Introduction:. Private equity (PE) investment in health care has increased more than 250% between 2010 and 2020. This is mirrored by an increasing number of published materials in medical journals. The objective of our study was to identify and characterize trends and key themes seen within publications discussing the topic of PE investment into orthopaedic surgery practices and bias within those publications. Methods:. The inclusion criteria for our study required an article to be published between the years 2003 and June 2024 and to contain the phrase “orthopaedic surgery” or “orthopedics” in addition to “PE” or “PE investment.” Based on these parameters, 15 articles met the criteria for inclusion. Articles were then evaluated to assess various themes related to general views expressed regarding PE firms, reasons for PE attraction to orthopaedics, and most cited positives/negatives of PE investment and potential conflicts of interest with respect to underlying relationships/associations with PE firms at the time of publication. Results:. Of the 15 publications meeting the inclusion criteria, 4 (26.7%) expressed positive views on the topic of PE ownership of orthopaedic practices, while 4 (26.7%) expressed a neutral view and 7 (46.7%) expressed a negative outlook. Four (26.7%) of the articles had authors who were either employed or had ownership in a practice that was purchased by a PE firm. Of these 4 articles, none disclosed this potential conflict of interest. Three of the 4 articles had either a positive or neutral view of PE. The most cited reason for PE attraction to orthopaedics was revenue from ancillary services. The most cited upside of PE transactions was the possibility of benefiting from economies of scale, while the most cited downside was the misalignment of incentives. Conclusion:. The plurality of present studies views PE transactions negatively (46.7%). Our research unveiled 4 studies with undisclosed conflicts of interest (26.7%). In addition, orthopaedic surgeons should be wary of the numerous downsides of PE transactions, such as the misaligned incentives between themselves and PE firms. Clinical relevance:. Independent orthopaedic practices should be critical of the literature when evaluating the merits of potential partnerships with PE firms.

Abstract Introduction Locked knee caused by inferior patellar dislocation is considered rare in elderly patients. It was originally thought that, in the osteoarthritic knee, osteophytes on the pole of the patella become entrapped in the inter-condylar notch, which is managed by performing closed reduction and immobilization in a knee splint for three to four weeks. We present an unusual case of a locked knee with an impaction fracture. To the best of our knowledge, there have been no previous reports of such impaction fractures managed with arthroscopy. Case presentation We present an unusual case of an 88-year-old Caucasian woman with moderate arthritis who had a locked knee caused by an impaction fracture of the patella into the lateral femoral condyle. In this case report, we describe the need for arthroscopic surgery to prevent relocking of the knee in these patients. Conclusions This case report emphasizes the need for careful assessment of locked knees in elderly patients. Impaction fractures should be considered in all rare cases of patellar dislocation, and we advocate arthroscopic assessment of the articular cartilage in these patients. This is an important consideration, as the population demographics change and such impaction fractures may become more common in patients with degeneration in the knees.

Tools to calculate human movement patterns can benefit musculoskeletal clinicians and researchers for rehabilitation assessments. The research objective of this study was to compare two human pose estimation models (HRNet, MediaPipe) against the laboratory marker-based reference standard for joint angles and range of motion (ROM) for several movement parameters. Twenty-two healthy volunteers (Female n = 16, Male n = 6), participated to compare outputs for knee and elbow kinematics. Joint angles were calculated by selecting three marker points defining the joint and angle between them in Qualisys Track Manager software. Using predicted key points, pose estimation model calculations for the same musculoskeletal kinematic outputs were computed. Coefficient of Variation (CoV) was used as a variation statistic for joint angle during movements. All comparison results were under 10%, implying that both models compute reliable joint angle data during the five tested activities. When comparing ROM as a discrete parameter, CoV values remain low, though not all below 10%. Intra-class Correlation Coefficients were computed across the ROM data as a measure of statistical similarity. Each exercise displayed good-excellent and significant correlations for both models compared to Qualisys apart from left knee sit-to-stand. Exploration from this data sampling imply that flexion/extension exercises give stronger consistency results than full sit-to-stand movements when compared to 3D motion analysis, and there is little distinction between these two models. Finer tuning of models will give further reliability for in-depth analysis as these results are restricted, but valuable for a rehabilitative setting with limited objective analysis alternative., Competing Interests: Declaration of competing interest The authors confirm that the content of the manuscript has not been published or submitted for publication in another journal but has been presented as exploratory data and protocol testing at various meetings. Chronic, low-grade inflammation has been implicated in aging and age-dependent conditions, including Alzheimer's disease, cardiomyopathy, and cancer. One of the age-associated processes underlying chronic inflammation is protein aggregation, which is implicated in neuroinflammation and a broad spectrum of neurodegenerative diseases such as Alzheimer's, Huntington's, and Parkinson's diseases. We screened a panel of bioactive thiadiazolidinones (TDZDs) from our in-house library for rescue of protein aggregation in human-cell and C. elegans models of neurodegeneration. Among the tested TDZD analogs, PNR886 and PNR962 were most effective, significantly reducing both the number and intensity of Alzheimer-like tau and amyloid aggregates in human cell-culture models of pathogenic aggregation. A C. elegans strain expressing human Aβ 1-42 in muscle, leading to AD-like amyloidopathy, developed fewer and smaller aggregates after PNR886 or PNR962 treatment. Moreover, age-progressive paralysis was reduced 90% by PNR886 and 75% by PNR962, and "healthspan" (the median duration of spontaneous motility) was extended 29% and 62%, respectively. These TDZD analogs also extended wild-type C. elegans lifespan by 15-30% ( p < 0.001), placing them among the most effective life-extension drugs. Because the lead drug in this family, TDZD-8, inhibits GSK3β, we used molecular-dynamic tools to assess whether these analogs may also target GSK3β. In silico modeling predicted that PNR886 or PNR962 would bind to the same allosteric pocket of inactive GSK3β as TDZD-8, employing the same pharmacophore but attaching with greater avidity. PNR886 and PNR962 are thus compelling candidate drugs for treatment of tau- and amyloid-associated neurodegenerative diseases such as AD, potentially also reducing all-cause mortality.

Background: Although statistical errors have been shown to be prevalent in the medical literature, there has been a paucity of studies focusing on the prevalence of statistical errors in neurosurgery. We examined the contemporary neurosurgical literature for a common statistical mistake, namely, misinterpretations of null hypothesis significance testing results near the P-value threshold by conveying the idea of a trend., Methods: PubMed/Medline was used to identify all articles published in 6 major neurosurgical journals between 2000 and 2020. The abstracts of these articles were extracted and scrutinized to determine when the word "trend" was used to express near statistical significance., Results: This study included a total of 45,244 articles. Of those, 461 (1.02% [95% confidence interval, 0.86%-1.18%]) employed the word "trend" to indicate near statistical significance, for a total of 3.8 (95% confidence interval, 2.93-4.75) errors per issue per year. The error under study occurred more frequently in Journal of Neurosurgery: Spine and less frequently in Acta Neurochirurgica (P = 0.007). On an annual basis, there was no linear correlation between the total number of articles published per journal and the number of such errors in that journal (r = 0.34, P = 0.01)., Conclusions: Misinterpretations of null hypothesis significance testing results near the P-value threshold are present in at least 1% of the neurosurgical literature. While we believe that most statistical errors in medicine in general and neurosurgery in particular may be unintentional in nature, additional measures should be put in place to prevent the subsequent adoption of such undesirable methodological practices among future researchers., (Copyright © 2021 Elsevier Inc. Diagnosis of neurodegenerative diseases hinges on "seed" proteins detected in disease-specific aggregates. These inclusions contain diverse constituents, adhering through aberrant interactions that our prior data indicate are nonrandom. To define preferential protein-protein contacts mediating aggregate coalescence, we created click-chemistry reagents that cross-link neighboring proteins within human, APP Sw -driven, neuroblastoma-cell aggregates. These reagents incorporate a biotinyl group to efficiently recover linked tryptic-peptide pairs. Mass-spectroscopy outputs were screened for all possible peptide pairs in the aggregate proteome. These empirical linkages, ranked by abundance, implicate a protein-adherence network termed the "aggregate contactome." Critical hubs and hub-hub interactions were assessed by RNAi-mediated rescue of chemotaxis in aging nematodes, and aggregation-driving properties were inferred by multivariate regression and neural-network approaches. Aspirin, while disrupting aggregation, greatly simplified the aggregate contactome. This approach, and the dynamic model of aggregate accrual it implies, reveals the architecture of insoluble-aggregate networks and may reveal targets susceptible to interventions to ameliorate protein-aggregation diseases., (Published by Elsevier Inc.)

Highly charged metal ions are difficult to investigate in weakly coordinating ionic liquids (ILs) because of the insolubility of their solid forms, but the molecular liquid TiCl 4 offers a way to react tetravalent metal ions in an IL. Reactions of TiCl 4 with 1-butyl-3-methylimidazolium ([C 4 mim] + )-based ILs containing chloride or bromide lead to mixtures of highly metastable amorphous solids and small amounts of crystalline chlorotitanate salts including [C 4 mim] 2 [TiCl 6 ] and two polymorphs of [C 4 mim] 2 [Ti 2 Cl 10 ] in a manner not well correlated with stoichiometry or anion identity. The reaction of TiCl 4 with [C 4 mim][BF 4 ] yields crystals of the mixed fluoro-chloro complex [C 4 mim] 2 [Ti 4 F 6 Cl 12 ], indicating spontaneous reaction of the IL ions to generate HF in situ. These unusual behaviors are explained in terms of the exceptionally high acidity of Ti 4+ and the unusual behavior of TiCl 4 among metal halides as a nonpolar molecular compound.

A neutral molecular complex, Zn(ZnCl 4 ) 2 (Cho) 2 , has been isolated from the well-known choline chloride/ZnCl 2 deep eutectic system (DES) and its crystal structure has been determined. The structure demonstrates the possibility of isolating unusual coordination complexes from DES which departs from the well-established observation of such systems being formed from large, ionic metal complexes or oligomers.

The natural product fumagillin exhibits potent antiproliferative and antiangiogenic properties. The semisynthetic analog PPI-2458, [(3R,4S,5S,6R)-5-methoxy-4-[(2R,3R)-2-methyl-3-(3-methylbut-2-enyl)oxiran-2-yl]-1-oxaspiro[2.5]octan-6-yl] N-[(2R)-1-amino-3-methyl-1-oxobutan-2-yl]carbamate, demonstrates rapid inactivation of its molecular target, methionine aminopeptidase-2 (MetAP2), and good efficacy in several rodent models of cancer and inflammation with oral dosing despite low apparent oral bioavailability. To probe the basis of its in vivo efficacy, the metabolism of PPI-2458 was studied in detail. Reaction phenotyping identified CYP3A4/5 as the major source of metabolism in humans. Six metabolites were isolated from liver microsomes and characterized by mass spectrometry and nuclear resonance spectroscopy, and their structures were confirmed by chemical synthesis. The synthetic metabolites showed correlated inhibition of MetAP2 enzymatic activity and vascular endothelial cell growth. In an ex vivo experiment, MetAP2 inhibition in white blood cells, thymus, and lymph nodes in rats after single dosing with PPI-2458 and the isolated metabolites was found to correlate with the in vitro activity of the individual species. In a phase 1 clinical study, PPI-2458 was administered to patients with non-Hodgkin lymphoma. At 15 mg administered orally every other day, MetAP2 in whole blood was 80% inactivated for up to 48 hours, although the exposure of the parent compound was only ∼10% that of the summed cytochrome P450 metabolites. Taken together, the data confirm the participation of active metabolites in the in vivo efficacy of PPI-2458. The structures define a metabolic pathway for PPI-2458 that is distinct from that of TNP-470 ([(3R,4S,5S,6R)-5-methoxy-4-[(2R,3R)-2-methyl-3-(3-methylbut-2-enyl)oxiran-2-yl]-1-oxaspiro[2.5]octan-6-yl] N-(2-chloroacetyl)carbamate). The high level of MetAP2 inhibition achieved in vivo supports the value of fumagillin-derived therapeutics for angiogenic diseases.

Semisynthetic analogues of fumagillin, 1, inhibit methionine aminopeptidase-2 (MetAP2) and have entered the clinic for the treatment of cancer. An optimized fumagillin analogue, 3 (PPI-2458), was found to be orally active, despite containing a spiroepoxide function that formed a covalent linkage to the target protein. In aqueous acid, 3 underwent ring-opening addition of water and HCl, leading to four products, 4-7, which were characterized in detail. The chlorohydrin, but not the diol, products inhibited MetAP2 under weakly basic conditions, suggesting reversion to epoxide as a step in the mechanism. In agreement, chlorohydrin 6 was shown to revert rapidly to 3 in rat plasma. In an ex vivo assay, rats treated with purified acid degradants demonstrated inhibition of MetAP2 that correlated with the biochemical activity of the compounds. Taken together, the results indicate that degradation of the parent compound was compensated by the formation of active equivalents leading to a pharmacologically useful level of MetAP2 inhibition.

Background: Epilepsy is associated with precocious development of Alzheimer-type neuropathological changes, including appearance of senile plaques, neuronal loss and glial activation. As inheritance of APOE ε4 allele(s) is reported to favor this outcome, we sought to investigate neuronal and glial responses that differ according to APOE genotype. With an eye toward defining ways in which APOE ε3 alleles may foster neuronal well-being in epilepsy and/or APOE ε4 alleles exacerbate neuronal decline, neuronal and glial characteristics were studied in temporal lobectomy specimens from epilepsy patients of either APOE ε4,4 or APOE ε3,3 genotype., Methods: Tissue and/or cellular expressions of interleukin-1 alpha (IL-1α), apolipoprotein E (ApoE), amyloid β (Aβ) precursor protein (βAPP), synaptophysin, phosphorylated tau, and Aβ were determined in frozen and paraffin-embedded tissues from 52 APOE ε3,3 and 7 APOE ε4,4 (0.25 to 71 years) epilepsy patients, and 5 neurologically normal patients using Western blot, RT-PCR, and fluorescence immunohistochemistry., Results: Tissue levels of IL-1α were elevated in patients of both APOE ε3,3 and APOE ε4,4 genotypes, and this elevation was apparent as an increase in the number of activated microglia per neuron (APOE ε3,3 vs APOE ε4,4 = 3.7 ± 1.2 vs 1.5 ± 0.4; P < 0.05). This, together with increases in βAPP and ApoE, was associated with apparent neuronal sparing in that APOE ε4,4 genotype was associated with smaller neuron size (APOE ε4,4 vs APOE ε3,3 = 173 ± 27 vs 356 ± 45; P ≤ 0.01) and greater DNA damage (APOE ε4,4 vs APOE ε3,3 = 67 ± 10 vs 39 ± 2; P = 0.01). 3) Aβ plaques were noted at early ages in our epilepsy patients, regardless of APOE genotype (APOE ε4,4 age 10; APOE ε3,3 age 17)., Conclusions: Our findings of neuronal and glial events, which correlate with lesser neuronal DNA damage and larger, more robust neurons in epilepsy patients of APOE ε3,3 genotype compared to APOE ε4,4 genotype carriers, are consistent with the idea that the APOE ε3,3 genotype better protects neurons subjected to the hyperexcitability of epilepsy and thus confers less risk of AD (Alzheimer's disease).Please see related article: http://www.biomedcentral.com/1741-7015/10/36.

The air-stable complex Pd(η(3)-allyl)(DTBNpP)Cl (DTBNpP = di(tert-butyl)neopentylphosphine) serves as a highly efficient precatalyst for the arylation of amines and enolates using aryl bromides and chlorides under mild conditions with yields ranging from 74% to 98%. Amination reactions of aryl bromides were carried out using 1-2 mol % Pd(η(3)-allyl)(DTBNpP)Cl at 23-50 °C without the need to exclude oxygen or moisture. The C-N coupling of the aryl chlorides occurred at relatively lower temperature (80-100 °C) and catalyst loading (1 mol %) using the Pd(η(3)-allyl)(DTBNpP)Cl precatalyst than the catalyst generated in situ from DTBNpP and Pd(2)(dba)(3) (100-140 °C, 2-5 mol % Pd). Other Pd(DTBNpP)(2)-based complexes, (Pd(DTBNpP)(2) and Pd(DTBNpP)(2)Cl(2)) were ineffective precatalysts under identical conditions for the amination reactions. Both Pd(DTBNpP)(2) and Pd(DTBNpP)(2)Cl(2) precatalysts gave nearly quantitative conversions to the product in the α-arylation of propiophenone with p-chlorotoluene and p-bromoanisole at a substrate/catalyst loading of 100/1. At lower substrate/catalyst loading (1000/1), the conversions were lower but comparable to that of Pd(t-Bu(3)P)(2). In many cases, the tri-tert-butylphosphine (TTBP) based Pd(I) dimer, [Pd(μ-Br)(TTBP)](2), stood out to be the most reactive catalyst under identical conditions for the enolate arylation. Interestingly, the air-stable Pd(I) dimer, Pd(2)(DTBNpP)(2)(μ-Cl)(μ-allyl), was less active in comparison to [Pd(μ-Br)(TTBP)](2) and Pd(η(3)-allyl)(DTBNpP)Cl. The X-ray crystal structures of Pd(η(3)-allyl)(DTBNpP)Cl, Pd(DTBNpP)(2)Cl(2), Pd(DTBNpP)(2), and Pd(2)(DTBNpP)(2)(μ-Cl)(μ-allyl) are reported in this paper along with initial studies on the catalyst activation of the Pd(η(3)-allyl)(DTBNpP)Cl precatalyst.

The U.S. Army Night Vision and Electronic Sensors Directorate (NVESD) and the U.S. Army Research Laboratory have developed a terahertz (THz) -band imaging system performance model for detection and identification of concealed weaponry. The MATLAB-based model accounts for the effects of all critical sensor and display components and for the effects of atmospheric attenuation, concealment material attenuation, and active illumination. The model is based on recent U.S. Army NVESD sensor performance modeling technology that couples system design parameters to observer-sensor field performance by using the acquire methodology for weapon identification performance predictions. This THz model has been developed in support of the Defense Advanced Research Project Agencies' Terahertz Imaging Focal-Plane Technology (TIFT) program and is currently being used to guide the design and development of a 0.650 THz active-passive imaging system. This paper will describe the THz model in detail, provide and discuss initial modeling results for a prototype THz imaging system, and outline plans to calibrate and validate the model through human perception testing.

The first definitive high-resolution single-crystal X-ray structure for the coordination of the 1-methylimidazole (Meimid) ligand to UO2(Ac)2 (Ac = CH3CO2) is reported. The crystal structure evidence is confirmed by IR, Raman, and UV-vis spectroscopic data. Direct participation of the nitrogen atom of the Meimid ligand in binding to the uranium center is confirmed. Structural analysis at the DFT (B3LYP) level of theory showed a conformational difference of the Meimid ligand in the free gas-phase complex versus the solid state due to small energetic differences and crystal packing effects. Energetic analysis at the MP2 level in the gas phase supported stronger Meimid binding over H2O binding to both UO2(Ac)2 and UO2(NO3)2. In addition, self-consistent reaction field COSMO calculations were used to assess the aqueous phase energetics of combination and displacement reactions involving H2O and Meimid ligands to UO2R2 (R = Ac, NO3). For both UO2(NO3)2 and UO2(Ac)2, the displacement of H2O by Meimid was predicted to be energetically favorable, consistent with experimental results that suggest Meimid may bind uranyl at physiological pH. Also, log(Knitrate/KAc) calculations supported experimental evidence that the binding stoichiometry of the Meimid ligand is dependent upon the nature of the reactant uranyl complex. These results clearly demonstrate that imidazole binds to uranyl and suggest that binding of histidine residues to uranyl could occur under normal biological conditions.

A total of sixty-three choline derivative-based ionic liquids in the forms of chlorides, acesulfamates, and bis(trifluoromethylsulfonyl)imides have been prepared and their physical properties (density, viscosity, solubility, and thermal stability) have been determined. Thirteen of these salts are known chlorides: precursors to the 26 water-soluble acesulfamates, 12 acesulfamates only partially miscible with water, and 12 water-insoluble imides. The crystal structures for two of the chloride salts-(2-hydroxyethyl)dimethylundecyloxymethylammonium chloride and cyclododecyloxymethyl(2-hydroxyethyl)dimethylammonium chloride-were determined. The antimicrobial (cocci, rods, and fungi) activities of the new hydrophilic acesulfamate-based ILs were measured and 12 were found to be active. The alkoxymethyl(2-hydroxyethyl)dimethylammonium acesulfamates have been shown to be insect feeding deterrents and thus open up a new generation of synthetic deterrents based on ionic liquids. The alkoxymethyl(2-decanoyloxyethyl)dimethylammonium bis(trifluoromethylsulfonyl)imides have also been shown to act as fixatives for soft tissues and can furthermore be used as substitutes for formalin and also preservatives for blood.

Variation in operational temperatures has revealed differences in the partitioning behavior of probe solutes between the phases in aqueous biphasic systems (ABS) and the related aqueous biphasic extraction chromatographic resin (ABEC). This difference has been studied using the hydrophobic anion, 99TcO4-, as a probe and (NH4)2SO4 as the kosmotropic salt. Distribution of the hydrophobic anion 99TcO4- to the PEG-rich phase in a MePEG-5000/(NH4)2SO4 ABS increases with increasing temperature, but decreases are observed in batch uptakes of this anion to ABEC resins from (NH4)2SO4 solutions. Phase diagrams were constructed at five different temperatures from 10 to 50 degreesC using cloud point titration for the ABS and a correlation between the phase divergence, measured in terms of tie line length (TLL), and the temperature of the partitioning system was verified. Thermodynamic parameters (deltaHdegrees,deltaSdegrees, deltaGdegrees, ) as a function of temperature were calculated for the various systems studied and the results imply thermodynamic differences between partitioning in ABS versus ABEC.

The metal-ion complexing properties of the ligand EDTAM (ethylenediamine-N,N,N',N'-tetraacetamide) are investigated as a model for the role of amide oxygen donors in the binding sites of Ca-binding proteins. The structures of the complexes [Ca(EDTAM)NO3]NO3 (1), [La(EDTAM)(H2O)4](NO3)3.H2O (2), and [Cd(EDTAM)(NO3)]NO3 (3) are reported: 1 monoclinic, P2(1)/c, a = 10.853(2) angstroms, b = 12.893(3) angstroms, c = 13.407(3) angstroms, beta = 103.28(3) degrees, Z = 4, R = 0.0281; 2 triclinic, P, a = 8.695(2) angstroms, b = 9.960(2) angstroms, c = 16.136(3) angstroms, alpha = 95.57(3) degrees, beta = 94.84(3) degrees, gamma = 98.72(3) degrees, Z = 2, R = 0.0394; 3 monoclinic, P2(1)/c, a = 10.767(2) angstroms, b = 12.952(2) angstroms, c = 13.273(2) angstroms, beta = 103.572(3) degrees, Z = 4, R = 0.0167. Compounds 1 and 3 are isostructural, and the EDTAM binds to the metal ion through its two N-donors and four O-donors from the amide groups. Ca(II) in 1 is 8-coordinate with a chelating NO3- group, while Cd(II) in 3 may possibly be 7-coordinate, with an asymmetrically coordinated NO3- that is best regarded as unidentate. The La(III) in 2 is coordinated to the EDTAM in a manner similar to that of 1 and 3, but it is 10-coordinate with four water molecules coordinated to the La(III). The formation constants (log K1) for complexes of a variety of metal ions with EDTAM are reported in 0.1 M NaNO3 at 25.0 +/- 0.1 degrees C. These are compared to the log K1 values for en (ethylenediamine) and THPED (N,N,N',N'-tetrakis(2-hydroxypropyl)-ethylenediamine). For large metal ions, such as Ca2+ or La3+, log K1 increases strongly when the four acetamide groups are added to en to give EDTAM, whereas for a small metal ion, such as Mg2+, this increase is small. The log K1 values for EDTAM compared to THPED suggest that the amide oxygen is a much stronger base than the alcoholic oxygen. Structures of binding sites in 40 Ca-binding proteins are examined. It is shown that the Ca-O=C bond angles involving coordinated amides in these sites are large, commonly being in the 150-180 degrees range. This is discussed in terms of the idea that for purely ionic bonding the M-O=C bond angle will approach 180 degrees, while for covalent bonding the angle should be closer to 120 degrees. How this fact might be used by the proteins to control selectivity for different metal ions is discussed.

Readily prepared imidazole-based boronium ions form stable, hydrophobic, room-temperature ionic liquids (RTIL) with unique electronic and spectroscopic characteristics.

Polyethylene glycol (PEG)-aqueous biphasic systems (ABS) and PEG-grafted aqueous biphasic extraction chromatographic (ABEC) resins have been shown to remove inorganic species from environmental and nuclear wastes. The partitioning behavior of several iodide species (iodide, iodine, triiodide, iodate, and 4-iodo-2,6-dimethylphenol (I-DMP)) have been studied for PEG (MW 2000)-salt systems and ABEC resins. Iodide partitioning to PEG-rich phases or onto ABEC resins can be enhanced by derivatization with 2,6-dimethylphenol to form 4-iodo-2,6-dimethylphenol or by addition of I(2) to form triiodide. Conversely, iodide partitioning to the PEG-rich phase or onto ABEC resins is reduced by oxidation of iodide to IO(3)(-). Partitioning studies of iodide, iodate, and iodine in a PEG-ABS are compared to results using ABEC resins.

Background: We investigated the levels of transcription factors associated with activation of the mitogen-activated protein (MAP) kinase pathway in schizophrenics using postmortem brain samples. These studies were done to determine whether our previous findings of abnormal levels of the MAP kinases in the cerebellar vermis were linked to additional downstream targets of this signal transduction pathway., Method: We measured the protein levels of 3 transcription factors in nuclear fractions of postmortem samples from cerebellar vermis of 10 patients with schizophrenia and 13 control subjects: Elk-1, cyclic adenosine monophosphate (cAMP) response element binding protein (CREB), and activating transcription factor 2 (ATF-2). Studies in rats examined the postmortem stability and effect of haloperidol and risperidone on levels of Elk-1, cAMP, and ATF-2 proteins., Results: We found a significant increase in the protein levels of Elk-1 (mean+SD, 4489+/-659 vs 2915+/-583 arbitrary densitometric units [P<.001]), CREB (mean +/- SD, 2149 1061 vs 904+/-711 arbitrary densitometric units [P=.003]) and ATF-2 (mean+/-SD, 1421 854 vs 512+/-394 arbitrary densitometric units [P=.003]) in the cerebellar vermis of schizophrenic subjects. Complementary studies in rats indicate that these findings can not be attributed to subacute treatment with antipsychotic medications., Conclusion: Taken together with the alterations of MAP kinases previously reported, and the findings of elevations of downstream transcription targets, we suggest that the MAP kinase signal transduction pathway contributes to the cerebellar abnormalities in schizophrenia.

Aqueous biphasic systems (ABS) are comprised of both a polymer-rich phase (e.g., polyethylene glycol, PEG) and a salt-rich phase [e.g., (NH4)2SO4] such that both phases are 80% water on a molar basis. ABS have demonstrated applications as environmentally-friendly methods to separate relatively hydrophobic anionic species, such as pertechnetate and mercury halide anionic complexes, from high ionic strength solutions although partitioning of hydrated metal ions, such as Fe3+ and actinides, to the PEG-rich phase is negligible without the addition of a metal ion complexant to the system. Four naphthol- or resorcinol-based dyes; 1-(2-pyridylazo)-2-naphthol (PAN), 1-(thiazolylazo)-2-naphthol (TAN), 4-(2-pyridylazo)-resorcinol (PAR) and 4-(2-thiazolylazo)-resorcinol (TAR), each incorporating a naphthol or resorcinol with an ortho azo functional group, have been studied as metal ion extractants in ABS as a function of pH. In the PEG-2000/ (NH4)2SO4 ABS, the distribution ratios of Fe3+, Co2+ and Ni2- were enhanced by several orders of magnitude at high pH in contrast to the behavior of Cs+, Cd2+ and Eu3+ whose partitioning behavior was largely unaffected by the presence of these extractants at any pH. The three extracted metal ions, Fe3+, Co2+ and Ni2+, could be stripped by contact with a fresh salt phase at low pH.

Poly(ethylene glycol)-based aqueous biphasic systems (PEG-ABS) can be utilized to separate and recover metal ions in environmental and hydrometallurgical applications. A concurrent study was conducted comparing the partitioning of mercury between aqueous layers in an ABS [Me-PEG-5000/(NH4)2SO4] and partitioning of mercury from aqueous solutions to aqueous biphasic extraction chromatographic (ABEC-5000) resins. In ammonium sulfate solutions, mercury partitions to the salt-rich phase in ABS, but by using halide ion extractants, mercury will partition to the PEG-rich phase after formation of a chloro, bromo or iodo complex. Aqueous biphasic systems (ABS) are suitable for the separation of small organic molecules in industrial and environmental applications and thus, it is important to correlate partitioning behavior of model organic solutes with their structure in order to develop predictive models. The partitioning behavior of five, uncharged, substituted benzenes (benzene, toluene. chlorobenzene, 1,4-dichlorobenzene and 1,2,4-trichlorobenzene) were studied in ABS prepared from stock solutions of 40% (w/w) PEG-2000 and increasing concentrations of four water-structuring salts (K3PO4, K2CO3, (NH4)2SO4 and NaOH). For a given solute and a defined concentration of salt, the partition coefficients increase as the deltaGhyd value of the salt anion becomes more negative (e.g., Dbenzene increases in the order OH-

A wide variety of anatomic and histological alterations are common in brains of aged individuals. However, identification of intrinsic aging changes--as distinct from changes resulting from cumulative environmental insult--is problematic. Some degree of neuronal and volume loss would appear to be inevitable, but recent studies have suggested that the magnitudes of such changes are much less than previously thought, and studies of dendritic complexity in cognitively intact individuals suggest continuing neuronal plasticity into the eighth decade. A number of vascular changes become more frequent with age, many attributable to systemic conditions such as hypertension and atherosclerosis. Age-associated vascular changes not clearly linked to such conditions include hyaline arteriosclerotic changes with formation of arterial tortuosities in small intracranial vessels and the radiographic changes in deep cerebral white matter known as "leukoaraiosis." Aging is accompanied by increases in glial cell activation, in oxidative damage to proteins and lipids, in irreversible protein glycation, and in damage to DNA, and such changes may underlie in part the age-associated increasing incidence of "degenerative" conditions such as Alzheimer disease and Parkinson disease. A small number of histological changes appear to be universal in aged human brains. These include increasing numbers of corpora amylacea within astrocytic processes near blood-brain or cerebrospinal fluid-brain interfaces, accumulation of the "aging" pigment lipofuscin in all brain regions, and appearance of Alzheimer-type neurofibrillary tangles (but not necessarily amyloid plaques) in mesial temporal structures.

Solvent extraction utilizing an oil-water mixture (e.g., chloroform-water) and a suitable complexant, is a proven technology for the selective removal and recovery of metal ions from aqueous solutions. Aqueous biphasic systems (ABS), formed by mixing certain inorganic salts and water-soluble polymers, or by mixing two dissimilar water-soluble polymers, have been studied for more than 40 years for the gentle, non-denaturing separation of fragile biomolecules, yet ABS have been virtually ignored as a possible extraction technology for metal ions. In this report we review our metal ion partitioning work and discuss the three major types of partitioning: (1) those rare instances that the metal ion species present in a given solution partitions to the PEG-rich phase without an extractant; (2) the use of halide salts which produce a metal anion complex that partitions to the PEG-rich phase; and (3) the use of a water-soluble extractant which distributes to the PEG-rich phase. In addition, we correlate the partitioning behavior we observed with available thermodynamic data for metal ions and their complexes.

The purpose of these studies was to examine the axial evolution of the negative glow in a hollow cathode discharge. The time-average negative glow profiles along the central axis of a hollow cathode were recorded for 81 pulse widths over the range of 5-25 microseconds in increments of 0.25 microseconds at a constant interpulse delay of 206.4 microseconds. Subsequent numerical processing yielded the instantaneous negative glow profile. The negative glow was viewed through a mesh-covered slot along the length of the hollow cathode. The negative flow profiles were imaged using a vidicon video camera. A PC-based frame grabber digitized the video images and stored them for subsequent processing.

Sputtering changes the analytical performance of the hollow cathode as an emission source. Proper conditioning (aging) of the hollow with sputtering greatly improves precision. Conditioning decreases the blank emission signal and changes the shape of the hollow. Micrographs which show the evolution of the bottom of the hollow into a stable bulb shape are presented. The blank emission signal decreases with aging of the hollow and approaches a constant value when the hollow is properly conditioned. A short-term stability (n = 5) of approximately 1.8% RSD has been observed frequently. An average long-term stability over several days (n = 45) of 4.5% for Li and 3.5% for Na in the emission signal from microsamples (less than 50 nL) deposited in the hollow cathode discharge source is reported here. Instrumentation, operation, and sample preparation procedures are described.

We present a general expression for the transmission function of the discrete frequency-versus-radius reticle and compare such a reticle with the more common continuous reticle. A discrete form of the frequency-versus-radius reticle has an integer number of chopping cycles on a single radius. The discreteform limits the resolution of the reticle in the radial direction, but this limit is not severe for small-target images. However, since no phase reversal occurs, electronic processing is simplified.

A significant improvement in the precision of the hollow cathode as an emission source is reported. Precision of 1% or less has been observed several times over periods of several hours. An average long-term stability of 4.3% for Ll and 6.0% for Na in the emission signal from microsamples (less than 50 nL) deposited in the hollow cathode discharge source is reported. The improved precision is attributed primarily to the introduction of electronics that hold the discharge current more nearly constant and to the shielding of all wiring to the source from the power supply. A current-controlled switch that is capable of driving a hollow cathode discharge in either dc or pulsed mode is described. This switch is capable of generating current pulses as short as 2 microseconds through a resistive load and greatly improves the discharge stability and repeatability at turn-on in both the dc and pulsed modes. The characteristics of pulses produced by this switch are presented; however, analytical performance is reported only for the dc mode. Temporal current plots are presented for the new instrumentation and compared to plots taken with commonly used current-controlled power supplies. Instrumentation, operation, and sample preparation procedures are described. Typical temporal profiles of the emission signal from microsamples deposited in AI and stainless steel hollow cathodes are given.
