Abstract

SUMMARY: Flavan‐3‐ols are prominent phenolic compounds found abundantly in the young leaves of tea plants. The enzymes involved in flavan‐3‐ol biosynthesis in tea plants have been extensively investigated. However, the localization and associations of these numerous functional enzymes within cells have been largely neglected. In this study, we aimed to investigate the synthesis of flavan‐3‐ols in tea plants, particularly focusing on epigallocatechin gallate. Our analysis involving the DESI‐MSI method to reveal a distinct distribution pattern of B‐ring trihydroxylated flavonoids, primarily concentrated in the outer layer of buds. Subcellular localization showed that CsC4H, CsF3′H, and CsF3′5′H localizes endoplasmic reticulum. Protein–protein interaction studies demonstrated direct associations between CsC4H, CsF3′H, and cytoplasmic enzymes (CHS, CHI, F3H, DFR, FLS, and ANR), highlighting their interactions within the biosynthetic pathway. Notably, CsF3′5′H, the enzyme for B‐ring trihydroxylation, did not directly interact with other enzymes. We identified cytochrome b5 isoform C serving as an essential redox partner, ensuring the proper functioning of CsF3′5′H. Our findings suggest the existence of distinct modules governing the synthesis of different B‐ring hydroxylation compounds. This study provides valuable insights into the mechanisms underlying flavonoid diversity and efficient synthesis and enhances our understanding of the substantial accumulation of B‐ring trihydroxylated flavan‐3‐ols in tea plants. Significance Statement: Our work reveals the mechanism of flavonoid diversity and efficient synthesis in tea plants from the perspective of metabolon. Different metabolons modules provide a new perspective on the scientific problem of why tea plants accumulate high levels of EGCG. 19. Characterizations of PM Synchronous Motor With an Integrated Common-Mode Voltage Filter

Abstract

The high-frequency common-mode voltage generated by switching of semiconductor devices may cause premature failure of the insulation system and metallic bearings in the motor drive systems, the false trigger of the relay devices, and electromagnetic interference. In this article, an integrated filter is proposed to reduce the common-mode voltage in the permanent magnet synchronous motor. In this filter, partial motor windings, which serve as inductance, are integrated with the passive components. Compared to the system with a conventional passive filter, the omitted filtering inductor of this integrated common-mode voltage filter reduces the weight and volume of the whole motor drive system. The peak-to-peak value and the high-order harmonic components of common-mode voltage can be reduced to up to 10% and 2% of those in motor without the integrated filter, respectively. Additionally, the root mean square value of the common-mode voltage has been reduced by 27%. The benefits of the integrated filter in suppressing common-mode voltage have been validated in both simulation results and experimental data.

20. Fault-Tolerant Control for Single-Phase Open-Circuit and Short-Circuit Fault in Five-Phase PMSM With Third-Order Harmonic Back EMF Using Coefficients Reconfiguration

Abstract

For a five-phase permanent magnet synchronous machine (PMSM), the third-order harmonic back EMF is increased by changing the shape of the permanent magnet for increasing average torque in healthy operation. Additionally, in the open-circuit (OC) and short-circuit (SC) faults, the effect of injected third-order harmonic back EMF also needs to be considered. Therefore, this paper proposes a fault-tolerant control method with original transformation matrixes and coefficients reconfiguration in fundamental and third-order harmonic subspaces for both single-phase OC and SC faults. Fault tolerance can be applied by reconfiguring the relative coefficients after an OC or SC fault occurs, depending on the type of fault. Besides, the proposed method aims to optimize the OC fault tolerance strategies and effectively solve the single-phase SC fault, considering the influence in the third-order harmonic back EMF. According to experimental results, the torque fluctuation after applying the proposed method is similar to that in the healthy stage and much lower than that using most fault-tolerant control methods without considering the third-order harmonic back EMF.

Abstract

Five-phase machines represent a widely adopted choice for fault-tolerant applications due to their extra degrees of freedom. This article deals with the single-phase short-circuit fault tolerant control using original transformation matrix for five-phase permanent-magnet machines. The proposed method first follows the fundamental phase current injection (FCI) method based on the calculated SC current. Moreover, third-order harmonic phase current injection (THCI) method is presented to reduce copper loss. The computational method of derating factor is also adopted for industrial applications. The fault tolerance and dynamic performance are also verified by the experimental results. A reduction of copper loss in THCI method by about 15% is achieved, compared with general and FCI methods.

Abstract

Background and purpose: Liver fibrosis is a critical risk factor for the progression from chronic liver injury to hepatocellular carcinoma. Clinically, there is a lack of therapeutic drugs for liver fibrosis. Previous studies have confirmed that GL‐V9, a newly synthesized flavonoid derivative, exhibits anti‐inflammatory activity, but whether it has anti‐hepatic fibrosis actions remains unclear. This study aimed to investigate the anti‐fibrotic activities and potential mechanisms of GL‐V9. Experimental approach: Bile duct ligation (BDL) and carbon tetrachloride (CCl4) challenges were used to assess the anti‐fibrotic effects of GL‐V9 in vivo. Mouse primary hepatic stellate cells (pHSCs) and the human HSC line LX2 also served as a liver fibrosis model in vitro. Cellular functions and molecular mechanism were analysed using senescence‐associated beta‐galactosidase staining, real‐time PCR, western blotting, immunofluorescence, and co‐immunoprecipitation. Key results: GL‐V9 attenuated hepatic histopathological injury and collagen accumulation, as well as decreasing the expression of fibrotic genes in vivo. GL‐V9 promoted senescence and inhibited the expression of fibrogenic genes in HSCs in vitro. Mechanistic studies revealed that GL‐V9 induced senescence by upregulating GATA4 expression in HSCs. Further studies confirmed that GL‐V9 stabilized GATA4 by promoting autophagic degradation of P62. Conclusion and implications: GL‐V9 exerted potent anti‐fibrotic effects both in vivo and in vitro by stabilizing GATA4, thereby promoting the senescence of HSCs, and by avoiding its activation and ultimately inhibiting liver fibrosis. This action indicated that the flavonoid GL‐V9 is a potential therapeutic candidate for the treatment of liver fibrosis. [ABSTRACT FROM AUTHOR]

50. A Low-Complexity Modulated Model Predictive Torque and Flux Control Strategy for PMSM Drives Without Weighting Factor

Abstract

This article proposes a modulated model predictive torque and flux control (M2PTFC) method with low complexity for a two-level voltage source inverter (2L-VSI)-fed permanent magnet synchronous motor (PMSM). The proposed strategy aims to reduce the computation burden and simplify the control implementation of the conventional M2PTFC scheme by reducing the number of candidate voltage vectors at every control sample to the minimum (i.e., only two candidates) and eliminating the weighting factor of the cost function and its corresponding tuning procedures. For these purposes, the proposed method devotes different control objectives to the duty modulation and the cost function evaluation processes while executing them sequentially. First, aiming at torque ripple reduction, a duty modulation strategy is proposed based on the analysis of torque deviations produced by different voltage vectors, which ensures proper selection of the two candidates and restricts active voltage vectors (AVVs) that cause high deviations. Then, these candidates are evaluated based on a cost function assigned to achieve the stator flux control objective. The effectiveness of the proposed control scheme is verified through comparative assessment with the conventional M2PTFC and two existing simplified methods by means of simulation and experimental results.
