Source analysis of Electroencephalography (EEG) data requires the computation of the scalp potential induced by current sources in the brain. This so-called EEG forward problem is based on an accurate estimation of the volume conduction effects in the human head, represented by a partial differential equation which can be solved using the finite element method (FEM). FEM offers flexibility when modeling anisotropic tissue conductivities but requires a volumetric discretization, a mesh, of the head domain. Structured hexahedral meshes are easy to create in an automatic fashion, while tetrahedral meshes are better suited to model curved geometries. Tetrahedral meshes thus offer better accuracy, but are more difficult to create. Methods: We introduce CutFEM for EEG forward simulations to integrate the strengths of hexahedra and tetrahedra. It belongs to the family of unfitted finite element methods, decoupling mesh and geometry representation. Following a description of the method, we will employ CutFEM in both controlled spherical scenarios and the reconstruction of somatosensory evoked potentials. Results: CutFEM outperforms competing FEM approaches with regard to numerical accuracy, memory consumption and computational speed while being able to mesh arbitrarily touching compartments. Conclusion: CutFEM balances numerical accuracy, computational efficiency and a smooth approximation of complex geometries that has previously not been available in FEM-based EEG forward modeling., Comment: 9 pages, 8 figures

Transfer Entropy, a generalisation of Granger Causality, promises to measure "information transfer" from a source to a target signal by ignoring self-predictability of a target signal when quantifying the source-target relationship. A simple example for signals with such self-predictability are narrowband signals. These are both thought to be intrinsically generated by the brain as well as commonly dealt with in analyses of brain signals, where band-pass filters are used to separate responses from noise. However, the use of Transfer Entropy is usually discouraged in such cases. We simulate simplistic examples where we confirm the failure of classic implementations of Transfer Entropy when applied to narrow-band signals, as made evident by a flawed recovery of effect sizes and interaction delays. We propose an alternative approach based on a whitening of the input signals before computing a bivariate measure of directional time-lagged dependency. This approach solves the problems found in the simple simulated systems. Finally, we explore the behaviour of our measure when applied to delta and theta response components in Magnetoencephalography (MEG) responses to continuous speech. The small effects that our measure attributes to a directed interaction from the stimulus to the neuronal responses are stronger in the theta than in the delta band. This suggests that the delta band reflects a more predictive coupling, while the theta band is stronger involved in bottom-up, reactive processing. Taken together, we hope to increase the interest in directed perspectives on frequency-specific dependencies.

Introduction: Identifying neurobiological differences between patients suffering from Major Depressive Disorder (MDD) and healthy individuals has been a mainstay of clinical neuroscience for decades. However, recent meta- and mega-analyses have raised concerns regarding the replicability and clinical relevance of brain alterations in depression. Methods: Here, we systematically investigate healthy controls and MDD patients across a comprehensive range of modalities including structural magnetic resonance imaging (MRI), diffusion tensor imaging, functional task-based and resting-state MRI under near-ideal conditions. To this end, we quantify the upper bounds of univariate effect sizes, predictive utility, and distributional dissimilarity in a fully harmonized cohort of N=1,809 participants. We compare the results to an MDD polygenic risk score (PRS) and environmental variables. Results: The upper bound of the effect sizes range from partial eta squared = .004 to .017, distributions overlap between 89% and 95%, with classification accuracies ranging between 54% and 55% across neuroimaging modalities. This pattern remains virtually unchanged when considering only acutely or chronically depressed patients. Differences are comparable to those found for PRS, but substantially smaller than for environmental variables. Discussion: We provide a large-scale, multimodal analysis of univariate biological differences between MDD patients and controls and show that even under near-ideal conditions and for maximum biological differences, deviations are extremely small and similarity dominates. We sketch an agenda for a new focus of future research in biological psychiatry facilitating quantitative, theory-driven research, an emphasis on computational psychiatry and multivariate machine learning approaches, as well as the utilization of ecologically valid phenotyping., Comment: 12 pages, 3 figures

Sorption of gases in micro- and mesoporous materials is typically interpreted on the basis of idealized structural models where real structure effects such as defects and disorder are absent. For covalent organic frameworks (COFs) significant discrepancies between measured and simulated adsorption isotherms are often reported but rarely traced back to their origins. This is because little is known about the real structure of COFs and its effect on the sorption properties of these materials. In the present work molecular simulations are used to obtain adsorption isotherms of argon, nitrogen, and carbon dioxide in the COF-LZU1 at various temperatures. The (perfect) model COF has a BET surface that is higher than the experimental BET surface by a factor of approximately 1.33, suggesting defects or inclusions are present in the real structure. We find that the saturation adsorption loading of small gaseous species in COF-LZU1, as determined from grand canonical Monte Carlo simulations, is also higher by approximately the same factor compared to the experimental saturation loading. The influence of interlayer slipping on the shape of the adsorption isotherm and the adsorption capacity is studied. Comparison between simulation and experiment at lower loadings suggests the layers to be shifted instead of perfectly eclipsed. The sensitivity of the adsorption isotherms in this regime towards the underlying framework topology shows that real structure effects have significant influence on the gas uptake. Accounting for layer slipping is important to applications such as catalysis, gas storage and separation.

Electroconvulsive Therapy (ECT) is arguably the most effective intervention for treatment-resistant depression. While large interindividual variability exists, a theory capable of predicting individual response to ECT remains elusive. To address this, we posit a quantitative, mechanistic framework of ECT response based on Network Control Theory (NCT). Then, we empirically test our approach and employ it to predict ECT treatment response. To this end, we derive a formal association between Postictal Suppression Index (PSI) - an ECT seizure quality index - and whole-brain modal and average controllability, NCT metrics based on white matter brain network architecture, respectively. Exploiting the known association of ECT response and PSI, we then hypothesized an association between our controllability metrics and ECT response mediated by PSI. We formally tested this conjecture in N=50 depressive patients undergoing ECT. We show that whole-brain controllability metrics based on pre-ECT structural connectome data predict ECT response in accordance with our hypotheses. In addition, we show the expected mediation effects via PSI. Importantly, our theoretically motivated metrics are at least on par with extensive machine learning models based on pre-ECT connectome data. In summary, we derived and tested a control-theoretic framework capable of predicting ECT response based on individual brain network architecture. It makes testable, quantitative predictions regarding individual therapeutic response, which are corroborated by strong empirical evidence. Our work might constitute a starting point for a comprehensive, quantitative theory of personalized ECT interventions rooted in control theory.

We propose a predictive Density Functional Theory (DFT) for the calculation of solvation free energies. Our approach is based on a Helmholtz free-energy functional that is consistent with the perturbed-chain SAFT (PC-SAFT) equation of state. This allows a coarse-grained description of the solvent, based on an inhomogeneous density of PC-SAFT segments. The solute, on the other hand, is described in full detail by atomistic Lennard-Jones interaction sites. The approach is entirely predictive, as it only takes the PC-SAFT parameters of the solvent and the force-field parameters of the solute as input. No adjustable parameters or empirical corrections are involved. The framework is applied to study self-solvation of n-alkanes and to the calculation of residual chemical potentials in binary solvent mixtures. Our DFT approach accurately predicts solvation free energies of small molecular solutes in three different solvents. Additionally, we show the calculated solvation free energies agree well with those obtained by molecular dynamics simulations and with the residual chemical potential calculated by the bulk PC-SAFT equation of state. We observe higher deviations for the solvation free energy of systems with significant solute-solvent Coulomb interactions.

A classical density functional theory (cDFT) based on the PC-SAFT equation of state is proposed for the calculation of adsorption equilibria of pure substances and their mixtures in covalent organic frameworks (COFs). Adsorption isotherms of methane, ethane, n-butane and nitrogen in the COFs TpPa-1 and 2,3-DhaTph are calculated and compared to results from grand canonical Monte Carlo (GCMC) simulations. Mixture adsorption is investigated for the methane/ethane and methane/n-butane binary systems. Excellent agreement between PC-SAFT DFT and GCMC is obtained for all adsorption isotherms up to pressures of 50 bar. The cDFT formalism accurately predicts the selective accumulation of longer hydrocarbons for binary mixtures in the considered COFs. This application shows substantial predictive power of PC-SAFT DFT solved in three-dimensional geometries and the results suggest the method can in the future also be applied for efficient optimization of force field parameters or of structural properties of the porous material based on an analytical theory as opposed to a stochastic simulation.
