we investigated the fishing pressure and the ethnozoological knowledge of turtles along the Benin coast. A questionnaire-based field survey was undertaken in 48 villages on the Atlantic coast through structured interviews. In total, 479 fishermen and 15 traditional healers were interviewed between July and August, 2020. The Use Value in terms of the types of utilisation of the body parts of the turtles, and the frequency of citation, were calculated. More than eighty percent of the interviewees confirmed the use of turtles for food, medicine, and artisanal purposes. Communities demonstrated nonspecific preference for the four-known species – leatherback turtle, Olive Ridley turtle, green turtle, or hawksbill turtle --within the research area. The flesh is the most used body part (Use Value = 1.5 for food). Body parts of turtles such as the fat, bones etc. are used to treat several diseases, particularly asthma, fever, hypertension, cough, colds, epilepsy, brittle bones in children, and paralysis. The test used shows that the Use Values of sea turtles vary significantly at the 5 % threshold from one sociolinguistic group to another. Our research indicates that integrating traditional ecological knowledge on animal-based food and medicines could be extremely useful when developing strategies for sustainable management and conservation of marine species, and in particular sea turtles.

Résumé Objectif : Etudier les formes cliniques et aspects thérapeutiques des tendinopathies des membres pelviens (TMP) chez les patients consultant dans le service de rhumatologie du CNHU/HKM de Cotonou (Bénin). Méthodes : Il s’est agi d’une étude transversale descriptive et analytique ayant porté sur 94 patients atteints de TMP. Les informations ont été recueillies sur une fiche de collecte ou d’enquête préparée à cet effet. L’analyse des données a été effectuée avec le logiciel Epi-info version 7.2.1.0. Résultats : Quatre-vingt-quatorze (94) patients ont été recrutés dont 85,11% de femmes et l’âge moyen était de 57,55±3,64ans. Cliniquement la cheville et le pied étaient les sièges les plus touchés dans 40,42% des cas. Les diagnostics les plus retenus étaient par ordre d’importance : la talagie plantaire (28,72%), la tendinopathie de la patte d’oie (25,53%), les tendinopathies tibiales (14,89%), les tendinopathies du moyen fessier (14,89%), les tendinopathies fibulaires (13,83%). Les anti-inflammatoires non stéroïdiens ont donné de bons résultats thérapeutiques (76,60%). Le stress et le facteur majeur lié à l’évolution du traitement p=0,004. Conclusion : Les TMP atteignent en majorité les femmes d’âge mur. Elles touchent préférentiellement la cheville et le pied. Le stress est un facteur majeur lié à l’évolution du traitement. Il convient donc de lutter efficacement contre le stress pour limiter les poussées douloureuses. Mots clés : Tendinopathie, membre pelvien, traitement, Bénin.

Human respiratory syncytial virus (hRSV) is the most common cause of bronchiolitis and pneumonia in newborns, with all children being infected before the age of two. Reinfections are very common throughout life and can cause severe respiratory infections in the elderly and immunocompromised adults. Although vaccines and preventive antibodies have recently been licensed for use in specific subpopulations of patients, there is still no therapeutic treatment commonly available for these infections. Here, we investigated the potential antiviral activity of Retro-2.2, a derivative of the cellular retrograde transport inhibitor Retro-2, against hRSV. We show that Retro-2.2 inhibits hRSV replication in cell culture and impairs the ability of hRSV to form syncytia. Our results suggest that Retro-2.2 treatment affects virus spread by disrupting the trafficking of the viral de novo synthetized F and G glycoproteins to the plasma membrane, leading to a defect in virion morphogenesis. Taken together, our data show that targeting intracellular transport may be an effective strategy against hRSV infection.

PurposeIn National Wilms Tumor Study 5 (NWTS-5), tumor-specific combined loss of heterozygosity of chromosomes 1p and 16q (LOH1p/16q) was associated with adverse outcomes in patients with favorable histology Wilms tumor. The AREN0533/AREN0532 studies assessed whether augmenting therapy improved event-free survival (EFS) for these patients. Patients with stage I/II disease received regimen DD4A (vincristine, dactinomycin and doxorubicin) but no radiation therapy. Patients with stage III/IV disease received regimen M (vincristine, dactinomycin, and doxorubicin alternating with cyclophosphamide and etoposide) and radiation therapy.MethodsPatients were enrolled through the AREN03B2 Biology study between October 2006 and October 2013; all underwent central review of pathology, surgical reports, and imaging. Tumors were evaluated for LOH1p/16q by microsatellite testing. EFS and overall survival were compared using the log-rank test between NWTS-5 and current studies.ResultsLOH1p/16q was detected in 49 of 1,147 evaluable patients with stage I/II disease (4.27%) enrolled in AREN03B2; 32 enrolled in AREN0532. LOH1p/16q was detected in 82 of 1,364 evaluable patients with stage III/IV disease (6.01%) in AREN03B2; 51 enrolled in AREN0533. Median follow-up for 83 eligible patients enrolled in AREN0532/0533 was 5.73 years (range, 2.84 to 9.63 years). The 4-year EFS for patients with stage I/II and stage III/IV disease with LOH1p/16 was 87.3% (95% CI, 75.1% to 99.5%) and 90.2% (95% CI, 81.8% to 98.6%), respectively. These results are improved compared with the NWTS-5 updated 4-year EFS of 68.8% for patients with stage I/II disease (P = .042), and 61.3% for patients with stage III/IV disease (P = .001), with trends toward improved 4-year overall survival. The most common grade 3 or higher nonhematologic toxicities with regimen M were febrile neutropenia (39.2%) and infections (21.6%).ConclusionAugmentation of therapy improved EFS for patients with favorable histology Wilms tumor and LOH1p/16q compared with the historical NWTS-5 comparison group, with an expected toxicity profile.

The mentoring and demonstration plot program with the application of technology carried out by the Faculty of Agriculture Universitas Gadjah Mada Team was proven to be able to increase the productivity and quality of soybeans, although some were only applied partially (not completely) from the recommended technology package, this was constrained by many factors in each mentoring area. In the Cilacap area, the demonstration plot results showed a fairly high potential reaching 2.24 tons/ha in Bantarsari, 2.94 and 3.77 tons/ha in Kawunganten, while in Majenang before harvesting soybeans showed good potential with 130-140 pods. per plant, this is of course the potential yield can reach above 3.0 tons/ha, but unfortunately it was affected by the flood due to the broken embankment, so the soybeans were inundated and damaged (rotten) in a relatively short time. The process of technology transfer and adoption that occurred went well, and many farmers gave testimony that this mentoring program was good to be developed and continued, so that under conditions according to the growing season, they would be able to show even better potential.

Crop diseases cause significant yield losses, and the use of resistant cultivars can effectively mitigate these losses and control many plant diseases. Most plant resistance (R) genes encode immune receptors composed of nucleotide-binding and leucine-rich repeat (NLR) domains. These proteins mediate the specific recognition of pathogen avirulence effectors to induce defence responses. However, NLR-triggered immunity can be associated with a reduction in growth and yield, so-called fitness costs. Recent data have shown that plants use an elaborate interplay of different mechanisms to control NLR gene transcript levels, as well as NLR protein abundance and activity, to avoid the associated cost of resistance in the absence of a pathogen. In this review, we discuss the different levels of NLR regulation (transcriptional, post-transcriptional and at the protein level). We address the apparent need for plants to maintain diverse modes of regulation. A recent model suggesting an equilibrium ON/OFF state of NLR proteins, in the absence of a pathogen, provides the context for our discussion.

Purpose The National Wilms Tumor Study (NWTS) treatment of favorable histology Wilms tumor with lung metastases was vincristine/dactinomycin/doxorubicin (DD4A) and lung radiation therapy (RT). The AREN0533 study applied a new risk stratification and treatment strategy to improve event-free survival (EFS) while reducing exposure to lung RT. Methods Patients with favorable histology Wilms tumor and isolated lung metastases showing complete lung nodule response (CR) after 6 weeks of DD4A continued receiving chemotherapy without lung RT. Patients with incomplete response (IR) or loss of heterozygosity at chromosomes 1p/16q received lung RT and four cycles of cyclophosphamide/etoposide in addition to DD4A drugs (Regimen M). AREN0533 was designed to preserve a 4-year EFS of 85% for lung nodule CR and improve 4-year EFS from 75% to 85% for lung nodule IR. Results Among 292 assessable patients, 133 had CR and 159 had IR. For patients with CR, 4-year EFS and overall survival (OS) estimates were 79.5% (95% CI, 71.2% to 87.8%) and 96.1% (95% CI, 92.1% to 100%), respectively. Expected versus observed event rates were 15% and 20.2% ( P = .052), respectively. For patients with IR, 4-year EFS and OS estimates were 88.5% (95% CI, 81.8% to 95.3%) and 95.4% (95% CI, 90.9% to 99.8%), respectively. Expected versus observed event rates were 25% and 12.2% ( P < .001), respectively. Overall, 4-year EFS and OS were 85.4% (95% CI, 80.5% to 90.2%) and 95.6% (95% CI, 92.8% to 98.4%) compared with 72.5% (95% CI, 66.9% to 78.1%; P < .001) and 84.0% (95% CI, 79.4% to 88.6%; P < .001), respectively, in the predecessor NWTS-5 study. Conclusion Excellent OS was achieved after omission of primary lung RT in patients with lung nodule CR, although there were more events than expected. EFS was significantly improved, with excellent OS, in patients with lung nodule IR using four cycles of cyclophosphamide/etoposide in addition to DD4A drugs. The overall AREN0533 treatment strategy yielded EFS and OS estimates that were superior to previous studies.

This article explores the subjectivity of two Francophone black African women's in/visibility. Through Ousmane Sembene's film La noire de ... and Henriette Akofa's novel Une esclave moderne, I pivot the ways the two works mirror the protagonists' isolating experience emblematized by the in/visibility that many migratory black women live across transnational spaces on both sides of the Atlantic, a crisis that takes its most palpable and sometimes deadly turn through a socially and ethically oppressive in/visibility. Inspired in part by the work of Renee Larrier (2000) and Frangoise Lionnet (1995) in postcolonial Francophone contexts and theoretical works in Anglophone spaces like Samantha Pinto's Difficult Diasporas: The Transnational Feminist Aesthetic of the Black Atlantic (2013). I examine how laboring black women's oppression born of transnational migration informs the states and spaces of in/visibility. Fluconazole is an effective agent for prophylaxis of invasive candidiasis in premature infants. The objective of this study was to characterize the population pharmacokinetics (PK) and dosing requirements of fluconazole in infants with birth weights of 2,000 ng/ml in 80% of simulated infants at week 1 and 59% at week 4 of treatment. Developmental changes in fluconazole clearance are best predicted by serum creatinine in this population. A twice-weekly dose of 6 mg/kg achieves appropriate levels for prevention of invasive candidiasis in extremely premature infants.

Background: Patients with stage IV favorable histology Wilms tumor (FHWT) with extrapulmonary metastases (EPM) constitute a small subset of patients with FHWT. Because of their rarity and heterogeneity, optimal FHWT treatment is not well understood. Children's Oncology Group protocol AREN0533 assigned patients with FHWT and EPM to intensified chemotherapy, regimen M, after initial DD‐4A chemotherapy. To improve understanding of prognostic factors and best therapies, experiences of patients with EPM on AREN0533, as well as on protocols AREN03B2 and NWTS‐5, were reviewed. Methods: Combined outcomes for patients with EPM from NWTS‐5, AREN0533, and AREN03B2 were determined. Those treated on AREN0533 were compared with those treated on NWTS‐5. Prognostic factors were explored in the pooled cohort. Results: Forty‐seven patients with FHWT with EPM enrolled on AREN0533, 37 enrolled on NWTS‐5, and 64 were followed only on AREN03B2. The pooled cohort of all 148 patients demonstrated a 4‐year event‐free survival (EFS) of 77.3% (95% CI, 70.8–84.4) and 4‐year overall survival of 88.9% (95% CI, 83.9–94.2). Four‐year EFS of patients with EPM treated on AREN0533 was 76.0% (95% CI, 64.6–89.4) vs 64.9% (95% CI, 51.7–82.2) on NWTS‐5; hazard ratio, 0.64, p =.26; no difference in overall survival was observed. Increasing linear age and slow incomplete lung response were associated with worse EFS in a pooled cohort. Conclusions: Outcomes for patients with EPM are among the lowest for children with FHWT. Further trials with standardized surgical and radiation treatment to metastatic sites, and prospectively collected biologic and treatment details are needed. Bean anthracnose caused by the hemibiotrophic fungus Colletotrichum lindemuthianum is one of the most important diseases of common bean (Phaseolus vulgaris) in the world. In the present study, the whole transcriptome of common bean infected with C. lindemuthianum during compatible and incompatible interactions was characterized at 48 and 72 hpi, corresponding to the biotrophy phase of the infection cycle. Our results highlight the prominent role of pathogenesis-related (PR) genes from the PR10/Bet vI family as well as a complex interplay of different plant hormone pathways including Ethylene, Salicylic acid (SA) and Jasmonic acid pathways. Gene Ontology enrichment analysis reveals that infected common bean seedlings responded by down-regulation of photosynthesis, ubiquitination-mediated proteolysis and cell wall modifications. In infected common bean, SA biosynthesis seems to be based on the PAL pathway instead of the ICS pathway, contrarily to what is described in Arabidopsis. Interestingly, ~30 NLR were up-regulated in both contexts. Overall, our results suggest that the difference between the compatible and incompatible reaction is more a question of timing and strength, than a massive difference in differentially expressed genes between these two contexts. Finally, we used RT-qPCR to validate the expression patterns of several genes, and the results showed an excellent agreement with deep sequencing.

Pituitary adenylyl cyclase-activating polypeptide (PACAP) is a member of the vasoactive intestinal polypeptide (VIP)-the secretin-glucagon family of neuropeptides. They act through two classes of receptors: PACAP type 1 (PAC1) and type 2 (VPAC1 and VPAC2). Among their pleiotropic effects throughout the body, PACAP functions as neuromodulators and neuroprotectors, rescuing neurons from apoptosis, mostly through the PAC1 receptor. To explore the potential protective effect of endogenous PACAP against Noise-induced hearing loss (NIHL), we used a knockout mouse model lacking PAC1 receptor expression (PACR1−/−) and a transgenic humanized mouse model expressing the human PAC1 receptor (TgHPAC1R). Based on complementary approaches combining electrophysiological, histochemical, and molecular biological evaluations, we show PAC1R expression in spiral ganglion neurons and in cochlear apical cells of the organ of Corti. Wild-type (WT), PAC1R−/−, and TgHPAC1R mice exhibit similar auditory thresholds. For most of the frequencies tested after acute noise damage, however, PAC1R−/− mice showed a larger elevation of the auditory threshold than did their WT counterparts. By contrast, in a transgene copy number-dependent fashion, TgHPAC1R mice showed smaller noise-induced elevations of auditory thresholds compared to their WT counterparts. Together, these findings suggest that PACAP could be a candidate for endogenous protection against noise-induced hearing loss.

Yield assessment of new released genotypes of cotton (Gossypium hirsutum L.) bred in Benin. This paper aimed at identifying the most productive upland cotton genotypes among seven new releases (P 625-1, P 746-1, Q 674-2, Q 723-4, Q 792-2, R 663-4 et R 689-1). The controls were the 3 varieties currently in use in Benin: ANG 956, OKP 768, and KET 782. Overall, 146 variety trials have been set over 3 years. All the new releases were evaluated on station, whereas only P 625‑1 and P 746‑1 have been tested on farmer field trials. Experiments were run according to a randomized complete block design. Data were analysed with a linear mixed model. The Genotype x Macro-environment interaction was retrieved from an Additive Main effects and Multiplicative Interactions (AMMI) model. A formal Mentorship Program within the Children's Oncology Group (COG) was established to pair young investigators (mentees) with established COG members (mentors). Despite the American Academy of Pediatrics policy statement promoting mentorship programs, there are no publications describing and evaluating national mentorship programs in pediatric subspecialties. In this study, a series of internal program evaluations were performed using surveys of both mentors and mentees. Responses were deidentified and analyzed to determine the utility of the program by both participant satisfaction and self-reported academic productivity. Results indicated that mentees were generally satisfied with the program. Mentor-mentee pairs that met at least quarterly demonstrated greater academic productivity than pairings that met less frequently. This formal mentorship program appeared to have subjective and objective utility for the development of academic pediatric subspecialists.
