Arbuscular mycorrhizal fungi (AMF) perform key soil ecosystem services and, because of their symbiotic relationship with plant roots, may be exposed to the plant protection products (PPPs) applied to soils and crops. In 2017, the European Food Safety Authority (EFSA) released a scientific opinion addressing the state of the science on risk assessment of PPPs for in-soil organisms, recommending the inclusion of AMF ecotoxicological testing in the PPP regulatory process. However, it is not clear how this can be implemented in a tiered, robust, and ecologically relevant manner. Through a critical review of current literature, we examine the recommendations made within the EFSA report and the methodologies available to integrate AMF into the PPP risk assessment and provide perspective and commentary on their agronomic and ecological relevance. We conclude that considerable research questions remain to be addressed prior to the inclusion of AMF into the in-soil organism risk assessment, many of which stem from the unique challenges associated with including an obligate symbiont within the PPP risk assessment. Finally, we highlight critical knowledge gaps and the further research required to enable development of relevant, reliable, and robust scientific tests alongside pragmatic and scientifically sound guidance to ensure that any future risk-assessment paradigm is adequately protective of the ecosystem services it aims to preserve. Earthworms are important ecosystem engineers, and assessment of the risk of plant protection products toward them is part of the European environmental risk assessment (ERA). In the current ERA scheme, exposure and effects are represented simplistically and are not well integrated, resulting in uncertainty when the results are applied to ecosystems. Modeling offers a powerful tool to integrate the effects observed in lower tier laboratory studies with the environmental conditions under which exposure is expected in the field. This paper provides a summary of the (In)Field Organism Risk modEling by coupling Soil Exposure and Effect (FORESEE) Workshop held 28-30 January 2020 in Düsseldorf, Germany. This workshop focused on toxicokinetic-toxicodynamic (TKTD) and population modeling of earthworms in the context of ERA. The goal was to bring together scientists from different stakeholder groups to discuss the current state of soil invertebrate modeling and to explore how earthworm modeling could be applied to risk assessments, in particular how the different model outputs can be used in the tiered ERA approach. In support of these goals, the workshop aimed at addressing the requirements and concerns of the different stakeholder groups to support further model development. The modeling approach included 4 submodules to cover the most relevant processes for earthworm risk assessment: environment, behavior (feeding, vertical movement), TKTD, and population. Four workgroups examined different aspects of the model with relevance for risk assessment, earthworm ecology, uptake routes, and cross-species extrapolation and model testing. Here, we present the perspectives of each workgroup and highlight how the collaborative effort of participants from multidisciplinary backgrounds helped to establish common ground. In addition, we provide a list of recommendations for how earthworm TKTD modeling could address some of the uncertainties in current risk assessments for plant protection products. Earthworms are considered ecosystem engineers and, as such, they are an integral part of the soil ecosystem. The movement of earthworms is significantly influenced by environmental factors such as temperature and soil properties. As movement may directly be linked to food ingestion, especially of endogeic species like Aporrectodea caliginosa, changes in those environmental factors also affect life history traits such as growth and reproduction.In our laboratory studies, earthworms showed a decrease in burrowing activity with decreasing moisture levels and, to some extent, the organic matter content. The burrowing activity of earthworms was also affected by temperature, for which the casts produced per earthworm was used as a proxy in laboratory experiments. We integrated changes in earthworm movement and life histories in response to temperature, soil organic matter content and the moisture level, as observed in our experiment and reported in the literature, through dynamic energy budget (DEB) modelling. The joint parametrization of a DEB model for A. caliginosa based on movement and life history data revealed that food ingestion via movement is an integral part of the earthworms’ energy budgets. Our findings highlight the importance of soil properties to be considered in the model development for earthworms. Furthermore, by understanding and incorporating the effect of environmental factors on the physiology, this mechanistic approach can help assess the impact of environmental changes such as temperature rise or drought.

Intact soil food webs are pivotal to maintaining essential soil functions, such as carbon recycling, sequestering, and biomass production. Although the functional role of micro- (e.g., bacteria and fungi) and macrofauna (e.g., earthworms) is comparatively well established, the importance of the mesofauna community (e.g., abundance and diversity of Acari and Collembola) in maintaining soil functionality is less clear. We investigated this question in a six-month field experiment in arable soil by actively manipulating mesofauna abundance and biodiversity through the application of two legacy insecticides (lindane and methamidophos) at sufficiently high doses to reduce mesofauna abundance (well above previously registered application rates; 2.5 and 7.5 kg a.s./ha for lindane, and 0.6 and 3 kg a.s./ha for methamidophos) and measure the impact on organic matter degradation. Our results demonstrate that both insecticides had reduced Collembola and Acari abundances by up to 80% over the study's six-month duration. In addition, we observed less pronounced and more complex changes in mesofauna biodiversity over time. These included insecticide-dependent temporal fluctuations (both reduction and increase) for different estimates (indices) of local (alpha)-diversity over time and no lasting impact for most estimates after six months. Even at these exceptionally high field rates, Collembola and Acari diversity was observed to generally recover by six months. In contrast, considering organic matter breakdown, we found no evidence of a treatment-related effect. These results suggest that organic matter breakdown in arable soils is likely driven by other trophic levels (e.g., microorganisms or earthworms) with only a limited influence of the mesofauna community. We discuss these findings with regard to their implications for our current understanding of soil food web function and future European soil risk assessments. A comprehensive multiyear monitoring program was conducted to assess the exposure, effects, and long-term risk of the fungicide bixafen to earthworms in cereal fields. The realistic exposure of bixafen in soil was assessed at 10 representative field sites in Germany after a period of up to 8 years of use with five different products containing bixafen, followed by annual measurements from 2017 to 2019. The measured exposure concentrations were compared with modeled predicted environmental concentrations in soil (PECsoil) that are derived in the context of the European risk assessment of plant protection products. It was shown that the model assumptions, in particular the kinetic parameters describing the background accumulation, provided a conservative description of the observed residue data. This demonstrates that the exposure modeling tools are adequate for use in soil risk assessment. Laboratory and field ecotoxicological studies were performed to provide a comprehensive risk assessment on the long-term use of bixafen-based fungicides in cereals. While a laboratory reproduction study with the earthworm Eisenia fetida indicated a potential risk at the Tier 1 risk assessment for the end-use product Skyway XPro®, a 2.5-year field study showed no unacceptable long-term effects on natural earthworm populations. The exposure in this study exceeded the maximum recommended field rate of Skyway XPro® by a factor of 3 and the maximum measured bixafen concentrations from exposure monitoring study by a factor of 5.2. International audience; The aim of the environmental risk assessment of chemicals is the prevention of unacceptable adverse effects on the environment. Therefore, the risk assessment for in-soil organisms, such as earthworms, is based on two key elements: the exposure assessment and the effect assessment. In the current risk assessment scheme, these two elements are not linked. While for the exposure assessment, advanced exposure models can take the spatial and temporal scale of substances into account, the effect assessment in the lower tiers considers only a limited temporal and spatial variability. However, for soil organisms, such as earthworms, those scales play a significant role as species move through the soil in response to environmental factors. To overcome this gap, we propose a conceptual integration of pesticide exposure, ecology, and toxicological effects on earthworms using a modular modeling approach. An essential part of this modular approach is the environment module, which utilizes exposure models to provide spatially and temporally explicit information on environmental variables (e.g., temperature, moisture, organic matter content) and chemical concentrations. The behavior module uses this information and simulates the feeding and movement of different earthworm species using a trait-based approach. The resulting exposure can be processed by a toxico kinetic-toxicodynamic (TKTD) module. TKTD models are particularly suitable to make effect predictions for time-variable exposure situations as they include the processes of uptake, elimination, internal distribution, and biotransformation of chemicals and link the internal concentration to an effect at the organism level. The population module incorporates existing population models of different earthworm species. The modular approach is illustrated using a case study with an insecticide. Our results emphasize that using a modular model approachwill facilitate the integration of exposure and effects and thus enhance the risk assessment of soil organisms.

In the first step of earthworm risk assessment for plant protection products (PPPs), the risk is assessed by comparing the no-observed effect levels (NOELs) from laboratory reproduction tests with the predicted exposure of the PPP in soil, while applying a trigger value (assessment factor [AF]) to cover uncertainties. If this step indicates a potential risk, field studies are conducted. However, the predicted environmental concentration in soil, which can be calculated, for example, for different soil layers (ranging from 0-1 cm to 0-20 cm), and the AF determine the conservatism that is applied in this first step. In this review paper, the tier 1 earthworm risk assessment for PPPs is calibrated by comparing the NOEL in earthworm reproduction tests with effect levels on earthworm populations under realistic field conditions. A data set of 54 pairs of studies conducted in the laboratory and in the field with the same PPP was compiled, allowing a direct comparison of relevant endpoints. The results indicate that a tier 1 AF of 5 combined with a regulatory relevant soil layer of 0 to 5 cm provides a conservative tier 1 risk assessment. A risk was identified by the tier 1 risk assessment in the majority of the cases at application rates that were of low risk for natural earthworm populations under field conditions. Increasing the conservatism in the tier 1 risk assessment by reducing the depth of the regulatory relevant soil layer or by increasing the tier 1 AF would increase the number of false positives and trigger a large number of additional field studies. This increased conservatism, however, would not increase the margin of safety for earthworm populations. Within the regulatory framework for authorisation of plant protection products (PPPs) (EU Directive 91/414/1991/EEC replaced by Regulation (EC) 1107/2009), higher tier risk assessments and earthworm field tests are conducted in different countries across Europe. This paper describes dominant earthworm species for regulatory and biogeographical regions in agricultural landscapes across Europe and examines regional differences in earthworm communities and densities and their respective response to a toxic reference. For the assessment of earthworm abundance and species distribution, data of untreated control plots from 30 earthworm field studies were analysed; each conducted according to the ISO 11268–3 (1999) guideline by European Crop Protection Association member companies in the context of registration of PPPs. For the evaluation of the response to PPPs under different regional and climatic conditions, the effect on earthworm abundance was assessed by comparing plots treated with toxic references with untreated control plots. Additionally, a comparative literature review was included providing an overview of earthworm species composition and densities in agricultural crops from 14 European countries. The assessment of earthworm field studies from six different European countries revealed that common earthworm species of anecic and endogeic ecological groups are present at most field sites. Dominant species groups of endogeic and anecic earthworms can be defined that are abundant in all assessed countries. These are the endogeic species Aporrectodea caliginosa, Aporrectodea rosea and Allolobophora chlorotica, and the anecic species Lumbricus terrestris (Northern and Central Europe) and Lumbricus friendi (Southern Europe). Taking into account the high variability in total earthworm abundances, it can be concluded that the variability within regions was larger than the variability between regions. Analysis of the earthworm community and data of toxic references lead to the conclusion that testing in different zones is not considered necessary.

Energy crops are increasingly cultivated in agricultural management systems world-wide. A substitution of food crops (e.g. cereals) by energy crops may generally alter the biological activity and litter decomposition in soil due to their varying structural and chemical composition and subsequently modify soil functioning. A soil microcosm experiment was performed to assess the decomposition and microbial mineralization of different energy crop residues in soil compared to a food crop, with or without earthworms. Residues of the energy crops winter rape (Brassica napus), maize (Zea mays), miscanthus (Miscanthus giganteus) and the food crop oat (Avena sativa) were each provided as food source for a mixed earthworm population, each consisting of one individual of Lumbricus terrestris, Aporrectodea caliginosa, and Octolasion tyrtaeum. After 6 weeks, the rate of litter loss from the soil surface, earthworm biomass, microbial biomass-C and -N, microbial activity, and enzyme activities were determined. The results emphasized, that litter loss and microbial parameters were predominantly promoted by earthworms and were additionally influenced by the varying structural and chemical composition of the different litter. Litter decay by earthworms was highest in N-rich maize litter treatment (C–N ratio 34.8) and lowest in the case of miscanthus litter (C–N ratio 134.4). As a consequence, the microbial biomass and basal respiration in soils with maize litter were higher, relative to other litter types. MBC–MBN ratio in soil increased when earthworms were present, indicating N competition between earthworms and microorganisms. Furthermore, enzyme activities responded in different ways on the varying types of litter and earthworm activity. Enzymes involved in the N-cycle decreased and those involved in the C-cycle tended to increase in the presence of earthworms, when litter with high C–N ratio was provided as a food source. Especially in the miscanthus treatments, less N might remain for enzymatic degradation, indicating that N competition between earthworms and microorganisms may vary between different litter types. Especially, an expansion of miscanthus in agricultural management systems might result in a reduced microbial activity and a higher N deficit for microorganisms in soil.

To assess the impact of different types of soil tillage on the density, biomass, and community composition of earthworms, a long-term field study was performed in which soils were tilled in different ways for ten years. This study included five different types of tillage: (i) plough, (ii) grubber, (iii) disc harrow, (iv) mulch sowing, and (v) direct sowing. At the end of the experiment the earthworm density, biomass, and community composition, and the SOC (soil organic carbon) content were determined. The results show that density, biomass, and community composition of earthworm populations varied in relation to the type of soil tillage used. The density of anecic earthworm species decreased when soils were managed by conventional ploughing, relative to reduced tillage practices, whereas conversely the density of endogeic species increased. Additionally, the varying types of soil tillage influenced the abundance and biomass of different earthworm species in different ways. The density of Aporrectodea caliginosa was positively influenced by ploughing, whereas Aporrectodea longa, Lumbricus castaneus, and Satchellius mammalis showed a positive relationship to the grubber and Allolobophora chlorotica to direct sowing. We attribute these changes to modifications in the vertical distribution of SOC and varying potentials for mechanical damage of earthworms by tillage. A decrease in tillage intensity modified the vertical SOC distribution in the topsoil and consequently revealed positive effects on earthworm biodiversity, thus sustaining soil functioning.

A laboratory experiment was performed to assess the impact of ecologically different earthworm species on soil water characteristics, such as soil tension, water content, and water infiltration rate. Three earthworm species ( Lumbricus rubellus , Aporrectodea caliginosa , Lumbricus terrestris ) were exposed in soil columns (diameter 30 cm, height 50 cm) for 100 days with a total fresh earthworm biomass of 22.7 ± 0.4 g per column, each in duplicate. Each column was equipped with tensiometers at 10 and 40 cm and FD-probes at 10 cm depth, to continuously measure the temporal development of soil tension and soil moisture. Additionally, 30 g of sieved and rewetted horse manure was placed on the soil surface as a food source. Precipitation events (10 mm) were simulated at day 28 and day 64. At the end of the experiment the water infiltration rate and the runoff at 55 cm depth were determined. The results showed considerable evidence, that ecologically different earthworms modify soil water characteristics in different ways. The anecic L. terrestris and the endogeic A. caliginosa showed the tendency to enhance the drying of the topsoil and subsoil. Their intensive and deep burrowing activity might enhance the exchange of water vapor due to a better aeration in soil. In contrast, the epigeic L. rubellus tended to enhance the storage of soil moisture in the topsoil, which might be linked to lower rates of litter loss from soil surface and thus a thicker litter layer remaining. A. caliginosa led to considerable higher water infiltration rates and faster water discharges in the subsoil, relative to the other species, probably due to a high soil dwelling activity.

A soil microcosm experiment was performed to assess (1) the C- and N- turnover of residues from biogas plants in soils in the presence of three earthworm species ( Lumbricus terrestris , Aporrectodea longa and Aporrectodea caliginosa ) and (2) the resulting changes in soil chemical and microbiological properties when using these residues as fertilizer in comparison to conventional slurry. Earthworms were exposed in soils, fertilized with an equivalent amount of 120 kg of NH 4 -N ha −1 from: (1) conventional cattle slurry and (2) a fermented residue derived from cattle slurry, grass (silage) and maize. Additional treatments without slurry and earthworms were used as controls. There was considerable evidence that soils fertilized by fermented slurry comprised fewer amounts of readily available nutrients for microbial C and N turnover. We observed significant stimulation of microbial biomass, basal respiration and nitrification in treatments with conventional slurry, especially in the presence of earthworms. However, the stimulation of microbial activity by manure and earthworms were significantly lower in treatments with fermented slurry. Moreover, the results showed clear interactions between different earthworm species and manures. While the biomass of the anecic species ( L. terrestris and A. longa ) increased in both slurry treatments, the biomass of A. caliginosa (endogeic) decreased, with a significantly stronger biomass decline in treatments with fermented slurry. The metabolic quotients revealed microbial stress metabolism in fermented slurry treatments, predominantly in treatments with A. caliginosa . We conclude that particularly A. caliginosa and soil microorganisms competed for labile C sources in treatments with fermented slurry. An application of these residues as fertilizer might result in a reduction of microbial activity in agricultural soils and in a decline of endogeic earthworms.

A soil microcosm experiment was performed to assess the uptake of Hg from various Hg-spiked food sources (soil, leaf litter and root litter of Trifolium alexandrinum) by two earthworm species, Lumbricus terrestris (anecic) and Octolaseon cyaneum (endogeic). Treatments were applied in which one of the three food sources was Hg spiked and the other two were not. Additional treatments in which all or none of the food sources were Hg spiked were used as controls. Uptake of Hg from soil into tissues of both earthworm species was significantly higher than uptake of Hg from leaf litter or root litter, indicating that soil may be the most important pool for the uptake of Hg into earthworms. In addition, the anecic L. terrestris significantly accumulated Hg from all Hg-spiked food sources (leaf litter, root litter and soil), whereas the endogeic O. cyaneum took up Hg mainly from soil particles. Interestingly, there was no further increase in Hg in L. terrestris when all food sources were Hg spiked compared to the single Hg-spiked sources. This may be attributed to the relatively high Hg content in the soil, which may have influenced the feeding behavior of the earthworms, although their biomass did not significantly decline. We suggest that, in addition to the physiological differences, feeding behavior may also play a role in the contrasting uptake of Hg by the two earthworm species.

Collembola are soil dwelling organisms that provide important ecosystem services within soils. To increase realism in evaluating potential effects of plant protection products a Collembola two-generation study was developed. This test assesses the potential for recovery of Collembola when exposed to plant protection products. Juvenile individuals of Folsomia candida (Willem, Ann Soc Entomol Belg 46:275-283, 1902) which hatched under conditions of exposure to a test substance in a modified OECD 232 bioassay were introduced into a second consecutive bioassay containing the same test substance aged in soil. This test system determines whether a population which was initially impacted by a substance in a 1st bioassay shows normal reproduction or survival in a 2nd bioassay after aging of the test substance in soil. An intermediate period for juvenile growth is included between the 1st and 2nd bioassay in order to reduce the control treatment variability in reproduction and mortality to fulfill the validity criteria according to the OECD 232 guideline. The Collembola two-generation study is able to differentiate between substances showing either a potential long-term risk or comprising a low risk. Comparing the results of this two generation study with data from semi-field or field studies indicates a high degree of conservatism when this test is considered within a tiered risk assessment scheme. This approach represents a valuable tool which makes the risk assessment more efficient by providing an alternative refinement option for highly conservative tier 1 Collembola risk assessment.

Earthworms are used in an increasing number of microcosm experiments that investigate their behaviour and biology or that consider earthworms an environmental factor that influences soil properties and biological interactions. However, there exists no standardized protocol for performing comparable studies. After giving a short overview of the different experimental approaches using earthworms as model organisms, the present paper provides recommendations for the planning and execution of earthworm experiments that help in achieving comparable results. The recommendations, Summarized in a workflow diagram, pertain to the acquisition, treatment and description of earthworms for experimentation, the description and preparation of test soils and the criteria that should be met for valid experimental results. Bioaccumulation of Hg, Cd and Pb by eight ecophysiologically distinct earthworm species was studied in 27 polluted and uncontaminated forest soils. Lowest tissue concentrations of Hg and Cd occurred in epigeic Lumbricus rubellus and highest in endogeic Octolasion cyaneum . Soils dominated by Dendrodrilus rubidus possess a high potential of risk of Pb biomagnification for secondary predators. Bioconcentration factors (soil–earthworm) followed the sequence ranked Cd > Hg > Pb. Ordination plots of redundancy analysis were used to compare HM concentrations in earthworm tissues with soil, leaf litter and root concentrations and with soil pH and CEC. Different ecological categories of earthworms are exposed to Hg, Cd and Pb in the topsoil by atmospheric deposition and accumulate them in their bodies. Species differences in HM concentrations largely reflect differences in food selectivity and niche separation.
